5. Transient receptor potential vanilloid 4-dependent calcium influx, ATP release and expression of inflammatory factors in enteric glial cells

Abstract

Objective To investigate the role and physiological significance of transient receptor potential vanilloid receptor 4 (TRPV4) in regulation of calcium influx in rat intestinal glial cells. Methods The expression of TRPV4 at mRNA and protein levels and its localization were detected by RT-qPCR, Western blotting and immunofluorescence assay in rat enteric glial cell line CRL-2690, rat intestinal epithelial cell line IEC6 and human embryonic kidney cell line HEK293T. The CRL-2690 cells were divided into GSK1016790A (GSK, TRPV4-specific agonist) group and GSK+HC067047 (HC, TRPV4-specific inhibitor) group, hypotonic group and hypotonic+HC group, GdCl3 group and GdCl3+HC group, and CaCl2 group and CaCl2+HC group, while the activity of TRPV4 channel was assessed by Fura-2 fluorescent probe and single-cell fluorescent calcium ion assay. CRL-2690 cells were also divided into control group, GSK group and GSK+HC group, and the level of cellular ATP release was measured by luciferin-luciferase assay, and the mRNA levels of inflammatory genes GFAP, IL-1β, IL-6 and IL-10 were detected by RT-qPCR. Results TRPV4 was expressed at mRNA and protein levels in rat intestinal glial cell line CRL-2690. GSK and low osmolarity stimulation significantly enhanced intracellular calcium fluorescence intensity in CRL-2690 cells, which was inhibited by HC (0.430 38±0.063 65 vs 0.004 23±0.005 78, P < 0.000 1). Calcium-sensing receptor (CaSR) activation-induced Ca2+ influx was also inhibited by HC (P < 0.05). Functionally, activation of TRPV4 promoted ATP release, expression of EGC reactive proliferation marker, GFAP, and inflammatory factors IL-1 β as well as IL-6, and inhibited the expression of anti-inflammatory factor IL-10 respectively. But all these effects were inhibited by HC treatment (P < 0.05). Conclusion Activation of TRPV4 induces extracellular Ca2+ influx and promotes ATP release and expression of inflammatory cytokines in enteric glial cell, which may be closely involved in the underlying mechanisms of bowel inflammation.

Abstract

Electrified railroad has the characteristics of high safety factors, high comfort, large transportation capacity, and less time-consuming, which is an effective means to solve traffic congestion. However, with the rapid development of China’s electrified railroad, harmonic, negative sequence and other power quality problems have also been widely concerned, while more and more regenerative braking energy in the traction power supply system can not be effectively utilized. The input of railroad power regulators makes the power quality problem be solved effectively, this paper studies the control strategy of traditional railroad power regulators. Combined with the actual working conditions of railroad vehicles, the simulation model of traction network, vehicles, and railroad power conditioner is established, and the load characteristics of traction substation are simulated by this model. Secondly, for a domestic substation field collection data processing to obtain the actual train operation load changes, to the battery energy storage device life within the maximum economic returns as the optimization goal, for the peak load reduction judgment threshold and the capacity of the battery energy storage device through the genetic algorithm to process the data of previous years to optimize the design.

Abstract

Background: This study aimed to evaluate the clinical application of the preoperative prealbumin-to-fibrinogen ratio (PFR) in the clinical diagnosis of hepatocellular carcinoma (HCC) patients and its prognostic value. Methods: The clinical and laboratory data of 269 HCC patients undergoing surgical treatment from January 2012 to January 2017 in Taizhou Hospital were retrospectively analysed. The Cox regression model was used to analyse the correlation between the PFR and other clinicopathological factors in overall survival (OS) and disease-free survival (DFS). Results: Cox regression analysis showed that the PFR (hazard ratio (HR)=2.123; 95% confidence interval (95% CI), 1.271-3.547; P=0.004) was an independent risk factor affecting the OS of HCC patients. Furthermore, a nomogram was built based on these risk factors. The C-index for the OS nomogram was 0.715. Conclusions: Nomograms based on the PFR can be recommended as the correct and actual model to evaluate the prognosis of patients with HCC.

