Abstract

The Eastern Mediterranean resides on the border between the temperate and semi-arid and arid climate zones, and is thus influenced by both mid-latitude and sub-tropical weather systems. Precipitation and extreme weather in this region are mainly associated with either Cyprus Lows or the “wet” Red Sea Troughs. Current regional climate projections indicate that the region may become warmer and drier in future decades. Here, we analyze the influence of enhanced greenhouse gas forcing on the climatological properties of the ‘wet’ and ‘dry’ Red Sea Trough (WRST & DRST, respectively). With this aim, a regional synoptic classification and a downscaling algorithm based on past analogs are applied to eighteen rain stations over the main ground water basins in Israel. The algorithms are applied to the NCEP/NCAR reanalysis data for 1986–2005 and to eight CMIP5 model simulations for the historical (1986–2005) and end of the century (2081–2100) climate conditions according to the RCP8.5 scenario. For the historical period, the CMIP5 models are largely able to represent the characteristics of the Red Sea Trough. Based on the multi-model mean, significant changes are found for WRST and DRST for the late XXI Century. First, an increase in the meridional pressure gradient is found for both the WRST and the DRST, implying stronger horizontal winds. Furthermore, a significant decrease in the occurrence of the WRST (− 20%) and a significant increase in the frequency of the DRST (+ 19%) are identified. Accordingly, the persistence of the WRST decreases (− 9%), while for DRST increases (+ 9%). The decline in the frequency of WRST occurs primarily in the transition seasons, while the increase for DRST is found throughout the wet season. In total, the daily rainfall associated with the WRST system is projected to significantly decline (− 37%) by the end of the XXI century. These results document the projected changes in a dominant synoptic system in this area, which can facilitate a better estimation of the arising challenges, e.g., related to shortage of water resources and associated political unrest, reduced agricultural potential, and increased air pollution and forest fires. Such a pathway can ultimately foster novel mitigation strategies for water resources management and regional climate change adaptation.

Abstract

Large Battery Energy Storage (BES) facilities may provide significant dynamic operation benefits for electric utilities. One area in which a BES facility could be useful is the frequency regulation requirement. This feature is significantly important for island power systems. The purpose of this paper is to quantify the effects of a 30 MW battery on the frequency regulation in the Israeli isolated power system. The study was performed on a single area model representing the whole power system and containing a first order transfer function that represented the BES performance.

Abstract

Alteration of platelet function contributes to microthrombus formation and may play an important role in the pathogenesis of diabetic micro- and macroangiopathies. However, the molecular mechanism for platelet dysfunction observed in patients with diabetes has not been fully elucidated. In this study, the direct effects of hyperglycemia on platelet function in vitro were investigated. Hyperglycemia increased reactive oxygen species generation in human platelets, and this effect was additive with that of collagen. Thenoyltrifluoroacetone (TTFA), an inhibitor of mitochondrial electron transport chain complex II, and carbonyl cyanide m-chlorophenylhydrazone (CCCP), an uncoupler of oxidative phosphorylation, completely prevented the effects of hyperglycemia, suggesting that reactive oxygen species arise from the mitochondrial electron transport chain. Hyperglycemia potentiated both platelet aggregation and the subsequent release of platelet-derived growth factor AB induced by a nonaggregating subthreshold concentration of collagen, which were also completely inhibited by TTFA or CCCP. Furthermore, hyperglycemia was found to inhibit protein tyrosine phosphatase (PTP) activity and increase phosphorylation of the tyrosine kinase Syk in platelets exposed to collagen. Hyperglycemia-induced PTP inhibition and Syk phosphorylation were found to be completely prevented by TTFA, CCCP, or Mn(III)tetrakis (4-benzoic acid) porphyrin, a stable cell-permeable superoxide dismutase mimetic. These results suggest that hyperglycemia-induced mitochondrial superoxide generation may play an important role in platelet dysfunction observed in patients with diabetes. [ABSTRACT FROM AUTHOR]

34. Quantitation of chemopreventive synergism between (-)-epigallocatechin-3-gallate and curcumin in normal, premalignant and malignant human oral epithelial cells.

