The article reports on ion-exchange columns used for the improved ancient deoxyribonucleic acid (aDNA) purification for polymerase chain reaction (PCR). Thirteen PCR-resistant ancient bone samples were tested to extract aDNA using a silica-based method and an ion-exchange column method to prove aDNA purification's effectivity on PCR-amplifiable DNA extraction. The results show that further purification of silica-based aDNA extracts using ion-exchange columns improved PCR amplification.
