Abstract Background The C-terminal-binding protein 1/brefeldin A ADP-ribosylation substrate (CtBP1/BARS) acts both as an oncogenic transcriptional co-repressor and as a fission inducing protein required for membrane trafficking and Golgi complex partitioning during mitosis, hence for mitotic entry. CtBP1/BARS overexpression, in multiple cancers, has pro-tumorigenic functions regulating gene networks associated with “cancer hallmarks” and malignant behavior including: increased cell survival, proliferation, migration/invasion, epithelial-mesenchymal transition (EMT). Structurally, CtBP1/BARS belongs to the hydroxyacid-dehydrogenase family and possesses a NAD(H)-binding Rossmann fold, which, depending on ligands bound, controls the oligomerization of CtBP1/BARS and, in turn, its cellular functions. Here, we proposed to target the CtBP1/BARS Rossmann fold with small molecules as selective inhibitors of mitotic entry and pro-tumoral transcriptional activities. Methods Structured-based screening of drug databases at different development stages was applied to discover novel ligands targeting the Rossmann fold. Among these identified ligands, N-(3,4-dichlorophenyl)-4-{[(4-nitrophenyl)carbamoyl]amino}benzenesulfonamide, called Comp.11, was selected for further analysis. Fluorescence spectroscopy, isothermal calorimetry, computational modelling and site-directed mutagenesis were employed to define the binding of Comp.11 to the Rossmann fold. Effects of Comp.11 on the oligomerization state, protein partners binding and pro-tumoral activities were evaluated by size-exclusion chromatography, pull-down, membrane transport and mitotic entry assays, Flow cytometry, quantitative real-time PCR, motility/invasion, and colony assays in A375MM and B16F10 melanoma cell lines. Effects of Comp.11 on tumor growth in vivo were analyzed in mouse tumor model. Results We identify Comp.11 as a new, potent and selective inhibitor of CtBP1/BARS (but not CtBP2). Comp.11 directly binds to the CtBP1/BARS Rossmann fold affecting the oligomerization state of the protein (unlike other known CtBPs inhibitors), which, in turn, hinders interactions with relevant partners, resulting in the inhibition of both CtBP1/BARS cellular functions: i) membrane fission, with block of mitotic entry and cellular secretion; and ii) transcriptional pro-tumoral effects with significantly hampered proliferation, EMT, migration/invasion, and colony-forming capabilities. The combination of these effects impairs melanoma tumor growth in mouse models. Conclusions This study identifies a potent and selective inhibitor of CtBP1/BARS active in cellular and melanoma animal models revealing new opportunities to study the role of CtBP1/BARS in tumor biology and to develop novel melanoma treatments.

En este artículo se analiza la escritura como un recurso de producción científico – académica, en la formación de profesionales de la Psicología y la salud mental. Partiremos de la noción de ethos discursivo, a fin de analizar las condiciones precisas para la formulación de propuestas de investigación en estudiantes universitarios/as. Para ello, una experiencia didáctico -pedagógica que contempló la implementación de encuestas, con dos muestras de trabajo: en primer lugar, una muestra de 120 estudiantes de la carrera de Licenciatura en Psicología, de la Facultad de Psicología de La Plata; en segundo lugar, una muestra de 24 estudiantes de la misma carrera en la Sede de la localidad de Chivilcoy. La encuesta, como técnica de recolección de datos, se administró en los espacios de trabajos prácticos (durante el período marzo – noviembre de 2023), a fin de relevar recursos necesarios para la planificación didáctico – pedagógica de la enseñanza de la metodología de la investigación, y al mismo tiempo delimitar las estrategias necesarias para fortalecer la escritura académica de los/as estudiantes. En el presente artículo contemplaremos la metodología, en tanto una disciplina científica en sí misma, en la que se conjugan aspectos epistemológicos propios de la Psicología, y aspectos técnicos para el relevamiento de datos. La especificidad epistemológica también se relaciona con los procesos de lectura y escritura académica, en primer lugar, a través de las características que Bajtín (1982) señala en torno al discurso académico – científico, cuyo principal objetivo es aportar información acerca de un objeto de estudio, en un campo determinado; en segundo lugar, abrevando en la definición de alfabetización académica que propone Paula Carlino (2013, 2017), la lectura y escritura académica contempla la especificidad disciplinaria. La autora toma tanto los desarrollos del movimiento “escribir a través del curriculum” (Bazerman et al., 2005; Russell, 2002. En Carlino, 2013), y los aportes en Australia de “enseñar escritura en contexto”, señalando la importancia de la función epistémica en la escritura académica. Se presentan entonces, las principales dificultades que refieren los/as estudiantes al momento de desarrollar la escritura académica mediante la elaboración de una propuesta de diseño de campo. Delimitaremos también los recursos adquiridos, en tanto competencias de alfabetización académica, por medio de estrategias de edición de los distintos momentos lógicos del diseño de una propuesta de investigación. En el artículo se proponen algunos recursos de trabajo para propiciar estrategias de lectura y escritura en el contexto áulico, con el fin de promover competencias de investigación científico - académico en el campo de las Ciencias Humanas, y la Psicología en particular.