18. Preoperative lymphocyte-to-monocyte ratio versus platelet-to-lymphocyte ratio as a prognostic predictor for non-small cell lung cancer patients

Abstract

Background: We investigated the prognostic value of the preoperative lymphocyte-to-mononuclear ratio (LMR) and platelet-to-lymphocyte ratio (PLR) in a large cohort of patients with non-small cell lung cancer (NSCLC). Methods: Clinical-pathological data from 507 NSCLC patients at Taizhou Hospital of Zhejiang Province between 2010 and 2016 were retrospectively evaluated. X-tile software was used to assess the optimal cutoff levels for LMR and PLR. Univariate and multivariate Cox regression models were used to assess the prognostic factors. Results: The median follow-up duration after surgical resection was 34.5 months. Patients were stratified into 2 groups by LMR (2.6 and > 2.6) and PLR (179.6 and > 179.6). Our results revealed that lower LMR (HR = 3.163 (1.821-5.493), P = 0.000), age (HR = 2.252 (1.412-3.592), P = 0.001), T stage (HR = 3.749 (2.275-6.179), P = 0.000), N stage (HR = 3.106 (1.967-4.902), P = 0.000), and cut edge (HR = 3.830 (1.077-13.618), P = 0.038) were considered to be independent indicators for overall survival (OS) of NSCLC patients. For disease-free survival (DFS), age, sex, T stage, N stage, LMR and cut edge were verified to be independent prognostic factors in patients with NSCLC. Conclusions: In the study cohort, reduced LMR was a robust independent predictor for both OS and DFS in patients with NSCLC who underwent surgical resection.

However, the long-term observation of patients’ liver functions after discharge has not been investigated. This study intends to analyze the abnormal liver function in patients one year after they are discharged.Methods: Serum liver function tests were analyzed for the first time immediately after hospitalization (T1), before discharge (T2), a median of 14.0 (14.0, 15.0) days after discharge (T3) and 1 year (356.0 (347.8, 367.0) days) after discharge (T4). Patients with at least one serum parameter (ALT, AST, ALP, GGT and TB) exceeding the upper limit of reference range were defined as having abnormal liver function.Results: For the 118 COVID-19 patients with a median follow-up time of 376.0 (71.5, 385.3) days from onset to the end of the follow-up after discharge, the proportion with abnormal liver function in T1, T2, T3 and T4 were 32.2%, 45.8%, 54.8% and 28.8%, respectively. The proportion of patients with at least once abnormal liver function detected from T1 to T2, T1 to T3, T1 to T4 was 60.2%, 77.4% and 88.9%, respectively. From T1 to T4, the ALT, AST, GGT and BMI at admission were significantly higher in the patients with persistently abnormal liver function than in the patients with persistently normal liver function. Abstract

The relationship among the gut microbiome, global fecal metabolites and rheumatoid arthritis (RA) has not been systematically evaluated. In this study, we performed 16S rDNA sequencing and liquid chromatography-tandem mass spectrometry (LC-MS/MS)-based nontargeted metabolomic profiling on feces of 26 untreated RA patients and 26 healthy controls. Twenty-six genera and forty-one MS2-identified metabolites were significantly altered in the RA patients. Klebsiella, Escherichia, Eisenbergiella and Flavobacterium were more abundant in the RA patients, while Fusicatenibacter, Megamonas and Enterococcus were more abundant in the healthy controls. Function prediction analysis demonstrated that the biosynthesis pathways of amino acids, such as L-arginine and aromatic amino acids, were depleted in the RA group. In the metabolome results, fecal metabolites including glycerophospholipids (PC(18:3(9Z,12Z,15Z)/16:1(9Z)), lysoPE 19:1, lysoPE 18:0, lysoPC(18:0/0:0)), sphingolipids (Cer(d18:0/16:0), Cer(d18:0/12:0), Cer(d18:0/14:0)), kynurenic acid, xanthurenic acid and 3-hydroxyanthranilic acid were remarkably altered between the RA patients and healthy controls. Dysregulation of pathways, such as tryptophan metabolism, alpha-linolenic acid metabolism and glycerophospholipid metabolism, may contribute to the development of RA. Additionally, we revealed that the gut microbiome and metabolites were interrelated in the RA patients, while Escherichia was the core genus. By depicting the overall landscape of the intestinal microbiome and metabolome in RA patients, our study could provide possible novel research directions regarding RA pathogenesis and targeted therapy.

