Abstract

Currently, there is no widely accepted criterion for judging the efficacy of CRT treatment. Patients underwent CRT implantation have most often been classified as either responders or non-responders based on the occurrence or absence of reverse remodelling following CRT. Based on this definition, nearly one-third of patients are considered as non-responders with a less favourable prognosis compared to responders. Recent recommendations have revised the previous terminologies. Accordingly, the CRT non-responder definition have-been re-defined as those who deteriorate (progressors) and remain stable for years (non-progressor) after CRT implantation. Considering the progressive nature of HF, non-progression could still represent a response to the device therapy and thereby classifying these patients as non-responders might be misleading. The background of the suboptimal CRT response is multifactorial, requiring a multidisciplinary approach for improving the effectiveness of CRT treatment. Therefore, we care must be taken on the appropriate patient selection and optimisation of the medications prior to implantation, currently available technical options and innovations during the intervention, device optimisation post-implantation, as well as on the management of comorbidities.

15. How codependency affects dyadic coping, relationship perception and life satisfaction

Abstract

In this study, we interpret codependency as a dysfunctional pattern of relating to others, and based on this approach, we hypothesized an association with negative forms of dyadic coping, relationship problems and life satisfaction. A total of 246 Hungarian participants (167 females, 79 males), aged 18-72 years (M = 35.3, SD = 11.6) completed our online survey including measures of codependency, dyadic coping, perceptions of relationship, and life satisfaction. In our cross-sectional research, the Spann-Fischer Codependency Scale (SF-CDS), the Dyadic Coping Inventory (DCI), the Shortened Marital Stress Scale (MSS-R), and the Satisfaction With Life Scale (SWLS) were used. Correlational and pathway analyzes were applied to confirm our hypotheses. Codependency was associated with negative dyadic coping, while we found no measurable influence on positive dyadic coping. Individuals with higher codependency rated both their own and their partner's negative dyadic coping more pronounced, while at the same time they characterized their relationships as more problematic. Structural Equation Modelling proved that codependent attitudes, along with the emergence of negative dyadic coping forms and perception of relationship problems, reduce a person's life satisfaction. Overall, it can be stated that the more codependent the participants were, the more negative their own and partner's behaviour was perceived in stressful situations and the more problematic their intimate relationship was found to be. Abstract

The Narcissistic Personality Inventory is a widely used measure of narcissism; however, several different conflicting factor structures have been proposed. The present study aimed to untangle those differences by offering a bifactor model of narcissism with an underlying general narcissism factor and several specific factors. Our objective was to estimate the variance explained by the general and the specific factors. Therefore, we applied a set of confirmatory factor analyses on three independent studies (N = 791; N = 319 and N = 237), and also tested the construct validity of these factors with other contemporary measures of grandiose and vulnerable narcissism, big five personality traits, explicit self-esteem, gender and well-being. Our results suggest that the bifactor model with three specific factors yielded acceptable fit to our data in all of the different single-stimulus response formats and languages used in our studies and modelling three specific factors offered more insight into the adaptive and maladaptive characteristics of narcissism. 36. Treatments with Diquat Reveal the Relationship between Protein Phosphatases (PP2A) and Oxidative Stress during Mitosis in Arabidopsis thaliana Root Meristems.

Abstract

Reversible protein phosphorylation regulates various cellular mechanisms in eukaryotes by altering the conformation, activity, localization, and stability of substrate proteins. In Arabidopsis thaliana root meristems, histone post-translational modifications are crucial for proper cell division, and they are also involved in oxidative stress signaling. To investigate the link between reactive oxygen species (ROS) and mitosis, we treated various Arabidopsis genotypes, including wild-types and mutants showing dysfunctional PP2A, with the ROS-inducing herbicide diquat (DQ). Studying the c3c4 double catalytic subunit mutant and fass regulatory subunit mutants of PP2A provided insights into phosphorylation-dependent mitotic processes. DQ treatment reduced mitotic activity in all genotypes and caused early mitotic arrest in PP2A mutants, likely due to oxidative stress-induced damage to essential mitotic processes. DQ had a minimal effect on reversible histone H3 phosphorylation in wild-type plants but significantly decreased phospho-histone H3 levels in PP2A mutants. Following drug treatment, the phosphatase activity decreased only in the stronger phenotype mutant plants (fass-5 and c3c4). Abstract

