The European Food Safety Authority (EFSA) has a goal to efficiently conduct aggregate exposure assessments (AEAs) for chemicals using both exposure models and human biomonitoring (HBM) data by 2030. To achieve EFSA's vision, a roadmap for action for advancing aggregate exposure (AE) in the EU was developed. This roadmap was created by performing a series of engagement and data collection activities to map the currently available methods, data, and tools for assessing AE of chemicals, against the needs and priorities of EFSA. This allowed for the creation of a AEA framework, identification of data and knowledge gaps in our current capabilities, and identification of the challenges and blockers that would hinder efforts to fill the gaps. The roadmap identifies interdependent working areas (WAs) where additional research and development are required to achieve EFSA's goal. It also proposes future collaboration opportunities and recommends several project proposals to meet EFSA's goals. Eight proposal projects supported by SWOT analysis are presented for EFSA's consideration. The project proposals inform high‐level recommendations for multi‐annual and multi‐partner projects. Recommendations to improve stakeholder engagement and communication of EFSA's work on AEA were gathered by surveying stakeholders on specific actions to improve EFSA's communication on AE, including webinars, virtual training, social media channels, and newsletters. Drivers of phytoplankton and zooplankton dynamics vary spatially and temporally in estuaries due to variation in hydrodynamic exchange and residence time, complicating efforts to understand controls on food web productivity. We conducted approximately monthly (2012–2019; n = 74) longitudinal sampling at 10 fixed stations along a freshwater tidal terminal channel in the San Francisco Estuary, California, characterized by seaward to landward gradients in water residence time, turbidity, nutrient concentrations, and plankton community composition. We used multivariate autoregressive state space (MARSS) models to quantify environmental (abiotic) and biotic controls on phytoplankton and mesozooplankton biomass. The importance of specific abiotic drivers (e.g., water temperature, turbidity, nutrients) and trophic interactions differed significantly among hydrodynamic exchange zones with different mean residence times. Abiotic drivers explained more variation in phytoplankton and zooplankton dynamics than a model including only trophic interactions, but individual phytoplankton–zooplankton interactions explained more variation than individual abiotic drivers. Interactions between zooplankton and phytoplankton were strongest in landward reaches with the longest residence times and the highest zooplankton biomass. Interactions between cryptophytes and both copepods and cladocerans were stronger than interactions between bacillariophytes (diatoms) and zooplankton taxa, despite contributing less biovolume in all but the most landward reaches. Our results demonstrate that trophic interactions and their relative strengths vary in a hydrodynamic context, contributing to food web heterogeneity within estuaries at spatial scales smaller than the freshwater to marine transition. Background: Allergic rhinitis includes a certain degree of autonomic imbalance. However, no information is available on how daily changes in allergy burden affect autonomic imbalance. We aimed to estimate associations between daily allergy burden (allergy symptoms and mood) and daily heart rate characteristics (resting heart rate and sample entropy, both biomarkers of autonomic balance) of adults with allergic rhinitis, based on real‐world measurements with a wearable telemonitoring system. Methods: Adults with a tree pollen allergy used a smartphone application to self‐report daily allergy symptoms (score 0–44) and mood (score 0–4), and a Mio Alpha 2 wristwatch to collect heart rate characteristics during two pollen seasons of hazel, alder and birch in Belgium. Associations between daily allergy burden and heart rate characteristics were estimated using linear mixed effects distributed lag models with a random intercept for individuals and adjusted for potential confounders. Results: Analyses included 2497 participant‐days of 72 participants. A one‐point increase in allergy symptom score was associated with an increase in next‐day resting heart rate of 0.08 (95% CI: 0.02–0.15) beats per minute. A one‐point increase in mood score was associated with an increase in same‐day sample entropy of 0.80 (95% CI: 0.34–1.26) × 10−2. No associations were found between allergy symptoms and heart rate sample entropy, nor between mood and resting heart rate. Conclusion: Daily repeated measurements with a wearable telemonitoring system revealed that the daily allergy burden of adults with allergic rhinitis has systemic effects beyond merely the respiratory system. Background and purpose: Enterovirus infections pose a serious threat for patients with humoral deficiencies and may be lethal, whilst the efficacy of proposed treatment options such as corticosteroids, intravenous immunoglobulins and fluoxetine remains debated. Methods: Viral clearance was investigated in a patient with rituximab‐induced B‐cell depletion and chronic echovirus 13 (E13) meningoencephalitis/myofasciitis in response to intravenous immunoglobulins and fluoxetine using sequential semi‐quantitative E13 viral load measurements by real‐time reverse transcription polymerase chain reaction. Fluoxetine concentrations in plasma and cerebrospinal fluid were determined by liquid chromatography mass spectrometry. Results: Intravenous immunoglobulins appeared ineffective in this case of E13 infection, whereas virus clearance in cerebrospinal