OBJECTIVE To establish pilot data on the plasma concentrations of SC amikacin at 2 doses in red-eared sliders and evaluate concurrent plasma biochemistry parameters. ANIMALS 8 adult red-eared sliders (Trachemys scripta elegans). METHODS Amikacin was administered SC at target doses of 5 and 10 mg/kg with a 3-week washout period. Blood samples were collected at 0, 24, 48, 72, and 96 hours postadministration. Plasma amikacin concentrations were quantified using liquid chromatography tandem mass spectrometry. Plasma biochemistry analyses were performed before amikacin administration, 1 week post 5-mg/kg administration, and 1 week post 10-mg/kg administration. RESULTS Mean maximum amikacin plasma concentrations were recorded 24 hours after 5-mg/kg and 10-mg/kg dosing and were 17.5 ± 2.32 µg/mL and 23.6 ± 2.92 µg/mL, respectively. Mean plasma concentrations after 5-mg/kg dosing steadily decreased to 9.1 ± 0.92 µg/mL by 96 hours postadministration. Amikacin remained detectable in all plasma samples 3 weeks post 5-mg/kg dosing with a mean plasma concentration of 1.04 ± 0.22 µg/mL. Mean plasma concentrations after 10-mg/kg dosing did not decrease over the 96-hour study period. There were no clinically relevant changes in biochemistry parameters. CLINICAL RELEVANCE Amikacin persists at detectable plasma levels for at least 3 weeks after SC administration of a 5-mg/kg dose in redeared sliders, which has not previously been reported in any species. No biochemistry changes consistent with renal toxicity occurred after either dose. OBJECTIVE To improve the current recommendations for the diagnosis of canine heartworm (Dirofilaria immitis) disease. ANIMALS Blood samples collected from 35 shelter dogs in the Republic of Korea. METHODS Samples were tested for the presence of microfilaria using the modified Knott (MK) test and D immitis DNA using species-specific loop-mediated isothermal amplification (LAMP) PCR. The blood samples were additionally assessed for the presence of heartworm antigens using the Antigen Rapid Canine Heartworm AG Test Kit 2.0 (Bionote Co). The performance of the MK test and LAMP PCR was assessed through statistical analysis, with a paired McNemar test utilized for comparison. RESULTS The heartworm antigen was detected in 28.5% of the subjects. Of the 10 positive animals, the MK test detected microfilaria in 4 of 35 (11.4%) animals, and LAMP PCR detected D immitis DNA in 6 of 35 (17.1%). The results of this study indicate that the LAMP PCR showed more positive results in samples compared to the conventional MK test. CLINICAL RELEVANCE The D immitis–specific LAMP PCR assay has the potential to function as an alternative to current detection methods. The incidence and prevalence of breast cancer in Iraq are alarming. Breast cancer is one of the most common cancers among Iraqi women, and its rates have been steadily increasing over the years. The exact reasons for the high incidence are not yet fully understood, but it is believed to be influenced by a combination of genetic, environmental, and lifestyle factors. The research objectives of this study revolve around two main goals. Firstly, the study aims to establish baseline values for the amount of uranium present in blood samples. Secondly, the study aims to assess the potential relationship between uranium levels in blood and the development of cancer. The investigation includes 16 blood samples from women diagnosed with breast cancer and 20 blood samples from women without breast cancer. The nuclear fission track analysis method using CR-39 solid-state nuclear track detectors will be employed to analyze the uranium contents in women's cancer blood (CB) samples. The methodology adopted for this study involved utilizing the SPSS program to conduct a comprehensive statistical analysis. The results of the study indicate that there is a variation in uranium concentration among both the patient women and healthy women. The uranium concentration among patient women ranged from 3.259 ppb to 1.918 ppb, while among healthy women, it varied from 2.105 ppb to 0.59 ppb. The aim of the study was to develop a molecular diagnosis protocol of monocytic ehrlichiosis and granulocytic anaplasmosis in dogs by conventional polymerase chain reaction (PCR) and to compare the results from PCR and a rapid serological test. One hundred and six blood samples from dogs were tested by the rapid serological test SNAP 4Dx Plus (IDEXX Laboratories, Westbrook, ME) and by conventional PCR. Sixty-two of them (58.49%) were positive for antibodies to Ehrlichia canis/Ehrlichia ewingii and 14 (13.21%) for antibodies to Anaplasma phagocytophilum/Anaplasma platys. In 24 (22.64%) blood samples, antibodies against both pathogens were simultaneously detected. Six (5.66%) samples were seronegative. Fourty-six of the 106 tested samples (43.4%) were positive for a 345 bp segment of the ribosomal gene of family Anaplasmataceae. In 28 of them the presence of a 444 bp fragment of the ankA gene of A. phagocytophilum was detected, and in 26: a 409 bp fragment of the gene of E. canis. Nine samples were simultaneously positive for genetic sequences of E. canis and A. phagocytophilum. The target DNA fragments specific for the two studied pathogens were not detected in one of the Anaplasmataceae-positive samples. In the remaining 60 cases (56.6%), the presence of a 345 bp segment of the ribosomal gene was not detected. Montmorency cherry (MC) can improve endurance performance, but optimal pre‐exercise timing of supplementation and influence of training status on efficacy are unknown. We investigated the effect of MC concentrate ingestion between 30‐ and 150‐min pre‐exercise in trained and recreational cyclists on 15‐km time trial (TT) performance and exercise economy. Twenty participants (10 recreationally active, RA; 10 trained, T) completed 10 min of steady‐state exercise (SSE) at 40%Δ (SSE) and a TT on four separate occasions following an unsupplemented (US), 30‐, 90‐ or 150‐min pre‐exercise Montmorency cherry concentrate (MCC) supplementation conditions (MCC30/90/150min). Venous and capillary blood samples were taken at regular intervals pre‐ and post‐SSE and TT. MCC significantly improved TT performance, but not exercise economy. The greatest improvement in performance occurred following MCC90min compared to US (US 1603.1 ± 248 s vs. MCC90min 1554.8 ± 226.7 s, 2.83% performance improvement). Performance was significantly enhanced for trained (US 1496.6 ± 173.1 s vs. MCC90min 1466.8 ± 157.6 s) but not recreationally active participants. Capillary [lactate] and heart rate were significantly greater during the TT for the 90‐min dose timing (p < 0.05). In the MCC30min and MCC90min conditions, plasma ferulic (US 8.71 ± 3.22 nmol. L−1 vs. MCC30min 15.80 ± 8.69 nmol. L−1, MCC90min 12.65 ± 4.84 nmol. L−1) and vanillic acid (US 25.14 ± 10.91 nmol.L−1 vs. MCC30min 153.07 ± 85.91 nmol. L−1, MCC90min 164.58 ± 59.06 nmol. L−1) were significantly higher pre‐exercise than in US and MCC150min conditions (p < 0.05). There was no significant change in muscle oxygenation status or plasma nitrite/nitrate concentration. MCC supplementation enhanced endurance exercise performance optimally when consumed ∼90 min pre‐exercise producing maximal plasma phenolic metabolites during exercise. Bovine leukemia virus (BLV) is the etiological agent of enzootic bovine leukosis and causes a persistent infection that can leave cattle with no symptoms. Many countries have been able to successfully eradicate BLV through improved detection and management methods. However, with the increasing novel molecular detection methods there have been few efforts to standardize these results at global scale. This study aimed to determine the interlaboratory accuracy and agreement of 11 molecular tests in detecting BLV. Each qPCR/ddPCR method varied by target gene, primer design, DNA input and chemistries. DNA samples were extracted from blood of BLV-seropositive cattle and lyophilized to grant a better preservation during shipping to all participants around the globe. Twenty nine out of 44 samples were correctly identified by the 11 labs and all methods exhibited a diagnostic sensitivity between 74 and 100%. Agreement amongst different assays was linked to BLV copy numbers present in samples and the characteristics of each assay (i.e., BLV target sequence). Finally, the mean correlation value for all assays was within the range of strong correlation. This study highlights the importance of continuous need for standardization and harmonization amongst assays and the different participants. The results underscore the need of an international calibrator to estimate the efficiency (standard curve) of the different assays and improve quantitation accuracy. Additionally, this will inform future participants about the variability associated with emerging chemistries, methods, and technologies used to study BLV. The skin is an anatomical reservoir for African trypanosomes, yet the prevalence of extravascular parasite carriage in the population at risk of gambiense Human African Trypanosomiasis (gHAT) remains unclear. Here, we conducted a prospective observational cohort study in the HAT foci of Forecariah and Boffa, Republic of Guinea. Of the 18,916 subjects serologically screened for gHAT, 96 were enrolled into our study. At enrolment and follow-up visits, participants underwent a dermatological examination and had blood samples and superficial skin snip biopsies taken for examination by molecular and immuno-histological methods. In seropositive individuals, dermatological symptoms were significantly more frequent as compared to seronegative controls. Trypanosoma brucei DNA was detected in the blood of 67% of confirmed cases (22/33) and 9% of unconfirmed seropositive individuals (3/32). However, parasites were detected in the extravascular dermis of up to 71% of confirmed cases (25/35) and 41% of unconfirmed seropositive individuals (13/32) by PCR and/or immuno-histochemistry. Six to twelve months after treatment, trypanosome detection in the skin dropped to 17% of confirmed cases (5/30), whereas