ABSTRACT Objective Methods Results Conclusions This study investigated the role of autophagy on osteogenesis of DFCs under inflammatory microenvironment during tooth eruption.DFCs were isolated and identified. Lipopolysaccharide (LPS) was used to construct the inflammatory microenvironment in vitro and in vivo. Cell viability was examined by CCK‐8 assay. Osteogenic differentiation was evaluated by alkaline phosphatase (ALP) staining, alizarin red S (ARS) staining. The gene and protein levels were examined using qRT‐PCR and western blot analysis, respectively. We observed the process of tooth eruption after local LPS injection by micro‐CT and HE staining. Osteogenesis and autophagy were monitored through qRT‐PCR, western blot and histological staining of specific markers.LPS at the indicated concentrations did not produce toxic effects on DFCs, and significantly promoted the inflammatory gene expression. LPS inhibited osteogenic differentiation and activated autophagy in DFCs. Blocking autophagy with 3‐MA reversed the expression of osteogenic markers in LPS‐treated DFCs. Additionally, the eruption of LPS‐treated teeth was accelerated and their DFs exhibited an increased expression of TNF‐α and Beclin1, and decreased expression of ALP and RUNX2.Autophagy was involved in the suppression of the DFCs osteogenesis in an LPS‐induced inflammatory condition, suggesting the pivotal role of autophagy in inflammation‐induced premature tooth eruption. Background: The prevalence of supernumerary teeth is increasing in modern dental practice. However, the presence of multiple supernumerary teeth should be further investigated. Proper diagnosis of an underlying syndrome might save the patient from future health hazards through early diagnosis and optimal follow‐up screening. Case Presentation: A 13‐year‐old female patient presented with multiple retained deciduous teeth and delayed eruption of permanent teeth. Although the medical and family history of the patient did not raise any concerns, the clinical and radiographic examinations yielded intriguing findings. The patient presented with a total of 11 supernumerary teeth, which impeded the normal eruption of permanent dentition in addition to the presence of retained deciduous teeth. Additional clinical and laboratory investigations were conducted in response to the case's complexity, resulting in the diagnosis of Trichorhinophalangeal Syndrome (TRPS) Type I. The patient underwent a precise treatment plan and then was followed up for 6 months postoperatively to monitor the eruptive movement of the permanent teeth. Conclusion: When a syndrome is the underlying cause, monitoring unusual cases, such as those with multiple supernumerary teeth, can be lifesaving or aid in the early diagnosis of more serious complications. Objectives: Esthetic crown lengthening (ECL) is commonly advocated to treat patients with altered passive eruption (APE). Since the introduction of the minimally invasive surgical concept, a limited number of studies have investigated this technique in a standardized manner, with further studies required to verify the validity and predictability of the minimally invasive FL‐technique. The current randomized trial compares a minimally invasive (ECL), using piezosurgery with flapless‐approach (FL), versus an open‐flap (OF) approach in the management of patients with APE Type 1B. Materials and Methods: Twenty‐four patients diagnosed with APE Type 1B were randomly assigned into test (FL) with tunneling approach or control (OF) group with minimally invasive flap reflection (n = 12/group). Postoperative pain was assessed during the first 48 h. Gingival margin (GM) level relative to a custom‐made stent (rGM) and patient satisfaction were assessed preoperative, immediately after surgery, at 3 and 6 months postsurgically. Postoperative swelling was reported for the first week postsurgically. Plaque index (PI), bleeding on probing (BoP), clinical attachment level (CAL), pocket depth (PD) and pink esthetic score (PES), were evaluated at baseline and 6 months. Linear regression analysis was conducted for pain. Results: OF‐group reported significantly higher pain and swelling scores than FL‐group during the first 48 h (p < 0.05). FL‐group showed no significant differences regarding rGM between 3 and 6 months, in contrast to OF‐group, where a significant decrease in rGM was notable (p < 0.05). No significant differences in PI, BoP, CAL, PD, PES, and patient satisfaction scores were evident between groups (p > 0.05). Regression analysis demonstrated that treatment and gender were significant predictors for pain (p < 0.05). Conclusions: Within the current study's limitations, piezo‐surgical ECL with FL‐approach presented significantly lower postoperative pain, swelling, and early GM stability compared to OF‐approach. Clinical Significance: Piezosurgical ECL with a FL‐approach can be considered a predictable technique with advantages over the OF‐approach in the management of patients with APE Type1B. Background: Down syndrome (DS), a common chromosomal anomaly caused by trisomy of chromosome 21, is characterized by a broad spectrum of phenotypic characteristics across multiple organ systems, including cardiac defects and leukemia. Dermatological findings are prevalent among individuals with DS; however, these issues are frequently underrecognized and inadequately researched, resulting in a significant gap in the provision of comprehensive healthcare strategies. Given the increased life expectancy of patients with DS and delayed manifestation of many dermatoses, physicians are increasingly encountering dermatological findings in this population. Objective: This study aimed to assess the prevalence and types of dermatological findings in individuals with DS, compare them with those in a control group, and emphasize the necessity of incorporating dermatological evaluations into routine health monitoring. Methods: This prospective cross‐sectional study was conducted from June 2023 to June 2024 and involved 100 genetically confirmed individuals with DS and 100 age‐ and sex‐matched controls. Comprehensive demographic, clinical, and karyotype data were collected for the DS group, and all the participants underwent detailed morphological evaluations. Results: The DS group had a mean age of approximately 6.37 years, whereas the controls were around 7 years old, with no significant differences in age or sex distribution between the groups. Karyotype analysis showed that trisomy 21 was present in 92% of the cases, mosaicism in 6%, and translocation in 2%. Common dermatological findings in the DS group included xerosis cutis (49%), thin and sparse hair (48%), dental caries (34%), delayed tooth eruption (28%), nail dystrophy (25%), fissured tongue (23%), and cheilitis (18%). Significant differences were noted in the prevalence of scabies, bacterial infections, and café au lait macules between the DS and control groups (p < 0.01). Dysmorphic findings in the DS group included epicanthal folds (97%), upslanted palpebral fissures (97%), brachycephaly (91%), and single transverse palmar crease (89%). Significant gender differences were noted in the prevalence of brachycephaly and the sandal gap (p < 0.01). Conclusions: This study highlights the importance of regular dermatological care in enhancing the health management and quality of life of individuals with DS due to the prevalence and variability of dermatological conditions. The present study aimed to evaluate whether epigenetic markers are expressed in the dental follicles surrounding ectopically erupting teeth. Twenty‐one dental follicles were collected in 20 adolescent children through surgical exposure of ectopic teeth. The epigenetic modifications of DNA methylation and histone acetylation were evaluated by immunohistochemistry. The results showed cells positive for DNA‐methyltransferase 1 (DNMT1), DNA methyltransferase 3 beta (DNMT3B), ten‐eleven translocation‐2 (TET2), acetyl‐histone H3 (AcH3), acetyl‐histone H4 (AcH4), 5‐methylcytosine (5mC), and 5‐hydroxymethylcytosine (5hmC) were present in all the samples. The levels of epigenetic markers representing active chromatin (5hmC, AcH3, AcH4, and TET2) were statistically significantly higher than those of markers representing inactive chromatin (5mC, DNMT3B, DNMT1). In conclusion, follicles in ectopic teeth display