Abstract

An in vitro model for oral cancer was used to examine the growth inhibitory effects of chemopreventive agents when used singly and in combination. The model consists of primary cultures of normal oral epithelial cells, newly established cell lines derived from dysplastic leukoplakia and squamous cell carcinoma. Two naturally occurring substances, (-)-epigallocatechin-3-gallate (EGCG) from green tea and curcumin from the spice turmeric were tested. Cells were treated singly and in combination and effects on growth determined in 5-day growth assays and by cell cycle analysis. Effective dose 50s and the combination index were calculated with the computerized Chou-Talalay method which is based on the median-effect principle. Agents were shown to differ in their inhibitory potency. EGCG was less effective with cell progression; the cancer cells were more resistant than normal or dysplastic cells. In contrast, curcumin was equally effective regardless of the cell type tested. Cell cycle analysis indicated that EGCG blocked cells in G1, whereas curcumin blocked cells in S/G2M. The combination of both agents showed synergistic interactions in growth inhibition and increased sigmoidicity (steepness) of the dose-effect curves, a response that was dose and cell type dependent. Combinations allowed for a dose reduction of 4.4-8.5-fold for EGCG and 2.2-2.8-fold for curcumin at ED50s as indicated by the dose reduction index (DRI). Even greater DRI values were observed above ED50 levels. Our results demonstrate that this model which includes normal, premalignant and malignant oral cells can be used to analyse the relative potential of various chemopreventive agents. Two such naturally-occurring agents, EGCG and curcumin, were noted to inhibit growth by different mechanisms, a factor which may account for their demonstrable interactive synergistic effect. Abstract

Leptin, a circulating hormone secreted mainly from adipose tissues, is involved in the control of body weight. The plasma concentrations are correlated with body mass index, and are reported to be high in patients with insulin resistance, which is one of the major risk factors for cardiovascular disease. However, the direct effect of leptin on vascular wall cells is not fully understood. In this study, we investigated the effects of leptin on reactive oxygen species (ROS) generation and expression of monocyte chemoattractant protein-1 (MCP-1) in bovine aortic endothelial cells (BAEC). We found that leptin increases ROS generation in BAEC in a dose-dependent manner and that its effects are additive with those of glucose. Rotenone, thenoyltrifluoroacetone (TTFA), carbonyl cyanide m-chlorophenylhydrazone (CCCP), Mn(III)tetrakis (4-benzoic acid) porphyrin (MnTBAP), uncoupling protein-1 (UCP1) HVJ-liposomes, or manganese superoxide dismutase (MnSOD) HVJ-liposomes completely prevented the effect of leptin, suggesting that ROS arise from mitochondrial electron transport. Leptin increased fatty acid oxidation by stimulating the activity of carnitine palmitoyltransferase-1 (CPT-1) and inhibiting that of acetyl-CoA carboxylase (ACC), pace-setting enzymes for fatty acid oxidation and synthesis, respectively. Leptin-induced ROS generation, CPT-1 activation, ACC inhibition, and MCP-1 overproduction were found to be completely prevented by either genistein, a tyrosine kinase inhibitor, H-89, a protein kinase A (PKA) inhibitor, or tetradecylglycidate, a CPT-1 inhibitor. Leptin activated PKA, and the effects of leptin were inhibited by the cAMP antagonist Rp-cAMPS. These results suggest that leptin induces ROS generation by increasing fatty acid oxidation via PKA activation, which may play an important role in the progression of atherosclerosis in insulin-resistant obese diabetic patients.

Abstract

The concentration of apolipoproteins was measured by quantitative immunoelectrophoresis in rat serum, in the lipoprotein-free ultracentrifugal fraction (density greater than 1.21) of serum, and in renal lymph. The A-IV and arginine-rich apolipoproteins were present in high concentrations (greater than 18.5% of serum concentration) both in the fraction of density greater than 1.21 and in renal lymph, whereas the other apolipoproteins were found in low concentrations ( less than 7% of serum concentration). The major apolipoproteins of renal lymph were similar to those found in the fraction of density greater than 1.21; however, the apolipoproteins composition of the renal lymph was very different from the apolipoprotein composition of serum lipoproteins. The presence of certain apolipoproteins in the fraction of density greater than 1.21 and in renal lymph suggests that these apolipoproteins might be present in the circulation as "free" apolipoproteins. The possible physiological importance of these specific apolipoproteins is postulated.

Abstract

Apoproteins of high density lipoproteins were detected in the urine of normal subjects after the urinary proteins were highly concentrated. By immunoelectrophoresis, all of the urinary apoproteins gave precipitin lines with similar electrophoretic mobility. This suggests that the various apoproteins are present in the same particle. The apoproteins were present only in the ultracentrifugal fraction of density greater than 1.24 g/ml. Neither apoprotein B nor apoprotein E were detected in the urine, suggesting that very low density and low density lipoproteins are not excreted in the urine of normal subjects.
