Ergothioneine (EGT) is a rare thiohistidine derivative with exceptional antioxidant properties. The blood level of EGT is considered highly reliable predictors for cardiovascular diseases and mortality, yet animals lack the ability to synthesize this compound. Free plasmids have been previously used to overexpress genes involved in the EGT biosynthetic pathway of Mycolicibacterium neoaurum. Here, we tentatively introduced a putative transporter gene mfsT1 into high‐copy plasmids and sharply increased the ratio of extracellular EGT concentration from 18.7% to 44.9%. Subsequently, an additional copy of egtABCDE, hisG, and mfsT1 was inserted into the genome with a site‐specific genomic integration tool of M. neoaurum, leading a 2.7 times increase in EGT production. Co‐enhancing the S‐adenosyl‐L‐methionine regeneration pathway, or alternatively, the integration of three copies of egtABCDE, hisG and mfsT1 into the genome further increased the total EGT yield by 16.1% (64.6 mg/L) and 21.7% (67.7 mg/L), respectively. After 168‐h cultivation, the highest titer reached 85.9 mg/L in the latter strain with three inserted copies. Background: Pulsed field ablation (PFA) has emerged as a novel non‐thermal modality with highly myocardium‐specific. However, the PFA catheter based on contact force (CF)‐sensing has not been reported. The study aimed to evaluate the efficacy and safety of a novel CF‐sensing PFA catheter. Methods: First, different CF (5, 15, 25, and 35 g) of the novel PFA catheter were evaluated on lesion dimensions during ablation on right and left ventricle in two pigs. Next, this catheter was further evaluated on four typical sites of superior vena cava (SVC), cavotricuspid isthmus (CTI), right superior pulmonary vein (RSPV), and right inferior pulmonary vein (RIPV) for atrial ablation in another six pigs. Electrical isolation was evaluated immediately after ablation and 30‐day survival. Chronic lesions were assessed via histopathology after euthanasia. Acute and chronic safety outcomes were observed peri‐ and post‐procedurally. Results: In ventricular ablation, increased CF from 5 to 15 g produced significantly greater lesion depth but nonsignificant increases from 15 to 35 g. In atrial ablation, the novel CF‐sensing PFA deliveries produced an acute attenuation of local electrograms and formation of a continuous line of block in all 6 pigs. The ablation line remained sustained blockage at the 30‐day survival period. The CF of SVC, CTI, RSPV, and RIPV was 9.4 ± 1.5, 14.5 ± 3.2, 17.2 ± 2.6, and 13.4 ± 2.8 g, respectively. Moreover, no evidence of damage to esophagus or phrenic nerve was observed. Idiopathic pulmonary fibrosis (IPF) is defined as a specific form of chronic, progressive fibrosing interstitial pneumonia. It is unknown why fibrosis in IPF distributes in the peripheral or named sub‐pleural area. Migration of pleural mesothelial cells (PMC) should contribute to sub‐pleural fibrosis. Calpain is known to be involved in cell migration, but the role of calpain in PMC migration has not been investigated. In this study, we found that PMCs migrated into lung parenchyma in patients with IPF. Then using Wt1tm1(EGFP/Cre)Wtp/J knock‐in mice, we observed PMC migration into lung parenchyma in bleomycin‐induced pleural fibrosis models, and calpain inhibitor attenuated pulmonary fibrosis with prevention of PMC migration. In vitro studies revealed that bleomycin and transforming growth factor‐β1 increased calpain activity in PMCs, and activated calpain‐mediated focal adhesion (FA) turnover as well as cell migration, cell proliferation, and collagen‐I synthesis. Furthermore, we determined that calpain cleaved FA kinase in both C‐terminal and N‐terminal regions, which mediated FA turnover. Lastly, the data revealed that activated calpain was also involved in phosphorylation of cofilin‐1, and p‐cofilin‐1 induced PMC migration. Main observation and conclusion: Anisotanols A—D (1—4), four new compounds possessing an unprecedented sesquiterpenoid skeleton with a congested tricyclic 6/3/5 ring system, were obtained from Anisodus tanguticus. Their structures were elucidated by comprehensive spectroscopic techniques, and the absolute configurations were confirmed via ECD calculations and single‐crystal X‐ray diffractions. A putative