Los movimientos sociales y los colectivos feministas remiten necesariamente a las lógicas de acción e identificación colectiva, en cuanto a los primeros incluyen en sus lógicas el argumento antipatriarcal, y en el caso de los segundos se organizan específicamente por los reclamos y reivindicaciones frente al patriarcado como un sistema de subordinación de las mujeres sustentado y legitimado en la desigualdad estructural. Consideramos al feminismo como un movimiento en el cual se presentan dos dificultades, la primera es el desafío de las retóricas del movimiento, y la segunda, las diferencias ideológicas que se plantean en el terreno de la política.

En este artículo analizaremos una experiencia de educación disidente en contexto de encierro punitivo. Se trata de una serie de talleres y grupos terapéuticos que se desarrollaron desde el año 2017 al año 2019 en la Unidad N.°45 de la localidad de Melchor Romero, en el marco del Programa Masculinidades disidentes: jóvenes y salud mental en cárceles, que cuenta con dos organizaciones de la sociedad civil copartícipes: Grupo de Estudios de Educación en Cárceles (GESEC) y la Asociación Civil de Familiares de Detenidos en Cárceles Federales (ACIFAD). La experiencia tuvo como destinatarios a varones jóvenes de 18 a 23 años de edad que participaron de una experiencia de trabajo que consistía en un primer momento de reflexiones recursivas sobre temáticas vinculadas con las masculinidades, y un segundo momento, de grupo terapéutico en donde la escritura del primer momento se inscribía en el so(gra)ma (cuerpo-lenguaje) de las masculinidades en disidencia (Colanzi, 2021). A los efectos de establecer un recorte específico, trabajaremos entonces con algunos interrogantes: ¿cuál es la posición de las masculinidades en cárceles frente a los lazos sexo-afectivos? ¿cómo se vivencias los lazos sexo-afectivos entre varones privados de libertad? ¿cómo los lazos sexo-afectivos operan en tanto una estrategia de cuidado amoroso y affidamento masculino entre varones? Palabras clave: masculinidades – cárceles – lazos sexo – afectivos – giro emocional – salud mental.

The Golgi complex has a central role in the secretory traffic. In vertebrate cells it is generally organized in polarized stacks of cisternae that are laterally connected by membranous tubules, forming a structure known as Golgi ribbon. The steady state ribbon arrangement results from a dynamic equilibrium between formation and cleavage of the membrane tubules connecting the stacks. This balance is of great physiological relevance as the unlinking of the ribbon during G2 is required for mitotic entry. A block of this process induces a potent G2 arrest of the cell cycle, indicating that a mitotic “Golgi checkpoint” controls the correct pre-mitotic segregation of the Golgi ribbon. Then, after mitosis onset, the Golgi stacks undergo an extensive disassembly, which is necessary for proper spindle formation. Notably, several Golgi-associated proteins acquire new roles in spindle formation and mitotic progression during mitosis. Here we summarize the current knowledge about the basic principle of the Golgi architecture and its functional relationship with cell division to highlight crucial aspects that need to be addressed to help us understand the physiological significance of the ribbon and the pathological implications of alterations of this organization.