up to 25% of unconfirmed, hence untreated, seropositive individuals (4/16) were still found positive. Dermal trypanosomes were observed in subjects from both transmission foci, however, the occurrence of pruritus and the PCR positivity rates were significantly higher in unconfirmed seropositive individuals in Forecariah. The lower sensitivity of superficial skin snip biopsies appeared critical for detecting trypanosomes in the basal dermis. Reproductive strategies of hibernating insectivorous bats and reproduction phenology parameters can vary considerably as they will be influenced by ecological seasonality. While the exact timing of greater mouseeared bat (Myotis myotis) reproduction is described through morphological assessment of dissected embryos, most recent studies have focused only on population or behavioural aspects of reproduction. In our study, non-invasive ultrasound examination and non-lethal blood sampling were used to assess basic reproduction parameters of M. myotis from two nursery colonies over two gestation periods with different weather conditions. We found that ovulation probably occurred before female emergence from the hibernacula between late March and mid-April, triggered by a short period of high ambient temperature lasting at least three days. On the other hand, the estimated date of blastocyst implantation only varied within four days despite weather conditions differing significantly over the two years. Though specific foetal growth velocity did not differ between localities, the date of parturition was influenced by shelter temperature in May (difference between localities) and average ambient temperature in April (difference between years). The progesterone level does not appear to be a suitable indicator of pregnancy stage as it changed significantly during pregnancy and did not correlate with foetus size. Impedance aggregometry is an alternative to light transmission aggregometry that allows analysis of platelet function in whole blood samples. We hypothesized (1) impedance aggregometry would produce repeatable results, (2) inhibition of cyclooxygenase with aspirin would attenuate aggregation responses to collagen and abolish the aggregation response to arachidonic acid (AA), and (3) thromboxane receptor antagonism (terutroban) would attenuate the aggregation response to AA. Venous blood was obtained from 11 participants three times separated by at least 2 weeks. One sample followed 7‐day‐aspirin intervention (81 mg once daily; ASA), the others no intervention (control). Aggregation was induced using 1 μg/mL collagen ([col 1]), 5 μg/mL collagen ([col 5]), and 50 mM AA via impedance aggregometry to determine total aggregation (AUC) analyzed for intra‐test repeatability, inter‐test repeatability, intervention (ASA or control), and incubation (saline or terutroban). [col 1] showed high intra‐test (p ≤ 0.03 visit 1 and 2) and inter‐test repeatability (p < 0.01). [col 5] and AA showed intra‐ ([col 5] p < 0.01 visit 1 and 2; AA p < 0.001 visit 1 and 2) but not inter‐test repeatability ([col 5] p = 0.48; AA p = 0.06). ASA attenuated AUC responses to [col 1] (p < 0.01), [col 5] (p = 0.03), and AA (p < 0.01). Terutroban attenuated AUC in response to AA (p < 0.01). [col 1] shows sufficient repeatability for longitudinal investigations of platelet function. Background: Malaria continues to be an important threat to public health and infects millions of children under 5 years of age each year. Although Ethiopia has set targets for at-risk group interventions to eradicate and manage malaria, the illness is still a serious public health problem in areas where it is endemic, especially in the unique lowlands in the Borena zone. Objective: This study aimed to determine the prevalence of malaria and associated factors among children in Borena's pastoral communities, Oromia Regional State, southern Ethiopia, in 2022. Methods: A community-based cross-sectional study was conducted from 1 March to 30 April 2022 among 437 randomly selected households with children under 5 years of age in pastoral communities in the Borena zone. Data were collected through face-to-face interviews with structured and pretested questionnaires and blood sample examination using microscopy. Thick and thin blood smears were prepared and examined under a microscope at a health center to confirm malaria cases. The data were analyzed using SPSS version 25. Bivariate and multivariable logistic regression analyses were used to identify factors associated with malaria, and a p-value <0.05 was used to declare statistical significance. Result: The prevalence of malaria among children under 5 years of age was 27.8% (95% CI = 23.5-32.1), and the prevalence rates of Plasmodium falciparum, Plasmodium vivax, and mixed malaria were 68.4%, 25.6%, and 6%, respectively. Regarding the proportion of malaria among age groups, 81% of children under 5 years of age between 48 and 59 months were malaria-positive. In this study, fever within the last week (AOR = 13.34, 95% CI = 6.37-27.95) and not sleeping under insecticide-treated nets (ITNs) (AOR = 3.10, 95% CI =1.95-4.92) were significantly associated with malaria. The age of the children was negatively associated with malaria prevalence. Conclusion: The prevalence of malaria among children under 5 years old was high during the rainy season in this pastoral region of Ethiopia. Factors such as fever within the last week and not sleeping in insecticide-treated nets were significantly associated with malaria. Therefore, to reduce malaria-related infections and deaths among children under 5 years of age, the government ought to enhance the availability and utilization of insecticide-treated nets (ITNs). This study aimed to determine the sequence type (ST) of Bartonella henselae infecting small Indian mongooses from Saint Kitts via multi-locus sequence typing (MLST). This investigation used stored EDTA blood (n = 22) samples from mongooses previously identified as positive for B. henselae. Chocolate agar plates were enriched with Bartonella alpha-Proteobacteria growth medium (BAPGM) to culture and isolate Bartonella from the blood samples. To perform MLST, DNA was extracted and purified from isolates followed by amplification by conventional PCR (300–500 bp) for eight genes (16S rDNA, batR, gltA, groEL, ftsZ, nlpD, ribC, and rpoB). Bartonella henselae STs were deposited in the PubMLST repository. Out of 22 B. henselae-positive blood samples, isolates were obtained from 12 mongooses (54.5%; 12/22). Each mongoose was infected with one ST. The studied mongoose population was infected with sequence types ST2, ST3, ST8, and a novel ST represented by ST38. Bartonella henselae ST2, ST3 and ST8 infecting mongooses are known to circulate in humans and cats, with ST2 and ST8 associated with Cat Scratch Disease (bartonellosis) in humans. In this study, the effects of exercise applied to sedentary individuals and the use of pomegranate–black carrot juice on minerals, fatty acids and some biochemical parameters were examined. Twenty healthy sedentary men participated in this study. This research consisted of three stages. Blood samples were taken from the participants before this study (Baseline), after the participants exercised (60 min/day) for 10 days (Exerciseonly), after the participants were given pomegranate–black carrot juice mixture (100 mL/100 mL) along with exercise (60 min/day) for 10 days (Exercise+supp). While AST and ALT levels increased in the Exerciseonly phase, they showed a relative decrease in the Exercise+supp phase. It was determined that Mg level in the Exerciseonly phase decreased compared to the Baseline and the Exercise+supp phase. It was determined that the Zn level in the Exercise+supp phase increased compared to the Zn level in the Exerciseonly phase. It was determined that 6:0, 12:0 and 14:0 fatty acid levels increased in the Exerciseonly phase compared to the Baseline. A decrease was detected in the Exerciseonly phase compared to the Baseline 18:2n6c, 18:3n6 and 18:3n3 fatty acid levels. It can be said that exercise and the use of pomegranate–black carrot juice mixture in sedentary individuals have supportive and corrective effects on serum mineral, fatty acids and some biochemical parameters. Heat shock stress induces genome-wide changes in transcription regulation, activating a coordinated cellular response to enable survival. We noticed many heat shock genes are up-regulated in blood samples from individuals with trisomy 21. We characterized the immediate transcriptional response to heat shock of two lymphoblastoid cell lines derived from brothers with and without trisomy 21. Bombali virus (BOMV) is a novel Orthoebolavirus that has been detected in free-tailed bats in Sierra Leone, Guinea, Kenya, and Mozambique. We screened our collection of 349 free-tailed bat lungs collected in Côte d'Ivoire and Tanzania for BOMV RNA and tested 228 bat blood samples for BOMV antibodies. We did not detect BOMV-specific antibodies but found BOMV RNA in a Mops condylurus bat from Tanzania, marking the first detection of an ebolavirus in this country. Inflammation appears to be a critical mechanism in the development of alcohol use disorder (AUD) and a consequence of chronic alcohol use. The potential anti‐inflammatory properties of cannabis may modulate the proinflammatory effects of alcohol. This study sought to extend previous work investigating the relationship between alcohol consumption, cannabis use and circulating interleukin (IL)‐6 levels in a sample with AUD. One hundred and thirty‐three individuals with an AUD provided blood samples to assess IL‐6 and answered questions regarding alcohol and cannabis use. An ordinary least squares multiple regression analysis was conducted to assess the effect of alcohol and cannabis use on IL‐6. A moderation analysis examined cannabis use as a potential moderator of the relationship between alcohol use and circulating IL‐6 levels. Alcohol use was predictive of higher log IL‐6 levels (standardized β = 0.16, p = 0.03), while cannabis use was not predictive of log IL‐6 levels (p = 