Summary: In this paper, valorisation of pumpkin oil cake as source of new protein carrier material for encapsulation was examined in order to enrich real food products. Cookies were used as a model system for addition of encapsulates at three different levels: 10%, 15% and 20%. The physicochemical and sensory properties were investigated, as well as conditions of stability after storage, in terms of total phenolic and betalain content. Furthermore, antioxidant, α‐glucosidase and ACE inhibitory potential of cookies was evaluated after simulated gastrointestinal digestion. The cookies showed good technological quality as well as acceptable sensorial properties. The addition of encapsulate increases the total phenolic and betalain contents and shoved significant stability during storage time. Novel peptides were produced after gastrointestinal digestion, which enhanced bioactive properties of enriched products through synergistic action with active compounds from beetroot juice. Obtained digest shoved significant antioxidant, antidiabetic and antihypertension activities, hence ensuring a potential healing effect that could be reached by consuming these cookies. The objective of this study was to show that biologically active hydrolysates can be obtained by simulated human gastrointestinal digestion (HGD) of transglutaminase cross-linked pumpkin oil cake protein (Tg-C) which was previously reported as a potential functional food additive. A two-stage in vitro digestion model system (by pepsin and α chymotrypsin and trypsin, simultaneously) was used to simulate the process of HGD on native and Tg-C major storage pumpkin oil seed/cake protein, cucurbitin (C). The biologically active potential of the digests was evaluated, measuring the angiotensin-converting-I enzyme (ACE) inhibitory and anti-oxidant capacity. The ACE inhibitory activity was determined in both final digests, with IC = 0.30 ± 0.04 mg/ml for C and IC = 0.28 ± 0.01 for Tg-C. The anti-oxidant potency of the examined proteins was enhanced by the digestion process. The 2,2-diphenyl-1-picrylhydrazyl and 2,2′-azino-bis(3-ethylbenzothiazoline-6-sulphonic acid) radical cation activities and reducing power testing showed that all the hydrolysates act as a radical quencher and reducing agents. Overall, the results showed that the cross-linking by Tg did not influence the digestion process, as well as having no effect on the biological activity of the hydrolysates. These also indicate that Tg-C, if used as functional food additive, after food consumption can be digested and become a source of peptides exerting positive effects on human health. The primary goal of this study is to improve our understanding of the extent of influence of climatic factors in Serbia and high-molecular-weight glutenin subunit (HMW-GS) composition upon wheat end-use quality. In-depth analyses were performed on four bread wheat cultivars that are the most common in agricultural practice in Serbia. Total glutenin content showed significant difference between the production years, in opposition to gliadins. Cluster analysis of different percentages of glutenin and gliadin subunit molecular weight ranges (<40,000, 40,000-80,000, 81,000-120,000, and >120,000) indicated that the year of production and the cultivar did not have a significant effect on the percentage ranges for glutenins. However, they had a considerable impact on the percentage ranges for gliadins. Production year and the interaction of year and cultivar had the strongest influences on the percentage of SDS-unextractable polymeric proteins. A synergistic effect of the HMW-GS composition and climatic conditions revealed that all eight samples with HMW-GS composition 2*, 5 + 10, 7 + 9 along with the highest Glu 1 score of 9 (out of a maximum of 10) produced in the year 2011 belonged to two clusters with the best wheat end-use quality. Furthermore, the climate conditions in 2011 made it possible for the wheat cultivars with HMW-GS composition -, 2 + 12, 7 + 9 to possess similar qualities as cultivars with HMW-GS composition 2*, 5 + 10, 7 + 9 produced in 2012. BACKGROUND: The aim of the present study was to investigate the ability of the small- and large-deformation fundamental rheological tests to monitor the changes during short-term post-harvest storage of wheat. RESULTS: At the end of wheat storage period, wet gluten quantity decreased, while gluten index increased in comparison to those of freshly harvested samples. The rheological properties of dough changed over the pre-defined period of storage in terms of becoming more elastic and less extensible in comparison to that of freshly harvested wheat. Visco-elastic properties of weaker flour samples changed more markedly during storage than those of stronger flours, indicating that the intensity of dough rheological changes during wheat storage might be dependent on gluten quality and were the characteristic of wheat variety. CONCLUSION: It was shown that small deformation dynamic oscillation and large deformation creep-recovery tests can be successfullyemployed to monitor the changes inflour quality duringwheat storage andthat requiredstorage period afterwheat harvesting has to be defined according to wheat variety initial rheological properties and its gluten quality. In this study, the fat phase of chocolate samples which contained cocoa butter from Ghana and cocoa butter equivalent (CBE) of moderate hardness was analyzed. Physical properties and shelf life of chocolate depend on the fat phase behavior as well as the amount and composition of added CBE. The laboratory-made chocolate samples were tempered at three different pre-crystallization temperatures (25, 27 and 29 °C), with three different concentrations of CBE, amounting to 3, 5 and 7 %, calculated on chocolate. The color and other physical attributes of chocolate samples were investigated by the following analytical methods: thermoreographic measurement, solid fat content (SFC) of chocolate, Blooming test (thermo-cycle test 32/20 °C) and color measurement. It was found that using CBE of moderate hardness did not change the melting properties of chocolate in relation to the investigated cocoa butter from Ghana (of moderate hardness). It was found that all three applied temperatures of pre-crystallization are optimum for the chocolate mass with the addition of the investigated CBE under the given measurement conditions. Sucrose-free spreads containing different share of maltitol (100, 70 and 30% as a bulking agents) are investigated with the aim to improve energy. Spreads with maltitol were produced in ball mill. The quality of produced spread mass is determined by moisture, size of the largest cocoa particle, yield stress and Casson plastic viscosity. The lowest energy values have spreads containing 100% maltitol. Spreads with maltitol do not have hygroscopic nature (moisture parameters are constant). Regardless of the sweetener type, spreads produced at maximum mixer speed rotation (50 r/min) have the lowest size of largest particle. Spreads containing 100% maltitol have greater plastic viscosity and significant viscous properties. Dynamic oscillatory measurements showed the spreads with different maltitol share are the viscoelastic solids, with dominate elastic component. The viscous component is dominant at lower frequencies (frequency <2 Hz). According to the rheology parameters, the best and the worst solution is if sucrose is replaced with maltitol, as a bulking agent, 100 and 30%, respectively. Practical Application Sugar-free confectionary products have become very popular nowadays. The energy value of spreads could be improved by selection of appropriate sweetener (maltitol). Hence, the maltitol share in this study would benefit the reducing energy value of spreads. It would be also useful for the future research to find the optimal maltitol share in spreads under different process conditions.