major epigenetic modifications. In the follicles, epigenetic markers associated with the activation of bone‐related genes are more abundant than markers associated with the inactivation of bone‐related genes. Age estimation methods through developmental atlases are simplified by image comparison using radiographs. Atlas‐based methods have been utilized to create population‐specific charts addressing population diversity. To tackle the absence of a dedicated atlas for the Brazilian population, the objective of the present study was to create a new atlas for dental development in Brazil based on the London Atlas chart. The new atlas (RP Atlas) is based on the median of tooth development, eruption, and resorption in individuals aged 5 to 23. The sample (567 individuals) was divided between females (n = 290) and males (n = 277). The RP Atlas was handcrafted, scanned, colored, and clustered in two atlases divided by sex. The development of a specific atlas for the Brazilian population has the potential to enhance the accuracy of age estimation in forensic scenarios, notably in a mixed country like Brazil. Further research is needed to assess the suitability of the newly developed atlas for the Brazilian population. Objectives: This study aimed to identify studies exploring oral and systemic conditions using Singapore data. Methods: Studies were searched using the databases PubMed, Embase and Web of Science, with no publication date or language restrictions. Studies analysing Singaporean data were included in this review. Results: Six domains were identified: pregnancy and gingivitis; tooth eruption, dental caries and early‐life factors; Crohn's disease and oral microbiome; diabetes and periodontal diseases; number of teeth, chewing ability and cognitive status; and oral health and pneumonia. Using data from Singapore, oral‐systemic studies have prompted reflections on the aetiopathogenesis of oral disorders, such as common causes connecting oral and systemic chronic conditions. Moreover, it is speculated whether oral conditions could be used as a marker to predict future systemic diseases or whether early‐life factors could affect the development of oral and systemic immune responses. Conclusions: While Singapore provides opportunities to explore challenges connected to healthy ageing, it also explores health development in many stages of life. Background: To investigate the association between tooth loss and oral potentially malignant disorders and oral squamous cell carcinoma, focusing on epidemiological factors and genetic variants. Methods: Case–control study, including histologically confirmed oral potentially malignant disorders and oral squamous cell carcinoma cases and healthy controls. Unadjusted and adjusted odds ratios for this association were calculated. Single‐nucleotides polymorphisms were tested for individuals with and without missing teeth. Results: Case individuals were more edentulous while controls had fewer missing teeth (p = 0.006). There was an increased risk for the outcomes associated with edentulism (OR = 6.95, p = 0.000), even after adjustments for educational level (OR = 4.7, p = 0.034) and smoking habits (OR = 5.01, p = 0.022). Among individuals with tooth loss, rs1533767 (WNT11), rs3923087, and rs11867417 (AXIN2) were associated with the outcomes (OR = 1.67, p = 0.03, OR = 0.53, p = 0.05, and OR = 0.42, p = 0.00, respectively). Objective: The aim of this study was to explore the regulatory effect of RUNX2 mutation on dental follicle cells (DFCs) senescence and clarify the underlying mechanism. This study aimed to explore the basis for a novel mechanism of delayed permanent tooth eruption in cleidocranial dysplasia (CCD) patients. Materials and Methods: Dental follicles were collected from a CCD patient and healthy controls. Senescence‐associated β‐galactosidase (SA‐β‐gal) staining, Ki67 staining, cell cycle assays, and senescence‐related gene and protein expression assays were performed to assess DFCs senescence. Western blotting was performed to detect the activation of mitogen‐activated protein kinase (MAPK) signalling pathways, and the molecular mechanism underlying RUNX2 regulating in DFCs senescence was explored. Results: RUNX2 mutation inhibited the cellular senescence of DFCs from the CCD patient compared with healthy controls. Ki67 staining showed that mutant RUNX2 promoted DFCs proliferation, and cell cycle assays revealed that the healthy control‐derived DFCs arrested at G1 phase. RUNX2 mutation significantly downregulated senescence‐associated gene and protein expression. RUNX2 mutation suppressed ERK signalling pathway activation, an ERK inhibitor decreased healthy control‐derived DFCs senescence, and an ERK activator promoted CCD patient‐derived DFCs senescence. Conclusions: RUNX2 mutation delayed DFCs senescence through the ERK signalling pathway, which may be responsible for delayed permanent tooth eruption in CCD patients. [ABSTRACT FROM AUTHOR]

Background/Aim: Frondoside A is a sea cucumber extract which is well known for its anti‐inflammatory and immunomodulatory properties. The purpose of this study was to evaluate the effect of Frondoside A application in the alveolar socket on inflammatory responses after delayed replantation in rat teeth. Materials and Methods: Human periodontal ligament cells were cultured and exposed to Frondoside A. Cell‐counting kit‐8 assay was performed to evaluate the cell viability and nitric oxide assay was performed to assess the anti‐inflammatory effect of Frondoside A. Molars were extracted from 32 Sprague–Dawley rats and randomly divided into control and Frondoside A groups. After 30 min of extra‐oral dry time, molars were replanted. In the Frondoside A group, Frondoside A solution was applied in the alveolar socket before replantation. The animals were sacrificed after 28 days and histologically and immunohistochemically evaluated. Results: 0.5 μM Frondoside A showed higher cellular viability at 6 h and lower production of nitric oxide compared with other Frondoside A solutions (p <.05). The Frondoside A group demonstrated lower inflammatory resorption scores in both middle 1/3 and apical 1/3 of root compared to the control group (p <.05). The Frondoside A group showed lower levels of expression in both cathepsin K and CD45 compared with the control group (p <.05). Conclusions: Within the limits of this study, intra‐alveolar delivery of Frondoside A alleviates inflammatory root resorption in delayed replantation of rat teeth. [ABSTRACT FROM AUTHOR]

Background: Mutations in PTH1R are associated with Jansen‐type metaphyseal chondrodysplasia (JMC), Blomstrand osteochondrodysplasia (BOCD), Eiken syndrome, enchondroma, and primary failure of tooth eruption (PFE). Inheritance of the PTH1R gene can be either autosomal dominant or autosomal recessive, indicating the complexity of the gene. Our objective was to identify the phenotypic differences in members of a family with a novel PTH1R mutation. Methods: The proband was a 13‐year, 6‐month‐old girl presenting with short stature, abnormal tooth eruption, skeletal dysplasia, and midface hypoplasia. The brother and father of the proband presented with short stature and abnormal tooth eruption. High‐throughput sequencing was performed on the proband, and the variant was confirmed in the proband and other family members by Sanger sequencing. Amino acid sequence alignment was performed using ClustalX software. Three‐dimensional structures were analyzed and displayed using the I‐TASSER website and PyMOL software. Results: High‐throughput genome sequencing and Sanger sequencing validation showed that the proband, her father, and her brother all carried the PTH1R (NM_000316) c.1393G>A (p.E465K) mutation. The c.1393G>A (p.E465K) mutation was novel, as it has not been reported in the literature database. According to the American College of Medical Genetics and Genomics (ACMG) guidelines, the p.E465K variant was considered to have uncertain significance. Biological information analysis demonstrated that this identified variant was highly conserved and highly likely pathogenic. Conclusions: We identified a novel heterozygous mutation in the PTH1R gene leading to clinical manifestations with incomplete penetrance that expands the spectrum of known PTH1R mutations. Tooth eruption is a pivotal milestone for children's growth and