Giardia is a zoonotic gastrointestinal parasite responsible for a substantial global public health burden, and quantitative microbial risk assessment (QMRA) is often used to forecast and manage this burden. QMRA requires dose–response models to extrapolate available dose–response data, but the existing model for Giardia ignores valuable dose–response information, particularly data from several well‐documented waterborne outbreaks of giardiasis. The current study updates Giardia dose–response modeling by synthesizing all available data from outbreaks and experimental studies using a Bayesian random effects dose–response model. For outbreaks, mean doses (D) and the degree of spatial and temporal aggregation among cysts were estimated using exposure assessment implemented via two‐dimensional Monte Carlo simulation, while potential overreporting of outbreak cases was handled using published overreporting factors and censored binomial regression. Parameter estimation was by Markov chain Monte Carlo simulation and indicated that a typical exponential dose–response parameter for Giardia is r = 1.6 × 10−2 [3.7 × 10−3, 6.2 × 10−2] (posterior median [95% credible interval]), while a typical morbidity ratio is m = 3.8 × 10−1 [2.3 × 10−1, 5.5 × 10−1]. Corresponding (logistic‐scale) variance components were σr = 5.2 × 10−1 [1.1 × 10−1, 9.6 × 10−1] and σm = 9.3 × 10−1 [7.0 × 10−2, 2.8 × 100], indicating substantial variation in the Giardia dose–response relationship. Compared to the existing Giardia dose–response model, the current study provides more representative estimation of uncertainty in r and novel quantification of its natural variability. Several options for incorporating variability in r (and m) into QMRA predictions are discussed, including incorporation via Monte Carlo simulation as well as evaluation of the current study's model using the approximate beta‐Poisson. Pd0‐catalyzed Mizoroki–Heck reactions traditionally exhibit poor reactivity with polysubstituted, unbiased alkenes. Intermolecular reactions with simple, all‐carbon tetrasubstituted alkenes are unprecedented. Herein we report that pendant carboxylic acids, combined with bulky monophospine ligands on palladium, can direct the arylation of tri‐ and tetrasubstituted olefins. Quaternary carbons are established at high Fsp3 attached‐ring junctures and the carboxylate directing group can be removed after coupling. The case report by Agass and Fraser in this issue ([1]) describes the diagnosis and successful surgical treatment of an osteochondroma in a 3-year-old Polo pony mare. An osteochondroma is derived from a portion of aberrant cartilaginous tissue which proliferates and separates from the edge of an epiphyseal growth plate during bone growth. This tissue separation is thought to occur secondary to osteoclastic activity in the periosteum of the metaphysis, which serves to reduce the outer diameter of the metaphysis and remodel it into the narrower diaphysis as the bone elongates (Jerome I et al i . Histopathological findings in the case reported by Agass and Fraser ([1]) were typical of an osteochondroma in that the tissue was covered in a smooth layer of cartilage, with evidence of endochondral ossification. Treatment of the reported fracture by internal fixation is optimal to provide rigid stabilisation, interfragmentary compression and to minimise the risk of fracture propagation and secondary tarsometatarsal osteoarthritis. Fracture configuration, temperament of the animal, a surgeon experienced in fracture repair and a competent surgical team are all vital components of successful standing fracture repair. Summary: A yearling Thoroughbred colt was presented for investigation of neck stiffness and episodes of intermittent neck pain without neurological signs. Osteochondrosis (OCD) of the cervical articular process joints (APJs) was diagnosed with the aid of radiography and computed tomography. An articular osteochondral fragment of the left fourth caudal cervical articular process was removed arthroscopically following a cut down approach to the joint capsule of the affected APJ. Surgical removal resulted in resolution of clinical signs at 4 weeks. However, subsequently the horse was markedly ataxic 6 weeks post surgery after being found cast in its stable. Cervical stenotic myelopathy was considered the most likely cause based on clinical and radiographic signs and the horse was subjected to euthanasia due to a poor prognosis for racing. Post‐mortem examination identified atypical cartilage within several cervical APJs with histological changes consistent with OCD. This case report supports OCD of the APJs as a cause of neck pain and confirms the clinical practicality and short‐term effectiveness of arthroscopic fragment removal. Surgical treatment for cervical OCD should be considered in horses without neurological signs, although case selection is important and the underlying pathology remains a risk factor for the development of subsequent neurological signs. Anaerobic digestion can inactivate zoonotic pathogens present in cattle manure, which reduces transmission of these pathogens from farms to humans through the environment. However, the variability of