Background: Gastric fluid plays a key role in food digestion and drug dissolution, therefore, the amount of gastric fluid present in a fasted state may influence subsequent digestion and drug delivery. We aimed to describe intra‐ and interindividual variation in fasted gastric content volume (FGCV) and to determine the association with age, sex, and body size characteristics. Methods: Data from 24 MRI studies measuring FGCV in healthy, mostly young individuals after an overnight fast were pooled. The analysis included 366 participants who had up to 6 repeated measurements, with a total of 870 measurements. Linear mixed model analysis was performed to calculate intra‐ and interindividual variability and to assess the effects of age, sex, weight, height, weight*height as a proxy for body size, and body mass index (BMI). Results: FGCV ranged from 0 to 156 mL, with a mean (± SD) value of 33 ± 25 mL. The overall coefficient of variation within the study population was 75.6%, interindividual SD was 15 mL, and the intraindividual SD was 19 mL. Age, weight, height, weight*height, and BMI had no effect on FGCV. Women had lower volumes compared to men (MD: −6 mL), when corrected for the aforementioned factors. Conclusion: FGCV is highly variable, with higher intraindividual compared to interindividual variability, indicating that FGCV is subject to day‐to‐day and within‐day variation and is not a stable personal characteristic. Early life history traits of small pelagic fish are usually affected by environmental conditions. Patagonian sprat, Sprattus fuegensis, is a small pelagic fish that inhabits the continental shelf, fjords and channels of Patagonia. Their larvae are usually exposed to large variations in physical and biological conditions. We evaluated the effects of environmental conditions on early life history traits of this key species encompassing over 20 years of information. The characterization of oceanographic features, larval abundance and distribution was carried out in western Patagonia between 1996 and 2019, while the growth and mortality rates were estimated for the austral spring 2019 between 49°54′S and 53°55′S. Larval abundance decreased three to ten times from 1996 to 1997 to the rest of the study period (2008–2010 and 2014–2019), with larger abundances being found over the shelf. In spring 2019, the growth rate was 0.20 ± 0.05 mm d−1 for larvae between 9 and 19 days after hatching. The growth rate was similar in partially stratified (7–50 J m−3) and stratified waters (50–100 J m−3), but slower in mixed waters (< 7 J m−3). GAM modelling showed that the faster larval growth occurred in partially stratified waters with temperatures above 7.5°C. Natural mortality rates were higher in partially stratified waters (24% of daily loss), lower in stratified areas (14%) and the lowest in the mixed water column (12%). Accordingly, larvae living in channels had faster growth rates but higher daily mortality, while those that hatched or were advected to water parcels on the continental shelf had slower growth but lower daily mortality. Recently, certain biological field experiments and research findings have revealed that predators not only reduce prey populations through direct predation (lethal effect) but also indirectly affect the growth rate of prey by inducing fear; these nonlethal effects can be carried over seasons or generations. In this paper, we proposed an autonomous and a nonautonomous Leslie–Gower model incorporating some biological factors such as fear and its carry‐over effects, prey refuge, and nonlinear predator harvesting. In the autonomous model, first, we examined the well‐posedness, positivity solutions, and their boundedness. Also, it is shown that new equilibrium points emerge and disappear with the change in intrinsic growth rate of predator. Further, we compute and analyzed the local and global stability at the interior equilibrium points. Furthermore, Hopf bifurcation and direction and stability of limit cycle at interior equilibrium points are established. Moreover, the sensitivity analysis of the biological parameters is carried out numerically with two statistical methods via Latin hypercube sampling and partial rank correlation coefficients. It was found that at the small values of fear factor, carry‐over effect, and refuge behavior of prey parameters, the autonomous system exhibits oscillatory behavior, whereas for small values of prey birth rate and harvesting effort, the system remains stable. However, when intrinsic growth rate of predator increases from a low value to a higher value, the system shows transition from stability to instability and back to stability. We also explore the effects of seasonal variations in biological parameters in the nonautonomous model. Additionally, we investigate the theoretical existence of positive periodic solutions using the continuation theorem. The influences of seasonal variations in some parameters such as prey's birth rate, fear, and its carry‐over effects; refuge behavior of prey; intrinsic growth rate of predator; and harvesting effort are then numerically investigated. Background: Regression describes the relationship of results from two analyzers, and the generated equation can be used to harmonize results. Point‐of‐care (POC) analyzers cannot be calibrated by the end user, so regression offers an opportunity for calculated harmonization. Harmonization (uniformity) of laboratory results facilitates the use of common reference intervals and medical decision thresholds. Objective: Our aims were to characterize the relationship of results for multiple biochemistry analytes on a POC and a commercial laboratory analyzer (CL) with three regression techniques and to use regression equations to harmonize the POC results with those of the CL. Harmonized results were assessed by recognized quality goals. We used harmonized results to assess the regression techniques. Methods: After analyzer imprecision assessments, paired clinical samples were assessed with one dataset to calculate regression parameters that were applied to a second dataset. Three regression techniques were performed, and each was used to harmonize the POC results with those from the CL. POC results were assessed for bias and the number of results reaching quality goals before and after harmonization. Results: All regression techniques could be used to harmonize most analytes so that 95% of results were within ASVCP TEa guidelines. Harmonization could be further improved with alternate regression techniques or exclusions. Conclusions: Regression offers a means to harmonize POC and CL analyzers. Further work is needed to assess how few samples can reliably be used and to assess likely species differences. Background: Point‐of‐care (POC) biochemistry analyzers are widely used in small animal clinical practice but infrequently independently assessed for performance. Objective: To assess the performance of two current model point‐of‐care biochemistry analyzers (Heska Element DC and IDEXX Catalyst) compared with a commercial laboratory analyzer (Cobas 8000). Methods: One hundred twenty‐one cats from a feline hospital population were sampled with plasma results from a single lithium heparin tube assessed on all three analyzers. Plasma biochemistry results from each POC analyzer were compared with the commercial laboratory analyzer using Bland–Altman difference plots and by determining whether the limits of agreement (LOAs) (95% of differences) fell within various quality goals after correcting for inherent bias. Results: Only 7 of 14 analytes on the Heska analyzer and 2 analytes on the IDEXX analyzer attained the most stringent LOA quality goal, which was being within desirable total error based on biologic variation (TEdes). The number of analytes achieving quality goals increased with less stringent standards such as American Society of Veterinary Clinical Pathologists allowable total error (ASVCP TEA) guidelines or if <95% of clinical comparisons reaching these quality goals is considered acceptable. Widespread bias was found between both POC analyzers and the commercial laboratory analyzer. Conclusions: The performance of both POC biochemistry analyzers was variable compared with a commercial laboratory analyzer. Performance goals were only able to be attained after the bias for each analyzer was accounted for by offsetting the LOAs and quality goals set by the mean bias for each analyte on each analyzer. Preclinical evidence implicating cannabinoid receptor 2 (CB2) in various diseases has led researchers to question whether CB2 genetics influence aetiology or progression. Associations between conditions and genetic loci are often studied via single nucleotide polymorphism (SNP) prevalence in case versus control populations. In the CNR2 coding exon, ~36 SNPs have high overall population prevalence (minor allele frequencies [MAF] ~37%), including non‐synonymous SNP (ns‐SNP) rs2501432 encoding CB2 63Q/R. Interspersed are ~27 lower frequency SNPs, four being ns‐SNPs. CNR2 introns also harbour numerous SNPs. This review summarises CB2 ns‐SNP molecular pharmacology and evaluates