Fracture healing is a complex staged repair process in which the mechanical environment plays a key role. Bone tissue is very sensitive to mechanical stress stimuli, and the literature suggests that appropriate stress can promote fracture healing by altering cellular function. However, fracture healing is a coupled process involving multiple cell types that balance and limit each other to ensure proper fracture healing. The main cells that function during different stages of fracture healing are different, and the types and molecular mechanisms of stress required are also different. Most previous studies have used a single mechanical stimulus on individual mechanosensitive cells, and there is no relatively uniform standard for the size and frequency of the mechanical stress. Analyzing the mechanisms underlying the effects of mechanical stimulation on the metabolic regulation of signaling pathways in cells such as in bone marrow mesenchymal stem cells (BMSCs), osteoblasts, chondrocytes, and osteoclasts is currently a challenging research hotspot. Grasping how stress affects the function of different cells at the molecular biology level can contribute to the refined management of fracture healing. Therefore, in this review, we summarize the relevant literature and describe the effects of mechanical stress on cells associated with fracture healing, and their possible signaling pathways, for the treatment of fractures and the further development of regenerative medicine. Aims: Despite refinements in balloon pulmonary angioplasty (BPA), total occlusion remains a challenge in chronic thromboembolic pulmonary hypertension (CTEPH). Owing to their low success and high complication rates, most interventional cardiologists are reluctant to address total occlusion, and there is a paucity of literature on BPA performance in total occlusion. We aimed to classify total occlusion according to morphology and present an illustrative approach for devising a tailored treatment strategy for each distinct type of total occlusion. Methods and results: All patients diagnosed with CTEPH who underwent BPA between May 2018 and May 2022 at Fuwai Hospital in Beijing, China, were included retrospectively. A total of 204 patients with CTEPH who underwent BPA were included in this study. Among these, 38 occluded lesions were addressed in 33 patients. Based on the morphology, we categorized the lesions into three groups: pointed‐head, round‐head, and orifice occlusions. Pointed‐head occlusion could be successfully addressed using soft‐tip wire, round‐head occlusion warranted hard‐tip wire and stronger backup, and orifice occlusion warranted the strongest backup force. The success rates for each group were as follows: pointed‐head (95.45%), round‐head (46.15%), and orifice occlusion (33.33%), with orifice occlusion having the highest complication rate (50%). The classification of occlusion was associated with BPA success (round‐head occlusion vs. pointed‐head occlusion, OR 24.500, 95% CI 2.498–240.318, P = 0.006; orifice occlusion vs. pointed‐head occlusion, OR 42.000, 95% CI 3.034–581.434, P = 0.005). Conclusions: Occlusion morphology has a significant impact on BPA success and complication rates. Inflammation contributes to development of idiopathic pulmonary arterial hypertension (IPAH), and tumor biomarkers can reflect inflammatory and immune status. We aimed to determine the value of tumor biomarkers in IPAH comprehensively. We enrolled 315 patients with IPAH retrospectively. Tumor biomarkers were correlated with established indicators of pulmonary hypertension severity. Multivariable Cox regression found that AFP (hazard ratio [HR]: 1.587, 95% confidence interval [CI]: 1.014–2.482, p = 0.043) and CA125 (HR: 2.018, 95% CI: 1.163–3.504, p = 0.013) could independently predict prognosis of IPAH. The changes of AFP over time were associated with prognosis of patients, each 1 ng/mL increase in AFP was associated with 5.4% increased risk of clinical worsening (HR: 1.054, 95% CI: 1.001–1.110, p = 0.046), enabling detection of disease progression. Moreover, beyond well‐validated PH biomarkers, CA125 was still of prognostic value in the low‐risk patients (HR: 1.014, 95% CI: 1.004–1.024, p = 0.004), allowing for more accurate risk stratification and prediction of