inactivation across farms and over time is unknown because most studies have examined pathogen inactivation under ideal laboratory conditions or have focused on only one or two full-scale digesters at a time. In contrast, we sampled seven full-scale digesters treating cattle manure in Wisconsin for 9 mo on a biweekly basis (n = 118 pairs of influent and effluent samples) and used real-time quantitative polymerase chain reaction to analyze these samples for 19 different microbial genetic markers. Overall, inactivation of pathogens and fecal indicators was highly variable. When aggregated across digester and season, log-removal values for several representative microorganisms--bovine Bacteroides, Bacteroidales-like CowM3, and bovine polyomavirus--were 0.78 ± 0.34, 0.70 ± 0.50, and 0.53 ± 0.58, respectively (mean ± SD). These log-removal values were up to two times lower than expected based on the scientific literature. Thus, our study indicates that full-scale anaerobic digestion of cattle manure requires optimization with regard to pathogen inactivation. Future studies should focus on identifying the potential causes of this suboptimal performance (e.g., overloading, poor mixing, poor temperature control). Our study also examined the fate of pathogens during manure separation and found that the majority of microbes we detected ended up in the liquid fraction of separated manure. Either central (brain) or peripheral (body physiological system) control mechanisms, or a combination of these, have been championed in the last few decades in the field of Exercise Sciences as how physiological activity and fatigue processes are regulated. In this review, we suggest that the concept of ‘central’ or ‘peripheral’ mechanisms are both artificial constructs that have ‘straight-jacketed’ research in the field, and rather that competition between psychological and physiological homeostatic drives is central to the regulation of both, and that governing principles, rather than distinct physical processes, underpin all physical system and exercise regulation. As part of the Integrative Governor theory we develop in this review, we suggest that both psychological and physiological drives and requirements are underpinned by homeostatic principles, and that regulation of the relative activity of each is by dynamic negative feedback activity, as the fundamental general operational controller. Because of this competitive, dynamic interplay, we propose that the activity in all systems will oscillate, that these oscillations create information, and comparison of this oscillatory information with either prior information, current activity, or activity templates create efferent responses that change the activity in the different systems in a similarly dynamic manner. Changes in a particular system are always the result of perturbations occurring outside the system itself, the behavioural causative ‘history’ of this external activity will be evident in the pattern of the oscillations, and awareness of change occurs as a result of unexpected rather than planned change in physiological activity or psychological state. This study investigated performance and physiology to understand pacing strategies in elite Paralympic athletes with cerebral palsy ( CP). Six Paralympic athletes with CP and 13 able-bodied ( AB) athletes performed two trials of eight sets of 10 shuttles (total 1600m). One trial was distance-deceived ( DEC, 1000 m + 600 m) one trial was nondeceived ( N- DEC, 1600 m). Time (s), heart rate ( HR, bpm), ratings of perceived exertion ( RPE, units), and electromyography of five bilateral muscles ( EMG) were recorded for each set of both trials. The CP group ran slower than the AB group, and pacing differences were seen in the CP DEC trial, presenting as a flat pacing profile over the trial ( P < 0.05). HR was higher and RPE was lower in the CP group in both trials ( P < 0.05). EMG showed small differences between groups, sides, and trials. The present study provides evidence for a possible pacing strategy underlying exercise performance and fatigue in CP. The results of this study show (1) underperformance of the CP group, and (2) altered pacing strategy utilization in the CP group. We proposed that even at high levels of performance, the residual effects of CP may negatively affect performance through selection of conservative pacing strategies during exercise. Reasons for performing study Knowledge of imaging anatomy, surgical anatomy and disorders affecting the sphenopalatine sinus are currently lacking. Objectives To describe the computed tomographic ( CT) and surgical anatomy of the sphenopalatine sinus and diagnosis, treatment and outcome in clinical cases with sphenopalatine sinus disease. Study design Cadaver observational study and retrospective case series. Methods The sphenopalatine sinuses of 10 normal cadaver heads were examined with digital radiography, CT and sinoscopic examination prior to anatomical sectioning. Sphenopalatine sinus anatomy was described and compared between cadaver specimens across the imaging modalities. Medical records (January 2004-2014) of cases diagnosed with sphenopalatine sinus disease were reviewed. Results