Multiplex imaging in the second near-infrared window (NIR-II, 1000-1700 nm) provides exciting opportunities for more precise understanding of biological processes and more accurate diagnosis of diseases by enabling real-time acquisition of images with improved contrast and spatial resolution in deeper tissues. Today, the number of imaging agents suitable for this modality remains very scarce. In this work, we have synthesized and fully characterized, including theoretical calculations, a series of dimeric Ln III /Ga III metallacrowns bearing Ru II polypyridyl complexes, LnRu-3 (Ln=Y III , Yb III , Nd III , Er III ). Relaxed structures of YRu-3 in the ground and the excited electronic states have been calculated using dispersion-corrected density functional theory methods. Detailed photophysical studies of LnRu-3 have demonstrated that characteristic emission signals of Yb III , Nd III and Er III in the NIR-II range can be sensitized upon excitation in the visible range through Ru II -centered metal-to-ligand charge transfer (MLCT) states. We have also showed that these NIR-II signals are unambiguously detected in an imaging experiment using capillaries and biological tissue-mimicking phantoms. By combining advantages of two series of lanthanide(III)/zinc(II) metallacrowns (MCs) assembled using pyrazine- (pyzHA 2- ) and quinoxaline- (quinoHA 2- ) hydroximate building blocks ligands, we created here water-soluble mixed-ligand MCs with extended absorption to the visible range. The Yb III analogue demonstrated improved photophysical properties in the near-infrared (NIR) range in cell culture media, facilitating its application for NIR optical imaging in living HeLa cells., (© 2023 The Authors. Drug-loaded liposomes are typical examples of nanomedicines. We show here that doxorubicin, the anti-cancer agent in the liposomal drug Doxil, can sensitize Ytterbium (Yb 3+ ) and generate its near-infrared (NIR) emission. When doxorubicin and amphiphilic Yb 3+ chelates are incorporated into liposomes, the sensitized emission of Yb 3+ is dependent on the integrity of the particles, which can be used to monitor drug release. Herein, the synthesis and characterization of the first family of multipodal ligands with a Tröger's base framework designed for the preparation of luminescent lanthanide(III) complexes are reported. Eight ligands were designed and synthesized using different strategies, including alkylation reactions, amide couplings, and Ugi multicomponent reactions. All the ligands bear carboxylate groups for the coordination of the lanthanide(III) ions, with the lanthanide(III)-sensitizing units consisting of the Tröger's base framework itself or attached benzamides. Upon irradiation of the chromophoric ligands, green terbium(III) emission was efficiently generated, whereas europium(III) emission was negligible. The geometry and substitution pattern of the ligands allow control of the stoichiometry of the species formed and the Tb III luminescence sensitization efficiency, showing that para-substitution patterns are more efficient than meta substitution for the formation of coordination compounds with lower Tb III /ligand ratio. Nanothermometry is the study of temperature at the submicron scale with a broad range of potential applications, such as cellular studies or electronics. Molecular luminescent-based nanothermometers offer a non-contact means to record these temperatures with high spatial resolution and thermal sensitivity. A luminescent-based molecular thermometer comprised of visible-emitting Ga 3+ /Tb 3+ and Ga 3+ /Sm 3+ metallacrowns (MCs) achieved remarkable relative thermal sensitivity associated with very low temperature uncertainty of S r =1.9 % K -1 and δT<0.045 K, respectively, at 328 K, as an aqueous suspension of polystyrene nanobeads loaded with the corresponding MCs. The interest in ratiometric luminescent probes that detect and quantify a specific analyte is growing. Owing to their special luminescence properties, lanthanide(III) cations offer attractive opportunities for the design of dual-color ratiometric probes. Here, the design principle of hetero-bis-lanthanide peptide conjugates by using native chemical ligation is described for perfect control of the localization of each lanthanide cation within the molecule. Two zinc-responsive probes, r-LZF1 Tb|Cs124|Eu and r-LZF1 Eu|Cs124|Tb are described on the basis of a zinc finger peptide and two DOTA (DOTA=1,4,7,10-tetraaza-cyclododecane-1,4,7,10-tetraacetic acid) complexes of terbium and europium. Both display dual-color ratiometric emission in response to the