Abstract

Cydalima perspectalis (Walker 1859), the boxwood moth, is one of the important invasive pests spreading rapidly throughout Europe within a few years. The objective of the current research was to determine an alternative biological control agent for the control of C. perspectalis. Beauveria bassiana, an entomopathogenic fungus with high insecticidal activity, was isolated from the larvae of the pest that causes substantial economic losses on boxwood (Buxus sempervirens) in the eastern Black Sea region of Turkey, and its virulence was tested on it. Based on the morphological data and molecular characteristics, including the partial sequence of ITS (ITS1- 5.8S-ITS2 gene region) and EF1, the fungus was identified as an isolate of B. bassiana (CP1). In larval stage susceptibility tests, it was determined that larval mortality was higher in older stages. In concentration-response experiments using 1 × 104-8 conidia/ml, the LC50 value of the new isolate was calculated to be 1.7 × 104 conidia/ml within 10 days against C. perspectalis 6th instar larvae under laboratory conditions. This is a significant study to describe a novel B. bassiana CP1 isolate, and preliminary data suggests that the fungal isolate has considerable potential as an effective biopesticide for controlling C. perspectalis. Abstract

Amaç: Bu çalışmada, 2010-2020 yılları arasında Trabzon Büyükşehir Belediyesi İtfaiye Dairesi Başkanlığı tarafından müdahale edilen yangınların incelenmesi ve elde edilen sonuçlara göre bilimsel öneriler sunulması amaçlanmıştır.Gereç ve Yöntem: Veriler, Trabzon Büyükşehir Belediyesi İtfaiye Dairesi Başkanlığına ait 1 Ocak 2010 ile 1 Ağustos 2020 tarihleri arasında kayıt altına alınan olay raporları incelenerek elde edilmiştir. Çalışmada, personel, araç sayıları, müdahale edilen olay sayıları, ölü ve yaralı sayıları, yıllık yangın sayıları, yangın türleri, yapı türleri, yangın nedenleri, yangınların aylara göre dağılım, yanma dereceleri ve eğitim sayıları incelenmiş ve basit frekans analizi ile değerlendirilmiştir. Bulgular: Trabzon Büyükşehir Belediyesi İtfaiye Dairesi Başkanlığının 2010 yılında 66 personel (%3,8) ve 16 (%2,8) araç ile 2020 yılında ise 265 personel (%15,1) ve 77 (%13,4) araç ile olaylara müdahale ettiği tespit edilmiştir. Meydana gelen yangınların dağılımı ve yanma dereceleri incelendiğinde; yangın olayının en fazla 1668 (%13,8) adet ile 2019 yılında yaşandığı tespit edilmiştir. Meydana gelen yangınlar, yangın türlerine göre incelendiğinde; ilk üç sırayı sırasıyla, 5135 (%56,27) adet ile bina yangınlarının 2070 (%22,68) adet ile ot-saman-ekin yangınlarının ve 844 (%9,25) adet ile motorlu araç yangınlarının aldığı tespit edilmiştir.Sonuç: Elde edilen bulgular ile uygulamaya, literatüre ve politika üreticilerine öneriler geliştirilecektir. Yapılan raporlamalar ışığında bu konulardaki eksiklikler belirlenebilir ve bu konularda farkındalık eğitimleri düzenlenebilir., Objective: In this study; it was aimed to examine the fires intervened by Trabzon Metropolitan Municipality Fire Department between the years of 2010-2020 and to present scientific suggestions according to the obtained results.Materials and Methods: The data were obtained by examining the incident reports of Trabzon Metropolitan Municipality Fire Department, recorded between January 1, 2010 and August 1, 2020. In the study, the number of personnel, vehicle numbers, the number of intervened incidents, the number of deaths and injuries, the number of annual fires, types of fires, types of buildings, fire causes, distribution of fires by months, degrees of burning and number of training were examined and evaluated with simple frequency analysis.Results: It was determined that Trabzon Metropolitan Municipality Department of Fire Brigade intervened with 66 people (3.8%) and 16 (2.8%) vehicles in 2010 with 265 personnel (15.1%) and 77 (13.4%) vehicles in 2020. When the distribution of fires and their burning degrees are examined; it was determined that the most fire incidents occurred in 2019 with the number of 1668 (13.8%). Examining the occurred fires according to their types; it was identified that the first three ranks were 5135 (56.27%) building fires, 2070 (22.68%) grass-straw-crop fires and 844 (9.25%) motor vehicle fires.Conclusion: With the obtained findings, suggestions for practice, literature and policy makers will be developed. In the light of the reports, deficiencies on these issues can be identified and awareness training can be organized on these issues.

