Drought is a critical abiotic stress which leads to crop yield and a decrease in quality. Annexins belong to a multi-gene family of calcium- and lipid-binding proteins and play diverse roles in plant growth and development. Herein, we report a rice annexin protein, OsANN9, which in addition to regular annexin repeats and type-II Ca2+ binding sites, also consists of a C2H2-type zinc-finger domain. We found that the expression of OsANN9 was upregulated by polyethylene glycol (PEG) or water-deficient treatment. Moreover, plants that overexpressed OsANN9 had increased survival rates under drought stress, while both OsANN9-RNAi and osann9 mutants showed sensitivity to drought. In addition, the overexpression of OsANN9 increased superoxide dismutase (SOD), peroxidase (POD) and catalase (CAT) activities, which regulate reactive oxygen species homeostasis. Collectively, these findings indicate that OsANN9 may function as a positive regulator in response to drought stress by modulating antioxidant accumulation. Interestingly, the setting rates of osann9 mutant rice plants significantly decreased in comparison to wild-type plants, suggesting that OsANN9 might be involved in other molecular mechanisms in the rice seed development stage. Background: Increasing studies have demonstrated potential disproportionate functional and ecological contributions of rare taxa in a microbial community. However, the study of the microbial rare biosphere is hampered by their inherent scarcity and the deficiency of currently available techniques. Sample-wise cross contaminations might be introduced by sample index misassignment in the most widely used metabarcoding amplicon sequencing approach. Although downstream bioinformatic quality control and clustering or denoising algorithms could remove sequencing errors and non-biological artifact reads, no algorithm could eliminate high quality reads from sample-wise cross contaminations introduced by index misassignment, making it difficult to distinguish between bona fide rare taxa and potential false positives in metabarcoding studies. Results: We thoroughly evaluated the rate of index misassignment of the widely used NovaSeq 6000 and DNBSEQ-G400 sequencing platforms using both commercial and customized mock communities, and observed significant lower (0.08% vs. 5.68%) fraction of potential false positive reads for DNBSEQ-G400 as compared to NovaSeq 6000. Significant batch effects could be caused by stochastically introduced false positive or false negative rare taxa. These false detections could also lead to inflated alpha diversity of relatively simple microbial communities and underestimated that of complex ones. Further test using a set of cow rumen samples reported differential rare taxa by different sequencing platforms. Correlation analysis of the rare taxa detected by each sequencing platform demonstrated that the rare taxa identified by DNBSEQ-G400 platform had a much higher possibility to be correlated with the physiochemical properties of rumen fluid as compared to NovaSeq 6000 platform. Community assembly mechanism and microbial network correlation analysis indicated that false positive or negative rare taxa detection could lead to biased community assembly mechanism and identification of fake keystone species of the community. Conclusions: We highly suggest proper positive/negative/blank controls, technical replicate settings, and proper sequencing platform selection in future amplicon studies, especially when the microbial rare biosphere would be focused. The potential application value of superhydrophobic surfaces with anti-icing and de-icing properties in aerospace, telecommunications cables, energy and other fields has always been an important research topic. In this paper, a kind of superhydrophobic surface with stable structure, sufficient heat, excellent heat transfer performance and mass production was prepared based on elastic material. In this study, the multi-stage micro-nano rough structure is synthesized by ingenious pressing and mold closing control, and the surface contact angle of the prepared sample can reach 159°. The multi-stage micro-nano rough structure on the surface of the experimental sample can make the water droplets form a large number of cavitation, which can delay the icing of static water droplets. The height difference on both sides of (Un)pressed mesh surface makes the water droplets freeze asymmetrically, so (Un)pressed mesh surface has excellent anti-icing ability. The experimental sample is connected with 24 V