The anatomy of the sphenopalatine sinus was variable. The borders of the sphenopalatine sinus were not identifiable on plain radiographs, whereas CT provided useful anatomical information. The palatine portion of the sphenopalatine sinus was consistently accessible sinoscopically and the sphenoidal portion was accessible in 6/10 cadaver heads. Fourteen cases of sphenopalatine sinus disease were identified, presenting with one or more clinical signs of exophthalmos, blindness, unilateral epistaxis or unilateral nasal discharge. Diagnoses included neoplasia (7), progressive ethmoidal haematoma (4), sinus cyst (2) and empyema (1). Computed tomography provided diagnostic information but could not differentiate the nature of soft tissue masses. Standing sinoscopic access to the palatine portion of the sphenopalatine sinus was possible for evaluation, biopsy and resection of abnormal soft tissues. Surgical access to the sphenoidal portion was limited. Eight horses were alive at 1 year after diagnosis, with a worse outcome associated with CT evidence of bone loss and a diagnosis of neoplasia. Conclusions Sphenopalatine sinus disease should be considered a rare cause of the clinical signs described. Knowledge of the anatomical variation of the sphenopalatine sinus is vital for interpreting CT images. A combination of CT and sinoscopy provides the most comprehensive approach for diagnosis and treatment of sphenopalatine sinus disease. [ABSTRACT FROM AUTHOR]

Since the 1950s, veterinary practitioners have included two separate dorsoproximal--palmarodistal oblique (DPr--PaDiO) radiographs as part of a standard series of the equine foot. One image is obtained to visualise the distal phalanx and the other to visualise the navicular bone. However, rapid development of computed radiography and digital radiography and their post-processing capabilities could mean that this practice is no longer required. The aim of this study was to determine differences in perceived image quality between DPr--PaDiO radiographs that were acquired with a computerised radiography system with exposures, centring and collimation recommended for the navicular bone versus images acquired for the distal phalanx but were subsequently manipulated post-acquisition to highlight the navicular bone. Thirty images were presented to four clinicians for quality assessment and graded using a 1--3 scale (1=textbook quality, 2=diagnostic quality, 3=non-diagnostic image). No significant difference in diagnostic quality was found between the original navicular bone images and the manipulated distal phalanx images. This finding suggests that a single DPr--PaDiO image of the distal phalanx is sufficient for an equine foot radiographic series, with appropriate post-processing and manipulation. The type and location of deep digital flexor tendon (DDFT) lesions may be important in predicting outcome. The objectives of this study were to determine the frequency of different types of DDFT lesions within the hoof capsule and to determine whether lesion type predicts return to athletic activity. Lesions of the DDFT were divided into: core lesions, dorsal border lesions and parasagittal splits. Lesion location was documented, and follow-up information was obtained by telephone survey at least 18 months after diagnosis. Of 168 horses with primary DDFT injury, 54 horses had dorsal border lesions, 59 had parasagittal splits and 55 had core lesions. Twenty-five per cent of all horses returned to previous levels of athletic activity within 18 months of MRI evaluation. Horses with complete splits or core lesions of the DDFT were significantly less likely to return to some level of athletic activity than horses with dorsal border lesions P<0.001. Dorsal border lesions of the DDFT appear to have a better prognosis than core lesions or parasagittal splits. Identifying the genes underlying phenotypic variation in natural populations can provide novel insight into the evolutionary process. The candidate gene approach has been applied to studies of a number of traits in various species, in an attempt to elucidate their genetic basis. Here, we test the application of the candidate gene approach to identify the loci involved in variation in gastrointestinal parasite burden, a complex trait likely to be controlled by many loci, in a wild population of Soay sheep. A comprehensive literature review, Gene Ontology databases, and comparative genomics resources between cattle and sheep were used to generate a list of candidate genes. In a pilot study, these candidates, along with 50 random genes, were then sequenced in two pools of Soay sheep; one with low gastrointestinal nematode burden and the other high, using a Nimble Gen sequence capture experiment. Further candidates were identified from single nucleotide polymorphisms ( SNPs) that were highly differentiated between high- and low-resistance sheep breeds. A panel of 192 candidate and control SNPs were then typed in 960 individual Soay sheep to examine whether they individually explained variation in parasite burden, as measured as faecal egg count, as well as two immune measures ( Teladorsagia circumcincta-specific antibodies and antinuclear