Abstract

The cytoplasmic Purkinje onconeural antigen cdr2 found to down-regulate c-Myc function by sequestering c-Myc in neuron cytoplasm is discussed. A study has shown that cdr2 interacts specifically with the helix-leucine zipper (HLZ) motif of c-Myc and that cdr2 is a cytoplasmic protein, an onconerual antigen, which has an HLZ motif as well. An immune response to cdr2, associated with tumor immunity and autoimmune cerebeller degeneration, is led to by expression of cdr2, normally expressed in cerebellar Purkinje neurons. Autoantibodies in PCD, paraneoplastic cerebellar degeneration, may contribute to Purkinje neuronal death. These findings have implications for tumor cell and neuronal cell survival.

Abstract

Abstract Background Accumulation of amyloid-β (Aβ) peptide in the brain is thought to play a key pathological role in Alzheimer's disease. Many pharmacological targets have therefore been proposed based upon the biochemistry of Aβ, but not all are equally tractable for drug discovery. Results To search for novel targets that affect brain Aβ without causing toxicity, we screened mouse brain samples from 1930 novel gene knock-out (KO) strains, representing 1926 genes, using Aβ ELISA assays. Although robust Aβ lowering was readily apparent in brains from a BACE1 KO strain, none of the novel strains exhibited robust decreases in brain Aβ, including a GPR3 KO strain, which had previously been proposed as an Aβ target. However, significantly increased Aβ was observed in brain samples from two KO strains, corresponding to genes encoding the glycosylphosphatidylinositol mannosyl transferase PIGZ and quinolinate phosphoribosyltransferase (QPRT). Conclusions Thus, gene ablations that are permissive for mouse survival and that also have a robust effect on Aβ levels in the brain are rare.

Abstract

The Adult Mouse Anatomical Dictionary was developed to provide an ontology for standardized nomenclature for anatomical terms in the postnatal mouse. The ontology will be used to annotate and integrate different types of data pertinent to anatomy., We have developed an ontology to provide standardized nomenclature for anatomical terms in the postnatal mouse. The Adult Mouse Anatomical Dictionary is structured as a directed acyclic graph, and is organized hierarchically both spatially and functionally. The ontology will be used to annotate and integrate different types of data pertinent to anatomy, such as gene expression patterns and phenotype information, which will contribute to an integrated description of biological phenomena in the mouse.

Abstract

The BCL6 transcription repressor (B-cell lymphoma 6, BCL6) protein has been shown to be a key molecular driver of diffuse large B-cell lymphoma (DLBCL). Somatic mutations of the BCL6gene include gross- or cryptic-chromosome translocations and point mutations that have been shown to result in the deregulation of BCL6expression. These BCL6 abnormalities also contribute to a subgroup of high-risk (HR) aggressive double- and triple-hit (DH/TH) lymphomas with very poor outcomes. We have developed highly specific, potent and orally bioavailable BCL6 PROteolysis TArgeting Chimera (PROTAC TM) degraders that demonstrate potent in-vitroand in-vivoefficacy in multiple pre-clinical DLBCL models.
