Abstract Entrepreneurship is of great significance to individuals, families and society. Recently, intrapreneurship, i.e., business ventures within established organisations, has also attracted widespread interest among individuals and organisations. However, we still know little about how individuals make decisions when they face diverse types of entrepreneurial activities. Based on theories of entrepreneurial action and conservation of resources and the literature on family embeddedness, this paper proposes an integrated framework for entrepreneurial choice—including intrapreneurship, self-employment and non-entrepreneurship, and examines the roles of socio-cognitive traits and family contingency factors in the entrepreneurial choice process. By using secondary and survey data, the empirical results show that (a) entrepreneurial alertness (EA) and self-efficacy (ESE) both positively affect individuals’ choice towards intrapreneurship and self-employment, with a stronger effect on the latter; (b) the interaction between EA and ESE has a negative effect on intrapreneurship but a positive effect on self-employment; (c) family-to-work conflict weakens the aforementioned interactive effect on both intrapreneurship and self-employment, whereas work-to-family conflict strengthens its effect on self-employment; (d) household income strengthens the interactive effect on both intrapreneurship and self-employment. Overall, these findings contribute to a nuanced understanding of the relationship among individual cognitive traits, family contingencies and entrepreneurial choice. The theoretical and practical implications are discussed.

Abstract Background A consensus has not been reached on the value of prostate-specific antigen density (PSAD) as a predictor of biochemical recurrence of prostate cancer. This meta-analysis aimed to evaluate the association between PSAD and biochemical recurrence of prostate cancer after primary treatment. Methods Two authors systematically searched PubMed, Web of Science, and Embase databases (up to August September 10, 2023) to identify studies that assessed the value of pretreatment PSAD in predicting biochemical recurrence after primary treatment (radical prostatectomy or radiotherapy) of prostate cancer. A random effect model was used to pool adjusted hazard ratios (HR) with 95% confidence intervals (CI) for biochemical recurrence. Results Nine studies with 4963 patients were eligible for the meta-analysis. The reported prevalence of biochemical recurrence ranged from 4 to 55.1%. For patients with higher PSAD compared to those with low PSAD, the pooled HR of biochemical recurrence was 1.59 (95% CI 1.21–2.10). Subgroup analysis showed that the pooled HR of biochemical recurrence was 1.80 (95% CI 1.34–2.42) for patients who received radical prostatectomy, and 0.98 (95% CI 0.66–1.45) for patients who received radiotherapy. Conclusions Elevated pretreatment PSAD may be an independent predictor for biochemical recurrence of prostate cancer after radical prostatectomy. Determining PSAD could potentially improve the prediction of biochemical recurrence in patients with prostate cancer.

Abstract Background Lung cancer is a highly prevalent malignancy and has the highest mortality rate among all tumors due to lymph node metastasis. Bone marrow and umbilical cord-derived mesenchymal stem cells (MSCs) have demonstrated tumor-suppressive effects on lung cancer. This study investigated the effects of DPSC lysate on proliferation, apoptosis, migration and invasion of cancer cells were studied in vivo and in vitro. Methods The proliferation, apoptosis, and migration/metastasis were evaluated by cell counting kit-8 assay, Annexin-V and propidium iodide staining, and the transwell assay, respectively. The expression levels of apoptosis-, cell cycle-, migration-, and adhesion-related mRNA and proteins were measured by qRT-PCR and western blot. The level and mRNA expression of tumor markers carcino embryonic antigen (CEA), neuron-specific enolase (NSE), and squamous cell carcinoma (SCC) were measured by Enzyme-linked immunosorbent assay (ELISA) and qRT-PCR. Finally, a tumor-bearing mouse model was constructed to observe the tumor-suppressive effect of DPSC lysate after intraperitoneal injection. Results DPSC lysate decreased the viability of A549 cells and induced apoptosis in lung cancer cells. Western blot confirmed that levels of Caspase-3, Bax, and Bad were increased, and Bcl-2 protein levels were decreased in A549 cells treated with DPSC lysate. In addition, DPSC lysate inhibited the migration and invasion of A549 cells; downregulated key genes of the cell cycle, migration, and adhesion; and significantly suppressed tumor markers. Xenograft results showed that DPSC lysate inhibited tumor growth and reduced tumor weight. Conclusions DPSC lysate inhibited proliferation, invasion, and metastasis; promoted apoptosis in lung cancer cells; and suppressed tumor growth- potentially providing a cell-based alternative therapy for lung cancer treatment. Abstract Stimuli‐responsive polymers have found applications as shape‐memory materials, optical switches, and sensors, but the installation of these responsive properties in non‐polar and inert polyolefins is challenging. In this contribution, a series of spiropyran (SP)‐based comonomers are synthesized and copolymerized with ethylene or ethylene/cyclic monomers. In addition to great mechanical and surface properties, these functionalized polyolefins responded to light, heat, and force, which induced changes in the polymer structure to transmit color or mechanical signals. These interesting responsive properties are also installed in a series of commercial polyolefin materials through reactive extrusion, making the scalable production of these materials possible.

Objective: This meta-analysis aimed to assess the influence of comorbidity, as assessed by the Charlson comorbidity index (CCI), on survival outcomes in patients with prostate cancer (PCa). Methods: We conducted a comprehensive search of the PubMed, Web of Science, and Embase databases to identify studies that examined the association between CCI-defined comorbidity and survival outcomes in PCa patients. We employed a random effect model to merge adjusted hazard ratios (HR) with 95 % confidence intervals (CI) for survival outcomes. Results: Sixteen studies reporting on 17 articles, which collectively included 457,256 patients. For the presence (CCI score ≥1) versus absence (CCI score of 0) of comorbidity, the pooled HR was 1.59 (95 % CI 1.43–1.77) for all-cause mortality, 0.98 (95 % CI 0.90–1.08) for PCa-specific mortality, and 1.88 (95 % CI 1.61–2.21) for other-cause mortality. When compared to a CCI score of 0, the pooled HR of all-cause mortality was 1.30 (95 % CI 1.18–1.44) for a CCI score of 1, 1.65 (95 % CI 1.37–2.00) for a CCI score ≥2, and 1.75 (95 % CI 1.57–1.95) for a CCI score ≥3. Additionally, the pooled HR of other cause mortality was 1.53 (95 % CI 1.41–1.67) for a CCI score of 1, 1.93 (95 % CI 1.74–2.75) for a CCI score ≥2, and 3.95 (95 % CI 2.13–7.34) for a CCI score ≥3. Conclusions: Increased comorbidity, as assessed by the CCI, significantly predicts all-cause and other-cause mortality in patients with PCa, but not PCa-specific mortality. The risk of all-cause and other-cause mortality increases with the burden of comorbidity.