direct current, and the average resistivity is 1.403 Ω·cm. The temperature can be raised to about 60 °C in one minute. The prepared sample has stable temperature, good electrical conductivity, sufficient heat generation, excellent heat transfer performance and high anti-icing efficiency. Aircraft anti-icing and de-icing, aimed at reducing energy consumption, is a compelling technology that enables a plethora of eco-friendly energy applications to overcome long-standing crisis challenges. In this study, we present a tri-conversion strategy, inspired by superhydrophobicity, photothermal conversion, and electrothermal conversion, to develop an all-weather, high-efficiency, and low-energy rotor leading edge anti-icing and de-icing patch. The experimental samples not only possess stable superhydrophobic photothermal conversion capabilities but also achieve a 610-second delay in droplet icing, a 1-second nitrogen blow for frost, and a low ice adhesion strength of 11.6 KPa. Under one sun illumination, it can achieve a stable 54-second static melt and 37-second dynamic melt. With a connection to a 24 V DC stabilized power supply, it showcases an excellent 146-second de-icing capability. The synergistic action of multiple barriers enhances the anti-icing and de-icing performance. This work not only provides rational design principles for high-energy consumption de-icing but also offers insights into harnessing the power of nature for photothermal conversion de-icing. Ecosystem stability is threatened by multiple global change factors such as drought and elevated nitrogen deposition. Yet, it is still poorly understood whether soil organisms can buffer against such perturbations. Here we focus on arbuscular mycorrhizal fungi (AMF), a common and widespread group of soil fungi. AMF form symbiotic associations with the majority of terrestrial land plants and promote a range of ecosystem services including plant production, diversity and nutrient cycling.We tested whether AMF have the ability to enhance the resistance and resilience of plant communities under soil moisture deficit (hereby drought) and nitrogen deposition. Grassland microcosms with 11 different plant species were established with and without AMF and exposed to elevated nitrogen levels and to an intermittent period of drought.Drought strongly reduced plant productivity and nitrogen cycling, but had limited effects on plant diversity. Nitrogen enrichment reduced plant diversity and increased nitrogen leaching and N2O emissions. The presence of AMF enhanced plant productivity, plant diversity and reduced nitrogen losses. AMF facilitated the resistance of plant productivity and nitrogen cycling to drought and the recovery of the plant community structure back to its pre drought state. Furthermore, AMF also mitigated the adverse effects of nitrogen enrichment on the resistance of multiple ecosystem functions to drought.Synthesis. Our work highlights the integral role of AMF for the stability of ecosystem functioning; AMF are not only able to promote resistance to harsh conditions of global change but also improve resilience by enabling plant communities to recover. Summary: Escherichia coli O157:H7, a causative agent of haemolytic uremic syndrome, can enter into a viable but non‐culturable (VBNC) state in response to harsh stress. Bacteria in this state can retain membrane integrity, metabolic activity and virulence expression, which may present health risks. However, virulence expression and resuscitation ability of the VBNC state are not well understood. Here, we induced E. coli O157:H7 into a VBNC state by high temperature, which is commonly used to prevent the proliferation of pathogens in process of soil solarization, composting and anaerobic digestion of organic wastes. The virulence genes were highly expressed in the VBNC state and resuscitated daughter cells. The resuscitation of VBNC cells occurred after the removal of heat stress in Luria‐Bertani medium. In addition, E. coli O157: H7 cells can leave the VBNC state and resuscitate with the clearance of protein aggregates. Notably, with the accumulation of protein aggregation and increased levels of reactive oxygen species, cells lost their ability to resuscitate. The results of this study not only can facilitate a better understanding of the health risks associated with the VBNC state but also have the potential to provide a theoretical basis for thermal disinfection processing. Global climate change is one of the most pressing conservation challenges; in particular, changes