Astroparticle physics is undergoing a profound transformation, due to a series of extraordinary new results, such as the discovery of high-energy cosmic neutrinos with IceCube, the direct detection of gravitational waves with LIGO and Virgo, and many others. This white paper is the result of a collaborative effort that involved hundreds of theoretical astroparticle physicists and cosmologists, under the coordination of the European Consortium for Astroparticle Theory (EuCAPT). Abstract

In this paper, which is of programmatic rather than quantitative nature, we aim to further delineate and sharpen the future potential of the LISA mission in the area of fundamental physics. Given the very broad range of topics that might be relevant to LISA, we present here a sample of what we view as particularly promising directions, based in part on the current research interests of the LISA scientific community in the area of fundamental physics. We organize these directions through a "science-first" approach that allows us to classify how LISA data can inform theoretical physics in a variety of areas. For each of these theoretical physics classes, we identify the sources that are currently expected to provide the principal contribution to our knowledge, and the areas that need further development. Abstract

Teleparallel gravity and its popular generalization $f(T)$ gravity can be formulated as fully invariant (under both coordinate transformations and local Lorentz transformations) theories of gravity. Several misconceptions about teleparallel gravity and its generalizations can be found in the literature, especially regarding their local Lorentz invariance. We describe how these misunderstandings may have arisen and attempt to clarify the situation. In particular, the central point of confusion in the literature appears to be related to the inertial spin connection in teleparallel gravity models. While inertial spin connections are commonplace in special relativity, and not something inherent to teleparallel gravity, the role of the inertial spin connection in removing the spurious inertial effects within a given frame of reference is emphasized here. The careful consideration of the inertial spin connection leads to the construction of a fully invariant theory of teleparallel gravity and its generalizations. Indeed, it is the nature of the spin connection that differentiates the relationship between what have been called good tetrads and bad tetrads and clearly shows that, in principle, any tetrad can be utilized. The field equations for the fully invariant formulation of teleparallel gravity and its generalizations are presented and a number of examples using different assumptions on the frame and spin connection are displayed to illustrate the covariant procedure. Abstract

The Kosambi-Cartan-Chern (KCC) theory represents a powerful mathematical method for the analysis of dynamical systems. In this approach one describes the evolution of a dynamical system in geometric terms, by considering it as a geodesic in a Finsler space. By associating a non-linear connection and a Berwald type connection to the dynamical system, five geometrical invariants are obtained, with the second invariant giving the Jacobi stability of the system. The Jacobi (in)stability is a natural generalization of the (in)stability of the geodesic flow on a differentiable manifold endowed with a metric (Riemannian or Finslerian) to the non-metric setting. In the present paper we review the basic mathematical formalism of the KCC theory, and present some specific applications of this method in general relativity, cosmology and astrophysics. In particular we investigate the Jacobi stability of the general relativistic static fluid sphere with a linear barotropic equation of state, of the vacuum in the brane world models, of a dynamical dark energy model, and of the Lane-Emden equation, respectively. 5. Can dark matter be a Bose-Einstein condensate?

Abstract

We consider the possibility that the dark matter, which is required to explain the dynamics of the neutral hydrogen clouds at large distances from the galactic center, could be in the form of a Bose-Einstein condensate. To study the condensate we use the non-relativistic Gross-Pitaevskii equation. By introducing the Madelung representation of the wave function, we formulate the dynamics of the system in terms of the continuity equation and of the hydrodynamic Euler equations. Hence dark matter can be described as a non-relativistic, Newtonian Bose-Einstein gravitational condensate gas, whose density and pressure are related by a barotropic equation of state. In the case of a condensate with quartic non-linearity, the equation of state is polytropic with index $n=1$. To test the validity of the model we fit the Newtonian tangential velocity equation of the model with a sample of rotation curves of low surface brightness and dwarf galaxies, respectively. We find a very good agreement between the theoretical rotation curves and the observational data for the low surface brightness galaxies. The deflection of photons passing through the dark matter halos is also analyzed, and the bending angle of light is computed. The bending angle obtained for the Bose-Einstein condensate is larger than that predicted by standard general relativistic and dark matter models. 6. Galactic dark matter as a bulk effect on the brane