disease outcomes. This paper selects 141 countries from 2009 to 2020 to investigate the impact of genetic distance on bilateral foreign direct investment (FDI). Through theoretical and empirical analysis, we get the following conclusions. First, genetic distance has a significant negative impact on bilateral FDI by increasing transaction costs. This conclusion still holds after several robust checks. Second, genetic distance has different effects on bilateral FDI for different countries: (1) If at least one of the two countries is a low‐income country, genetic distance will have a significant negative impact on their FDI, but no significant impact on both high‐income countries; (2) if at least one of the two countries is not a member of the Belt and Road Initiative, genetic distance will have a significant negative impact, but no significant impact on both member countries; and (3) genetic distance has a significant negative impact only when both countries are World Trade Organization members. Third, further analysis shows that a smaller genetic distance has no significant effect on bilateral FDI, and only when the genetic distance is larger than the average would a significant negative impact exists. Objective: Intertrochanteric fracture of the femur is a common fracture in older people. Due to the poor systemic condition and prognosis of elderly patients, it is prone to more complications. We introduce the bone‐setting concept in the design of the robots, which are used for intertrochanteric fracture of the femur reduction. The purpose of this study is to compare the effect of bone‐setting robots and conventional reduction in the treatment of intertrochanteric fracture of the femur (IFF). Methods: From June 2021 to January 2023, 60 patients with IFF who were treated surgically were assigned to bone‐setting robots group and conventional reduction methods group in this retrospective study. The reduction time, operation time, total time, intraoperative blood loss, incision length, fluoroscopy time, and the follow‐up time were reviewed. The visual analogue scale (VAS) and Harris scores were used for functional assessment. For continuous variables, independent t‐tests were applied; for categorical data, the chi‐square test was applied. The significance level as p < 0.05. Results: Among the 60 patients with IFF, 31 were assigned to the bone‐setting robots group, and 29 were assigned to the conventional reduction methods group. Both groups with a similar baseline in the number, gender, age, and classification (p > 0.05). The reduction time, operation time, total time, intraoperative blood loss, and fluoroscopy time were less than those in the bone‐setting robots reduction group compared to the conventional reduction group. In the bone‐setting robots reduction group, the preoperative VAS score was 6.2 ± 1.3, the Harris score was 35.3 ± 3.1, 1 week after surgery VAS score was 3.3 ± 1.2, the Harris score was 57.3 ± 3.7, and at the last follow‐up VAS score was 2.4 ± 0.8, and the Harris score was 88.7 ± 3.4. While in the conventional reduction group, the preoperative VAS score was 6.3 ± 1.3, the Harris score was 35.9 ± 2.9, 1 week after surgery VAS score was 4.8 ± 1.4, the Harris score was 46.8 ± 2.8, and at the last follow‐up VAS score was 2.6 ± 0.8, and the Harris score was 87.3 ± 3.3. There were no significant differences between the two groups at the preoperative and 6‐month postoperative follow‐ups in VAS score and Harris score (p > 0.05, p > 0.05, respectively). But the difference was statistically significant at the one‐week postoperative follow‐up in VAS and Harris scores (p < 0.001). Conclusion: The bone‐setting robots can better protect the "fracture environment" and have the advantages of being precise, minimally invasive, simple, short time, low radiation, and rapid fracture recovery. Pulmonary artery (PA) dilatation is commonly observed in patients with pulmonary hypertension (PH). However, the clinical aspects of PA dilatation in various etiology of PH remain unknown. In this study, we investigated the clinical and imaging characteristics of 1018 patients with different subtypes of pulmonary arterial hypertension (PAH) and chronic thromboembolic pulmonary hypertension (CTEPH). The independent determinants for all‐cause death