fluid was obtained after 167 days of oral fluoxetine. Since treatment with corticosteroids resulted in a flare of symptoms, rechallenge with viral load measurements was not attempted. Conclusion: In this report of a patient with rituximab‐associated chronic echovirus 13 meningoencephalitis, viral clearance in response to single treatment options is assessed for the first time. Our observations further support the in vivo efficacy of fluoxetine against enteroviral infections. More research is needed to establish its efficacy in different enterovirus strains. Ecosystem metabolism is a key ecological attribute and easy to describe, but quantifying metabolism in estuaries is challenging. Properly scaling measurements through time and space requires consideration of hydrodynamics and mixing water from heterogeneous sources, making any estimation uncertain. Here, we compared three methods for modeling ecosystem metabolism in a portion of the Sacramento‐San Joaquin Delta. Metabolism estimates based on laboratory incubations, continuous in situ buoys, and an oxygen isotope approach all indicated the system was net heterotrophic, and calculated rates were comparable in magnitude when averaged over the 2‐month study. Daily metabolic rates based on in situ buoys were the most variable, likely due to horizontal and vertical advection and poor portrayal of the dissolved oxygen budget. After temporally averaging in situ buoy estimates or smoothing the dissolved oxygen time series for tidal effects, rates were more comparable to the other methods, which may be necessary to account for tidal advection and unbalanced contributions from subhabitats within the metabolic footprint. Incubation‐based rates represent the finest temporal and spatial scale and only account for pelagic processes, which may explain why incubation‐based rates were lower than the other two methods. The oxygen isotope method provided temporally and spatially integrated rates that were bracketed by the other two methods and may be a valuable tool in systems matching the model requirements. Because uncertainty arises in each method from a number of assumptions and scaling calculations, the resolution of metabolic rates in estuaries is likely coarser and more variable than in other aquatic ecosystems. In Belgium, variations in thyroid cancer incidence were observed around the major nuclear sites. The present ecological study investigates whether there is an excess incidence of thyroid cancer among people living in the vicinity of the four nuclear sites at the smallest Belgian geographical level. Rate ratios were obtained from a Bayesian hierarchical model for areas of varying sizes around the nuclear sites. Focused hypothesis tests and generalized additive models were performed to test the hypothesis of a gradient in thyroid cancer incidence with increasing levels of surrogate exposures. No evidence was found for more incident cases of thyroid cancer near the two nuclear power plants. Regarding the two industrial and research nuclear sites, no evidence for a higher incidence in the vicinity of Mol‐Dessel was observed, whereas a slightly nonsignificant higher incidence was found in the close vicinity of Fleurus. In addition, significant gradients for thyroid cancer incidence were observed with the different types of surrogate exposure considered in the 20 km area around the site of Fleurus (decreasing distance, increasing wind direction frequency and increasing exposure to estimated hypothetical radioactive discharges of iodine‐131). In the investigation at the smallest Belgian geographical level, variations in thyroid cancer incidence were found around the Belgian nuclear sites. Significant exposure–response relationships were also observed for the site of Fleurus. Further investigations into these findings could be useful to allow inferring causal relationships on the origin of variations in incidence and to provide information at the individual level. What's new? Potential cancer risk associated with living near nuclear installations has long been a public concern. In Belgium, a previous study found a higher incidence around the two nuclear sites with research and industrial activities, but not around the two nuclear power plants. Exposure misclassification due to the large geographical scale could not be excluded, however. The present study, which uses data available at the smallest Belgian geographical level, confirms the previously‐described incidence patterns around the nuclear power plants and for one of the research and industrial sites. Abstract: Natural marks can provide a noninvasive approach to identify individual fishes, such as the endangered Delta Smelt Hypomesus transpacificus. We evaluated the feasibility of chromatophores as natural marks in cultured adult Delta Smelt and used the dorsal view of the head, where chromatophores are particularly abundant. Digital images of the head were compared among three photo sessions conducted at nearly 6‐week intervals in winter–spring 2013 (n = 292–234). Images were evaluated visually (naked eye) and by automated image recognition (TinEye Application Programming Interface [API]). Visual analysis enabled us to correctly identify all 60 fish examined between each pair of photo sessions. The TinEye API also correctly identified 100% of the individuals when it was able to, but the software did not identify 6–41% of the fish between photo sessions; the longer the period between photo sessions, the lower was the percentage of fish identified by the TinEye API. Fish exposure to outdoor ambient light constricted chromatophores, but the percentage of image matching was not influenced by ambient light, gender, or spawning condition. We conclude that it is feasible to