The use of the term ‘living lab’ is now widespread, and in agrifood studies has been incorporated into research funding requirements. However, the methodological skills and prerequisites for successful living labs are not particularly well articulated. Based on our experience (five national and EU research projects) and our research backgrounds in (interdisciplinary) social science and participatory research, we highlight several risks associated with the use of living labs in agrifood studies without having the resources, reflectivity or skills required. These risks include: 1) imposing predefined agendas that are masked as participatory processes, 2) placing an overemphasis on confidentiality or knowledge sharing, and 3) getting lost in researcher roles. We argue that there is a need to discuss and improve understanding of these risks in order to produce successful living labs in agrifood studies, as well as avoid conceptual dilution or confusion.

BACKGROUND AND AIMS: Epidemiological studies have shown that subnormal levels of vitamin D (25(OH)D) are associated with a more aggravated clinical course of ulcerative colitis (UC). Despite an increased focus on the therapeutic importance of vitamin D and vitamin D receptor (VDR) signaling, the mechanisms underlying the effects of the vitamin D-VDR axis on UC remain elusive. Therefore, we aimed to investigate whether exposure to active vitamin D (1,25(OH)2D3)/VDR signaling in human organoids could influence the maintenance of the colonic epithelium.METHODS: Intestinal VDR expression was studied by immunohistochemistry, RNA expression arrays, and single-cell RNA sequencing of colonic biopsy specimens obtained from patients with UC and healthy individuals. To characterize the functional and transcriptional effects of 1,25(OH)2D3, we used patient-derived colonic organoids. The dependency of VDR was assessed by knocking out the receptor with CRISPR/Cas9.RESULTS: Our results suggest that 1,25(OH)2D3/VDR stimulation supports differentiation of the colonic epithelium and that impaired 1,25(OH)2D3/VDR signaling thereby may compromise the structure of the intestinal epithelial barrier, leading to flares of UC. Furthermore, a transcriptional response to VDR activity was observed primarily in fully differentiated cells at the top of the colonic crypt, and this response was reduced during flares of UC.CONCLUSIONS: We identified an important role of vitamin D signaling in supporting differentiated cell states in the human colonic epithelium, and thereby maintenance of the intestinal barrier integrity. This makes the vitamin D-VDR signaling axis an interesting target for therapeutic efforts to achieve and maintain remission in patients with UC.

Purpose: This study aimed to investigate patients' experiences with electronic Patient-Reported Outcome Measures (ePROMs) during follow-up consultations with registered nurses and surgeons in breast cancer care. Methods: A qualitative approach was employed, using focused ethnography with participant observations during patient consultations at a Plastic and Breast Surgery outpatient clinic, followed by individual interviews with the patients. Data were analysed using reflexive thematic analysis by Braun and Clarke. The discussion was informed by theory on person-centred practice. Results: A total of 38 participants were included. ePROMs strengthened person-centred practice by nudging patients to reflect on their health, voice their concerns and engage in active dialogue during consultations, particularly regarding body image issues. The relevance and impact of ePROMs were highly dependent on the timing of the patient's treatment trajectory. Patients found that ePROMs were more meaningful and contextually appropriate when completed during late follow-up. This could be because patients faced challenges with ePROMs, including not understanding their purpose, the need for assistance from relatives and instances when ePROMs were not integrated into consultations. Conclusion: ePROMs can empower patients by facilitating meaningful discussions about body image and other concerns during postoperative follow-up consultations. However, their effectiveness relies on clear communication and proper integration into clinical practice. The results add to existing litterature by highlighting the dual impact of ePROMs, as they can enhance patient-centred care but also present challenges when their purpose is unclear. The implications for future practice include the need to develop strategies that engage diverse patient groups in the ePROM process, supporting equal access to follow-up care and addressing health disparities.

This study investigated the efficacy of a 10-session nature-based therapeutic intervention for people with post-concussion symptoms. The intervention involved physical and vestibular exercises, sensory training, relaxation, and psychoeducation, all of which were integrated with the natural environment in a forest therapy garden. This study was designed with a passive control period followed by the intervention (n = 30). The Mental Fatigue Scale (MFS) was the primary outcome measure. The secondary outcome measures were the Warwick–Edinburg Mental Wellbeing Scale and the short version of the Quality of Life after Brain Injury. A Likert scale was used to examine the mental strain of the sessions themselves. The MFS (primary outcome) exhibited a significant decrease with a medium-sized effect from before to after the intervention. The secondary outcomes exhibited significant increases from the beginning to the end of the intervention. All outcomes were sustained at follow-up ten weeks later. No significant difference was found from the control period. This study indicates that the described nature-based intervention is a feasible treatment for reducing prolonged post-concussion symptoms. However, it should be studied more in-depth to understand the impact of the natural environment and to validate the results on a larger representative population.