were identified using univariate and multivariate Cox proportional hazard models. PA dilatation was identified in 88.8% of the patients, and 27.2% had a PA diameter/ascending aorta diameter ratio greater than 1.5. PA diameter was shown to be significantly correlated with hemodynamic parameters and symptom duration in idiopathic PAH patients. PA diameter only correlated with pulmonary circulation volume in patients with PAH associated with congenital heart disease. PA diameter correlated with symptom duration and right ventricular end‐diastolic dimension in CTEPH patients. PA diameter correlated with right ventricular end‐diastolic dimension in patients with PAH associated with connective tissue disease. Only 6‐min walk distance, but not PA dilatation, predicts all‐cause death independently. In conclusion, PA dilatation is a common finding in PH patients. BACKGROUND: The natural fermentation of multispecies microbial communities is responsible for unique flavors of winery regions of the eastern foothills of the Ningxia Helan Mountains in China. However, the participation of different microorganisms in the metabolic network for the development of important flavor substances is not clearly defined. Microbial population and diversity on different fermentation phases of Ningxia wine were analyzed by metagenomic sequencing approach. RESULTS: Gas chromatography–mass spectrometry and ion chromatography were used to identify flavor components, and 13 esters, 13 alcohols, nine aldehydes and seven ketones were detected in volatile substances with odor activity values > 1, and eight organic acids were detected as important flavor components in young wine. Thus, 52 238 predicted protein‐coding genes from 24 genera were identified in the Kyoto Encyclopedia of Genes and Genomes level 2 pathways of global and overview maps, and the genes were primarily involved in amino acid metabolism and carbohydrate metabolism. Major microbial genera (Saccharomyces, Tatumella, Hanseniaspora, Lactobacillus, and Lachancea) were closely related to self‐characteristic compound metabolism and further contributed to wine flavor. CONCLUSION: This study clarifies the different metabolic roles of microorganisms in flavor formation during Ningxia wine spontaneous fermentation. Saccharomyces, dominant fungi involved in glycolysis and pyruvate metabolism, produces not only ethanol but also two important precursors, pyruvate and acetyl‐CoA, which are necessary for the tricarboxylic acid cycle, fatty acid metabolism, amino acid metabolism, and flavor formation. Lactobacillus and Lachancea, dominant bacteria involved in lactic acid metabolism. Tatumella, dominant bacteria involved in amino acid metabolism, fatty acid metabolism, and acetic acid metabolism to produce esters in the Shizuishan City region samples. These findings provide insights into the use of local functional strains to generate unique flavor formation, as well as improved stability and quality, in wine production. © 2023 Society of Chemical Industry. BACKGROUND: The widespread incidence of "false smut" disease in rice has caused extensive ustiloxin contamination around the world. Until now there has been a lack of knowledge regarding the natural occurrence of ustiloxins in paddy. The development of efficient removal methods is also still a challenge that remains unexplored. RESULTS: In the current study, three main ustiloxins – ustiloxin A (UA), ustiloxin B (UB), and ustiloxin G (UG) – were determined simultaneously by ultraperformance liquid chromatography‐tandem mass spectrometry (UPLC–MS/MS) in 206 paddy samples collected in 2021 from five rice‐producing provinces in China. The predominant ustiloxin was UA with an occurrence of 46.1% and an average concentration of 49.71 μg kg−1. This was followed by UB (31.1%, 13.31 μg kg−1) and UG (18.4%, 9.19 μg kg−1). No targeted ustiloxins were detected in white rice samples randomly collected from supermarkets in Shanghai. To reveal the causes, two approaches were tested for the removal of the ustiloxins: most of the targeted ustiloxins (>93%) were removed in brown rice by husking and, subsequently, all targeted ustiloxins (100%) were removed by whitening. CONCLUSION: A wide distribution of ustiloxins was discovered in paddy samples in this study. The UA contaminations