To operationalize the extent to which firms place a strategic emphasis on human capital and human resource management (HRM), we draw on the strategy field's dominant logic theory and on the emerging literature on executive compensation for the heads of major firm functions, such as HRM. Specifically, we investigate whether the pay of human resource executives (HREs) relative to other members of top management teams (TMTs) is related to firm performance. After controlling for the endogeneity of HRE status as one of the highest paid executives on the TMT, we find in a comprehensive sample of the largest US firms that HRE relative pay is positively related to firm financial performance, specifically Tobin's Q. We also find that the relationship of HRE relative pay with Tobin's Q is stronger in relatively smaller and younger firms in our sample. Background: Previous studies have highlighted the crucial role of Wnt7B in the development of various cancers, including breast, pancreatic, and gastric cancers. However, research into the involvement of Wnt7B is often confined to specific tumor types, with a noticeable lack of comprehensive studies spanning multiple cancer forms. The potential of Wnt7B as a diagnostic or prognostic cancer biomarker has not been fully explored. Methods: In this study, we combined bioinformatics and immunohistochemistry analyses to examine the expression patterns and functions of Wnt7B in cancerous and adjacent noncancerous tissues across a range of tumors. Results: Our data indicate that Wnt7B may serve as a novel prognostic biomarker and therapeutic target in certain cancers. Conclusion: We found significant upregulation of Wnt7B expression levels in the majority of cancer cases examined. Furthermore, Wnt7B can influence cancer prognosis by modulating the tumor microenvironment, immune cell infiltration, and tumor stemness, among other factors. High‐throughput synthesis and screening of chemical libraries play pivotal roles in drug discovery. Click chemistry has emerged as a powerful strategy for constructing highly modular chemical libraries. However, the development of new click reactions and unlocking new clickable building blocks remain exceedingly challenging. Herein, we describe a double‐click strategy that enables the sequential ligations of widely available carboxylic acids and amines with fluorosulfuryl isocyanate (FSO2NCO) via a modular amidation/SuFEx (sulfur‐fluoride exchange) process. This method provides facile access to chemical libraries of N‐fluorosulfonyl amides (RCONHSO2F) and N‐acylsulfamides (RCONHSO2NR′R′′) in near‐quantitative yields under simple and practical conditions. The robustness and efficiency of this double click strategy is showcased by the facile construction of chemical libraries in 96‐well microtiter plates from a large number of carboxylic acids and amines. Preliminary biological activity screening reveals that some compounds exhibit high antimicrobial activities against Gram‐positive bacterium S. aureus and drug‐resistant MRSA (MIC up to 6.25 μg ⋅ mL−1). The pleiotropic effect of cancer‐associated fibroblasts (CAFs) on tumour progression depends on the environment. circFARP1 is critical for CAFs‐induced gemcitabine (GEM) resistance in pancreatic cancer. Its specific role and mechanism in non–small cell lung cancer (NSCLC) have not been reported yet. We prepared a cancer‐associated fibroblasts‐conditioned medium (CAF‐CM) to incubate the A549 cells. Quantitative real‐time polymerase chain reaction was used to detect RNA levels. We detected protein expression by immunohistochemistry, immunocytochemistry, western blot and immunofluorescence. We also detected the targeting impact between circFARP1, miR‐338‐3p and SRY‐box transcription factor 4 (SOX4) by using dual‐luciferase reporter and RNA pull‐down assays. We determined cell proliferation, migration and invasion capabilities through Cell Counting Kit‐8 and transwell assays. In addition, we measured tumour volume and weight in vivo by establishing a xenograft tumour model. CircFARP1 levels were remarkably high in the CAFs. The transfection experiments found that circFARP1 downregulation in CAFs caused migration, proliferation and invasion inhibition of CAFs and A549 cells, whereas inhibiting miR‐38‐3p or overexpressing SOX4 in CAFs could significantly reverse the inhibition. In vivo study in nude mice confirmed that CAFs could promote NSCLC tumour growth and knockdown of circFARP1 could inhibit tumour growth of NSCLC, whereas miR‐38‐3p downregulation or SOX4 overexpression could significantly reverse the inhibition. circFARP1 promotes NSCLC development by stimulating miR‐338‐3p/SOX4 signalling axis to regulate CAFs. Introduction: This study was designed to investigate whether renal α‐klotho levels are associated with renal pathology. This is the first report on patients with chronic kidney disease (CKD). Methods: We conducted a retrospective observational study. A total of 65 CKD patients were enrolled. Serum and renal biopsy samples were collected. Estimated glomerular filtration rate (eGFR) was examined by biochemical test. And α‐klotho expressions were assessed by RT‐PCR and immunohistochemistry. In addition, detailed microscopic findings were reviewed. Results: Renal α‐klotho levels are associated positively with eGFR, and negatively with renal pathology, including interstitial fibrosis, inflammatory cell infiltration, and tubular atrophy. The residual stress in low‐carbon high‐strength steel is not effectively relaxed during alloy carbide precipitation but is fully relaxed during manganese partitioning. The specific microscopic mechanisms responsible for this observed phenomenon have yet to be fully elucidated. This study employs first‐principles calculations in conjunction with experiments to demonstrate that alloy carbide precipitation leads to the formation of dense dislocation tangles that hinder carbon diffusion and carbide precipitation. Conversely, manganese partitioning aids carbon diffusion by altering dislocation morphology while also causing plastic deformation. The simultaneous occurrence of partitioning plasticity and precipitation plasticity during manganese partitioning results in a more significant relaxation of residual stress compared to alloy carbides precipitation: The plasticity coefficient for alloy carbide precipitation is 1.303 × 10−5, while the plasticity coefficient for manganese partition is 2.691 × 10−5. During alloy carbide precipitation, the elastic strain energy decreases to 43.48% of its initial value, whereas it can be further reduced to 25.29% after manganese partitioning. In this work, MgO‐MgAl2O4 ceramic filters were fabricated from microporous MgO powder and α‐Al2O3 micro‐powder, in which microporous MgO powder was prepared using Mg(OH)2 as initial raw material. With the α‐Al2O3 micro‐powder content increased from 0 to 15 wt%, the tight packing and reaction sintering between MgO microparticles and Al2O3 microparticles promoted the formation and growth of magnesium aluminate spinel necks, reduced the apparent porosity of ceramic filter skeletons, decreased the pore size, and improved the strength of the filters. When the α‐Al2O3 micro‐powder content further raised to 20 wt%, the thermal mismatch between MgO microparticles and spinel microparticles caused a small amount of crack generation, which lowered the strength of the filter. Besides, after introducing the α‐Al2O3 micro‐powder, more spinel necks were produced due to a small amount of MgO solid dissolved in the spinel lattice during the thermal shock test, which enhanced the thermal shock resistance of the filters. When the α‐Al2O3 micro‐powder content was 15 wt%, the filter had excellent comprehensive performance, the bulk density and apparent porosity of the filter skeleton were 3.09 g/cm3 and 10.7%, and the compressive strength before and after thermal shock test was 3.48 and 3.70 MPa, respectively. Nasopharyngeal carcinoma (NPC), a squamous cell carcinoma originating in the nasopharynx, is a leading malignancy in south China and other south and east Asia areas. It is frequently associated with Epstein‐Barr virus (EBV) infection, while there are also some NPC patients without EBV infection. Here, it is shown that the EBV+ (EBV positive) and EBV‐ (EBV negative) NPCs contain both