development. This process involves with the formation of the tooth root, the periodontal ligament (PDL) and the alveolar bone, as the tooth crown penetrates the bone and gingiva to enter the oral cavity. This review aims to outline current knowledge of the adverse dental effects of antiresorptive medications. Recently, paediatric indications for antiresorptive medications, such as bisphosphonates (BPs), have emerged, and these agents are increasingly used in children and adolescents to cure pathological bone resorption associated with bone diseases and cancers. Since tooth eruption is accompanied by osteoclastic bone resorption, it is expected that the administration of antiresorptive medications during this period affects tooth development. Indeed, several articles studying human patient cohorts and animal models report the dental defects associated with the use of these antiresorptive medications. This review shows the summary of the possible factors related to tooth eruption and introduces the future research direction to understand the mechanisms underlying the dental defects caused by antiresorptive medications. Background: Stem/progenitor cells from human exfoliated deciduous teeth (SHED) show remarkable pluripotent, regenerative, and immunological capacities. During in vivo regenerative processes, there could be the presence of SHED in the surrounding inflammatory microenvironment, through toll‐like receptors (TLRs). Aim: The aim of this paper was to present a characteristic TLR expression profile on SHED for the first time. Design: Cells were harvested from extracted primary teeth (n = 10), anti‐STRO‐1 immunomagnetically sorted and cultivated, through colony‐forming units (CFUs). SHED were examined for mesenchymal stem/progenitor cell traits, including the expression of clusters of differentiation (CDs) 14, 34, 45, 73, 90, 105, and 146, and their multilineage differentiation aptitude. TLRs 1–10 expression was investigated for SHED in uninflamed and inflamed (25 ng/mL IL‐1β, 103 U/mL IFN‐γ, 50 ng/mL TNF‐α, and 3 × 103 U/mL IFN‐α; SHED‐i) microenvironmental conditions. Results: SHED were negative for CDs 14, 34, and 45, but were positive for CDs 73, 90, 105, and 146, and demonstrated characteristic multilineage differentiation. In an uninflamed microenvironment, SHED expressed TLRs 1, 2, 3, 4, 6, 8, 9, and 10. The inflammatory microenvironment downregulated TLR7 significantly on gene level and upregulated TLR8 on gene and protein levels (p <.05; Wilcoxon signed‐rank test). Conclusion: There appears to be a unique TLR expression profile on SHED, which could modulate their immunological and regenerative abilities in oral tissue engineering approaches. The article describes the case of a nine-year-old boy with maternal history of delayed tooth eruption (DTE). It discusses the unexpected spontaneous eruption of lower right first permanent molar (FPM) almost three years following surgical exposure. Also mentioned are the challenge in determining DTE aetiology, the negative effect of delaying FPM extraction, and a longer period of observation as a possible option presented to families of affected children.

Objective: To describe a strategy using digital technologies for improving the diagnosis, treatment planning, and surgical execution of patients with excessive gingival display (EGD) due to altered passive eruption (APE). Clinical Considerations: An important component for successful patient's management is to fulfill their esthetic expectations whilst delivering predictable and long‐term therapeutic outcomes. To achieve this goal in patients with excessive gingival display due to altered passive eruption, it is essential to perform an accurate diagnosis and to communicate to the patient the expected customized results using digital technologies. Computer‐aided designed and manufactured multifunctional anatomical prototypes (MAPs) may contribute to these purposes. Additionally, they can guide the surgical crown lengthening procedure or serve as a reference during the surgical guide fabrication providing information of the required anatomical landmarks. Conclusions: This novel strategy protocol for diagnosis, communication, and treatment management of patients with excessive gingival display follows functional and biological principles within the frame of a digital workflow, which improves the diagnostic capabilities, enhances communication, and guides the surgical treatment as shown in the 12 months follow‐up of the reported case. Clinical Significance: Developing a virtual patient by combining multiple digital data sets including cone‐beam computed tomography (CBCT), intra‐oral scans and digital photography, supports the clinician and the patient to achieve a comprehensive diagnosis and to better communicate the expected results to the patient. Furthermore, this digital treatment exercise based on anatomical and biological principles will facilitate the surgical precision and the achievement of successful outcomes, thus fulfilling the patient needs and expectations. Among developmental odontogenic cysts, the dentigerous type is the second most prevailing one. It is a benign intraosseous lesion commonly affecting the mandibular region. Dentigerous cysts present a high prevalence in children as they can be caused by the eruption of permanent teeth or the infection of deciduous ones. The adopted treatment modalities include enucleation (cystectomy), marsupialization, and decompression. Decompression maintains communication between the cyst and the oral medium through a sutured fixed device, namely an acrylic stent or a pretrimmed disposable suction tube. In the mixed dentition, the extraction of the affected primary teeth and the decompression approach is recommended, especially since children and parents are more tolerant of conservative treatments. We report in this study, a case of a 9-year-old boy complaining of a painful swelling in the left mandibular region. Intraoral and radiological examination revealed an expansion of the buccal and lingual cortical plates associated with teeth #73, #74, and #75 and a well-limited, unilocular radiolucent image extending from the distal aspect of tooth #31 to the mesial aspect of tooth #36 involving the crowns of the unerupted teeth #33, #34, and #35. The preliminary diagnosis was in favor of a dentigerous cyst. The treatment was to extract the deciduous teeth and to use a sterile tube for decompression. The patient was followed up for 5 years, a complete remission of the cyst was observed and the teeth #33, #34, and #35 re-erupted normally on the mandibular arch. [ABSTRACT FROM AUTHOR]

Tooth eruption is an important and unique biological process during craniofacial development. Both the genetic and environmental factors can interfere with this process. Here we aimed to find the failure pattern of tooth eruption among five genetic diseases. Both systematic review and meta‐analysis were used to identify the genotype–phenotype associations of unerupted teeth. The meta‐analysis was based on the characteristics of abnormal tooth eruption in 223 patients with the mutations in PTH1R, RUNX2, COL1A1/2, CLCN7, and FAM20A respectively. We found all the patients presented selective failure of tooth eruption (SFTE). Primary failure of eruption patients with PTH1R mutations showed primary or isolated SFTE1 in the first and second molars (59.3% and 52% respectively). RUNX2 related cleidocranial dysplasia usually had SFTE2 in canines and premolars, while COL1A1/2 related osteogenesis imperfecta mostly caused SFTE3 in the maxillary second molars (22.9%). In CLCN7 related osteopetrosis, the second molars and mandibular first molars were the most affected. While FAM20A related enamel renal syndrome most caused SFTE5 in the second molars (86.2%) and maxillary canines. In conclusion, the SFTE was the common characteristics of most genetic diseases with abnormal isolated or syndromic tooth eruption. The selective pattern of unerupted teeth was gene‐dependent. The aim was to characterise the endocannabinoid system (ECS) in the dental pulp of teeth at different stages of eruption. Pulp of: erupted premolars (EPM), third molars in pre‐eruptive (PThM), intraosseous (IThM) and eruptive stages (EThM) (n = 12 each group) were used. Messenger RNA expression of components of the ECS as cannabinoid receptors (CBr1 and CBr2), and anandamide synthetizing (NAPE–PLD) and degradation (FAAH) enzymes were measured by RT–PCR. Data were analysed using Student's t‐test for comparisons between two groups and one‐way analysis of variance and Tukey's post‐test for multiple comparisons (statistical significance: p < 0.05). mRNA expression of CBr2, NAPE–PLD and FAAH was similar in the studied stages, was lower in IThM than in PThM and EThM, and the lowest in EThM (p < 0.01); of note, CBr2 mRNA expression was not detected in EThM. CBr1 mRNA did not differ significantly between IThM and PThM but was lower in EThM (p < 0.01). The absence of CBr2 and presence of CBr1 in EThM suggest the involvement of the ECS via CBr1 as a mediator of tooth and bone tissue homeostasis during tooth eruption. [ABSTRACT FROM AUTHOR]

