Dendritic spines are small, bulbous protrusions along neuronal dendrites where most of the excitatory synapses are located. Dendritic spine density in normal human brain increases rapidly before and after birth achieving the highest density around 2-8 years. Density decreases during adolescence, reaching a stable level in adulthood. The changes in dendritic spines are considered structural correlates for synaptic plasticity as well as the basis of experience-dependent remodeling of neuronal circuits. Alterations in spine density correspond to aberrant brain function observed in various neurodevelopmental and neuropsychiatric disorders. Dendritic spine initiation affects spine density. In this review, we discuss the importance of spine initiation in brain development, learning, and potential complications resulting from altered spine initiation in neurological diseases. Current literature shows that two Bin Amphiphysin Rvs (BAR) domain-containing proteins, MIM/Mtss1 and SrGAP3, are involved in spine initiation. We review existing literature and open databases to discuss whether other BAR-domain proteins could also take part in spine initiation. Finally, we discuss the potential molecular mechanisms on how BAR-domain proteins could regulate spine initiation.

How will the coronavirus disease 2019 (COVID-19) pandemic develop in the coming months and years? Based on an expert survey, we examine key aspects that are likely to influence the COVID-19 pandemic in Europe. The challenges and developments will strongly depend on the progress of national and global vaccination programs, the emergence and spread of variants of concern (VOCs), and public responses to non-pharmaceutical interventions (NPIs). In the short term, many people remain unvaccinated, VOCs continue to emerge and spread, and mobility and population mixing are expected to increase. Therefore, lifting restrictions too much and too early risk another damaging wave. This challenge remains despite the reduced opportunities for transmission given vaccination progress and reduced indoor mixing in summer 2021. In autumn 2021, increased indoor activity might accelerate the spread again, whilst a necessary reintroduction of NPIs might be too slow. The incidence may strongly rise again, possibly filling intensive care units, if vaccination levels are not high enough. A moderate, adaptive level of NPIs will thus remain necessary. These epidemiological aspects combined with economic, social, and health-related consequences provide a more holistic perspective on the future of the COVID-19 pandemic. (c) 2021 The Author(s). Vertaisarvioitu. Aivojen hermosolujen väliset kiihdyttävät signaalit saapuvat vastaanottaviin soluihin pääosin tuojahaarakkeiden okasten kautta. Yksilön kehittyessä hermosolujen välisten synapsiyhteyksien ja tuojahaarakkeiden okasten määrä alkuun lisääntyy, kunnes niiden määrä saavuttaa huippunsa noin yhden vuoden iässä. Tämän jälkeen okaset karsiutuvat aikuisikään asti ja muovautuvat dynaamisesti läpi aikuisiän. Synapsien lukumäärän lisäksi hermosolujen välisten kiihdyttävien ja estävien signaalien tasapaino on tärkeää normaalille aivotoiminnalle. Autismikirjon häiriöitä sairastavien aivoja tutkittaessa on todettu, että heidän okasensa ovat usein poikkeavan muotoisia ja niitä on normaalia enemmän. Autismikirjon häiriöön liitetyt poikkeavuudet synapsiyhteyksien ja okasten valinnassa sekä kiihdyttävien ja estävien synapsien tasapainossa ovat todennäköisesti yhteydessä aivojen tiedonkäsittelyn muutoksiin.