0.36). Days of cannabis use moderated the relationship between alcohol use and IL‐6 levels, such that the relationship between alcohol use and IL‐6 levels was only significant in individuals with AUD without recent cannabis use. This study extends previous work to a clinical sample with an AUD and underscores the importance of considering cannabis use in studies on alcohol use and inflammation. This study aims to examine the possibility and impact of utilizing data science on blood samples to rapidly and proactively identify underlying health issues. By utilizing effective algorithms, models will be constructed to address these problems and determine potential healthcare options based on geographical location. Once data is gathered, health officials will be notified of major diseases and individuals at risk or already affected. Authentic blood samples are used to ensure the credibility and validity of the proposed system. The data was collected during a volunteer-led hemoglobin blood test camp specifically for women residing in impoverished areas, resulting in a total of 551 samples. The effectiveness of this technique has been assessed through experimental results based on Hb, RDW%, MCV, RBC, and M-Index. The proposed data analysis and deep learning algorithm achieved average values of haemoglobin count 11.67 g/dL with a 1.33 standard deviation, RDW 14.59%, MCV 81.45, RBC 4.37 per microliter with a variance of 0.5, and M-Index 19.56. The experimental results achieved 97.60% accuracy, demonstrating the effectiveness of the proposed technique for classifying anemia and its subtypes. Malaria is a significant health problem in Africa, primarily due to the Plasmodium falciparum species, but this is not the only etiological factor responsible for malaria on the continent. The goal of the present research was to describe asymptomatic malaria cases and to identify Plasmodium species responsible for malaria in the BaAka Pygmies, inhabitants of the Central African Republic (CAR). Screening was realised in the period of August–September 2021 among 308 people, including 74 children and 234 adults reporting to a healthcare facility in Monasao (southwest CAR), an area inhabited by a semi-nomadic tribe of BaAka Pygmies. The study consisted of two phases. Phase I, which was conducted in Africa, consisted of performing malaria rapid diagnostic tests (mRDTs), taking haemoglobin measurements and collecting blood samples onto Whatman FTA cards for molecular diagnostics. Phase II, which was conducted in Poland, involved molecular tests (RT-PCR) to confirm or rule out malaria infections and to identify Plasmodium species responsible for the infections. mRDTs detected Plasmodium infections in 50.3% of children and 17.1% of adults participating in the study, whereas RT-PCR assays yielded positive results for 59.5% children and 28.6% adults. Molecular tests detected multiple Plasmodium falciparum infections but also three infections with P. malariae, three with P. ovale and one with P. vivax. The obtained results have confirmed numerous asymptomatic Plasmodium infections among the BaAka Pygmies. The rates of asymptomatic malaria cases in adults were twice as high as those in children, which may be indicative of the gradual acquisition of protective immunity with age. Metabolomics plays a crucial role in understanding metabolic processes within biological systems. Using specific pulse sequences, NMR-based metabolomics detects small and macromolecular metabolites that are altered in blood samples. Here we proposed a method called spectral editing neural network, which can effectively edit and separate the spectral signals of small and macromolecules in 1H NMR spectra of serum and plasma based on the linewidth of the peaks. We applied the model to process the 1H NMR spectra of plasma and serum. The extracted small and macromolecular spectra were then compared with experimentally obtained relaxation-edited and diffusion-edited spectra. Correlation analysis demonstrated the quantitative capability of the model in the extracted small molecule signals from 1H NMR spectra. The principal component analysis showed that the spectra extracted by the model and those obtained by NMR spectral editing methods reveal similar group information, demonstrating the effectiveness of the model in signal extraction. 1H NMR-based metabolomics can detect small and macromolecular metabolites simultaneously from complex biological samples, however, signaling overlap remains a challenge for accurate molecular identification and quantification. Aims Methods Results Conclusion Intraoperative hypotension is a risk factor for kidney, heart and cognitive postoperative complications. Literature suggests that the use of low‐dose peripheral norepinephrine (NOR) reduces organ dysfunction, yet its administration remains unstandardized. In this work we develop a pharmacokinetic (PK)/pharmacodynamic (PD) model of NOR and its effect on mean arterial pressure (MAP).From June 2018 to December 2021, we included patients scheduled for elective neurosurgery and requiring vasopressors for intraoperative hypotension management at Lariboisière Hospital, Paris. Low doses of NOR were administered peripherally, and successive arterial blood samples were collected to track its plasmatic concentration. We used a compartmental modelling approach for NOR PK. We developed and compared 2 models for NOR PD on MAP. Model comparison was done using Bayes information criteria. The resulting PK/PD model parameters were fitted over the entire population and linked to age, weight, height and sex.We included 29 patients (age 52 [46–64] years, 69% female). NOR median time to peak effect on MAP was 74 [53–94] s. After bolus administration, MAP increased by 24% (15–31%). A 2‐comparment model with depot best captured NOR PK. NOR PD effect on MAP was well represented by both Emax and Windkessel models, with better results for the former. We found that age, height and weight as well as history of smoking and hypertension were correlated with model parameters.We have developed a PK/PD model to accurately track norepinephrine plasma concentration and its effect on MAP over time, which could serve for target‐controlled infusion. Background: This study aims to analyze the pathogenic gene in a Chinese family with non-syndromic hearing loss and identify a novel mutation site in the TNC gene. Methods: A five-generation Chinese family from Anhui Province, presenting with autosomal dominant non-syndromic hearing loss, was recruited for this study. By analyzing the family history, conducting clinical examinations, and performing genetic analysis, we have thoroughly investigated potential pathogenic factors in this family. The peripheral blood samples were obtained from 20 family members, and the pathogenic genes were identified through whole exome sequencing. Subsequently, the mutation of gene locus was confirmed using Sanger sequencing. The conservation of TNC mutation sites was assessed using Clustal Omega software. We utilized functional prediction software including dbscSNV_AdaBoost, dbscSNV_RandomForest, NNSplice, NetGene2, and Mutation Taster to accurately predict the pathogenicity of these mutations. Furthermore, exon deletions were validated through RT-PCR analysis. Results: The family exhibited autosomal dominant, progressive, post-lingual, non-syndromic hearing loss. A novel synonymous variant (c.5247A > T, p.Gly1749Gly) in TNC was identified in affected members. This variant is situated at the exon–intron junction boundary towards the end of exon 18. Notably, glycine residue at position 1749 is highly conserved across various species. Bioinformatics analysis indicates that this synonymous mutation leads to the disruption of the 5' end donor splicing site in the 18th intron of the TNC gene. Meanwhile, verification experiments have demonstrated that this synonymous mutation disrupts the splicing process of exon 18, leading to complete exon 18 skipping and direct splicing between exons 17 and 19. Conclusion: This novel splice-altering variant (c.5247A > T, p.Gly1749Gly) in exon 18 of the TNC gene disrupts normal gene splicing and causes hearing loss among HBD families. [ABSTRACT FROM AUTHOR]

Simple Summary: Miranda's donkey is an autochthonous Portuguese breed that is important for the culture and economy of the country's northern region. Knowledge of the physiological characteristics of the breed is crucial for its preservation, and even if studies have been conducted, none have focused on its coagulation profile. The aim of this study was to establish reference intervals for coagulation and assess the influence of sex and age. For this, seven parameters involved in coagulation were analyzed in blood samples from 75 clinically healthy animals. Statistical differences were only found between ages in two parameters, platelet count and plateletcrit, both of which were higher in young animals. The reference intervals described here can help monitor health and guide the diagnosis and treatment of sick Miranda's donkey, contributing to its preservation. Miranda's donkey, originating in northern Portugal, is an autochthonous breed that is deeply intertwined with the region's culture and economy. No significant differences were found between sexes (p > 0.05), but statistically significant differences were found between ages (p < 0.05) for platelet count and plateletcrit (both higher in young animals). The RIs described here can help monitor health and guide the diagnosis and treatment of diseased Miranda's donkeys, contributing to their preservation. Thyroid cancer (TC) represents a significant health burden globally, with follicular thyroid cancer (FTC) posing diagnostic challenges despite advancements. This pilot study aimed to evaluate the utility of a liquid biopsy with cell-free DNA (cfDNA) in patients with FTC. Blood samples were collected from 13 patients diagnosed with FTC, DNA extraction was performed, and cfDNA was analyzed using the Illumina's TruSight Oncology 500 High-Throughput panel. The results revealed low tumor mutational burden