shared and distinct genetic abnormalities, among the latter are increased mutations in TP53. To investigate the functional roles of NPC‐associated genetic alterations, primary, orthotopic, and genetically defined NPC models were developed in mice, a key tool missed in the field. These models, initiated with gene‐edited organoids of normal nasopharyngeal epithelium, faithfully recapitulated the pathological features of human disease. With these models, it is found that Trp53 and Cdkn2a deficiency are crucial for NPC initiation and progression. And latent membrane protein1 (LMP1), an EBV‐coding oncoprotein, significantly promoted the distal metastasis. Further, loss of TGFBR2, which is frequently disrupted both in EBV‐ and EBV+ NPCs, dramatically accelerated the progression and lung metastasis of NPC probably by altering tumor microenvironment. Despite docetaxel combined with cisplatin and 5‐fluorouracil (TPF) being the established treatment for advanced nasopharyngeal carcinoma (NPC), there are patients who do not respond positively to this form of therapy. However, the mechanisms underlying this lack of benefit remain unclear. DCAF7 is identified as a chemoresistance gene attenuating the response to TPF therapy in NPC patients. DCAF7 promotes the cisplatin resistance and metastasis of NPC cells in vitro and in vivo. Mechanistically, DCAF7 serves as a scaffold protein that facilitates the interaction between USP10 and G3BP1, leading to the elimination of K48‐linked ubiquitin moieties from Lys76 of G3BP1. This process helps prevent the degradation of G3BP1 via the ubiquitin‒proteasome pathway and promotes the formation of stress granule (SG)‐like structures. Moreover, knockdown of G3BP1 successfully reversed the formation of SG‐like structures and the oncogenic effects of DCAF7. Significantly, NPC patients with increased levels of DCAF7 showed a high risk of metastasis, and elevated DCAF7 levels are linked to an unfavorable prognosis. The study reveals DCAF7 as a crucial gene for cisplatin resistance and offers further understanding of how chemoresistance develops in NPC. Excitation of photoactive electron donor−acceptor (EDA) complexes to generate radical is a promising approach in radical chemistry. In this study, we introduce a new model of H‐bonding EDA complexes for the selective hydrothiolation and hydroxysulfenylation of carbonyl‐activated alkenes with diverse thiols under visible light conditions. The reliability of this H‐bonding EDA complex model has been confirmed by meticulous experimental and theoretical calculations. Mechanistic investigations have revealed the significant influence of the solvent in determining whether the excitation of photoactive H‐bonding EDA complex leads to charge transfer (CT) or energy‐charge transfer (En‐CT), thereby controlling Markovnikov and anti‐Markovnikov selectivity. Notably, the Quantum Theory of Atoms in Molecules (QTAIM) analysis clearly shows that the excited state of the C=O−H−S EDA complex involves closed‐shell partially covalent interactions. Summary: Protein posttranslational modifications play crucial roles in plant immunity through modulating a complicated signaling network mediated by different hormones. We previously demonstrated that OsATL32, an ATL‐type E3 ligase, negatively contributes to rice immunity against Magnaporthe oryzae.Here, we show that OsATL32 forms a loop with OsPPKL2 and OsGSK2 through distinct protein posttranslational modifications to modulate rice immunity.OsATL32 ubiquitinates OsPPKL2, a protein phosphatase with Kelch‐like repeat domains that exerts positive roles in regulating rice immunity against M. oryzae and chitin‐triggered immune responses, for degradation. The glycogen synthase kinase 2 (OsGSK2), which acts as a negative regulator of rice immunity against M. oryzae and chitin‐triggered immune responses, phosphorylates OsATL32 to elevate its protein stability and E3 ligase activity on OsPPKL2. Moreover, OsPPKL2 directly dephosphorylates OsGSK2, affecting its kinase activity on substrates including OsATL32 for phosphorylation. Like OsGSK2 as a BR signaling repressor, OsATL32 negatively regulates BR signaling; conversely, OsPPKL2 plays a positive role in BR signaling.These findings provide a molecular mechanism in which OsATL32 serves as a node connecting BR signaling and immunity by associating with OsPPKL2 and OsGSK2, assembling into a distinct protein posttranslational modifications‐linked loop that functions in rice BR signaling and immunity. Polymer dielectrics have gained significant usage in electrical engineering due to their unique merits, such as good flexibility, easy processing, and low cost. However, it is a huge challenge that polymer dielectrics maintain high breakdown strength meanwhile high permittivity for enhanced energy density. Here, sandwiched polymer films are constructed for enhanced energy density. Increased permittivity has been achieved via blending BaTiO3@polyfluorosiloxane with a polymer as the top and bottom layer. Furthermore, the neat polymer as the middle layer is without critically sacrificing breakdown strength. The energy storage density increases from 7.8 J/cm3 of the neat polymer to 10.41 J/cm3 of the sandwich structure at 460 MV/m. Precipitation δ18O has offered valuable insights into the evolution of the Asian monsoon. Recent researches focusing on precipitation Δ′17O has enhanced our understanding by offering new perspectives beyond those of δ18O, revealing insights into vapor sources and continental recycling. Nevertheless, there remains a lack of interannual triple oxygen isotope data, particularly in the Asian monsoon region. In this study, we analyzed the triple oxygen isotopes and hydrogen isotopes in monthly precipitation samples collected from Chongqing in Southwest China between 2019 and 2022 A.D. Seasonal variations in δD, δ18O, δ17O, and d‐excess values were observed, with lower values during the rainy season and higher values during the dry season, highlighting the impact of changes in moisture sources and local meteorological conditions on seasonal shifts in δD, δ18O, and δ17O. While, mean Δ′17O values were higher in rainy season and lower in dry season. Notably, during rainy season, there is a negative correlation between monthly Δ′17O values and the RH of the vapor source area, as well as a positive correlation with d‐excess. Recalculated Δ′17O values based on RH of oceanic moisture source, are higher than the measured values for this period, indicating the contribution of terrigenous moisture to precipitation in SW China. Precipitation Δ′17O values provide a more precise reflection of changes in moisture source, continental recycling, and evapotranspiration processes that drive water cycling compared Integrating modeling works in future will facilitate the use of precipitation Δ′17O values to quantify the impact of different moisture source on precipitation. Seasonal variations in δD, δ18O, δ17O, and d‐excess revealed low values during the rainy season (May–October) and significantly higher values during the dry season (November–April). These variations are primarily attributed to different water vapor sources in distinct seasons. Moreover, we observed a negative correlation between precipitation Δ′17O and relative humidity in the source area of the water vapor during the rainy season, suggesting that Δ′17O contains valuable information about the water vapor's source area. Chiral carbon dots (CDs) with circularly polarized luminescence (CPL) are one of the most dynamic areas of modern science. However, the design, preparation, and ambiguity mechanism of solid‐state CPL‐active CDs remains a formidable challenge. Herein, for the first time, CDs with customized chiroptical activities in the solid state, especially CPL, are transcribed from chiral metal–organic framework (CMOFs) via a bottle‐around‐ship strategy. Within these CMOFs⊃CDs assemblies, CDs inherited the chirality of the host CMOFs through host–guest interactions, which is revealed by density functional theory (DFT) simulations and experimental results, and amplified the luminescence dissymmetry factor (glum) by effective artificial chiral light‐harvesting systems. Impressively, CMOFs⊃CDs in pairs generated color‐tunable CPL and white CPL with chromaticity coordinates of (0.32, 