Long‐term ecological research can resolve effects of disturbance on ecosystem dynamics by capturing the scale of disturbance and interactions with environmental changes. To quantify how disturbances interact with long‐term directional changes (sea‐level rise, freshwater restoration), we studied 17 yr of monthly dissolved organic carbon (DOC), total nitrogen (TN), and phosphorus (TP) concentrations and bacterioplankton productivity across freshwater‐to‐marine estuary gradients exposed to multiple disturbance events (e.g., droughts, fire, hurricanes, and low‐temperature anomalies) and long‐term increases in water levels. By studying two neighboring drainages that differ in hydrologic connectivity, we additionally tested how disturbance legacies are shaped by hydrologic connectivity. We predicted that disturbance events would interact with long‐term increases in water levels in freshwater and marine ecosystems to increase spatiotemporal similarity (i.e., synchrony) of organic matter, nutrients, and microbial activities. Wetlands along the larger, deeper, and tidally influenced Shark River Slough (SRS) drainage had higher and more variable DOC, TN, and TP concentrations than wetlands along the smaller, shallower, tidally restricted Taylor River Slough/Panhandle (TS/Ph) drainage. Along SRS, DOC concentrations declined with proximity to coast, and increased in magnitude and variability following drought and flooding in 2015 and a hurricane in 2017. Along TS/Ph, DOC concentrations varied by site (higher in marine than freshwater wetlands) but not year. In both drainages, increases in TN from upstream freshwater marshes occurred following fire in 2008 and droughts in 2010 and 2015, whereas downstream increases in TP occurred with coastal storm surge from hurricanes in 2005 and 2017. Decreases in DOC:TN and DOC:TP were explained by increased TN and TP. Increases in bacterioplankton productivity occurred throughout both drainages following low‐temperature events (2010 and 2011) and a hurricane (2017). Long‐term TN and TP concentrations and bacterioplankton productivity were correlated (r > 0.5) across a range of sampling distances (1–50 km), indicating spatiotemporal synchrony. DOC concentrations were not synchronized across space or time. Our study advances disturbance ecology theory by illustrating how disturbance events interact with long‐term environmental changes and hydrologic connectivity to determine the magnitude and extent of disturbance legacies. Intestinal tracts are among the most densely populated microbial ecosystems. Gut microbiota and their influence on the host have been well characterized in terrestrial vertebrates but much less so in fish. This is especially true for coral reef fishes, which are among the most abundant groups of vertebrates on earth. Surgeonfishes (family: Acanthuridae) are part of a large and diverse family of reef fish that display a wide range of feeding behaviours, which in turn has a strong impact on the reef ecology. Here, we studied the composition of the gut microbiota of nine surgeonfish and three nonsurgeonfish species from the Red Sea. High-throughput pyrosequencing results showed that members of the phylum Firmicutes, especially of the genus Epulopiscium, were dominant in the gut microbiota of seven surgeonfishes. Even so, there were large inter- and intraspecies differences in the diversity of surgeonfish microbiota. Replicates of the same host species shared only a small number of operational taxonomic units ( OTUs), although these accounted for most of the sequences. There was a statistically significant correlation between the phylogeny of the host and their gut microbiota, but the two were not completely congruent. Notably, the gut microbiota of three nonsurgeonfish species clustered with some surgeonfish species. The microbiota of the macro- and microalgavores was distinct, while the microbiota of the others (carnivores, omnivores and detritivores) seemed to be transient and dynamic. Despite some anomalies, both host phylogeny and diet were important drivers for the intestinal microbial community structure of surgeonfishes from the Red Sea. [ABSTRACT FROM AUTHOR]

The Red Sea is a unique marine ecosystem with contrasting gradients of temperature and salinity along its north-to-south axis. It is an extremely oligotrophic environment that is characterized by perpetual year-round water column stratification, high annual solar irradiation, and negligible riverine and precipitation inputs. In this study, we investigated whether the contemporary environmental conditions shape community assemblages by pyrosequencing 16S rRNA genes of bacteria in surface water samples collected from the northeastern half of this water body. A combined total of 1855 operational taxonomic units (OTUs) were recovered from the 'small-cell' and 'large-cell' fractions. Here, a few major OTUs affiliated with Cyanobacteria and Proteobacteria accounted for ∼93% of all sequences, whereas a tail of 'rare' OTUs represented most of the diversity. OTUs allied to Surface 1a/b SAR11 clades and Prochlorococcus related to the high-light-adapted (HL2) ecotype were the most widespread and predominant sequence types. Interestingly, the frequency of taxa that are typically found in the upper mesopelagic zone was