Background: Reported as being expressed by mono‐ and binucleate placental cells, pregnancy‐associated glycoproteins (PAGs) are released into the blood circulation from the ruminant placenta. Circulating gestational PAGs levels may differ between sheep breeds. Objective: This study was aimed at the close monitoring of the serum PAGs profiles of Karya and Konya Merino sheep during early pregnancy. Methods: Fifteen Karya and 15 Konya Merino ewes were synchronized by a 12‐day treatment with progesterone‐impregnated intravaginal sponges. After the sponges were withdrawn, the ewes were administered 400 IU of equine chorionic gonadotropin. The ewes were allowed to mate naturally, and all animals were sampled for blood as of the day of mating (day 0) at weeks 1, 2, 3, 4 and 5. Pregnancy diagnoses were made by transabdominal ultrasonography at week 5. At weeks 6, 7, 8, 9 and 11, blood samples were collected only from the pregnant ewes. The blood samples were centrifuged at 3000 × g, and extracted sera were stored at −20°C until being used for laboratory analyses. Serum PAGs levels were determined with the aid of a commercial PAG‐enzyme‐linked immunosorbent assay test originally developed for pregnancy diagnosis in cattle. Differences in the between the PAGs levels throughout pregnancy and the group effect (Karya and Konya Merino) were determined with a two‐way mixed analysis of variance. Pairwise comparisons were made using a Bonferroni adjustment. Results: PAGs levels showed a linear increase with the advance of pregnancy in both Karya and Konya Merino sheep. No difference was detected between the breeds for serum PAGs levels. The serum PAGs levels of the pregnant and non‐pregnant ewes differed as of the fourth week. Conclusion: The serum PAGs levels of the Karya and Konya Merino ewes were similar during the first 11 weeks of gestation, and pregnancy diagnosis could be made based on serum PAGs levels as of the 4th week in both breeds. The objective of the study was to evaluate the effectiveness of the digital rectal palpation (DRP) technique for early pregnancy diagnosis and to compare the results of experiments I and II in the Red Sokoto goat. Experiment I had 68 goats with 52 does and 16 bucks. Does were divided into prostaglandin F2‐alpha (PGF2α; n = 18), progesterone pessaries (P4P; n = 18), and Control (n = 16) groups as oestrus synchronizaton methods. Bucks were used for oestrus detection (n = 6) and breeding (n = 10). Comparative pregnancy diagnosis was carried out on day 21 post‐breeding using ultrasonography (US), DRP, progesterone assay (PA), ballottement (BL), and non‐return‐to‐heat (NRH). Experiment II was a repeat but had 51 animals with 42 does and nine bucks; three bucks for oestrus detection and six for breeding. Does were divided into recto‐vaginal artificial insemination (AI) by DRP (n = 14), vaginal speculum AI (n = 14), and Control‐natural service (n = 14) groups, indicating breeding methods. Oestrus was synchronized with PGF2α and pregnancy diagnosis was carried out on day 21 using US, DRP, PA, and NRH. The results for both experiments were similar. Pregnancy rates for PGF2α group were 66.2%, 66.7%, 64.8%, 62.1%, and 63.0% for US, DRP, PA, BL, and NRH, respectively; P4P had 81.5%, 81.5%, 42.6%, 20.3%, and 42.6% for US, DRP, PA, BL, and NRH, respectively; while Control was 73.6%, 79.2%, 70.9%, 73.6%, and 73.6% for US, DRP, PA, BL, and NRH, respectively. Proportions are significantly (p <.000) different in the P4P group. It was concluded that the DRP technique was effective and consistent in early pregnancy diagnosis on day 21 and comparable to US, PA, BL, and non‐return‐to‐oestrus in both experiments in Red Sokoto goat does. Ultrasound technology has led to new lines of research in equine reproduction, and it has helped to greatly improve clinical diagnosis and reproductive outcomes in equine practice. This review aims to discuss the potential clinical uses and new approaches of ultrasonography in equine reproduction. Doppler modalities are usually used to evaluate the vascularization of the follicles, corpus luteum (CL), and the uterus in the mare for diagnostic purposes. Inclusion of Doppler ultrasound in artificial insemination and embryo transfer programs could improve the reproductive outcome of these techniques. Better selection of recipients based on CL functionality, early pregnancy diagnosis 7–8 days postovulation of the donor before flushing or diagnosis of mares with endometritis with pathological increases of blood flow are examples of clinical applications in the mare. In the stallion, colour Doppler ultrasound has improved the diagnostic potential of B‐mode ultrasound, improving the differential diagnosis of pathologies such as testicular torsion (decrease or absence of blood flow in the cord) and orchitis (increased blood flow in the cord). The incorporation of pulsed Doppler ultrasound into the