Summary Salinity significantly inhibits plant growth and development. While the recretohalophyte Limonium bicolor can reduce its ion content by secreting salt, the metabolic pathways it employs to adapt to high salt stress remain unclear. This study aims to unravel this enigma through integrated transcriptomic and metabolomic analyses of L. bicolor under salt stress conditions. The results showed that compared to the control (S0), low salt treatment (S1) led to a significant increase in plant growth, photosynthesis efficiency and antioxidant enzyme activity but caused no significant changes in organic soluble substance and ROS contents. However, high salt treatments (S3 and S4) led to a significant decrease in plant growth, photosynthesis efficiency and antioxidant enzyme activity, accompanied by a significant increase in organic soluble substance and ROS contents. A significant increase in phenolic compounds, such as caffeoyl shikimic acid and coniferin, upon the treatments of S1, S3 and S4, and a decrease and increase in flavonoids upon the treatments of S1 and S3 were also observed, respectively. This study also demonstrated that the expression patterns of key genes responsible for the biosynthesis of these metabolites are consistent with the observed trends in their accumulation levels. These results suggest that under low salt stress conditions, the halophyte L. bicolor experiences minimal osmotic and oxidative stress. However, under high salt stress conditions, it suffers severe osmotic and oxidative stress, and the increase in organic soluble substances and flavonoids serves as a key response to these stresses and also represents a good strategy for the alleviation of them. Neutral sphingomyelinase‐2 (nSMase2), gene name sphingomyelin phosphodiesterase‐3 (Smpd3), is a key regulatory enzyme responsible for generating the sphingolipid ceramide. The function of nSMase2 in the brain is still controversial. To better understand the functional roles of nSMase2 in the aging mouse brain, we applied RNA‐seq analysis, which identified a total of 1462 differentially abundant mRNAs between +/fro and fro/fro, of which 891 were increased and 571 were decreased in nSMase2‐deficient mouse brains. The most strongly enriched GO and KEGG annotation terms among transcripts increased in fro/fro mice included synaptogenesis, synapse development, synaptic signaling, axon development, and axonogenesis. Among decreased transcripts, enriched annotations included ribosome assembly and mitochondrial protein complex functions. KEGG analysis of decreased transcripts also revealed overrepresentation of annotations for Alzheimer's disease (AD), Parkinson's disease (PD), and Huntington disease (HD). Ingenuity Pathway Analysis (IPA) tools predicted lower susceptibility to these neurodegenerative disorders, as well as predictions agreeing with stronger synaptic function, learning, and memory in fro/fro mice. The IPA tools identified signaling proteins, epigenetic regulators, and microRNAs as likely upstream regulators of the broader set of genes encoding the affected transcripts. It also revealed 16 gene networks, each linked to biological processes identified as overrepresented annotations among the affected transcripts by multiple analysis methods. Therefore, the analysis of these RNA‐seq data indicates that nSMase2 impacts synaptic function and neural development, and may contribute to the onset and development of neurodegenerative diseases in middle‐aged mice. Background: Trigeminal trophic syndrome (TTS) is a rare condition caused by damage to the trigeminal nervous system, resulting in sensory disturbances and ulcers on the face. Treating TTS is complex and often requires medical or surgical intervention like flap reconstruction. However, there is limited research on surgical treatments for TTS ulcers. Methods: We report the case of a 19‐year‐old man with TTS. We employed an innovative surgical technique involving dual cross‐face nerve grafts. In the initial procedure, corneal neuralization was accomplished using supraorbital and cross‐face infraorbital nerve graft. The subsequent operation utilized auricular composite tissue flap transplantation repair and cross‐face mental nerve graft. Results: This procedure led to rapid and sustained healing, as well as aesthetic improvement. Extracellular vesicles (EVs) are shed from the plasma membrane, but the regulation and function of these EVs remain unclear. We found that oxidative stress induced by H2O2 in Hela cells stimulated filopodia formation and the secretion of EVs. EVs were small (150 nm) and labeled for CD44, indicating that they were derived from filopodia. Filopodia‐derived small EVs (sEVs) were enriched with the sphingolipid ceramide, consistent with increased ceramide in the plasma membrane of filopodia. Ceramide was colocalized with neutral sphingomyelinase 2 (nSMase2) and acid sphingomyelinase (ASM), two sphingomyelinases generating ceramide at the plasma membrane. Inhibition of nSMase2 and ASM prevented oxidative stress‐induced sEV shedding but only nSMase2 inhibition prevented filopodia formation. nSMase2 was S‐palmitoylated and interacted with ASM in filopodia to generate ceramide for sEV shedding. sEVs contained