Abstract Background For early screening and diagnosis of non-small cell lung cancer (NSCLC), a robust model based on plasma proteomics and metabolomics is required for accurate and accessible non-invasive detection. Here we aim to combine TMT-LC-MS/MS and machine-learning algorithms to establish models with high specificity and sensitivity, and summarize a generalized model building scheme. Methods TMT-LC-MS/MS was used to discover the differentially expressed proteins (DEPs) in the plasma of NSCLC patients. Plasma proteomics-guided metabolites were selected for clinical evaluation in 110 NSCLC patients who were going to receive therapies, 108 benign pulmonary diseases (BPD) patients, and 100 healthy controls (HC). The data were randomly split into training set and test set in a ratio of 80:20. Three supervised learning algorithms were applied to the training set for models fitting. The best performance models were evaluated with the test data set. Results Differential plasma proteomics and metabolic pathways analyses revealed that the majority of DEPs in NSCLC were enriched in the pathways of complement and coagulation cascades, cholesterol and bile acids metabolism. Moreover, 10 DEPs, 14 amino acids, 15 bile acids, as well as 6 classic tumor biomarkers in blood were quantified using clinically validated assays. Finally, we obtained a high-performance screening model using logistic regression algorithm with AUC of 0.96, sensitivity of 92%, and specificity of 89%, and a diagnostic model with AUC of 0.871, sensitivity of 86%, and specificity of 78%. In the test set, the screening model achieved accuracy of 90%, sensitivity of 91%, and specificity of 90%, and the diagnostic model achieved accuracy of 82%, sensitivity of 77%, and specificity of 86%. Conclusions Integrated analysis of DEPs, amino acid, and bile acid features based on plasma proteomics-guided metabolite profiling, together with classical tumor biomarkers, provided a much more accurate detection model for screening and differential diagnosis of NSCLC. In addition, this new mathematical modeling based on plasma proteomics-guided metabolite profiling will be used for evaluation of therapeutic efficacy and long-term recurrence prediction of NSCLC.

The Lower Silurian Longmaxi Formation is the favorable target area for deep shale gas exploration and development in southeastern Sichuan Basin. Based on whole-rock X-ray diffraction analysis, scanning electron microscope, reservoir evolution thermal simulation experiment and nitrogen adsorption experiment, the diagenetic characteristics of deep shale reservoir in Longmaxi Formation were analyzed, and the reservoir pore evolution law was clarified. The results show that: ①The diagenetic minerals of the deep shale in the Longmaxi Formation are mainly quartz and clay minerals, with a small amount of carbonate minerals and feldspar. The primary inorganic pores are mainly controlled by mechanical compaction and cementation (quartz, carbonate, clay, pyrite). The organic pores are mainly controlled by the thermal maturity of organic matter, dissolution and later compaction. ②In the process of thermal simulation experiment, the organic pores of shale show a process of change from scratch, from small to large and then from large to small. Later, the organic matter is affected by compaction and graphitization, and the volume of micropores and mesopores begins to decrease. ③The shale pores of Longmaxi Formation have undergone several evolutionary stages. In the early stage of diagenesis, compaction caused a large number of inorganic pores to disappear. In the middle stage of diagenesis, kerogen hydrocarbon generation occupied pores, dissolution and cementation transformed pores. In the late diagenetic period, liquid hydrocarbon cracking gas and pressurization promote the development of organic pores.

Background: Banxia Xiexin decoction (BXD) is a classic traditional Chinese medicine (TCM) formula clinically used to treat chronic gastritis, gastric ulcers, gastric cancer, and many other gastrointestinal diseases. Long noncoding RNAs (lncRNAs) have been shown to play an important role in maintaining the malignant phenotype of tumors. However, no relevant studies have shown whether Banxia Xiexin decoction regulates and controls lncRNA TUC338, and the effect of TUC338 on the regulation of gastric cancer invasion and metastasis remains unclear. Purpose: To investigate the ability of the traditional Chinese medicine (TCM) Banxia Xiexin decoction (BXD) to inhibit the migration and invasion of human gastric cancer AGS cells by regulating the long noncoding RNA (lncRNA) TUC338. Methods: UHPLC‒MS/MS was used to analyze the chemical components of BXD. MTT was performed to determine the effects of BXD on the proliferation of AGS cells. qRT‒PCR was used to determine the expression of lncRNA TUC338 in gastric cancer tissues, paracarcinoma tissues, AGS human gastric cancer cells and GES-1 normal gastric mucosa cells and to evaluate the effects of BXD on the expression of lncRNA TUC338 in AGS cells. Lentiviral transfection was used to establish human gastric cancer AGS cells with knocked down lncRNA TUC338 expression. The effects of lncRNA TUC338 knockdown on the migration and invasion of AGS cells were observed by a scratch assay and Transwell migration assay, respectively. Western blotting was performed to analyze the effects of lncRNA TUC338 knockdown on epithelial-to-mesenchymal transition (EMT) in AGS cells. We performed quality control on three batches of BXD. We used UHPLC‒MS/MS to control the quality of three random batches of BXD used throughout the study. Results: Ninety-five chemical components were identified from the water extract of BXD, some of which have anticancer effects. The expression of TUC.338 in gastric cancer tissues was higher than that in para-carcinoma tissues. BXD inhibited the invasion and migration of gastric cancer cells by inhibiting the expression of lncRNA TUC338, which reduced EMT. After knockdown of lncRNA TUC338, the migration and invasion of AGS cells were reduced; the expression of the EMT-related protein E-cadherin was increased, and the expression of N-cadherin and vimentin was reduced. Conclusions: The present results suggest that BXD has potential as an effective treatment for gastric cancer through the inhibition of lncRNA TUC338 expression.

Impact craters are extensively researched geological features that contribute to various aspects of lunar science, such as evaluating the model age, regolith thickness, etc. The method for identifying impact craters has gradually transitioned from manual counting to automated identification. Automatic crater detection based on the digital elevation model (DEM) is commonly used to detect larger craters. However, using only DEM has limitations in discerning smaller craters (diameter < ~1 km). This study utilizes an improved Faster R-CNN algorithm and the Kaguya Terrain Camera (TC) morning map to detect small impact craters in the Chang’e-5 (CE-5) landing site. It uses model fusion to improve the precision of small crater identification. The results show a recall rate of 96.33% and a precision value of 90.19% for craters with diameters exceeding 200 m. The model found a total of 187,101 impact craters in the CE-5 region. The spatial distribution density of impact craters with diameters ranging from 100 m to 200 m is approximately 2.5706/km2. For craters with diameters ranging from 200 m to 1 km, the average spatial distribution density is about 0.9016/km2. By the unbiased impact crater density of chronological analysis, the model age of the Im2 and Em4 geological units in the CE-5 region is 3.78 Ga and 2.07 Ga, respectively.

