Molecular markers can serve as diagnostic tools to support pathological analysis in thyroid neoplasms. However, because the same markers can be observed in some benign thyroid lesions, additional approaches are necessary to differentiate thyroid tumor subtypes, prevent overtreatment and tailor specific clinical management. This applies particularly to the recently described variant of thyroid cancer referred to as noninvasive follicular thyroid neoplasm with papillary‐like nuclear features (NIFTP). This variant has an estimated prevalence of 4.4% to 9.1% of all papillary thyroid carcinomas worldwide. We studied 60 thyroid lesions: 20 classical papillary thyroid carcinoma (CPTC), 20 follicular variant of PTC (FVPTC) and 20 NIFTP. We examined morphological and molecular features to identify parameters that can differentiate NIFTP from the other PTC subtypes. When blindly investigating the nuclear architecture of thyroid neoplasms, we observed that NIFTP has significantly longer telomeres than CPTC and FVPTC. Super‐resolved 3D‐structured illumination microscopy demonstrated that NIFTP is heterogeneous and that its nuclei contain more densely packed DNA and smaller interchromatin spaces than CPTC and FVPTC, a pattern that resembles normal thyroid tissue. These data are consistent with the observed indolent biological behavior and favorable prognosis associated with NIFTP, which lacks BRAFV600E mutations. Of note, next‐generation thyroid oncopanel sequencing was unable to distinguish the thyroid cancer histotypes in our study cohort. Robotics in agriculture faces several challenges, such as the unstructured characteristics of the environments, variability of luminosity conditions for perception systems, and vast field extensions. To implement autonomous navigation systems in these conditions, robots should be able to operate during large periods and travel long trajectories. For this reason, it is essential that simultaneous localization and mapping algorithms can perform in large‐scale and long‐term operating conditions. One of the main challenges for these methods is maintaining low memory resources while mapping extensive environments. This work tackles this issue, proposing a localization and mapping approach called VineSLAM that uses a topological mapping architecture to manage the memory resources required by the algorithm. This topological map is a graph‐based structure where each node is agnostic to the type of data stored, enabling the creation of a multilayer mapping procedure. Also, a localization algorithm is implemented, which interacts with the topological map to perform access and search operations. Results show that our approach is aligned with the state‐of‐the‐art regarding localization precision, being able to compute the robot pose in long and challenging trajectories in agriculture. In addition, we prove that the topological approach innovates the state‐of‐the‐art memory management. The proposed algorithm requires less memory than the other benchmarked algorithms, and can maintain a constant memory allocation during the entire operation. This consists of a significant innovation, since our approach opens the possibility for the deployment of complex 3D SLAM algorithms in real‐world applications without scale restrictions. Microbial evolution experiments provide a powerful tool to unravel the molecular basis of adaptive evolution but their outcomes can be difficult to interpret, unless the selective forces that are applied during the experiment are carefully controlled. In this respect, experimental evolution in continuous cultures provides advantages over commonly used sequential batch‐culture protocols because continuous cultures allow for more accurate control over the induced selective environment. However, commercial continuous‐culture systems are large and expensive, while available DIY continuous‐culture systems are not versatile enough to allow for multiple sensors and rigorous stirring.We present a modular continuous‐culture system that adopts the commonly used GL45 glass laboratory bottle as a bioreactor vessel. Our design offers three advantages: first, it is equipped with a large head plate, fitting two sensors and seven input/output ports, enabling the customization of the system for many running modes (chemostat, auxostat, etc.). Second, the bioreactor is small (25–250 ml), which makes it feasible to run many replicates in parallel. Third, bioreactor modules can be coupled by uni‐ or bi‐directional flows to induce spatiotemporal variation in selection. These features result in a particularly flexible culturing platform that facilitates the investigation of a broad range of evolutionary and ecological questions.To illustrate the versatility of our culturing system, we outline two evolution experiments that impose a temporally or spatially variable regime of selection. The first experiment illustrates how controlled temporal variation in resource availability can be utilized to select for anticipatory switching. The second experiment illustrates a spatially structured morbidostat setup that is designed to probe epistatic interactions between adaptive mutations. Furthermore, we demonstrate how sensor data can be used to stabilize selection pressures or track evolutionary adaptation.Evolution experiments in which populations are exposed to controlled spatiotemporal variation, are essential to gain insight into the process of adaptation and the mechanisms that constrain evolution. Continuous‐culture systems, like the one presented here, offer control over key environmental parameters and establish a well‐defined regime of selection. Beach erosion poses significant threat to small island economies which are generally highly dependent