nSMase2 and ASM and decreased the level of these two enzymes in oxidatively stressed Hela cells. A novel metabolic labeling technique for EVs showed that oxidative stress induced secretion of fluorescent sEVs labeled with NBD‐ceramide. NBD‐ceramide‐labeled sEVs transported ceramide to mitochondria, ultimately inducing cell death in a proportion of neuronal (N2a) cells. In conclusion, using Hela cells we provide evidence that oxidative stress induces interaction of nSMase2 and ASM at filopodia, which leads to shedding of ceramide‐rich sEVs that target mitochondria and propagate cell death. Purpose: This work proposes a robot‐assisted augmented reality (AR) surgical navigation system for mandibular reconstruction. The system accurately superimposes the preoperative osteotomy plan of the mandible and fibula into a real scene. It assists the doctor in osteotomy quickly and safely under the guidance of the robotic arm. Methods: The proposed system mainly consists of two modules: the AR guidance module of the mandible and fibula and the robot navigation module. In the AR guidance module, we propose an AR calibration method based on the spatial registration of the image tracking marker to superimpose the virtual models of the mandible and fibula into the real scene. In the robot navigation module, the posture of the robotic arm is first calibrated under the tracking of the optical tracking system. The robotic arm can then be positioned at the planned osteotomy after the registration of the computed tomography image and the patient position. The combined guidance of AR and robotic arm can enhance the safety and precision of the surgery. Results: The effectiveness of the proposed system was quantitatively assessed on cadavers. In the AR guidance module, osteotomies of the mandible and fibula achieved mean errors of 1.61 ± 0.62 and 1.08 ± 0.28 mm, respectively. The mean reconstruction error of the mandible was 1.36 ± 0.22 mm. In the AR‐robot guidance module, the mean osteotomy errors of the mandible and fibula were 1.47 ± 0.46 and 0.98 ± 0.24 mm, respectively. The mean reconstruction error of the mandible was 1.20 ± 0.36 mm. Conclusions: The cadaveric experiments of 12 fibulas and six mandibles demonstrate the proposed system's effectiveness and potential clinical value in reconstructing the mandibular defect with a free fibular flap. Ultra‐deep marine carbonate reservoirs is an important field for oil and gas exploration, and it is also a hot research topic in recent years. However, their hydrocarbon episodic charging model is still under debate up to now. A great breakthrough has been made in the newly discovered ultra‐deep fault‐karst oil reservoir in the Shunbei area of Tarim Basin. Based on quantitative gas chromatogram‐mass spectrometry, stable carbon isotopes of reservoir oils, microthermometry and confocal laser scanning analyses of fluid inclusions, the reservoir oil origin and hydrocarbon charging history in Shunbei No. 1, No. 5 and No. 7 strike‐slip fault zones are expounded. The results showed that the Ordovician reservoir oil derives from the source rocks within the Cambrian Yuertusi Formation deposited in a highly anoxic bottom water environment. There were four stages of hydrocarbon charging in the study area, in the late Caledonian, late Hercynian–Indosinian, middle Yanshanian and late Himalayan. During this period, the force‐field strength of the charged oil gradually decreased, and the early‐stage charged crude oil with low maturity and high force‐field strength preferentially accumulated in the Yijianfang Formation, while the late‐stage charged crude oil with high maturity and low force‐field strength preferentially accumulated in the Yingshan Formation. This study reveals the history and differences of petroleum accumulation in the Shunbei fault zones, which is significance for understanding the hydrocarbon distribution in ultra‐deep marine carbonate reservoirs. The formation of extracellular vesicles (EVs) is induced by the sphingolipid ceramide. How this pathway is regulated is not entirely understood. Here, we report that the ceramide transport protein (CERT) mediates a non‐vesicular transport of ceramide between the endoplasmic reticulum (ER) and the multivesicular endosome at contact sites. The process depends on the interaction of CERT's PH domain with PI4P generated by PI4KIIα at endosomes. Furthermore, a complex is formed between the START domain of CERT, which carries ceramide, and the Tsg101 protein, which is part of the endosomal sorting complex required for transport (ESCRT‐I). Inhibition of ceramide biosynthesis reduces CERT‐Tsg101 complex formation. Overexpression of CERT increases EV secretion while its inhibition reduces EV formation and the concentration of ceramides and sphingomyelins in EVs. In conclusion, we discovered a function of CERT in regulating the sphingolipid composition and biogenesis of EVs, which links ceramide to the ESCRT‐dependent pathway. [ABSTRACT FROM AUTHOR]