Accurately grasping the distribution and area of cotton for agricultural irrigation scheduling, intensive and efficient management of water resources, and yield estimation in arid and semiarid regions is of great significance. In this paper, taking the Xinjiang Shihezi oasis agriculture region as the study area, extracting the spectroscopic characterization (R, G, B, panchromatic), texture feature (entropy, mean, variance, contrast, homogeneity, angular second moment, correlation, and dissimilarity) and characteristics of vegetation index (normalized difference vegetation index/NDVI, ratio vegetation index/DVI, difference vegetation index/RVI) in the cotton flowering period before and after based on GF-6 image data, four models such as the random forests (RF) and deep learning approach (U-Net, DeepLabV3+ network, Deeplabv3+ model based on attention mechanism) were used to identify cotton and to compare their accuracies. The results show that the deep learning model is better than that of the random forest model. In all the deep learning models with three kinds of feature sets, the recognition accuracy and credibility of the DeepLabV3+ model based on the attention mechanism are the highest, the overall recognition accuracy of cotton is 98.23%, and the kappa coefficient is 96.11. Using the same Deeplabv3+ model based on an attention mechanism with different input feature sets (all features and only spectroscopic characterization), the identification accuracy of the former is much higher than that of the latter. GF-6 satellite image data in the field of crop type recognition has great application potential and prospects.

Characteristic construction is an important part of material data analysis. In the big data environment, the development of material data science will have implications for multidimensional data analysis methods. Among these, the machine learning method for multidimensional data models can be widely applied to material data types, including element ratios, atomic compositions, electronic arrangements, molecular structures, and energy distributions. For high-throughput computing materials, it is recommended in material data science to judge and extract the main characteristics influencing material properties and predict novel functional materials using the discovered laws. Consequently, we considere the characteristic construction, learning prediction, feature extraction, and high-order analysis of computational materials as the main research purposes, and construct a composite analysis model of the material system by combining data preprocessing, data mining, data evaluation, and knowledge representation as the main steps of data analysis. This demonstrates that a method to comprehensively judge the properties of materials by constructing the characteristics of materials in different dimensions is essential.

Abstract Background To discussed the risk factor and the management of vitrectomy for long-term endophthalmitis developing after intraocular lens (IOL) implantation in children. Methods We retrospectively investigated the clinical characteristics and surgical outcomes of long-term endophthalmitis developing after IOL implantation in children. Results Four eyes of four children were included in the study. The mean time to endophthalmitis development after IOL implantation was 3.0 ± 0.8 years. The corneal or scleral sutures may have been caused the infection. All 4 patients underwent vitrectomy and received intravitreal antibiotics with or without IOL removal. At the last follow-up, the outcomes were satisfactory; the fundus was clear, the retina remained attached, the visual acuity improved, and there were no severe complications. Conclusions The use of scleral sutures and the exposure of conceal sutures may induce the onset of long-term endophthalmitis after IOL implantation. Complete vitrectomy and appropriate use of antibiotics are effective in the treatment of long-term endophthalmitis developing after IOL implantation in children.

Here the authors show an ionic anchoring support strategy that is highly versatile, generally applicable to different systems, and enables strong catalyst-support interactions while tolerating various polar functional groups.

Remote sensing technology has become a popular tool for crop classification, but it faces challenges in accurately identifying crops in areas with fragmented land plots and complex planting structures. To address this issue, we propose an improved method for crop identification in high-resolution remote sensing images, achieved by modifying the DeepLab V3+ semantic segmentation network. In this paper, the typical crop area in the Jianghuai watershed is taken as the experimental area, and Gaofen-2 satellite images with high spatial resolutions are used as the data source. Based on the original DeepLab V3+ model, CI and OSAVI vegetation indices are added to the input layers, and MobileNet V2 is used as the backbone network. Meanwhile, the upper sampling layer of the network is added, and the attention mechanism is added to the ASPP and the upper sampling layers. The accuracy verification of the identification results shows that the MIoU and PA of this model in the test set reach 85.63% and 95.30%, the IoU and F1_Score of wheat are 93.76% and 96.78%, and the IoU and F1_Score of rape are 74.24% and 85.51%, respectively. The identification accuracy of this model is significantly better than that of the original DeepLab V3+ model and other related models. The proposed method in this paper can accurately extract the distribution information of wheat and rape from high-resolution remote sensing images. This provides a new technical approach for the application of high-resolution remote sensing images in identifying wheat and rape.

Abstract Background This study investigated the role of TP73 gene polymorphism, rs1801173on risk of gastric cancer. Methods We conducted a case-controlled study including 577 primary gastric cancer and 678 normal control cases. The target gene fragment was amplified using PCR using blood samples collected from patients. Allele analysis and genotyping were performed using snapshot method. Results The findings showed that the control group had consistent genotype frequency distribution and presented Hardy–Weinberg equilibrium. The results showed no significant differences in sex, drinking history and age distributions between subjects with the polymorphism and subjects in the control group. Smoking status was correlated with incidence of gastric cancer (P = 0.006). The rs1801173 locus of TP73 gene contained 3 genotypes including: TT, CT, and CT. Logistic regression analysis showed that distribution of recessive model and dominant model was comparable between the two groups before (P = 0.688; 0.937) or after (P = 0.703; 0.990) adjusting for confounders. The distribution frequency in case group was not significantly different relative to that of the control group (P = 0.763). Conclusion Smoking can independently influence the risk of gastric cancer. TP73 gene rs1801173 polymorphism was not significantly correlated with risk of gastric cancer.

Aims. This study explores the effects of curcumin as a therapeutic agent against oral squamous cell carcinoma (OSCC). Methods. We acquired the targets of curcumin from three digital databases, including the Comparative Toxicogenomics Database, Search Tool for Interactions of Chemicals, and SwissTargetPrediction. Then, we identified the differentially expressed genes (DEGs) and the weighted gene coexpression network analysis-based key modules using the expression profiles of GSE23558 to acquire the OSCC-related genes. Additionally, the GeneCards and Online Mendelian Inheritance in Man databases were also used to identify the OSCC-related genes. Finally, curcumin-OSCC interaction genes were obtained by overlapping curcumin targets and OSCC-related genes. The enrichment analysis was performed by the ClusterProfiler algorithm and Metascape, respectively. Then, a protein-protein interaction network was created, and the maximal clique centrality algorithm was used to identify the top 10 hub genes. Besides, we examined the expression levels of hub genes in OSCC using The Cancer Genome Atlas database. Results. 927 DEGs were identified, including 308 upregulated ones and 619 downregulated ones. The cluster one-step network construction function of the WGCNA algorithm recognized a soft-thresholding power of 6, and 9083 genes were acquired. 2591 OSCC-related genes were obtained by overlapping the GSE23558-identified genes and the OSCC-related genes from disease target bases. Finally, we identified 70 candidate drug-disease interaction genes by overlapping the disease-related genes with the curcumin target. The enrichment analysis suggested that response to oxidative stress, epithelial cell proliferation, and AGE/RAGE pathway might involve in the effect of curcumin on OSCC. The topologic study identified the ten hub genes, including VEGFA, AKT1, TNF, HIF1A, EGFR, JUN, STAT3, MMP9, EGF, and MAPK3. A significant difference was observed in VEGFA, AKT1, TNF, HIF1A, EGFR, MMP9, EGF, and MAPK3 expression levels between head and neck squamous cell carcinoma and the normal controls. However, no significant difference was observed in JUN (P=0.14) and STAT3 (P=0.054). Conclusion. This study provided an overview and basis for the potential mechanism of curcumin against OSCC. The following experiments should be performed to further understand the effectiveness and safety of curcumin in treating OSCC.