reproductive evaluation of the stallion has enabled early identification of stallions with testicular dysfunction, thus allowing administration of timely treatment and subsequent improvements of the fertility prognosis for these animals. In addition, this technique has been used in the monitoring of patients undergoing medical and surgical treatments, thus verifying their efficacy. Recently, computer‐assisted pixel analysis using specific software has been performed in research work in order to semi‐quantitatively evaluate the vascularization (colour and power Doppler) and echotexture of different organs. These softwares are now being developed for clinical purposes, as is the case with Ecotext, a computer program developed for the evaluation of testicular echotexture, providing information on testicular functionality. Contents: As sheep produce similar pregnancy‐associated glycoproteins (PAGs) to cattle, a commercially available bovine visual pregnancy test based on the detection of PAGs (visual‐PAG‐test) was evaluated in sheep. The test was performed with whole blood (WhB), plasma (P) and serum (S) of 163 pregnant and 153 non‐pregnant ewes. Additionally, 11 pregnant ewes were tested weekly from day 14 to 49 of gestation and monthly from day 60 to day 149. Ten ewes were sampled weekly from the date of lambing until day 63 post‐partum (p.p.). The sensitivity in mid‐pregnancy (n = 163) was 98.16% (WhB), 99.39% (P) and 99.39% (S) compared to transabdominal ultrasonography as the gold standard while the specificity (n = 153) was 94.12% (WhB), 76.47% (P) and 93.46% (S), respectively. During early pregnancy, all 11 ewes were correctly identified as pregnant on day 42 (100%); however, the test sensitivity decreased to 54.6% (WhB) and 63.6% (S and P, respectively) at day 49. The ewes were again consistently identified as pregnant on day 63 (P) or on day 119 (S, WhB). The test was consistently negative from day 42 p.p. onwards in eight out of ten ewes. Two ewes remained consistently positive until the last sample on day 63 p.p. In conclusion, the test could be used to accurately select pregnant ewes at day 42 with a drop in sensitivity at day 49. The sensitivity of the visual‐PAG‐test was good in mid to late pregnancy, and early detection of pregnancy was possible in individual animals. In some ewes, the PAGs were however detectable for more than 63 days p.p.—the previous breeding history should therefore always be taken into account for correct interpretation of the test results. Abstract: The present study was conducted to determine the difference in plasma prostaglandin F2α metabolite concentrations following oxytocin (OT) challenge between pregnant and non‐pregnant cows. Experiment 1: cows were subjected to the OT challenge test on days 12, 14 or 16 (day of estrus = day 0) with or without prior insemination and plasma 13,14‐dihydro‐15‐keto prostaglandin F2α (PGFM) concentrations were measured from −30 to 180 min after OT injection. On day 16, the increment of plasma PGFM concentrations in response to OT injection was significantly smaller in pregnant than that in cyclic cows. On days 12 and 14, there was little OT‐induced PGFM secretion and no difference in PGFM increase between the pregnant and cyclic cows. Experiment 2: cows were inseminated on day 0 and subjected to the OT challenge test on day 16. Cows were classified into non‐pregnant/early embryonic death (NP/EED), late embryonic death (LED) and pregnant (PREG) groups. The increment of PGFM concentrations in response to OT injection was less in both PREG and LED groups than that in the NP/EED group. In conclusion, plasma PGFM secretion induced by OT is suggested as the base of pregnancy diagnosis prior to returning estrus in cows. [ABSTRACT FROM AUTHOR]

Improper or delayed pregnancy diagnosis has significant impact over animal production, particularly in buffaloes which inherently suffer from several reproductive inefficiencies. Thus the present study has undertaken to identify serum protein markers pertaining to early pregnancy diagnosis in buffaloes. Serum samples were collected from 10 pregnant Murrah Buffalo heifers at weekly intervals from days 0-35 post-artificial insemination and from 12 inseminated non-pregnant cyclic buffalo heifers on days 0, 7, 14 and 21. Two-dimensional gel electrophoresis and densitometric analysis revealed the presence of five protein spots showing average density fold change of ≥4 during early pregnancy. Mass spectrometry analysis identified these up-regulated proteins as anti-testosterone antibody light chain, apolipoprotein A-II precursor, serum amyloid A, cytokeratin type II, component IV isoform 1, which are have established roles in embryogenesis, but over-expression of the fifth identified protein immunoglobulin lambda light chain in pregnancy has been elucidated as a novel finding in the current study. Further, with bioinformatics analysis, potential antigenic B-cell epitopes were predicted for all these five proteins. An antibody cocktail-based approach involving antibodies against all these five up-regulated entire proteins or their epitopes could be developed for early detection of pregnancy in buffaloes. © 2016 Japanese Society of Animal Science [ABSTRACT FROM AUTHOR]