on coastal tourism. This work investigates the evolution of the low‐lying sandy coast of Boa Vista through an integrated characterization of coastline and shoreline indicators (over the past four decades) based on aerial imagery. It was found that tandem use of the two indicators was important to obtain a reliable perspective of the Boa Vista low‐lying coastal evolution across a wide range of coastal environments. Results indicate that between 1968 and 2010 the coast was relatively stable, although some spatial variability was recognized. The largest changes were observed at the tips of embayed beaches and a clear coastal progradation was found at the southern (downwind) coastal sectors. Coastal evolution has been dominated by sediment budget and the results put in evidence the sedimentary connections between the beaches across the island, either through bypass and overpass processes. Findings show that understanding coastal evolution at low‐lying islands should be supported on island‐scale observations, being the only scale capable to capture the sedimentary connections between beach systems, that often control coastal evolution. © 2019 John Wiley & Sons, Ltd. © 2019 John Wiley & Sons, Ltd. A sustainable society will have to largely refrain from the use of fossil carbon deposits. In such a regime, renewable electricity can be harvested as a primary source of energy. However, as for the synthesis of carbon‐based materials from bulk chemicals, an alternative is required. A sustainable approach towards this is the synthesis of commodity chemicals from CO2, water and sunlight. Multiple paths to achieve this have been designed and tested in the domains of chemistry and biology. In the latter, the use of both chemotrophic and phototrophic organisms has been advocated. 'Direct conversion' of CO2 and H2O, catalyzed by an oxyphototroph, has excellent prospects to become the most economically competitive of these transformations, because of the relative ease of scale‐up of this process. Significantly, for a wide range of energy and commodity products, a proof of principle via engineering of the corresponding production organism has been provided. In the optimization of a cyanobacterial production organism, a wide range of aspects has to be addressed. Of these, here we will put our focus on: (1) optimizing the (carbon) flux to the desired product; (2) increasing the genetic stability of the producing organism and (3) maximizing its energy conversion efficiency. Significant advances have been made on all these three aspects during the past 2 years and these will be discussed: (1) increasing the carbon partitioning to >50%; (2) aligning product formation with the growth of the cells and (3) expanding the photosynthetically active radiation region for oxygenic photosynthesis. Shape-Memory Alloys (SMAs) are a class of metal materials that exhibit two outstanding properties, the superelastic and the shape-memory effects. Taking advantage of these properties, several SMA actuator wires and plates have been proposed over the last years in robotic, automotive, and biomedical engineering. In this paper, the feasibility of a novel design concept of SMA-reinforced laminated glass panels is proposed, based on an adaptive embedded system built up of SMA wires. Glass panels used as cladding walls in facades have in fact typically high size-to-thickness ratios, hence, major restrictions in design are represented by prevention of glass failure and large deflections. Log-normal is a very common propagation model used to estimate the path loss introduced by a given environment. However, in most cases the parameters of the model are antenna specific. In this work, it is shown that the path loss exponent is highly dependent on the antenna pair used in the communication system. Large errors are introduced in the prediction of the path loss if the antennas are different from those used to obtain the parameters of the propagation model. An improved model is developed for the 2.4 GHz industrial, scientific and medical band to estimate the path loss exponent for different antenna directivities knowing the path loss exponent of a reference antenna. Protein investment costs are considered a major driver for the choice of alternative metabolic strategies. We tested this premise in L actococcus lactis, a bacterium that exhibits a distinct, anaerobic version of the bacterial Crabtree/ Warburg effect; with increasing growth rates it shifts from a high yield metabolic mode [mixed-acid fermentation; 3 adenosine triphosphate ( ATP) per glucose] to a low yield metabolic mode (homolactic fermentation; 2 ATP per glucose). We studied growth rate-dependent relative transcription and protein ratios, enzyme activities, and fluxes of L . lactis in glucose-limited chemostats, providing a high-quality and comprehensive data set. A three- to fourfold higher growth rate rerouted metabolism from acetate to lactate as the main fermentation product. However, we observed hardly any changes in transcription, protein levels and enzyme activities. Even levels of ribosomal proteins, constituting a major investment in cellular machinery, changed only slightly. Thus, contrary to the original hypothesis, central metabolism in this organism appears to be hardly regulated at the level of gene expression, but rather at the metabolic level. We conclude that L . lactis is either poorly adapted to growth at low and constant glucose concentrations, or that protein costs play a less important role in fitness than hitherto assumed. ZigBee/IEEE 802.15.4 is one of the most used standards for low‐power applications. However, full function devices must be always active to route data in a mesh network. The objective of this work is to implement a sleeping technique at the application layer that enables sleep mode for all nodes of a ZigBee network. A time synchronisation mechanism to deal with the clock drift of the sensor nodes was developed. The technique also enables the recovery of lost messages. A large network is organised into smaller groups to reduce latency and packet collisions. The active interval of each node is dynamically adapted to the network operation to optimise the energy consumption. The proposed technique was applied to a real testbed and the increase in energy efficiency was evaluated. The results demonstrated energy savings of about 95% for networks containing up to 20 nodes per group and wake up periods longer than 2 min. [ABSTRACT FROM AUTHOR]