A green‐emitting perovskite first‐order distributed feedback (DFB) laser based on the methylammonium lead bromide (MAPbBr3) with high stability is demonstrated for the first time. The laser achieves stable lasing at 550 nm with a full width at half maximum of 0.4 nm. Low lasing threshold of 60 μJ cm−2 under nanosecond pulsed excitation and 3.1 μJ cm−2 under femtosecond pulsed excitation is observed, showing a much lower lasing threshold compared with the second‐order DFB cavities, which are fabricated on the same substrate. By optimizing the antisolvent treatment and encapsulating with poly(methyl methacrylate), the laser lifetime, resistance to moisture, lasing threshold, and intensity are significantly improved. The lasers are fabricated with a complementary metal‐oxide‐semiconductor‐compatible process, thus offer promising potential for the integrated photonic devices.

Porosity is a key parameter in shale gas reservoir evaluation and reserve calculation and its accurate test is the basis for calculating geological reserves of shale gas and determining development plans. In order to clarify the differences between different porosity test methods and their influences on the calculation results of shale gas reserves, we collected 65 shale samples of Lower Silurian Longmaxi Formation from six shale gas wells in the Zhaotong National Shale Gas Demonstration Area of the southern Sichuan Basin for comparative experiments using three porosity test method, including gas injection porosimetry (GIP) method, water immersion porosimetry (WIP) method and nuclear magnetic resonance (NMR) method. Then, these three methods were comparatively analyzed based on the test results. Finally, it was proposed to optimize the key parameters of these three shale porosity test methods. And the following research results were obtained. First, in terms of the GIP method, the particle size of shale sample shall be in the range of 20–60 mesh and the helium saturation equilibrium time shall be over 1800 s. Second, in terms of the WIP method, the sample shall be dried for at least 48 h under 110 °C and saturated for 24 h under the confining pressure of 15 MPa. Third, in terms of the NMR method, NMR porosity calculation shall not be conducted until the NMR signal of the dried sample is deducted, on the basis of echo time and waiting time optimization. Fourth, porosity average and median value obtained by these three shale porosity test methods follow the relationship of WIP porosity > NMR porosity > particle GIP porosity > plunger GIP porosity. Fifth, different shale porosity test methods have greater influences on the calculation results of shale gas geological reserves, whose difference can reach 20%. In conclusion, during the application of NMR method and WIP method, fluid is introduced for saturation, which may damage the shale pores. However, the particle GIP porosity can reflect the entire space of shale more comprehensively and is not influenced by the properties of the applied fluid. Therefore, it is suggested to adopt the particle GIP method to calculate shale gas geological reserves.

BackgroundConflicting results have been reported on the value of the Geriatric Nutritional Risk Index (GNRI) in predicting adverse outcomes in patients with peripheral artery disease (PAD). The objective of this meta-analysis was to evaluate the association of GNRI with adverse outcomes in patients with lower extremity PAD.MethodsRelevant studies were comprehensively searched in PubMed and Embase databases until December 31, 2021. Eligible studies should evaluate the value of GNRI in predicting major adverse cardiovascular and leg events (MACLEs), all-cause mortality, and amputation in patients with lower extremity PAD.ResultsEight studies reporting on 9 articles involving 5,541 patients were included. A fixed-effect model meta-analysis showed that patients with PAD with low GNRI had an increased risk of MACLEs [adjusted risk ratio (RR) 2.26; 95% confidence interval (CI) 1.54–3.31] and all-cause mortality (RR 2.38; 95% CI 1.71–3.31) compared with those with high GNRI. When analysis of GNRI is by continuous data, 10 units of GNRI decrease was associated with 36% and 44% higher risk of MACLEs and all-cause mortality, respectively. However, per 10 units GNRI score decrease was not significantly associated with a higher risk of amputation (p = 0.051).ConclusionLow GNRI may be an independent predictor of adverse outcomes in patients with lower extremity PAD. Routine screening of nutritional status using the GNRI may provide important prognostic information in patients with PAD.

Natural gas is a clean and efficient energy source. Shale gas, one of the unconventional natural gases, is becoming an indispensable part of natural gas. Compared with marine facies shale gas, which has large-scale exploitation, marine and continental transitional facies shale gas have greater development potential. It could be the new direction of development in the future. In this article, the basic geological characteristics, sedimentary environment, and reservoir characteristics of organic-rich transitional facies shale in the southern Sichuan basin are analyzed by lithologic characteristics, chemical element analysis, reservoir space, gas bearing characteristics, and so on and are compared with domestic and abroad transitional shale gas. The results show that the sedimentary period of transitional shale of Upper Permian Leping Formation has a high paleoproductivity level and deposition rate favorable for the accumulation of organic matter in southern Sichuan basin. It was warm and humid paleoclimatic by Sr, V, Cr, Ni, Co, and other trace elements analysis. According to the experimental test analysis of reservoir characteristics, the organic matter is mainly type Ⅲ kerogen, and the thermal evolution degree is mainly overmature in the reservoir. The reservoir space is mainly of microfractures and micropores. The main types of micropore development are intergranular pore, intragranular pore, and organic pore. Compared with other shale gas regions that form industrial airflow, the study region with large thickness, highly brittle mineral content, high level of organic carbon content, and excellent gas-bearing characteristics, it has favorable conditions for shale gas accumulation. The southern Sichuan basin could be the key area for breakthroughs in the exploration and development of transitional shale gas in the next step.

Proliferative diabetic retinopathy (PDR) accounts for severe impact on vision, its mechanism is still poorly understood. To compare the differences of vitreous protein profiles in PDR patients before and after a complete anti-vascular endothelial growth factor (VEGF) loading dose with ranibizumab treatment. Twelve vitreous humor (VH) samples were collected from six PDR patients before (set as pre group) and after (set as post group) intravitreal injection of ranibizumab (IVR) treatment. LC–MS/MS and bioinformatics analysis were performed to identify differentially expressed proteins. Proteins were validated with targeted proteomics using parallel reaction monitoring (PRM) in a validation set consisting of samples from the above patients. Objective: To explore the repair effect of the prepared drug-loaded AM1241 poly(ethylene glycol)–dithiothreitol (PEG-DTT) hydrogel on cranial bone defects in SD rats.Methods: The PEG-DTT hydrogel under borax catalysis was quickly prepared, and the characterization of the material was observed by a scanning electron microscope. The effect of AM1241 on cell activity and bone tissue differentiation was tested. The SD rat model of cranial bone defect was established, and the defect was repaired by injecting the prepared hydrogel into the defect. The defect was divided into four groups, namely, sham group, blank group, PEG-DTT group, and PEG-DTT + AM1241 group. The rats were euthanized, and whole cranial bone was taken out for micro-CT and histological observation.Results: The prepared hydrogel is porous; it is liquid when heated to 80°C and a hydrogel when cooled to 25°C. 5–10 μM AM1241 increased osteoblast activity. A moderate amount of AM1241 can promote osteogenic differentiation. Both the PEG-DTT group and PEG-DTT + AM1241 group showed obvious new bone tissue formation, but the PEG-DTT + AM1241 group had a better effect. In addition, the new bone tissue in the PEG-DTT + AM1241 group was significantly more than that in the other groups.Conclusion: The prepared AM1241-loaded PEG-DTT hydrogel showed a good repair effect on SD rats with cranial bone defects. It can be used as materials for cranial bone repair in SD rats with cranial bone defects, but the repair effect is weaker than that of normal bone. These results provide a theoretical and practical basis for its further clinical application.