identify individual adult Delta Smelt throughout the spawning season without having to tag them, but further development of automated image recognition is needed. ABSTRACT In order to illustrate the significance of a new anatomical finding, distortion of the interhemispheric fissure ( DIHF) associated with impacted medial borders of the frontal lobes, we report a retrospective observational study of 13 fetuses in which DIHF was identified on prenatal imaging. In 10 cases there were associated anatomical anomalies, including mainly midline anomalies (syntelencephaly (n = 2), lobar holoprosencephaly (n = 1), Aicardi syndrome (n = 2)), but also schizencephaly (n = 1), cortical dysplasia (n = 1) and more complex cerebral malformations (n = 3), including neural tube defect in two cases. Chromosomal anomaly was identified in two cases, including 6p deletion in a case without associated central nervous system anomalies and a complex mosaicism in one of the cases with syntelencephaly. In two cases, the finding was apparently isolated on both pre- and postnatal imaging, and the children were doing well at follow-up, aged 4 and 5 years. The presence of DIHF on prenatal imaging may help in the diagnosis of cerebral anomalies, especially those involving the midline. If DIHF is apparently isolated on prenatal ultrasound, magnetic resonance imaging is recommended for careful analysis of gyration and midline, especially optic and olfactory structures. We used pattern recognition algorithms and image processing to identify individual Chinook salmon Oncorhynchus tshawytscha. Using melanophore spot patterns located on the dorsal head region, algorithms ranked all database images against each other. We coupled this technology with a graphical user interface to visually confirm or reject top-ranked algorithm results and tested this process on 295 juvenile Chinook salmon in seven photo sessions over a 251-d period. Juveniles began developing spots, identifiable in photo images, between 167 and 197 d after conception (52.7-mm fork length [FL]). Unique spot patterns appeared 197–232 d from conception, beginning at approximately 104-mm FL. Of 254 fish surviving the experimental period, 106 (42%) demonstrated identifiable patterns, 102 (40%) developed spots but patterns were insufficient for identification, and 46 (18%) exhibited a complete lack of spots. Spot patterns continued developing on individual fish by study end. On average, fish that developed recognizable spot patterns did so at approximately 140-mm FL. Once they did, reidentification was 100% correct in up to four subsequent trials. Patterns remained identifiable even after a 25–32% size increase over a 55-d period and as juveniles went through smoltification. Although patterns occurred at sizes typically larger than salmon observed at some California Central Valley monitoring locations, this technique provides a potentially valuable, noninvasive method of identifying individual salmon during emigration. Improved image collection techniques and use of body areas exhibiting identifiable patterns at earlier developmental stages may increase fish available for pattern identification. These results demonstrate the indexing of a large database using pattern recognition algorithms for Chinook salmon. The utility of such an approach may be valuable for addressing specific biological questions associated with mass-produced (wild and hatchery), migratory salmonids, especially as individuals develop, grow, and move through the various habitats available to them. In the gut ecosystem, nitric oxide (NO) has been described to have damaging effects on the energy metabolism of colonocytes. Described mechanisms of NO production are microbial reduction of nitrate via nitrite to NO and conversion of l-arginine by NO synthase. The aim of this study was to investigate whether dietary compounds can stimulate the production of NO by representative cultures of the human intestinal microbiota and whether this correlates to other processes in the intestinal tract. We have found that the addition of a reduced sulfur compound, i.e. cysteine, contributed to NO formation. This increase was ascribed to higher sulfide concentrations generated from cysteine that in turn promoted the chemical conversion of nitrite to NO. The NO release from nitrite was of the order of 4‰ at most. Overall, it was shown that two independent biological processes contribute to the chemical formation of NO in the intestinal tract: (i) the production of sulfide by fermentation of sulfur containing amino acids or reduction of sulfate by sulfate reducing bacteria, and (ii) the reduction of nitrate to nitrite. Our results indicate that dietary thiol compounds in combination with nitrate may contribute to colonocytes damaging processes by promoting NO formation. [ABSTRACT FROM AUTHOR]

The degree of hydrological connectivity is mainly determined by the spatial organisation of heterogeneity. A meaningful and aggregate abstraction of spatial patterns is one of the promising means to gain fundamental insights into this complex interaction and can, moreover, be used as a tool to acquire a profound understanding of the major controls of catchment hydrology. In order to disclose such controls, pattern-process relationships and the explanatory power of landscape metrics were tested by simulating the runoff of differently patterned virtual basins, generated by neutral landscape models and fractal networks and solved by a surface hydrological model composed of kinematic wave routing and Green-Ampt infiltration. A total of 23 landscape metrics quantified the spatial patterns and were subsequently related to the functional connectivity, assessed as the proportion of internal runoff generation