0.32). Furthermore, benefiting from excellent processability, as luminescent coatings and 3D printing inks, a white circularly polarized light‐emitting diode, and an extended 3D model “light bulb” featuring white CPL are successfully fabricated, respectively. Using nuclear power to replace electricity generated from fossil fuels is an effective strategy to reduce global carbon dioxide emissions and also spurs the search for new sources of nuclear fuel. Extracting uranium from seawater has a significant reserve advantage, although its ultralow concentration presents substantial challenges. Here, inspired by the fractal structure of cardiac trabeculae on the inner surface of the heart, a uranium enrichment electrode with a superhydrophilic and uranium‐affinitive fractal surface is developed. This innovative design enhances rapid charge/ion transfer, ensures complete surface wetting, and provides numerous adsorption sites. By synergistically integrating the advantages of electric‐assisted processes and bioinspired microstructures predicated on chemical coordination principles, the electrode demonstrates a uranium adsorption capacity of 13.2 mg g−1 following a 7‐d exposure to natural seawater. This research not only demonstrates an effective strategy for the development of advanced uranium enrichment electrodes but also provides more possibilities for innovative approaches in sustainable energy technology. As a modality of optical imaging, fluorescence imaging of biochemical analytes holds significant potential in the field of disease diagnosis and therapy. However, the non‐responsive fluorophores are not able to selectively and sensitively detect lesions, resulting in a poor signal‐to‐background ratio and compromised accuracy. The utilization of activatable sensors, in contrast, is capable of effectively overcoming these limitations by displaying variations in fluorescence intensity and spectral wavelength in response to physiological parameters. Aggregation‐induced emission luminogens (AIEgens), particularly those emitting fluorescence in the near‐infrared (NIR, 700–1700 nm) window, have practical application prospects in disease theranostics due to their excellent photostability, deeper penetration depth, and enhanced fluorescence intensity in physiological environments. In this review, an overview of the advancements in research on NIR AIEgens for internal stimuli‐activated diagnosis and treatment of diseases is provided. The design principles of responsive NIR AIEgens and their biomedical applications are exemplified with representative examples, categorized based on the type of stimuli. Finally, the review concludes with a brief discussion of the challenges, opportunities, and future prospects of activatable NIR AIEgens in the field of disease photo theranostics. Statins, the first‐line medication for dyslipidemia, are linked to an increased risk of type 2 diabetes. But exactly how statins cause diabetes is yet unknown. In this study, a developed short‐term statin therapy on hyperlipidemia mice show that hepatic insulin resistance is a cause of statin‐induced diabetes. Statin medication raises the expression of progesterone and adiponectin receptor 9 (PAQR9) in liver, which inhibits insulin signaling through degradation of protein phosphatase, Mg2+/Mn2+ dependent 1 (PPM1α) to activate ERK pathway. STIP1 homology and U‐box containing protein 1 (STUB1) is found to mediate ubiquitination of PPM1α promoted by PAQR9. On the other hand, decreased activity of hepatocyte nuclear factor 4 alpha (HNF4α) seems to be the cause of PAQR9 expression under statin therapy. The interventions on PAQR9, including deletion of PAQR9, caloric restriction and HNF4α activation, are all effective treatments for statin‐induced diabetes, while liver specific over‐expression of PPM1α is another possible tactic. The results reveal the importance of HNF4α‐PAQR9‐STUB1‐PPM1α axis in controlling the statin‐induced hepatic insulin resistance, offering a fresh insight into the molecular mechanisms underlying statin therapy. Osteoarthritis is a highly prevalent progressive joint disease that still requires an optimal therapeutic approach. Intermittent fasting is an attractive dieting strategy for improving health. Here this study shows that intermittent fasting potently relieves medial meniscus (DMM)‐ or natural aging‐induced osteoarthritic phenotypes. Osteocytes, the most abundant bone cells, secrete excess neuropeptide Y (NPY) during osteoarthritis, and this alteration can be altered by intermittent fasting. Both NPY and the NPY‐abundant culture medium of osteocytes (OCY‐CM) from osteoarthritic mice possess pro‐inflammatory, pro‐osteoclastic, and pro‐neurite outgrowth effects, while OCY‐CM from the intermittent fasting‐treated osteoarthritic mice fails to induce significant stimulatory effects on inflammation, osteoclast formation, and neurite outgrowth. Depletion of osteocyte NPY significantly attenuates DMM‐induced osteoarthritis and abolishes the benefits of intermittent fasting on osteoarthritis. This study suggests that osteocyte NPY is a key contributing factor in the pathogenesis of osteoarthritis and intermittent fasting represents a promising nonpharmacological antiosteoarthritis method by targeting osteocyte NPY. Motor learning (ML), which plays a fundamental role in growth and physical rehabilitation, involves different stages of learning and memory processes through different brain regions. However, the neural mechanisms that underlie ML are not sufficiently understood. Here, a previously unreported neuronal projection from the dorsal hippocampus (dHPC) to the zona incerta (ZI) involved in the regulation of ML behaviors is identified. Using recombinant adeno‐associated virus, the projections to the ZI are surprisingly identified as originating from the dorsal dentate gyrus (DG) and CA1 subregions of the dHPC. Furthermore, projection‐specific chemogenetic and optogenetic manipulation reveals that the projections from the dorsal CA1 to the ZI play key roles in the acquisition and consolidation of ML behaviors, whereas the projections from the dorsal DG to the ZI mediate the retrieval/retention of ML behaviors. The results reveal new projections from the dorsal DG and dorsal CA1 to the ZI involved in the regulation of ML and provide insight into the stages over which this regulation occurs. Synthetic Aperture Radar (SAR) has emerged as a pivotal technology in geosciences, offering unparalleled insights into Earth's surface. Indeed, its ability to provide high‐resolution, all‐weather, and day‐night imaging has revolutionized our understanding of various geophysical processes. Recent advancements in SAR technology, that is, developing new satellite missions, enhancing signal processing techniques, and integrating machine learning algorithms, have significantly broadened the scope and depth of geosciences. Therefore, it is essential to summarize SAR's comprehensive applications for geosciences, especially emphasizing recent advancements in SAR technologies and applications. Moreover, current SAR‐related review papers have primarily focused on SAR technology or SAR imaging and data processing techniques. Hence, a review that integrates SAR technology with geophysical features is needed to highlight the significance of SAR in addressing challenges in geosciences, as well as to explore SAR's potential in solving complex geoscience problems. Spurred by these requirements, this review comprehensively and in‐depth reviews SAR applications for geosciences, broadly including various aspects in air‐sea dynamics, oceanography, geography, disaster and hazard monitoring, climate change, and geosciences data fusion. For each applied field, the scientific advancements produced because of SAR are demonstrated by combining the SAR techniques with characteristics of geophysical phenomena and processes. Further outlooks are also explored, such as integrating SAR data with other geophysical data and conducting interdisciplinary research to offer comprehensive insights into geosciences. With the support of deep learning, this synergy will enhance the capability to model, simulate, and forecast geophysical phenomena with greater accuracy and reliability. SAR has been providing high‐resolution images for many decades and they have been applied