and minimal pathogenic variants in cfDNA, indicating challenges such as low DNA yield and poor material quality despite adequate coverage. Background: Arterial blood gas analysis (ABGA) plays a vital role in emergency and intensive care, which is affected by many factors, such as different instrumentation, temperature, and testing time. However, there are still no relevant reports on the difference in discarding different blood volumes on ABGA values. Methods: We enrolled 54 patients who underwent thoracoscopic surgery and analysed differences in blood gas analysis results when different blood volumes were discarded from the front line of the arterial heparin blood collector. A paired t test was used to compare the results of the same patient with different volumes of blood discarded from the samples. The difference was corrected by Bonferroni correction. Results: Our results demonstrated that the PaO2, PaCO2, and THbc were more stable in the 4th ml (PaO2 = 231.3600 ± 68.4878 mmHg, PaCO2 = 41.9232 ± 7.4490 mmHg) and 5th ml (PaO2 = 223.7600 ± 12.9895 mmHg, PaCO2 = 42.5679 ± 7.6410 mmHg) blood sample than in the 3rd ml (PaO2 = 234.1000 ± 99.7570 mmHg, PaCO2 = 40.6179 ± 7.2040 mmHg). Raising small ruminants is the main source of income for farmers in Pakistan especially in rural areas of Dera Ghazi Khan in Punjab. Despite having large sheep population, the prevalence of intra-erythrocytic protozoa, Theileria (T.) lestoquardi, has never been reported from this area. This study was conducted to fill this knowledge gap and 333 blood samples of apparently healthy small ruminants (168 sheep and 165 goats) along with their epidemiological data were collected from Dera Ghazi Khan district during August till November 2022. The polymerase chain reaction (PCR) analysis amplified a 785 base pair amplicon specific for the Merozoite surface antigen (ms 1–2) gene of T. lestoquardi in 2 out of the 168 (3.3%) sheep blood samples, while no goat blood sample out of 165 was found to be infected with T. lestoquardi. DNA sequencing confirmed the presence of Theileria lestoquardi in both samples and phylogenetic analysis revealed that these amplicon resembled the partial ms 1–2 gene sequences detected in small ruminants from Pakistan, India Iran and Egypt. All the studied epidemiological factors (age, sex, breed, size of herd, dogs with herd, composition of herd, size of herd and Tick burden on sheep) were not found associated with the prevalence of T. lestoquardi. In conclusion, this study reports a low prevalence of T. lestoquardi infection in the Dera Ghazi Khan District of Punjab, Pakistan. Objective: People regularly contact emergency medicine services concerned that they have been exposed to drink spiking, i.e., exposure to drugs without their knowledge or permission. We identified drugs in blood and urine samples from patients suspecting exposure to drink spiking, with special consideration for drugs not reported taken by the patient (unreported drugs). Methods: From September 2018 to May 2019, we collected blood and urine samples from patients 16 years or older presenting at an emergency clinic in Oslo, Norway, within 48 hours of suspected exposure to drink spiking. We also collected information on ethanol ingestion and drugs taken. Blood samples were analyzed for 20 classical recreational drugs using ultra-high performance liquid chromatography–tandem mass spectrometry (UHPLC-MS/MS) and an automated enzymatic method for ethanol. Urine samples were analyzed using immunoassay methods and a specific gas chromatography mass spectrometry (GCMS) method for gammahydroxybutyrate (GHB). Results: From 100 included patients (median age 24 years, 62 females), we collected 100 blood samples and 72 urine samples. Median time since exposure was 5 hours. Unreported drugs were found in 15 patients. Unreported drugs in the blood samples were clonazepam in 3, methylenedioxymethamphetamine (MDMA) in 3, amphetamine in 2, tetrahydrocannabinol (THC) in 2, tramadol in 1, cocaine in 1, and methamphetamine in 1. Unreported drugs in the urine samples were cocaine in 5, amphetamine in 4, ecstasy in 3, and cannabis in 2. Ethanol was found in 69 patients, all reporting ethanol ingestion. Median blood ethanol concentration was higher in patients with no unreported drugs detected, 1.00‰ (interquartile range (IQR) 0–1.52) vs. 0‰ (IQR 0–0.46) (p<0.001). GHB was not detected. Conclusion: Unreported drugs, possibly used for drink spiking, were found in 15% of patients. Blood ethanol concentration was higher when no unreported drugs were found. Flow cytometry of blood samples is a very valuable clinical and research tool to monitor the immune response in human patients. Furthermore, it has been successfully applied in cats, such as for infections with feline immune deficiency virus (FIV). However, if cells are not isolated and frozen, analysis of anticoagulated blood samples requires mostly prompt processing following blood collection, making later analysis of stored full blood samples obtained in clinical studies often impossible. The SMART Tube system (SMART TUBE Inc., California, United States; SMT) allows fixation and long-term preservation of whole blood samples at −80°C. However, this system has so far only been applied to human biological samples. In the present study, a new flow cytometry SMART Tube protocol adapted for feline whole blood samples was successfully established allowing quantification of T-helper cells, cytotoxic T-cells, B-cells, monocytes, and neutrophils up to 2  years post sampling. Results obtained from frozen stabilized and fresh blood samples were compared for validation purposes and correlated to differential blood counts from a conventional hematology analyzer. Clinical applicability of the new technique was verified by using samples from a treatment study for feline infectious peritonitis (FIP). Using the new SMT protocol on retained samples, it could be demonstrated that long-term storage of these SMT tubes is also possible. In summary, the newly adapted SMT protocol proved suitable for performing flow cytometry analysis on stored feline whole blood samples, thus opening up new avenues for veterinary research on a variety of aspects of clinical interest. Le Dantec virus (LDV), assigned to the species Ledantevirus ledantec, genus Ledantevirus, family Rhabdoviridae has been associated with human disease but has gone undetected since the 1970s. We describe the detection of LDV in a human case of undifferentiated fever in Uganda by metagenomic sequencing and demonstrate a serological response using ELISA and pseudotype neutralisation. By screening 997 individuals sampled in 2016, we show frequent exposure to ledanteviruses with 76% of individuals seropositive in Western Uganda, but lower seroprevalence in other areas. Serological cross-reactivity as measured by pseudotype-based neutralisation was confined to ledanteviruses, indicating population seropositivity may represent either exposure to LDV or related ledanteviruses. We also describe the discovery of a closely related ledantevirus in blood from the synanthropic rodent Mastomys erythroleucus. Ledantevirus infection is common in Uganda but is geographically heterogenous. Further surveys of patients presenting with acute fever are required to determine the contribution of these emerging viruses to febrile illness in Uganda. Author summary: Understanding the viruses capable of human infection is important for outbreak prevention and early intervention in viral epidemics. Le Dantec virus (LDV) is a member of the viral genus Ledantevirus (Rhabodoviridae) and has previously been isolated only once previously in a child with febrile illness in Senegal in 1965. We detected the genome of LDV in blood sampled from a patient presenting with febrile illness in Western Uganda in 2012. To estimate the extent to which LDV may be causing human infection in the region, we tested stored blood samples collected in 2016 for evidence of antibodies to LDV, revealing that up to 76% of Ugandans in some areas had previously been exposed to either to LDV or a closely related virus. Seroprevalence was highest in Western Uganda, an area of high biodiversity where several other ledanteviruses have been isolated in association with ectoparasites of bats. To investigate potential ecological reservoirs of zoonotic viruses we tested blood from wild rodents inhabiting areas close to human settlements. We detected a new ledantevirus, closely related to LDV, in blood from a wild rodent Mastomys erythroleucus. Our work shows that ledanteviruses are a common cause of human infection in Uganda. Background: Cattle and buffaloes can contract cysticercosis, an infection of the muscles brought on by Taenia saginata larvae. Despite having a global spread, cysticercosis is more prevalent in impoverished nations due to impaired hygiene standards. It has been discovered that Taenia saginata cysticercosis routine visual diagnosis is not very effective, especially in mild infections. Therefore, a more trustworthy in vivo test might be used as an alternative in slaughterhouses and epidemiological studies. Biochemical assays are possibly utilized as an alternative to detect cysticercosis inside a topical environment. Aim: Investigating serum biochemical alterations in cattle with cysticercosis was the goal of the current research. As a further method of diagnosis, it was also determined how Cysticercus bovis affected pro-inflammatory cytokines and histopathology. Methods: Blood samples from 42 slaughtered cattle (21 healthy and 21 sick animals) were taken from Assiut abattoir. Using an ELISA and spectrophotometer, respectively, their serum's pro-inflammatory cytokines and biochemical profile were evaluated. These cattle were chosen between March 2023 and February 2024. Results: A percentage of 4.6% of the 455 cattle examined after being slaughtered had T. saginata cysticerci infections. All values in the serum biochemistry were considerably different (p < 0.01), whereas the majority of biochemical parameters increased significantly (p < 0.01) in infected animals. In