Metasurface absorber (MA) has been a research hotspot in the field of artificial electromagnetic structural material due to its dual advantages of high performance and compact design. Usually, the design of MA depends on the designer's professional knowledge, experience and physical inspiration. The desired optical response can be obtained by using electromagnetic simulation software to carry out hundreds or thousands of numerical calculations. Thus, it is still a challenge to quickly retrieve the optimal structure according to the desired optical response and realize the on‐demand inverse design. Besides, limited by the inner physics of the MA, it is not always possible to find the structural parameters corresponding to the desired spectrum. This paper not only takes the planar geometry and thickness of the structures into account but also realizes the probability classification of the desired spectra through the classification network. According to the classification results, the prediction network of the corresponding is selected to realize the on‐demand inverse design of MA. The proposed network model can design MA rapidly and accurately in a data‐driven way and can be flexibly applied to the design of other data‐enabled photonic devices, which is promising to become a comprehensive and effective design tool. Introduction: Innovative strategy is urgently needed to precisely discover novel natural products as lead compounds for development of new drugs against orphan diseases such as triple‐negative breast cancer (TNBC). Herein, we describe a targeting pharmacophore with probe‐reactivity‐guided strategy for the discovery of electrophilic sesquiterpene (ES), a class of bioactive natural product. Objective: This study aimed to identify pharmacophore, based on pharmacophore with probe‐reactivity‐guided strategy for precisely discovering ESs from ethyl acetate extract of Eupatorium chinense L. (EEEChL) Methodology: MTT assay combined with ultra‐performance liquid chromatography (UPLC) analysis was used to identify pharmacophore. UPLC‐mass spectrometry (MS) was applied to carefully compare the intrinsic reactivity characteristics of two chemoselective nucleophilic probes: glutathione (GSH) and 4‐bromothiophenol (BTP) reaction with ESs. ESs was isolated and identified from EEEChL by phytochemical methods. Furthermore, stoichiometric ratio and binding site of one typical ES 8β‐[4′‐hydroxytigloyloxy]‐5‐desoxy‐8‐desacyleuparotin (HDDE) reaction with BTP were studied by UPLC‐quadrupole time‐of‐flight (Q‐TOF)‐MS and two‐dimensional nuclear magnetic resonance (NMR). Results: Eleven ESs were identified from EEEChL, MTT assay illustrated that all of the 11 ESs possess fairly good anti‐TNBC activity Conclusions: Electrophilic groups were confirmed as pharmacophore of bioactive compounds contained in EEEChL. An optimised halogenated aromatic probe BTP furnishes ES‐BTP conjugates that are highly conspicuous via MS by virtue of a unique isotopic bromine signature, conjugates also have a considerable separation on C18 column. The new probe‐reactivity‐guided strategy can effectively improve the traditional bioassay‐guided approaches, and significantly increase the probability of obtaining designated bioactive compounds. Based on the fact that the Hankel matrix representing clean seismic data is low rank, low‐rank approximation methods have been widely utilized for removing noise from seismic data. A common strategy for real seismic data is to perform the low‐rank approximations for small local windows where the events can be approximately viewed as linear. This raises a fundamental question of selecting an optimal rank that best captures the number of events for each local window. Gavish and Donoho proposed a method to select the rank when the noise is independent and identically distributed. Gaussian matrix by analysing the statistical performance of the singular values of the Gaussian matrices. However, such statistical performance is not available for noisy Hankel matrices. In this paper, we adopt the same strategy and propose a rule that computes the number of singular values exceed the median singular value by a multiplicative factor. We suggest a multiplicative factor of 3 based on simulations which mimic the theories underlying Gavish and Donoho in the independent and identically distributed Gaussian setting. The proposed optimal rank selection rule can be incorporated into the classical low‐rank approximation method and many other recently developed methods such as those by shrinking the singular values. The low‐rank approximation methods with optimally selected rank rule can automatically suppress most of the noise while preserving the main features of the seismic data in each window. Cyclin-dependent protein kinases (CDKs) are required for various cell cycle events both in mitosis and in meiosis. During the meiotic prophase of Saccharomyces cerevisiae, only one CDK, Cdc28, which forms a complex with B-type cyclins, Clb5 or Clb6, promotes not only the onset of premeiotic DNA replication but also the formation of meiotic double-strand breaks (DSBs). In this study, we showed that Cdc28 exhibits punctate staining on chromosomes during meiotic prophase I. Chromosomal localization of Cdc28, dependent on Clb5 and/or Clb6, is frequently observed in zygotene and pachytene, when formation of the synaptonemal complex (SC) occurs. Interestingly, the CDK localization is independent of DSB formation, but rather dependent on meiosis-specific chromosome components such as Red1, Hop1 and a cohesin subunit Rec8. Compromised CDK activity in meiotic prophase leads to defective SC formation without affecting DSB formation. These results suggest that CDK-dependent phosphorylation regulates meiotic chromosome morphogenesis. [ABSTRACT FROM AUTHOR]