to many fields in geosciences. This work comprehensively overviews the application of SAR in geosciences, including oceanography, geography, geodesy, climatology, seismology, meteorology, and environmental science. Moreover, this review paper highlights the significance of SAR in various aspects of geosciences, summarizes recent advancements in SAR technology, and demonstrates unique insights and important contributions of SAR in understanding and solving geophysical questions. Future directions and outlooks include integrating SAR with other geophysical data and interdisciplinary applications for complex questions. This review serves as an up‐to‐date guide to the cutting‐edge uses of SAR technology in comprehensive geophysical studies. It is aimed at researchers and practitioners in geosciences, as well as policymakers and stakeholders interested in leveraging SAR for geosciences. Epoxy resins, as important thermosetting polymers, exhibit excellent adhesion to various substrates. In view of this, reticulate coating of triglycidyl isocyanate with triethylenetetramine was introduced onto the surface of poly(styrene‐divinylbenzene) utilizing amine curing reaction to obtain poly(styrene‐divinylbenzene)@triglycidyl isocyanate‐triethylenetetramine composite microspheres. The amino groups and epoxy groups of triglycidyl isocyanate‐triethylenetetramine endowed poly(styrene‐divinylbenzene) with good reactivity, which could be quaternized under mild conditions to obtain an anion exchange chromatographic stationary phase. The quaternized poly(styrene‐divinylbenzene)@triglycidyl isocyanate‐triethylenetetramine was characterized by scanning electron microscope, Fourier‐transform infrared spectroscopy, N2 adsorption‐desorption experiment, et al. The chromatographic performance of the customized column was evaluated by separating seven conventional anions, organic weak acids, and carbohydrates. Poly(styrene‐divinylbenzene)@triglycidyl isocyanate‐triethylenetetramine possesses the uniform size of poly(styrene‐divinylbenzene) microspheres and good reactivity of triglycidyl isocyanate‐triethylenetetramine, which offers a flexible strategy for the preparation of anion exchange stationary phase. The column exhibits excellent chemical and mechanical stability and chromatographic performance. Finally, the column was successfully applied for the determination of nitrite in pickles. 4,8‐Dihydroxy‐1‐tetrahydronaphthone (4,8‐DHT), an allelochemical, was isolated from the Carya cathayensis epicarp and was discovered to have phytotoxicity in laboratory tests. In this study, field experiments were designed to assess the phytotoxicity of 4,8‐DHT on weeds in Cerasus yedoensis and Lagerstroemia indica forestlands after pre‐ and post‐emergence application, in addition to physiological effect on Lindernia procumbens were also measured. The phytotoxic results of 4,8‐DHT demonstrated that the total inhibition rate of weeds was 81.3% and 68.2% in C. yedoensis forestland and L. indica forestland after pre‐emergence applied for 24 days in 2021, and enhanced by 9.9% and 27.8% compared with post‐emergence treatment. Especially, in C. yedoensis forestland, a high concentration of 4,8‐DHT sprayed at the pre‐emergence stage after 8 days had a significant inhibition rate on all weeds except Cyperus rotundus, and was still above 80.0% after 24 days of exposure. Comprehensive control efficacy of 4,8‐DHT indicates that pre‐emergence application is more favorable for weeds control. Physiological analysis showed that 4,8‐DHT stimulated the contents of thiobarbituric acid reactive substances (TBARS), hydrogen peroxide (H2O2), and proline and declined in the activities of antioxidant enzymes, meanwhile, accompanied by a decrease in the root vitality and chlorophyll content in L. procumbens. Type IV collagen is an integral component of basement membranes. Mutations in COL4A1, one of the key genes encoding Type IV collagen, can result in a variety of diseases. It is clear that a significant proportion of mutations that affect splicing can cause disease directly or contribute to the susceptibility or severity of disease. Here, we analyzed exonic mutations and intronic mutations described in the COL4A1 gene using bioinformatics programs and identified candidate mutations that may alter the normal splicing pattern through a minigene system. We identified seven variants that induce splicing alterations by disrupting normal splice sites, creating new ones, or altering splice regulatory elements. These mutations are predicted to impact protein function. Our results help in the correct molecular characterization of variants in COL4A1 and may help develop more personalized treatment options. Summary: Paocai, as one of the most widely consumed traditional fermented foods in China, is dominated by lactic acid bacteria (LAB), which exert a significant influence on the dynamics of metabolites, microecological changes, and sensory quality. In this study, five different paocai groups were designed for fermentation using Lactiplantibacillus plantarum HYY‐DB9 (LP), Levilactobacillus brevis GIM 1.773 (LB), a mixture of two LABs (LP + LB), pasteurisation (PST), and natural fermentation (NF). Nitrite content, pH, titratable acidity, viable cell counts (LAB and yeasts), metabolites (organic acids and volatile substances), and sensory evaluation were determined for each group. The results demonstrated that the mixed fermentation with two LABs (LP + LB) significantly reduced nitrite content. Organic acids produced by LAB played a crucial role in the degradation process, particularly lactic acid which exhibited the most pronounced effect on nitrite degradation. Furthermore, artificial inoculation of LAB fermentation resulted in improved taste and flavour of paocai along with increased production of alcohols, esters, and acids compared to traditional natural fermentation. These findings have important implications for enhancing paocai quality control and managing nitrite content while also providing an excellent fermentation agent for fermented paocai. BACKGROUND: Whey protein isolate (WPI) generally represents poor functional properties such as thermal stability, emulsifying activity and antioxidant activity near its isoelectric point or high temperatures, which limit its application in the food industry. The preparation of WPI–polysaccharide covalent conjugates based on Maillard reaction is a promising method to improve the physical and chemical stability and functional properties of WPI. In this research, WPI–inulin conjugates were prepared through wet heating method and ultrasound method and their structural and functional properties were examined. RESULTS: In conjugates, the free amino acid content was reduced, the high molecular bands were emerged at sodium dodecyl sulfate‐polyacrylamide gel electrophoresis (SDS‐PAGE), new C–N bonds were formed in Fourier‐transform infrared (FTIR) spectroscopy, and fluorescence intensity was reduced compared with WPI. Furthermore, the result of circular dichroism (CD) spectroscopy also showed that the secondary structure of conjugates was changed. Conjugates with ultrasound treatment had better structural properties compared with those prepared by wet heating treatment. The functional properties such as thermal stability, emulsifying activity index (EAI), emulsion stability (ES) and antioxidant activity of conjugates with wet heating treatment were significantly improved compared with WPI. The EAI and ES of conjugates with ultrasound treatment were the highest, but the thermal stability and antioxidant activity were only close to that of the conjugates with wet heating treatment for 2 h. CONCLUSION: This study revealed that WPI–inulin conjugates prepared with ultrasound or wet heating method not only changed the structural characteristics of WPI but also could promote its functional properties including thermal stability, EAI, ES and antioxidant activity. © 2024 Society of Chemical Industry. The accuracy of fault interpretation is generally influenced by the quality of seismic images. Because of the blurring effect of the migration process, faults with small throws may not be clearly imaged in seismic images, which will impose limitations on the fault detection. To address this issue, we propose a deep learning‐based