presence of Zn 2+ . By using a screening approach, anthracene was identified for the sensitization of the luminescence of two near-infrared-emitting lanthanides, Yb 3+ and Nd 3+ . Herein, the design, synthesis, and characterization of a phenhomazine ligand are described. The ligand has six pendant acetate arms designed for the combined coordination of copper(II) and lanthanide(III) ions, with the perspective of developing a "turn-off" copper sensor. The key step for the ligand preparation was the one-step endomethylene bridge fission of a diamino Tröger's base with a concomitant alkylation. Fluorescence and absorption spectroscopies as well as nuclear magnetic resonance (NMR) experiments were performed to analyze and understand the coordination properties of the ligand. Transition metal coordination was driven by the synergistic effect of the free nitrogen atoms of the diazocinic core and the two central acetate arms attached to those nitrogen atoms, whereas lanthanide coordination is performed by the external acetate arms, presumably forming a self-assembled 2:2 metallosupramolecular structure. The terbium complex shows the typical green emission with narrow bands and long luminescence lifetimes. The luminescence quenching produced by the presence of copper(II) ions was analyzed. Multimodal probes capable of combining imaging modalities within a single molecule are in high demand today as they can provide information at both molecular and anatomical levels. Herein, a study was conducted on a series of gallium(III)/lanthanide(III) bis(12-MC-4) metallacrowns (MCs) with the general composition {Ln[12-MC Ga III N(shi) -4]} 2 (iph) 4 (Ln-I x , x=0, 4, 8, 12), where shi and iph are salicylhydroximate and isophthalate ligands, respectively, or their iodinated derivatives. For Yb-I x , the attenuation in X-ray computed tomography (XCT) imaging and near-infrared (NIR) luminescence properties can be finely tuned by controlled structural modifications based on iodo groups. Solutions of Yb-I x appear to be 22-40 times more efficient as XCT agents in comparison to the commercially available iobitridol, while providing an intense emission signal in the NIR range with total quantum yields up to 8.6 %, which are among the highest values reported so far. Intrigued by the unexpected room-temperature dual visible/near-infrared (NIR) luminescence observed for fast-relaxing erbium complexes embedded in triple-stranded helicates, in this contribution, we explore a series of six tridentate N-donor receptors L4-L9 with variable aromaticities and alkyl substituents to extricate the stereoelectronic features responsible for such scarce optical signatures. Detailed solid-state (X-ray diffraction, differential scanning calorimetry, optical spectroscopy) and solution (speciations and thermodynamic stabilities, spectrophotometry, NMR and optical spectroscopy) studies of mononuclear unsaturated [Er(Lk) 2 ] 3+ and saturated triple-helical [Er(Lk) 3 ] 3+ model complexes reveal that the stereoelectronic changes induced by the organic ligands affect inter- and intramolecular interactions to such an extent that 1) melting temperatures in solids, 2) the affinity for trivalent erbium in solution, and 3) optical properties in luminescent complexes can be rationally varied and controlled. A new family of [3.3.1] metallacryptates with the general composition [LnGa 6 (H 2 shi)(Hshi)(shi) 7 (C 5 H 5 N)] (Ln-1; shi 3- =salicylhydroximate; Ln = Pr, Nd, Sm-Yb) has been synthesized and characterized. Ln-1 display both interesting magnetic and luminescent properties. Sm-1 has sharp emission bands in the visible and the near-infrared (NIR) regions with quantum yield values (QSmL ) of 1.64(9) and 5.5(2) . 10 -2  %, respectively. Tb-1 exhibits a weak green emission (QTbL =1.89(3) . 10 -1  %) while Pr-1, Nd-1, Ho-1, Er-1, and Yb-1 possess emission bands in the NIR range with QPrL =3.7(2) . 10 -3  %, QNdL =1.71(5) . 10 -1  %, QHoL =1.1(2) . 10 -3  %, QErL =7.1(2) . 