9. Characterization and pathogenicity of Alternaria species associated with leaf spot on Plantago lanceolata in Sichuan Province, China.

Abstract

Plantago lanceolata has been increasingly cultivated in recent years as a forage crop due to its high feeding quality, good adaptability and medicinal benefits. Between 2015 and 2023, Alternaria leaf spot (ALS), appearing as reddish‐brown necrotic spots with a greyish white to pale brown centre, was commonly observed in two forage germplasm nurseries of P. lanceolata of Sichuan Province, China. Initially, 139 Alternaria strains were isolated from leaf lesions, and then identified using the identity of rDNA internal transcribed spacer (ITS) sequences in the NCBI database and the similarity of morphological characteristics. By pure culture technique, six representative isolates were finally obtained for the present study. Based on both multilocus phylogeny (SSU, LSU, ITS, gapdh, rpb2, tef1, endoPG, OPA10‐2, alt a 1, KOG1058 and KOG1077) and morphology, four Alternaria species were identified: three new records of A. alternata, A. arborescens species complex and A. gossypina on this host in China, belonging to section Alternaria, and a new species of section Porri, namely, A. lanceolatae sp. nov. Pathogenicity tests by inoculation on both detached leaves and whole plants revealed that A. lanceolatae was the most important causal agent of ALS, while the other three species were nonpathogenic to P. lanceolata. This study characterized a novel pathogenic Alternaria species of section Porri from P. lanceolata in China and could contribute as useful research for possible ALS management. In the first part of the experiment, rats with CIP were divided into the immunofluorescence group and the coimmunoprecipitation (Co-IP) group (n = 6). Rats in the immunofluorescence group were injected with the retrograde tracer CB conjugated with Alexa Fluor 594 into the lateral ventricle two days before the injection of CFA into the plantar surface of the left paw. Three days later, rats that exhibited hyperalgesia were perfused, and their brains were extracted and used for double immunofluorescence staining of the CSF-contacting nucleus. Rats in the Co-IP group were anesthetized and dissected 3 days after CFA injection, and fresh brain segments containing the CSF-contacting nucleus were collected for Co-IP to assess the colocalization of TRPV1 and GABAB1 in the CSF-contacting nucleus (n = 6). In the second part of the experiment, SD rats were divided into the normal saline group (control group) and the CFA group. Fresh CSF-contacting nucleus-containing tissues were collected for Western blot analysis 3 days after plantar injection to observe the changes in TRPV1 and GABAB1 expression in the CSF-contacting nucleus.Results: TRPV1 and GABAB1 were co-expressed in the CSF-contacting nucleus in rats with CIP, and their expression was upregulated.Conclusion: TRPV1 and GABAB1 in the CSF-contacting nucleus are jointly involved in CIP in rats, and there is a direct or indirect link between TRPV1 and GABAB1.Keywords: cerebrospinal fluid-contacting nucleus, TRPV1 receptor, GABAB1 receptor, chronic inflammatory pain

Therefore, this study was carried out for finding research trends and fronts in this field using Citespace and VOSviewer.Methods: Publications about acupuncture therapy for LDH were extracted from the Web of Science database and PubMed with an unlimited time frame. A bibliometric analysis and visualization of results was conducted using CiteSpace 6.1.R3 and VOSviewer 1.6.18 on the information of the annual publication, countries, journals, institutions, authors, references, and keywords.Results: A total of 127 publications were included, and the number of publications had increased noticeably over the past 30 years and reached a peak in the past 3 years. The most productive country with the most publications was China, whose Medical University was the institution with the highest volume of publications. The most productive author was Chen Rixin, while the most-cited author was Kreiner DS. Chinese Acupuncture and Moxibustion was the journal with the most publications, and Spine Journal was the most frequently cited journal. 47. Advances in research on microbial leaching of uranium

Abstract

With the increasing demand of natural uranium, the exploitation of low-grade or complex uranium ores has been attracting more and more attention. Biological leaching is one of the important techniques for exploitation of such uranium resources. At present, heap leaching and in-situ leaching are the main processes for uranium recovery. A number of laboratory experiments and field trials show that the microbial leaching is a feasible technique for industrial production of uranium from low-grade or refractory ores. The tolerance of microorganisms which have been mutated and acclimatised can be improved obviously and it can be adapted to high fluorine uranium ores, low temperature and high acidity environments. For example, the acclimatised bacteria can grow well in mining tailings with fluorine content of 2-3.98 g/l, under the conditions of a low temperature of 8 degrees C and in a solution with pH 1.2-2.0 or even in a higher acidity solution (pH 0.6). Acclimatisation and induction are common technical ways to improve the adaptability of bacteria species to a leaching environment and indigenous bacteria often have better adaptability to native mining areas. The results also show that there is a synergistic effect between mixed strains and the leaching effect is better than that of a single strain. The indirect action of microbial leaching on uranium ore is clear, but the view on direct action of microorganisms on uranium ore is still controversial. The mechanism of uranium leaching by microorganism, the study of tolerance, the breeding of bacteria and research on the practical application of microbial leaching uranium technology were reviewed, especially in the past 20 years.