Semi-synthetic cannabinoids (SSCs) are becoming more popular as legal replacement of tetrahydrocannabinol (THC). Products containing hexahydrocannabinol (HHC), hexahydrocannabiphorol (HHC-P), tetrahydrocannabiphorol (THC-P) and tetrahydrocannabidiol (H4-CBD) are readily available in e.g. kiosks or online shops. As soon as a compound becomes illegal, both as narcotics and regarding driving under the influence of drugs (DUID), the next analogue takes over the market. Therefore, it is difficult to predict what should be detectable in DUID cases. In Copenhagen, both analysis of seized drugs and biological samples are being performed at the Section of Forensic Chemistry. The information regarding which compounds are found in seized materials can be valuable for the toxicological investigations of forensic cases. At our section, HHC was first observed in seized materials in 2022. It was present in 135 seized materials received from police and customs in Eastern Denmark in 2022–2023. In the same period, HHC-P, THC-P and H4-CBD were detected in 20, 9 and 53 seized materials, respectively. With this information in mind, we developed an analytical strategy for including SSCs in our LC-MS/MS analysis of biological samples, including those from DUID cases. HHC was scheduled as a narcotic substance in Denmark in May 2023 followed by HHC-P, THC-P and H4-CBD in January 2024. Of the earlier mentioned seized materials containing HHC, 33% of these were collected after it was declared illegal. HHC-P, THC-P and H4-CBD only became prominent after illegalization of HHC, where 94% of the mentioned cases were confiscated after HHC was prohibited. Targeted screening of the new SSCs were included in our routine LC-MS/MS analysis for quantification of THC in blood from DUID cases. Glucagon-like peptide 1 (GLP-1) receptor agonists can decrease alcohol intake by central mechanisms that are still poorly understood. The lateral septum (LS) and the ventral/caudal part of the hippocampus are enriched in GLP-1 receptors, and activity in these regions was shown to modulate reward-related behaviors. Using microinfusions of the GLP-1 receptor agonist exendin-4 in mice trained to self-administer oral alcohol in an operant assay, we tested whether pharmacological stimulation of GLP-1 receptors in hippocampus and LS decrease alcohol self-administration. We report that infusion of exendin-4 in the ventral hippocampus or LS was sufficient to reduce alcohol self-administration with as large effect sizes as we previously reported with systemic exendin-4 administration. Infusion of exendin-4 into the nucleus accumbens also reduced alcohol self-administration, as anticipated based on earlier reports, while infusion of exendin-4 into the caudate-putamen (dorsal striatum) had little effect, consistent with lack of GLP-1 receptor expression in this region. The distribution of exendin-4 after infusion into the LS or caudate putamen was visualized using a fluorescently labeled ligand. Transitioning towards more sustainable food products, such as plant proteins, requires a change in practice by several actors in the agrifood system. Change of this kind involves everyday choices about what food to produce, sell, prepare, and eat. Inspired by science and technology studies (STS) thinking, we investigate how such choices are influenced by socio-material practices of valuing. We use the case of fresh grain legumes for human consumption to explore how valuing is simultaneously affected by and shapes the agrifood system. Through interviews with 24 actors in the Danish agrifood system, we identify valuing parameters ranging from taste, nitrogen fixation, durability, and nutrition to price. The study reveals differences regarding what and how actors value depending on the actors’ position in the agrifood system and how the fresh grain legumes travel from field to plate. Where values conflict, we observe how some valuing practices have the power to exclude others and thereby prevent specific enactments of the fresh grain legumes. We argue that looking for valuing practices can help us understand how agrifood systems come into being, and that valuing differently can represent active involvement, both academically and practically, in encouraging change in the agrifood system. By using STS-thinking, the study brings novel insights about barriers towards more plant-based diets and contributes to the diversification of theoretical perspectives on sustainable transitions.

Introduction Behavioural and psychological symptoms of dementia (BPSD) should only rarely and briefly be treated with antipsychotics. Despite recommendations to the contrary, the use of antipsychotics in nursing home residents with dementia is widespread and followed by serious adverse effects. Intervention studies on methods to reduce the use of antipsychotics in persons with dementia are few and needed. The aim of this protocol is to describe the rationale and content of the intervention DEprescribing and Care to reduce Antipsychotics in DEmentia (DECADE)-a hybrid effectiveness-implementation pilot study. Materials and methods This is a protocol of a prospective hybrid effectiveness-implementation pilot study. The primary aim of DECADE is to reduce the use of antipsychotic drugs by 50% in 50% of nursing home residents with dementia while maintaining or improving BPSD. The intervention is implemented in six nursing homes including approximately 190 residents with dementia and consists of Academic Detailing, medication review, education of nursing home staff, and care plans. The evaluation of feasibility and potential effectiveness is an overall assessment of all clinical and process outcomes. Logistic regression analyses will be used to investigate factors characterizing situations with prescription of antipsychotics. BPSD is analysed with a before- and after design using self-controlled case series methods and the use of antipsychotics is analysed as interrupted time series. Discussion This protocol describes a study that will provide an indication of DECADE effectiveness and a model for upscaling and further evaluation in a controlled design.