Abstract Background Angiogenesis is an important parameter in the development of diabetic retinopathy (DR), and it is indicative of an early stage evolving into a late phase. Therefore, examining the role of angiogenic factors in early DR is crucial to understanding the mechanism of neovascularization. Methods The present study identified hub genes and pathways associated with angiogenesis in early DR using bioinformatics analysis. Genes from published literature and Gene Expression Omnibus (GEO) were collected and analysed. Results We collected 73 genes from 70 published studies in PubMed, which were referred to as DR-related gene set 1 (DRgset1). The gene expression profile of GSE12610 was downloaded, and 578 differentially expressed genes (DEGs) between diabetic and normal samples were identified. DEGs and DRgset1 were further combined to create DR-related gene set 2 (DRgset2). After an enrichment analysis, we identified 12 GO terms and 2 pathways associated with neovascularization in DRgset1, and 8 GO terms and 2 pathways in DRgset2. We found 39 new genes associated with angiogenesis and verified 8 candidate angiogenesis-related genes in DR cells using real-time PCR: PIK3CB, ALDH3A1, ITGA7, FGF23, THBS1, COL1A1, MAPK13, and AIF1. We identified 10 hub genes associated with neovascularization by constructing a protein-protein interaction (PPI) network: TNF, VEGFA, PIK3CB, TGFB1, EDN1, MMP9, TLR4, PDGFB, MMP2, and THBS1. Conclusions The present study analysed angiogenesis-related genes and pathways in early DR in a comprehensive and systematic manner. PIK3CB, ALDH3A1, ITGA7, FGF23, THBS1, COL1A1, MAPK13, and AIF1 may be the candidate genes to further explore the mechanisms of angiogenesis in early DR. TNF, PIK3CB, TGFB1, EDN1, MMP9, TLR4, PDGFB, MMP2, and THBS1 may be new targets for early neovascularization therapy in the future.

Based on exploration and development results and evaluation of marine shale gas in South China in the past ten years, in view of the features of “high maturity, strong tectonic reformation and high shear stress” of the shale in Zhaotong exploration zone in the Yunnan and Guizhou Plateau, as well as the key issues of long time diffusion and leakage of shallow shale gas, and the preservation conditions, the factors controlling shallow shale gas sweet spot and key zone selection evaluation technology of shale gas are investigated. From 2017 to 2018, the first significant exploration breakthrough was made in the Taiyang anticline at a buried depth of 700 to 2 000 m, discovering large-scale proved geological reserves of shallow shale gas. By examining the accumulation conditions and sweet spot control factors of the shallow shale gas in this area, it is found that the accumulation and productivity potential of shale gas in the mountainous area with complex structure outside basin are controlled by five factors: (1) The gas-rich area has weak tectonic reformation and good preservation conditions on the whole, taking on typical anticline trap occurrence mode. (2) The gas-rich area is in over-pressure state and high in shale gas content. (3) The gas-rich area has high quality shale and thus superior source rock condition. (4) The gas-rich area has high quality reservoirs dominated by class I. (5) The shale gas reservoir in the gas-rich area has high content of brittle minerals and small difference between maximum and minimum horizontal stresses which are conducive to hydraulic fracturing. The innovative practice and core technologies formed during the exploration and production capacity construction of shallow shale gas in the Zhaotong demonstration zone have great reference significance for shallow shale gas exploration and development in other areas. Background: From previous studies, we found that there are more than 100 types of RNA modifications in RNA molecules. m6A methylation is the most common. The incidence rate of adenocarcinoma of the esophagogastric junction (AEG) at home and abroad has increased faster than that of stomach cancer at other sites in recent years. Here, we systematically analyze the modification pattern of m6A mRNA in adenocarcinoma at the esophagogastric junction.Methods: m6A sequencing, RNA sequencing, and bioinformatics analysis were used to describe the m6A modification pattern in adenocarcinoma and normal tissues at the esophagogastric junction.Results: In AEG samples, a total of 4,775 new m6A peaks appeared, and 3,054 peaks disappeared. The unique m6A-related genes in AEG are related to cancer-related pathways. There are hypermethylated or hypomethylated m6A peaks in AEG in differentially expressed mRNA transcripts.Conclusion: This study preliminarily constructed the first m6A full transcriptome map of human AEG. This has a guiding role in revealing the mechanism of m6A-mediated gene expression regulation.

Abstract It is important to investigate the mechanical performances of (Gd0.9Yb0.1)2Zr2O7 (GYbZ) materials deposited on irregular substrates for improving new thermal barrier coatings. Three-point bending fracture characteristics of freestanding GYbZ coating prepared by supersonic plasma sprayed (SPS) technique were investigated with the help of digital image correlation technique. The cracking time, crack propagation path, and mechanical properties of GYbZ coating were obtained. Meanwhile, the X-ray computed tomography technique was introduced to scan the microstructure of freestanding GYbZ coatings, which are used to establish three-dimensional (3D) finite element model by using the Avizo software. The brittle cracking criterion was applied to describe the bending fracture process of GYbZ coatings. The critical cracking strain was estimated as 0.36%±0.03% by repeatedly comparing the difference between the experimental and simulated curves. The results would be extended to predict the dangerous region and failure mechanisms of GYbZ coatings deposited on irregular substrate during finite element simulations.

Abstract Analogous to DNA methylation and histone modification, RNA modification, as another epigenetic layer, plays an important role in many diseases, especially in tumours. As the most common form of RNA modification, m6A methylation has attracted increasing research interest in recent years. m6A is catalysed by RNA methyltransferases METTL3, METTL14 and WTAP (writers), m6A is removed by the demethylases FTO and ALKBH5 (erasers) and interacts with m6A-binding proteins, such as YT521-B homology (YTH) domain-containing proteins. This article reviews recent studies on methylation modification of m6A in gastrointestinal tract cancers.