This article analyzes writing as a resource for scientific-academic production, in the training of professionals in Psychology and mental health. We will start from the notion of discursive ethos, in order to analyze the precise conditions for the formulation of research proposals in university students. For this, a didactic-pedagogical experience that included the implementation of surveys, with two work samples: first, a sample of 120 students from the Bachelor's degree in Psychology, from the Faculty of Psychology of La Plata; Secondly, a sample of 24 students from the same career at the Headquarters in the town of Chivilcoy. The survey, as a data collection technique, was administered in the practical work spaces, in order to collect resources necessary for the didactic-pedagogical planning of teaching the research methodology, and at the same time delimit the necessary strategies to strengthen students' academic writing. In this article we will consider the methodology, as a scientific discipline in itself, in which epistemological aspects typical of Psychology are combined, and technical aspects for data collection. Epistemological specificity is also related to the processes of academic reading and writing, firstly, through the characteristics that Bakhtin (1982) points out about academicscientific discourse, whose main objective is to provide information about an object of study., in a given field; Secondly, based on the definition of academic literacy proposed by Paula Carlino (2013, 2017), academic reading and writing contemplates disciplinary specificity. The author takes both the developments of the "writing across the curriculum" movement (Bazerman et al., 2005; Russell, 2002. In Carlino, 2013), and the contributions in Australia of "teaching writing in context", pointing out the importance of The epistemic function in academic writing. The main difficulties that students refer to when developing academic writing through the development of a field design proposal are then presented. We will also delimit the acquired resources, as academic literacy skills, through editing strategies of the different logical moments of the design of a research proposal. The article proposes some work resources to promote reading and writing strategies in the classroom context, in order to promote scientific-academic research skills in the field of Human Sciences, and Psychology in particular. Intracellular mono-ADP-ribosyltransferases (mono-ARTs) catalyze the covalent attachment of a single ADP-ribose molecule to protein substrates, thus regulating their functions. PARP10 is a soluble mono-ART involved in the modulation of intracellular signaling, metabolism and apoptosis. PARP10 also participates in the regulation of the G1- and S-phase of the cell cycle. However, the role of this enzyme in G2/M progression is not defined. In this study, we found that genetic ablation, protein depletion and pharmacological inhibition of PARP10 cause a delay in the G2/M transition of the cell cycle. Moreover, we found that the mitotic kinase Aurora-A, a previously identified PARP10 substrate, is actively mono-ADP-ribosylated (MARylated) during G2/M transition in a PARP10-dependent manner. Notably, we showed that PARP10-mediated MARylation of Aurora-A enhances the activity of the kinase in vitro. Consistent with an impairment in the endogenous activity of Aurora-A, cells lacking PARP10 show a decreased localization of the kinase on the centrosomes and mitotic spindle during G2/M progression. Taken together, our data provide the first evidence of a direct role played by PARP10 in the progression of G2 and mitosis, an event that is strictly correlated to the endogenous MARylation of Aurora-A, thus proposing a novel mechanism for the modulation of Aurora-A kinase activity.

Un aspecto de la obra de Amparo Dávila (México 1928-2020) que no ha sido muy estudiado es el que tiene que ver con el horror gastronómico, la representación de la comida como fuente de abyección y de espanto. El primer libro de Amparo Dávila, Tiempo destrozado (1959), incluye dos cuentos en los que la comida y la cocina están codificadas en un registro gótico: «Alta cocina» y el relato homónimo «Tiempo destrozado». En estos cuentos la comida se vuelve extraña y repulsiva, atraviesa inquietantes metamorfosis y genera incertidumbre intelectual acerca de su naturaleza. En este ensayo me concentraré en «Alta cocina», que es el texto donde se desarrolla más la experiencia del horror alimenticio, para indagar la manera en que el cuento hace estallar la frontera entre lo humano y lo animal y pone en entredicho el binarismo civilización/barbarie. Asimismo, analizaré la forma en que un espacio históricamente asociado a la mujer como la cocina se presenta oscuro, siniestro y amenazante.