in precipitation regimes have already substantially influenced terrestrial ecosystems. However, the mechanisms influencing precipitation changes on individual plants and the plant communities in desert grasslands have yet to be fully elucidated. We therefore examine the influence of increased precipitation on plant community compositions in the Gurbantunggut Desert, Xinjiang, northwestern China, from 2005 to 2009. We found that growth of all plant species and the community productivities increased markedly with enhanced water input. Cover of ephemeral synusia also significantly increased due to increased precipitation, implying that the role of the ephemeral community for stabilization of sand dunes was strengthened by increased precipitation. The response of plant community compositions to increased precipitation was primarily reflected as changes in plant density, while increased precipitation did not affect plant species richness and the diversity index. Dominant species drove the response of plant density to increasing precipitation during the five‐year study period. However, the relative responses of rare species were stronger than those of the dominant species, thereby potentially driving species turnover with long‐term increased precipitation. This finding improved our understanding of how increased precipitation drives the changes in plant community composition in desert grasslands and will help to better predict changes in the community composition of ephemerals under future global climate change scenarios. Compared to conventional high internal phase emulsions (HIPEs), ultrastable HIPEs have recently attracted much attention due to much better stability and protection for bioactive compounds. Here, novel food-grade ultrastable HIPEs were successfully constructed. By simply mixing two individual unstable HIPEs (instability in 3 months in open atmosphere) stabilized by ternary complexes of protein (i.e., lactoferrin), two polyphenols (i.e., (−)-epigallocatechin-3-gallate and curcumin) and two anionic polysaccharides (i.e., κ-carrageenan and low methoxylated pectin), respectively, ultrastable mixed-HIPEs (very stable for 1 year or more in open atmosphere) were achieved unexpectedly. The two individual HIPEs underwent liquid-to-gel and viscous-to-elastic transitions during mixing, exhibiting strong structural synergism. The stabilization was ascribed to the electrostatic repulsive interactions and the formed interpenetrating polymer networks hydrogels in continuous phases; their contribution varied according to the binding sequences of protein, polyphenol, and polysaccharide (for ternary complexes) and the combination modes of individual HIPEs (for mixed-HIPEs). The excellent soft material properties of ternary complexes can provide guarantee for the stsability of emulsion droplets, avoiding their rupture caused by strong squeezing in mixed-HIPEs. This finding greatly expands the type of food grade ultrastable HIPEs. Fine particulate matter (PM 2.5) has been verified to augmented the incidence of pneumonia, asthma, pulmonary fibrosis, and other pulmonary diseases. Airway inflammation is the pathological basis of the respiratory system, and understanding the molecular mechanisms responsible for airway inflammation may thus support the diagnosis and treatment of respiratory diseases. In our study, human bronchial epithelial cells (BEAS-2B) were exposed to various concentrations of PM 2.5 for 48 h. PM 2.5 entered the cells, resulting in increased production of interleukin 6 (IL-6) and interleukin 8 (IL-8) and decreased the expression of circular RNA 406961 (circ_406961). Further, PM 2.5 with a concentration of 75 μg/mL was applied to mechanism study. Functional experiments further confirmed that circ_406961 inhibited PM 2.5 -induced BEAS-2B cell inflammation. RNA pull-down and mass spectrometry showed that circ_406961 interacted with interleukin enhancer-binding factor 2 (ILF2), which could regulate phosphorylation of signal transducer and activator of transcription 3 (STAT3) and mitogen-activated protein kinase 8 (MAPK8, JNK). Our studies showed that circ_406961 inhibited activation of STAT3/JNK pathways via interacting with ILF2 protein, thereby inhibiting the PM 2.5 -induced inflammatory reaction. [ABSTRACT FROM AUTHOR]