10 -3  % and QYbL =0.65(3) %. Nd-1, Dy-1, and Yb-1 display slow magnetization relaxation in an applied field, where only Dy-1 has been observed to follow an Orbach process (U eff =12.7 K). A family of dimeric Ln III [12-MC Ga(III)N(shi) -4] metallacrowns (MCs) (Ln III =Pr, Nd, Sm, Eu, Gd, Tb, Dy, Ho, Er, Tm, and Yb) was synthesized using the isophthalate group (ip 2- ) as a linker. The [LnGa 4 ] 2 complexes exhibit remarkable photophysical properties, with large molar absorptivities of ≈4×10 4  m -1  cm -1 , high quantum yields and long luminescence lifetimes with values of (i) 31.2(2)% and 1.410(1) ms, respectively for the visible-emitting [TbGa 4 ] 2 complex and (ii) 2.43(6)% and 30.5(1) μs for the near-infrared (NIR) emitting [YbGa 4 ] 2 in the solid state. The NIR emission was obtained not only from Yb, Nd, and Er complexes but also from the less frequently observed emitters such as Pr and Ho. In addition, emission in both visible and NIR domains could be detected for Dy and Sm MCs. ESI-MS and UV/Vis data revealed that the complexes are highly stable in dimethylsulfoxide (DMSO) solution with the 1 H- and COSY-NMR spectra of the diamagnetic [YGa 4 ] 2 analogue providing evidence for long-term solution stability. In this work, we describe a novel DOTA (1,4,7,10-tetraazacyclododecane-1,4,7,10-tetraacetic acid) based ligand with a chromophoric tropone coordinating sidearm ( 1 ). Ln 3+ complexes of 1 have one inner sphere water molecule. The r 1 relaxivity of Gd 1 is similar to that of the commercial Gd-based MRI agents. The neutral O-donor atom of the tropone moiety slows down the water exchange rate by a factor of 3 compared to GdDOTA. In addition, Nd 1 and Yb 1 complexes exhibit significant NIR emission in aqueous solutions indicating that the tropone unit is an efficient sensitizer for these Ln 3+ -ions. Therefore, this new ligand is a promising platform for the design of Ln 3+ based dual MR/optical imaging probes.

Responsive luminescent probes emitting in the near-infrared (NIR) are in high demand today for biological applications as they allow for the easy and unambiguous discrimination of autofluorescence. Due to their luminescence properties, lanthanide ions offer an interesting alternative to classical organic fluorescent dyes. This has stimulated the development of lanthanide-based responsive probes. Nevertheless, responsive probes that can operate in water with NIR-emitting lanthanide ions are scarce. In this communication, zinc fingers are shown to be versatile scaffolds to elaborate a variety of Zn 2+ -responsive probes based on lanthanide emission and featuring desirable properties for the selective detection of Zn 2+ in experimental conditions close to cellular. The development of efficient sensors for the determination of the water content in organic solvents is highly desirable for a number of chemical industries. Presented herein is a Mg(2+) metal-organic framework (MOF), which exhibits the remarkable capability to rapidly detect traces of water (0.05-5 % v/v) in various organic solvents through an unusual turn-on luminescence sensing mechanism. We report the synthesis of a cyclen-based ligand (4,10-bis[(1-oxidopyridin-2-yl)methyl]-1,4,7,10-tetraazacyclododecane-1,7-diacetic acid=L1) containing two acetate and two 2-methylpyridine N-oxide arms anchored on the nitrogen atoms of the cyclen platform, which has been designed for stable complexation of lanthanide(III) ions in aqueous solution. Relaxometric studies suggest that the thermodynamic stability and kinetic inertness of the Gd(III) complex may be sufficient for biological applications. A detailed structural study of the complexes by (1) H NMR spectroscopy and DFT calculations indicates that they adopt an anti-Δ(λλλλ) conformation in aqueous solution, that is, an anti-square antiprismatic (anti-SAP) isomeric form, as demonstrated by analysis of the (1) H NMR paramagnetic shifts induced by Yb(III) . The water-exchange rate of the Gd(III) complex is ${k{{298\hfill \atop {\rm ex}\hfill}}}$=6.7×10(6)  s(-1) , about a quarter of that for the mono-oxidopyridine analogue, but still about 50 % higher than the ${k{{298\hfill \atop {\rm ex}\hfill}}}$ of GdDOTA (DOTA=1,4,7,10-tetraazacyclododecane-1,4,7,10-tetraacetic acid). The 2-methylpyridine N-oxide chromophores can be used to sensitize a wide range of Ln(III) ions emitting in both the visible (Eu(III) and Tb(III) ) and NIR (Pr(III) , Nd(III) , Ho(III) , Yb(III) ) spectral regions. The emission quantum yield determined for the Yb(III) complex (${Q{{{\rm L}\hfill \atop {\rm Yb}\hfill}}}$=7.3(1)×10(-3) ) is among the highest ever reported for complexes of this metal ion in aqueous solution. This work, based on the synthesis and analysis of chemical compounds, describes a kinetic approach for identifying intramolecular intermetallic energy-transfer processes operating in discrete polynuclear lanthanide complexes, with a special emphasis on europium-containing entities. When all coordination sites are identical in a (supra)molecular complex, only heterometallic communications are experimentally accessible and a Tb → Eu energy transfer could be evidenced in [TbEu(L5)(hfac)6] (hfac = hexafluoroacetylacetonate), in which the