method to enhance faults in poststack seismic images. We generate abundant training samples by convolving the three‐dimensional point‐spread functions with the noisy reflectivity models. The corresponding labels are synthesized using the one‐dimensional seismic wavelet convolution method, simulating conditions with perfect illumination. To train the network for optimal performance, we investigate the impact of different loss functions. Ultimately, we employ a mixed loss function combining structural similarity index measure and gradient difference loss, since the gradient difference loss focuses more on geological edge information, and the structural similarity index measure possesses excellent image perceptual capability and optimization property. Results from one synthetic seismic image and three real seismic data demonstrate that our proposed method can effectively restore the sharpness of fault surfaces, particularly for faults with small displacements. Compared to the structural smoothing method, the network we trained achieves optimal fault enhancement. Furthermore, coherence‐based fault images indicate that seismic images enhanced using our method can improve the accuracy of fault interpretation and yield more continuous fault maps. The fir‐tree tenon/mortise structure in areo‐engine suffers from the combined high and low cycle fatigue (CCF) loads during service. The structural integrity of the tenon/mortise structure is significantly affected by the local strain distribution, while the strain value under the CCF loads is difficult to acquire. In this study, a simulated specimen of tenon/mortise structure is designed, and the CCF test is carried out using a divided‐path loading fixture to avoid interference between the high cycle fatigue (HCF) loads and the low cycle fatigue (LCF) loads. The high speed digital image correlation (DIC) method is adopted to acquire the real‐time strain distribution during CCF test. According to the strain results, the critical position of the mortise and its strain variation are determined. The high cycle strain of the mortise is proportional to the vibration amplitude of the tenon. The experimental results can provide basis for strength and life assessment. Background and Objectives: This study explored the impact of nitrogen (N) application on yield and breadmaking as well as arabinoxylan (AX) content of wheat varieties Zhongmai 578 and Jimai 22 across varied environments, aiming to optimize N levels for improved outcomes. Findings: Significant N effects were observed on yield, flour properties, and breadmaking quality traits. Both varieties displayed high yield potential and stability, with Zhongmai 578 excelling in thousand kernel weight and Jimai 22 in spike number. Optimal N rates varied by location, with some achieving peak yields at reduced N levels. Processing traits like dough properties and bread‐making quality were influenced by E × N interactions. Arabinoxylans profiles was not affected by different N input. Conclusions: Customizing N applications to specific environment is critical for maximizing wheat yield potential and quality. Environmental considerations are integral for achieving desired processing traits and yield, highlighting the need for tailored N strategies in wheat cultivation. Background and Aims: For patients with high‐risk non‐ST elevation myocardial infarction (NSTEMI), current guidelines recommend an early invasive strategy within 24 h. New‐onset acute ischemic stroke (NAIS) is a rare but fatal complication of percutaneous coronary intervention (PCI). However, the effect of the timing of PCI and the risk of NAIS in NSTEMI is poorly defined. Methods: Patients with NSTEMI who underwent PCI were queried from the National Inpatient Sample Database (2016–2019) and stratified into three groups: early (<24 h), medium (24–72 h), and late (>72 h) PCI. Multivariate logistic regression models were used to determine the association between timing of PCI and NAIS. Results: Among 633,115 weighted hospitalizations, patients in the late PCI group had a higher incidence of NAIS (1.3%) than those in the early (0.67%) and medium (0.71%) PCI groups. Patients undergoing late PCI were older, more likely to be female, and had a greater incidence of comorbidities (e.g., diabetes mellitus, chronic pulmonary and renal illness, and atrial fibrillation) than those undergoing early or medium PCI. After adjustment, only late PCI was significantly associated with a 54% increased NAIS risk (adjusted odds ratio: 1.54 [95% confidence interval: 1.29–1.84]). Additionally, there was heterogeneity in the magnitude of risk by age and sex. Younger people (<65 years) (p for interaction <0.001) and men (interaction‐value p = 0.040) were more likely to encounter NAIS. Background: Atopic dermatitis (AD) is a prevalent chronic inflammatory and highly pruritic skin condition characterized by the infiltration of immune cells, notably eosinophils and mast cells. Mast cells (MCs) critically participate in the complex pathogenesis of AD through multiple pathways and have recently garnered growing attention in research. Despite the abundance of related studies published over the years, a comprehensive bibliometric analysis on this topic remains lacking. Objective: Our objective was to perform an up‐to‐date bibliometric analysis of the literature focusing on the relationship between MCs and AD. This analysis would provide valuable insights through a thorough bibliometric review, enabling a clearer understanding of the current research landscape, pinpointing key studies, and detecting emerging trends within this field. Methods: We searched the Web of Science Core Collection (WoSCC) database on 15 July 2024. The data retrieval strategy was structured as follows: #1: TS = ("mast cells") OR TS = ("mast cell") OR TS = ("mastocyte"); #2: TS = ("atopic dermatitis") OR TS = ("atopic eczema") Final data: (#1 AND #2). A total of 2272 items published between 2001 and 2024 were included. Several scientometric visualization tools, including VOSviewer, R‐bibliometrix, CiteSpace and an online analytical platform, were utilized to conduct text mining and to visualize the bibliometric data, facilitating a comprehensive analysis of research trends and patterns. Results: Out of the initial 2272 articles retrieved, 2168 were selected for analysis after applying inclusion and exclusion criteria based on publication type. The findings indicate a steady and substantial exponential growth in the annual number of publications focused on the relationship between over the years. The South Korea (547/2168), USA (465/2168) and Japan (436/2168) were the major contributors within this field, collectively constituting more than half of the total publications. To clarify the underlying mechanisms and role of MCs in the pathogenesis of AD and to make MCs prime targets for therapeutic intervention have garnered the most attention in this field. According to references analysis, the research emphasis has shifted to developing MC‐related therapeutics and intervention and regulating the immune system of AD patients through modulating the activity of various immune cells. On the basis of keywords analysis, we outlined the following research frontiers and hotpots in the future: the role of oxidative stress in the pathogenesis; imbalance in the different types of T helper (Th) cells during immune response; skin barrier and barrier dysfunction; improving quality of life; sensory neurons; biological agents and small‐molecule drugs. Furthermore, IL‐13, IL‐4, NFKB1, BCGF‐1 and CD4 ranked as the top five genes that have received the most investigative attention in the intersection of MCs and AD. Conclusion: In a word, this analysis would greatly benefit from a thorough bibliometric review to gain a deeper understanding of the current research landscape, identify pivotal studies and pinpoint emerging trends in the field of MCs and AD. Meanwhile, our findings offered researchers a holistic perspective of ongoing developments, serving as a valuable resource for guiding future research and informing decision‐making for both researchers and policymakers in this area. Membrane vesicles (MVs) are produced by species across all domains of life and have diverse physiological functions as well as promising