were significantly different depending on their origin, with the highest occurrence in paddy from Shanghai and Jiangsu, southeast coast provinces in China. Contamination by UG was also found in paddy for the first time and was strongly correlated with those of UA and UB. A combination of husking and whitening has been verified to be a practicable and promising way to ensure efficient removal and food safety. © 2023 Society of Chemical Industry. Rainfall fluctuations are the major threats caused by climate change. This paper analyses the impact of seasonal rainfall fluctuations on rural poverty using panel data for 992 counties, which are located in humid zone in the south of China from 2000 to 2018. The findings are as follows: (1) Rainfall fluctuations significantly affect rural poverty, specifically, increased rainfall significantly alleviates rural poverty in the dry season, but aggravates rural poverty in the rainy season. (2) The effect of rainfall fluctuations on rural poverty is significantly heterogeneous in different regions. (3) Rainfall fluctuations put low‐income areas at greater risk of returning to poverty. (4) The mechanism through which rainfall fluctuations affect rural poverty is agricultural production, and market regulation can mitigate the shock of rainfall fluctuations. Aims: The prognosis is poor for patients with pulmonary artery stenosis‐associated pulmonary hypertension (PAS‐PH). Identifying predictors of prognosis in PAS‐PH is crucial to preventing premature death, which has rarely been investigated. We aimed to explore the cardiopulmonary exercise testing (CPET) parameters to predict the prognosis of these patients. Methods: We prospectively included all patients with PAS‐PH who underwent CPET between September 2014 and June 2021 in Fuwai Hospital (ClinicalTrials.gov ID: NCT02061787). The primary outcome was clinical worsening, including death, rehospitalization for heart failure, or deterioration of PH. Results: Seventy‐two patients were included in this study. A median of 2‐year follow‐up revealed that 18 (25%) patients experienced clinical worsening. The 1‐year, 3‐year, and 5‐year event‐free survival rates were 92.5%, 81.7%, and 62.7%, respectively. Patients with clinical worsening demonstrated significantly worse baseline haemodynamics and poorer exercise capacity than their counterparts. Multivariable Cox regression identified that peak O2 pulse could independently predict clinical worsening [hazard ratio: 0.344, 95% confidence interval (CI) 0.188–0.631, P < 0.001], outperforming other parameters. Peak O2 pulse correlated with PH severity. Incorporating peak O2 pulse into the simplified 2015 European Society of Cardiology/European Respiratory Society risk stratification improved the accuracy for predicting clinical worsening (pre vs. post area under the curve: 0.727 vs. 0.846, P < 0.001; net reclassification index: 0.852, 95% CI 0.372–1.332, P < 0.001; integrated discrimination index 0.133, 95% CI 0.031–0.235, P = 0.011). Conclusions: The prognosis is poor for PAS‐PH, and exercise intolerance and ventilation inefficiency are commonly observed. Peak O2 pulse independently predicted the prognosis of these patients. Objective: Although widely used in clinical practice, vertebral augmentation procedure (VAP) for osteoporotic vertebral compression fracture (OVCF) is not supported. Recently, the effect of recombinant human parathyroid hormone (1–34) (rhPTH) has been paid great attention for its efficacy in anti‐osteoporosis and bone union. This study aims to explore the outcome of rhPTH on acute OVCF and compare it with VAP to clarify its therapeutic advantages. Methods: The retrospective study comprised 71 acute OVCF patients from January 2015 to March 2020: 22 received rhPTH treatment (rhPTH group) and 49 underwent VAP (VAP group). The rhPTH group was 15 women and seven men with an average of 76.18 years, and the VAP group were 35 women and 14 men with an average of 73.63 years. The thoracic/lumbar vertebrae were 14/8 in the rhPTH group and 29/20 in the VAP group. The average follow‐up period was 14.05 months in the rhPTH group and 13.82 months in the VAP group. The two groups were assessed regarding the visual analog score (VAS), Oswestry Disability Index (ODI), OVCF bone union, bone mineral