contrast, there was a substantial (p < 0.01) decline in HDL-c, SOD, CAT, and GSH. On the other hand, procytokine inflammatory indices for both TNF-α and IL-1β indicated a substantial increase (p < 0.01) in infected cattle. Additionally, the histological results revealed significant alterations in the tissues of infected livestock. Conclusion: This has been inferred cysticercosis possesses negative impacts on cattle's plasma biochemical profiles, indicating the field applicability of biochemical measures in outbreaks of bovine cysticercosis. This study was performed to analyze fingertip capillary blood sampling in pediatric patients using microcapillary blood collection tubes and microhematocrit tubes and to compare the blood cell analysis results obtained via these two blood collection methods. Finger capillary blood was collected from 110 outpatients using microcapillary blood collection tubes and microhematocrit tubes and complete blood count analysis was performed with a Sysmex XS-900i hematology analyzer and manual microscopy for blood cell morphology. Paired data was evaluated for agreement and bias using the microhematocrit samples as the reference group and the samples from the microcapillary blood collection tubes as the observation group. The two blood collection methods demonstrated good agreement for measuring red blood cell (RBC) parameters (i.e., RBC, Hb, Hct, MCV, MCH and MCHC), wherein the relative bias was > allowable total error (TEa) in 0.91%, 1.82%, 11.82%, 1.82%, 0.91% and 8.18% of the parameter measures, respectively. According to industry requirements, the proportion of samples meeting the acceptable bias level should be > 80%. Additionally, the estimated biases at each medical decision level were within clinically acceptable levels for RBC, Hb, Hct, and MCV. However, the proportion of WBC and PLT counts with relative bias > TEa was 25.45% and 35.45%, respectively. Furthermore, the relative bias of the WBC count at the medical decision level of 0.5 × 109/L and that of the PLT counts at the medical decision levels of 10 × 109/L and 50 × 109/L were clinically significant. Bland–Altman analysis further showed a mean bias of 0.66 × 109/L (95% LoA, − 0.79 to 2.11) for the WBC count and 39 × 109/L (95% LoA, − 46 to 124) for the PLT count from the blood samples collected in the microcapillary blood collection tubes compared with the counts of those collected in the microhematocrit tubes. Neutrophil, monocyte, lymphocyte, eosinophil, and PLT counts increased significantly in the microcapillary blood collection tubes compared with those in the microhematocrit tubes, along with an elevated number of instrument false alarms (P < 0.05). The two capillary blood collection devices exhibit performance differences. Therefore, clinicians should pay attention to the variation in results caused by different blood collection methods. The avian life cycle is composed by a progressive sequence of life history stages (LHS). Changes in energy expenditure and exposure to stressors at different LHS require corresponding changes in behavior, physiology, and morphology. Variation in hematological parameters, such hematocrit (Hct), hemoglobin (Hb), and heterophil to lymphocyte ratio (H/L ratio), can have permissive, stimulatory, and preparative actions to help maintain homeostasis through different LHS. Few studies have examined differences in these parameters among different LHS in free‐living birds, with most of them restricted to temperate zones. We collected blood samples and measured hematological parameters every week for over a year from a population of a common resident bird species in southeastern Brazil, the pale‐breasted thrush Turdus leucomelas. Hematocrit and hemoglobin concentration were highest during the onset of the reproduction and lowest during molt. Furthermore, H/L ratios were higher at the end of the reproduction, indicating that the breeding season could be the most stressful period of the year for this population of thrushes. There was no difference between sexes for any hematological parameter at any LHS. These results show that there is a permissive physiological effect for Hct and Hb to facilitate LHS transitions and that reproduction could be the most stressful event for this species. The treatment of early triple-negative breast cancer (eTNBC) has improved patients' prognosis but often leads to adverse events and sequelae affecting quality of life (QoL). Pain Neuroscience Education (PNE) is a promising non-pharmacological intervention in this field. Preliminary data have shown the beneficial effect of PNE in BC survivors. However, there are still gaps in knowledge regarding its optimal use in eTNBC. To address this issue, a prospective pilot study will enroll 30 consecutive patients diagnosed with eTNBC at IRCCS Humanitas Research Hospital. The PNE program will consist of 10 weekly sessions to be started within 4 weeks of the onset or worsening of a pain syndrome (PS). QoL, pain, and disability will be assessed before, during, at the end of, and 6 months after PNE using validated questionnaires. Peripheral venous blood samples will be taken before and at the end of PNE to evaluate inflammatory serum biomarker levels. The primary objective is to evaluate whether PNE leads to clinical improvement in QoL and pain. If successful, it will be validated in a larger multi-centric cohort, potentially leading to its widespread implementation as a standard pain management tool for eTNBC patients. Objective: Bartonella are emerging bacterial zoonotic pathogens. Utilization of clotted blood samples for surveillance of these bacteria in wildlife has begun to supersede the use of tissues; however, the efficacy of these samples has not been fully investigated. Our objective was to compare the efficacy of spleen and blood samples for DNA extraction and direct detection of Bartonella spp. via qPCR. In addition, we present a protocol for improved DNA extraction from clotted, pelleted (i.e., centrifuged) blood samples obtained from wild small mammals. Results: DNA concentrations from kit-extracted blood clot samples were low and A260/A280 absorbance ratios indicated high impurity. Kit-based DNA extraction of spleen samples was efficient and produced ample DNA concentrations of good quality. We developed an in-house extraction method for the blood clots which resulted in apposite DNA quality when compared to spleen samples extracted via MagMAX DNA Ultra 2.0 kit. We detected Bartonella in 9/30 (30.0%) kit-extracted spleen DNA samples and 11/30 (36.7%) in-house-extracted blood clot samples using PCR. Our results suggest that kit-based methods may be less suitable for DNA extraction from blood clots, and that blood clot samples may be superior to tissues for Bartonella detection. A cross-sectional study has been undertaken in El-Minia province to survey an outbreak of foot-and-mouth disease (FMD) in cattle and identify the associated risk factors. From November 2021 to October 2022, researchers collected 100 blood samples, ten tongue epithelium specimens, and saliva swabs from animals of various ages and sexes in different locations. Additionally, the animals were examined for FMD signs. The overall seroprevalence was tested via SNT, which was 67%. Tissue samples and swabs were isolated and examined using conventional RT-PCR for detection of the FMD serotype, revealing serotype A. Statistically, no significant variation (P > 0.05) was observed in the prevalence of FMD in relation to age, sex, season, or locality. The findings of this study indicated that the recent FMD outbreak has been caused by serotype A in the studied locations and that FMD is widespread in cattle populations in El-Minia province. These findings underline the necessity for immediate attention and future research to detect FMDV strains spreading in the region that aid in executing effective prevention strategies. Background: Anti‐Müllerian hormone (AMH) holds potential as a biomarker for assessing the superovulation (SO) response in cattle. Nonetheless, there exists scant information regarding this aspect in the literature concerning dairy heifers. Given this gap, our objective is to explore the viability of AMH as an indicator for gauging the SO response specifically in Holstein heifers. Furthermore, our aim encompasses examining the variations in AMH levels within the same individuals before and after undergoing SO. Methods: The study included 41 Holstein heifers. All heifers were superovulated and blood samples were taken both before and after the SO protocol. Results: The findings revealed that the mean values of serum AMH concentrations before and after SO were 0.122 ng/mL (0.093–0.248 ng/mL) and 0.119 ng/mL (0.084–0.170 ng/mL), respectively. AMH concentrations in heifers were stratified into low (<0.106 ng/mL), medium (0.107–0.126 ng/mL) and high (>0.127 ng/mL) categories both before and after SO. Conclusions: There was no significant correlation between AMH levels in the heifers both before and after SO treatment with the number of follicles, corpora lutea, total embryos collected or embryos transferred (p > 0.05). Furthermore, this study showed that serum AMH concentrations in Holstein heifers did not change after SO treatment. In this study, as AMH levels in Holstein heifers were in a narrow range, a relationship between AMH and SO response could not be determined. In future studies, we believe that it would be more useful to plan more studies in Holstein donor heifers, taking into account the number of animals and AMH levels. [ABSTRACT FROM AUTHOR]