applications. While the mechanisms for vesiculation in Gram‐negative bacteria are well‐established, the genetic determinants and regulatory factors responsible for MV biogenesis in Gram‐positive bacteria remain largely unknown. Here, we demonstrate that a Q225P substitution in the alternative sigma factor B (SigB) triggers MV production in Staphylococcus aureus strain Newman by hindering the specific binding of SigB to the asp23 promoter, thereby repressing expression of alkaline shock protein 23 (Asp23). Isogenic deletion of asp23 also promotes MV formation in Newman, confirming the critical roles played by sigB and asp23 in modulating S. aureus vesiculation. While bacterial growth and cytoplasmic membrane fluidity are not impaired, mutation of asp23 weakens the cell wall and enhances autolysis, consistent with decreased expression of alpha‐type psm and lrgAB that modulate murein hydrolase activity. TEM and proteomic analysis show that Newman and asp23 deletion mutant generate MVs with nearly identical morphology and composition, but virulence‐associated factors are significantly enriched in MVs from the asp23 mutant. Mantle cell lymphoma (MCL) is a rare and aggressive form of non‐Hodgkin lymphoma. Challenges in its treatment include relapse, drug resistance, and a short survival period. The Hedgehog/GLI1 (Hh/GLI1) and Wnt/β‐catenin pathways are crucial in cancer cell proliferation, survival, and drug resistance, making them significant targets for anticancer research. This study aimed to assess the effectiveness of combining inhibitors for both pathways against MCL and investigate the underlying molecular mechanisms. The co‐expression of key proteins from the Hh/GLI1 and Wnt/β‐catenin pathways was observed in MCL. Targeting the Hh/GLI1 pathway with the GLI1 inhibitor GANT61 and the Wnt/β‐catenin pathway with the CBP/β‐catenin transcription inhibitor ICG‐001, dual‐target therapy was demonstrated to synergistically suppressed the activity of MCL cells. This approach promoted MCL cell apoptosis, induced G0/G1 phase blockade, decreased the percentage of S‐phase cells, and enhanced the sensitivity of MCL cells to the drugs adriamycin and ibrutinib. Both GANT61 and ICG‐001 downregulated GLI1 and β‐catenin while upregulating GSK‐3β expression. The interaction between Hh/GLI1 and Wnt/β‐catenin pathways was mediated by GANT61‐dependent Hh/GLI1 inhibition. Moreover, GLI1 knockdown combined with ICG‐001 synergistically induced apoptosis and increased drug sensitivity of MCL cells to doxorubicin and ibrutinib. GANT61 attenuated the overexpression of β‐catenin and decreased the inhibition of GSK‐3β in MCL cells. Overall, the combined targeting of both the Hh/GLI1 and Wnt/β‐catenin pathways was more effective in suppressing proliferation, inducing G0/G1 cycle retardation, promoting apoptosis, and increasing drug sensitivity of MCL cells than mono treatments. To unveil the charge storage mechanisms and interface properties of electrode materials is very challenging for Na‐ion storage. In this work, we report that the novel layered perovskite Bi2TiO4F2@reduced graphene oxides (BTOF@rGO) serves as a promising anode for Na‐ion storage in an ether‐based electrolyte, which exhibits much better electrochemical performance than in an ester‐based electrolyte. Interestingly, BTOF@rGO possesses a prominent specific capacity of 458.3–102 mAh g−1/0.02–1 A g−1 and a high initial coulombic efficiency (ICE) of 70.3%. Cross‐sectional morphology and depth profile surface chemistry indicate not only a denser reactive interfacial layer but also a superior solid electrolyte interface film containing a higher proportion of inorganic components, which accelerates Na+ migration and is an essential factor for the improvement of ICE and other electrochemical properties. Electrochemical tests and ex situ measurements demonstrate the triple hybridization Na‐ion storage mechanism of conversion, alloying, and intercalation for BTOF@rGO in the ether‐based electrolyte. Furthermore, the Na‐ion batteries assembled with the BTOF@rGO anode and the commercial Na3V2(PO4)2F3@C cathode exhibit remarkable energy densities and power densities. Background: The therapeutic advantage of thoracic radiotherapy (tRT) as an adjunct to first‐line immunotherapy and chemotherapy in patients with extensive‐stage small cell lung cancer (ES‐SCLC) remains unclear. We sought to elucidate this in a retrospective cohort study comparing the effectiveness and safety of tRT in combination with first‐line immunotherapy and chemotherapy. Methods: Our retrospective study included patients with ES‐SCLC, treated at the West China Hospital between January 2019 and December 2022. They received first‐line immunotherapy and chemotherapy and were categorized into two cohorts based on the administration of tRT. The primary outcomes were overall survival (OS) and progression‐free survival (PFS). Cox regression analysis was utilized to identify potential independent predictors of prognosis and to compare the treatment outcomes across various patient subgroups. Treatment‐related toxicities across both cohorts were compared using the Chi‐squared test. Results: A total of 99patients were eligible for the study, out of which 55 received tRT. The medianduration of follow‐up was 39 months. Remarkably, patients who received tRTdemonstrated superior OS and PFS in comparison to those who did not (P < 0.05). Subgroup analysis further confirmed these findings. Multivariate analysisidentified treatment group and liver metastasis as independent prognosticfactors (P < 0.05). The incidence of grade 3‐4 adverse events showed nostatistically significant difference between the two cohorts. Conclusions: Thus, weconfirmed that the addition of tRT to the conventional regimen of first‐linechemotherapy and immunotherapy yields better survival outcomes without asignificant increase in toxicity. Aims/Introduction: To evaluate the differences in cardiac autonomic function, cardiac structure and diastolic function between individuals with diabetic foot (DF) and those with diabetes but without DF. Materials and Methods: A total of 413 individuals with DF and 437 without DF who underwent a 24‐h electrocardiogram Holter and a Doppler echocardiogram were included. The heart rate variability parameters to evaluate cardiac autonomic function, and the indices for the assessment of cardiac structure and left ventricular (LV) diastolic function, including left atrium, LV posterior wall thickness, interventricular septum and E/e′ ratio, were measured or calculated. Propensity score matching was used for the sensitivity analysis to minimize potential imbalance. Results: In both the crude and propensity score matching analyses, significant differences were observed in heart rate variability between individuals with and without DF, as evidenced by lower standard deviation of the normal sinus interval, lower low‐frequency power/high‐frequency power ratio, lower standard deviation of the 5‐min average RR intervals, lower low‐frequency power, lower percentage of normal adjacent RR interval difference >50 ms, lower root mean square of successive RR interval differences and lower high‐frequency power (all P < 0.05). In multivariate analysis, DF showed an independent negative correlation with the aforementioned indices of heart rate variability (all P < 0.05). Individuals with DF showed higher left atrium, LV posterior wall thickness, interventricular septum and a higher E/e′ ratio than those without DF in the crude analysis (all P < 0.05), whereas these indices were no longer associated with DF in the multivariate analysis and the propensity score matching analyses. Conclusions: Cardiac autonomic modulation was more severely impaired in individuals with DF than in their counterparts without DF. Aim: To determine the association of the presence of diabetes and, among persons with diabetes, the age at type 2 diabetes mellitus (T2DM) onset, BMI and the interactive effect with the subsequent thyroid cancer risk. Materials and Methods: We conducted a population register‐based longitudinal cohort study in Shanghai, including 428 568 persons with new‐onset T2DM matched with the general population. The risk of thyroid cancer among subgroups was calculated based on standardized incidence ratio (SIR), hazard ratio (HR) and Cox