evidence from ~70 studies investigating CB2 genetic variants with proposed linkage to disease. Although CNR2 genetic variation has been associated with a wide variety of conditions, including osteoporosis, immune‐related disorders, and mental illnesses, further work is required to robustly validate CNR2 disease links and clarify specific mechanisms linking CNR2 genetic variation to disease pathophysiology and potential drug responses. Biological hallmarks of splenic marginal zone lymphoma (SMZL) remain poorly described. Herein, we performed in‐depth SMZL characterization through multimodal single‐cell analyses of paired blood/spleen samples. The 3'‐single‐cell RNA‐sequencing, Cellular Indexing of Transcriptomes and Epitopes by sequencing, and 5'‐V(D)J single‐cell RNA‐sequencing datasets were integrated to characterize SMZL transcriptome profiles, including B‐cell receptor and T‐cell receptor repertoires. Hyperexpanded B‐cell clones in the spleen were at a memory‐like stage, whereas recirculating tumor B‐cells in blood encompassed multiple differentiation stages, indicating an unexpected desynchronization of the B‐cell maturation program in SMZL cells. Spatial transcriptomics showed the enrichment of T‐effector and T‐follicular helper (TFH) signatures in the nodular subtype of SMZL. This latter also exhibited gene‐based cell–cell interactions suggestive of dynamic crosstalk between TFH and cancer cells in transcriptomics, further substantiated by using imaging mass cytometry. Our findings provide a comprehensive high‐resolution description of SMZL biological hallmarks and characterize, for the first time in situ, inter‐ and intra‐patient heterogeneity at both transcriptomic and protein levels. © 2024 The Authors. Integrating multiple single‐cell datasets is essential for the comprehensive understanding of cell heterogeneity. Batch effect is the undesired systematic variations among technologies or experimental laboratories that distort biological signals and hinder the integration of single‐cell datasets. However, existing methods typically rely on a selected dataset as a reference, leading to inconsistent integration performance using different references, or embed cells into uninterpretable low‐dimensional feature space. To overcome these limitations, a reference‐free method, Beaconet, for integrating multiple single‐cell transcriptomic datasets in original molecular space by aligning the global distribution of each batch using an adversarial correction network is presented. Through extensive comparisons with 13 state‐of‐the‐art methods, it is demonstrated that Beaconet can effectively remove batch effect while preserving biological variations and is superior to existing unsupervised methods using all possible references in overall performance. Furthermore, Beaconet performs integration in the original molecular feature space, enabling the characterization of cell types and downstream differential expression analysis directly using integrated data with gene‐expression features. Additionally, when applying to large‐scale atlas data integration, Beaconet shows notable advantages in both time‐ and space‐efficiencies. In summary, Beaconet serves as an effective and efficient batch effect removal tool that can facilitate the integration of single‐cell datasets in a reference‐free and molecular feature‐preserved mode. Background and Purpose: Conclusions from prior literature regarding the impact of sex, age, and height on spinal cord (SC) MRI morphometrics are conflicting, while the effect of body weight on SC morphometrics has been found to be nonsignificant. The purpose of this case‐control study is to assess the associations between cervical SC MRI morphometric parameters and age, sex, height, and weight to establish their potential role as confounding variables in a clinical study of people with multiple sclerosis (MS) compared to a cohort of healthy volunteers. Methods: Sixty‐nine healthy volunteers and 31 people with MS underwent cervical SC MRI at 3 Tesla field strength. Images were centered at the C3/C4 intervertebral disc and processed using Spinal Cord Toolbox v.4.0.2. Mixed‐effects linear regression models were used to evaluate the effects of biological variables and disease status on morphometric parameters. Results: Sex, age, and height had significant effects on cord and gray matter (GM) cross‐sectional area (CSA) as well as the GM:cord CSA ratio. There were no significant effects of body weight on morphometric parameters. The effect of MS disease duration on cord CSA in the C4 level was significant when controlling for all other variables. Conclusions: Studies of disease‐related changes in SC morphometry should control for sex, age, and height to account for physiological variation. [ABSTRACT FROM AUTHOR]

Myalgic encephalomyelitis/chronic fatigue syndrome (ME/CFS) is a chronic disease presenting with severe fatigue, post‐exertional malaise, and cognitive disturbances—among a spectrum of symptoms—that collectively render the patient housebound or bedbound. Epigenetic studies in ME/CFS collectively confirm alterations and/or malfunctions in cellular and organismal physiology associated with immune responses, cellular metabolism, cell death and proliferation, and neuronal and endothelial cell function. The sudden onset of ME/CFS follows a major stress factor that, in approximately 70% of cases, involves viral infection, and ME/CFS symptoms overlap with those of long COVID. Viruses primarily linked to ME/CFS pathology are the symbiotic herpesviruses, which follow a bivalent latent–lytic lifecycle. The complex interaction between viruses and hosts involves strategies from both sides: immune evasion and persistence by the viruses, and immune activation and viral clearance by the host. This dynamic interaction is imperative for herpesviruses that facilitate their persistence through epigenetic regulation of their own and the host genome. In the current article, we provide an overview of the epigenetic signatures demonstrated in ME/CFS and focus on the potential strategies that latent viruses—particularly Epstein–Barr virus—may employ in long‐term epigenetic reprograming in ME/CFS. Epigenetic studies could aid in elucidating relevant biological pathways impacted in ME/CFS and reflect the physiological variations among the patients that stem from environmental triggers, including exogenous viruses and/or altered viral activity. Humans have substantially transformed the global land surface, resulting in the decline in variation in biotic communities across scales, a phenomenon known as "biological homogenization." However, different biota are affected by biological homogenization to varying degrees, but this variation and the underlying mechanisms remain little studied, particularly in soil systems. To address this topic, we used metabarcoding to investigate the biogeography of soil protists and their prey/hosts (prokaryotes, fungi, and meso‐ and macrofauna) in three human land‐use ecosystem types (farmlands, residential areas, and parks) and natural forest ecosystems across subtropical and temperate regions in China. Our results showed that the degree of community homogenization largely differed between taxa and functional groups of soil protists, and was strongly and positively linked to their colonization ability of human land‐use systems. Removal analysis showed that the introduction of widespread, generalist taxa (OTUs, operational taxonomic units) rather than the loss of narrow‐ranged, specialist OTUs was the major cause of biological homogenization. This increase in generalist OTUs seemingly alleviated the negative impact of land use on specialist taxa, but carried the risk of losing functional diversity. Finally, homogenization of prey/host biota and environmental conditions were also important drivers of biological homogenization in human land‐use systems, with their importance being more pronounced in phagotrophic than parasitic and phototrophic protists. Overall, our study showed that the variation in biological homogenization strongly depends on the colonization ability of taxa in human land‐use systems, but is also affected by the homogenization of resources and environmental conditions. Importantly, biological homogenization is not the major cause of the decline in the diversity of soil protists, and conservation and study efforts should target at taxa highly sensitive to local extinction, such as parasites. Cereals are some of the most important global crops that contribute directly and indirectly to the production of food for human consumption. Cereal aphids can cause significant damage to wheat, barley and oats, particularly via the transmission of plant viruses that cause devastating plant diseases, such as yellow dwarf disease. High levels of yellow dwarf disease can result in yield losses of around 20%, rising to 80% if infection is severe. Yellow dwarf disease is caused by multiple viruses, including viruses within the families Tombusviridae and Solemoviridae. These include yellow dwarf virus species within the genus Luteovirus (Barley yellow dwarf virus) and Polerovirus (Cereal yellow