Abstract

The behavior of the angular velocity of a test particle moving in a stable circular orbit in the vacuum on the brane is considered. In the brane world scenario, the four dimensional effective Einstein equation acquire extra terms, called dark radiation and dark pressure, respectively, which arise from the embedding of the 3-brane in the bulk. A large number of independent observations have shown that the rotational velocities of test particles gravitating around galaxies tend, as a function of the distance from the galactic center, toward constant values. By assuming a constant tangential velocity, the general solution of the vacuum gravitational field equations on the brane can be obtained in an exact analytic form. This allows us to obtain the explicit form of the projections of the bulk Weyl tensor on the brane, and the equation of state of the dark pressure as a function of the dark radiation. The physical and geometrical quantities are expressed in terms of observable/measurable parameters, like the tangential velocity, the baryonic mass and the radius of the galaxy. We also analyze the dynamics of test particles by using methods from the qualitative analysis of dynamical systems, by assuming a simple linear equation of state for the dark pressure. 7. On Einstein clusters as galactic dark matter halos

Abstract

We consider global and gravitational lensing properties of the recently suggested Einstein clusters of WIMPs as galactic dark matter halos. Being tangential pressure dominated, Einstein clusters are strongly anisotropic systems which can describe any galactic rotation curve by specifying the anisotropy. Due to this property, Einstein clusters may be considered as dark matter candidates. We analyse the stability of the Einstein clusters against both radial and non-radial pulsations, and we show that the Einstein clusters are dynamically stable. With the use of the Buchdahl type inequalities for anisotropic bodies, we derive upper limits on the velocity of the particles defining the cluster. These limits are consistent with those obtained from stability considerations. The study of light deflection shows that the gravitational lensing effect is slightly smaller for the Einstein clusters, as compared to the singular isothermal density sphere model for dark matter. 8. Minimum mass-radius ratio for charged gravitational objects

Abstract

We rigorously prove that for compact charged general relativistic objects there is a lower bound for the mass-radius ratio. This result follows from the same Buchdahl type inequality for charged objects, which has been extensively used for the proof of the existence of an upper bound for the mass-radius ratio. The effect of the vacuum energy (a cosmological constant) on the minimum mass is also taken into account. Several bounds on the total charge, mass and the vacuum energy for compact charged objects are obtained from the study of the Ricci scalar invariants. 9. The Einstein-Elko system -- Can dark matter drive inflation?

Abstract

Recently, a spin one half matter field with mass dimension one was discovered, called Elko spinors. The present work shows how to introduce these fields into a curved spacetime by the standard covariantisation scheme. After formulating the coupled Einstein-Elko field equations, the spacetime is assumed to be homogeneous and isotropic in order to simplify the resulting field equations. Analytical ghost Elko solutions are constructed which have vanishing energy-momentum tensor without and with cosmological constant. The cosmological Elko theory is finally related to the standard scalar field theory with self interaction that gives rise to inflation and it is pointed out that the Elko spinors are not only prime dark matter candidates but also prime candidates for inflation., Comment: 24 pages

We propose that the Universe is filled with a massive vector field, non-minimally coupled to gravitation. The field equations of the model are consistently derived and their application to cosmology is considered. The Friedmann equations acquire an extra dark-energy component, which is proportional to the mass of the vector particle. This leads to a late-time accelerated de Sitter type expansion. The free parameters of the model (gravitational coupling constants and initial value of the cosmological vector field) can be estimated by using the PPN solar system constraints. 11. Bounds on the basic physical parameters for anisotropic compact general relativistic objects

Abstract

We derive upper and lower limits for the basic physical parameters (mass-radius ratio, anisotropy, redshift and total energy) for arbitrary anisotropic general relativistic matter distributions in the presence of a cosmological constant. The values of these quantities are strongly dependent on the value of the anisotropy parameter (the difference between the tangential and radial pressure) at the surface of the star. In the presence of the cosmological constant, a minimum mass configuration with given anisotropy does exist. Anisotropic compact stellar type objects can be much more compact than the isotropic ones, and their radii may be close to their corresponding Schwarzschild radii. Upper bounds for the anisotropy parameter are also obtained from the analysis of the curvature invariants. General restrictions for the redshift and the total energy (including the gravitational contribution) for anisotropic stars are obtained in terms of the anisotropy parameter. Abstract

We consider the astrophysical and cosmological implications of the existence of a minimum density and mass due to the presence of the cosmological constant. If there is a minimum length in nature, then there is an absolute minimum mass corresponding to a hypothetical particle with radius of the order of the Planck length. On the other hand, quantum mechanical considerations suggest a different minimum mass. These particles associated with the dark energy can be interpreted as the ``quanta'' of the cosmological constant. We study the possibility that these particles can form stable stellar-type configurations through gravitational condensation, and their Jeans and Chandrasekhar masses are estimated. From the requirement of the energetic stability of the minimum density configuration on a macroscopic scale one obtains a mass of the order of 10^55 g, of the same order of magnitude as the mass of the universe. This mass can also be interpreted as the Jeans mass of the dark energy fluid. Abstract