Objective To determine bovine fetal characteristics significantly associated with increasing gestational age as measured via transcutaneous ultrasonography over the right flank. Methods The length of gestation at date of pregnancy diagnosis via transcutaneous and transrectal ultrasonography was determined for 224 dairy cattle by estimation from subsequent calving dates. A separate dataset was created for each measurable fetal characteristic (i.e. thoracic diameter, abdominal diameter, umbilical diameter, placentome length and placentome height) and risk factors significantly associated with gestational age at pregnancy diagnosis within each dataset, including the fetal characteristic, were identified. Results Abdominal diameter was the most frequently observed fetal characteristic and thoracic diameter was the least. Gestational age at pregnancy diagnosis (d) was significantly associated with fetal thoracic diameter (P < 0.01), abdominal diameter (P < 0.01) and umbilical diameter (P = 0.02) when measured via transcutaneous ultrasound. Within each model, sire breed, dam breed, dam age and/or calf sex were also significantly associated with gestational age. Gestational age at pregnancy diagnosis was not significantly associated with either placentome height or length (P > 0.05). Conclusion Fetal thoracic diameter, abdominal diameter and umbilical diameter were found to be significantly associated with gestational age between approximate days 73 to 190 of gestation. Objective To investigate the practical application and image quality of transcutaneous ultrasonography across the right flank for the diagnosis of mid- to late-gestation in cattle. Procedure Dairy cows of known pregnancy status were used to establish criteria for the identification of mid- to late-stage pregnancy by transcutaneous ultrasonography. Factors involved in the practical application of transcutaneous ultrasound for routine use in pregnancy diagnosis in dairy cattle were also examined. Results Transcutaneous ultrasound, using a 3.5-MHz sector transducer applied over the caudodorsal right flank fold, allowed rapid and detailed visualisation of bovine pregnancy (i.e. fetus, fetal fluids, placentomes and/or fetal membranes) and could be used to establish criteria for the diagnosis of pregnancy. The commonly available cattle holding facilities of the herringbone dairy, rotary herringbone dairy, AI race or crush, and walk-through dairy all allow safe access to the right flank of a dairy or beef cow. However, transcutaneous scanning over the right flank is difficult in modern rotary turnstile dairies. Conclusion A 3.5-MHz sector transducer applied over the caudodorsal right flank fold allows detailed visualisation of the bovine fetus in most types of cattle-holding facilities. Further investigation of the sensitivity and specificity of this technique over the entire gestation period and its possible use in ageing of the bovine fetus is merited. [ABSTRACT FROM AUTHOR]

Objective Evaluate the sensitivity and specificity of transrectal ultrasound and transcutaneous ultrasonography across the right flank between days 31 and 196 of pregnancy in the dairy cow. Procedure Pregnancy status and stage of gestation at date of pregnancy diagnosis by transcutaneous and transrectal ultrasonography were determined for 1570 dairy cattle. Sensitivity and specificity values and probabilities of a correct diagnosis for both techniques were determined and compared. Possible effects of gestational age, herd, method of transcutaneous ultrasound diagnosis, cow age, cow and bull breed, and calf sex on the determination of a correct diagnosis of pregnancy status were evaluated for both techniques. Results The overall sensitivity and probability of a correct diagnosis of pregnancy status from days 31 to 196 of gestation were significantly higher when using transrectal ultrasound relative to transcutaneous ultrasound over the right flank. However, the sensitivity of transcutaneous ultrasound changed over the course of pregnancy: it was low (7–8%) before day 84, increased to 89% between days 141 and 154, and was close to 100% thereafter. The specificity of both methods did not differ significantly. Herd and gestational age at date of pregnancy diagnosis were significantly associated with making a correct test result using transcutaneous ultrasound, although only the latter predictor was significantly associated with making a correct test result using transrectal ultrasound. Conclusion Transcutaneous ultrasound can not be recommended as an accurate method