biosynthetic pathway for these compounds was proposed. Dual mobility (DM) total hip arthroplasty (THA) is associated with reduced dislocation rates; however, the kinematic mechanism of dislocation in DM THA constructs is still not well understood. This study hypothesizes that the difference in kinematics between DM THA and conventional THA designs contributes to reduced dislocation rates of DM THA. In addition, this study aims to quantify and compare those kinematic parameters between DM THA and conventional THA using a validated dual fluoroscopy imaging system (DFIS) and finite element (FE) modelling. Fresh frozen cadavers were measured to compare the impingement‐free range of motion and provocative subluxation kinematics among three THA constructs: (1) DM, (2) constrained liner (CS), and (3) 36 mm head diameter neutral liner (NL). The DFIS was used to measure the in vitro kinematics of the hip. Subject‐specific FE models were developed to assess the horizontal dislocation distance and resistive torque at dislocation. The DM construct head exhibited increased provocative anterior and posterior subluxation range of motion before dislocation when compared to CS constructs (p =.05; p =.03), as well as NL constructs (p =.05). The DM THA showed a significantly larger posterior horizontal dislocation distance, as well as smaller resistive torque at dislocation, when compared to NL (p =.05; p =.03) and CS constructs (p =.04; p =.01). Our findings demonstrate there was increased provocative subluxation range of motion as well as normalized jump distance for the DM constructs compared to the NL and CS constructs, suggesting the DM THA may provide increased stability hip during at‐risk functional hip positions. SET domain bifurcated protein 1 (SETDB1) is a histone lysine methyltransferase that promotes the silencing of some tumour suppressor genes and is overexpressed in many cancers. SETDB1 contains a unique tandem tudor domain (TTD) that recognizes histone H3 sequences containing both methylated and acetylated lysines. Beginning with the identification of a hit compound (Cpd1), we discovered the first potent and selective small molecule SETDB1‐TTD inhibitor (R,R)‐59 through stepwise structure‐guided optimization. (R,R)‐59 showed a KD value of 0.088±0.045 μM in the ITC assay. The high potency of (R,R)‐59 was well explained by the cocrystal structure of the (R,R)‐59‐TTD complex. (R,R)‐59 is an endogenous binder competitive inhibitor. Evidence has also demonstrated its cellular target engagement. Interestingly, the enantiomer (S,S)‐59 did not show activity in all the assays, highlighting the potential of (R,R)‐59 as a tool compound in exploring the biological functions of SETDB1‐TTD. Background: Rheumatoid arthritis‐associated interstitial lung disease (RA‐ILD) is an irreversible pathologic condition of unknown cause, commonly involving the joint and the lung with variable amounts of fibrotic change. In contrast to rheumatoid arthritis or other chronic interstitial lung diseases such as interstitial pulmonary fibrosis, there is so far no extensively accepted or implemented animal model for this disease. Aims: To provide guidance for those who are investigating the pathogenesis of RA‐ILD with animal models. Materials and Methods: An analysis of papers from PubMed during 1978‐2020. Results: We outline the present status quo for animal models of RA‐ILD about their modeling methods and pathogenesis, compare their pros and cons with respect to their ability to mimic the clinical and histological features of human disease and discuss their applicability for future research. Discussion: There is no doubt that these animal models do provide valuable information relating to the pathogenesis of RA‐ILD and the development of effective therapeutic drugs. Nevertheless, these animal models can not entirely recapitulate clinical pathology and have some limitations in experimental research application. Therefore, it should be emphasized that we should improve and explore animal models in more accordance with the pathogenesis and clinical characteristics of human RA‐ILD. Conclusion: These established animal models of the disease can significantly progress our understanding of the etiology of RA‐ILD, the fundamental mechanisms of its