density (BMD), kyphotic angle (KA), anterior and posterior border height (ABH and PBH, respectively), adverse events and the health‐related quality of life assessed by short form‐36 health survey scores (SF‐36). Categorical variables were analyzed by chi‐square test and continuous variables between groups were analyzed by independent samples t‐test or Mann–Whitney U test according to the normality. Results: During the follow‐up, the VAS was significantly lower in the rhPTH group than in the VAP group at month 3 (0.39 ± 0.6 vs 0.68 ± 0.651) (p = 0.047), month 6 (0.45 ± 0.60 vs 2.18 ± 1.22) (p < 0.001), and month 12 (0.45 ± 0.60 vs 2.43 ± 1.49) (p < 0.001). At month 12, the ODI was significantly lower in the rhPTH group (18.59 ± 3.33%) than in the VAP group (28.93 ± 16.71%) (p < 0.001). Bone bridge was detected on sagittal computed tomography images of all fractured vertebrae in the rhPTH group. The BMD was significantly higher in the rhPTH group (87.66 ± 5.91 Hounsfield units [HU]) than in the VAP group (68.15 ± 11.32HU) (p < 0.001). There were no significant differences in the changes in KA, ABH, and PBH between groups (all p > 0.05). The incidence of new OVCF was significantly lower in the rhPTH group than in the VAP group (p = 0.042). All scores of SF‐36 were significantly higher in the rhPTH group than in the VAP group (all p < 0.05). Conclusion: In acute OVCF patients, rhPTH was better than VAP in increasing spinal BMD to promote OVCF healing, reduce new OVCF, and improve back pain, physical ability, and health‐related quality of life. In this study, an eco‐friendly method was proposed to prepare carrageenan suitable for spinning into fibers. The extract from Hypnea cervicornis without alkali pretreatment was directly converted into the spinning solution, and carrageenan fibers were obtained via wet spinning. The improved procedure had a higher yield (61.8% ± 1.1%) than the traditional method (36.0% ± 1.3%). The chemical structure of the carrageenan was analyzed via fourier transform infrared spectroscopy (FTIR) and carbon nuclear magnetic resonance spectroscopy (13C NMR). It was confirmed that carrageenan extracted from Hypnea cervicornis was mainly ι‐carrageenan and its structure was not changed by alkali pretreatment. The properties of the spinning solution and fibers were characterized using dynamic light scattering (DLS), scanning electron microscope (SEM), energy dispersive spectrometer (EDS), and tensile tests. The results showed that the spinning solution prepared by the improved procedure was homogeneous and suitable for spinning, and the mechanical strength of the novel prepared carrageenan fibers was equal to the fibers spanned from traditional carrageenan. In this article, we discuss the numerical solution for the two‐dimensional (2‐D) damped sine‐Gordon equation by using a space–time continuous Galerkin method. This method allows variable time steps and space mesh structures and its discrete scheme has good stability which are necessary for adaptive computations on unstructured grids. Meanwhile, it can easily get the higher‐order accuracy in both space and time directions. The existence and uniqueness to the numerical solution are strictly proved and a priori error estimate in maximum‐norm is given without any space–time grid conditions attached. Also, we prove that if the mesh in each time level is generated in a reasonable way, we can get the optimal order of convergence in both temporal and spatial variables. This study determined the growth performance, correlations of linear body dimensions, slaughter performance and the fitting model of two generations of Leizhou Black Ducks. Fifteen (15) male and 225 female parents forming generation 0 were selected from the Leizhou duck population. Fifteen (15) families were created in the ratio of 1 male:15 females. Eggs from all the families were collected and numbered according to the family. Generation 1 ducklings were selected and grouped into families in the same ratio. Body weights (BWs) and linear body dimensions were recorded every 2 weeks from weeks 0 to 16. Logistic, Gompertz and Von Bertalanffy models were employed to ascertain the growth model of both sexes of Leizhou Black Ducks. Except for weeks 0–2, generation 1 families had greater BWs than the generation 0 