dwarf virus, Wheat yellow dwarf virus, Maize yellow dwarf virus). Some yellow dwarf virus species are primarily vectored by one aphid species whereas others can be transmitted by multiple vectors. Biological diversity within a given vector species (e.g., genotype, biotype) can influence virus transmission efficiency. However, it is unclear what biological factors drive this variation within a given vector species. Understanding how biological variation in vector populations influences virus transmission efficiency can help to identify biological traits that underpin successful transmission in competent vector populations. Here, the available literature on yellow dwarf virus transmission efficiency is synthesized and significant variation in yellow dwarf virus transmission efficiency is detected between different populations for several vector species. Background: Biochemical measurements are commonly evaluated using populationbased reference intervals; however, there is a growing trend toward reassessing results with within-subject variation (CVI). Objectives: We aimed to estimate the CVI of 16 biochemical analytes using a large database of dogs and cats, which refers to the results of routine health checkups. Methods: Pairs of sequential results for 16 analytes were extracted from a database of adult patients. The second result was divided by the first result to produce the ratio of sequential results (rr), and the frequency distribution of rr was plotted. From the plots, the coefficient of variation (CVrr) was calculated. Analytical variation (CVA) was calculated using quality control data, and CVI was estimated as follows: CVI = ([CVrr∕21∕2]² −CVA²)1∕2. Estimated CVI was compared with previously reported CVI using the Bland–Altman plot analysis. Results: From the database, 9078 data points from 3610 dogs and 3743 data points from 1473 cats were extracted, with 5468 data pairs for dogs and 2270 for cats. Sampling intervals ranged from 10 to 1970 days (median 366) for dogs and 23 to 1862 days (median 365) for cats. Bland–Altman analysis showed most CVI plots fell within the limits of agreement; however, positive fixed biases were observed in both dogs and cats. Conclusions: Our study introduces a novel approach of estimating CVI using routine health checkup data in dogs and cats. In this work, two new biscyclometalated Ir(III) complexes with diimine ligands were synthesized. The compounds were characterized by 1D and 2D NMR spectroscopy and HR ESI mass spectrometry. All complexes exhibit efficient phosphorescence with pronounced sensitivity to the presence of oxygen. The photophysical properties of the obtained compounds, including absorption, emission spectra, lifetimes and quantum yields of phosphorescence were measured in various aqueous and model biological media with variations in pH, temperature and at various O2 concentrations. The complexes exhibit a very good sensory response to the changes in oxygen concentration, resulting in an increase in quantum yields and lifetimes by 2.8–5.5 times upon oxygen removal from aerated aqueous solutions. Biological tests with CHO‐K1 cell line have shown that these compounds display low toxicity, rapid internalization into cells and localization predominantly in lysosomes. For the most promising complex the phosphorescence lifetime imaging experiments were conducted, revealing that this sensor markedly changes the phosphorescence lifetime values in cells from 1.8 to 4.1 μs upon transition from normoxia to simulated hypoxia. The obtained results indicate that this type of iridium chromophores can be effectively used for assessing the oxygen status of living objects by using time‐resolved luminescent microscopy. The ability to sex individuals is an important component of many behavioural and ecological investigations and provides information for demographic models used in conservation and species management. However, many birds are difficult to sex using morphological characters or traditional molecular sexing methods. In this study, we developed probabilistic models for sexing birds using quantitative PCR (qPCR) data. First, we quantified distributions of gene copy numbers at a set of six sex‐linked genes, including the sex‐determining gene DMRT1, for individuals across 17 species and seven orders of birds (n = 150). Using these data, we built predictive logistic models for sex identification and tested their performance with independent samples from 51 species and 13 orders (n = 209). Models using the two loci most highly correlated with sex had greater accuracy than models using the full set of sex‐linked loci, across all taxonomic levels of analysis. Sex identification was highly accurate when individuals to be assigned were of species used in model building. Our analytical approach was widely applicable across diverse neognath bird lineages spanning millions of years of evolutionary divergence. Unlike previous methods, our probabilistic framework incorporates uncertainty around qPCR measurements as well as biological variation within species into decision‐making rules. We anticipate that this method will be useful for sexing birds, including those of high conservation concern and/or subsistence value, that have proven difficult to sex using traditional approaches. Introduction: Establish the referenceintervals (RIs) and analyze biological variability (BV) to introduce the thrombin generation test (TGT) into clinical practice. Methods: To determine the RIs parameters of TGT, we analyzed platelet‐poor plasma (PPP) (n = 123), rich (PRP) (n = 76), and microparticle‐mediated TGT (MP‐TGT) (n = 32) in healthy participants. For the BV study, we collected samples from five participants over 5 weeks. A nested analysis of variance (ANOVA) was performed to evaluate the BV results. Results: The between‐individual variation (CVG), within‐individual variation (CVI), analytical variation (CVA) for TGT on PPP for all parameters were from 5.5% to 17.3%, 5.4% to 17.7%, and 2.6% to 5.3%, respectively. For PRP, the CVG, CVI, and CVA were ranged from 3.0% to 23.7%, 8.4% to 23.0%, and 4.1% to 6.9%, respectively. The index of individuality (II) ranged from 0.3 to 3.1 for PPP and from 0.3 to 4.5 for PRP. The reference change value (RCV) for PPP was from 19.8% to 50.1%, while for PRP, it was 27.2% to 66.5%. We recommend using the RIs for the parameters ETP (nM/min): 1101.6–2332.1 and Peak (nM): 163.5–381.3 for PPP and ETP (nM/min): 1088.5–2634.9; Peak (nM): 72.6–210.7 for PRP. The resulting MP‐TGT are highly dependent on age require a larger sample. Conclusion: For TGT on PPP and PRP the RIs developed on our population for Peak and ETP parameters can be used. This article discusses a case of pseudomyogenic hemangioendothelioma (PHE), a type of vascular tumor, with a specific gene fusion called EGFL7:FOSB. The first case of PHE with this fusion showed widespread metastasis, while the second case presented with a more typical clinical presentation without evidence of distant tumors or metastasis. Colour pattern variation provides biological information in fields ranging from disease ecology to speciation dynamics. Comparing colour pattern geometries across images requires colour segmentation, where pixels in an image are assigned to one of a set of colour classes shared by all images. Manual methods for colour segmentation are slow and subjective, while automated methods can struggle with high technical variation in aggregate image sets. We present recolorize, an R package toolbox for human‐subjective colour segmentation with functions for batch‐processing low‐variation image sets and additional tools for handling images from diverse (high‐variation) sources. The package also includes export options for a variety of formats and colour analysis packages. This paper illustrates recolorize for three example datasets, including high variation, batch processing and combining with reflectance spectra, and demonstrates the downstream use of methods that rely on this output. INTRODUCTION: Blood biomarkers have proven useful in Alzheimer's disease (AD) research. However, little is known about their biological variation (BV), which improves the interpretation of individual‐level data. METHODS: We measured plasma amyloid beta (Aβ42, Aβ40), phosphorylated tau (p‐tau181, p‐tau217, p‐tau231), glial fibrillary acidic protein (GFAP), and neurofilament light chain (NfL) in plasma samples collected weekly over 10 weeks from 20 participants aged 40 to 60 years from the European Biological Variation Study. We estimated within‐ (CVI) and between‐subject (CVG) BV, analytical variation, and reference change values (RCV). RESULTS: Biomarkers presented considerable variability in CVI and CVG. Aβ42/Aβ40 had the lowest CVI (≈ 3%) and p‐tau181 the highest (≈ 16%), while others ranged from 6% to 10%. Most RCVs ranged from 20% to 30% (decrease) and 25% to 40% (increase). DISCUSSION: BV estimates for AD plasma biomarkers can potentially refine their clinical and research interpretation. RCVs might be useful for detecting significant changes between serial measurements when monitoring early disease progression or