[Display omitted] Hexavalent chromium [Cr (VI)] is an important environmental pollutant and may cause lung injury when inhaled into the human body. Cr (VI) is genotoxic and can cause DNA damage, although the underlying epigenetic mechanisms remain unclear. To simulate the real-life workplace exposure to Cr (VI), we used a novel exposure dose calculation method. We evaluated the effect of Cr (VI) on DNA damage in human bronchial epithelial cells (16HBE and BEAS-2B) by calculating the equivalent real-time exposure dose of Cr (VI) (0 to 10 μM) in an environmental population. Comet experiments and olive tail moment measurements revealed increased DNA damage in cells exposed to Cr (VI). Cr (VI) treatment increased nuclear γ-H2AX foci and γ-H2AX protein expression, and caused DNA damage in the lung tissues of mice. An effective Cr (VI) dose (6 μM) was determined and used for cell treatment. Cr (VI) exposure upregulated circ_0008657, and knockdown of circ_0008657 decreased Cr (VI)-induced DNA damage, whereas circ_0008657 overexpression had the opposite effect. Mechanistically, we found that circ_0008657 binds to microRNA (miR)-203a-3p and subsequently regulates ATM serine/threonine kinase (ATM), a key protein involved in homologous recombination repair downstream of miR-203a-3p, thereby regulating DNA damage induced by Cr (VI). The present findings suggest that circ_0008657 competitively binds to miR-203a-3p to activate the ATM pathway and regulate the DNA damage response after environmental chemical exposure in vivo and in vitro. Background: Although anaerobic digestion for biogas production is used worldwide in treatment processes to recover energy from carbon-rich waste such as cellulosic biomass, the activities and interactions among the microbial populations that perform anaerobic digestion deserve further investigations, especially at the population genome level. To understand the cellulosic biomass-degrading potentials in two full-scale digesters, this study examined five methanogenic enrichment cultures derived from the digesters that anaerobically digested cellulose or xylan for more than 2 years under 35 or 55 °C conditions. Results: Metagenomics and metatranscriptomics were used to capture the active microbial populations in each enrichment culture and reconstruct their meta-metabolic network and ecological roles. 107 population genomes were reconstructed from the five enrichment cultures using a differential coverage binning approach, of which only a subset was highly transcribed in the metatranscriptomes. Phylogenetic and functional convergence of communities by enrichment condition and phase of fermentation was observed for the highly transcribed populations in the metatranscriptomes. In the 35 °C cultures grown on cellulose, Clostridium cellulolyticum-related and Ruminococcus-related bacteria were identified as major hydrolyzers and primary fermenters in the early growth phase, while Clostridium leptum-related bacteria were major secondary fermenters and potential fatty acid scavengers in the late growth phase. While the meta-metabolism and trophic roles of the cultures were similar, the bacterial populations performing each function were distinct between the enrichment conditions. Conclusions: Overall, a population genome-centric view of the meta-metabolism and functional roles of key active players in anaerobic digestion of cellulosic biomass was obtained. This study represents a major step forward towards understanding the microbial functions and interactions at population genome level during the microbial conversion of lignocellulosic biomass to methane. Introduction. Changes in eating habits have made gout a metabolic disease of increasing concern. Previous studies have indicated that there are significant differences in species composition and abundance of gut microbiome in gout patients compared with average. Considering that traditional Chinese medicine has a momentous effect in treating gout, the research study aimed to explore the differences of genomic and metabolomics of gut microbiome before and after traditional Chinese medicine treatment in patients with gout. Method. 