antibodies). The cumulative effect of the candidate and control SNPs were estimated by fitting genetic relationship matrices ( GRMs) as random effects in animal models of the three traits. No more significant SNPs were identified in the pilot sequencing experiment and association study than expected by chance. Furthermore, no significant difference was found between the proportions of candidate or control SNPs that were found to be significantly associated with parasite burden/immune measures. No significant effect of the candidate or control gene GRMs was found. There is thus little support for the candidate gene approach to the identification of loci explaining variation in parasitological and immunological traits in this population. However, a number of SNPs explained significant variation in multiple traits and significant correlations were found between the proportions of variance explained by individual SNPs across multiple traits. Although single nucleotide polymorphisms (SNPs) are increasingly being recognized as powerful molecular markers, their application to non-model organisms can bring significant challenges. Among these are imperfect conversion rates of assays designed from in silico resources and the enhanced potential for genotyping error relative to pre-validated, highly optimized human SNPs. To explore these issues, we used Illumina's GoldenGate assay to genotype 480 Antarctic fur seal ( Arctocephalus gazella) individuals at 144 putative SNPs derived from a 454 transcriptome assembly. One hundred and thirty-five polymorphic SNPs (93.8%) were automatically validated by the program GenomeStudio, and the initial genotyping error rate, estimated from nine replicate samples, was 0.004 per reaction. However, an almost tenfold further reduction in the error rate was achieved by excluding 31 loci (21.5%) that exhibited unclear clustering patterns, manually editing clusters to allow rescoring of ambiguous or incorrect genotypes, and excluding 18 samples (3.8%) with unreliable genotypes. After stringent quality filtering, we also found a counter-intuitive negative relationship between in silico minor allele frequency and the conversion rate, suggesting that some of our assays may have been designed from paralogous loci. Nevertheless, we obtained over 45 000 individual SNP genotypes with a final error rate of 0.0005, indicating that the GoldenGate assay is eminently capable of generating large, high-quality data sets for non-model organisms. P-glycoprotein (P-gp), the product of ABCB1 gene, is thought to play a role in the biliary excretion of a variety of drugs, but specific studies in dogs have not been performed. Because a number of endogenous (ABCB1 polymorphisms) and exogenous (pharmacological P-gp inhibition) factors can interfere with normal P-gp function, a better understanding of P-gp’s role in biliary drug excretion is crucial in preventing adverse drug reactions and drug–drug interactions in dogs. The objectives of this study were to compare biliary excretion of technetium-99m-sestamibi (99mTc-MIBI), a radio-labelled P-gp substrate, in wild-type dogs (ABCB1 wild/wild), and dogs with intrinsic and extrinsic deficiencies in P-gp function. Dogs with intrinsic P-gp deficiency included ABCB1 mut/mut dogs, and dogs with presumed intermediate P-gp phenotype (ABCB1 mut/wild). Dogs with extrinsic P-gp deficiency were considered to be ABCB1 wild/wild dogs treated with the P-gp inhibitor ketoconazole (5 mg/kg PO q12h × 9 doses). Results from this study indicate that ABCB1 mut/mut dogs have significantly decreased biliary excretion of 99mTc-MIBI compared with ABCB1 wild/wild dogs. Treatment with ketoconazole significantly decreased biliary excretion of 99mTc-MIBI in ABCB1 wild/wild dogs. P-gp appears to play an important role in the biliary excretion of 99mTc-MIBI in dogs. It is likely that concurrent administration of a P-gp inhibitor such as ketoconazole will decrease P-gp-mediated biliary excretion of other substrate drugs as well. [ABSTRACT FROM AUTHOR]

Tenascins are glycoproteins found in the extracellular matrix (ECM) of many tissues. Their role is not only to support the tissue structurally but also to regulate the fate of the different cell types populating the ECM. For instance, tenascins are required when active tissue modeling during embryogenesis or re-modeling after injury occurs. Interestingly, the four members of the tenascin family, tenascin-C, -X, -R and -W, show different and often mutually exclusive expression patterns. As a consequence, these structurally related proteins display distinct functions and are associated with distinct pathologies. The present review aims at presenting the four members of the tenascin family with respect to their structure, expression patterns and implications in diseases and tissue mechanics. Question: Does forest vegetation community structure reflect legislative land use designations? Location: Adirondack Park, New York, USA. Methods: The Adirondack Park, located in northern New York State, is a mixture of public and private lands, with state-owned Forest Preserve lands comprising ca. 42% of the 2.4 million ha, on which timber harvesting and many other forms of