interventions. Numerous biodiversity–ecosystem functioning (BEF) experiments have shown that plant community productivity typically increases with species diversity. In these studies, diversity is generally quantified using metrics of taxonomic, phylogenetic, or functional differences among community members. Research has also shown that the relationships between species diversity and functioning depends on the spatial scale considered, primarily because larger areas may contain different ecosystem types and span gradients in environmental conditions, which result in a turnover of the species set present locally. A fact that has received little attention, however, is that ecological systems are hierarchically structured, from genes to individuals to communities to entire landscapes, and that additional biological variation occurs at levels of organization above and below those typically considered in BEF research. Here, we present cases of diversity effects at different hierarchical levels of organization and compare these to the species‐diversity effects traditionally studied. We argue that when this evidence is combined across levels, a general framework emerges that allows the transfer of insights and concepts between traditionally disparate disciplines. Environmental variation can influence the reproductive success of species managed under human care and in the wild, yet the mechanisms underlying this phenomenon remain largely mysterious. Molecular mechanisms such as epigenetic modifiers are important in mediating the timing and progression of reproduction in humans and model organisms, but few studies have linked epigenetic variation to reproductive fitness in wildlife. Here, we investigated epigenetic variation in black‐footed ferrets (Mustela nigripes), an endangered North American mammal reliant on ex situ management for survival and persistence in the wild. Despite similar levels of genetic diversity in human‐managed and wild‐born populations, individuals in ex situ facilities exhibit reproductive problems, such as poor sperm quality. Differences across these settings suggest that an environmentally driven decline in reproductive capacity may be occurring in this species. We examined the role of DNA methylation, one well‐studied epigenetic modifier, in this emergent condition. We leveraged blood, testes, and semen samples from male black‐footed ferrets bred in ex situ facilities and found tissue‐type specificity in DNA methylation across the genome, although 1360 Gene Ontology terms associated with male average litter size shared functions across tissues. We then constructed gene networks of differentially methylated genomic sites associated with three different reproductive phenotypes to explore the putative biological impact of variation in DNA methylation. Sperm gene networks associated with average litter size and sperm count were functionally enriched for candidate genes involved in reproduction, development, and its regulation through transcriptional repression. We propose that DNA methylation plays an important role in regulating these reproductive phenotypes, thereby impacting the fertility of male ex situ individuals. Our results provide information into how DNA methylation may function in the alteration of reproductive pathways and phenotypes in artificial environments. Previous studies have shown that females typically outperform males on episodic memory tasks. In this study, we investigated if (1) there are differences between males and females in their connectome characteristics, (2) if these connectivity patterns are associated with memory performance, and (3) if these brain connectome characteristics contribute to the differences in episodic memory performance between sexes. In a sample of 655 healthy young subjects (n = 391 females; n = 264 males), we derived brain network characteristics from diffusion‐weighted imaging (DWI) data using models of crossing fibers within each voxel of the brain and probabilistic tractography (graph strength, shortest path length, global efficiency, and weighted transitivity). Group differences were analysed with linear models and mediation analyses were used to explore how connectivity patterns might relate to sex‐dependent differences in memory performance. Our results show significant sex‐dependent differences in weighted transitivity (d = 0.42), with males showing higher values. Further, we observed a negative association between weighted transitivity and memory performance (r = −0.12). Finally, these distinct connectome characteristics partially mediated the observed differences in memory performance (effect size of the indirect effect r = 0.02). Our findings indicate a higher interconnectedness in females compared to males. Additionally, we demonstrate that the sex‐dependent differences in episodic memory performance can be partially explained by the differences in this connectome measure. In medical, veterinary and forensic entomology, the ease and affordability of image data acquisition have resulted in whole‐image analysis becoming an invaluable approach for species identification. Krawtchouk moment invariants are a classical mathematical transformation that can extract local features from an image, thus allowing subtle species‐specific biological variations to be accentuated for subsequent analyses. We extracted Krawtchouk moment invariant features from binarised wing images of 759 male fly specimens from the Calliphoridae, Sarcophagidae and Muscidae families (13 species and a species variant). Subsequently, we trained the Generalized, Unbiased, Interaction Detection and Estimation random forests classifier using linear discriminants derived from these features and inferred the species identity of specimens from the test samples. Fivefold cross‐validation results show a 98.56 ± 0.38% (standard error) mean identification accuracy at the family level and a 91.04 ± 1.33% mean identification accuracy at the species level. The mean F1‐score of 0.89 ± 0.02 reflects good balance of precision and recall properties of the model. The present study consolidates findings from previous small pilot studies of the usefulness of wing venation patterns for inferring species identities. Thus, the stage is set for the development of a mature data analytic ecosystem for routine computer image‐based identification of fly species that are of medical, veterinary and forensic importance. The imprint left by niche evolution on the variation of biological diversity across spatial and environmental gradients is still debated among ecologists. Furthermore, understanding to what extent dispersal limitation may reinforce or blur such imprint is still a gap in the ecological knowledge. In this article we introduce a simulation approach coupled to approximate Bayesian computation (ABC) that parameterizes both the adaptation rate of species' niche positions over the evolution of a monophyletic lineage and the intensity of dispersal limitation associated with the variation of species alpha diversity among assemblages distributed across spatial and environmental gradients. The analytical tool was implemented in the R package 'mcfly' (www.r‐project.org). We evaluated the statistical performance of the analytical framework using simulated datasets, which confirmed the suitability of the analysis to estimate adaptation rate parameter but showed to be less precise in relation to the dispersal limitation parameter. Also, we found that increased dispersal limitation levels improved the parameterization of the adaptation rate of species' niche positions in simulated datasets. Further, we evaluated the role played by niche evolution and dispersal limitation on species alpha diversity variation of Phyllostomidae bats across the Neotropics. The framework proposed here shed light on the links between niche evolution, dispersal limitation and gradients of biological diversity, and thereby improved our understanding of evolutionary imprints on current biological diversity patterns. Objective: Interpreting thyroid function tests can be challenging due to inherent variation, and the need for tests rises with age. While age‐related changes in thyrotropin (TSH) levels are known, the biological variation in older adults remains unclear. Design: We recruited nineteen 65–99‐year‐old (older adults) without thyroid disease for monthly blood sampling for 1 year. Patients and Measurements: Serum was stored at −20C°, and TSH, total thyroxine (TT4) and total triiodothyronine (TT3) were analysed in random order in a single batch for each participant. Results were compared to test results from 15 euthyroid men aged 24–53 years (younger adults) collected previously using a similar methodology. Results: Interindividual coefficients of variation in older/younger adults were 46.7%/44.0% for TSH, 12.7%/19.5% for TT4 and 14.6%/22.4% for TT3. Intraindividual coefficients of variation (CVI) were 19.0%/25.4% for TSH, 5.5%/10.8% for TT4 and 6.9%/13.2% for TT3. The index of individuality was below 0.6 for all hormones in all age groups. The number of samples required to determine the homoeostatic set‐point at 10% precision in older adults was 14–21 for TSH and 2 for TT4 and TT3. TT4 in older adults was the only parameter in any group with comparable CVI between individuals (p =.22). Conclusions: CVI for TT4 and TT3 was halved in older compared to younger adults with two tests of TT4 needed to describe the individual set‐point. Similar CVI between older adults caused TT4 to provide a reliable estimate of thyroid function, and the added value of measuring thyroxine could improve clinical practice. [ABSTRACT FROM AUTHOR]