Generation of functionally mature organs requires exquisite control of transcriptional programs governing cell state transitions during development. Despite advances in understanding the behavior of adult intestinal stem cells and their progeny, the transcriptional regulators that control the emergence of the mature intestinal phenotype remain largely unknown. Using mouse fetal and adult small intestinal organoids, we uncover transcriptional differences between the fetal and adult state and identify rare adult-like cells present in fetal organoids. This suggests that fetal organoids have an inherent potential to mature, which is locked by a regulatory program. By implementing a CRISPR-Cas9 screen targeting transcriptional regulators expressed in fetal organoids, we establish Smarca4 and Smarcc1 as important factors safeguarding the immature progenitor state. Our approach demonstrates the utility of organoid models in the identification of factors regulating cell fate and state transitions during tissue maturation and reveals that SMARCA4 and SMARCC1 prevent precocious differentiation during intestinal development.

During intestinal organogenesis, equipotent epithelial progenitors mature into phenotypically distinct stem cells that are responsible for lifelong maintenance of the tissue. While the morphological changes associated with the transition are well characterized, the molecular mechanisms underpinning the maturation process are not fully understood. Here, we leverage intestinal organoid cultures to profile transcriptional, chromatin accessibility, DNA methylation, and three-dimensional (3D) chromatin conformation landscapes in fetal and adult epithelial cells. We observed prominent differences in gene expression and enhancer activity, which are accompanied by local changes in 3D organization, DNA accessibility, and methylation between the two cellular states. Using integrative analyses, we identified sustained Yes-Associated Protein (YAP) transcriptional activity as a major gatekeeper of the immature fetal state. We found the YAP-associated transcriptional network to be regulated at various levels of chromatin organization and likely to be coordinated by changes in extracellular matrix composition. Together, our work highlights the value of unbiased profiling of regulatory landscapes for the identification of key mechanisms underlying tissue maturation.

Drug concentrations in peripheral blood are often used to evaluate whether death was caused by drug intoxication. In some cases, peripheral blood is not available, and analytical results of alternative matrices should instead be used in the toxicological evaluation. However, reference concentrations of alternative matrices are few, which makes interpretation of results a challenge. In this study, concentrations of selected benzodiazepines, opioids, illicit drugs, and other commonly used drugs in postmortem femoral blood, cardiac blood, brain tissue, and muscle tissue are presented. Alternative matrix-to-femoral blood drug concentration ratios and correlations of blood and alternative matrix drug concentrations were calculated to examine which of the investigated alternative matrices were most suited to use for toxicological evaluation in cases where peripheral blood is not available. The results showed that concentrations in cardiac blood, brain tissue, and muscle tissue could be useful in the postmortem evaluation of most of the 19 selected analytes. In most cases, analytes were detected in all the alternative matrices. The median concentration ratios for the selected analytes in brain tissue, cardiac blood, and muscle tissue relative to femoral blood ranged from 0.57 to 3.42, 0.59 to 1.87, and 0.67 to 7.04, respectively. Overall, cardiac blood provided the concentrations most comparable with femoral blood concentrations, indicating that cardiac blood can be useful in cases where femoral blood is not available. People and places are entangled through material, social, discursive and emotional relations, which makes it complex to understand the daily living in a certain place. In this article, we explore such entanglements, with an outset in two underprivileged neighbourhoods in Denmark, by drawing on two sets of theoretical thinking: The framework of multiplicity, and the concept of place. We argue that the neighbourhoods cannot be defined as either good or bad places of living. Rather they exist in complex ways, where what makes some people feel at home, is what makes others feel insecure, and where such ambivalent feelings can co-exist inherently for individual residents. Furthermore, our analysis shows that manifold enactments of residents and places enable multiple trajectories of how to live and identify oneself in relation to the neighbourhood.

Research has confirmed that natural environments can offer many health benefits. However, a large group in society, people with mobility disabilities, are often hindered from gaining these benefits. Enhancing physical accessibility to natural environments is important, but in our opinion, not enough. This position paper addresses the importance of incorporating social inclusiveness in accessibility solutions in natural environments. We present a framework based on theories on affordances and behaviour settings that can guide the design process of inclusive, accessible, natural, and health-promoting environments for people with mobility disabilities. The development of the framework is based on quantitative and qualitative results achieved from a several-year-long research project concerning the design and use of a forest trail for people with mobility disabilities. The framework allows for the integration of two key opposing factors: naturalness and accessibility. The framework also integrates the important and often overlooked factor of social inclusion when designing health-promoting natural environments. Landscape architects must understand their project sites as not only physical spaces, but also as social and emotional spaces. Implementing this framework can support our endeavours to achieve the Sustainable Development Goals and the Leave No One Behind agenda.

Recent developments and research in modern machine learning have led to substantial improvements in the geospatial field. Although numerous deep learning models have been proposed, the majority of them have been developed on benchmark datasets that lack strong real-world relevance. Furthermore, the performance of many methods has already saturated on these datasets. We argue that shifting the focus towards a complementary data-centric perspective is necessary to achieve further improvements in accuracy, generalization ability, and real impact in end-user applications. This work presents a definition and precise categorization of automated data-centric learning approaches for geospatial data. It highlights the complementary role of data-centric learning with respect to model-centric in the larger machine learning deployment cycle. We review papers across the entire geospatial field and categorize them into different groups. A set of representative experiments shows concrete implementation examples. These examples provide concrete steps to act on geospatial data with data-centric machine learning approaches.