Objective: to investigate the impact of the severity of microcephaly caused by the Zika virus (MCZ) on tooth eruption and orofacial structures of children. Design: This case series study developed the research at the Mens Sana Rehabilitation Center, Arcoverde, Brazil. The study included 27 children diagnosed with MCZ. We performed the data collection in June 2018 through a questionnaire answered by the legal guardians, followed by a clinical examination of the children. The data were analyzed by the Mann–Whitney and Fisher's exact tests (p = 0.05). Results: The final sample was composed of 20 children. Of these, 13 (35.0%) had severe microcephaly, 5 (30.0%) had altered sequence of tooth eruption, 10 (50.0%) had delayed eruption, and the mean number of decayed teeth was 2.3. The most identified orofacial changes were teeth grinding habit (65.0%), difficult chewing (50.0%), and non‐nutritive sucking (50.0%). The Mann–Whitney test showed that the severity of microcephaly did not affect tooth eruption (p = 0.581). The Fisher's exact test showed that the severity of microcephaly was not associated with orofacial changes (p > 0.05). Conclusions: The severity of MCZ does not seem to influence changes in deciduous tooth eruption and the presence of orofacial anomalies. [ABSTRACT FROM AUTHOR]

This study investigated the influence of several covariates on the time and sequence of permanent dentition emergence following exfoliation of the deciduous teeth in puppies. The aim was to determine whether the emergence of permanent dentition can be used to assess whether a puppy that is traded across borders in the EU has reached the legally required minimum age of 15 weeks. The events were evaluated in a longitudinal study, with some cross‐sectional observations, of 440 puppies belonging to 82 breeds. No significant differences were found between the left and right sides of a puppy's dentition nor between male and female puppies. Breed size and skull type had a significant impact: both the appearance and the completion of tooth eruption occurred considerably later in small or toy breeds and brachycephalic skull type breeds. The sequence of emergence differed little between maxillary or mandibular quadrants or between breeds. The first emerging elements were the maxillary first premolars or first incisors or the mandibular first incisors. The emergence of a permanent tooth usually occurred within 3 days before or after exfoliation of its precursor, except for the canines, which demonstrated a wider tooth replacement interval. This study presents standards for age assessment based on the emergence of permanent dentition for breeds of different sizes, using median ages or cumulative emergence percentages. However, the legal age determination of puppies with an assumed age of 15 weeks cannot be performed based on permanent dentition emergence, as this occurred at a later age in a large proportion of dogs (at least 95%) in our studied population. Topics include dental alterations usually occuring as an outcome of disturbances during tooth development and modifying shape, number, size, and structure of the teeth as well as their eruption; and individuals diagnosed with ameloblastoma exhibited root resorptions in association with the lesion.

Dental trauma occurs frequently in toddlers and the treatment protocols vary depending on each case. Thus, clinicians must not only plan the treatment but also consider any possible sequelae and characteristics related to the child and the family. The aim of this paper was to report a case of a three‐year‐old boy who had a horizontal root fracture in the middle third of the primary maxillary right incisor root (tooth 51) and crown discoloration of the primary maxillary left incisor (tooth 61) which were successfully treated with conservative management. The management was established in agreement with the family and was based on active surveillance. The clinical and radiographic follow‐ups showed no changes besides pulp canal calcification of the left incisor throughout the eight years of follow‐up. The pulps remained normal with no pathological signs throughout their life cycle and the permanent successors erupted normally, presenting an excellent result of the non‐invasive approach adopted. Background and Objective: Teeth overeruption is a problem of clinical significance, but the underlying mechanism how changes in external occlusal force convert to the periodontium remodeling signals has been a largely under explored domain. And recently, periodontal ligament-associated protein-1 (PLAP-1)/ asporin was found to play a pivotal role in maintaining periodontal homeostasis. The aim of this study was to explore the function of PLAP-1 in the periodontally hypofunctional tissue turnover. Methods: After extracting left maxillary molars in mice, the left and right mandibular molars were distributed into hypofunction group (HG) and control group (CG), respectively. Mice were sacrificed for radiographic, histological, and molecular biological analyses after 1, 4 and 12 weeks. In vitro, dynamic compression was applied using Flexcell FX-5000 Compression System to simulate intermittent occlusal force. The expression of PLAP1 in loaded and unloaded human periodontal ligament cells (hP-DLCs) was compared, and its molecular biological effects were further explored by small interfering RNA (siRNA) targeting PLAP1. Results: In vivo, fiber disorder in periodontal ligament (PDL), bone apposition at furcation regions, and bone resorption in alveolar bone were illustrated in the HG compared with the CG. In addition, PLAP-1 positive area decreased significantly in PDL following occlusal unloading. In vitro, the loss of compressive loading relatively down-regulated PLAP1 expression, which was essential to promote collagen I but inhibit osterix and osteocalcin expression in hPDLCs. Conclusions: PLAP-1 presumably plays a pivotal role in occlusal force-regulated periodontal homeostasis by facilitating collagen fiber synthesis in hPDLCs and suppressing excessive osteoblast differentiation, further preventing teeth from overeruption. Nutritional restriction during developmental periods impairs organ physiology. Female rats were subjected to protein restriction during pregnancy and lactation to analyze dental and maxillary development. Four exposure groups were considered: normal‐protein diet during pregnancy and lactation (NP, 17% casein), low‐protein diet during lactation (LP‐L, 6% casein), low‐protein diet during pregnancy and lactation (LP), and low‐protein diet during pregnancy (LP‐G). Maxillae from 15‐day‐old male pups were collected. All protein‐restricted groups presented increased dentin thickness and reduced alveolar bone area. When protein restriction was applied during both gestation and lactation (LP), harmful effects were observed in the form of loss of protective OPG (osteoprotegerin) in tooth epithelium‐mesenchyme, due to higher RANKL expression, delay in odontoblast maturation, less dental pulp vascularity, reduction in amount of alveolar bone, and less matrix mineralization. In the LP‐L group, effects of protein restriction seemed less harmful, and despite less alveolar bone, the enhancement in BMP‐7, VEGF, and RANKL seems a compensatory signal to maintain maxillary osteogenesis. In LP‐G animals, Dspp expression was higher, suggesting a delay in odontoblast maturation or expression recuperation. In conclusion, maternal protein restriction affects dental and maxillary development. A low‐protein diet only in gestation allows for normal development. Objectives: This study aimed to determine the sequence and eruption chronology of permanent teeth in school children and adolescents of Chennai and compare the findings with an existing standard table. Additionally, the study also attempted to explore the influence of sex, body mass index (BMI), and socioeconomic status (SES) on tooth eruption patterns., Methods: A cross-sectional study was designed, and 12,650 children aged 5-18 years were selected from thirty-five schools using a multistage random sampling method. The clinical eruption status of all permanent teeth was evaluated using a new grading system. The chronology of permanent teeth eruption was determined using probit regression analysis while the intergroup comparison was carried out using the unpaired t-test and anova test., Results: The study sample included 6414 males and 6236 females. The mandibular first molar emerged as the first tooth at 5.5 years, whereas the maxillary second molar was the last to erupt, appearing at 11.9 years. A statistically significant difference (p < 0.0001) was noted between the sexes, BMI, and SES categories for most teeth; however, the variation was only 1-3 months., Conclusion: An accelerated eruption pattern was observed for all teeth, except the mandibular canines, with a notable reversal in the maxillary sequence, where canines erupted before the first premolars. Tooth eruption occurred earlier in females, in the lower jaw, right side, in normal BMI, and high SES categories. The Cayo Santiago rhesus macaque colony is a renowned primate population that has experienced significant natural and anthropogenic ecological variation in their 85-year history. Demographic and familial information is also tracked and collated for the majority of monkeys. Thus, the health history of rhesus macaques at Cayo Santiago should reflect the impacts of both environmental and genetic factors. In this study, we utilized a sample of skeletal remains comprised of 2787 individuals (1571 females, 1091 males), born between 1938 and 2017 from the derived skeletal collection of the primate colony to assess variation in survivorship, pathology, bone mineral density (BMD), and dental eruption status, in the context of hurricane impacts, nutritional fluctuations, and matriline genealogy. Results demonstrated that rhesus macaques at Cayo Santiago exhibit a range of skeletal pathologies that encompass biomedical and archaeological significance, multiple etiologies, severities, locations, and types, in addition to a secular trend of declining BMD that is hypothesized to reflect decreasing physical activity levels under increasing population densities. Specifically, hurricane impacts were found to increase the rate of systemic disease, decrease BMD in young adults, and delay eruption of the primary dentition. Certain matrilines exhibited heightened levels of systemic disease at early ages while others exhibited greater rates of congenital disease. Early-life adversity, through the experience of major hurricanes, may enhance inflammatory pathways, heightening the risk of disease and accelerating the aging process leading to reduced BMD. Such impacts may underly greater levels of observed infection post-hurricane through intensification of pathogen transmission and disease rates brought on by hurricane-adaptive social strategies that favor closer proximity. Familial susceptibility to disease indicates heritable host genetic factors are likely influencing disease patterning in the population. Aim: The purpose of this research was to present an artificial intelligence (AI) model, which can automatically segment and detect ectopic eruption of first permanent molars (EMMs) in early mixed dentition on panoramic radiographs using the no‐new‐Net (nnU‐Net) model. Design: A total of 438 EMMs obtained from 285 panoramic radiographs were included in this study. An AI model based on nnU‐Net was trained to segment and detect EMMs. The performance of the model was evaluated by the intersection over union (IoU), precision, F1‐score, accuracy and FROC. Furthermore, the detecting performance of nnU‐Net was compared with that of three dentists with different years of experience using the McNemar chi‐squared test. The reliability of different dentists was evaluated by intraclass correlation coefficients (ICCs). Results: The nnU‐Net yielded an IoU of 0.834, a precision of 0.845, an F1‐score of 0.902 and an accuracy of 0.990, whereas the dentists yielded a mean IoU of 0.530, a mean precision of 0.539, a mean F1‐score of 0.699 and a mean accuracy of 0.811. The ICC of different dentists was 0.776. The statistical analysis of the McNemar chi‐squared test showed that the nnU‐Net results were statistically significant and superior to those of dentists (p <.05). Background. Early diagnosis and interceptive treatment of the maxillary canine impaction is crucial as it reduces treatment complexity and decreases complications and adverse outcomes. Aim and Objectives. To determine the mean maxillary canine position among 9-10-year-old children and predict the risk of impaction of the maxillary canines. Methodology. Panoramic radiographs (PANs) of 289 healthy children aged between 9 and 10 years were observed where the average position of maxillary canines was related to the lateral incisor, sector locations, and angulations to the bicondylar line were traced. The average position was obtained by using descriptive statistics. One sample Wilcoxon signed-rank test is done to predict the risk of canine impaction by comparing the data obtained to the average position from prior studies. Results. A total of 289 PANs (126 males and 163 females) were utilized for the analysis. The findings showed that the average position of the maxillary canines in our population was statistically different from the average position of nonimpacted canines in previous studies. However, on average, more than 85% of canines in our population were still located within the safe range of satisfactory position, with females showing slight predominance outside of the acceptable range. The mean scores of the angles between the right canine and lateral incisor were significantly higher among females than males (p = 0.001). Similarly, females had a significantly higher mean angle of the left canine than males (p < 0.001). In regard to the angles between the bicondylar line and permanent maxillary canine, the mean scores were not significantly different (p > 0.05) on both the left and right side. Conclusion. There is a low risk of impaction of maxillary canines in the Malaysian population. Background: There are diverse opinions among dentists about managing compromised first permanent molars (cFPMs) in children and a perceived lack of guidance to help them evaluate prognosis. Aim: To evaluate the current management of cFPM in children referred to a UK hospital centre and to report the severity of the affected teeth. Design: A service evaluation was undertaken, based on case records of medically fit children (6‐11 years) referred to for the management of cFPMs. The presence of hypomineralisation, post‐eruptive breakdown and the proposed care plans were recorded. Radiographic signs of severity were scored using the ICDAS index (intra/inter‐rater kappa 0.96/0.82). Results: From 349 records screened over a 4‐month period, 249 met the selection criteria. Almost 81% were planned to have extraction of at least one cFPM, whereas 19.3% were managed without extraction. More than half of the extraction cases (n = 105) had radiographic radiolucencies not exceeding the middle third of dentine in the worst‐affected FPM. At the time of extraction, the mean age of the patients was 9.8 years (±0.9). GA was used in 196 (97.5%) cases, and 40.8% had not received previous treatment in any of their cFPMs. Conclusion: Potentially restorable cFPMs in children is, most of the time, in a cohort of UK patients referred for tier 3 services, being managed by timed extractions under general anasethesia. Orthodontic extrusion preserves the natural root system and may convert the tooth indicated for extraction into useful tooth with good prognosis and a low risk of failure. Orthodontic tooth eruption can be an alternative to treatment such as surgical crown lengthening, especially in esthetic areas, and provides more favorable conditions for prosthodontic coronal restorations by guaranteeing