Vegetation dynamics in arid and semi-arid regions have an important impact on carbon cycle, water cycle, and energy exchange at local, regional, and global scales. Therefore, it is of great significance for scientists to grasp the changes of vegetation cover in arid and semi-arid regions timely and accurately. Based on this, the applicability of ESTARFM model in the complex terrain area of arid and semi-arid Xinjiang was explored using Landsat and MODIS data fusion, and the overall change characteristics of vegetation cover (FVC) and the distribution and change patterns of different terrains in the study area in the past 15 years were analyzed by combining the dimidiate pixel model, unary linear regression and digital elevation model. The results show that: (1) the NDVI data fused by ESTARFM Model has high consistency with the real NDVI data, and it can be used for subsequent FVC estimation. (2) From 2006 to 2020, the inter FVC was at a high level as a whole, and the average annual FVC showed a weak increasing trend in fluctuation; there are obvious differences in spatial distribution, which is characterized by high distribution in the north and low in the south. (3) The improved area of vegetation cover in the study area is greater than the degraded area, accounting for 52.3% and 47.7% respectively; (4) In the elevation range of 2000 to 3500 m, the FVC showed a slight degradation trend on 25° to 45° slopes and south and southeast slopes, and the rest showed a slight improvement trend. ESTARFM-based model enables monitoring of vegetation cover changes in complex terrain areas of the arid and semi-arid regions in Xinjiang over a long time series. The overall FVC level in the study area is high, and there both are serious degradation and improvement phenomena.

ObjectivesMacrophage-derived exosomes (Mφ-Exos) are involved in tumor onset, progression, and metastasis, but their regulation in oral squamous cell carcinoma (OSCC) is not fully understood. RBPJ is implicated in macrophage activation and plasticity. In this study, we assessed the role of Mφ-Exos with RBPJ overexpression (RBPJ-OE Mφ-Exos) in OSCC.Materials and MethodsThe long non-coding RNA (lncRNA) profiles in RBPJ-OE Mφ-Exos and THP-1-like macrophages (WT Mφ)-Exos were evaluated using lncRNA microarray. Then the functions of Mφ-Exo-lncRNA in OSCC cells were assessed via CCK-8, EdU, and Transwell invasion assays. Besides, luciferase reporter assay, RNA immunoprecipitation, and Pearson’s correlation analysis were adopted to confirm interactions. Ultimately, a nude mouse model of xenografts was used to further analyze the function of Mφ-Exo-lncRNAs in vivo.ResultsIt was uncovered that lncRNA LBX1-AS1 was upregulated in RBPJ-OE Mφ-Exos relative to that in WT Mφ-Exos. RBPJ-OE Mφ-Exos and LBX1-AS1 overexpression inhibited OSCC cells to proliferate and invade. Meanwhile, LBX1-AS1 knockdown boosted the tumor to grow in vivo. The effects of RBPJ-OE Mφ-Exos on OSCC cells can be reversed by the LBX1-AS1 knockdown. Additionally, mechanistic investigations revealed that LBX1-AS1 acted as a competing endogenous RNA of miR-182-5p to regulate the expression of FOXO3.ConclusionExo-LBX1-AS1 secreted from RBPJ-OE Mφ inhibits tumor progression through the LBX1-AS1/miR-182-5p/FOXO3 pathway, and LBX1-AS1 is probably a diagnostic biomarker and potential target for OSCC therapy.

Increasing evidence indicated that the tumor microenvironment (TME) played a crucial role in cancer initiation and progression. Ubiquitin-conjugating enzyme E2C (UBE2C) was differentially expressed in many cancer types. However, the immunological and prognostic roles of UBE2C were unclear. Differentially expressed genes (DEGs) of 29 cancer types were downloaded from GEPIA2 and 4 cancer types failed to download owing to no DEGs. Furthermore, the gene expression profiles, mutation data, and survival data of 33 cancer types were obtained from UCSC Xena. Clinical stage relevance, tumor mutational burden (TMB), TME relevance analysis, and gene set enrichment analysis (GSEA) of DEGs in 33 cancer types were performed. And DEGs were identified in oral squamous cell carcinoma (OSCC) by biological experiments. Previous studies indicated that UBE2C was related to the prognosis of many cancers. In our study, the higher UBE2C expression level meant a terminal clinical stage in 8 cancer types and the expression level of UBE2C was related to TMB in 20 cancer types. In addition, both immune relevance analysis and GSEA showed that UBE2C might participate in immune response in many cancers. Furthermore, the UBE2C mRNA level and protein level were all identified as upregulated in OSCC cell lines and tissues. UBE2C was differentially expressed in many cancer types and related to the pathogenesis and TME of many cancers, which might be a potential diagnostic and therapeutic biomarker.

LINC01355 has been demonstrated to be dysregulated in several cancers. However, the exact molecular function of LINC01355 in the pathogenesis of OSCC remains unstudied. Here, we reported the effect of LINC01355 in OSCC and investigated the mechanisms. Firstly, we found that the results indicated LINC01355 was increased in OSCC cells. Knockdown of LINC01355 repressed OSCC cell proliferation, migration, and invasion. Recently, immunotherapy is a significant method for the treatment of cancers, in which CD8+ T cells exhibit a significant role. The influence of LINC01355 on the antitumor activity of CD8+ T cells was also focused in this study. As shown, the silence of LINC01355 could repress OSCC tumor growth via inducing CD8+ T cell immune responses. In addition, we found that downregulation of LINC01355 significantly restrained CD8+ T cell apoptosis, induced CD8+ T cell percentage, and enhanced the cytolysis activity when cocultured with OSCC cells. It has been reported that the Notch pathway represses CD8+ T cell activity in cancer patients. In our present study, we displayed that lack of LINC01355 suppressed OSCC malignant behaviors and enhanced the antitumor activity of CD8+ T cells via inactivating Notch signaling. We showed that decreased LINC01355 significantly restrained the Notch signal via a decrease of Notch-1, JAG-1, and HES-1. Repression of Notch1 reversed the effect of LINC01355 in OSCC cells. In conclusion, it was implied that LINC01355 might induce the development of OSCC via modulating the Notch signal pathway, which could provide a candidate therapeutic target for OSCC.

Background. Oral squamous cell carcinoma (OSCC) is the most common type of oral cancer, which remains a major cause of morbidity and mortality in patients with head and neck cancers. However, the critical immune-related signatures and their prognostic values have rarely been investigated. Materials and Methods. Gene differential analysis was used to measure the differences of gene expression between the groups. Correlation analysis was used to assess the association between the gene expression levels and immune-related risk score/DNA methylation levels. The gene set enrichment analysis (GSEA) was used to identify the pathways or cell types enriched by those identified differentially expressed genes (DEGs). Results. In this study, we identified four immune-related gene signatures, including CTSG, TNFRSF4, LCORL, and PLAU, that were significantly associated with the overall survival in OSCC patients from the Cancer Genome Atlas (TCGA) OSCC cohort. Moreover, these four immune-related signatures were differentially expressed between the OSCC and nontumor tissues. The two groups (high and low risk) stratified by the immune-related risk scores had significantly different OS and mortality rates. The gene expression patterns and prognostic values of these immune-related signatures were also verified in two independent validation cohorts. Furthermore, the downregulated genes in the high-risk group (which were also upregulated in the low-risk group) were significantly enriched in the cell type-specific signatures of type 2 T helper cell (Th2), plasmacytoid dendritic cell (pDC), and memory B cell. In contrast, the upregulated genes in the high-score group were enriched in growth factor receptor-related signaling pathways, such as the VEGFA-VEGFR2 signaling pathway, PI3K-Akt signaling pathway, focal adhesion-PI3K-Akt-mTOR signaling pathway, and PDGF pathway, suggesting that those pathways were inversely correlated with immune cell infiltration. Conclusion. In summary, the immune-related signatures had the potential for predicting the risk of OSCC patients. Moreover, the present study also improved our understanding of the association between the growth factor receptor pathways and immune cell infiltration in OSCC.