This article offers a review of research and policy on climate change in Portugal and is organized into three main themes: scientific knowledge and assessment of climate change; policy analysis and evaluation; and public engagement. Modern scientific research on meteorology and climatology started in Portugal in the 1950s and a strong community of researchers in climate science, vulnerabilities, impacts, and adaptation has since developed, particularly in the last decade. Nevertheless, there are still many gaps in research, especially regarding the economic costs of climate change in Portugal and costs and benefits of adaptation. Governmental policies with a strong emphasis on mitigation were introduced at the end of the 1990s. As greenhouse gas emissions continued to rise beyond its Kyoto target for 2012, the country had to resort to the Kyoto Flexibility Mechanisms in order to comply. Climate change adaptation policies were introduced in 2010 but are far from being fully implemented. Regarding public engagement with climate change, high levels of concern contrast with limited understanding and rather weak behavioral dispositions to address climate change. Citizens display a heavy reliance on the media as sources of information, which are dominated by a techno-managerial discourse mainly focused on the global level. The final part of the article identifies research gaps and outlines a research agenda. Connections between policy and research are also discussed. Approximate Bayesian computation (ABC) substitutes simulation for analytic models in Bayesian inference. Simulating evolutionary scenarios under Kimura’s stepping stone model (KSS) might therefore allow inference over spatial genetic process where analytical results are difficult to obtain. ABC first creates a reference set of simulations and would proceed by comparing summary statistics over KSS simulations to summary statistics from localities sampled in the field, but: comparison of which localities and stepping stones? Identical stepping stones can be arranged so two localities fall in the same stepping stone, nearest or diagonal neighbours, or without contact. None is intrinsically correct, yet some choice must be made and this affects inference. We explore a Bayesian strategy for mapping field observations onto discrete stepping stones. We make Sundial, for projecting field data onto the plane, available. We generalize KSS over regular tilings of the plane. We show Bayesian averaging over the mapping between a continuous field area and discrete stepping stones improves the fit between KSS and isolation by distance expectations. We make Tiler Durden available for carrying out this Bayesian averaging. We describe a novel parameterization of KSS based on Wright’s neighbourhood size, placing an upper bound on the geographic area represented by a stepping stone and make it available as m Vector. We generalize spatial coalescence recursions to continuous and discrete space cases and use these to numerically solve for KSS coalescence previously examined only using simulation. We developed a spatially explicit model of a bioinvasion and used an approximate Bayesian computation (ABC) framework to make various inferences from a combination of genetic (microsatellite genotypes), historical (first observation dates) and geographical (spatial coordinates of introduction and sampled sites) information. Our method aims to discriminate between alternative introduction scenarios and to estimate posterior densities of demographically relevant parameters of the invasive process. The performance of our landscape-ABC method is assessed using simulated data sets differing in their information content (genetic and/or historical data). We apply our methodology to the recent introduction and spatial expansion of the cane toad, Bufo marinus, in northern Australia. We find that, at least in the context of cane toad invasion, historical data are more informative than genetic data for discriminating between introduction scenarios. However, the combination of historical and genetic data provides the most accurate estimates of demographic parameters. For the cane toad, we find some evidence for a strong bottleneck prior to introduction, a small initial number of founder individuals (about 15), a large population growth rate (about 400% per generation), a standard deviation of dispersal distance of 19 km per generation and a high invasion speed at equilibrium (50 km per year). Our approach strengthens the application of the ABC method to the field of bioinvasion by allowing statistical inferences to be made on the introduction and the spatial expansion dynamics of invasive species using a combination of various relevant sources of information. Purpose: Acute anterior uveitis (AAU) is one of the most common extra‐articular manifestations of spondyloarthritis (SpA), causing significant burden in quality of life (QoL). Golimumab (GLM) is a tumor necrosis factor‐inhibitor proven to be effective and