The equations describing the adiabatic, small radial oscillations of general relativistic stars are generalized to include the effects of a cosmological constant. The generalized eigenvalue equation for the normal modes is used to study the changes in the stability of the homogeneous sphere induced by the presence of the cosmological constant. The variation of the critical adiabatic index as a function of the central pressure is studied numerically for different trial functions. The presence of a large cosmological constant significantly increases the value of the critical adiabatic index. Abstract

In this paper, which is of programmatic rather than quantitative nature, we aim to further delineate and sharpen the future potential of the LISA mission in the area of fundamental physics. Given the very broad range of topics that might be relevant to LISA, we present here a sample of what we view as particularly promising directions, based in part on the current research interests of the LISA scientific community in the area of fundamental physics. We organize these directions through a "science-first" approach that allows us to classify how LISA data can inform theoretical physics in a variety of areas. For each of these theoretical physics classes, we identify the sources that are currently expected to provide the principal contribution to our knowledge, and the areas that need further development. The classification presented here should not be thought of as cast in stone, but rather as a fluid framework that is amenable to change with the flow of new insights in theoretical physics.

Abstract

In this paper, which is of programmatic rather than quantitative nature, we aim to further delineate and sharpen the future potential of the LISA mission in the area of fundamental physics. Given the very broad range of topics that might be relevant to LISA,we present here a sample of what we view as particularly promising fundamental physics directions. We organize these directions through a “science-first” approach that allows us to classify how LISA data can inform theoretical physics in a variety of areas. For each of these theoretical physics classes, we identify the sources that are currently expected to provide the principal contribution to our knowledge, and the areas that need further development. The classification presented here should not be thought of as cast in stone, but rather as a fluid framework that is amenable to change with the flow of new insights in theoretical physics.

Abstract

Mechanisms of regulatory cell volume increase following cell shrinkage include accumulation of organic osmolytes such as betaine, taurine, sorbitol, glycerophosphorylcholine (GPC) and myo-inositol. Myo-inositol is taken up by the sodium-myo-inositol-transporter SMIT1 (SLC5A3) expressed in a wide variety of cell types. Hypertonicity induces the transcription of the SMIT1 gene upon binding of the transcription factor tonicity enhancer binding protein (TonEBP) to tonicity responsive enhancers (TonE) in the SMIT1 promoter region. However, little is known about post-translational regulation of the carrier protein. In this study we show that SMIT1 is modulated by the serum- and glucocorticoid-inducible kinase SGK1, a protein genomically up-regulated by hypertonicity. As demonstrated by two-electrode voltage-clamp in the Xenopus oocyte expression system, SMIT1-mediated myo-inositol-induced currents are up-regulated by coexpression of wild type SGK1 and constitutively active (S422D)SGK1 but not by inactive (K127N)SGK1. The increase in SMIT1 activity is due to an elevated cell surface expression of the carrier while its kinetic properties remain unaffected. According to the decay of SMIT1 activity in the presence of brefeldin A, SGK1 stabilizes the SMIT1 protein in the plasma membrane. The SGK isoforms SGK2, SGK3 and the closely related protein kinase B (PKB) are similarly capable of activating SMIT1 activity. SMIT1-mediated currents are decreased by coexpression of the ubiquitin-ligase Nedd4-2, an effect counteracted by additional coexpression of SGK1. In conclusion, the present observations disclose SGK isoforms and protein kinase B as novel regulators of SMIT1 activity.

Contains fulltext : 48079.pdf (Publisher’s version ) (Open Access) Renal calcium reabsorption involves the epithelial calcium channel ECaC1 (TRPV5) which is tightly regulated by 1,25(OH)2D3. As shown recently, TRPV5 is activated by the serum and glucocorticoid inducible kinase SGK1, a kinase transcriptionally upregulated by 1,25(OH)2D3. This stimulatory effect is due to enhanced TRPV5 abundance in the plasma membrane and requires the presence of the scaffold protein NHERF2 (sodium hydrogen exchanger regulating factor 2). The present study aims to define the molecular requirements for the interaction of TRPV5 with SGK1 and NHERF2. Pull-down experiments and overlay assays revealed that the TRPV5 C-tail interacts in a Ca2+-independent manner with NHERF2. Deletion of the second but not of the first PDZ domain in NHERF2 abrogates the stimulating effect of SGK1/NHERF2 on TRPV5 protein abundance in the plasma membrane as quantified by chemiluminescence and electrophysiology. Thus, the second PDZ domain in NHERF2 is required for stabilization at or TRPV5 targeting to the plasma membrane. The experiments demonstrate the significance of SGK1 and NHERF2 as TRPV5 modulators which are likely to participate in the regulation of calcium homeostasis by 1,25(OH)2D3.