proportional hazards models. Results: In total, 1142 thyroid cancer cases were identified during 8 years of follow‐up, with an incidence rate of 59.01/100 000 person‐years and a higher risk (SIR = 1.21) compared with the general population. The earlier age at T2DM onset and higher BMI were associated with an increasing risk of thyroid cancer independently (onset age <50, SIR: 1.46; BMI ≥30.0 kg/m2, SIR: 1.93), with the highest risk in patients with both BMI ≥30.0 kg/m2 and onset age <50 years (SIR = 3.91, HR = 3.04). Among patients with T2DM onset age <60 years, SIR increased with higher BMI, while there were no trends when onset age ≥60 years. Among patients with BMI ≥25.0 kg/m2, SIR increased with an earlier onset age, whereas no trends were shown in the BMI <24.9 kg/m2 groups. Obese (BMI ≥30.0 kg/m2) patients had a significantly higher HR of thyroid cancer only when T2DM onset age <60 years. Conclusions: Both earlier age of T2DM onset (<50 years) and higher BMI (≥30 kg/m2) contributed to the higher risk of thyroid cancer. Summary: Cone enlargement is a crucial process for seed production and reproduction in gymnosperms. Most of our knowledge of cone development is derived from observing anatomical structure during gametophyte development. Therefore, the exact molecular mechanism underlying cone enlargement after fertilization is poorly understood. Here, we demonstrate that sucrose promotes cone enlargement in Torreya grandis, a gymnosperm species with relatively low rates of cone enlargement, via the TgNGA1‐TgWRKY47‐TgEXPA2 pathway.Cell expansion plays a significant role in cone enlargement in T. grandis. 13C labeling and sucrose feeding experiments indicated that sucrose‐induced changes in cell size and number contribute to cone enlargement in this species. RNA‐sequencing analysis, transient overexpression in T. grandis cones, and stable overexpression in tomato (Solanum lycopersicum) suggested that the expansin gene TgEXPA2 positively regulates cell expansion in T. grandis cones.The WRKY transcription factor TgWRKY47 directly enhances TgEXPA2 expression by binding to its promoter. Additionally, the NGATHA transcription factor TgNGA1 directly interacts with TgWRKY47. Objectives: Sinonasal inverted papilloma (SNIP) is a noncancerous tumor that develops in the mucous membrane of the nasal sinuses. Many malignancies are tightly linked to autophagy, an intracellular self‐degradation mechanism. HMGB1 has demonstrated its ability to modulate autophagy in many pathological conditions. This work investigates how HMGB1 and other genes involved in autophagy contribute to SNIP. Material and Methods: The study included 45 patients with SNIP and a control group consisting of 28 individuals. In each group, qPCR was employed to examine the mRNA expression levels of genes correlated with autophagy and HMGB1. HMGB1 and genes associated with autophagy were examined for protein expression levels via Western Blot and immunohistochemical staining assays. At the same time, the association between HMGB1 and genes involved in autophagy was discovered through correlation analysis. Furthermore, Krouse staging was utilized for investigating the expression levels of HMGB1 and other autophagy‐related genes at various stages in clinically staged SNIP patients. Results: LC3B, ATG5, and Beclin1 autophagy‐related genes and HMGB1 were substantially expressed in SNIP. Additionally, there was a positive correlation between HMGB1 and these genes. During various phases of SNIP, the levels of HMGB1 expression and autophagy‐related genes were notably elevated at stage T4 compared with stage T2. Conclusion: Clinical staging in SNIP is correlated with HMGB1 expression in conjunction with autophagy‐related genes LC3B, ATG5, and Beclin1, suggesting the possibility of novel prognostic indicators. Purpose: Teratozoospermia is the main pathogenic factor of male infertility. However, the genetic etiology of teratozoospermia is largely unknown. This study aims to clarify the relationship between novel variations in TENT5D and teratozoospermia in infertile patients. Materials and methods: Two infertile patients were enrolled. Routine semen analysis of patients and normal controls was conducted with the WHO guidelines. Whole‐exome sequencing (WES) was conducted to identify pathogenic variants in the two patients. Morphology and ultrastructure analysis of spermatozoa in the two patients was determined by Papanicolaou staining, scanning electron microscopy (SEM), and transmission electron microscopy (TEM). The functional effect of the identified variants was analyzed by immunofluorescence staining and western blotting. The expression of TENT5D in different germ cells was detected by immunofluorescence staining. Results: Two new hemizygous variations, c.101C > T (p.P34L) and c.125A > T (p.D42V), in TENT5D were detected in two patients with male infertility. Morphology analysis showed abnormalities in spermatozoa morphology in the two patients, including multiple heads, headless, multiple tails, coiled, and/or bent flagella. Ultrastructure analysis showed that most of the spermatozoa exhibited missing or irregularly arranged '9+2' structures. Further functional experiments confirmed the abrogated TENT5D protein expression in patients. In addition, both p.P34L and p.D42V substitutions resulted in a conformational change of the TENT5D protein. We precisely analyzed the subcellular localization of TENT5D in germ cells in humans and mice. And we found that TENT5D was predominantly detected in the head and flagellum of elongating spermatids and epididymal spermatozoa. Conclusions: Our results showed further evidence of a relationship between TENT5D mutation and human male infertility, providing new genetic insight for use in the diagnosis and treatment of male infertility. Objectives: The study aimed to explore the internal pathway of the association between activities of daily living and subjective well‐being among Chinese older adults using a large and representative sample. Background: There is still a lack of systematic investigation into the association between activities of daily living and subjective well‐being among Chinese older adults. Materials and methods: The study included 10 578 participants from the Chinese Longitudinal Healthy Longevity Survey. We applied multiple linear regression analysis and Spearman correlation analysis to examine the association between activities of daily living and subjective well‐being and PROCESS macro to determine the mediating effect of leisure activities on the relationship between them. Results: The mean total score of subjective well‐being was 30.5 ± 4.4. The prevalence of basic activities of daily living and instrumental activities of daily living limitations was 18.0% and 60.8%, respectively. Correlation analysis showed that activities of daily living score were negatively correlated with subjective well‐being. Leisure activities were positively correlated with subjective well‐being. The effect of instrumental activities of daily living on subjective well‐being was partially mediated by leisure activities and leisure activities completely mediated the association between basic activities of daily living and subjective well‐being. Conclusions: The effect of instrumental activities of daily living on subjective well‐being was partially mediated by leisure activities and leisure activities completely mediated the association between basic activities of daily living and subjective well‐being. Abnormal relaxation and contraction of intestinal smooth muscle can cause various intestinal diseases. Diarrhea is a common and important public health problem worldwide in epidemiology. Zingiber officinale Roscoe (fresh ginger) has been found to treat diarrhea, but the material basis and mechanism of action that inhibits intestinal peristalsis remain unclear. Metabolomics and serum pharmacology were used to identify differential metabolites, metabolic pathways, and pharmacodynamic substances, and were then combined with network pharmacology to explore the potential targets of ginger that inhibit intestinal peristalsis during diarrhea treatment, and the targets identified were verified using molecular docking and molecular dynamic simulation. We found that 25 active components of ginger (the six most relevant