30 patients with gout and 29 matched controls were recruited of which 16 patients took H treatment and 14 patients took T treatment. Stools were collected twice for patients before and after treatment and only once for controls. A total of 89 samples were annotated with metagenomic species and functions, and the enrichment analysis of differential genes and KO pathway was carried out. Result. The results showed a decrease in the diversity of gut microbiome in gout patients and the gene abundance and metabolomics had great differences among study groups. The number of bacterial genera also had significant differences among treatment groups. Moreover, among different groups, the regulation of different species was variously correlated. The correlation between species and clinical laboratory indicators in the rising group was stronger than that in the decreasing group and the upregulation of some strain was related to the content of urea nitrogen. Conclusion. After the traditional Chinese medicine treatment, the glutathione pathway was significantly enriched and some pathogenic bacteria were significantly inhibited. We aimed at tabulating and characterizing a flavan-3-ol isolated from date seeds into date seed pills (DSP) as a commercial healthcare pill, and deeply studying their anti-glycoxidation impacts on bovine serum albumin using multi-dimensional approaches SDS-PAGE/NBT, UPLC-ESI-Q-TOF-MS, antiradical via Fenton-type reaction, multispectral, and molecular docking. Results showed that DSP were successfully tabulated (an encapsulation efficiency of 98.78 %) with high antioxidant values via noncovalently binding with whey proteins supported by FTIR and lib-dock results. DSP dose-dependently decayed fluorescent-AGEs by 16.77, 5.32, and 37.41 % in DSP500-BSA-G/MGO/GO models, respectively, later confirmed by SDS-PAGE and initial precursors mitigation results. The docking results confirmed that DSP could bind with glycoxidation sites of BSA-subunits, namely Lys431, Arg458, and Arg196 via H-bonds. DSP trapped both di-carbonyls and reduced CML-level, where new adducts such as epicatechin-mono/MGO-GO, were identified. 500 µg mL−1 DSP reduced OH-induced fluorescence intensity of AGEs by 43.32 % in BSA-G/Fe2+/H 2 O 2. Carbon black nanoparticles (CBNPs) and cadmium (Cd) are major components of various air pollutants and cigarette smoke. Autophagy and inflammation both play critical roles in understanding the toxicity of particles and their components, as well as maintaining body homeostasis. However, the effects and mechanisms of CBNPs and Cd (CBNPs-Cd) co-exposure on the human respiratory system remain unclear. In this study, a CBNPs-Cd exposure model was constructed to explore the respiratory toxicity and combined mechanism of these chemicals on the autophagy-lysosome pathway in the context of respiratory inflammation. Co-exposure of CBNPs and Cd significantly increased the number of autophagosomes and lysosomes in human bronchial epithelial cells (16HBE) and mouse lung tissues compared to the control group, as well as the groups exposed to CBNPs and Cd alone. Autophagic markers, LC3II and P62 proteins, were up-regulated in 16HBE cells and mouse lung tissues after CBNPs-Cd co-exposure. However, treatment with Cq inhibitor (an indicator of lysosomal acid environment) resulted in a substantial decreased co-localization fluorescence of LC3 and lysosomes in the CBNPs-Cd combination group compared with the CBNPs-Cd single and control groups. No difference in LAMP1 protein expression was observed among the exposed groups. Adding 3 MA alleviated inflammatory responses, while applying the Baf-A1 inhibitor aggravated inflammation both in vitro and in vivo following CBNPs-Cd co-exposure. Factorial analysis showed no interaction between CBNPs and Cd in their effects on 16HBE cells. We demonstrated that co-exposure to CBNPs-Cd increases the synthesis of autophagosomes and regulates the acidic environment of lysosomes, thereby inhibiting autophagy-lysosome fusion and enhancing the inflammatory response in both 16HBE cells and mouse lung. The problem of surface icing poses a serious threat to people's economy and safety, especially in the fields of aerospace, wind power generation and circuit transmission. Super-hydrophobic has excellent anti-icing performance, so it has been widely studied. As the most promising anti-icing technology, superhydrophobic anti-icing surface should not only be simple to prepare, but also have excellent comprehensive performance, which can meet the anti-icing task under harsh working conditions and long-term durability. This paper summarizes the basic performance requirements of superhydrophobic surface for anti-icing operation, and then summarizes the preparation methods and existing problems of superhydrophobic surface in recent years. Background: Lung cancer has high morbidity and mortality worldwide with non-small cell lung cancer (NSCLC) accounting for 85% of the cases. Therapies for lung cancer have relatively poor outcomes and further improvements are required. Circular RNAs have been reported to participate in the occurrence and progression of cancer. Information on the functions and mechanism