families (p <.05). Males from both generations had significantly higher (p <.01) BWs than females at 16 weeks old. Significant positive correlations were observed between BWs and measured body dimensions for both sexes except for between BW and pelvis width (PW) where negative correlations (p >.05) were observed for males at week 10 and females at 16 weeks old. There was a significant negative correlation (p <.01) between body length and PW in males at 10 weeks. The accuracy rate of Logistic, Gompertz and Von Bertalanffy models was at 0.972 and similar was obtained by all three models with Von Bertalanffy being the best model. Live weight of generation 1 before slaughter was significantly higher (p <.01) than generation 0, whereas no significant differences were seen in the other carcass traits. This study was conducted to evaluate the effects of dietary sodium butyrate (SB) supplementation on growth performance, the development of gastrointestinal tract and immune organs (thymus, spleen and bursa of fabricius), and serum antibody titer after Newcastle disease (ND) vaccination in broilers. The total of 288 1‐day‐old broilers were randomly allocated to four groups with six replications according to initial body weight. Four treatment groups were designed as follows and fed the indicated diets: CON, basal diet; T1, basal diet supplemented with 0.3 g/kg SB; T2, basal diet supplemented with 0.6 g/kg SB; T3, basal diet supplemented with 1.2 g/kg SB. During days 1–21, broilers fed the T2 diet had higher (p <.05) average daily gain (ADG) than broilers fed the CON diet. On day 21, dietary SB supplementation showed linear increase (p <.05) in relative weight of the duodenum, jejunum, ileum, small intestine (the sum weight of duodenum, jejunum and ileum), pancreas and thymus, and linear increase (p <.05) in relative length of the duodenum, jejunum, ileum, small intestine (the sum length of duodenum, jejunum and ileum) and caeca. Meanwhile, dietary SB supplementation showed linear increase in the antibody titer against ND on days 14, 21, 28 and 35. In conclusion, dietary SB supplementation improved the development of gastrointestinal by increasing the relative weight and length, as well as enhanced the immune response of ND vaccine. According to our previous studies, bta‐miR‐152, PRKAA1 and UCP3 are differentially expressed in mammary gland tissues of high milk fat and low milk fat cows, and the trend in bta‐miR‐152 expression is opposite from those of PRKAA1 and UCP3. To further identify the function and regulatory mechanism of bta‐miR‐152 in milk fat metabolism, we investigated the effect of bta‐miR‐152 on cellular triglyceride content in bovine mammary epithelial cells cultured in vitro, on the basis of bta‐miR‐152 overexpression and inhibition assays. The target genes of bta‐miR‐152 were identified through qPCR, Western blotting and dual luciferase reporter gene detection. Compared with that in the control group, the expression of UCP3 was significantly lower in the bta‐miR‐152 mimic group, the expression of PRKAA1 was decreased, and the intracellular TAG content was significantly increased. After transfection with bta‐miR‐152 inhibitor, the expression of UCP3 increased significantly, and the expression of PRKAA1 decreased, but the difference was not significant; in addition, the intracellular TAG content decreased significantly. The ban on the use of antibiotic in feed encouraged nutritionists to using alternatives to maintain growth performance and intestinal function of broilers. This study was conducted to evaluate the effects of Yupingfeng polysaccharides (YP) supplementation on growth performance and expression of SGLT1, GLUT2 and GLUT5 in Qingyuan partridge chicken. Experiment 1: a total of 540 chickens were randomly allocated to five groups with six replication. Dietary treatments were: (1) CON (control group), basal diet; (2) T1, CON + 0.5 g kg−1 YP; (3) T2, CON + 1 g kg−1 YP; (4) T3, CON + 2 g kg−1YP; (5) T4, CON + 4 g kg−1YP. Experiment 2, a total of 162 were randomly allocated to three groups with three replication. Dietary treatments were: (1) CON, basal diet; (2) T1, CON + 0.5 g kg−1YP; (3) T2, CON + 1 g kg−1YP. From days 1 to 14 and overall, chicken fed T1 diet had higher ADG. On day 42, there was increased villus height