English summary

The axon initial segment (AIS) is the site of action potential initiation and plays an important role in maintaining neuronal polarity. Recent advances in super-resolution microscopy revealed the presence of an intricate membrane-associated periodic lattice in the AIS that contains sub-membranous actin rings periodically spaced ~190 nm and connected to a spectrin-ankyrin lattice. The precise function of these actin rings in unclear, as well as details of their structure and dynamics. The insensitivity of the AIS to actin-disrupting drugs led to the long-held view that actin is not a critical component of AIS structure. Here I show that the AIS contains a population of relatively stable, latrunculin-resistant actin filaments that are decorated by the tropomyosin isoform Tpm3.1. Disrupting these filaments through the perturbation of Tpm3.1 function led to the loss of accumulation of ankyrin G and other AIS markers, disruption of neuronal polarity, the loss of the clustering of voltage-gated sodium channels, and a rapid reduction in firing frequency. The findings I present in this thesis suggest that actin plays an important role in maintaining AIS structure and function, more important than previously appreciated. Hermosolut ovat pitkälle erikoistuneita soluja ja niiden eri haarakkeilla on erilaiset tehtävät. Tuojahaarakkeet tuovat signaalin muista hermosoluista hermosolun keskukseen, soomaan. Soomasta lähtee viejähaarake, joka vie signaalin taas eteenpäin. Viejähaarakkeen alussa, heti soomasta lähtiessä, on erityinen alue, jota suomeksi kutsutaan viejähaarakkeen alkuosaksi (englanniksi axon initial segment). Viejähaarakkeen alkuosa on tärkeä, sillä siinä päätetään, lähteekö signaali eteenpäin seuraaville hermosoluille. Viejähaarakkeen alkuosa myös säätelee mitkä solun rakennusaineet kuljetetaan viejähaarakkeisiin ja mitkä pidetään soomassa tai tuojahaarakkeissa. Näin solu pystyy erilaistamaan solun eri osat eri tehtäviä varten, tuoja- ja viejähaarakkeet on rakennettu eri rakennuskomponenteista. Viejähaarakkeen rakenne ja proteiinien järjestys on ensiarvoisen tärkeää viejähaarakkeen alkuosan tehokkaalle toiminnalle. Mikroskopiatekniikoiden kehittyminen viime vuosina on mahdollistanut viejähaarakkeen aktiinitukirangan rakenteen tarkemman tarkastelun. Aktiinitukiranka on solujen luusto ja lihakset, aktiinimolekyylit järjestäytyvät aktiinisäikeiksi, joista voi rakentaa soluun erilaisia rakenteita. Säätelystä riippuu miten pysyviä tai dynaamisia nämä rakenteet ovat. Väitöskirjassani selvitin viejähaarakkeen aktiinitukirangan rakennetta ja aktiinisäikeiden dynamiikkaa. Havaitsin että viejähaarakkeen alkuosan aktiinisäikeet ovat hyvin stabiileja. Tutkin myös miten aktiinisäikeisiin sitoutuva tropomyosiini 3.1 proteiini säätelee viejähaarakkeen alkuosan aktiinitukirankaa. Yllätyksekseni tämä tropomyosiini oli hyvin keskeinen koko viejähaarakkeen alkuosan rakenteen ylläpidolle. Kun poistin tropomyosiini 3.1 proteiinin, tai estin sen toimintaa hermosoluissa, tuhosin samalla viejähaarakkeen rakenteen ja toimivuuden, sekä rakennuskomponenttien kuljetuksen säätelyn, että signaalin eteenpäin viemisen. Aiemmin luultiin että aktiinitukiranka ei ole tärkeä viejähaarakkeen alkuosan rakenteelle tai toiminnalle. Tulokseni kuitenkin osoittavat, että aktiinitukirangan normaali rakenne on edellytys koko viejähaarakkeen rakenteen ja toiminnan ylläpidolle.

This thesis explores the role of several actin-binding proteins in the regulation of brain physiology with a focus on dendritic spines. Dendritic spines are considered the plausible physical substrate for learning and memory, as their morphology allows for modulating incoming signals. Disruptions in spine density and morphology are also often associated with neuropsychiatric disorders. The two cellular processes representing neuronal learning are long-term potentiation (LTP) and long-term depression (LTD). Here, I show that the actin-severing protein gelsolin transiently relocates to dendritic spines upon LTD induction, but not LTP induction or spontaneous neuronal activity. It is plausible that the modest – but relatively long-lasting – LTD-induced elevation of Ca2+ concentration increases the affinity of gelsolin to F-actin, thus inducing the relocalization