The majority of the melanocytic neoplasms are considered malignant and highly metastatic. However, a subset of the melanocytic tumors has a more favorable prognosis and the identification of precise prognostic markers for this neoplasm may be useful to guide treatment. The collagen architecture and density have been shown to correlate with tumor progression in human breast cancer and canine mast cell tumors. The purpose of the present study was to investigate the prognostic value of the intratumoral collagen index (ICI) as an indicator of postsurgical survival and its relation with other prognostic markers for canine oral melanomas (OMs). Twenty-two cases were tested for intratumoral collagen density using Masson's trichrome stain and morphometry. No differences were found between dogs regarding survival. The ICI was not correlated with proliferative activity or nuclear atypia. The results presented herein indicate that the quantity of intratumoral collagen in canine OMs is not an efficient indicator of postsurgical survival. Complementary studies about the expression and activity of enzymes that are capable of degrading extracellular matrix (ECM) components are necessary. [ABSTRACT FROM AUTHOR]

Concomitantemente, observa-se que doenças associadas à longevidade, como o câncer, tem se tornado cada vez mais frequentes. Diante do fato de que atualmente os animais de companhia têm desempenhado papéis de membros da família, é natural que seus tutores temam a ocorrência de distúrbios neoplásicos e, experimentem sensações de medo, angústia, ansiedade entre outras, tanto no momento da confirmação do diagnóstico de câncer quanto no momento da indicação do tratamento, especialmente se a quimioterapia antineoplásica fizer parte do protocolo terapêutico preconizado. Neste estudo, tutores de pacientes com câncer responderam a dois questionários, de avaliação psicológica padronizados (HADS e BRUMS), bem como outros dois questionários para avaliação de suas percepções com relação ao diagnóstico e tratamento do câncer. As entrevistas foram feitas em 2 momentos: (1) antes do início da terapia e (2) ao longo do tratamento. O objetivo desta pesquisa foi avaliar o perfil psicológico e as percepções dos tutores de pacientes com câncer com relação ao diagnóstico e tratamento de seu pet. Dos 103 tutores que participaram deste estudo, 92% descreveram seu cão/gato como membro da família. Esperança (93%), tristeza (77%) e medo (73%) foram os principais sentimentos relatados no momento do diagnóstico. Em 55% dos casos, estes afirmaram sentir esperança com relação à terapia e 65% deles descreveram os efeitos adversos como seus maiores medos frente ao tratamento. Apenas 31% dos efeitos adversos esperados foram efetivamente observados e 78% dos tutores reportaram qualidade de vida boa/excelente durante a tratamento. O mesmo foi considerado benéfico por 82% dos tutores avaliados e 89% sentiam-se felizes por tê-lo feito. Antes do tratamento 54% dos tutores apresentavam sinais de ansiedade. Contudo, apenas 28% dos mesmos ainda os descreviam ao longo do tratamento, havendo associação da redução da ansiedade com a menor ocorrência de efeitos adversos bem como com a melhora da qualidade de vida dos pacientes durante o tratamento. Constatou-se que a receptividade por parte dos tutores, frente ao tratamento instituído neste pacientes, foi positiva, visto que inicialmente, a maioria dos entrevistados descreveram sinais de ansiedade, principalmente relacionados a expectativa dos efeitos adversos, bem como a queda da qualidade de vida durante o tratamento. Sendo o sofrimento do pet diante dos efeitos adversos e a perda do mesmo em decorrência da terapia, os principais medos apontados pelos tutores entrevistados na presente pesquisa. As the bonds between dogs, cats and humans become tighter, and therefore the care towards pets, we may observe that these species have shown longer life spans. Concurrently, we have observed that illnesses related to longevity, such as cancer, have become more frequent. Given that, nowadays, pets often play the role of family members, it is natural that their owners fear the occurrence of neoplastic diseases and suffer from fear, anxiety and similar feelings both at the diagnosis and during the early stages of the treatment, specially if antineoplastic chemotherapy is part of the recommended therapeutic protocol. Owners of cancer patients answered two standardized psychological questionnaires (HADS and BRUMS), as well as two other questionnaires to assess their perceptions regarding the diagnosis and treatment of cancer. The interviews happened in two moments (1) before the beginning of the therapy and, (2) in the middle of the treatment. The goals of this study were to evaluate the psychological profile and perceptions of tutors of cancer patients regarding the diagnosis and treatment of their pet. 103 clients participated in this survey, 92% described their dog/cat as a family member. Hope (93%), sadness (77%) and fear (73%) were the main feelings reported at the time of diagnosis. In 55% of the cases, they stated they were feeling hopeful about the therapy and 65% of them described the adverse effects as their greatest fears. Only 31% of expected adverse effects were observed and 78% of those owners reported good/excellent quality of life during the treatment. It was considered beneficial by 82% of the evaluated owners and 89% were feeling happy for choose it. Before the treatment 54% of these clients had signs of anxiety. In contrast, only 28% of those still described them during the treatment. There was an association between the reduction of anxiety signs and the lower occurrence of adverse effects, as well as the improvement of patients' quality of life during treatment. In conclusion, there was a positive balance according to the tutors regarding the treatment of their dog cat. Initially, most of these described signs of anxiety, mainly related to the expectation of adverse effects, as well as the decrease of the quality of life during the treatment. The pet's suffering due to adverse effects and the lost of him/her due to the therapy, were the main fears highlighted by the tutors interviewed in the present survey.

10. Analysis of VEGFR-2 and PDGFR-β expression in canine splenic hemangiosarcoma to identify drug repositioning candidates.

- Abstract
Splenic tumors are very common in dogs, and canine hemangiosarcoma (HSA) is one of the most important malignant splenic tumors. Surgery followed by chemotherapy (anthracycline-based protocols) is recommended for treating canine HSA; however, patients still do not achieve long-term survival. Therefore, this research aimed to assess vascular endothelial growth factor receptor-2 ( VEGFR-2 ) and platelet-derived growth factor receptor-β ( PDGFR-β ) gene expression in formalin-fixed tissues, evaluate the quality of mRNA for quantitative polymerase chain reaction (qPCR) analysis and identify drug repositioning candidates based on VEGFR-2 and PDGFR-β. qPCR analysis identified the relative expression of heterogeneous VEGFR-2 and PDGFR-β, with samples showing no transcripts or very low expression and those with higher relative quantification for both genes. We then used immunohistochemistry to correlate the relative quantification of VEGFR-2 and PDGFR-β transcripts with respective higher protein expression to validate our results. In the next step, we evaluated drug repositioning candidates and identified small molecule inhibitors (i.e. sorafenib) and natural compounds (curcumin and resveratrol) with the ability to block VEGFR-2 and PDGFR-β genes. Overall, our results indicated that VEGFR-2 and PDGFR-β expression is highly variable among canine HSA samples and different drugs can block the expression of both genes.