of jejunum in T1 group. On days 14 and 28, there was decreased villus height of duodenum and jejunum in T2 group. In duodenum, the expression of SGLT1 (days 21, 35 and 42), GLUT2 (days 7, 14, 21, 28, 35 and 42) and GLUT5 (days 7, 14, 21 and 28) was increased with YP supplementation. In jejunum, the expression of SGLT1 (days 7, 14, 21, 28 and 35), GLUT2 (days 14, 21, 28, 35 and 42) and GLUT5 (days 7, 14, 21, 28, 35 and 42) was increased with YP supplementation. In this article, we consider the space-time continuous Galerkin (STCG) method for the viscoelastic wave equations. It allows variable temporal step-sizes, and the changing of the spatial grids in two adjacent time levels. The existence, uniqueness, and stability of the approximate solutions are demonstrated and the error estimates with global and local spatial mesh sizes in L∞ (L²) norm are derived without any restrictive assumptions on the space-time meshes. If the meshes in each time level satisfy some reasonable assumptions, then we can get the optimal order error estimates both in time and space. ABSTRACT Crosslinked nanocomposite films of sodium alginate (SA) and silicon dioxide (SiO2) with different SiO2 loading values were prepared by in situ synthesis. Biocomposite films were produced by solution casting and solvent evaporation with glycerol as the plasticizer and calcium chloride as the crosslinking agent. The effects of the addition of nano silicon dioxide (nano-SiO2) in SA on the microstructural, physical, mechanical, and optical properties of the nanocomposite films were characterized. The results show that nano-SiO2 was dispersed homogeneously in the SA matrix; it thereby formed a strong interfacial interaction between the nano-SiO2 particles and the matrix. The transparency of the bionanocomposite films was enhanced. Thermogravimetric analysis also revealed that nano-SiO2 improved the thermal stability of the SA films. The incorporation of SiO2 further reduced the water vapor permeability and swelling degree and significantly increased the tensile strength and elongation, which are parameters important for packaging industries. Finally, the lower light transmission of UV light from 200 to 250 nm indicated that SA/SiO2 nanocomposite films could potentially be used to prevent lipid damage by UV light in food conservation. © 2016 Wiley Periodicals, Inc. Chickens are susceptible to the highly pathogenic H5N1 strain of avian influenza virus (HPAIV), whereas ducks are not. Here, we used high-throughput sequencing to analyse the microRNA expression in the spleen, thymus and bursa of Fabricius of H5N1-HPAIV-infected and non-infected chickens and ducks. We annotated the genomic positions of duck microRNAs and we compared the microRNA repertoires of chickens and ducks. Our results showed that the microRNA expression patterns in the homologous immune organs of specific-pathogen-free (SPF) chickens and ducks diverge substantially. Moreover, there was larger divergence between the microRNA expression patterns in immune organs of HPAIV-infected chickens than HPAIV-infected ducks. Lentinula edodes, one of the most cultivated edible fungi in the world, are usually neglected for mycotoxins contamination due to the initial thinking of its resistance to mycotoxingenic molds. In the present study, a sensitive and reliable liquid chromatography with tandem mass spectrometry method was developed for the simultaneous quantification of 33 mycotoxins in L. edodes. Targeted mycotoxins were extracted using a quick, easy, cheap, effective, rugged, and safe procedure without any further clean-up step, and analyzed by liquid chromatography with tandem mass spectrometry on an Agilent Poroshell 120 EC-C18 column (100 × 3 mm, 2.7 μm) with a linear gradient elution program using water containing 5 mM ammonium acetate and methanol as the mobile phase. After validation by determining linearity ( R2 > 0.99), sensitivity (LOQ ≤ 20 ng/kg), recovery (73.6-117.9%), and precision (0.8-19.5%), the established method has been successfully applied to reveal the contamination states of various mycotoxins in L. edodes. Among the 30 tested samples, 22 were contaminated by various mycotoxins with the concentration levels ranging from 3.3-28 850.7 μg/kg, predicting that the edible fungus could be infected