significantly elevated in the northern transects compared with those in the central, presumably as a direct effect of deep convective mixing in the Gulf of Aqaba and water exchange with the northern Red Sea. Although temperature was the best predictor of species richness across all major lineages, both spatial and environmental distances correlated strongly with phylogenetic distances. Our results suggest that the bacterial diversity of the Red Sea is as high as in other tropical seas and provide evidence for fundamental differences in the biogeography of pelagic communities between the northern and central regions. Six strains of novel bacteria were isolated from profundal sediment of Lake Constance, a deep freshwater lake in Germany, by direct dilution of the sediment in mineral agar medium containing a background lawn of the hydrogen-scavenging Methanospirillum hungatei as a syntrophic partner. The numbers of colony-forming units obtained after incubation for more than 2 months were in the same range as those of total bacterial counts determined by DAPI staining (up to 108 cells per millilitre) suggesting that these organisms were dominant members of the community. Identical dilution series in the absence of methanogenic partners yielded numbers that were lower by two to three orders of magnitude. The dominant bacteria were isolated in defined co-culture with M. hungatei, and were further characterized. Growth was slow, with doubling times of 22–28 h at 28°C. Cells were small, 0.5 × 5 μm in size, Gram-positive, and formed terminal oval spores. At 20°C, glucose was fermented by the co-culture strain BoGlc83 nearly stoichiometrically to 2 mol of acetate and 1 mol of methane plus CO2. At higher temperatures, also lactate and traces of succinate were formed. Anaerobic growth depended strictly on the presence of a hydrogen-scavenging partner organism and was inhibited by bromoethane sulfonate, which together indicate the need for a syntrophic partnership for this process. Strain BoGlc83 grew also aerobically in the absence of a partner organism. All enzymes involved in ATP formation via glycolysis and acetyl CoA were found, most of them at activities equivalent to the physiological substrate turnover rate. This new type of sugar-fermenting bacterium appears be the predominant sugar utilizer in this environment. Members of the phylum Planctomycetes are found in aquatic and terrestrial habitats. Here we show that the highest density of Planctomycetes in natural environments (2.6 × 109 cells ml−1) is encountered in the hindgut of soil-feeding termites ( Cubitermes spp.), where they constitute up to one-third of the bacteria in the alkaline P3 compartment detected by fluorescent in situ hybridization (FISH). A 16S-rRNA-based approach revealed that the planctomycete community is very diverse and falls into three major clusters representing novel, deeply branching lineages. Terminal restriction fragment length polymorphism (T-RFLP) analysis and FISH with cluster-specific oligonucleotide probes confirmed that most of the lineages are also present in other gut compartments, albeit in much lower numbers, but absent from the food soil. The majority of planctomycetes in the gut belong to a large clade, the ‘Termite planctomycete cluster’, which consists exclusively of clones from termite guts and seems to be represented in all termite species. Differences in microenvironment and interactions of microorganisms within and across habitat boundaries should influence structure and diversity of the microbial communities within an ecosystem. We tested this hypothesis using the well characterized gut tract of the European subterranean termiteReticulitermes santonensisas a model. By cloning and sequencing analysis and molecular fingerprinting (terminal restriction fragment length polymorphism), we characterized the bacterial microbiota in the major intestinal habitats– the midgut, the wall of the hindgut paunch, the hindgut fluid and the intestinal protozoa. The bacterial community was very diverse (> 200 ribotypes) and comprised representatives of several phyla, includingFirmicutes(mainly clostridia, streptococci andMycoplasmatales-related clones),Bacteroidetes,Spirochaetesand a number ofProteobacteria, all of which were unevenly distributed among the four habitats. The largest group of clones fell into the so-called Termite group 1 (TG-1) phylum, which has no cultivated representatives. The majority of the TG-1 clones were associated with the protozoa and formed two phylogenetically distinct clusters, which consisted exclusively of clones previously retrieved from the gut of this and otherReticulitermesspecies. Also the other clones represented lineages of microorganisms that were exclusively recovered from the intestinal tract of termites. The termite specificity of these lineages was underscored by the finding that the closest relatives of the bacterial clones obtained fromR. santonensiswere usually derived also from the most closely related termites. Overall, differences in diversity between the different gut habitats and the uneven distribution of individual phylotypes support conclusively that niche heterogeneity is a strong determinant of the structure and spatial organization of the microbial community in the termite gut.