intermetallic separation amounts to 12.6 Å. In the presence of different coordination sites, as found in the trinuclear complex [Eu3(L2)(hfac)9], homometallic communication can be induced by selective laser excitation and monitored with the help of high-resolution emission spectroscopy. The narrow and non-degenerated character of the Eu((5)D0 ↔ (7)F0) transition excludes significant spectral overlap between donor and acceptor europium cations. Intramolecular energy-transfer processes in discrete polynuclear europium complexes are therefore limited to short distances, in agreement with the Fermi golden rule and with the kinetic data collected for [Eu3(L2)(hfac)9] in the solid state and in solution. Consequently, trivalent europium can be considered as a valuable local structural probe in discrete polynuclear complexes displaying intermetallic separation in the sub-nanometric domain, a useful property for probing lanthanido-polymers., (© 2014 WILEY-VCH Verlag GmbH & Co. We report herein the synthesis of a luminescent polynuclear dendritic structure (Sm(III)-G3P-2,3Nap) in which eight Sm(III) ions are sensitized by thirty-two 2,3-naphthalimide chromophores. Upon a single excitation wavelength, the dendrimer complex exhibits two types of emission in the visible and in the near-infrared (NIR) ranges. Sm(III)-G3P-2,3Nap was non-cytotoxic after 24 h of incubation and up to 2.5 μM. The ability of the Sm(III)-based probe to be taken up by cells was confirmed by confocal microscopy. Epifluorescence microscopy validated Sm(III)-G3P-2,3Nap as a versatile probe, capable of performing interchangeably in the visible or NIR for live-cell imaging. As both emissions are obtained from a single complex, the cytotoxicity and biodistribution are inherently the same. The possibility for discriminating the sharp Sm(III) signals from autofluorescence in two spectral ranges increases the reliability of analysis and reduces the probability of artifacts and instrumental errors.

Calcium plays a vital role in the human body and especially in the central nervous system. Precise maintenance of Ca(2+) levels is very crucial for normal cell physiology and health. The deregulation of calcium homeostasis can lead to neuronal cell death and brain damage. To study this functional role played by Ca(2+) in the brain noninvasively by using magnetic resonance imaging, we have synthesized a new set of Ca(2+) -sensitive smart contrast agents (CAs). The agents were found to be highly selective to Ca(2+) in the presence of other competitive anions and cations in buffer and in physiological fluids. The structure of CAs comprises Gd(3+)-DO3A (DO3A=1,4,7-tris(carboxymethyl)-1,4,7,10-tetraazacyclododecane) coupled to a Ca(2+) chelator o-amino phenol-N,N,O-triacetate (APTRA). The agents are designed to sense Ca(2+) present in extracellular fluid of the brain where its concentration is relatively high, that is, 1.2-0.8 mM. The determined dissociation constant of the CAs to Ca(2+) falls in the range required to sense and report changes in extracellular Ca(2+) levels followed by an increase in neural activity. In buffer, with the addition of Ca(2+) the increase in relaxivity ranged from 100-157%, the highest ever known for any T1-based Ca(2+)-sensitive smart CA. The CAs were analyzed extensively by the measurement of luminescence lifetime measurement on Tb(3+) analogues, nuclear magnetic relaxation dispersion (NMRD), and (17)O NMR transverse relaxation and shift experiments. Herein, we discuss how, why, and when cascade complexation reactions produce stable, mononuclear, luminescent ternary complexes, by considering the binding of hexafluoroacetylacetonate anions (hfac(-)) and neutral, semi-rigid, tridentate 2,6-bis(benzimidazol-2-yl)pyridine ligands (Lk) to trivalent lanthanide atoms (Ln(III)). The solid-state structures of [Ln(Lk)(hfac)(3)] (Ln=La, Eu, Lu) showed that [Ln(hfac)(3)] behaved as a neutral six-coordinate lanthanide carrier with remarkable properties: 1) the strong cohesion between the trivalent cation and the didentate hfac anions prevented salt dissociation; 2) the electron-withdrawing trifluoromethyl substituents limited charge-neutralization and favored cascade complexation with Lk; 3) nine-coordination was preserved for [Ln(Lk)(hfac)(3)] for the complete lanthanide series, whilst a counterintuitive trend showed that the complexes formed with the smaller lanthanide elements were destabilized. Thermodynamic and NMR spectroscopic studies in solution confirmed that these characteristics were retained for solvated molecules, but the operation of concerted anion/ligand