To this end cRNA encoding TRPV5 has been injected into Xenopus oocytes with or without additional injection of SGK1, its isoforms SGK2 and SGK3, constitutively active (S422D)SGK1, inactive (K127N)SGK1, constitutively active (T308D,S473D)PKB and/or the Na+/H+ exchanger regulating factor NHERF2. In Xenopus laevisoocytes expression of TRPV5 increases uptake of tracer Ca(S422D;) and induces a Ca2+ current (ICa). In the presence of Cl-, TRPV5 mediated Ca2+ entry leads to secondary activation of Ca(2+)-sensitive Cl- channels (ICl(Ca)). Coexpression of TRPV5 with both (S422D)SGK1 and NHERF2 stimulates tracer Ca2+ entry, ICa and ICl(Ca). The effect of (S422D)SGK1 on TRPV5 and NHERF2 expressing oocytes is mimicked by SGK1 and SGK3, but not by SGK2, constitutively active (T308D,S473D)PKB or inactive (K127N)SGK1. Abstract

The New Mexico State University Satellite (NMSUSat) is part of the University Nanosat 3 program managed by the Air Force Research Laboratory and it is being developed at New Mexico State University. The planned Science Mission for the satellite is to perform Near Ultra Violet emission intensity measurements of the earth's upper atmosphere over the night side of the earth. The Engineering Mission is to demonstrate techniques for distributed data relaying over the Internet and to conduct an energy storage experiment to assess the operational characteristics of double layer capacitors. The Educational Mission of the program to assist in the further development of the aerospace engineering concentration area in the College of Engineering and to develop multi-disciplinary capstone and design classes for students in engineering departments, computer science, and the engineering physics program. This paper will discuss the preliminary design for the satellite components and how the mission segments will be worked among the participating departments at New Mexico State University.

Abstract

PURPOSE. The serum- and glucocorticoid-inducible kinase SGK1 regulates several epithelial channels and transporters, the related protein kinase B (PKB) regulates glucose transport. SGK1 is expressed in the brain and could thus regulate glial and/or neuronal transport processes. The present study explores whether SGK1 is expressed in the retina and whether it regulates EAAT3, a Na+-coupled glutamate transporter. EAAT3 is expressed in retinal ganglion cells and accomplishes the clearance of glutamate from synaptic clefts. METHODS. Immunohistochemistry was performed to test for retinal SGK1 expression. For functional analysis, cRNA encoding EAAT3 was injected into Xenopus oocytes with or without additional injection of wild-type SGK1, constitutively active S422DSGK1, inactive K127NSGK1, and/or constitutively active T308D,S473DPKB. Glutamate induced current (IGLU) was taken as a measure for transport. RESULTS. SGK1 is indeed expressed in several retinal cells including retinal ganglion cells where it is colocalized with EAAT3. In EAAT3-expressing Xenopus oocytes, glutamate-induced current was stimulated by coexpression of wild-type SGK1, constitutively active S422DSGK1, and constitutively active T308D,S473DPKB, but not by inactive K127NSGK1. CONCLUSIONS. SGK1 and EAAT3 are coexpressed in retinal neurons, and SGK1 serves to stimulate EAAT3. This function is shared by protein kinase B (PKB). The experiments reveal a novel mechanism regulating EAAT3, which may be essential for the function of the retinal ganglion cells.