The Golgi Complex is the central hub in the endomembrane system and serves not only as a biosynthetic and processing center but also as a trafficking and sorting station for glycoproteins and lipids. In addition, it is an active signaling hub involved in the regulation of multiple cellular processes, including cell polarity, motility, growth, autophagy, apoptosis, inflammation, DNA repair and stress responses. As such, the dysregulation of the Golgi Complex-centered signaling cascades contributes to the onset of several pathological conditions, including cancer. This review summarizes the current knowledge on the signaling pathways regulated by the Golgi Complex and implicated in promoting cancer hallmarks and tumor progression.

The Golgi complex of mammalian cells is organized in a ribbon-like structure often closely associated with the centrosome during interphase. Conversely, the Golgi complex assumes a fragmented and dispersed configuration away from the centrosome during mitosis. The structure of the Golgi complex and the relative position to the centrosome are dynamically regulated by microtubules. Many pieces of evidence reveal that this microtubule-mediated dynamic association between the Golgi complex and centrosome is of functional significance in cell polarization and division. Here, we summarize findings indicating how the Golgi complex and the centrosome cooperate in organizing the microtubule network for the directional protein transport and centrosome positioning required for cell polarization and regulating fundamental cell division processes.

Helicobacter pylori, a dominant member of the gastric microbiota, shares co-evolutionary history with humans. This has led to the development of genetically distinct H. pylori subpopulations associated with the geographic origin of the host and with differential gastric disease risk. Here, we provide insights into H. pylori population structure as a part of the Helicobacter pylori Genome Project (HpGP), a multi-disciplinary initiative aimed at elucidating H. pylori pathogenesis and identifying new therapeutic targets. We collected 1011 well-characterized clinical strains from 50 countries and generated high-quality genome sequences. We analysed core genome diversity and population structure of the HpGP dataset and 255 worldwide reference genomes to outline the ancestral contribution to Eurasian, African, and American populations. We found evidence of substantial contribution of population hpNorthAsia and subpopulation hspUral in Northern European H. pylori. The genomes of H. pylori isolated from northern and southern Indigenous Americans differed in that bacteria isolated in northern Indigenous communities were more similar to North Asian H. pylori while the southern had higher relatedness to hpEastAsia. Notably, we also found a highly clonal yet geographically dispersed North American subpopulation, which is negative for the cag pathogenicity island, and present in 7% of sequenced US genomes. We expect the HpGP dataset and the corresponding strains to become a major asset for H. pylori genomics. The bacterium Helicobacter pylori, often found in the human stomach, can be classified into distinct subpopulations associated with the geographic origin of the host. Here, the authors provide insights into H. pylori population structure by collecting over 1,000 clinical strains from 50 countries and generating and analyzing high-quality bacterial genome sequences. El objetivo del artículo es problematizar la noción de cuidado, desde una dimensión política (Faur, 2014) en el contexto de encierro punitivo. Se presenta un análisis de entrevistas con diferentes actores abocados al desarrollado de intervenciones frente a las mujeres madres encarceladas con sus hijos/as en la Unidad N°33 de la localidad de Los Hornos, en la provincia de Buenos Aires. El ejercicio del cuidado en el caso de las mujeres encarceladas, cumple una función doble, por un lado el control informal (Gentile & Tabbush, 2013) del sistema patriarcal que asigna de manera obligatoria el ejercicio del mismo por parte de las mujeres, aún en el caso de las mujeres madres detenidas. Por otro lado, en función de las características del aumento de prisionización de las mujeres por delitos de tenencia de estupefacientes, se indaga de estas en la economía ilegal, y se intenta establecer algunas reflexiones sobre el mencionado delito y la posibilidad de conciliar estrategias de supervivencia económica y cuidado.