by the mycotoxins-producing fungi. Background Monoclonal antibodies generally obtained through the classic mouse hybridoma system were requisite for the establishment of various immunoassays. In this study, a new rabbit monoclonal antibody ( RabMAb) against sulfonamides ( SAs) was first produced via hybridoma technique in rabbit. The related enzyme-linked immunosorbent assay ( ELISA) was then developed and applied to real sample analysis. Results A sensitive competitive indirect ELISA method based on a novel RabMAb for rapid detection of sulfonamides was first established. The obtained half-maximum inhibition concentration ( IC50) values for four SAs were all below 10 ng mL−1, with 0.68 ng mL−1 sulfathiazole ( STZ), 1.11 ng mL−1 sulfadiazine ( SD), 1.15 ng mL−1 sulfapyridine ( SP) and 5.27 ng mL−1 sulfamethoxazole ( SMX). Desirable recoveries when detecting different spiked swine urine and milk samples were achieved ranging from 92.6% to 104.3% and from 61.1% to 81.6%, respectively. MicroRNAs (miRNAs) are small noncoding regulatory RNAs that play key roles in many diverse biological processes such as spermatogenesis. However, no study has been performed on the miRNA transcriptome of developing porcine testes. Here, we employed Solexa deep sequencing technology to extend the repertoire of porcine testis miRNAs and extensively compare the expression patterns of sexually immature and mature porcine testes. Solexa sequencing of two small RNA libraries derived from immature (30 days) and mature (180 days) pig testis samples yielded over 25 million high-quality reads. Overall, the two developmental stages had significantly different small RNA compositions. A custom data analysis pipeline identified 398 known and/or homologous conserved porcine miRNAs, 15 novel pig-specific miRNAs and 56 novel candidate miRNAs. We further observed multiple mature miRNA variants and identified a new bidirectional transcribed miRNA locus, ssc-mir-181a. A total of 122 miRNAs were differentially expressed in the immature and mature testes, and 10 were validated using quantitative RT-PCR. Furthermore, GO and KEGG pathway analyses of the predicted miRNA targets further illustrate the likely roles for these differentially expressed miRNAs in spermatogenesis. This study is the first comparative profile of the miRNA transcriptome in immature and mature porcine testes using a deep sequencing approach, and it provides a useful resource for future studies on the role of miRNAs in spermatogenesis and male infertility treatment. Background: Biallelic variants of the CASQ2 are known to cause the autosomal recessive form of catecholaminergic polymorphic ventricular tachycardia (CPVT), an inherited disease that predisposes young individuals to syncope and sudden cardiac death. To date, only about 24 CASQ2 variants have been reported in association with CPVT pathogenesis; furthermore, studies in Asians, especially in the Chinese population, are relatively rare. The aim of this study was to detect CASQ2 variants in Chinese patients with CPVT. Methods: We used targeted next‐generation sequencing (NGS) to identify CASQ2 variants in Chinese patients with CPVT. A screening process was performed to prioritize rare variants of potential functional significance. Sanger sequencing was conducted to conform the candidate variants and determine the parental origin. Results: We identified seven different CASQ2 variants, of which three (c.1074_1075delinsC, c.1175_1178delACAG, and c.838+1G>A) have not been previously reported. The variants exhibited autosomal recessive inheritance, and were detected in four unrelated Chinese families with CPVT. They included a nonsense variant c.97C>T (p.R33*) and a missense variant c.748C>T (p.R250C) in Family 1 with three CPVT patients; two heterozygous frameshift variants, c.1074_1075delinsC (p.G359Afs*12) and c.1175_1178delACAG (p.D392Vfs*84), in Family 2 with one CPVT patient; one pathogenic homozygous variant c.98G>A (p.R33Q) of CASQ2 in the CPVT patient of Family 3; and two heterozygous splicing variants, (c.532+1G>A) and (c.838+1G>A), in Family 4 with one CPVT patient. Conclusion: To our knowledge, this is the first systematic study of Chinese children with CASQ2 variants. Our work further expands the genetic spectrum of CASQ2‐associated CPVT.