components), 35 potential key targets (three core targets), 40 differential metabolites (four key metabolites), and four major metabolic pathways were involved in the process by which ginger inhibits intestinal peristalsis during diarrhea treatment. This study reveals the complex mechanism of action and pharmacodynamic material basis of ginger in the inhibition of intestinal peristalsis, and this information helps in the development of new Chinese medicine to treat diarrhea and lays the foundation for the clinical application of ginger. Background: IKZF1 deletion (IKZF1del) is associated with poor prognosis in B‐cell precursor acute lymphoblastic leukemia (BCP‐ALL). But the prognosis of IKZF1del combined with other prognostic stratification factors remains unclear. Whether intensified treatment improves BCP‐ALL prognosis has not been determined. Method s : A retrospective analysis was performed on 1291 pediatric patients diagnosed with BCP‐ALL and treated with the South China Children's Leukemia 2016 protocol. Patients were stratified based on IKZF1 status for comparison of characteristics and outcome. Additionally, IKZF1del patients were further divided based on chemotherapy intensity for outcome assessments. Results: The BCP‐ALL pediatric patients with IKZF1del in south China showed poorer early response. Notably, the DFS and OS for IKZF1del patients were markedly lower than IKZF1wt group (3‐year DFS: 88.7% [95% CI: 83.4%–94.0%] vs. 93.5% [95% CI: 92.0%–94.9%], P =.021; 3‐year OS: 90.7% [95% CI: 85.8% to 95.6%] vs. 96.1% [95% CI: 95% to 97.2%, P =.003]), with a concurrent increase in 3‐year TRM (6.4% [95% CI: 2.3%–10.5%] vs. 2.9% [95% CI: 1.9%–3.8%], P =.025). However, the 3‐year CIR was comparable between the two groups (5.7% [95% CI: 1.8%–9.5%] vs. 3.7% [95% CI: 2.6%–4.7%], P =.138). Subgroup analyses reveal no factor significantly influenced the prognosis of the IKZF1del cohort. Noteworthy, intensive chemotherapy improved DFS from 85.7% ± 4.1% to 94.1% ± 0.7% in IKZF1del group (P =.084). Particularly in BCR::ABL positive subgroup, the 3‐year DFS was remarkably improved from 53.6% ± 20.1% with non‐intensive chemotherapy to 100% with intensive chemotherapy (P =.026). Conclusions: Pediatric BCP‐ALL patients with IKZF1del in South China manifest poor outcomes without independent prognostic significance. While no factor substantially alters the prognosis in the IKZF1del group. Intensified chemotherapy may reduce relapse rates and improve DFS in patients with IKZF1del subset, particularly in IKZFdel patients with BCR::ABL positive. [ABSTRACT FROM AUTHOR]

BACKGROUND: This study explored the denaturation of 11S globulin, a protein known for its diverse functional properties in soy protein applications, at pH 3.0 and pH 10.0, followed by a gradual return to pH 7.0 to facilitate renaturation. It investigated the structural and functional changes during renaturation induced by a change in pH, revealing the stabilization mechanism of 11S globulin. RESULTS: The findings revealed that during pH adjustment to neutral, the denatured soybean 11S globulin – resulting from alkaline (pH 10.0) or acidic (pH 3.0) treatments – experienced a refolding of its extended tertiary structure to varying extents. The particle size and the proportions of α‐helix and β‐sheet in the secondary structure aligned progressively with those of the natural‐state protein. However, for the alkali‐denatured 11S, the β‐sheet content decreased upon adjustment to neutral, whereas an increase was observed for the acid‐denatured 11S. In terms of functional properties, after alkaline denaturation, the foaming capacity (FC) and emulsifying activity index (EAI) of 11S increased by 1.4 and 1.2 times, respectively, in comparison with its native state. The solubility, foamability, and emulsifiability of the alkali‐denatured 11S gradually diminished during renaturation but remained superior to those of the native state. Conversely, these properties showed an initial decline, followed by an increase during renaturation triggered by pH neutralization. CONCLUSIONS: This research contributes to the enhancement of protein functionality, offering a theoretical foundation for the development of functional soy protein products and expanding their potential applications. © 2024 Society of Chemical Industry. Preimplantation genetic testing (PGT) can minimize the risk of birth defects. However, the accuracy and applicability of routine PGT is confounded by uneven genome coverage and high allele drop‐out rate from existing single‐cell whole genome amplification methods. Here, a method to diagnose genetic mutations and concurrently evaluate embryo competence by leveraging the abundant mRNA transcript copies present in trophectoderm cells is developed. The feasibility of the method is confirmed with 19 donated blastocysts. Next, the method is applied to 82 embryos from 26 families with monogenic defects for simultaneous mutation detection and competence assessment. The accuracy rate of direct mutation detection is up to 95%, which is significantly higher than DNA‐based method. Meanwhile, this approach correctly predicted seven out of eight (87.5%) embryos that failed to implant. Of six embryos that are predicted to implant successfully, four met such expectations (66.7%). Notably, this method is superior at conditions for mutation detection that are challenging when using DNA‐based PGT, such as when detecting pathogenic genes with a high de novo rate, multiple pseudogenes, or an abnormal expansion of CAG trinucleotide repeats. The placenta links feto‐maternal circulation for exchanges of nutrients, gases, and metabolic wastes between the fetus and mother, being essential for pregnancy process and maintenance. The allantois and mesodermal components of amnion, chorion, and yolk sac are derived from extraembryonic mesoderm (Ex‐Mes), however, the mechanisms contributing to distinct components of the placenta and regulation the interactions between allantois and epithelium during chorioallantoic fusion and labyrinth formation remains unclear. Isl1 is expressed in progenitors of the Ex‐Mes and allantois the Isl1 mut mouse line is analyzed to investigate contribution of Isl1+ Ex‐Mes / allantoic progenitors to cells of the allantois and placenta. This study shows that Isl1 identifies the Ex‐Mes progenitors for endothelial and vascular smooth muscle cells, and most of the mesenchymal cells of the placenta and umbilical cord. Deletion of Isl1 causes defects in allantois growth, chorioallantoic fusion, and placenta vessel morphogenesis. RNA‐seq and CUT&Tag analyses revealed that Isl1 promotes allantoic endothelial, inhibits mesenchymal cell differentiation, and allantoic signals regulated by Isl1 mediating the inductive interactions between the allantois and chorion critical for chorionic epithelium differentiation, villous formation, and labyrinth angiogenesis. This study above reveals that Isl1 plays roles in regulating multiple genetic and epigenetic pathways of vascular morphogenesis, provides the insight into the mechanisms for placental formation, highlighting the necessity of Isl1 for placenta formation/pregnant maintenance. The recent discovery of anti‐chiral edge states in magnetic semimetals linking two unequal‐frequency bulk Dirac nodes manifests a paradoxical picture in contrast to the chiral behavior in Chern insulators, implying an untapped phase may exist. Here, a unique Chern metal, exhibiting a nontrivial Chern number but without a complete bandgap is proposed, and the complete topological phase diagram in the modified Haldane model is obtained. Subsequently, the Chern metal in a 2D honeycomb‐lattice gyromagnetic photonic crystal by simultaneously breaking parity and time‐reversal symmetries with opposite and unequal magnetic biases at different sublattices is realized. For the paired complementary‐shape boundaries, such as zigzag/bearded, it is experimentally revealed that either clockwise or counter‐clockwise chiral character is switchable in Chern metals relying on different boundary choices, a remarkable property deemed as bi‐chiral edge propagation. The topological Chern metal with hidden bulk‐boundary correspondence is articulated to bridge the chiral and anti‐chiral paradox gap between Chern insulators and magnetic semimetals.