N6-Methyladenosine (m6A) modification is one of the commonest chemical modifications in eukaryotic mRNAs, which has essential effects on mRNA translation, splicing, and stability. Currently, there is a rising concern on the regulatory role of m6A in tumorigenesis. As a known component in the m6A methyltransferase complex, METTL3 (methyltransferase-like 3) plays an essential role in m6A methylation. Till now, the functions of METTL3 in oral squamous cell carcinoma (OSCC) and its relative mechanism remain to be explored. In this research, through the GEPIA database, we found that high METTL3 expression has a correlation with poor prognosis of squamous cell carcinoma of head and neck. qRT-PCR displayed that METTL3 was highly expressed in OSCC cells. Functionally, METTL3 knockdown reduced the invasion, migration, and proliferation competence of OSCC cells and attenuated the activation of CD8+ T cells. In contrast, METTL3 overexpression resulted in opposite results. GEPIA, UALCAN, and SRAMP databases, PCR, western blot, and m6A RNA methylation assay confirmed the m6A modification of PRMT5 and PD-L1 mediated by METTL3. In conclusion, our results displayed that METTL3 intensified the metastasis and proliferation of OSCC by modulating the m6A amounts of PRMT5 and PD-L1, suggesting that METTL3 may be a therapeutic target for OSCC patients.

lncRNAs are related to the progression of various diseases, including oral squamous cell carcinoma (OSCC), which is a common squamous cell carcinoma of the head and neck. Tumor-associated macrophages and tumor cells are significant components of tumor microenvironment. M2 polarization of tumor-associated macrophages is a crucial actor in tumor malignancy and metastasis. In this study, we studied the molecular mechanism of lncRNA DCST1-AS1 in OSCC. Here, we reported that DCST1-AS1 was significantly increased in OSCC cells. We found that loss of DCST1-AS1 obviously inhibited the proliferation, migration, and invasion of OSCC cells and xenograft tumor growth. Meanwhile, silencing of DCST1-AS1 also repressed the percentage of macrophages expressing M2 markers CD206 and CD11b. DCST1-AS1 shRNA enhanced the percentage of macrophages expressing M1 markers CD80 and CD11c. Then, we observed that loss of DCST1-AS1 suppressed OSCC progression via inactivating NF-κB signaling. As well established, NF-κB signaling exerts critical roles in tumor progression, and our study proved that DCST1-AS1 could regulate NF-κB signaling. We proved that blocking the NF-κB pathway using antagonists greatly downregulated OSCC progression and M2 macrophage polarization induced by the overexpression of DCST1-AS1. To sum up, we reported that DCST1-AS1 plays an important role in modulating OSCC tumorigenicity and M2 macrophage polarization through regulating the NF-κB pathway.

Background/Aims: Emerging evidence points towards an important role of long noncoding RNAs (lncRNAs) in the tumorigenesis and progression of gastric cancer (GC). MT1JP has recently been reported to be differentially expressed and act as a tumor suppressor in different tumors, with its mechanisms not fully understood in GC. Methods: RT-qPCR was used to detect the expression of MT1JP, miR-214-3p and RUNX3 in tumor tissues and cell lines of GC. The CCK-8 assay, colony formation, Transwell assay and wound healing assay were used to evaluate the proliferation, invasion and migration of GC cells, respectively. Bioinformatics analysis and luciferase reporter assay were performed to disclose the interaction between MT1JP, miR-214-3p and RUNX3. Western blot and immunofluorescence were applied to assess the downstream signaling of RUNX3. Results: MT1JP was found downregulated in GC tissues and cells. Low expression of MT1JP was significantly correlated with advanced TNM stage and lymphatic metastasis. The expression of plasma MT1JP was also found decreased in GC patients compared to healthy controls, with an area under the ROC curve (AUC) of 0.649 for diagnosis of GC. Gain- and loss-of-function of MT1JP revealed that MT1JP functioned as a ceRNA for miR-214-3p to facilitate RUNX3 expression and then upregulated p21 and Bim levels suppressing GC cell proliferation, invasion and migration, and promoting apoptosis. Furthermore, MT1JP overexpression suppressed tumor growth and inhibited the expression of miR-214-3p and proliferation antigen Ki-67, but increased the expression of RUNX3, p21 and Bim in vivo. Conclusions: Our results suggest a potential ceRNA regulatory network involving MT1JP regulates RUNX3 expression by competitively binding endogenous miR-214-3p in tumorigenesis and progression of GC. This mechanism may contribute to a better understanding of GC pathogenesis and provide potential therapeutic strategy for GC.

Abstract Background Preoperative treatment of anti-vascular endothelial growth factor (VEGF) agents is extensively used in proliferative diabetic retinopathy (PDR), but the molecular mechanism is not fully understood. The objective of this research is to observe change of protein profile induced by ranibizumab (an anti-VEGF agent) in vitreous humor from PDR patients and reveal the effects of anti-VEGF treatment on PDR. Methods A proteomic method was used to identify differentially expressed proteins in vitreous humor. Untreated PDR patients were defined as PDR group, while those who treated with intravitreal injection of ranibizumab (IVR) were defined as IVR. Gene Ontology (GO) annotation and REACTOME pathways were obtained from DAVID Bioinformatics Resources. Intravitreal level of apolipoprotein C-I (APOC1), serpin peptidase inhibitor clade A member 5 (SERPINA5), tissue inhibitor of metalloproteinases (TIMP2), and keratin 1 (KRT1) were determined by enzyme-linked immuno sorbent assay (ELISA). Results 339 differentially expressed proteins were identified in response to IVR. The most notable GO annotation describes the altered proteins was “innate immune response”. The most notable REACTOME pathway was “platelet degranulation”. ELISA result showed increased level of APOC1, SERPINA5, KRT1 and a decreased level of TIMP2 in PDR group compared with IVR. Conclusions In addition to decreasing VEGF level, ranibizumab is associated with change of human vitreous protein profile in patients with PDR, in which the differential proteins are involved in immune response, platelet degranulation, complement activation etc., suggesting that the effects of VEGF are involved in these signaling pathways.