pathogenesis and the identification of new bio‐markers, and can contribute to the development and implementation of novel treatment strategies. It is more convenient and safer to employ wireless power transfer (WPT) systems to charge the battery pack of electric scooters (ESs) than conventional plug‐in systems. A charging equaliser suitable for ESs is proposed to achieve constant current (CC) output without communication link between the transmitter side and the receiver side. The full‐bridge rectifier and select switches utilised at the receiver side are employed to be operated once to change from the balancing mode to charging modes. The characteristics of the load‐independent current output in the CC mode are achieved by properly selecting the parameters of the circuit so that no sophisticated control strategies are required to regulate the output as per the charging profile. The experimental results show that when the voltage of battery pack is charged at 32 or 4.2 V, the current is 1.02 or 1.12 A, respectively. The traditional pure switched‐capacitor equaliser brings the large inrush current and low energy density. This study proposes a series of resonant switched‐capacitor (ReSC) voltage equaliser, which realises energy transferred directly from source cells to target cells by the series ReSC converter. The ReSC converter eliminates the inrush current and improves the capacitors' energy density to increase the balancing speed. Meanwhile, all the switches are controlled by a pair of complementary PWM signals at a fixed operational frequency without voltage monitors. Both simulation and experiment are used to verify the system feasibility and theoretical analysis of the proposed circuit. In the same experimental condition, the proposed series resonant circuit reduces the inrush current. Previous studies shown that myeloperoxidase (MPO) level is higher in patients with atrial fibrillation (AF); however, no genetic evidence between MPO and AF risk in human population was observed. Therefore, the present study was aimed to investigate the association between rs2243828, a variant in promoter region of MPO and the risk of AF in Chinese GeneID population. The results demonstrated that the minor G allele of rs2243828 showed a significant association with AF in two independent population (GeneID‐north population with 694 AF cases and 710 controls, adjusted P‐adj = 6.25 × 10−3 with an odds ratio was 0.77, GeneID‐central population with 1106 cases and 1501 controls, P‐adj = 9.88 × 10−5 with an odds ratio was 0.75). The results also showed G allele was significantly associated with lower plasma concentration of myeloperoxidase in general population. We also observed a significant difference of odds ratio between subgroups of hypertension and non‐hypertension. Therefore, our findings identified variant in MPO associated with risk of AF and it may give strong evidence to link the inflammation with the incidence of AF. [ABSTRACT FROM AUTHOR]

Background: Anhydramnios results from the poor development of the placenta or problems with intrauterine development of the kidneys or urinary tract. Complete lack of amniotic fluid indicates a severe problem with the organs of the urinary system. The genes associated with anhydramnios show very diversity and are not yet well defined. Methods: Whole‐exome sequencing (WES) was used for an aborted male case around the 20th week of gestation diagnosed with anhydramnios. The resulted deleterious variants were verified by Sanger sequencing. Pathogenicity of deleterious variants was explored by in silico analysis. Results: A maternally inherited deleterious frameshift variant, c.1454_1455insC, p.(S486Ffs29) in exon 9 and two paternally inherited missense variants c.1037C > G, p.(Ser346Trp) in exon 7 and c.1465A > G, p.(Asn489Asp) in exon 9 of Angiotensin‐I‐Converting Enzyme (ACE) gene were found and confirmed by Sanger sequencing. c.1454_1455insC, p.(S486Ffs29) and c.1037C > G, p.