for early pregnancy diagnosis in the dairy cow, particularly on a whole-herd basis. However, if pregnancy testing is undertaken in mid to late gestation, this technique could provide an accurate and rapid alternative to transrectal ultrasound or manual palpation per rectum. Aim: To clarify the location of primordial germ cells (PGC) in an embryo of target-age and to examine the culture environment of the PCG. Methods: The days of ovulation and fertilization were estimated by measuring the serum concentration of estrogen. Pregnancy was confirmed by measurement of the serum concentration of the beta subunit of macaque chorionic gonadotropin and by ultrasonography. We also examined the location of PGC in the embryo at the time of retrieval. Results: Results showed that PGC in an embryo were in the hindguts at day 30 postfertilization, arrived at the genital ridges via mesenteries at approximately day 33 postfertilization, and colonized the gonads by day 36 postfertilization. Conclusions: In conclusion, embryos collected on day 33 postfertilization are more suitable for obtaining PGC from cynomolgus monkeys. The PGC collected from cynomolgus monkey fetuses were cultured under conditions for the derivation and culture of human embryonic germ cells; enzymatically dispersed single cells were cultured on a SIM thioguanine-resistant ouabain-resistant cells (STO) feeder layer with recombinant human leukemia inhibitory factor, recombinant human basic fibroblast growth factor and forskolin. The cells from genital ridges and mesenteries at day 33 postfertilization had alkaline phosphatase (ALP) activity in vitro for a maximum of 13 days. In contrast, ALP activity had been held for 2 months under the same culture condition when the cells were derived from the gonads at day 66 postfertilization. The incidence of ectopic pregnancy in the United States has been reported to be as high as 20 per 1000 pregnancies, a more than 4-fold increase over the last 20 years. Clinical presentation can range from subtle, nonspecific abdominal complaints to acute onset pain or hemorrhagic bleeding. This article reviews the associated risk factors, pathophysiology, diagnosis, and management of ectopic pregnancy. Clinicians must maintain a high index of suspicion whenever women who might be pregnant have abdominal complaints. [Copyright &y& Elsevier]

We measured metabolites of progesterone (progestins) in fecal samples collected from captive Arabian oryx (Oryx leucoryx) females in postpartum (n=8), nonpregnant (n=9), and pregnant (n=8) reproductive stages between 1996 and 1998. We analyzed progestins using enzyme-immunoassays for pregnanediol and 20-oxo-pregnanes, respectively. Progestin concentrations were elevated for 3 days after parturition and then decreased to basal anestrous concentrations. Ovarian cyclicity resumed 25±2.4 days after parturition in 5 of the 8 females monitored. In nonpregnant females, excretion of fecal progestins followed a cyclic pattern increasing 6- to 12-fold from the follicular to the luteal phase. Fecal progestin concentrations allowed discrimination between pregnant and nonpregnant females after 3 months of gestation (P<0.01), mean concentration of the tested hormone metabolites being at least 3 times higher during mid and later stages of gestation (>3 months) than during early pregnancy (0-3 months). These data were subsequently used to set criteria for designation of a cow as pregnant in 55 free-ranging Arabian oryx in the reserve of Mahazat as-Sayd, Saudi Arabia sampled in 1998-1999 and 2003. The proportion of pregnant and nonpregnant oryx correctly identified by the test was 81% and 83%, respectively, when using both progestin assays. Despite a limited sample size, our results provide evidence that fecal progestin analysis is a reliable non- invasive method to determine the reproductive status of captive Arabian oryx and that it also can provide reasonably accurate physiological indices of pregnancy status in free- ranging specimens. The radioreceptorassay developed to determine serum luteinizing hormone level in the cynomolgus monkey was evaluated for its usefulness in early pregnancy diagnosis by the detection of serum chorionic gonadotropin (CG). Blood samples were collected at weekly intervals from the 1st to the 5th week after conception to determine changes in circulating levels of CG. In the pregnancy cases, serum CG levels increased to above 50 μg/ml in almost all animals. By the determination of CG 3 weeks after conception, 86% of all pregnant cases exhibited a positive response. Cases that were negative 3 weeks after conception were followed by a repeated test in the next week. According to this test schedule, 95% of the pregnant cases were detected by 4 weeks after conception, and 5% were undetected as negative responses because their CG levels were low., (Copyright © 1987 Wiley‐Liss, Inc., A Wiley Company.)