constituting the hydrological response at the outlet. Landscape metrics allowed the identification of dominant features of heterogeneity that explained the observed connectivity, and to disclose changes in control with class abundance. Therefore, landscape metrics are a useful tool for basin comparison and classification in terms of the dominant processes and the corresponding model structure requirements. BACKGROUND: Spider mites are important crop pests that rapidly develop resistance to acaricides. To investigate whether acaricide resistance is a threat to greenhouse rose culture in the Netherlands, the susceptibility of 15 strains of Tetranychus urticae was tested to several currently used acaricides, and resistance mechanisms were investigated. RESULTS: Although the observed levels of susceptibility differed between strains and acaricides, resistance was detected in most strains. The activity of detoxifying enzymes was significantly increased in most field-collected strains, and a number of amino acid substitutions known to be involved in resistance were detected. CONCLUSIONS: Resistance levels to traditional acaricides such as bifenthrin and abamectin were prominent, and might result in control failure under field conditions. Resistance to more recently registered compounds was detected in several populations. Resistance levels were generally unstable in the laboratory without selection pressure. The toxicological, biochemical and genetic data in this study will be essential in devising an efficient resistant management for Dutch rose culture. Bifenazate is a recently developed acaricide that is mainly used to control spider mites on a variety of crops. Although first thought to be a neurotoxin, genetic evidence obtained from bifenazate resistant Tetranychus urticae strains suggested an alternative mode of action as a Qo pocket inhibitor of the mitochondrial complex III. In this study, we reveal how bifenazate resistance in strains of Panonychus citri is maternally inherited and can confer cross-resistance to the known Qo inhibitor acequinocyl. The mitochondrial genome of P. citri was sequenced and Qo pocket mutations were shown to be linked with the resistant trait. Parallel evolution of cytochrome b mediated bifenazate resistance corroborates the alternative mode of action and yet again illustrates that care should be taken when employing Qo inhibitors as crop protection compounds. Hippocampal deep brain stimulation (DBS) is an experimental therapy for patients with pharmacoresistant temporal lobe epilepsy (TLE). Despite the successful clinical application of DBS, the optimal stimulation parameters are undetermined. We evaluate the efficacy of a new form of DBS, using continuous stimuli with Poisson distributed intervals (Poisson distributed stimulation, PDS) in the kainate (KA) rat model, a validated model for human TLE. Status epilepticus was elicited by injection of KA (i.p.). After development of spontaneous seizures, rats were implanted with hippocampal DBS- and depth electroencephalography (EEG) electrodes. After baseline EEG monitoring, one group of rats (n = 13) was treated with PDS and a second (n = 11) received regular high frequency stimulation (HFS) at 130 Hz. Stimulation intensity was 100 μA below the threshold for induction of epileptiform EEG activity. Stimulation intensity was significantly lower for PDS (156 ± 20 μA) than HFS (207 ± 23 μA; p < 0.02). Seven (54%) of 13 rats treated with PDS and 5 (45%) of 11 rats treated with HFS experienced a significant reduction in seizure frequency. In PDS-improved rats, seizure frequency was reduced to 33% (p < 0.01) of baseline value and in HFS-improved rats to 50% (p < 0.01). After termination of PDS, seizure rate returned to baseline value. Continuous hippocampal PDS significantly reduces the number of spontaneous seizures. Compared to regular HFS, there is a slightly larger number of improved rats and a larger efficacy at a considerably lower stimulus intensity. The article presents a study on the effect of bifenazate resistance mutations to cross-resistance between acequinocyl and fluacrypyrim. It mentions that the combination of two specific mutations in the cytochrome b Q0 pocket has caused and inherited the cross-resistance to acequinocyl in two bifenazate-resistant strains. It suggests the careful use of bifenazate and acequinocyl in control programmes to avoid resistance building up. Further, they should not be used in direct alteration since they have similar mode of action. It also reveals that fluacrypyrim is highly monogenic and dominant. It states that the resistance to fluacrypyrim was stimulated by an altered target-site protein since there is no evidence found on the increased in its metabolism.

Abstract: Two studies are reported that tested the fear-avoidance (FA) model using path analytic techniques. In study 1, 429 employees with back pain at baseline and back pain at 18months follow-up completed questionnaires assessing sociodemographic information, pain severity, negative affect, pain-related fear, and disability. Results indicated that pain severity at baseline predicted pain-related fear and disability at follow-up, and that pain-related fear is rather a consequence than an antecedent of pain severity. Results further revealed that the disposition to experience negative affect has a low impact upon pain severity and disability, and is best viewed as a precursor of pain-related fear. Study 2 included 238 employees without back pain at baseline, but who developed back pain at 1year follow-up. A similar model as in study 1 was tested. Overall, results are in line with those of study 1. Results are discussed in terms of theoretical relevance and clinical implications. [ABSTRACT FROM AUTHOR]
