Objective: This study will explore and optimize the conditions for the construction of the mouse model of endometriosis using the estrogen-releasing thermo-sensitive hydrogel system. Methods: (1) The mice were injected subcutaneously with a gradient concentration of estrogen-releasing thermo-sensitive hydrogel systems, and the blood estrogen concentration was measured 3 days later. And then we draw the curve between mouse blood estrogen concentration and thermo-sensitive hydrogel systems concentration, selecting the most appropriate concentration of estrogen-releasing thermo-sensitive hydrogel systems. (2) To explore the release pattern of estrogen-releasing thermo-sensitive hydrogel system, the mice were injected subcutaneously with the optimal concentration of estrogen-releasing thermo-sensitive hydrogel system, and the blood estrogen concentration was measured on the 1st, 3rd, 5th, 7th and 10th day respectively. (3) We transplanted uteruses from donor mice into the abdominal cavities of recipient mice. The recipient mice were divided into three groups named a, b, and c. Group a was intraperitoneally injected with PBS everyday after transplantation, and group b was intraperitoneally injected with estrogen solution everyday after transplantation. And estrogen-releasing thermo-sensitive hydrogel systems mentioned above were injected subcutaneously to group c once a week. Mice were killed 21 days later to observe the success rate of the endometriosis models. Results: (1) The concentration of estrogen in mice was linear with the concentration of estrogenin the estrogen-releasing thermo-sensitive hydrogel systems. And 0.8 mg/ml was the most appropriate concentration of the estrogen-releasing thermo-sensitive hydrogel system. (2) The thermo- sensitive hydrogel systems including 0.8 mg/ml estrogen can release estrogen in mice for 7 days and can achieve effective blood estrogen concentration. (3) All mice in group a, b, and c survived. All mice in group b and c showed ectopic cysts, and no cyst was observed in group a. Conclusions: The construction of the mouse model of endometriosis requires exogenous estrogen. We have found that the novelestrogen-releasing thermo-sensitive hydrogel systems can slowly release estrogen after subcutaneously injection in mice, and the value of the subcutaneously injection of estrogen-releasing thermo-sensitive hydrogel system (0.8 mg/ml) per week to the construction of the mouse model is certain.The estrogen-releasing thermo-sensitive hydrogel system has high application value and is worthy of promotion.

Objective: To evaluate the feasibility and efficacy of arterial duct stenting in neonates with pulmonary atresia and intact ventricular septum., Methods: Eleven neonatal pulmonary atresia with intact ventricular septum patients received arterial duct stenting in our hospital from December 2007 to September 2010 were involved in this study. The average age was (8.20 +/- 2.90) days (ranged from 3 to 13 days). The average weight was (3.41 +/- 0.29) kg (ranged from 3.00 to 3.88 kg). The stents were selected according to digital subtracted angiography measurements. After checking for correct position by angiography, the balloon was inflated to expand the stent to desired diameter. Oxygen saturation was monitored, echocardiography was measured and stent diameter and location were observed by chest Xray. Patients were followed up at 1, 3, 6 and 12 months post procedure., Results: Stents were successfully implanted in all 11 patients. The preoperative peripheral oxygen saturation was (63.27 +/- 8.47)%, while increased to (82.73 +/- 5.59)% after alprostadil application and to (86.18 +/- 3.19)% after operation (all P < 0.01). After the operation, the peripheral oxygen saturation was higher than alprostadil application (P < 0.05). The intraoperative narrowest diameter of patent ductus arteriosus was (1.69 +/- 0.37) mm, the length was (16.72 +/- 2.37) mm. The internal diameter of implant stents was 4 mm, the length was (20.18 +/- 3.40) mm. After the operation, surgical B-T shunt operation was performed in one patient due to stent shift and pulse oxygen saturation decrease. One patient died post operation with unknown reason, another patient received stent balloon dilatation due to pulse oxygen saturation decrease at 4 months after the surgery. Pulmonary atresia with intact ventricular septum surgeries were performed in 2 patients at 5 and 7 months after stent implantation., Conclusion: The neonatal pulmonary atresia with intact ventricular septum arterial stent implantation was a feasible and effective procedure and this method could be used as preferred treatment in pulmonary atresia and intact ventricular septum for neonates.