proper sealing and esthetics and preserving periodontal tissue health. The aim of this case report was to explain a multidisciplinary approach that successfully preserve and treat the teeth with subgingival carious lesion. Aim: To describe a novel surgical method (crown rotation surgery) to manage inversely impacted central incisors with immature roots. Methodology: Two young patients each presented with an inversely impacted maxillary central incisor. To protect the apical tissues, the two impacted incisors were rotated downwards to a relatively normal position without extraction from their bony sockets. Results: After crown rotation surgery, spontaneous eruption, continuous root development, and periodontal healing of the rotated incisors were observed. The pulp retained vitality and blood flow was normal. Moreover, there were no obvious signs of pulp canal obliteration (PCO), as indicated by Cone Beam Computed Tomography (CBCT) imaging. Conclusions: By optimising protection of the vital pulp and apical tissues, crown rotation surgery represents a minimally invasive, conservative, and practical surgical technique for treating inversely impacted incisors with developing roots. Objectives: This study was done to determine the timing of eruption of permanent teeth by sex and the number of permanent teeth erupted at different ages and to assess its association with malocclusion. Material and Methods: The sample for this study consisted of 633 healthy subjects (317 boys and 316 girls) aged between 5.0 and 15.0 years. The subjects were divided into subgroups considering an age interval of 1 year. Results: The mean eruption times were lower for girls compared with boys except for the maxillary and mandibular second premolars and second molars, which were found to erupt earlier in boys. All the permanent mandibular teeth, except the second premolars, tended to erupt earlier than their maxillary antagonists in both sexes. The sequence of eruption differs between girls and boys in the maxillary canine and maxillary second premolars. In Class III malocclusion, all the permanent mandibular teeth erupt earlier than their maxillary antagonists. In the entire sample, the correlation between the number of erupted teeth and age was strong (p <.05), but the differences between sexes were not significant. Conclusions: It is important to know the normal eruption time of permanent teeth in the population due to diagnosis and better treatment planning in pediatric dentistry and orthodontics. Mutations in LTBP3 are associated with Dental Anomalies and Short Stature syndrome (DASS; MIM 601216), which is characterized by hypoplastic type amelogenesis imperfecta, hypodontia, underdeveloped maxilla, short stature, brachyolmia, aneurysm and dissection of the thoracic aorta. Here we report a novel (p.Arg545ProfsTer22) and a recurrent (c.3107‐2A > G) LTBP3 variants, in a Turkish family affected with DASS. The proband, who carried compound heterozygous variant c.3107‐2A > G, p.Arg545ProfsTer22, was most severely affected with DASS. The proband's father, who carried the heterozygous variant c.3107‐2A > G had short stature and prognathic mandible. The mother and brother of the proband carried the heterozygous variant p.Arg545ProfsTer22, but only the mother showed any DASS characteristics. The c.3107‐2A > G and the p.Arg545ProfsTer22 variants are expected to result in abnormal LTPB3 protein, failure of TGFβ‐LAP‐LTBP3 complex formation, and subsequent disruption of TGFβ secretion and activation. This is the first report of heterozygous carriers of LTBP3 variants showing phenotypes. The new findings of DASS found in this family include taurodontism, single‐rooted molars, abnormal dentin, calcified dental pulp blood vessels, prognathic mandible, failure of mandibular tooth eruption, interatrial septal aneurysm, secundum atrial septal defect, tricuspid valve prolapse, and a recurrent glenohumeral joint dislocation. Objective: The aim of this case report is to present a novel digital approach of guiding orthodontics and periodontal surgery by creating interim restorations that represent the final shape of the teeth before starting the treatment. Clinical considerations: The patient presented with worn dentition, super‐erupted maxillary and mandibular anterior teeth, and altered passive eruption, all this creating a gummy smile. Following a digital wax‐up design, a digital orthodontic set‐up, and the digital bracket bonding, temporary restorations, and bracket indirect bonding trays were fabricated. Restorations' cementation, bracket bonding, and periodontal surgery were performed the same day. After 14 months of orthodontic treatment, teeth were restored with final ceramic restorations. One year and 6 months was the total treatment duration. Conclusion: Prosthetically guided orthodontic concept allows the use of digital technology to simulate the final outcome of the patient treatment before starting dental therapy. This technique not only improves the communication between specialists, it also reduces treatment time and increases precision and predictability. Clinical significance: The prosthetically guided orthodontics (PGO) concept has the goal to start interdisciplinary therapy with a simulation of the final outcome. Giving the worn teeth their proper size through interim restorations prior to orthodontic therapy will guide the orthodontist from the beginning of the treatment and will improve communications between all the specialists. Objectives: (1) To investigate sex differences in molar wear in known‐age Cayo Santiago rhesus macaques (Macaca mulatta) and, (2) To explore sex differences in body weight and molar eruption timing as factors influencing sex differences in molar wear. Materials and methods: Data set I comprises wear scores, ages and body weights of 212 living monkeys included in the 1985 roundup. Data set II consists of molar wear measurements taken on 2D images of 103 of these monkeys' dental remains. Ordinal logistic regression was used to analyze the first data set. General linear models were used to analyze the second. Results: Males generally exhibited more wear than females at equivalent chronological ages, though results varied by tooth type for the second data set. Male body weight in the full 1985 living sample was significantly related to dental wear, when age was taken into account; however, when males less than 8 years of age were eliminated from the sample, the association between dental wear and weight became statistically insignificant. Analysis of the second data set suggested no statistically significant sex difference in dental wear for third molars, despite the approximately 2 year sex difference in eruption age for this tooth type. Discussion: This study suggests that body weight in males might be a predictor of dental wear and that if it is, body weight might also influence sex differences in dental wear. Aims: To describe oral manifestations in children born with microcephaly attributed to congenital Zika virus syndrome (CZS). Methods: Data was collected in semiannual intervals from 2017 to 2019, by oral exams of the children and interview with caregivers at a Public Dental Center in Rio de Janeiro, Brazil. A single calibrated examiner performed clinical examinations. Results: Of 38 eligible children, 34 were followed‐up from 12 to 30 months of age, 20 boys and 14 girls. The mean age of emergence of their first primary tooth was 12.4 months (SD = 2.9). By 30 months of age only 14.7% (n = 5) had complete primary dentition. Alteration in the sequence of tooth emergence was observed in 41.1% (n = 14). Radiographic examination demonstrated dental agenesis (14.7% n = 5). Dental developmental alterations (38.2%, n = 13), enamel defects (14.7%, n = 5), eruption cysts/hematoma (23.5%, n = 8), gingival bleeding (55.8%, n = 19), narrow palate, and bruxism (64.7%, n = 22) were also observed. The article describes the case of a 2-year-old boy with pre-eruptive intracoronal resorption defect of erupting primary molar leading to facial cellulitis. A localized periodontal infection associated with pericoronitis was diagnosed and oral antibiotics were prescribed. A facial swelling arose two days after completion of the antibiotics. Topics covered include the differential diagnoses, the results of the examination and the outcome of the treatment.