11. Determination of fungal pathogens of Hypera postica(Gyllenhall) (Coleoptera: Curculionidae): isolation, characterization, and susceptibility

Abstract

Fungal pathogens of Hypera postica(Gyllenhall) (Coleoptera: Curculionidae) were collected from the vicinities of Adana and Igdir in Turkey. The pathogenicity of the fungal isolates against the pest were investigated. According to morphologic (colony morphology, spore shape) and molecular (sequences of ITS1-5.8S ITS2 region and EF1-α, Bloc, and RPB1 genes) characterizations, the isolates were identified as Beauveria bassiana(HpI-2, HpI-6, HpI-7, HpI-10, HpA-3, HpA-4, HpA-5) and Beauveria pseudobassiana(HpI-4). All these strains were isolated from H. posticafor the first time. In order to determine pathogenesis of all isolates on the target pest, bioassays were conducted against larvae and adults, as screening of (1 × 107conidia/ml) and dose-response (1 × 105, 1 × 106, 1 × 107, 1 × 108, conidia/ml), under laboratory conditions. The fungal isolates, closely related to each other, yielded significantly varied mortalities on larvae and adults. H. posticalarvae were found more susceptible than adults to the fungal isolates in all tests. The highest mortality rates (100 and 98%) for larvae and adults, respectively, were obtained by B. bassianastrain HpA-5 within 14 days at 1 × 108conidia/ml concentration. The median lethal concentration (LD50) of HpA-5 required to kill the larvae and adults of H. posticaat concentrations of 2.37 × 104and 1.4 × 105conidia/ml, respectively. These results are promising; therefore, the B. bassianastrain HpA-5 can potentially be used against H. postica.

12. Development of novel betabaculovirus (HycuGV-Hc1) as a biopesticide (HycuGV-TR61) and its efficacy on the fall webworm, Hyphantria cuneaDrury (Lepidoptera: Erebidae) larvae

Abstract

Background: The aim of this study was to develop an oil formulation from a local betabaculovirus. Hyphantria cuneaDrury (Lepidoptera: Erebidae) granulovirus (HycuGV-Hc1) was isolated from the infected larvae to test its efficacy on the pest. The oil formulation was prepared by mixing the viral suspension with sunflower oil and some adjuvants and named HycuGV-TR61. Crude virus and the formulations were carried out on third instar H. cunealarvae using 1 × 104–8OBs /ml concentrations and were exposed to temperatures (28, 35, and 42 °C) and UV-B light at different periods (0, 1, 3, 5 h). Results: The mortality rate, which was 50% at the lowest concentration (1 × 104OB/ml), reached 99.86% at the highest concentration (1 × 108OB/ml). LC50values of fresh and old forms were calculated as 0.64 × 104and 0.87 × 104OBs/ml, respectively. Application of shelf life showed that there was non-significant change in the pathogenic activity of the formulation with time. In the experiments, it was observed that the activity decreased as the temperature and time of exposure increased. Significantly difference in larval mortality was observed when fresh and old formulations were exposed to 0, 1, 3 and 5 h to UV-B, (old: 96.7, 86, 80 and 60%; fresh: 97.1, 90, 85 and 62%, respectively). Conclusions: The results revealed superior aspects of HycuGV-TR61, which was developed as a local viral biopesticide, its resistance to abiotic factors and its potential to be used in pest control.