An individual's telomere length early in life may reflect or contribute to key life‐history processes sensitive to environmental variation. Yet, the relative importance of genetic and environmental factors in shaping early‐life telomere length is not well understood as it requires samples collected from multiple generations with known developmental histories. We used a confirmed pedigree and conducted an animal model analysis of telomere lengths obtained from nestling house sparrows (Passer domesticus) sampled over a span of 22 years. We found significant additive genetic variation for early‐life telomere length, but it comprised a small proportion (9%) of the total biological variation. Three sources of environmental variation were important: among cohorts, among‐breeding attempts within years, and among nestmates. The magnitude of variation among breeding attempts and among nestmates also differed by cohort, suggesting that interactive effects of environmental factors across time or spatial scales were important, yet we were unable to identify the specific causes of these interactions. The mean amount of precipitation during the breeding season positively predicted telomere length, but neither weather during a given breeding attempt nor date in the breeding season contributed to an offspring's telomere length. At the level of individual nestlings, offspring sex, size and mass at 10 days of age also did not predict telomere length. Environmental effects appear especially important in shaping early‐life telomere length in some species, and more focus on how environmental factors that interact across scales may help to explain some of the variation observed among studies. Background: In treatment planning for proton therapy a constant Relative Biological Effectiveness (RBE) of 1.1 is used, disregarding variations with linear energy transfer, clinical endpoint, or fractionation. Purpose: To present a methodology to analyze the variation of RBE with fractionation from clinical data of tumor control probability (TCP) and to apply it to study the response of prostate cancer to proton therapy. Methods and materials: We analyzed the dependence of the RBE on the dose per fraction by using the LQ model and the Poisson TCP formalism. Clinical tumor control probabilities for prostate cancer (low and intermediate risk) treated with photon and proton therapy for conventional fractionation (2 Gy(RBE)×37 fractions), moderate hypofractionation (3 Gy(RBE)×20 fractions) and hypofractionation (7.25 Gy(RBE)×5 fractions) were obtained from the literature and analyzed aiming at obtaining the RBE and its dependence on the dose per fraction. Results: The theoretical analysis of the dependence of the RBE on the dose per fraction showed three distinct regions with RBE monotonically decreasing, increasing or staying constant with the dose per fraction, depending on the change of (α, β) values between photon and proton irradiation (the equilibrium point being at (αp/βp) = (αX/βX)(αX/αp)). An analysis of the clinical data showed RBE values that decline with increasing dose per fraction: for low risk RBE≈1.124, 1.119, and 1.102 for 1.82 Gy, 2.73 Gy and 6.59 Gy per fraction (physical proton doses), respectively; for intermediate risk RBE≈1.119 and 1.102 for 1.82 Gy and 6.59 Gy per fraction (physical proton doses), respectively. These values are nonetheless very close to the nominal 1.1 value. Conclusions: In this study, we have presented a methodology to analyze the RBE for different fractionations, and we used it to study clinical data for prostate cancer and evaluate the RBE versus dose per fraction. The analysis shows a monotonically decreasing RBE with increasing dose per fraction, which is expected from the LQ formalism and the changes in (α, β) values between photon and proton irradiation. However, the calculations in this study have to be considered with care as they may be biased by limitations in the modeling assumptions and/or by the clinical data set used for the analysis. Background: Laboratory results are influenced by presence and severity of disease, as well as preanalytical factors, analytical variation, and biological variation. Biological variation data for urinary protein: creatinine ratio (UPC) and urine specific gravity (USG) in cats are lacking. Objectives: Determine the biological variation of UPC and USG in cats. Animals: Eighty healthy client‐owned cats. Methods: Prospective study. Urine was collected on days 0, 14, and 56 from all 80 cats to investigate the persistence of borderline or overt proteinuria or suboptimal urine concentration. In 15 of these cats, urine was collected weekly from day 0 to 42 to calculate the index of individuality (II) and reference change value (RCV), and on days 56 and 57 to evaluate day‐to‐day variability of UPC and USG. Results: Borderline or overt proteinuria (UPC ≥0.2) was present in 18/80 (23%) cats at baseline and persisted on 3 occasions in 2 months in 8/18 (44%) cats. Urine concentration was suboptimal at inclusion (USG <1.035) in 8/80 (10%) cats and at all 3 time points during 2 months in 3/8 (38%) cats. The II of UPC and USG indicated intermediate individuality. The 1‐sided RCV was 82% for UPC and 36% for USG. Proteinuria substage was identical on 2 consecutive days in 13/15 (87%) cats, and urine concentrating ability remained the same in all 15 cats. Conclusions and Clinical Importance: A >82% increase in UPC in a healthy cat is not solely attributable to physiological and analytical variation. The response of immunogenecity anti‐drug antibody (ADA) generally includes biological and analytical variability. The nature of biological and analytical variations may lead to a variety of symmetric and asymmetric ADA data. As a result, current statistical methods may yield unreliable results because these methods assume special types of symmetric or asymmetric ADA data. In this paper, we survey and compare parametric models that are useful for analyzing a variety of asymmetric data that have rarely been used to calculate assay cut points. These models include symmetric distributions as limiting case; therefore, they are useful in the analysis of a variety of symmetric data. We also investigate two nonparametric approaches that have received little attention in screening cut point calculations. A simulation study was conducted to compare the performance of the methods. Background and Objectives: Room temperature‐stored platelets (RTPs) maximize platelet viability but limit shelf life. The aims of this study were to investigate the impact of donor variability on cold‐stored platelets (CSPs) and RTP, to determine whether RTP quality markers are appropriate for CSP. Materials and Methods: Double platelet donations (n = 10) were collected from consented regular male donors stored in 100% plasma. A full blood count, donor age, weight, height and body mass index (BMI) were collected at the time of donation. Platelet donations were split equally into two bags, and assigned to non‐agitated CSP or agitated RTP. The quality and function of platelets were assessed throughout the standard 7 days of storage and at expiry (day 8). Non‐parametric statistical analyses were used to analyse results given the small sample size. Results: As expected, there were significant differences between CSP and RTP throughout storage including a reduction in CSP concentration as well as a loss of swirling. Furthermore, a significant increase in CSP exhibiting activation and apoptotic markers was observed. Platelet concentrations were further impacted by donor BMI, and donors with the highest BMI (>29) had the lowest platelet concentration and activation response at the end of CSP storage. Conclusion: Platelet quality and functionality play a vital role in transfusion outcomes; however, blood components are inherently variable. This study demonstrated, for the first time, the specific impact of donor BMI on CSP quality and function and highlights the requirement for novel quality markers for assessing CSPs. Introduction: People with a low or high haemoglobin glycation index (HGI) have lower or higher HbA1c than other people with the same FPG. This study compared the prevalence of prediabetes based on FPG, 2hOGTT and HbA1c in people with low, moderate or high HGI. Methods: Prediabetes was diagnosed based on ADA cutpoints in 10,488 NHANES participants without self‐reported diabetes. HGI was calculated as the difference between a participant's observed HbA1c and a predicted HbA1c where predicted HbA1c = 0.024 FPG + 3.1. Participants were divided into low (HGI < −0.15%), moderate (HGI −0.15% to +0.15%) and high (HGI > +0.15%) HGI subgroups. Results: The prevalence of prediabetes was 42.4% based on FPG, 27.2% based on HbA1c and 17.2% based on 2hOGTT. FPG and HbA1c thus overdiagnosed prediabetes by 25.2% and 10.0%, respectively, compared to the OGTT gold standard. Prevalence was (1) similar in low, moderate and high HGI participants based on 2hOGTT, (2) highest in low HGI participants based on FPG, and (3) highest in high HGI participants based on HbA1c. Among participants with mismatched FPG and HbA1c, OGTT was normal in (1) 79.5% of participants with normal FPG but prediabetic HbA1c (mean HGI = +0.53%), and (2) 75.2% of participants with normal HbA1c but prediabetic FPG (mean HGI = −0.30%). Conclusions: FPG overdiagnosed prediabetes in people with low HGI. HbA1c overdiagnosed prediabetes in people with high HGI. Background: Cats commonly develop thyroid disease but little is known about the long‐term biological variability of serum thyroid hormone and thyrotropin (thyroid‐stimulating hormone; TSH) concentrations. Objectives: We aimed to determine the long‐term biological variation of thyroid hormones and TSH in clinically healthy cats. Methods: A prospective, observational study was carried out. Serum samples for analysis of total thyroxine (T4, by radioimmunoassay [RIA] and homogenous enzyme immunoassay [EIA]), triiodothyronine (T3), free T4 (by dialysis), and TSH were obtained every 8 weeks for 1 year from 15 healthy cats, then frozen until single‐batch analysis. Coefficients of variation (CV) within individual cats (CVI) and among individual cats (CVG), as well as the variation between duplicates (ie, analytical variation [CVA]) were determined with restricted maximum likelihood estimation. The indices of individuality (IoI) and reference change values (RCVs) for each hormone were calculated. Results: Some thyroid hormones showed similar (total T4 by EIA) or greater (TSH) interindividual relative to intraindividual variation resulting in intermediate to high IoI, consistent with previous studies evaluating the biological variation of these hormones weekly for 5–6 weeks. By contrast, total T4 (by RIA) and free T4 had a low IoI. Total T3 had a high ratio of CVA to CVI; therefore, interindividual variation could not be distinguished from analytical variation. No seasonal variability in the hormones could be demonstrated. Conclusions: Clinicians might improve the diagnosis of feline thyroid disease by establishing baseline concentrations for analytes with intermediate‐high IoI (total T4, TSH) for individual cats and applying RCVs to subsequent measurements. Background: Biological variation (BV) of urinary (U) biochemical analytes has not been described in absolute terms, let alone as a ratio of the U‐creatinine or fractional excretion in healthy dogs. These analytes are potential diagnostic tools for different types of kidney damage and electrolyte disorders in dogs. Objectives: We aimed to investigate the BV of specific gravity, osmolality, creatinine, urea, protein, glucose, chloride, sodium, potassium, calcium, and phosphate in urine from healthy pet dogs. Methods: Blood and urine samples from 13 dogs were collected once weekly for 8 weeks. Samples were analyzed in duplicate and in randomized order. For each sample, U‐analyte and serum concentrations were measured, and U‐analyte/U‐creatinine and fractional excretion (FE) were calculated. Components of variance, estimated by restricted maximum likelihood, were used to determine within‐subject variation (CVI), between‐subject variation (CVG), and analytical variation (CVA). Index of individuality (II) and reference change values were calculated. Results: CVI for all urine analytes varied between 12.6% and 35.9%, except for U‐sodium, U‐sodium/U‐Cr, and FE‐sodium, which had higher CVIs (59.5%‐60.7%). For U‐protein, U‐sodium, U‐potassium, U‐sodium/U‐creatinine, FE‐urea, FE‐glucose, FE‐sodium, FE‐potassium, and FE‐phosphate II were low, indicating that population‐based RIs were appropriate. The remaining analytes had an intermediate II, suggesting that population‐based RIs should be used with caution. Conclusion: This study presents information on the biological variation of urinary and serum biochemical analytes from healthy dogs. Genealogical samples that couple dental data and documented relatedness information provide unique opportunities to examine the biological foundations of tooth variation. Over the past century, these resources have been critical for examining the various factors that influence dental phenotypes—the same traits that anthropologists regularly apply to reconstructions of past phenomena. Genealogical samples are uniquely suited to test long‐standing assumptions underlying bioanthropological practice, for example, biodistance and phylogenetic analysis, which commonly reference aspects of tooth size and form as proxies for latent genetic information. This article provides an overview of published genealogical research, with a focus on the practical implications of quantitative genetic and environmental studies of (non)human primate dentitions. To highlight the utility of genealogical samples for understanding the influence of specific non‐genetic factors on dental characters, we also present novel data on gestational hormone effects in opposite‐sex dizygotic twin pairs as a test of the twin testosterone transfer (TTT) hypothesis. This article discusses fruitful next steps in genealogical dental research, as well as important ethical considerations surrounding the use of associated datasets, which are sensitive in nature. As we forge ahead in an age of phenomics, genealogical samples are likely to play a key role in generating comprehensive genotype–phenotype maps of the dentition and in refining bioanthropological methods. Monitoring programmes worldwide use biota to assess the "health" of water bodies. Indices based on biota are used to describe the change in status of sites over time, to identify progress against management targets and to diagnose the causes of biological degradation. A variety of numerical stressor‐specific biotic indices have been developed based on the response of biota to differences in stressors among sites. Yet, it is not clear how variation in pressures within sites, over what time period, and in what combination has the greatest impact on different biotic groups. An understanding of how temporal variation in pressures influences biological assessment indices would assist in setting achievable targets and help focus catchment‐scale mitigation strategies to ensure that they deliver the desired improvements in biological condition.Hydrochemical data provided by a network of high‐frequency (15 or 30 min) automated monitoring stations over 3 years were matched to replicated biological data to understand the influence of spatio‐temporal variation in pollution pressures on biological indices. Hydrochemical data were summarised in various ways to reflect central tendency, peaks, troughs and variation over 1–90 days before the collection of each biological sample. An objective model selection procedure was used to determine which hydrochemical determinand, and over what time period, best explained variation in the biological indices.Stressor‐specific indices derived from macroinvertebrates which purportedly assess stress from low flows, excess fine sediment, nutrient enrichment, pesticides and organic pollution were significantly inter‐correlated and reflected periods of low oxygen concentration, even though only one index (ASPTWHPT, average score per taxon) was designed for this purpose. Changes in community composition resulting from one stressor frequently lead to confounding effects on stressor‐specific indices.Variation in ASPTWHPT was best described by dissolved oxygen calculated as Q5 over 10 days, suggesting that low oxygen events had most influence over this period. Longer‐term effects were apparent, but were masked by recovery. Macroinvertebrate abundance was best described by Q95 of stream velocity over 60 days, suggesting a slower recovery in numbers than in the community trait reflected by ASPTWHPT.Although use of ASPTWHPT was supported, we recommend that additional independent evidence should be used to corroborate any conclusions regarding the causes of degradation drawn from the other stressor‐specific indices. The use of such stressor‐specific indices alone risks the mistargeting of management strategies if the putative stressor‐index approach is taken to be more reliable than the results herein suggest. Suboptimal conditions during development can shorten telomeres, the protective DNA caps on the end of chromosomes. Shorter early‐life telomere length (TL) can indicate reduced somatic maintenance, leading to lower survival and shorter lifespan. However, despite some clear evidence, not all studies show a relationship between early‐life TL and survival or lifespan, which may be due to differences in biology or study design (e.g., survival period measured). In superb fairy‐wrens (Malurus cyaneus), we assessed whether early‐life TL predicts mortality across different life‐history stages (fledgling, juvenile, adult). However, in