of circRNAs in lung cancer is limited and needs more exploration. Methods: We detected expression of genes and proteins by qPCR and western blot. Function of circSATB2 was investigated using RNA interference and overexpression assays. Location of circSATB2 was assessed by fluorescence in situ hybridization (FISH). Interaction of circSATB2, miR-326 and FSCN1 was confirmed by dual-luciferase reporter assay. Results: Data from the investigation showed that circSATB2 was highly expressed in NSCLC cells and tissues. circSATB2 positively regulated fascin homolog 1, actin-bundling protein 1 (FSCN1) expression via miR-326 in lung cancer cells. Furthermore, circSATB2 can be transferred by exosomes and promote the proliferation, migration and invasion of NSCLC cells, as well as induce abnormal proliferation in normal human bronchial epithelial cells. Also, circSATB2 was highly expressed in serumal exosomes from lung cancer patients with high sensitivity and specificity for clinical detection and was related to lung cancer metastasis. Conclusions: circSATB2 participated in the progression of NSCLC and was differentially expressed in lung cancer tissue and serumal exosomes. circSATB2 may be potential biomarker for the diagnosis of NSCLC. Mesenchymal stem cell (MSC)-based therapy was an ideal strategy for therapeutic angiogenesis in ischemic tissues. However, limitations such as heterogeneous proangiogenic nature and low cell survival following transplantation is a key challenge for clinical applications. Biofunctional materials such as extracellular matrix (ECM) and hydrogels are considered with great promise for improved therapeutics by constructing a supportable 3D microenvironment niche for transplanted cells. Here, we employ the RGD (Arg-Gly-Asp)-modified self-assembling D-form peptide hydrogel (Nap-DFDFKGRGD) to serve as the co-transplantation platform for human placental MSCs (hP-MSCs). Our data showed that this D-form hydrogel possessed excellent biocompatibility and biostability, exerting an anti-apoptosis capacity and improved cell viability in vitro. Furthermore, we found that the self-assembled hydrogel improved cell survival, as well as promoted collateral vessel revascularization by bioluminescence imaging (BLI) and micro-CT imaging analysis. Further study confirmed significant up-regulation of proangiogenic cytokines of hP-MSCs in vivo. These results demonstrated the improvements of therapeutic efficacy of hP-MSCs in terms of tissue regeneration and functional recovery in the murine hindlimb ischemia (HLI) model. Benzo[ a ]pyrene (B[ a ]P) is a class I carcinogen and hazardous environmental pollutant with genetic toxicity. Understanding the molecular mechanisms underlying genetic deterioration and epigenetic alterations induced by environmental contaminants may contribute to the early detection and prevention of cancer. However, the role and regulatory mechanisms of circular RNAs (circRNAs) in the B[ a ]P-induced DNA damage response (DDR) have not been elucidated. In this study, human bronchial epithelial cell lines (16HBE and BEAS-2B) were exposed to various concentrations of B[ a ]P, and BALB/c mice were treated with B[ a ]P intranasally. B[ a ]P exposure was found to induce DNA damage and upregulate circular RNA hsa_circ_0057504 (circ_0057504) expression in vitro and in vivo. In addition, B[ a ]P upregulated TMEM194B mRNA and circ_0057504 expression through inhibition of DNA methyltransferase 3 alpha (DNMT3A) expression in vitro. Modulation (overexpression or knockdown) of circ_0057504 expression levels using a lentiviral system in human bronchial epithelial cells revealed that circ_0057504 promoted B[ a ]P-induced DNA damage. RNA pull-down and western blot assays showed that circ_0057504 interacted with non-POU domain-containing octamer-binding (NONO) and splicing factor proline and glutamine rich (SFPQ) proteins and regulated formation of the NONO-SFPQ protein complex. Thus, our findings indicate that circ_0057504 acts as a novel regulator of DNA damage in human bronchial epithelial cells exposed to B[ a ]P. The current study reveals novel insights into the role of circRNAs in the regulation of genetic damage, and describes the effect and regulatory mechanisms of circ_0057504 on B[ a ]P genotoxicity. Exposure to high concentrations of copper is associated with pulmonary inflammation and chronic respiratory disease (CRD). Epigenetic modulation of noncoding RNAs contributes to the development of several CRDs. It is unknown whether epigenetic modulation is involved in