Abstract

Surface expression of the glial glutamate transporter EAAT1 is stimulated by insulin-like growth factor 1 through activation of phosphatidylinositol-3-kinase. Downstream targets include serum and glucocorticoid-sensitive kinase isoforms SGK1, SGK2 and SGK3, and protein kinase B. SGK1 regulates Nedd4-2, a ubiquitin ligase that prepares cell membrane proteins for degradation. To test whether Nedd4-2, SGK1, SGK3 and protein kinase B regulate EAAT1, cRNA encoding EAAT1 was injected into Xenopus oocytes with or without additional injection of wild-type Nedd4-2, constitutively active S422DSGK1, inactive K127NSGK1, wild-type SGK3 and/or constitutively active T308D,S473DPKB. Glutamate induces a current in Xenopus oocytes expressing EAAT1, but not in water-injected oocytes, which is decreased by co-expression of Nedd4-2, an effect reversed by additional co-expression of S422DSGK1, SGK3 and T308D,S473DPKB, but not K127NSGK1. Site-directed mutagenesis of the SGK1 phosphorylation sites in the Nedd4-2 protein ( S382A,S468ANedd4-2) and in the EAAT1 protein (T482AEAAT1, T482DEAAT1) significantly blunts the effect of S422DSGK1. Moreover, the current is significantly larger in T482DEAAT1- than in T482AEAAT1-expressing oocytes, indicating that a negative charge mimicking phosphorylation at T482 increases transport. The experiments reveal a powerful novel mechanism that regulates the activity of EAAT1. This mechanism might participate in the regulation of neuronal excitability and glutamate transport in other tissues.

The amino acid transporter SN1 with substrate specificity identical to the amino acid transport system N is expressed mainly in astrocytes and hepatocytes where it accomplishes Na+-coupled glutamine uptake and efflux. To characterize properties and regulation of SN1, substrate-induced currents and/or radioactive glutamine uptake were determined in Xenopus oocytes injected with cRNA encoding SN1, the ubiquitin ligase Nedd4-2, and/or the constitutively active serum and glucocorticoid inducible kinase S422DSGK1, its isoform SGK3, and the constitutively active protein kinase B T308D,S473DPKB. The substrate-induced currents were enhanced by increasing glutamine and/or Na+ concentrations, hyperpolarization, and alkalinization (pH 8.0). They were inhibited by acidification (pH 6.0). Coexpression of Nedd4-2 downregulated SN1-mediated transport, an effect reversed by coexpression of S422DSGK1, SGK3, and T308D,S473DPKB. It is concluded that SN1 is a target for the ubiquitin ligase Nedd4-2, which is inactivated by the serum and glucocorticoid inducible kinase SGK1, its isoform SGK3, and protein kinase B.

33. Substance P receptor binding sites are expressed by glia in vivo after neuronal injury.

Abstract

In vitro studies have demonstrated that glia can express functional receptors for a variety of neurotransmitters. To determine whether similar neurotransmitter receptors are also expressed by glia in vivo, we examined the glial scar in the transected optic nerve of the albino rabbit by quantitative receptor autoradiography. Receptor binding sites for radiolabeled calcitonin gene-related peptide, cholecystokinin, galanin, glutamate, somatostatin, substance P, and vasoactive intestinal peptide were examined. Specific receptor binding sites for each of these neurotransmitters were identified in the rabbit forebrain but were not detected in the normal optic nerve or tract. In the transected optic nerve and tract, only receptor binding sites for substance P were expressed at detectable levels. The density of substance P receptor binding sites observed in this glial scar is among the highest observed in the rabbit forebrain. Ligand displacement and saturation experiments indicate that the substance P receptor binding site expressed by the glial scar has pharmacological characteristics similar to those of substance P receptors in the rabbit striatum, rat brain, and rat and canine gut. The present study demonstrates that glial cells in vivo express high concentrations of substance P receptor binding sites after transection of retinal ganglion cell axons. Because substance P has been shown to regulate inflammatory and immune responses in peripheral tissues, substance P may also, by analogy, be involved in regulating the glial response to injury in the central nervous system.

Abstract

The teleparallel equivalent of general relativity is an intriguing alternative formulation of general relativity. In this thesis, we examine theories of teleparallel gravity in detail, and explore their relation to a whole spectrum of alternative gravitational models, discussing their position within the hierarchy of Metric Affine Gravity models. Consideration of alternative gravity models is motivated by a discussion of some of the problems of modern day cosmology, with a particular focus on the dark energy problem. Modifications of gravity in the teleparallel framework are examined as potential models to alleviate some of these issues and the relationships between various teleparallel and non-teleparallel modifed gravity models are analysed in depth. In particular f(T;B) gravity, where T is a torsion scalar and B is a derivative of a torsion vector, is introduced as a way of analysing both f(T) gravity and f(R) gravity, where R is the Ricci scalar, within the same unified framework. Various theoretical issues of all of these theories are discussed. In a similar way, teleparallel scalar-tensor models are analysed, taking into account coupling between torsion and a scalar field, with dynamical systems techniques utilised to analyse the cosmology of these models. An interesting conformal relationship is found to hold between teleparallel scalar-tensor models and f(T;B) gravity.