Background: Piroplasmosis is a common and prevalent tick‐borne disease that affects equids. Objectives: To determine the infection and molecular characteristics of the piroplasms in donkeys from Xinjiang, northwestern China, we undertook a cross sectional study by collecting representative samples across several counties within the region. Methods: A total of 344 blood samples were collected from adult domestic donkeys from 13 counties in Xinjiang. PCR was conducted to test for T. equi and B. caballi in the blood samples based on the equine merozoite antigen‐1 (Ema‐1) gene and the 48 kDa rhoptry protein (BC48) gene, respectively. Results: Sixteen blood samples tested positive for piroplasms and the overall infection rate was 4.7% (16/344). Seven of the 13 counties were positive for piroplasms. Among the 16 piroplasm‐positive samples, 15 were singly infected with T. equi with an infection rate of 4.4% (15/344), and coinfection with T. equi and B. caballi was detected in one sample (0.3%, 1/344) from Wushi. Four T. equi sequence genotypes were identified and grouped into different branches of the evolutionary trees. Conclusion: These findings suggest that the infection rate of piroplasms is low in domestic donkeys in southern Xinjiang and that T. equi genotypes have a regional distribution. Oral submucous fibrosis (OSF) affecting populations is considered a public health issue in South/Southeast Asia. The purpose of this study was to analyze the scientometric characteristics and research trends of OSF. All the papers on OSF were comprehensively retrieved from the Scopus database. Regional comparison (India versus outside of India) and chronological comparison (before 2015 versus after 2015) were performed. Among all the 1357 papers on OSF, 930 (68.5%) were from India. In India, biology research on antioxidant, oxidative stress, angiogenesis, and extracellular matrix were distinctive keywords. Risk factors of smokeless tobacco and gutkha and the roles of saliva and blood sampling were also distinctive keywords in India. In outside of India, biology research on myofibroblast, alpha smooth muscle actin, microRNA, long untranslated RNA, and protein p53 were distinctive keywords. The trend of biology research on connective tissue, genotype, genetic predisposition, messenger RNA, and cytology before 2015 has changed to research on myofibroblast, biological marker, microRNA, epithelial mesenchymal transition, extracellular matrix, and oxidative stress after 2015. The trend of clinical aspects of surgery and mouth hygiene before 2015 has changed to the aspects of adverse event/effects, complication, and quality of life after 2015. This scientometric study elucidated the current scenario and research trends of OSF, and would help in improving in reciprocal collaboration and communication for this disease control in South/Southeast Asia. Objective: To compare the diagnostic efficacy of multiplex polymerase chain reaction (PCR), Mycobacterium leprae-specific repetitive element (RLEP) PCR and loop-mediated isothermal amplification (LAMP) PCR in the diagnosis of pediatric leprosy as an alternative to slit-skin smear (SSS) examination. Methods: A cross-sectional study was performed on 26 children aged 0–18 years with characteristic skin lesions of leprosy. SSS examination for acid fast bacilli (AFB) was performed for all children. Additionally, urine, stool and blood samples were tested by three PCR techniques - multiplex, RLEP and LAMP. The results of these tests were compared with each other and with results of SSS examination for acid fast bacilli (AFB) using appropriate statistical tests. Results: Out of 26 patients studied, SSS examination was positive for AFB in 7 cases (26.9%). In blood samples, the positivity of multiplex PCR, RLEP PCR and LAMP PCR was 84.6%, 80.8%, and 80.8%, respectively. Multiplex PCR in blood samples was positive in 100% (n = 7) of SSS positive cases and 84.2% (16 out of 19) of the SSS negative cases (P < 0.001). The positivity of all PCR methods in urine and stool samples was significantly lesser than in blood. Conclusion: Multiplex PCR in blood sample is a superior diagnostic tool for pediatric leprosy compared to RLEP PCR and LAMP PCR as well as SSS examination. [ABSTRACT FROM AUTHOR]

The present study was conducted to investigate the internal and external biosecurity levels and their association with Mycoplasma gallisepticum infection in broiler farms in Khartoum State, Sudan. Sixty-six broiler farms were selected. A questionnaire of 77 questions on internal and external biosecurity measures was administered during each farm visit. A total of 360 blood samples were collected and used for serological examination to detect antibodies against M. gallisepticum. The overall score of biosecurity was 0.57, and the average score of internal biosecurity (0.75) was higher (p<0.01) than the average score of external biosecurity (0.51). Poor internal biosecurity practices were observed in 23.4% of broiler farms, whereas most farms (93.6%) recorded poor external biosecurity. A significant (p<0.01) positive correlation was encountered between the external biosecurity and the number of houses (0.62), as well as the number of workers (rs= 0.54). Both internal and external biosecurity displayed significant positive correlations with the number of birds (rs= 0.64, 0.35; p< 0.05). Fifty percent of the farms were positive for M. gallisepticum. The disease incidence correlated positively with the spread of manure on neighboring farms (rs= 0.44, p<0.05). A negative correlation was observed between the presence of Mycoplasma and the distance between farms (rs= -0.50, p<0.01) as well as farm enclosure (rs= -0.44, p<0.05). It is concluded that broiler farms in Khartoum State exhibited low biosecurity measures and a high seroprevalence of M. gallisepticum infection. This research paper demonstrates a metamaterial (MTM) based sensing technique to detect various blood samples by analyzing their dielectric properties. The performance of this MTM-based sensor is evaluated with the help of mimicked human blood samples that closely resemble the dielectric properties of actual human blood samples. Moreover, the ISM band frequency of 2.4 GHz is chosen as one of the reference resonance frequencies due to its various industrial and medical applications. The resonating patch is developed on the FR-4 substrate with a dimension of 10 × 20 mm2 that provides sharp reference resonances of 2.4 and 4.72 GHz for the spectra of the transmission coefficient with a good quality factor (Q-factor). The MTM sensor can detect the mimicked blood samples with a maximum frequency deviation of up to 650 MHz at 2.4 GHz and up to 850 MHz at 4.72 GHz, with maximum sensitivity of 0.917 and 0.707, respectively. The measured results using the prototype of the sensor support the simulation result with good agreement, indicating high sensing capability. Due to its high sensitivity, figure of merit (FoM), and frequency shifting with dielectric property changes in blood samples, the developed MTM-based sensor can be implemented effectively for quick sensing of infected blood samples and biomedical applications. Background: Toxoplasmosis is a common disease caused by an apicomplexian protozoa, Toxoplasma gondii; one of the potential sources of human zoonotic infection is meat handling and consumption. Objectives: This study aims to determine the prevalence of T. gondii in the blood of butchers and slaughtered cattle by using the B1 gene and demonstrate the role of slaughtered cattle in transmitting toxoplasmosis to humans. Methods: The study involved 100 blood samples collected randomly from butchers (male) and 100 from cattle before slaughtering from September 2022 to September 2023 and examined by real-time PCR technique. Results: The results revealed total infection rates of 28% and 12% in butchers and cattle, respectively, with the highest rate recorded in butchers aged 18-30 years old and the lowest at age >40 years. While in cattle, the highest rate is seen at age (2-4 years) and the lowest at age (1-2 years) without significant differences. Calves reported a higher infection rate (14.28%) compared with cows (8.10%), with a significant difference (P>0.05). Conclusion: Cattle are an important source of zoonotic transmission of T. gondii infection to humans by handling meat and consuming raw or uncooked meat. We recommended a control program by awareness of butchers during meat handling, avoiding consuming raw or uncooked meat, and appropriate heat treatments for beef. [ABSTRACT FROM AUTHOR]

BACKGROUND: The need for arterial blood data in quantitative PET research limits the wider usability of this imaging method in clinical research settings. Image-derived input function (IDIF) approaches have been proposed as a cost-effective and non-invasive alternative to gold-standard arterial sampling. However, this approach comes with its own limitations-partial volume effects and radiometabolite correction among the most important-and varying rates of success, and the use of IDIF for brain PET has been particularly troublesome. MAIN BODY: This paper summarizes the limitations of IDIF methods for quantitative PET imaging and discusses some of the advances that may make IDIF extraction more reliable. The introduction of automated pipelines (both commercial and open-source) for clinical PET scanners is discussed as a way to improve the reliability of IDIF approaches and their utility for quantitative purposes. Survey data gathered from the PET community are then presented to understand whether the fields opinion of the usefulness and validity of IDIF is improving. Finally, as the introduction of next-generation PET scanners with long axial fields of view, ultra-high sensitivity, and improved spatial and temporal resolution, has also brought IDIF methods back into the spotlight, a discussion of the possibilities offered by these state-of-the-art scanners-inclusion of large vessels, less partial volume in small vessels, better description of the full IDIF kinetics, whole-body modeling of radiometabolite production-is included, providing a pathway for future use of IDIF. CONCLUSION: Improvements in PET scanner technology and software for automated IDIF extraction may allow to solve some of the major limitations associated with IDIF, such as partial volume effects and poor temporal sampling, with the exciting potential for accurate estimation of single kinetic rates. Nevertheless, until individualized radiometabolite correction can be performed effectively, IDIF approaches remain confined at best to a few tracers.