safe in SpA. The GO‐EASY Study provided evidence that GLM decreases the AAU occurrence rate in SpA. Our aim is to study the impact of GLM in the change of vision‐related (VR) QoL in subjects with SpA and past or current AAU. Methods: Ongoing prospective multicentre observational study (including 8 centres in Portugal) of SpA patients with history of AAU treated with GLM followed‐up for 12 months. We intend to recruit in total 30 patients and we report herein the 6 months outcomes for the first 9 patients enrolled. The occurrence of AAU was assessed in the 2 years before GLM treatment was started and the first 6 months of follow‐up and calculated for the period at risk for a new AAU. VR QoL was assessed with the self‐administered National Eye Institute Visual Functioning Questionnaire‐25 (NEI VFQ‐25). Adverse events were noted. Results: Nine patients (66.7% female, 100% TNFi‐naive, mean age 46.1 ± 14.4 years (range 22–65)) have completed 6 months of follow‐up. Three patients (33%) were also under oral methotrexate. The mean number of AAU flares in the 2 years preceding the start of GLM was 2.2 ± 1.3 (1–4). During the first 6 months of GLM treatment, the AAU incidence rate was reduced from 1.54 to 0.11 per 100 patient‐years (incidence ratio‐ratio 13.46, 95% CI 2.15–558.00; p < 0.01). At baseline and at 24 weeks after GLM onset, the mean overall index NEI VFQ‐25 total score was 70.2 and 83.2, respectively; improvement in the NEI VFQ‐25 total score was +13 ± 18.2. No significant or new adverse events occurred. Conclusions: Preliminary data from the GO‐VISION study suggest that GLM is safe and effective in patients with SpA and history of AAU, being able to reduce the AAU occurrence rate and potentially increasing VR QoL. Abstract: We have reported previously on the ability of Lactobacillus reuteri to produce a compound with vitamin activity. Here we report on the chemical characterisation of this corrinoid-like molecule. High performance liquid chromatography coupled to an ultraviolet diode array detector, mass spectrometry and nuclear magnetic resonance spectroscopy has enabled us to identify the compound as Co-[-(7-adenyl)]-Co-cyanocobamide or pseudovitamin . This molecule differs from cobalamin in the -ligand, where it has adenine instead of 5,6-dimethylbenzimidazole bound in a -glycosidic linkage to C-1 of ribose. L. reuteri is the first lactic acid bacterium in which the production of a cobalamin-like molecule has been identified and the first microorganism reported to produce exclusively pseudo-. We use linked data for 1460 workplaces and 19,853 employees from the Workplace Employee Relations Survey 1998 to analyse the incidence and duration of employee training in Britain. We find training to be positively associated with having a recognized vocational qualification and current union membership. However, being non-white, having shorter current-job tenure and part-time or fixed-term employment statuses are all associated with less training. Furthermore, in line with recent non-competitive training models, higher levels of wage compression (measured in absolute or relative terms) are positively related to training. Summary. We report a case of Sezary syndrome with two abnormal CD4+ T-cell populations detected in the peripheral blood by flow cytometry immunophenotyping and DNA cell content, suggesting a biclonal T-cell lymphoproliferative disorder. Despite these findings, molecular analysis of the T-cell receptor genes was consistent with a monoclonal T-cell proliferation, supporting the existence of intraclonal diversity rather than a true biclonal disease. The patient achieved a transient response with 2-deoxycoformycin, with a selective decrease of the larger/hyperploid T-cell population; later on, an increased representation of this T-cell population was observed concomitantly with clinical relapse. The use of dominant markers such as amplified fragment length polymorphism (AFLP) for population genetics analyses is often impeded by the lack of appropriate computer programs and rarely motivated by objective considerations. The point of the present note is twofold: (i) we describe how the computer program Geneland designed to infer population structure has been adapted to deal with dominant markers; and (ii) we use Geneland for numerical comparison of dominant and codominant markers to perform clustering. AFLP markers lead to less accurate results than bi-allelic codominant markers such as single nucleotide polymorphisms (SNP) markers but this difference becomes negligible for data sets of common size (number of individuals n≥100, number of markers L≥200). The article presents a case report of a one-year-old girl with a three day history of asthenia, dark urine, and skin pallor. It mentions that severe anaemia with mild reticulocytosis, total bilirubin, and elevated levels of serum were revealed by laboratory tests. It states that the evidence of intravascular haemolysis and C3d positivity of the Coomb's tests resulted for the diagnosis of paroxysmal cold haemoglobinuria.

We use linked data for 13 991 employees and 1494 workplaces to analyse the incidence of employer-provided training in Australia. We find potential experience, current job tenure, low education levels, skilled vocational training and part-time or fixed-term employment status are all associated with a lower probability of recent training. In contrast to studies for other countries, we find no evidence of discrimination on the basis of demographic characteristics in the provision of this job-related training. Finally, and in support of recent non-competitive training models, higher levels of wage compression are found to be positively related to a greater incidence of employee training.