This study was to investigate the relationship of serum mitochondrial coupling factor 6 (CF6) with blood lipid and glucose in patients with type 2 diabetic retinopathy. A total of 180 patients with type 2 diabetes enrolled in our hospital from January 2015 to September 2017 were selected as the research objects. Abstract Background Recent evidence indicates that long noncoding RNAs (lncRNAs) play pivotal roles in the regulation of cellular processes and are found to be dysregulated in a variety of cancers. LINC00261 is an lncRNA that is aberrantly expressed in gastric cancer (GC). The clinical role of LINC00261 in GC and molecular mechanisms remains to be found. Methods Real-time polymerase chain reaction (PCR) was used to examine LINC00261 expression in GC cell lines/tissues compared with normal epithelial cells/adjacent non-tumorous tissues. Gain and loss of function approaches were used to investigate the biological role of LINC00261 in GC cells. Purpose. To examine the difference in the vitreal protein profiles of patients with proliferative diabetic retinopathy (PDR) with and without preoperative intravitreal conbercept (IVC) treatment. Methods. Liquid chromatography-tandem mass spectrometry- (LC-MS/MS-) based proteomic methods were used to determine the protein profiles of the vitreous humor in patients with PDR treated with (IVC group; n=9) and without (PDR group; n=8) preoperative IVC. Gene ontology (GO) annotation and REACTOME pathway analysis were obtained to overview differentially expressed proteins between each group. Background: microRNAs (miRNAs) are small non-coding RNAs and have been shown to play a crucial role in the colorectal cancer (CRC) tumorigenesis and progression. The aim of this study was to investigate the clinical significance and prognostic value of miR-140-5p in CRC. The exact functions and the underlying molecular mechanisms of miR-140-5p in CRC was further determined. Methods: miR-140-5p expression was detected in CRC samples, their adjacent nontumor tissues as well as CRC cell lines by RT-qPCR. Cell proliferation was detected using CCK-8, and cell invasion and migration were evaluated using Transwell assay. The direct regulation of VEGFA by miR-140-5p was identified using luciferase reporter assay. Results: miR-140-5p was significantly dowregulated in CRC tissues and cell lines. Downregulation of miR-140-5p was significantly correlated with advanced CRC stage and poorer overall survival. Both gain-of-function and loss of function studies demonstrated that miR-140-5p acted as a tumor suppressor by inhibiting cell proliferation, migration and invasion. Integrated analysis identified VEGFA as a direct and functional target gene of miR-140-5p. Silencing VEGFA by small interfering RNA (siRNA) resembled the phenotype resulting from ectopic miR-140-5p expression, while overexpression of VEGFA attenuated the effect of miR-140-5p on CRC cells. Conclusions: Our results suggested a tumor suppressive role of miR-140-5p in CRC tumorigenesis and progression by targeting VEGFA.

Changes of Selenoprotein F (SELENOF) protein levels have been reported during selenium supplementation, stressful, and pathological conditions. However, the mechanisms of how these external factors regulate SELENOF gene expression are largely unknown. In this study, HEK293T cells were chosen as an in vitro model. The 5′-flanking regions of SELENOF were analyzed for promoter features. Dual-Glo Luciferase assays were used to detect promoter activities. Putative binding sites of Heat Shock Factor 1 (HSF1) were predicted in silico and the associations were further proved by chromatin immunoprecipitation (ChIP) assay. Selenate and tunicamycin (Tm) treatment were used to induce SELENOF up-regulation. The fold changes in SELENOF expression and other relative proteins were analyzed by Q-PCR and western blot. Our results showed that selenate and Tm treatment up-regulated SELENOF at mRNA and protein levels. SELENOF 5′-flanking regions from −818 to −248 were identified as core positive regulatory element regions. Four putative HSF1 binding sites were predicted in regions from −1430 to −248, and six out of seven primers detected positive results in ChIP assay. HSF1 over-expression and heat shock activation increased the promoter activities, and mRNA and protein levels of SELENOF. Over-expression and knockdown of HSF1 showed transcriptional regulation effects on SELENOF during selenate and Tm treatment. In conclusion, HSF1 was discovered as one of the transcription factors that were associated with SELENOF 5′-flanking regions and mediated the up-regulation of SELENOF during selenate and Tm treatment. Our work has provided experimental data for the molecular mechanism of SELENOF gene regulation, as well as uncovered the involvement of HSF1 in selenotranscriptomic for the first time.

Materials capable of circularly polarized luminescence (CPL) have attracted considerable attention for their promising potential applications. Bacterial cellulose (BC) was characterized as having a stable right-handed twist, which makes it a potential chiral host to endow luminophores with CPL. Then, the CPL-active BC composite film was constructed by simply impregnating bacterial cellulose pellicles with dilute aqueous solutions of luminophores (rhodamine B, carbon dots, polymer dots) and drying under ambient conditions. Simple encapsulation of luminophores renders BC with circularly polarized luminescence with a dissymmetry factor of up to 0.03. The multiple chiral centers of bacterial cellulose provide a primary asymmetric environment that can be further modulated by supramolecular chemistry, which is responsible for its circular polarization ability. We further demonstrate that commercial grade paper may endow luminophores with CPL activity, which reifies the universality of the method.

High incidence rate of Alzheimer’s disease (AD) is observed in patients with type 2 diabetes. Aggregated β-amyloid (Aβ) and hyperphosphorylated tau are the hallmarks of AD. Hyperphosphorylated tau has been detected in diabetic animals as well as in diabetic patients. Folates mediate the transfer of one carbon unit, required in various biochemical reactions. The effect of folate on tau phosphorylation in diabetic models still remains unknown. In this study, we investigated the effect and mechanism of folic acid on hyperphosphorylation of tau in streptozotocin (STZ)-induced diabetic mice. Diabetic mice induced by STZ, at the age of 10 weeks, were administered with three levels of folic acid: folic acid-deficient diet, diet with normal folic acid content, and 120 μg/kg folic acid diet for 8 weeks. Levels of serum folate and blood glucose were monitored. Tau phosphorylation, protein phosphatase 2A (PP2A) methylation, and Glycogen synthase kinase 3β (GSK-3β) phosphorylation were detected using Western blot. The S-adenosyl methionine:S-adenosyl homocysteine ratio (SAM:SAH) in brain tissues was also determined. DNA methyltransferase (DNMT) mRNA expression levels were detected using real-time PCR. Folic acid reduced tau hyperphosphorylation at Ser396 in the brain of diabetes mellitus (DM) mice. In addition, PP2A methylation and DNMT1 mRNA expression were significantly increased in DM mice post folic acid treatment. GSK-3β phosphorylation was not regulated by folic acid administration. Folic acid can reduce tau phosphorylation by regulating PP2A methylation in diabetic mice. These results support that folic acid can serve as a multitarget neuronal therapeutic agent for treating diabetes-associated cognitive dysfunction.

In this work, we studied propylene polymerization using some α-diimine palladium catalysts with systematically varied ligand sterics. In propylene polymerization, the ligand steric effect exhibits significant variations on the catalytic activity, polymer molecular weight, and branching density. However, the regio control for the polymer microstructure is poor. Furthermore, copolymerization of 1-octene with the highly challenging and biorenewable comonomer acrylic acid was investigated. High copolymer molecular weights and high comonomer incorporation ratios could be achieved in this system. This study provides a novel access for the direct synthesis of branched carboxylic acid functionalized polyolefins.