The Golgi complex (GC), in addition to its well-known role in membrane traffic, is also actively involved in the regulation of mitotic entry and progression. In particular, during the G2 phase of the cell cycle, the Golgi ribbon is unlinked into isolated stacks. Importantly, this ribbon cleavage is required for G2/M transition, indicating that a “Golgi mitotic checkpoint” controls the correct segregation of this organelle. Then, during mitosis, the isolated Golgi stacks are disassembled, and this process is required for spindle formation. Moreover, recent evidence indicates that also proper mitotic segregation of other organelles, such as mitochondria, endosomes, and peroxisomes, is required for correct mitotic progression and/or spindle formation. Collectively, these observations imply that in addition to the control of chromosomes segregation, which is required to preserve the genetic information, the cells actively monitor the disassembly and redistribution of subcellular organelles in mitosis. Here, we provide an overview of the major structural reorganization of the GC and other organelles during G2/M transition and of their regulatory mechanisms, focusing on novel findings that have shed light on the basic processes that link organelle inheritance to mitotic progression and spindle formation, and discussing their implications for tissue homeostasis and diseases.

BackgroundThe detection of Trypanosoma cruzi genetic material in clinical samples is considered an important diagnostic tool for Chagas disease. We have previously demonstrated that PCR using clot samples yields greater sensitivity than either buffy coat or whole blood samples. However, phenol-chloroform DNA extraction from clot samples is difficult and toxic. The objective of the present study was to improve and develop a more sensitive method to recover parasite DNA from clot samples for the diagnosis of Chagas disease.Methodology/principal findingsA total of 265 match pair samples of whole blood-guanidine (GEB) and clot samples were analyzed; 150 were from Chagas seropositive subjects. DNA was extracted from both whole blood-guanidine samples, using a previously standardized methodology, and from clot samples, using a newly developed methodology based on a combination of the FastPrep technique and the standard method for GEB extraction. A qPCR targeting the nuclear satellite sequences was used to compare the sample source and the extraction method. Of the 150 samples from Chagas positive individuals by serology, 47 samples tested positive by qPCR with DNA extracted by both GEB and clot, but an additional 13 samples tested positive only in DNA extracted from clot. No serology-negative samples resulted positive when tested by qPCR.ConclusionsThe new methodology for DNA extraction from clot samples improves the molecular diagnosis of Chagas disease.

The Golgi mitotic checkpoint couples Golgi inheritance with cell cycle transition, and regulates centrosomal recruitment of the mitotic kinase Aurora-A. Here the authors show that upon Golgi ribbon fragmentation in G2, Src phosphorylates Aurora-A at the Golgi, driving its localization to the centrosomes.

Um dos motivos é que essas empresas consideram os processos tradicionais extensos. Assim, normalmente, essas empresas são classificadas em nível 1 de maturidade do CMMI (Capability Maturity Model Integration). O objetivo deste artigo é apresentar uma instância do processo WAE (Web Application Extension), a fim de auxiliar especificamente as MPEs que desenvolvem software Web a alavancarem o nível de capacidade de seu processo de desenvolvimento de software. A instância foi validada durante sua aplicação no desenvolvimento de um software comercial com o apoio da Coordenadoria de Web da Unoeste e em outros três estudos de caso.

In Micro and Small Enterprises (MSE) of software, there is usuallynot a systematic software development process. One of the reasons is that these enterprises consider the traditional processes extensive. Thus, these enterprises are usually classified in level 1 of maturity of the CMMI (Capability Maturity Model Integration). The objective ofthis paper is to present a proposal for a software development process, in order to aid specifically MSEs that build Web software, to help them increase the capacity level of their processes. The process was validated during its application in development of software with the support of the Unoeste Web Coordination.

Many pathogenic bacteria utilize ADP-ribosylating toxins to modify and impair essential functions of eukaryotic cells. It has been previously reported that Neisseria meningitidis possesses an ADP-ribosyltransferase enzyme, NarE, retaining the capacity to hydrolyse NAD and to transfer ADP-ribose moiety to arginine residues in target acceptor proteins. Here we show that upon internalization into human epithelial cells, NarE gains access to the cytoplasm and, through its ADP-ribosylating activity, targets host cell proteins. Notably, we observed that these events trigger the disruption of the epithelial monolayer integrity and the activation of the apoptotic pathway. Overall, our findings provide, for the first time, evidence for a biological activity of NarE on host cells, suggesting its possible involvement in Neisseria pathogenesis.