transfers with the larger cations induced subtle structural variations. Combined with the strong red photoluminescence of [Eu(Lk)(hfac)(3)], the ternary system Ln(III)/hfac(-)/Lk is a promising candidate for the planned metal-loading of preformed multi-tridentate polymers., (Copyright © 2012 WILEY-VCH Verlag GmbH & Co. A series of novel triazole derivative pyridine-based polyamino-polycarboxylate ligands has been synthesized for lanthanide complexation. This versatile platform of chelating agents combines advantageous properties for both magnetic resonance (MR) and optical imaging applications of the corresponding Gd(3+) and near-infrared luminescent lanthanide complexes. The thermodynamic stability constants of the Ln(3+) complexes, as assessed by pH potentiometric measurements, are in the range log K(LnL)=17-19, with a high selectivity for lanthanides over Ca(2+), Cu(2+), and Zn(2+). The complexes are bishydrated, an important advantage to obtain high relaxivities for the Gd(3+) chelates. The water exchange of the Gd(3+) complexes (k(ex)(298)=7.7-9.3×10(6) s(-1)) is faster than that of clinically used magnetic resonance imaging (MRI) contrast agents and proceeds through a dissociatively activated mechanism, as evidenced by the positive activation volumes (ΔV(≠)=7.2-8.8 cm(3) mol(-1)). The new triazole ligands allow a considerable shift towards lower excitation energies of the luminescent lanthanide complexes as compared to the parent pyridinic complex, which is a significant advantage in the perspective of biological applications. In addition, they provide increased epsilon values resulting in a larger number of emitted photons and better detection sensitivity. The most conjugated system PheTPy, bearing a phenyl-triazole pendant on the pyridine ring, is particularly promising as it displays the lowest excitation and triplet-state energies associated with good quantum yields for both Nd(3+) and Yb(3+) complexes. Cellular and in vivo toxicity studies in mice evidenced the non-toxicity and the safe use of such bishydrated complexes in animal experiments. The ML(4) complexes formed by reaction between the bidentate azulene-based ligand diethyl 2-hydroxyazulene-1,3-dicarboxylate (HAz) and several lanthanide cations (Pr(3+), Nd(3+), Gd(3+), Ho(3+), Er(3+), Tm(3+), Yb(3+), and Lu(3+)) have been synthesized and characterized by elemental analysis, FT-IR vibrational spectroscopy and electrospray ionization mass spectroscopy. Spectrophotometric titrations have revealed that four Az(-) ligands react with one lanthanide cation to form the ML(4) complex in solution. Studies of the luminescence properties of these ML(4) complexes demonstrated that Az(-) is an efficient sensitizer for four different near-infrared emitting lanthanide cations (Nd(3+), Er(3+), Tm(3+), and Yb(3+)); the resulting complexes have high quantum yield values in CH(3)CN. The near-infrared emission arising from Tm(3+) is especially interesting for biologic imaging and bioanalytical applications since biological systems have minimal interaction with photons at this wavelength. Hydration numbers, representing the number of water molecules bound to the lanthanide cations, were obtained through luminescence lifetime measurements and indicated that no molecules of water/solvent are bound to the lanthanide cation in the ML(4) complex in solution. The four coordinated ligands protect well the central luminescent lanthanide cation against non-radiative deactivation from solvent molecules.

The synthesis, characterization, and metal-binding studies of chelate-functionalized dendrimers is reported. Salicylate, catecholate, and hydroxypyridinonate bidentate chelators have been coupled to the surface of both poly(propyleneimine) (Astramol) and poly(amidoamine) (Starburst, PAMAM) dendrimers up to the fifth generation (64 endgroups). A general method has been developed for the facile and high quality chromatographic purification of poly(propyleneimine) and poly(amidoamine) dendrimer derivatives. One- and two-dimensional (TOCSY) 1H NMR experiments and electrospray ionization mass spectrometry (ESI-MS) have confirmed the exhaustive coupling of these chelators to the primary amine functionalities of the dendrimers. Spectrophotometric titrations were used to investigate the metal binding ability of these macrochelates. Spectral analysis shows that ferric iron binding to these ligands is localized to the chelating endgroups. The ability of these dendritic polymers to bind large numbers of metal ions may lead to applications as metal sequestering agents for waste remediation technologies.