anthropogenic disturbance are prohibited. A survey of vegetation communities was conducted in eighteen upland catchments with differing land use history (managed and Forest Preserve), including overstory, understory, and dead wood (snags and downed woody debris) using randomly placed plots. Results: Mean overstory density and basal area were not significantly different between land uses, although mean overstory tree size was greater in Preserve catchments. Sapling densities were greater in managed catchments, while mean herb/shrub coverage was not affected by land use. Densities of 25% of common species were affected by land use, determined by GIS coverages constructed using an Inverse Distance Weighted estimation procedure. Discriminant Analysis of per-plot plant community data correctly classified 89% of both managed and Preserve plots. Conclusion: The success of the Discriminant Analysis in classifying land uses based on vegetation communities indicates its potential utility of this method in comparing forest vegetation to a reference condition in this and other areas. The analysis suggests that at least 85 years is required for Adirondack up-land catchments to recover following harvesting. A 19-year-old Hackney pony was referred to a veterinary teaching hospital for further investigation of left-sided scrotal enlargement. Both testes were normally positioned within the scrotum and there was no pain on palpation. Ultrasonography revealed multiple masses of soft tissue echogenicity, located adjacent to the testicular parenchyma. They were hyperechoic when compared to the testicular parenchyma and affected both testes. Surgical excision was performed by closed castration. Gross examination revealed firm, white, nodular masses of varying size on the surface of both testes. The cut surface of individual masses had a dense, whorling, fibrous and somewhat shiny appearance. The histopathological diagnosis was bilateral, multicentric, well differentiated leiomyoma of the tunica albuginea. The smooth muscle origin was confirmed by immunohistochemical staining with an anti-human monoclonal antibody reactive to smooth muscle actin. This is the first report of a bilateral, multicentric, leiomyoma in the testes of a horse confirmed by histopathology and immunohistochemistry. This case highlights that neoplasia should be considered as a differential for bilateral testicular enlargement. [ABSTRACT FROM AUTHOR]

The Grand Bruit Fault Zone of southern Newfoundland is a fundamental structure within Late Precambrian basement on the Gondwanan margin of the Appalachian orogen. Within the fault zone, a sequence of structures documents changes in the sense of ductile displacement from (1) reverse dip slip, to (2) dextral strike slip, to (3) sinistral oblique slip, and, finally, to (4) dextral lateral offsets. Fault movements along this structure were punctuated by emplacement of a variety of plutons and minor intrusions which, when precisely dated, allow these movements to be bracketed at between 571 Ma and 564 Ma, 497 Ma and 427 Ma, 424 Ma and 420 Ma, and 421 Ma and 387 Ma, respectively. The tectonic evolution of the Gondwanan inlier of southern Newfoundland is mirrored, in large part, by the record of mylonite development within the Grand Bruit Fault Zone. These tectonic events are attributable to well-constrained, regional orogenic events of both the Pan-African and Appalachian cycles. Newly formed shear zones in the fault zone reactivate parts of much older faults of similar regional orientation and are, in some cases, kinematically indistinguishable from the ancestral structures. Integration of precise geochronological data with the sequence of overprinted fault structures demonstrates that, although the role of progressive deformation in shear zone development was important, the observed disposition of structures and rock units is primarily a function of polyorogenic accretion. As a multiple-reactivated structural lineament in a Gondwanan basement inlier, the fault zone exerted fundamental control over the tectonic development of the leading edge of the convergent southeast margin of the orogen. Residents and staff of a nursing home in a metropolitan area were randomly assigned to three conditions to: (a) test whether staff's use of operant behavioural management strategies (Condition 1) would cause a greater increase in residents' self-care behaviour than staff's use of mutual goal setting (Condition 2) or routine nursing care (Condition 3), and (b) examine how useful group scores were, compared to individual scores, in determining change in individual residents' behaviour due to treatment. In-service training was provided to staff in Conditions 1 and 2 but not in Condition 3. Over a period of 22 weeks, nursing staff encouraged subjects to perform targeted self-care tasks independently. Data analysis indicated significantly greater change in self-care behaviours for subjects in Condition 1 than for subjects in Conditions 2 and 3. However, visual inspection of data for each case revealed that individual scores were more useful than averages and differences between groups for determining the effectiveness of the clinical interventions.