(Ser346Trp) were identified as two novel compound heterozygous deleterious variants. The pathogenicity of these deleterious variants was determined by in silico analysis and both the deleterious variants disrupt the structure of the ACE protein. Conclusion: Two novel compound heterozygous variants were identified in the case with anhydramnios, which may be associated with pathogenicity of anhydramnios. Background and Aims: Heat shock factor (HSF4) plays a vital role in carcinogenesis and tumour progression. However, its clinical significance implications in hepatocellular carcinoma (HCC) remained elusive. Methods: RT‐PCR and western blot were used to detect the HSF4 expression levels in HCC cells and tissues. Immunohistochemistry staining was performed on a tissue microarray containing 104 HCC patients received radical resection. In vitro effects of HSF4 on proliferation, migration and invasion were determined by colony formation and transwell assays in HCCLM3, Huh7, MHCC97L and SMMC7721 cells. Epithelial‐mesenchymal transition (EMT) was identified by RT‐PCR, WB and immunofluorescence in HCCLM3 and MHCC97L cells. AKT pathway activation was detected by WB and dual luciferase report system in HCCLM3 and MHCC97L cells. Results: HSF4 expression was higher in primary HCC tissues derived from recurrent patients, and positively correlated with invasiveness potentials of cell lines. Clinically, patients with high HSF4 expression had significant poorer prognosis. In vitro experiments showed HSF4 silencing inhibited HCC cell proliferation, migration and invasion, whereas HSF4 overexpression had inverse effects. Moreover, silence of HSF4 induced an epithelial‐like phenotype, whereas the overexpression of HSF4 resulted in a mesenchymal‐like phenotype in HCC by activating AKT pathway. Further experiments showed that HSF4 could activate AKT pathway in a hypoxia‐inducible factor‐1α (HIF‐1α) dependent, but transforming growth factor‐β (TGF‐β) independent manner. Conclusions: HSF4 is upregulated in HCC, resulting in greater proliferation, migration and invasion capacities. Moreover, high HSF4 expression is a promising predictive indicator of poor outcome after radical resection. Intramedullary stabilization is frequently used to treat long bone fractures. Since implant removal can become technically very challenging with the potential to cause further tissue damage, biodegradable materials are emerging as alternative options. Magnesium (Mg)‐based biodegradable implants have a controllable degradation rate and good tissue compatibility, which makes them attractive for musculoskeletal research. Herein, the degradation of Mg and steel implants, the pathological characteristics and osteoblast differentiation in mice femora were examined. To investigate the molecular mechanism, we analyzed the differentially expressed long noncoding RNAs (lncRNAs) and messenger RNAs (mRNAs) in Mg‐implanted or stain‐steel‐implanted callus tissues. lncRNA LOC103691336 was upregulated in Mg‐implanted tissues and most relevant to BMPR2, a kinase receptor of BMPs with an established role in osteogenesis. The knockdown of LOC103691336 attenuated Mg‐mediated osteogenic differentiation. Furthermore, miR‐138‐5p, previously reported to inhibit osteogenic differentiation, could bind to LOC103691336 and BMPR2 in bone marrow stromal cells (BMSCs). LOC103691336 competed with BMPR2 for miR‐138‐5p binding in BMSCs to attenuate the inhibitory effect of miR‐138‐5p on BMPR2 expression. Finally, the effect of LOC103691336 knockdown on Mg‐mediated osteogenic differentiation could be attenuated by miR‐138‐5p inhibition. In conclusion, we provided a novel mechanism of Mg implants mediating the osteogenesis differentiation and demonstrated that Mg implants may be promising for improving fracture healing. New Findings: What is the central question of this study? Can targeted ablation of cardiac sympathetic neurons suppress myocardial infarction‐induced adverse cardiac remodelling and left ventricular dysfunction? What is the main finding and its importance? Targeted ablation of cardiac sympathetic neurons significantly alleviated sympathetic remodelling and neuroendocrine activation, attenuated cardiac hypertrophy and fibrosis and improved left ventricular function. Thus, targeted ablation of cardiac sympathetic neurons might have a beneficial effect on adverse postinfarction remodelling and left ventricular dysfunction. Abstract: Sympathetic overactivation is crucial in the development and progression of adverse cardiac remodelling and dysfunction. Targeted ablation of cardiac sympathetic neurons (TACSN) is an effective approach to inhibit overactivation of the sympathetic nervous system. The aim of this study was to investigate whether TACSN could suppress myocardial infarction (MI)‐induced adverse cardiac remodelling and dysfunction, thereby producing protective effects. Thirty‐eight dogs were randomly assigned into the sham‐operated, MI or MI‐TACSN group. The TACSN was induced by injecting cholera toxin B subunit–saporin compound into the stellate ganglia 1 week after MI. Five weeks after MI surgery, echocardiographic and haemodynamic parameters of cardiac function were significantly improved in the TACSN group compared with the MI group. In addition, TACSN attenuated the extent of cardiac hypertrophy and fibrosis and suppressed the increase in the plasma concentrations of noradrenaline, nerve growth factor, atrial natriuretic peptide, brain natriuretic peptide, angiotensin II and aldosterone. Furthermore, TACSN alleviated the growth associated protein‐43‐positive and tyrosine hydroxylase‐positive nerve densities in the infarcted border zone and restored protein expression of the β1‐adrenergic receptor in the left ventricular myocardium. These findings indicate that TACSN might have a beneficial effect on adverse postinfarction remodelling and left ventricular dysfunction, which might be attributable, at least in part, to the attenuation of both sympathetic remodelling and the cardiac neuroendocrine system. Abstract: Introduction: Bulbus fritillariae cirrhosae (known as Chuān bèi mǔ in China, BFC) contain fritillaria steroidal alkaloids as the bioactive ingredients and are widely used as traditional Chinese medicine for the treatment of cough and phlegm. Due to limited wild resources, the cultivated species are becoming predominantly used in Chinese traditional medicine markets. Objective: To assess the impact of different growth stages on the alkaloids of cultivated BFC and establish a reference for quality control and guidance for appropriate harvesting practices. Methods: The ultra‐high performance liquid chromatography quadrupole time‐of‐flight mass spectrometry (UHPLC‐QTOF‐MS) metabolomic strategy was applied to determine potential chemical markers for the discrimination and quality control of cultivated BFC in different growth stages. The molecular feature extraction and multivariate statistical analysis were applied to alkaloid extraction and full metabolomic profiling of cultivated BFC for classification and marker compound characterisation. Result: This approach allowed the establishment of a fast and efficient comparative multivariate analysis of the metabolite composition of 42 samples covering growth of cultivated BFC ranging in age from one to seven years old. Four alkaloid compounds were identified in cultivated BFC based on accurate mass, retention time, and MS/MS fragments. These compounds may be used as potential chemical markers for the classification and discrimination of cultivated BFC samples indifferent growth stages. Conclusions: The proposed analytical method in combination with multivariate statistical analysis comprised a useful and powerful strategy to explore the chemical ingredients and transforming mechanisms of cultivated BFC and for quality evaluation and control. Perovskite solar cells (PSCs) may offer huge potential in photovoltaic conversion, yet their practical applications face one major obstacle: their low stability, or quick degradation of their initial efficiencies. Here, a new design scheme is presented to enhance the PSC stability by using low-temperature hydrothermally grown hierarchical nano-SnO2 electron transport layers (ETLs). The ETL contains a thin compact SnO2 layer underneath a mesoporous layer of SnO2 nanosheets. The mesoporous layer plays multiple roles of enhancing photon collection, preventing moisture penetration and improving the long-term stability. Through such simple approaches, PSCs with power conversion efficiencies of ≈13% can be readily obtained, with the highest efficiency to be 16.17%. A prototypical PSC preserves 90% of its initial efficiency even after storage in air at room temperature for 130 d without encapsulation. To achieve high performance for inverted structure perovskite solar cells, the design of hole-transporting layer (HTL) and related interfacial engineering are very important tasks. To avoid the hygroscopic characteristics of poly (3, 4-ethylenedioxythiophene):poly (styrenesulfonate) that may degrade the adjacent moisture-sensitive perovskite layer, here, a new CrO x-based hole-transport material has been introduced. The feasibility of fabrication efficient perovskite solar cells with CrO x and Cu-CrO x as HTLs is confirmed for the first time. Cu doping can modify the chromium ion contents and suppress the formation of surface hydroxylation and CrO3 in the CrO x film, which can increase work function, electrical conductivity, and carrier mobility of the CrO x films. Consequently, the power conversion efficiency of the corresponding device increases to 10.99% from its original value of 9.27%. This study not only provides a novel HTL system for high performance and decently stable optoelectronic devices but also reveals the importance of HTL doping for interface engineering. A specific and sensitive HPLC-MS/MS method was developed and validated for the simultaneously quantification of isoliquiritigenin (ISL) and neoisoliquiritin (NIS) in rat plasma by oral administration. Analytes were analyzed on an Agilent 6460 LC-MS/MS system (Agilent, USA) using an Agilent Zorbax SB-C18 column (4.6 × 150 mm, 5 μm). Gradient elution was applied for the analyte separation using a mobile phase composed of 0.1% formic acid aqueous solution and methanol at a flow rate of 1.0 mL/min with a total running time of 12 min. The calibration curves for ISL and NIS showed good linearity in the concentrations ranging from 0.001 to 4.000 μg/mL with correlation coefficients >0.998. The precision, accuracy, recovery and stability were deemed acceptable. The method was applied to the pharmacokinetics study of ISL and NIS in rats by single and combination administration. The result showed that Cmax and AUC0→ t of ISL were markedly increased from 0.53 to 1.20 μg/mL, and from 69.63 to 200.74 min μg/mL by combination administration. The mean t1/2 value was also prolonged from 64.55 to 203.74 min in the combination group. These results indicated that NIS may have been metabolized to ISL which increased the absorption and extended the elimination of ISL. However, little difference was found for NIS pharmacokinetics parameters between single NIS and the combination group, which suggested that there was no significant biotransformation of ISL to NIS. Novel waterborne polyurethane (WPU) composites based on NiAl-LDH and ZnO were successfully synthesized by in-situ polymerization. The nanostructured NiAl-LDH/ZnO was first grafted by isophorone diisocyanate (IPDI), forming the NiAl-LDH/ZnO-NCO complex with -NCO group on the surface, which promotes its homogeneous dispersion in WPU matrix. The microstructure and particle distribution of the composites were characterized. The mechanical properties and the water resistance of the composites were improved. ABSTRACT A specific and sensitive UHPLC-qTOF-MS method was developed and validated for quantification of fuziline in rat plasma after oral administration of three dosages. The analyte was separated on an Acquity UPLC BEH C18 column with a total running time of 3 min using a mobile phase of 0.1% formic acid aqueous solution and methanol (80:20, v/v) at a flow-rate of 0.25 mL/min. The calibration curves for fuziline showed good linearity in the concentrations ranging from 1 to 200 ng/mL with correlation coefficients >0.997. The precision, accuracy, recovery and stability were deemed acceptable. The method was applied to a pharmacokinetics study of fuziline in rats. The mean half-life was 5.93, 6.13 and 5.12 h for 1, 2 and 4 mg/kg oral administration of fuziline, respectively. The peak concentration and area under the concentration-time curve increased linearly with the doses. Main observation and conclusion: Anisotanols A—D (1—4), four new compounds possessing an unprecedented sesquiterpenoid skeleton with a congested tricyclic 6/3/5 ring system, were obtained from Anisodus tanguticus. Their structures were elucidated by comprehensive spectroscopic techniques, and the absolute configurations were confirmed via ECD calculations and single‐crystal X‐ray diffractions. A putative biosynthetic pathway for these compounds was proposed.