ABSTRACT Under vibration conditions, continuous alternating loads cause fatigue damage to structures, affecting their stability, durability, and overall safety. In this paper, the additional damping structures with entangled metallic wire material (EMWM) is proposed. Dynamic tests with different loading frequencies and loading amplitudes are carried out to evaluate the energy dissipation and stiffness characteristics of the different structures in terms of energy dissipation, loss factor, and average stiffness. Fatigue tests are conducted, and the fatigue life tests are applied with different loading amplitudes and loading frequencies using displacement control, and the fatigue properties are obtained under different numbers of cyclic loading. The test results are analyzed with respect to the energy dissipation mechanism of EMWM and the characteristics of the additional damping structure. The hysteresis curves under different loading times are identified by parameters, and the fatigue mechanical models are constructed. Comparison between the predicted data of the model and the hysteresis curve data of the test indicates that the model has high accuracy. [ABSTRACT FROM AUTHOR]

Little is known about the dynamic change and risk factors of intense thirst in neuro‐intensive care unit (Neuro‐ICU).The objective of this study was to explore the dynamic change and determine the risk factors of intense thirst in patients admitted to Neuro‐ICU, providing reference for personalized interventions of intense thirst.The study design is a prospective observational study. Demographic and disease‐related, treatment, physiological and biochemical data were collected for 230 patients from Neuro‐ICU of a hospital from May 2023 to November 2023. We assessed thirst intensity on numeric rating scale (NRS) of 0–10 (10 = worst possible thirst) at eight time points: 7:00, 9:00, 11:00, 13:00, 15:00, 17:00, 19:00 and 21:00 and a self‐designed general information questionnaire based on safety protocol for thirst management to analyse the risk factors of intense thirst in Neuro‐ICU patients. If thirst scores were more than 7, we defined it as intense thirst. This study followed the STROBE checklist for cross‐sectional studies.A total of 230 Neuro‐ICU patients were observed. The dynamic analysis results showed an overall downward trend in thirst intensity, with the highest NRS thirst scores at 07:00 (6.13 ± 2.14) and the lowest at 21:00 (4.02 ± 2.72). The investigation of the current situation showed that the incidence of intense thirst in Neuro‐ICU patients was 47.4%. Intense thirst in Neuro‐ICU patients was predicted by dysphagia (odds ratio [OR] = 1.436, 95% confidence interval [CI]:1.063–1.941), open mouth breathing (OR = 2.201, 95% CI:1.041–4.656), high glucose (OR = 2.584, 95% CI:1.097–6.087), xerostomia (OR = 3.049, 95% CI:1.950–4.767) (all p < .05).The intensity of thirst was dynamically changing and the incidence of intense thirst in Neuro‐ICU patients was relatively high. Timely assessment of Neuro‐ICU patients' thirst severity and identification of those at high risk can ensure the implementation of effective interventions based on patients' characteristics.Thirst is a pervasive distressing symptom often reported by critically ill patients. This study revealed that the clinical nurses need to enhance their focus on dynamic change of thirst, which is helpful for improving the efficiency of bundled thirst interventions at the suitable time. Wing dimorphism in Nilaparvata lugens is controlled by the insulin‐like growth factor 1 (IGF‐1) signaling – Forkhead transcription factors (IIS‐FoxO) pathway. However, the role of this signal in the wing development program remains largely unclear. Here, we identified 2 R‐SMAD proteins, NlMAD1 and NlMAD2, in the brown planthopper (BPH) transcriptome, derived from the intrinsic transforming growth factor‐β pathway of insect wing development. Both proteins share high sequence similarity and conserved domains. Phylogenetic analysis placed them in the R‐SMAD group and revealed related insect orthologs. The expression of Nlmad1 was elevated in the late instar stages of the macropterous BPH strain. Nlmad1 knockdown in nymphs results in malformed wings and reduced wing size in adults, which affects the forewing membrane. By contrast, Nlmad2 expression was relatively consistent across BPH strains and different developmental stages. Nlmad2 knockdown had a milder effect on wing morphology and mainly affected forewing veins and cuticle thickness in the brachypterous strain. NlMAD1 functions downstream of the IIS‐FoxO pathway by mediating the FoxO‐regulated vestigial transcription and wing morph switching. Inhibiting Nlmad1 partially reversed the long‐winged phenotype caused by NlFoxO knockdown. These findings indicate that NlMAD1 and NlMAD2 play distinct roles in regulating wing development and morph differentiation in BPH.