contrast to a similar study on a congener, early‐life TL did not predict mortality across any life stage in this species. We then performed a meta‐analysis including 32 effect sizes from 23 studies (15 birds and three mammals) to quantify the effect of early‐life TL on mortality whilst taking into consideration potential sources of biological and methodological variation. Overall, the effect of early‐life TL on mortality was significant, corresponding to a 15% reduction in mortality risk with each standard deviation increase in TL. However, the effect became weaker when correcting for publication bias. Contrary to our predictions, there was no evidence that effects of early‐life TL on mortality varied with species lifespan or the period over which survival was measured. However, negative effects of early‐life TL on mortality risk were pervasive throughout life. These results imply that effects of early‐life TL on mortality are more likely to be context‐dependent than age‐dependent, although substantial power and publication bias issues highlight the need for more research. Ecogeographic rules describe spatial patterns in biological trait variation and shed light on the drivers of such variation. In animals, a consensus is emerging that 'pioneering' traits may facilitate range shifts via a set of bold, aggressive and stress‐resilient traits. Many of these same traits are associated with more northern latitudes, and most range shifts in the northern hemisphere indicate northward movement. As a consequence, it is unclear whether pioneering traits are simply corollaries of existing latitudinal variation, or whether they override other well‐trodden latitudinal patterning as a unique ecogeographic rule of phenotypic variation.The tree swallow Tachycineta bicolor is a songbird undergoing a southward range shift in the eastern United States, in direct opposition of the poleward movement seen in most other native species' range shifts. Because this organic range shift countervails the typical direction of movement, this case study provides for unique ecological insights on organisms and their ability to thrive in our changing world.We sampled female birds across seven populations, quantifying behavioural, physiological and morphological traits. We also used GIS and field data to quantify a core set of ecological factors with strong ties to these traits as well as female performance.Females at more southern expansion sites displayed higher maternal aggression, higher baseline corticosterone and more pronounced elevation of corticosterone following a standardized stressor, contrary to otherwise largely conserved latitudinal patterning in these traits. Microhabitat variation explained some quantitative phenotypic variation, but the expansion and historic ranges did not differ in openness, distance to water or breeding density.This countervailing range shift therefore suggests that pioneering traits are not simply corollaries of existing latitudinal variation, but rather, they may override other well‐trodden latitudinal patterning as a unique ecogeographic rule of phenotypic variation. The ecological variation in biological and adult life‐table attributes of two populations of Aedes aegypti (Diptera: Culicidae) from the desert (Jodhpur) and coastal (Kolkata) regions of India are assessed to understand the reproductive and survival strategies. The results showed that females lived longer than males in both strains. The desert strain was more r‐strategist because of its higher intrinsic rate of increase (rm = 0.23), finite rate of increase (λ = 1.25), lower life expectancy of males (7.9 days) and females (14.4 days), mean generation time (T = 19.2 days) and doubling time (DT = 3.0 days). However, there was no difference in net reproductive rate (R0) between the desert and coastal strains. The coastal strain showed a longer female life expectancy (22.0 days) than the desert strain (14.4 days). However, the fecundity (eggs/female/day) was lower in the coastal strain (11.4) than in the desert strain (15.1). Conclusively, the desert (Jodhpur) strain is adapted to a better r‐strategy than the coastal (Kolkata) strain of Ae. aegypti, which might be helpful to flourish in harsh environmental conditions. Background: Biological variation helps determine whether population‐based or subject‐based reference intervals are more appropriate to assess changes in serial analytical values. Previous studies have investigated the biological variation of biochemical analytes weekly or with variable frequency over 5‐14 weeks in cats, but none have considered biological variation at less frequent intervals over 1 year. Objectives: We aimed to evaluate the long‐term biological variation of 19 biochemical analytes in clinically healthy cats. Methods: A prospective, observational study in which 15 clinically healthy, client‐owned cats were sampled for serum biochemical analyses every 8 weeks for 1 year. Frozen serum samples were single‐batch analyzed. Restricted maximum likelihood estimation was used to determine the coefficients of variation (CV), describing variation within each cat, between cats, and the analytical variation. These CVs were used to determine the indices of individuality and reference change values (RCVs). Results: Albumin, alkaline phosphatase, creatine kinase, and globulin had high indices of individuality, indicating that they are best evaluated by RCVs. Phosphorus, potassium, chloride, sodium, symmetric dimethylarginine, and total CO2 had low indices of individuality, indicating that population‐based reference intervals are appropriate. Alanine aminotransferase, aspartate aminotransferase, blood urea nitrogen, calcium, cholesterol, creatinine, glucose, total bilirubin, and total protein had intermediate indices of individuality, indicating that RCVs may provide additional insight into the interpretation of analyte measurements beyond the population‐based reference intervals. Conclusions: For many analytes, the biological variation detected was similar to that reported in prior studies. Animal behavior is motivated by the fundamental need to feed and reproduce, and these behaviors can be inferred from spatiotemporal variations in biological signals such as vocalizations. Yet, linking foraging and reproductive effort to environmental drivers can be challenging for wide‐ranging predator species. Blue whales are acoustically active marine predators that produce two distinct vocalizations: song and D calls. We examined environmental correlates of these vocalizations using continuous recordings from five hydrophones in the South Taranaki Bight region of Aotearoa New Zealand to investigate call behavior relative to ocean conditions and infer life history patterns. D calls were strongly correlated with oceanographic drivers of upwelling in spring and summer, indicating associations with foraging effort. In contrast, song displayed a highly seasonal pattern with peak intensity in fall, which aligned with the timing of conception inferred from whaling records. Finally, during a marine heatwave, reduced foraging (inferred from D calls) was followed by lower reproductive effort (inferred from song intensity). [ABSTRACT FROM AUTHOR]

The RPMS1 gene is the only member of the BamHI‐A rightward transcripts (BARTs) family for which a full‐length complementary DNA has been identified, and RPMS1 transcript has been confirmed in many Epstein–Barr virus (EBV)‐positive malignancies. However, the effects of sequence variations of RPMS1 in hematological malignancies and their biological significance are unclear. To explore the association between RPMS1 gene variations and hematological malignancy, the RPMS1 gene of 391 EBV‐positive samples from patients with EBV‐positive leukemia, myelodysplastic syndromes and lymphoma in northern China were sequenced. On the basis of phylogenetic tree and mutation characteristics of RPMS1, all the sequences were divided into five major types: RPMS1‐A, RPMS1‐B, RPMS1‐C, RPMS1‐E, and RPMS1‐F. RPMS1‐A type, similar to the prototype B95‐8, was identified in 71.87% (281/391) of samples and was the major type in all subpopulations. The frequency of RPMS1‐F type was significantly higher in all malignant hematopathy groups than in healthy donors. The Hodgkin lymphoma group contained more RPMS1‐F than other malignant hematopathy groups, and acute myeloid leukemia contained more RPMS1‐C type than other malignant hematopathy groups. Therefore, RPMS1‐A is the main type of RPMS1 gene in northern China, and RPMS1‐F may be associated with hematologic malignancies. [ABSTRACT FROM AUTHOR]