Abstract

Hot deformation behavior of Al–0.2Sc–0.04Zr (wt.%) aluminum alloy was investigated via conducting hot compression tests at temperatures of 440–600 °C and at strain rates, ranging from 0.001 to 5 s−1, at an interval of an order of magnitude. The characteristics of the true stress–strains acquired in the hot compression tests were investigated, and the influence of processing parameters on the microstructure of the deformed samples was observed by using optical microscope and electron back-scattered diffraction (EBSD). Two types of processing maps for Al–0.2Sc–0.04Zr alloy were developed using the Kumar–Prasad (K–P) and Murty–Rao (M–R) instability/stability criteria, respectively. The prediction results of two types of processing maps were examined. Based on the analysis of the M–R processing map and EBSD, a deformation mechanism map covering a wide range of temperature and strain rate was developed. The result shows that the flow stress had a rapid increase to the maximum stress, and subsequently, the flow stress decreases continually to be of steady-state type when the deformation continues on. The M–R criterion can more accurately predict the unstable region than the K–P criterion. The optimum temperature T and strain rate $$ \dot{\varepsilon } $$ combination conditions for Al–0.2Sc–0.04Zr alloy are T = 520–560 °C, $$ \dot{\varepsilon } $$ = 10−3 s−1 or $$ \dot{\varepsilon } $$ = 0.1–5 s−1. The microstructure evolution dominated by the predominant or complete dynamic recrystallization can be acquired by deformation at the combination conditions. However, it is dominated mainly by dynamic recovery at lower temperatures (T 520 °C). In addition, the microstructure of specimen deformed at 600 °C shows a growing and coarse trend. Moreover, the nano-dimension precipitate particles of Al3(Sc,Zr) are uniformly distributed throughout Al matrix. The efficiency parameter η value of Al–0.2Sc–0.04Zr alloy in processing maps is smaller in comparison with that in pure aluminum, and it could be attributed to these nano-dimension particles.

Abstract

The microstructure, deformation mechanisms, dynamic recrystallization (DRX) behavior, and texture evolution of AZ80 magnesium alloy were investigated by three-pass cyclic expansion-extrusion (CEE) tests. Optical microscopy (OM), electron back-scattered diffraction (EBSD), and X-ray diffraction (XRD) were employed to study microstructure, grain orientation, DRX mechanism, and texture evolution. The results show that the grain sizes decrease continuously with the increase of CEE pass. The grain refinement effect of the first pass is the most remarkable, and there appear a large number of twins. After three-pass CEE, a well-distributed structure with fine equiaxed grains is obtained. With the increase of CEE pass, the deformation mechanism changes from twinning to slipping and the DRX mechanism changes mainly from twinning-induced dynamic recrystallization (TDRX) to rotation dynamic recrystallization (RDRX) and then to continuous dynamic recrystallization (CDRX). The grain misorientation between the new grains and matrix grains deceases gradually, and a relatively small angle misorientation is obtained after three-pass CEE. Grain misorientations of the first two passes are attributed to TDRX and RDRX behaviors, respectively. The grain refinement changes the deformation and DRX mechanisms of CEE process, which leads the (0002) basal texture intensity first decrease and then increase suddenly. Eventually, the extremely strong basal texture is formed after three-pass CEE.