of gelsolin to dendritic spines. Proper spine regulation is crucial for learning in live animals. MIM is an I-BAR containing membrane curving protein, shown to be involved in dendritic spine initiation and dendritic branching in Purkinje cells in the cerebellum. Behavioral analysis of MIM knock-out (KO) mice revealed defects in both learning and reverse-learning, alterations in anxiety levels and reduced dominant behavior, and confirmed the previously described deficiency in motor coordination and pre-pulse inhibition. Anatomically, we observed a decreased density of thin dendritic protrusions, enlarged brain ventricles and decreased cortical volume. Genetic studies have pointed out that genes often disturbed in neuropsychiatric disorders encode synaptic actin regulators. We selected five genes encoding different actin-regulating proteins and induced ASD-associated de novo missense mutations in these proteins. These mutations induced changes in the localization of α-actinin-4, which localized less to dendritic spines, and for SWAP-70 and SrGAP3, which localized more to dendritic spines. Among the wild-type proteins studied, only α-actinin-4 expression caused a significant change in dendritic spine morphology by increasing mushroom spine density and decreasing thin spine density. We hypothesized that mutations associated with ASD shift dendritic spine morphology from mushroom to thin spines. An M554V mutation in α-actinin-4 (ACTN4) resulted in the expected shift in dendritic spine morphology by increasing the density of thin spines. In addition, we observed a trend toward higher thin spine density with mutations in myosin IXb and SWAP-70. Myosin IIb and myosin IXb expression increased the proportion of inhibitory synapses in spines. The expression of mutated myosin IIb (Y265C), SrGAP3 (E469K), and SWAP-70 (L544F) induced variable changes in inhibitory synapses. Dendriittien okaset ovat pieniä nystyjä hermosolujen dendriittien (haarakkeiden) pinnalla. Okasissa sijaitsevat synapsit, eli hermosolujen liitokset, vastaanottavat signaaleja muista hermosoluista ja ovat näin hermosolujen toiminnan ytimessä. Okasten lisäyksen ja poiston on huomattu olevan yhteydessä oppimiseen. Useissa sairauksissa on myös havaittu, että okasten tiheys tai muoto ei ole normaali. Ymmärtääksemme syyt patologisten okasmuutosten taustalla, meidän pitää ensin ymmärtää miten okasten tiheyttä ja muotoa säädellään normaalisti. Väitöskirjatyössäni keskityin tutkimaan miten aktiinitukirankaa säätelevät proteiinit muokkaavat okasia. Aktiinitukiranka on solujen luusto ja lihakset, aktiinimolekyylit muodostavat erilaisia dynaamisia rakenteita, jotka sekä ylläpitävät solun olemassa olevaa muotoa että muuttavat sitä. Okasten muoto muuttuu solun aktivoinnin seurauksena. Jos hermosolua aktivoidaan nopealla taajuudella, okaset kasvavat ja muuttuvat pysyviksi. Jos taas matalalla taajuudella, okaset pienenevät ja saattavat jopa surkastua pois. Väitöskirjatyössäni tutkin ensin gelsoliini nimisen proteiinin käyttäytymistä erilaisissa hermosoluaktivoinneissa. Huomasin että gelsoliini siirtyy okasiin vain silloin kun hermosolua aktivoidaan matalalla taajuudella. Tämä tulos auttaa meitä ymmärtämään miten hermosolu voi vastata eri taajuuksilla tapahtuviin aktivointeihin eri tavoin. Tutkin myös, miten okasten muodostumisessa tärkeäksi havaitun MIM proteiinin poisto vaikuttaa hiirien oppimiseen. Tämä työ auttaa meitä ymmärtämään miten solutason muutokset vaikuttavat eläimen käyttäytymiseen. Tässä osatyössä havaitsimme myös, että MIM proteiinin poisto johti hiirillä suurentuneisiin aivokammioihin. Lopuksi selvitin miten autismikirjon häiriöön liitetyt mutaatiot aktiinitukirankaa säätelevissä proteiineissa vaikuttavat hermosolujen okasiin, sekä synapseihin, jotka hillitsevät hermosolun aktivoitumista. Tämä työ osoitti, että yhden aminohapon (proteiinin rakennuspalikka) muutos voi vaikuttaa yksittäisen proteiinin toimintaan niin paljon, että hermosolun muoto ja toiminta muuttuvat. Useat tutkituista mutaatioista muuttivat okasten muotoa ohuempaan suuntaan. Jos löydämme solumuutoksia, jotka toistuvat eri autismikirjohäiriön mutaatioille, pääsemme lähemmäksi ymmärrystä siitä, onko autismikirjon häiriön taustalla yhteistä, yksittäistä, biologista syytä.