Objective To obtain the monitoring measurement range of quality indicators of red blood cells, plasma and derivatives and leukocyte-reduced apheresis platelets provided by blood stations in Hebei province, explore the distribution of monitoring values and the change of monitoring level, so as to further strengthen the homogenization construction of quality control laboratories in blood stations in Hebei. Methods In 2023, the sampling data of 12 blood stations in Hebei from 2015 to 2022 were collected, scatter plots were made and the range markers were set, and the "mean±SD" line was taken as the upper limit and lower limit of the measurement range. In 2024, the monitoring values in 2023 were added, and the changes of two measurement ranges were compared to analyze the stability and overall level. Results Comparison of the measurement range from 2015 to 2022 and the measurement range from 2015 to 2023 showed that the standard deviation of the content of deleukocyte suspension of red blood cells-hemoglobin, washed erythrocyte-hemoglobin, washed erythrocyte-supernatant protein, cryoprecipitate coagulation factor-FⅧ, fresh frozen plasma-FⅧ, leukocyte-reduced apheresis platelets-leukocyte residue and leukocyte-reduced apheresis platelet-red blood cell concentration decreased from 8.132 to 7.993, 6.252 to 6.104, 0.273 to 0.267, 57.506 to 56.276, 0.920 to 0.892, 0.653 to 0.644 and 2.653 to 2.603, respectively.The narrowing of the standard deviation range of the above items led to more concentrated monitoring values and reduced dispersion. Comparison of the measurement range from 2015 to 2022 and the measurement range from 2015 to 2023 showed that the mean value of leukocyte residue of the deleukocyte suspension of red blood cells, hemoglobin content of the wash erythrocyte, protein content of supernatant of the wash erythrocyte, hemolysis rate of the wash erythrocyte, FⅧ content of the cryoprecipitate coagulation factor, plasma protein content of the fresh frozen plasma, FⅧ content of the fresh frozen plasma, platelet content of the leukocyte-reduced apheresis platelets changed from 0.362 to 0.476, 44.915 to 44.861, 0.280 to 0.283, 0.137 to 0.142, 133.989 to 133.271, 60.262 to 60.208, 1.301 to 1.277 and 3.036 to 3.033, respectively, and was closer to the national standard line, which reflects an increase in the number of unqualified monitoring values or values close to the national standard line in 2023. The long-term qualified rate of coagulation items was low, and no improvement has been observed. The stability of biochemical items has been enhanced but overall deviation has occurred, with the average value close to the national standard line. The possibility of subsequent testing failure has increased. The counting items showed no obvious common characteristics. Conclusion The use of "mean±SD" in the analysis can visually display the distribution of monitoring values of different items in Hebei, forming an indicator measurement range covering the past nine years. It shows the characteristics of each item, and provides reference for subsequent quality control laboratory data analysis of each blood stations to takes active measures to improve the monitoring level.

Background: Effective testing for malaria, including the detection of infections at very low densities, is vital for the successful elimination of the disease. Unfortunately, existing methods are either inexpensive but poorly sensitive or sensitive but costly. Recent studies have shown that mid-infrared spectroscopy coupled with machine learning (MIRs-ML) has potential for rapidly detecting malaria infections but requires further evaluation on diverse samples representative of natural infections in endemic areas. The aim of this study was, therefore, to demonstrate a simple AI-powered, reagent-free, and user-friendly approach that uses mid-infrared spectra from dried blood spots to accurately detect malaria infections across varying parasite densities and anaemic conditions. Methods: Plasmodium falciparum strains NF54 and FCR3 were cultured and mixed with blood from 70 malaria-free individuals to create various malaria parasitaemia and anaemic conditions. Blood dilutions produced three haematocrit ratios (50%, 25%, 12.5%) and five parasitaemia levels (6%, 0.1%, 0.002%, 0.00003%, 0%). Dried blood spots were prepared on Whatman™ filter papers and scanned using attenuated total reflection-Fourier Transform Infrared (ATR-FTIR) for machine-learning analysis. Three classifiers were trained on an 80%/20% split of 4655 spectra: (I) high contrast (6% parasitaemia vs. negative), (II) low contrast (0.00003% vs. negative) and (III) all concentrations (all positive levels vs. negative). The classifiers were validated with unseen datasets to detect malaria at various parasitaemia levels and anaemic conditions. Additionally, these classifiers were tested on samples from a population survey in malaria-endemic villages of southeastern Tanzania. Results: The AI classifiers attained over 90% accuracy in detecting malaria infections as low as one parasite per microlitre of blood, a sensitivity unattainable by conventional RDTs and microscopy. These laboratory-developed classifiers seamlessly transitioned to field applicability, achieving over 80% accuracy in predicting natural P. falciparum infections in blood samples collected during the field survey. Crucially, the performance remained unaffected by various levels of anaemia, a common complication in malaria patients. Conclusion: These findings suggest that the AI-driven mid-infrared spectroscopy approach holds promise as a simplified, sensitive and cost-effective method for malaria screening, consistently performing well despite variations in parasite densities and anaemic conditions. The technique simply involves scanning dried blood spots with a desktop mid-infrared scanner and analysing the spectra using pre-trained AI classifiers, making it readily adaptable to field conditions in low-resource settings. In this study, the approach was successfully adapted to field use, effectively predicting natural malaria infections in blood samples from a population-level survey in Tanzania. Simple Summary: The metabolic profile implies simultaneous determination of carbohydrate, fat, protein, and mineral metabolism parameters as well as endocrinological parameters in the blood of cows. Blood is exposed to a variety of preanalytical factors during sampling, transport to the laboratory, and laboratory preparation for analysis, which may cause hemolysis of the sample. As hemolysis affects the values of the metabolic profile, the analyzed blood parameters may falsely increase or decrease, and the metabolic status of the cows may be misinterpreted. Preventing hemolysis is important because severe hemolysis requires discarding the sample and resampling, which is very resource-intensive. In this paper, three levels of hemolysis were determined for each blood parameter tested: (a) a hemolysis level that does not affect the values of the parameters and allows the results to be issued without restriction; (b) a hemolysis level that affects the values of the parameters but remains within the acceptable biological variability and permits the results to be issued along with a note in the form of a correction formula; and (c) a hemolysis level at which the obtained values of the parameters or the entire sample must be discarded. The results are presented graphically using interferograms, which can be easily implemented in every laboratory after validation. Hemolysis is a common cause of errors in laboratory tests as it affects blood parameters and leads to a positive or negative bias. This study aims to examine the relationship between the level of hemolysis (expressed as cell-free hemoglobin concentration, g/L) and the variability of metabolic and endocrine parameters and to determine the threshold level of hemolysis that causes an analytically and clinically significant bias for the twenty most frequently examined blood parameters in cows. Paired blood samples of 10 mL each were obtained from 30 cows. One was subjected to mechanical trauma and plasma was extracted directly from the other. Hemolyzed and non-hemolyzed samples from the same animal were mixed to obtain final samples with cell-free hemoglobin concentrations of 0, 1, 2, 4, 6, 8, and 10 g/L. Metabolic and endocrine parameters were measured in the samples and their deviation and the linear equation between the level of hemolysis and the deviation were determined. The following threshold values of hemolysis were determined, which correspond to the acceptable analytical (lower value) and clinical (upper value) levels of parameter variability: BHB 0.96 and 4.81; NEFA 0.39 and 3.31; GLU 0.38 and 3.90; ALB 1.12 and 6.11; TPROT 