Abstract

This PhD thesis is devoted to the study of dynamical systems appearing in theoretical models of dark energy. The quest for understanding the origin of the observed cosmic acceleration has led physicists to advance a large number of phenomenological explanations based on different fundamental theories. The best approach to analyse the background cosmological impli- cations of all these models consists in employing dynamical systems tech- niques. In this thesis, after reviewing elements of dynamical systems theory and basic cosmology, several dynamical systems, which arise in dark energy models ranging from scalar fields to modified gravity, will be studied using both analytical and numerical methods. The work is organised in order to present as many details as possible for the simpler and well known models, while outlining major results and referring to the literature for the less stud- ied ones. This choice aims at providing the reader with a complete overview and summary of dynamical systems in dark energy applications.

Abstract

Excess salt intake decreases peripheral glucose uptake, thus impairing glucose tolerance. Stimulation of cellular glucose uptake involves phosphatidylinositide-3-kinase (PI-3K)-dependent activation of protein kinase B/Akt. A further kinase downstream of PI-3K is serum- and glucocorticoid-inducible kinase (SGK)1, which is upregulated by mineralocorticoids and, thus, downregulated by salt intake. To explore the role of SGK1 in salt-dependent glucose uptake, SGK1 knockout mice (sgk1(-/-)) and their wild-type littermates (sgk1(+/+)) were allowed free access to either tap water (control) or 1% saline (high salt). According to Western blotting, high salt decreased and deoxycorticosterone acetate (DOCA; 35 mg/kg body wt) increased SGK1 protein abundance in skeletal muscle and fat tissue of sgk1(+/+) mice. Intraperitoneal injection of glucose (3 g/kg body wt) into sgk1(+/+) mice transiently increased plasma glucose concentration approaching significantly higher values ([glucose]p,max) in high salt (281 +/- 39 mg/dl) than in control (164 +/- 23 mg/dl) animals. DOCA did not significantly modify [glucose]p,max in control sgk1(+/+) mice but significantly decreased [glucose]p,max in high-salt sgk1(+/+) mice, an effect reversed by spironolactone (50 mg/kg body wt). [Glucose]p,max was in sgk1(-/-) mice insensitive to high salt and significantly higher than in control sgk1(+/+) mice. Uptake of 2-deoxy-d-[1,2-(3)H]glucose into skeletal muscle and fat tissue was significantly smaller in sgk1(-/-) mice than in sgk1(+/+) mice and decreased by high salt in sgk1(+/+) mice. Transfection of HEK-293 cells with active (S422D)SGK1, but not inactive (K127N)SGK, stimulated phloretin-sensitive glucose uptake. In conclusion, high salt decreases SGK1-dependent cellular glucose uptake. SGK1 thus participates in the link between salt intake and glucose tolerance.

Abstract

Phosphatidylinositol 3-kinase (PI3 kinase) inhibition disrupts the ability of insulin to stimulate GLUT1 and GLUT4 translocation into the cell membrane and thus glucose transport. The effect on GLUT4 but not on GLUT1 is mediated by activation of protein kinase B (PKB). The serum- and glucocorticoid-inducible kinase SGK1, a further kinase downstream of PI3 kinase, regulates several transporters by enhancing their plasma membrane abundance. GLUT1 contains a consensus site ((95)Ser) for phosphorylation by SGK1. Thus, the present study investigated whether GLUT1 is regulated by the kinase. Tracer-flux studies in Xenopus oocytes and HEK-293 cells demonstrated that GLUT1 transport is enhanced by constitutively active (S422D)SGK1. The effect requires the kinase catalytical activity since the inactive mutant (K127N)SGK1 failed to modulate GLUT1. GLUT1 stimulation by (S422D)SGK1 is not due to de novo protein synthesis but rather to an increase of the transporter's abundance in the plasma membrane. Kinetic analysis revealed that SGK1 enhances maximal transport rate without altering GLUT1 substrate affinity. These observations suggest that SGK1 regulates GLUT1 and may contribute to or account for the PI3 kinase-dependent but PKB-independent stimulation of GLUT1 by insulin.