BackgroundCongenital transmission is a major source of new Trypanosoma cruzi infections, and as vector and blood bank control continue to improve, the proportion due to congenital infection will grow. A major unanswered question is why reported transmission rates from T. cruzi-infected mothers vary so widely among study populations. Women with high parasite loads during pregnancy are more likely to transmit to their infants, but the factors that govern maternal parasite load are largely unknown. Better understanding of these factors could enable prioritization of screening programs to target women most at risk of transmission to their infants.Methodology/principal findingsWe screened pregnant women presenting for delivery in a large urban hospital in Bolivia and followed infants of infected women for congenital Chagas disease. Of 596 women screened, 128 (21.5%) had confirmed T. cruzi infection; transmission occurred from 15 (11.7%) infected women to their infants. Parasite loads were significantly higher among women who transmitted compared to those who did not. Congenital transmission occurred from 31.3% (9/29), 15.4% (4/26) and 0% (0/62) of women with high, moderate and low parasite load, respectively (χx2 for trend 18.2; pConclusions/significanceWe hypothesize that sustained vector-borne parasite exposure and repeated superinfection by T. cruzi may act as an immune booster, allowing women to maintain effective control of the parasite despite the down-regulation of late pregnancy.

Simple Summary: Mono-ADP-ribosylating PARP enzymes are involved in the regulation of several cellular pathways, including cell cycle. The mono-ADP-ribosyltransferase PARP10 is frequently amplified in different tumor types, and has been shown to promote in vitro cellular proliferation and in vivo tumoral growth. The aim of our study was to investigate the role of PARP10 in the regulation of G2/M transition. We found that PARP10 is involved in the mono-ADP-ribosylation of Aurora-A, an important mitotic kinase, during the G2/M transition. Moreover, PARP10-catalyzed modification enhances the kinase activity of Aurora-A. Consistently, the depletion of PARP10 affects the timely recruitment of Aurora-A on centrosomes and mitotic spindle, causing a delay in mitotic entry. Our discovery provides novel details in the cellular functions exerted by PARP10. Twenty to thirty percent of persons with Trypanosoma cruzi infection eventually develop cardiomyopathy. If an early indicator were to be identified and validated in longitudinal studies, this could enable treatment to be prioritized for those at highest risk. We evaluated cardiac and extracellular matrix remodeling markers across cardiac stages in T. cruzi infected (Tc+) and uninfected (Tc-) individuals.Participants were recruited in a public hospital in Santa Cruz, Bolivia and assigned cardiac severity stages by electrocardiogram and echocardiogram. BNP, NTproBNP, CKMB, troponin I, MMP-2, MMP-9, TIMP-1, TIMP-2, TGFb1, and TGFb2 were measured in specimens from 265 individuals using multiplex bead systems. Biomarker levels were compared between Tc+ and Tc- groups, and across cardiac stages. Receivers operating characteristic (ROC) curves were created; for markers with area under curve>0.60, logistic regression was performed.Analyses stratified by cardiac stage showed no significant differences in biomarker levels by Tc infection status. Among Tc+ individuals, those with cardiac insufficiency had higher levels of BNP, NTproBNP, troponin I, MMP-2, TIMP-1, and TIMP-2 than those with normal ejection fraction and left ventricular diameter. No individual marker distinguished between the two earliest Tc+ stages, but in ROC-based analyses, MMP-2/MMP-9 ratio was significantly higher in those with than those without ECG abnormalities.BNP, NTproBNP, troponin I, MMP-2, TIMP-1, and TIMP-2 levels rose with increasing severity stage but did not distinguish between Chagas cardiomyopathy and other cardiomyopathies. Among Tc+ individuals without cardiac insufficiency, only the MMP-2/MMP-9 ratio differed between those with and without ECG changes.