The mechanisms connecting environmental conditions to plasticity in biological aging trajectories are fundamental to understanding individual variation in functional traits and life history. Recent findings suggest that telomere biology is especially dynamic during early life stages and has long‐term consequences for subsequent reproduction and survival. However, our current understanding is mostly derived from studies investigating ecological and anthropogenic factors separately, leaving the effects of complex environmental interactions unresolved. American alligators (Alligator mississippiensis) are long‐lived apex predators that rely on incubation temperature during a discrete period during development and endocrine cues to determine sex, making them especially vulnerable to current climatic variability and exposure to anthropogenic contaminants interfering with hormone function. Here, we combine field studies with a factorial design to understand how the developmental environment, along with intrinsic biological variation contribute to persistent telomere variation. We found that exposure to a common endocrine disrupting contaminant, DDE, affects telomere length, but that the directionality is highly dependent upon incubation temperature. Variation in hatchling growth, underlies a strong clutch effect. We also assess concentrations of a panel of glucocorticoid hormones and find that contaminant exposure elicits an increase in circulating glucocorticoids. Consistent with emerging evidence linking stress and aging trajectories, GC levels also appear to trend with shorter telomere length. Thus, we add support for a mechanistic link between contaminants and glucocorticoid signalling, which interacts with ecological aspects of the developmental environment to alter telomere dynamics. The outcomes of biological control programs can be highly variable, with natural enemies often failing to establish or spread in pest populations. This variability has posed a major obstacle in use of the bacterial parasite Pasteuria penetrans for biological control of Meloidogyne species, economically devastating plant‐parasitic nematodes for which there are limited management options. A leading hypothesis for this variability in control is that infection is successful only for specific combinations of bacterial and nematode genotypes. Under this hypothesis, failure of biological control results from the use of P. penetrans genotypes that cannot infect local Meloidogyne genotypes. We tested this hypothesis using isofemale lines of M. arenaria derived from a single field population and multiple sources of P. penetrans from the same and nearby fields. In strong support of the hypothesis, susceptibility to infection depended on the specific combination of host line and parasite source, with lines of M. arenaria varying substantially in which P. penetrans source could infect them. In light of this result, we tested whether using a diverse pool of P. penetrans could increase infection and thereby control. We found that increasing the diversity of the P. penetrans inoculum from one to eight sources more than doubled the fraction of M. arenaria individuals susceptible to infection and reduced variation in susceptibility across host lines. Together, our results highlight genotype‐by‐genotype specificity as an important cause of variation in biological control and call for the maintenance of genetic diversity in natural enemy populations. The origin of functional heterogeneity among macrophages, key innate immune system components, is still debated. While mouse strains differ in their immune responses, the range of gene expression variation among their pre‐stimulation macrophages is unknown. With a novel approach to scRNA‐seq analysis, we reveal the gene expression variation in unstimulated macrophage populations from BALB/c and C57BL/6 mice. We show that intrinsic strain‐to‐strain differences are detectable before stimulation and we place the unstimulated single cells within the gene expression landscape of stimulated macrophages. C57BL/6 mice show stronger evidence of macrophage polarization than BALB/c mice, which may contribute to their relative resistance to pathogens. Adaptation to heterogeneous sensory environments has been implicated as a key parameter in speciation. Cichlid fish are a textbook example of divergent visual adaptation, mediated by variation in the sequences and expression levels of cone opsin genes (encoding the protein component of visual pigments). In some vertebrates including fish, visual sensitivity is also tuned by the ratio of vitamin A1/A2–derived chromophores (i.e., the light‐sensitive component of the visual pigment bound to the opsin protein), where higher proportions of A2 cause a more red‐shifted wavelength absorbance. This study explores the variation in chromophore ratios across multiple cichlid populations in Lake Victoria, using as a proxy the expression of the gene Cyp27c1, which has been shown to regulate the conversion of vitamin A1 into vitamin A2 in several vertebrates. This study focuses on sympatric Pundamilia cichlids, where species with blue or red male coloration co‐occur at multiple islands but occupy different depths and consequently different visual habitats. In the red species, we found higher cyp27c1 expression in populations from turbid waters than from clear waters, but there was no such pattern in the blue species. Across populations, differences between the sympatric species in cyp27c1 expression had a consistent relationship with species differences in opsin expression patterns, but the red/blue identity reversed between clear and turbid waters. To assess the contribution of heritable vs. environmental causes of variation, we tested whether light manipulations induce a change in cyp27c1 expression in the laboratory. We found that cyp27c1 expression was not influenced by experimental light conditions, suggesting that the observed variation in the wild is due to genetic differences. Nonetheless, compared to other cichlid species, cyp27c1 is expressed at very low levels in Pundamilia, suggesting that it may not be relevant for visual adaptation in this species. Conclusively, establishing the biological importance of this variation requires testing of actual A1/A2 ratios in the eye, as well as its consequences for visual performance. [ABSTRACT FROM AUTHOR]

Coronavirus disease 2019 (COVID‐19), caused by a highly pathogenic emerging virus, is called severe acute respiratory syndrome coronavirus 2 (SARS‐CoV‐2). Knowledge regarding the pathogenesis of this virus is in infancy; however, investigation on the pathogenic mechanisms of the SARS‐CoV‐2 is underway. In COVID‐19, one of the most remarkable characteristics is the wide range of disease manifestation and severity seen across individuals of different ethnic backgrounds and geographical locations. To effectively manage COVID‐19 in the populations, beyond SARS‐CoV‐2 detection, serological response assessment, and analytic techniques, it is critical to obtain knowledge about at‐risk individuals and comprehend the identified variations in the disease's severity in general and also in the populations' levels. Several factors can contribute to variation in disease presentation, including population density, gender and age differences, and comorbid circumstances including diabetes mellitus, hypertension, and obesity. Genetic factors presumably influence SARS‐CoV‐2 infection susceptibility. Besides this, COVID‐19 has also been linked with a higher risk of mortality in men and certain ethnic groups, revealing that host genetic characteristics may affect the individual risk of death. Also, genetic variants involved in pathologic processes, including virus entrance into cells, antiviral immunity, and inflammatory response, are not entirely understood. Regarding SARS‐CoV‐2 infection characteristics, the present review suggests that various genetic polymorphisms influence virus pathogenicity and host immunity, which might have significant implications for understanding and interpreting the matter of genetics in SARS‐CoV‐2 pathogenicity and customized integrative medical care based on population investigation. From the oasis of Cholistan, true branching heterocystous cyanobacterial strains were studied for, the cell arrangement in primary branches being mono‐ or bi‐seriate; the shape of cells in main filament large and irregular; profused secondary branching emerging on one or both sides and tapering along their length. In these observed traits, two clear morphological taxa were recognized, both well‐assorted from the previously described species of the genus Westiellopsis. Both strains showed culturing responses and were studied for antibacterial, cytotoxic, and anticancer potentials. The strain derived from the site B‐10 provenance exhibited antibacterial activity against Pseudomonas (18 mm), Klebsiella (15 mm), Staphylococcus (22 mm). On the contrary, the strain of site A‐44 showed no activity against any of the above‐mentioned bacterial strains. The cytotoxicity assay for the strain of the B‐10 site showed 36% larval mortality, whereas strain A‐44 showed 24% larval mortality. Performance of the strain B‐10 in MTT assay (assessed on HCT‐116 cell lines) revealed a dose‐dependent activity: at 200, 100, 50, and 25 µg/ml; achieving a growth inhibition of 50.15%, 40.22%, 33.72%, and 10.21%, respectively; and the strain of A‐44 could only exhibit a 30.06% growth inhibition at 200 µg/ml. The 16S rRNA sequencing revealed the sequence homology with Neowestiellopsis. Based on data presented here we report two diverse taxa of true branching Nostocales from Cholistan oasis, Pakistan.