Objective: To analyze published scientific evidence about the most common orofacial disorders in children with microcephaly associated with ZIKV infection. Methods: Through a scoping review, we respond to the search question was formulated: "What are the orofacial alterations in children with microcephaly associated with ZIKV?" It was performed a search in PubMed, EMBASE, Scopus, and Health Virtual Library. It was selected papers wrote in English, Portuguese, or Spanish. An evidence quality analysis was performed using the Agency for Healthcare Research and Quality classification. Results: It was included 11 studies performed in the Brazilian Northeast region. The studies were carried out between 2018 and 2020. The main related alterations were delay in the tooth‐eruption and deformation in the oral structures. Conclusion: The results observed in our work show moderate scientific evidence regarding the association of ZIKV with orofacial alterations in children with microcephaly due to the classifications of the quality of evidence of each one of the included studies. To report a case of cleidocranial dysplasia (CCD) with 15 supernumerary teeth and review the relevant literature. Diagnose and treat a CCD patient with 15 impacted supernumerary teeth in the maxilla and mandible. When CCD is accompanied by supernumerary teeth ambushed in the jaw and does not affect the health of the body, it can be regarded as a part of normal bone tissue. If X‐ray or computed tomography examination shows that supernumerary teeth affect permanent tooth eruption, the replacement process of adjacent teeth and permanent tooth eruption should be closely observed, and supernumerary teeth should be extracted in time if necessary. Dental follicles are involved in odontogenesis, periodontogenesis, and tooth eruption. Dental follicles are unique structures, considering that their remnants can persist within the jawbones after odontogenesis throughout life if the tooth does not erupt. Pathological changes may occur in these tissues as individuals age. The changes range from benign to life threatening. Thus, the assessment of age‐related changes in dental follicles associated with unerupted teeth is of paramount importance. In this review, we summarize the physiological roles and changes in dental follicles in odontogenesis, tooth eruption, and aging, in addition to the pathological changes associated with these structures. We encourage investigators to consider this peculiar tissue as a unique model and explore its potential to clarify its importance from the viewpoints of developmental biology, tissue physiology, and pathology. Wolf‐Hirschhorn syndrome is a polymalformative chromosomal disorder caused by a deletion in the distal region of the short arm of chromosome 4. The disease is considered rare (1/50,000 births) and predominantly affects females (2:1). In addition to the characteristic facial phenotype ("Greek warrior helmet"), its clinical manifestations include epilepsy, developmental and psychomotor delay, intellectual disability, cardiac and respiratory complications, and eating problems. The most prevalent oral manifestations are hypodontia, delayed tooth eruption, morphological dental abnormalities, dental malocclusions, cleft lip/palate and ogival palate. Based on our clinical experience, Wolf‐Hirschhorn syndrome does not represent an absolute contraindication for any type of dental procedure. The feasibility of dental treatment will depend mainly on the degree of epilepsy control and on the level of collaboration, this latter conditioned by the severity of the intellectual disability and communication difficulties. Unicystic ameloblastoma (UAM) in the pediatric population is a rare clinical entity that has not been well addressed in the literature. Radical approaches affect a growing young patient's physical and psychological development, so conservative approaches are widely used for management in children. This report describes the case of a 9‐year‐old girl with UAM of the mandible, which also involved the impaction of the first and second premolars. Marsupialization with orthodontic treatment was performed to shrink the lesion and upright the first premolar involved in the tumor. Ten months after marsupialization, the lesion had healed entirely. The combination of conservative surgery and orthodontic treatment effectively shrank the lesion, preserving mandibular growth, preserving the involved first and second premolars, and promoting the eruption of the teeth. Background: The second permanent molar plays an important role when considering extraction of first permanent molars severely affected by molar‐incisor hypomineralization (MIH). Aim: To assess the association between MIH and enamel hypomineralization of the second permanent molars in terms of presence and severity. Design: In this retrospective cross‐sectional study, permanent teeth were evaluated in 453 patients between the ages of 13 and 16 at a dental clinic, using intraoral photographs. A calibrated examiner classified enamel hypomineralization using the MIH index and dental caries experience using the DMFT and DMFS indices. Statistical analyses were performed using a linear generalized model and ordinal logistic regression. Results: The most commonly affected teeth by enamel hypomineralization were permanent molars, whereas incisors, premolars, and canines were less affected. The presence of severe defects in the first permanent molars was associated with mild defects in the second permanent molars (OR = 4.01; 95% CI: 2.50‐7.77). Enamel hypomineralization was associated with increased caries experience (PR = 5.54; 95% CI: 3.81‐9.06). Conclusion: Enamel hypomineralization mainly affects the first and second permanent molars. Mild defects in the second permanent molars tend to be more frequent in patients with severe MIH in the first permanent molars. Aim: To evaluate in a laboratory setting, the impact of three designs of endodontic access cavities on dentine removal and effectiveness of canal instrumentation in extracted maxillary first molars using micro‐computed tomography (micro‐CT). Methodology: A total of 30 extracted intact maxillary first molars were selected and scanned by micro‐CT with a voxel size of 24 µm and randomly distributed into three groups: the traditional endodontic cavity (TEC) group, the conservative endodontic cavity (CEC) group and the guided endodontic cavity (GEC) group. The pulp chambers of teeth in the groups were accessed accordingly. After root canal preparation, the teeth were rescanned. The volume of dentine removed after canal preparation, the noninstrumented canal areas, canal transportation and centring ratio were analysed. Data were analysed statistically using one‐way analysis of variance. Tukey's post hoc test was used for multiple comparisons. The significance level was set at p <.05. Results: The total volume of dentine removed was significantly greater in the TEC group after root canal preparation (p <.05). No significant differences in the volume of dentine removed occurred between the CEC and GEC groups (p >.05). The volume of dentine removed in the crown, pericervical dentine and coronal third of the canal was significantly lower in CEC and GEC groups when compared to that in the TEC group (p <.05), no difference was observed in the middle third of the canal and apical third of the canal amongst the three groups (p >.05). There was no significant difference in noninstrumented canal area, canal transportation and centring ratio amongst the TEC, CEC and GEC groups (p >.05). Conclusions: In extracted maxillary molars tested in a laboratory setting, CEC and GEC preserved more tooth tissue in the crown, pericervical dentine and coronal third of the canal compared with TEC after root canal preparation. The design of the endodontic access cavity did not impact on the effectiveness of canal instrumentation in terms of noninstrumented canal area, canal transportation and centring ratio. Studies of dental caries on the deciduous dentition can be a source of