Objective: To observed the post-seizure expression level of neuropeptides ghrelin and Nesfatin-1 between the untreated and treated groups in kainic acid-kindling rats and understand the significance of their basic expressions., Methods: The male S-D rats were divided randomly into NS and VPA groups after seizure. Each group was divided into 7 subgroups of 3 h, 6 h, 12 h, 24 h, 3 d, 7 d and 14 d. And the blank group (without NS or VPA) and sham group (normal saline instead of kainic acid) were established. RIA (radioimmunoassay), ELISA (enzyme-linked immunosorbent assay) and immunohistochemistry were used to detect the expression levels of ghrelin and Nesfatin-1 in the serum and hypothalamus of kainic acid-kindling rats., Results: Compared with the blank and sham groups, the seizure rats had a decreased expression of ghrelin while there was an elevated expression of Nesfatin-1 in hypothalamus. This trend was consistent with the serum expressions of deacylated ghrelin and Nesfatin-1. The expression of ghrelin became elevated at all stages in the VPA group. Especially the subgroups of 24 h (0.320 ± 0.007) and 7 d (0.284 ± 0.004) had a significant increase (P < 0.05) while their expression levels remained lowed than that of the blank group (0.342 ± 0.039). The expressions of Nesfatin-1 had a significant decrease (P < 0.05) in all VPA subgroups while they were higher than those in the blank group (0.124 ± 0.007)., Conclusion: An accurate detection of serum expression level of deacylated ghrelin and Nesfatin-1 may reflect the fluctuation trend of neuropeptides in hypothalamus. It may offer a simple, sensitive and noninvasive method of diagnosing and treating epilepsy.

Objective: To investigate the expression of Arc mRNA in insular electrical kindled and single electrical stimulated rats and its significance., Methods: Male SD rats were divided randomly into kindled group, single electrical stimulated group, sham-operated group and control group. Each group was divided into 2 sub-groups at different time points. Kindled group: establishing chronic insular electrical kindled model, decapitation to perform RT-PCR of Arc mRNA on hippocampus and to applying in situ hybridization of Arc mRNA on dentate gyrus; Single electrical stimulated group: using the same method as the kindled group with only electrical stimulation twice; Sham-operated group: using the same method to the kindled group without electrical stimulation;, Control Group: no surgery., Results: Expression of Arc mRNA in the hippocampus of single electrical stimulated, sham-operated and control groups were 0.72 ± 0.14, 0.75 ± 0.16 and 0.71 ± 0.14 respectively. They were significantly less than the kindled group of 1.78 ± 0.43(P < 0.01) at 3 h. The expression of Arc mRNA had no significant difference (P > 0.05) among 4 groups at 6 h; in situ hybridization (cell count) of Arc mRNA showed the expression of Arc mRNA in the kindled group was 112.8 ± 6.0. And it was significantly higher than the other three groups of 46.25 ± 4.35, 45.25 ± 6.23, 44.75 ± 6.49 (P < 0.01) after 3 h, there were no significant difference among 4 groups at 6 h (P > 0.05)., Conclusion: Insular epilepsy increase the expression of Arc mRNA in hippocampus. Arc may play an important role in the synaptic plasticity of insular epilepsy.

According to investigations, it was shown that peripheral blood WBC count increased, percentage of promyelocytes increased, differentiation features emerged, protein kinase C (PK-C) activity elevated, intracellular lysozyme content increased, CFU-F productivity increased and typical t (15; 17) disappeared, when induced differentiation therapy with all trans-retinoic acid (RA) was carried out in patients with acute promyelocytic leukemia. Comparison between RA group and RA+Harringtonin/Ara-C group showed no significant difference in most of the parameters; the curative effect was same in both groups. Complete remission (CR) rate was achieved in 92.6% (25/27 cases) of the patients in the entire group. In order to increase CR rate, prevention and treatment for haemorrhagic complications are critical. Patients with hyperleukocytosis (WBC > or = 100 x 10(9)/L) should be treated with intensive combined therapy as well as aggressive prevention of haemorrhage and pathological changes of lung and brain.