Turkey oak (Quercus cerris L.) is a deciduous tree species of the genus Quercus section Cerris (Fagaceae), widely distributed in Europe and Asia Minor. Throughout its vast distribution range, the species carries high phenotypic and genetic variability. Due to its high drought tolerance, Turkey oak may have great potential to be a key tree species in Central and Western Europe under climate change conditions. However, more detailed information on its phylogeny, phylogeography, phenotypic, and genetic variability is still needed for a more careful and reliable assessment of the species' adaptation potential. To this end, based on 41 reviewed articles, we collected detailed information to support the evaluation of Turkey oak's adaptation potential under climate change. In the reviewed articles, we observed a significantly lower number of genetic studies of the members of the section Cerris (especially the Euro-Mediterranean species) relative to the subgenus Quercus. We also identified research gaps with respect to the functional and population genetics of the species that should be addressed in the future. Nevertheless, the different evolutionary background and adaptation strategies of Turkey oak as compared to Central European white oaks, as well as its high phenotypic and genetic variability, may indeed represent a great potential for this species to support the climate adaptation of forestry in Central and Western Europe. Abstract

Primary demyelinating disorders of the central nervous system (CNS) include multiple sclerosis and the orphan conditions neuromyelitis optica spectrum disorder (NMOSD) and myelin oligodendrocyte glycoprotein IgG-associated disease (MOGAD). Curative technologies under development aim to selectively block autoimmune reactions against specific autoantigens while preserving the responsiveness of the immune system to other antigens. Our analysis focused on target patient selection for such developments, carefully considering the relevant clinical, regulatory, and market-related aspects. We found that the selection of patients with orphan conditions as target populations offers several advantages. Treatments for orphan conditions are associated with limited production capacity, qualify for regulatory incentives, and may require significantly shorter and lower-scale clinical programs. Furthermore, they may meet a higher acceptable cost-effectiveness threshold in order to compensate for the low numbers of patients to be treated. Finally, curative technologies targeting orphan indications could enter less competitive markets with lower risk of generic price erosion and would benefit from additional market protection measures available only for orphan products. These advantages position orphan conditions and subgroups as the most attractive target indications among primary demyelinating disorders of the CNS. The authors believe that after successful proof-of-principle demonstrations in orphan conditions, broader autoimmune patient populations may also benefit from the success of these pioneering developments. Abstract

Pertuzumab (Perjeta®), a humanized antibody binding to the dimerization arm of HER2 (Human epidermal growth factor receptor-2), has failed as a monotherapy agent in HER2 overexpressing malignancies. Since the molecular interaction of HER2 with ligand-bound EGFR (epidermal growth factor receptor) has been implied in mitogenic signaling and malignant proliferation, we hypothesized that this interaction, rather than HER2 expression and oligomerization alone, could be a potential molecular target and predictor of the efficacy of pertuzumab treatment. Therefore, we investigated static and dynamic interactions between HER2 and EGFR molecules upon EGF stimulus in the presence and absence of pertuzumab in HER2+ EGFR+ SK-BR-3 breast tumor cells using Förster resonance energy transfer (FRET) microscopy and fluorescence correlation and cross-correlation spectroscopy (FCS/FCCS). The consequential activation of signaling and changes in cell proliferation were measured by Western blotting and MTT assay. The autocorrelation functions of HER2 diffusion were best fitted by a three-component model corrected for triplet formation, and among these components the slowly diffusing membrane component revealed aggregation induced by EGFR ligand binding, as evidenced by photon-counting histograms and co-diffusing fractions. This aggregation has efficiently been prevented by pertuzumab treatment, which also inhibited the post-stimulus interaction of EGFR and HER2, as monitored by changes in FRET efficiency. Overall, the data demonstrated that pertuzumab, by hindering post-stimulus interaction between EGFR and HER2, inhibits EGFR-evoked HER2 aggregation and phosphorylation and leads to a dose-dependent decrease in cell proliferation, particularly when higher amounts of EGF are present. Consequently, we propose that EGFR expression on HER2-positive tumors could be taken into consideration as a potential biomarker when predicting the outcome of pertuzumab treatment. Abstract

The importance of highly monitored and analyzed processes, linked by information systems such as knowledge graphs, is growing. In addition, the integration of operators has become urgent due to their high costs and from a social point of view. An appropriate framework for implementing the Industry 5.0 approach requires effective data exchange in a highly complex manufacturing network to utilize resources and information. Furthermore, the continuous development of collaboration between human and machine actors is fundamental for industrial cyber-physical systems, as the workforce is one of the most agile and flexible manufacturing resources. This paper introduces the human-centric knowledge graph framework by adapting ontologies and standards to model the operator-related factors such as monitoring movements, working conditions, or collaborating with robots. It also presents graph-based data querying, visualization, and analysis through an industrial case study. The main contribution of this work is a knowledge graph-based framework that focuses on the work performed by the operator, including the evaluation of movements, collaboration with machines, ergonomics, and other conditions. In addition, the use of the framework is demonstrated in a complex use case based on an assembly line, with examples of resource allocation and comprehensive support in terms of the collaboration aspect between shop-floor workers.