information about changes in diet, which can be correlated with the time of eruption of the deciduous and permanent teeth. In this respect, the moment of transition from the maternal diet (mother's milk) to solid food is particularly interesting. The aim of this study was to assess changes in the diet of children who inhabited Radom, Poland, in the late Medieval/early Modern (LMP, 14th–17th c.) and Modern (MP, 18th–19th c.) periods by applying odontological and physicochemical methods. We analyzed the dental remains of 104 nonadult individuals (1.0–2.5 years, 3.0–5.5 years, and 6.0–8.0 years), with a total of 1044 deciduous and 108 permanent teeth. The concentration of stable δ15N isotopes in collagen isolated from the femur bones was assessed in 63 individuals (27 children, 36 adults). The diagnosis of dental caries was carried out through visual, radiographic, and fluorescent techniques. Dental caries were recorded in 30% (68/224) of the deciduous teeth in the LMP and 39% (321/820) of the deciduous teeth in the MP. Dental caries were found more often in the two earliest age groups of children in the MP than in the LMP. Early childhood caries (ECC) were diagnosed in 18% of the deciduous dentition of children from the MP. Isotopic analyses showed that the start of weaning occurred in the MP group almost 4 months earlier than in the LMP group (1.1 years in LMP vs. 0.7 years in MP). The final weaning of children from maternal milk in the LMP group took place at 2.5 years of age, whereas in the MP group it took place at 3.2 years of age. The study shows that the period of feeding children exclusively with the mother's milk in the MP was shorter. This means a faster introduction of solid food products to the diet. Background: Cellular cementum, a mineralized tissue covering apical tooth roots, grows by apposition to maintain the tooth in its occlusal position. We hypothesized that resident cementocytes would show morphological changes in response to cementum apposition, possibly implicating a role in cementum biology.Methods: Mandibular first molars were induced to super-erupt (EIA) by extraction of maxillary molars, promoting rapid new cementum formation. Tissue and cell responses were analyzed at 6 and/or 21 days post-procedure (dpp).Results: High-resolution micro-computed tomography (micro-CT) and confocal laser scanning microscopy showed increased cellular cementum by 21 dpp. Transmission electron microscopy (TEM) revealed that cementocytes under EIA were 50% larger than control cells, supported by larger pore sizes detected by micro-CT. Cementocytes under EIA displayed ultrastructural changes consistent with increased activity, including increased cytoplasm and nuclear size. We applied EIA to Hyp mutant mice, where cementocytes have perilacunar hypomineralization defects, to test cell and tissue responses in an altered mechanoresponsive milieu. Hyp and WT molars displayed similar super-eruption, with Hyp molars exhibiting 28% increased cellular cementum area versus 22% in WT mice at 21 dpp. Compared to control, Hyp cementocytes featured well-defined, disperse euchromatin and a thick layer of peripherally condensed heterochromatin in nuclei, indicating cellular activity. Immunohistochemistry (IHC) for cementum markers revealed intense dentin matrix protein-1 expression and abnormal osteopontin deposition in Hyp mice. Both WT and Hyp cementocytes expressed gap junction protein, connexin 43.Conclusion: This study provides new insights into the EIA model and cementocyte activity in association with new cementum formation. Introduction. Although primary teeth have a determinative role in development of normal occlusion, few studies about anomalies related to deciduous dentition have been conducted so far. Regarding recent improvements in common knowledge and the importance of maintaining primary teeth until eruption of succedaneous teeth, identifying the morphology of primary teeth and probable variations is of great importance to achieve optimal therapeutic outcome. This study aims to determine the prevalence of three-rooted mandibular primary molars in a population of northwestern Iran. Materials and Methods. In this descriptive cross-sectional study, periapical radiographs of patients attending private oral and maxillofacial radiology offices in the northwestern region of Iran from 2017 to 2019 were retrospectively reviewed. A total of 300 cases in the 3– 10 -year age range having bilateral periapical radiographs from mandibular primary molars were screened. First and second primary molars were observed meticulously. Gender, side of the mandible (right or left), symmetry, overall prevalence, and prevalence considering the type of molar (D or E) were recorded and analyzed. Results. Overall prevalence of three-rooted mandibular primary molars was 9.33 % ( 28 / 300), 92.9 % of which were unilateral. The prevalence of teeth showing supernumerary roots among all teeth examined was 2.5 % (30 / 1200). Considering symmetry and gender, the occurrence of these three-rooted primary molars did not differ significantly (respectively, p = 0.832 and p = 0.541). However, there was a significant relationship between the occurrence of supernumerary roots and left side for three-rooted first molars and right side for three-rooted second molars (p = 0.021). Conclusion. Obtained data show that three-rooted mandibular primary molars in children of northwest region of Iran have a prevalence of approximately 10 % which urges some specific considerations in exodontic and endodontic procedures. Sambar (Rusa unicolor), one of 6 deer species introduced to Australia, are poorly studied and declared vulnerable in their native range of India and southern Asia. In southeastern Australia sambar are increasing in abundance and distribution despite commercial venison harvest and unlimited public hunting. The ability to accurately age individuals is important for management of cervid populations because many life history events are age dependent. The techniques most widely used to estimate the age of cervids involve tooth eruption and molar wear (EW), and interpretation of cementum annuli (CA) in incisors. Aging techniques based on teeth are influenced by diet, body condition and metabolic stressors such as disease and reproduction. We compared age estimation derived by both EW and CA techniques and applied age estimates from both methods to lifetables and the Euler‐Lotka equation to predict population growth. Sambar have CA that are discernible and correspond to midwinter despite calving throughout the year in a temperate environment. Age estimation by eruption times of molar teeth appears reliable in sambar but estimation based on wear patterns of molar teeth is less accurate than CA. Assuming CA is accurate, aging by EW was less than 30% for sambar 3 years and older. For management purposes, however, estimation of rates of population growth using either the Euler‐Lotka equation or a lifetable provided similar outputs using ages estimated by either CA or EW. © 2021 The Wildlife Society. Fusion is a developmental anomaly characterised by the union of two adjacent teeth or tooth‐like substance. Odontomes are malformation of the dental tissue which arise during normal tooth development. They are usually asymptomatic but often associated with tooth eruption disturbance. In this paper, we report a rare case of fusion involving permanent mandibular second molar with an odontome, which led to a partial eruption of the molar as well as its devitalisation. Successful endodontic management was carried out in this case with a supplementary disinfection procedure using XP Endo finisher (FKG Dentaire, La Chaux‐de‐Fonds, Switzerland). The use of high‐end diagnostic imaging modalities such as cone beam computed tomography (CBCT) helped in making a confirmatory diagnosis; determining the treatment plan before undertaking the actual treatment; for better understanding of the fused tooth's complicated root morphology; and for its effective management and to follow up this unusual case for 12 months.
