1. A simple and fast HPLC method for determining the composition of fructooligosaccharides and xylooligosaccharides obtained by fungal enzymes

- Abstract
The growing demand for functional food products, particularly prebiotics, underscores the need for advancements in the production processes and analytics of both conventional and novel oligosaccharides. This study introduces a tailored HPLC-RID method for the detection of two prominent classes of prebiotics: fructooligosaccharides, a well-established group, and xylooligosaccharides, an emerging category. This method is the first described method for the analysis of more than one type of OS, with complete validation, and is preferred due to its ease of use and affordability. Rigorous validation of this method adheres to the International Council on Harmonization guidelines. Five components of FOS and six of XOS were quantified, with LOQ ranging from 0.55 to 1.64 mg/mL for FOS and from 0.16 to 1.00 mg/mL for XOS, and good linearity correlation (R2) 0.9807–0.9994 (FOS) or 0.9884–0.9999 (XOS). The calculated accuracy was in the range: 94.87–100.82 (FOS) or 97.87–105.41 (XOS). The proposed method was validated in successfully detecting and quantifying FOS and XOS samples, even within complex matrices enriched with interfering compounds, including a few food samples.