Dendritic spines are small bulbous protrusions extending from dendritic shafts of neurons. These compartments house most of the postsynaptic terminals of excitatory synapses in the mammalian central nervous system. Dendritic spines are formed during early development and their density and morphology undergoes significant changes during maturation. After maturation dendritic spines are not static structures but display constant changes in their morphology and stability. The shape and size of dendritic spines have been linked to synaptic transmission, coupling the form of spines to neuron function. Several neurological diseases and disabilities are characterized by abnormal spine density and morphology. The main structural component of the dendritic spines is the actin filament, F-actin. Actin filaments are dynamic polymers of the monomeric protein actin. The filaments are constantly turning over and reorganizing. Both processes are regulated by actin binding proteins. All structural changes and the maintenance of dendritic spines are dependent on actin dynamics. Current research indicates that the dynamics of actin filaments do not change following spine maturation. Maturation does lead to a decrease in the movement of spines and an increase in stability, indicating changes in F-actin dynamics. In this study I have shown that the dynamics of F-actin do change during maturation. The stable pool of F-actin increases in size and the turnover of the dynamic pool increases. One of the actin binding proteins with a potential to regulate actin stabilization is myosin IIb, a motor protein with capabilities to bind F-actin and to introduce contractility into the filament network. Myosin IIb has been shown to regulate dendritic spine development, size and shape and play a role in memory consolidation. In this study I have shown that myosin IIb regulates dendritic spine F-actin via two distinct mechanisms. Myosin IIb can bind F-actin and stabilize it without affecting the turnover of the dynamic filaments. Myosin IIb-mediated contractility on the other hand can facilitate the turnover of the dynamic filaments. These findings help us to understand the molecular mechanism behind dendritic spine structure regulation and possibly in the future how it is related to synaptic transmission and different pathological states. Due to their small size, dendritic spines pose unique challenges for the study of actin dynamics. Most of the available methods are based on advanced fluorescence microscopy. In this study I have made a critical evaluation of the methods used to measure F-actin turnover in dendritic spines and the analysis of the data. I have also developed a novel approach to use fluorescence anisotropy to measure the level of actin bundling. The method has been previously applied to measure actin polymerization. My findings have led to the conclusion that in actin-dense compartments, such as the dendritic spines, fluorescence anisotropy reflects actin bundling rather than polymerization and that conclusions based on earlier research using similar techniques should be re-evaluated. Suurin osa nisäkkäiden keskushermoston eksitatoristen synapsien post-synaptisista terminaaleista sijaitsee okasissa. Tuojahaarakkeiden okaset muodostuvat aikaisessa yksilönkehityksen vaiheessa ja käyvät läpi suuria muutoksia tiheydessä ja morfologiassa kehityksen aikana. Myöskään kehittyneet okaset eivät ole staattisia rakenteita vaan ne muuttavat muotoaan jatkuvasti. Okasten koko ja muoto ovat yhteydessä synapsien voimakkuuteen, luoden näin yhteyden muodon ja toiminnan välille. Epänormaali okasten tiheys sekä muoto ovat tunnusomaista useille neurologisille taudeille ja häiriötiloille. Tuojahaarakkeiden okasissa tapahtuvia muutoksia pidetään myös yksinä aivoissa tapahtuvina muutoksina jotka liittyvät muistiin ja oppimiseen. Tuojahaarakkeiden okasten pääasiallinen rakenteellinen komponentti on aktiinitukiranka. Aktiinitukiranka muodostuu aktiinifilamenteista, jotka ovat aktiini nimisen proteiinin muodostamia polymeerejä. Nämä filamentit ovat erittäin dynaamisia. Dynamiikkaa säätelevät aktiinia sitovat proteiinit. Kaikki muutokset tuojahaarakkeiden okasten koossa, muodossa ja stabiliteetissa riippuvat aktiinifilamenttien dynamiikasta. Aikaisemmissa tutkimuksissa on osoitettu että okasten kehittyminen ei johda muutoksiin aktiinifilamenttien dynamiikassa okasissa. Kehittyminen johtaa kuitenkin muutoksiin okasten koossa, liikkeessä ja stabiliteetissa jotka viittaavat muutoksiin aktiini filamenttien dynamiikassa. Tässä tutkimuksessa olen osoittanut että okasten sisältämien aktiinifilamenttien dynamiikka muuttuu kehityksen kuluessa. Stabiilien filamenttien osuus ja dynaamisten filamenttien muutosnopeus kasvavat. Myosiini IIb on aktiinia sitova proteiini, joka voi kontrolloida aktiinifilamenttien dynamiikkaa kehityksen aikana. Myosiini IIb:n on osoitettu vaikuttavan okasten kehitykseen, kokoon, muotoon sekä muistojen muistamiseen. Tässä tutkimuksessa olen osoittanut että myosiini IIb säätelee okasten aktiinitukirankaa kahdella erillisellä mekanismilla. Sitoutumalla aktiinifilamentteihin vakauttaen niitä sekä kohdistamalla supistavaa voimaa filamenttijärjestelmään lisäten filamenttien muutosnopeutta. Tuojahaarakkeiden okasten pieni koko luo haastellisen ympäristön aktiinifilamenttien dynamiikan tutkimukselle. Menetelmät dynamiikan mittaamiseen perustuvat edistyneeseen valomikroskopiaan. Tässä tutkimuksessa olen tehnyt kriittisen arvion käytetyistä menetelmistä sekä data-analyysistä. Lisäksi esittelen uuden, fluoresenssin anisotropian mittaamiseen perustuvan, menetelmän filamenttien järjestyksen tutkimiseen. Menetelmää on aiemmin käytetty aktiinin polymerisaation tutkimuksessa. Havaintoihini perustuen voidaan kuitenkin tehdä johtopäätös että menetelmää voidaan käyttää aktiini filamenttien järjestyksen tutkimiseen aktiini-tiheissä solunosissa kuten tuojahaarakkeiden okasissa.