Abstract

Trypanosoma brucei, causative for African sleeping sickness, relies exclusively on glycolysis for ATP production. Under anaerobic conditions, glucose is converted to equimolar amounts of glycerol and pyruvate, which are both secreted from the parasite. As we have shown previously, glycerol transport in T. brucei occurs via specific membrane proteins (Wille, U., Schade, B., and Duszenko, M. (1998) Eur. J. Biochem. 256, 245-250). Here, we describe cloning and biochemical characterization of the three trypanosomal aquaglyceroporins (AQP; TbAQP1-3), which show a 40-45% identity to mammalian AQP3 and -9. AQPs belong to the major intrinsic protein family and represent channels for small non-ionic molecules. Both TbAQP1 and TbAQP3 contain two highly conserved NPA motifs within the pore-forming region, whereas TbAQP2 contains NSA and NPS motifs instead, which are only occasionally found in AQPs. For functional characterization, all three proteins were heterologously expressed in yeast and Xenopus oocytes. In the yeast fps1Delta mutant, TbAQPs suppressed hypoosmosensitivity and rendered cells to a hyper-osmosensitive phenotype, as expected for unregulated glycerol channels. Under iso- and hyperosmotic conditions, these cells constitutively released glycerol, consistent with a glycerol efflux function of TbAQP proteins. TbAQP expression in Xenopus oocytes increased permeability for water, glycerol and, interestingly, dihydroxyacetone. Except for urea, TbAQPs were virtually impermeable for other polyols; only TbAQP3 transported erythritol and ribitol. Thus, TbAQPs represent mainly water/glycerol/dihydroxyacetone channels involved in osmoregulation and glycerol metabolism in T. brucei. This function and especially the so far not investigated transport of dihydroxyacetone may be pivotal for the survival of the parasite survival under non-aerobic or osmotic stress conditions.

Abstract

The chloride channel ClC-Kb is expressed in the basolateral cell membrane of the distal nephron and participates in renal NaCl reabsorption. Loss-of-function mutations of ClC-Kb lead to classic Bartter syndrome, a rare salt-wasting disorder. Recently, we identified the ClC-Kb(T481S) polymorphism, which confers a strong gain-of-function effect on the ClC-Kb chloride channel. The present study has been performed to explore the prevalence of the mutation and its functional significance in renal salt handling and blood pressure regulation. As evident from electrophysiological analysis with the 2-electrode voltage-clamp technique, heterologous expression of ClC-Kb(T481S) in Xenopus oocytes gave rise to a current that was 7-fold larger than the current produced by wild-type ClC-Kb. The prevalence of the mutant allele was significantly higher in an African population from Ghana (22%) than in whites (12%). As tested in 1 white population, carriers of ClC-Kb(T481S) were associated with significantly higher systolic (by approximately 6.0 mm Hg) and diastolic (by approximately 4.2 mm Hg) blood pressures and significantly higher prevalence (45% versus 25%) of hypertensive (> or =140/90 mm Hg) blood pressure levels. Individuals carrying ClC-Kb(T481S) had significantly higher plasma Na+ concentrations and significantly decreased glomerular filtration rate. In conclusion, the mutation ClC-Kb(T481S) of the renal epithelial Cl- channel ClC-Kb strongly activates ClC-Kb chloride channel function in vitro and may predispose to the development of essential hypertension in vivo.

Abstract

Purpose: The serum- and glucocorticoid-inducible kinase SGK1 regulates several epithelial channels and transporters, the related protein kinase B (PKB) regulates glucose transport. SGK1 is expressed in the brain and could thus regulate glial and/or neuronal transport processes. The present study explores whether SGK1 is expressed in the retina and whether it regulates EAAT3, a Na(+)-coupled glutamate transporter. EAAT3 is expressed in retinal ganglion cells and accomplishes the clearance of glutamate from synaptic clefts., Methods: Immunohistochemistry was performed to test for retinal SGK1 expression. For functional analysis, cRNA encoding EAAT3 was injected into Xenopus oocytes with or without additional injection of wild-type SGK1, constitutively active (S422D)SGK1, inactive (K127N)SGK1, and/or constitutively active (T308D,S473D)PKB. Glutamate induced current (I(GLU)) was taken as a measure for transport., Results: SGK1 is indeed expressed in several retinal cells including retinal ganglion cells where it is colocalized with EAAT3. In EAAT3-expressing Xenopus oocytes, glutamate-induced current was stimulated by coexpression of wild-type SGK1, constitutively active (S422D)SGK1, and constitutively active (T308D,S473D)PKB, but not by inactive (K127N)SGK1., Conclusions: SGK1 and EAAT3 are coexpressed in retinal neurons, and SGK1 serves to stimulate EAAT3. This function is shared by protein kinase B (PKB). The experiments reveal a novel mechanism regulating EAAT3, which may be essential for the function of the retinal ganglion cells.