copper mediated pulmonary inflammation and CRD. We conducted a case-control study of 101 CRD cases and 161 control subjects in Shijiazhuang, China, and evaluated circRNAs and cytokine levels (IL-6 and IL-8) by qPCR and ELISA. Urinary copper concentration was determined by inductively coupled plasma mass spectrometry. Linear mixed models and generalized linear mixed models were used to assess the associations of circRNAs with CRD, urinary copper, and cytokines. We exposed the human bronchial epithelial cell line, 16HBE, to copper and assessed the functional role of a circRNA, circ_0008882, by RNA overexpression. Cellular location of circ_0008882 was assessed by separation of nuclear and cytoplasmic RNAs. Nine circRNAs were associated with an increased risk for CRDs, while the relative expression of circ_0008882 was decreased after copper exposure in vitro and in vivo. Copper exposure stimulated 16HBE cells to release proinflammatory IL-6 and IL-8. The release of the cytokines was inhibited by overexpression of circ_0008882. These results suggest a role for circ_0008882 in the regulation of CRD associated inflammation following copper exposure. Advanced glycation end-products (AGEs), which instigate many disorders, are mostly mediated by dicarbonyl rearrangements. We studied the corresponding mechanisms of the anti-glycation effects of two anthocyanins purified from mulberry fruits, namely cyanidin 3-glucoside (C3G) and cyanidin 3-rutinoside (C3R), on glycated β-lactoglobulins (β-Lg). Both mulberry anthocyanins (MAs) inhibited the AGEs-formation in a dose-dependent manner, but the effect of C3R was significantly stronger than that of C3G (p < 0.05). MAs inhibited AGEs-formation by selectively trapping dicarbonyls, especially glyoxal. The UPLC-ESI-Q-TOF-MS results characterized that C3R formed mono- and di-glyoxal adducts, where C3G only created di-glyoxal adducts. Additionally, C3R could directly interact with some of the glycation sites of β-Lg. Overall, GO-trapping and β-Lg-MAs covalent/noncovalent binding are disclosed as the key mechanisms of the anti-AGEs activity of MAs on β-Lg, which could be valorised as effectual AGEs inhibitors in proteins-rich matrices. Circular RNAs are widely expressed in eukaryotic cells and associated with cancer. However, limited studies to date have focused on the potential role of circRNAs in progression of lung cancer. Data from the current investigation showed that circRNA 100146 is highly expressed in non-small cell lung cancer (NSCLC) cell lines and the chemically induced malignant transformed bronchial cell line, 16HBE-T, as well as 40 paired tissue samples of NSCLC. Suppression of circRNA 100146 inhibited the proliferation and invasion of cells and promoted apoptosis. Furthermore, circRNA 100146 could interact with splicing factors and bind miR-361-3p and miR-615-5p to regulate multiple downstream mRNAs. Our collective findings support a role of circRNA 100146 in the development of NSCLC and further demonstrate endogenous competition among circRNA 100146, SF3B3 and miRNAs, providing novel insights into the mechanisms underlying non-small cell lung cancer. circNOL10 is a circular RNA expressed at low levels in lung cancer, though its functions in lung cancer remain unknown. Here, the function and molecular mechanism of circNOL10 in lung cancer development are investigated using in vitro and in vivo studies, and it is shown that circNOL10 significantly inhibits the development of lung cancer and that circNOL10 expression is co‐regulated by methylation of its parental gene Pre‐NOL10 and by splicing factor epithelial splicing regulatory protein 1 (ESRP1). circNOL10 promotes the expression of transcription factor sex comb on midleg‐like 1 (SCML1) by inhibiting transcription factor ubiquitination and thus also affects regulation of the humanin (HN) polypeptide family by SCML1. circNOL10 also affects mitochondrial function through regulating the humanin polypeptide family and affecting multiple signaling pathways, ultimately inhibiting cell proliferation and cell cycle progression, and promoting the apoptosis of lung cancer cells, thereby inhibiting lung cancer development. This study investigates the functions and molecular mechanisms of circNOL10 in the development of lung cancer and reveals its involvement in the transcriptional regulation of the HN polypeptide family by SCML1. The results also demonstrate the inhibitory effect of HN on lung cancer cells growth. These findings may identify novel targets for the molecular therapy of lung cancer.