1.40 and 6.80; UREA 6.62 and 20.1; TBIL 0.75 and 5.65; AST 0.11 and 2.18; GGT 1.71 and 8.90, LDH 0.01 and 0.11, ALP 0.97 and 2.95; TGC 1.56 and 15.5; CHOL 1.29 and 8.56; Ca 5.68 and 25.7; P 0.57 and 8.43; Mg 1.10 and 8.47; INS 1.15 and 3.89; T3 8.19 and 15.6; T4 8.97 and 18.5; and CORT 2.78 and 11.22 g/L cell-free hemoglobin. Three decision levels are available for each metabolic and endocrine parameter: if hemolysis is below the lower (analytical) threshold value, results can be reported without restriction; if hemolysis is between the lower and upper thresholds, the results can be issued with guidance in the form of corrective linear equations; and if hemolysis is above the upper (clinical) threshold, the results and sample must be discarded. A pre-transfusion crossmatch test is crucial for ensuring safe blood transfusions by identifying the compatibility between donor and recipient blood samples. Conventional tube methods for crossmatching have limitations, including subjectivity in result interpretation and the potential for human error. In this study, we evaluated the diagnostic performance of a new crossmatch test using Microscanner C3, which can overcome these shortcomings. The crossmatch test results using the method were obtained in 323 clinical samples. The sensitivity, specificity, positive predictive value, negative predictive value, and concordance rate of the crossmatch test using Microscanner C3 were 98.20%, 100.00%, 100.00%, 98.11%, and 99.07%, respectively. The diagnostic performance of the new system offers a promising alternative to conventional tube methods for pre-transfusion crossmatch testing. Microscanner C3 could also increase the automation, standardization, and accuracy of crossmatch tests. The crossmatch test using Microscanner C3 is thought to increase the efficiency and reliability in identifying blood samples suitable for transfusion, thereby improving patient safety and optimizing the use of blood products in clinical settings. The study presents the importance of forensic bacteriology, its use in forensic cases, the methods for bacteriological sampling from corpses, the types and species of bacteria isolated from human and pig corpses, which are most commonly used in forensic biology. The microbial changes that occur after death remain unclear. Postmortem microbiology is a relatively new field of research. After death, the failure of the immune system and other physical barriers leads to the proliferation and spread of microbes. In order for bacteriological information to be accepted within the scope of forensic bacteriology, the court must find suspicion to be present about whether the bacteria seen on the body will contribute to solving the case. Experts must be appointed to examine the issue in line with this suspicion, and these experts must prepare and submit their reports to the court at the requested time. When considering the literature studies, forensic bacteriology has been suggested to be a scientific discipline in the developmental stage and to only be able to provide circumstantial evidence in forensic cases as opposed to primary evidence. According to the literature review, most bacterial studies isolated from corpses were conducted in Romania. Although bacterial samples were isolated from various parts of the corpses, bacteria were mostly isolated from their blood samples. According to literature searches from various scientific journal databases, no study has occurred with a list of the bacteria isolated from corpses. Simple Summary: Early pregnancy diagnosis is of major importance to the economics of dairy herds. The detection of placental proteins, termed pregnancy-associated glycoproteins (PAGs), represents a diagnostic tool to identify both pregnant and non-pregnant cows. However, as milk production correlates negatively with plasma levels of PAGs during early pregnancy, detection of PAGs may be hindered by high milk production. This study evaluates a newly developed commercial PAGs-based pregnancy test using whole blood or plasma samples during early pregnancy (28–55 days of gestation) in high-producing dairy cows. This study evaluated a commercial pregnancy-associated glycoproteins (PAGs)-based pregnancy test using whole blood or plasma samples during early pregnancy (28–55 days of gestation) in high-producing dairy cows. Transrectal ultrasonography was used as the gold standard method. The study population constituted of 284 cows. False positive diagnoses were recorded from Day 60 to 89 and from Day 60 to 99 postpartum in blood and plasma samples, respectively. In early pregnancy screening, correct positive diagnoses were recorded in 75% and 100% of blood and plasma samples, respectively. High milk production was associated with negative results in blood samples and with the lowest test line intensity in plasma samples. False positive or negative diagnoses were recorded in 0% of both types of samples in cows previously diagnosed as pregnant and showing signs of estrus. In conclusion, the use of plasma was more effective than the use of blood in early pregnancy diagnosis. In cows previously diagnosed as pregnant and showing signs of estrus, both types of samples showed the same results. Because of large individual variations, normal single pregnancies could not be differentiated from twin pregnancies, from pregnancies with a recently dead conceptus, or from pregnancies that experienced subsequent pregnancy loss. Currently, tandem mass spectrometry-based newborn screening (NBS), which examines targeted biomarkers, is the first approach used for the early detection of maple syrup urine disease (MSUD) in newborns, followed by confirmatory genetic mutation tests. However, these diagnostic approaches have limitations, demanding the development of additional tools for the diagnosis/screening of MUSD. Recently, untargeted metabolomics has been used to explore metabolic profiling and discover the potential biomarkers/pathways of inherited metabolic diseases. Thus, we aimed to discover a distinctive metabolic profile and biomarkers/pathways for MSUD newborns using untargeted metabolomics. Herein, untargeted metabolomics was used to analyze dried blood spot (DBS) samples from 22 MSUD and 22 healthy control newborns. Our data identified 210 altered endogenous metabolites in MSUD newborns and new potential MSUD biomarkers, particularly L-alloisoleucine, methionine, and lysoPI. In addition, the most impacted pathways in MSUD newborns were the ascorbate and aldarate pathways and pentose and glucuronate interconversions, suggesting that oxidative and detoxification events may occur in early life. Our approach leads to the identification of new potential biomarkers/pathways that could be used for the early diagnosis/screening of MSUD newborns but require further validation studies. Our untargeted metabolomics findings have undoubtedly added new insights to our understanding of the pathogenicity of MSUD, which helps us select the appropriate early treatments for better health outcomes. We investigated the pharmacokinetic pathway of berberine and its metabolites in vitro, in Caco-2 cells, and in human participants following the administration of dihydroberberine (DHB) and micellar berberine (LipoMicel®, LMB) formulations. A pilot trial involving nine healthy volunteers was conducted over a 24 h period; blood samples were collected and subjected to Ultra High-Performance Liquid Chromatography–High Resolution Mass Spectrometry (UHPLC-HRMS) analyses to quantify the concentrations of berberine and its metabolites. Pharmacokinetic correlations indicated that berberrubine and thalifendine follow distinct metabolic pathways. Additionally, jatrorrhizine sulfate appeared to undergo metabolism differently compared to the other sulfated metabolites. Moreover, berberrubine glucuronide likely has a unique metabolic pathway distinct from other glucuronides. The human trial revealed significantly higher blood concentrations of berberine metabolites in participants of the DHB treatment group compared to the LMB treatment group—except for berberrubine glucuronide, which was only detected in the LMB treatment group. Similarly, results from in vitro investigations showed significant differences in berberine metabolite profiles between DHB and LMB. Dihydroberberine, dihydroxy-berberrubine/thalifendine and jatrorrhizine sulfate were detected in LMB-treated cells, but not in DHB-treated cells; thalifendine and jatrorrhizine-glucuronide were detected in DHB-treated cells only. While DHB treatment provided higher blood concentrations of berberine and most berberine metabolites, both in vitro (Caco-2 cells) and in vivo human studies showed that treatment with LMB resulted in a higher proportion of unmetabolized berberine compared to DHB.