Abstract

Objective: Serum- and glucocorticoid-inducible kinase 1 (SGK1) inhibits the ubiquitin ligase neuronal cell expressed developmentally downregulated 4-2 (Nedd4-2), which retards the retrieval of the epithelial Na+ channel ENaC. Accordingly, SGK1 enhances ENaC abundance in the cell membrane. The significance of this effect is shown by an association of an E8CC/CT;I6CC polymorphism in the SGK1 gene with increased blood pressure. However, strong expression of SGK1 in enterocytes not expressing ENaC points to further functions of SGK1. This study was performed to test for regulation of Na+-coupled glucose transporter 1 (SGLT1) by Nedd4-2, SGK1, and/or the related kinases SGK3 and PKB. Additional studies searched for an association of the SGK1 gene with BMI., Research Methods and Procedures: mRNA encoding SGLT1, wild-type Nedd4-2, inactive (C938S)Nedd4-2, wild type SGK1, constitutively active (S422D)SGK1 or inactive (K127N)SGK1, wild-type SGK3, and constitutively active (T308DS473D)PKB or inactive (T308AS473A)PKB were injected into Xenopus oocytes, and glucose transport was quantified from glucose-induced current (I(glc)). BMI was determined in individuals with or without the E8CC/CT;I6CC polymorphism., Results: I(glc) was significantly decreased by coexpression of Nedd4-2 but not of (C938S)Nedd4-2. Abstract

Background: ClC anion channels are ubiquitous and have been identified in organisms as diverse as bacteria and humans. Despite their widespread expression and likely physiological importance, the function and regulation of most ClCs are obscure. The nematode Caenorhabditis elegans offers significant experimental advantages for defining ClC biology. These advantages include a fully sequenced genome, cellular and molecular manipulability, and genetic tractability., Results: We show by patch clamp electrophysiology that C. elegans oocytes express a hyperpolarization- and swelling-activated Cl(-) current with biophysical characteristics strongly resembling those of mammalian ClC-2. Double-stranded RNA-mediated gene interference (RNAi) and single-oocyte RT-PCR demonstrated that the channel is encoded by clh-3, one of six C. elegans ClC genes. CLH-3 is inactive in immature oocytes but can be triggered by cell swelling. However, CLH-3 plays no apparent role in oocyte volume homeostasis. The physiological signal for channel activation is the induction of oocyte meiotic maturation. During meiotic maturation, the contractile activity of gonadal sheath cells, which surround oocytes and are coupled to them via gap junctions, increases dramatically. These ovulatory sheath cell contractions are initiated prematurely in animals in which CLH-3 expression is disrupted by RNAi., Conclusions: The inwardly rectifying Cl(-) current in C. elegans oocytes is due to the activity of a ClC channel encoded by clh-3. Functional and structural similarities suggest that CLH-3 and mammalian ClC-2 are orthologs. CLH-3 is activated during oocyte meiotic maturation and functions in part to modulate ovulatory contractions of gap junction-coupled gonadal sheath cells.

Abstract

1. We report the molecular identification of a Na+-Pi (inorganic phosphate) cotransport system of the NaPi-II protein family from zebrafish intestine. Following a PCR-related strategy, a DNA fragment from intestine-derived RNA was isolated. Rapid amplification of cDNA ends (3'- and 5'-RACE) resulted in the complete sequence (2607 bp) containing an open reading frame of 1893 bp. 2. The NaPi-II-related protein was expressed in Xenopus laevis oocytes and the resulting transport activity was analysed by electrophysiological means. The apparent Km for Pi was 250 microM (96 mM Na+, -60 mV), and voltage-dependent binding of Na+ exhibited a Km of 67.1 mM (1 mM Pi, -60 mV). 3. Interestingly, the overall transport activity was almost insensitive to changes in the holding potential. The apparent affinity for Na+ decreased under hyperpolarizing conditions, whereas Pi binding showed no voltage dependence. Transport activity was inhibited at low pH, which is characteristic for renal NaPi-II isoforms. 4. The expression of the NaPi-II-related isoform was addressed by reverse-transcription PCR. The mRNA could be detected in intestine, liver, eye and kidney. Unexpectedly, a second NaPi-II-related isoform was identified and found to be expressed in kidney, intestine, liver, brain, eye and prominently in testis. In addition, a shorter amplicon was demonstrated to be an antisense transcript related to the NaPi-II intestinal isoform.
