Objective To examine the association of viruses with acute febrile respiratory disease in horses. Design Nasal swab and serum samples were collected from 20 horses with acute febrile upper respiratory disease that was clinically assessed to have a viral origin. Methods Each of the samples was inoculated onto equine fetal kidney, RK13 and Vero cell cultures, and viral nucleic acid was extracted for polymerase chain reaction (PCR) or reverse transcription PCR. PCR primers were designed to amplify nucleic acid from viruses known to cause or be associated with acute febrile respiratory disease in horses in Australia. A type specific ELISA was used to measure equine herpesvirus (EHV1 and EHV4) antibody, and serum neutralisation assays were used to measure equine rhinitis A virus (ERAV) and equine rhinitis B virus 1 and 2 (ERBV1 and ERBV2) antibody titres in serum samples. Results Virus was isolated from 4 of 20 nasal swab samples. There were three isolations of EHV4 and one of ERBV2. By PCR, virus was identified in the nasal swab samples of 12 of the 20 horses. Of those that were positive, 17 viruses were detected as follows: one triple positive (EHV4, EHV2, and EHV5), three double positives (EHV4, ERBV and EHV2, EHV5 in two horses) and eight single positives (EHV4 in two horses, EHV2 in one horse, EHV5 in six horses and ERBV in six horses). Conclusion By virus isolation and PCR, 17 viruses were identified in nasal swab samples from 12 of 20 horses that had acute febrile respiratory disease consistent with a diagnosis of virus infection. Specific-pathogen-free cats were immunized with a betapropiolactone-inactivated feline herpesvirus 1 (FHV1) vaccine, with or without alhydrogel adjuvant, and antibody and cell mediated immune responses were measured. Maximal antibody titres were obtained in 2 cats vaccinated with 2 doses of adjuvanted vaccine. Antibody persisted at low levels for at least 10 months in 3 of 4 vaccinated cats and there was a strong anamnestic antibody response in the 4 vaccinated cats following FHV1 challenge 10 months after vaccination. FHV1 antigen specific lymphocyte blastogenesis (LB), was detected following vaccination and challenge. A cell-mediated plaque reduction (CMPR) assay was developed using both autologous kitten kidney (AKK) cells, prepared by unilaterally nephrectomizing cats, and a feline embryo (FEmb) cell line. Leukocyte suspensions enriched for T lymphocytes mediated a major histocompatabily (MHC)-restricted, cytotoxic T (Tc) lymphocyte response. Buffy coat cells, with added plasma, mediated and MHC - unrestricted CMPR response which was interpreted as an antibody dependent cell mediated cytotoxicity (ADCC). Both null cells, presumably a feline killer (K) cell, and neutrophils were effector cells in ADCC. In 51Cr release cell mediated cytotoxicity (CMC) assay leukocytes from vaccinated cats were cytotoxic for both AKK- and FEmb-FHV1 infected target cell suspensions when tested 10 months post-vaccination and 3 days before FHV1 challenge. CMC was augmented in the vaccinated cats and a significant primary response was observed in an unvaccinated control cat on day 4 post challenge. The levels of cytotoxicity were higher in AKK than in FEmb cells and effector cells enriched for T lymphocytes were more cytotoxic for AKK cells indicative of a MHC-restricted Tc lymphocyte response. Zusammenfassung Antikörper- und zellvermittelte Reaktionen gegen Katzenherpesvirus 1 nach Impfung mit inaktivierter Vakzine und Testinfektion Spf-Katzen wurden mit betapropiolakton-inaktivierter Katzenherpesvirus-1(FHV 1)-Vakzine mit oder ohne Alhydrogel geimpft. Anschließend wurde die antikörper- und zellvermittelte Immunreaktion gemessen. Die maximalen Antikörpertiter hatten zwei Katzen, die mit 2 Dosen der Adjuvans-Vakzine geimpft worden waren. In drei von vier geimpften Katzen konnten geringe Antikörpertiter noch mindestens zehn Monate nach der Impfung festgestellt werden. 10 Monate nach der Impfung löste eine Belastung mit FHV 1 eine starke anamnestische Antikörperreaktion in den vier geimpften Katzen aus. FHV 1-antigenspezifische Lymphozytenblastogenese (LB) konnte bei geimpften und belasteten Katzen nachgewiesen werden. Ein zellvermittelter Plaque-Reduktionstest (CMPR) wurde mit autologen, von einseitig nephrektomierten Katzen gewonnenen Welpennierenzellen (AKK), und einer Katzenembryozellinie entwickelt. Leukozytensuspensionen, die mit T-Lymphozyten angereichert waren, vermittelten eine auf den MHC begrenzte zytotoxische T-Lymphozytenreaktion. In Plasma aufgeschwemmte 'Buffycoat' Zellen vermittelten einen MHC, der nicht auf die CMPR begrenzt blieb und als eine antikörperabhängige zellvermittelte Zytotoxizität interpretiert wurde. Sowohl Nullzellen, vermutlich eine Katzenwelpenkillerzelle, als auch Neutrophile waren als Effektorzellen in ADGG wirksam. Im Zytotoxizitätstest (CMC) mit 51Cr waren Leukozyten von geimpften Katzen sowohl für AKK- als auch für FEmb-FHVY-infizierte Targetzellensuspensionen zytotoxisch, und zwar 10 Monate nach der Impfung und 3 Tage vor der FHV 1 Belastung. CMC trat bei geimpften Katzen vermehrt auf, und eine signifikante Primärreaktion konnte, 4 Tage nach der Belastung, in einer ungeimpften Kontrollkatze festgestellt werden. Die Zytotoxizitätsreaktion war in AKK-Zellen stärker als in FEmb-Zellen. Betapropriolactone-inactivated feline calicivirus (FCV) vaccine stimulated homotypic virus neutralizing antibody in 4 vaccinated cats and heterotypic neutralizing antibody in 3 of 4 vaccinates. Fluctuation in the stimulation index (SI) of concanavalin A (Con-A)-induced lymphocyte blastogenesis (LB) was observed following vaccination. FCV antigen (Ag)-induced LB with SI>2 was detected in the 4 vaccinated cats by day 3 following first vaccination and an anamnestic LB response was evident in 3 of the 4 vaccinated cats following the second vaccination. Following FCV challenge 6 months post-vaccination there was a fall in the SI that was more obvious in washed blood cultures, while increased SI were evident in whole blood cultures later after challenge. Following challenge, anamnestic LB-responses occurred in the 3 vaccinated cats on day 7 post-challenge while a primary LB-response was detected on day 10 in an unvaccinated control cat. The onset of FCV antibody and LB responses correlated with recovery from FCV disease. Reduction in plaque number and diameter in autologous kitten kidney (AKK) and feline embryo (FEmb) cells infected with FCV was observed using peripheral blood leukocytes as effector cells. Leukocytes enriched for T lymphocytes mediated a major histocompatability complex (MHC)-restricted response since it was detected in AKK but not in FEmb cell monolayers. Separated mononuclear cells (SMC) mediated an MHC-unrestricted response in that it was detected in both AKK and FEmb target cells. Immunity to FCV was challenged 6 months after second vaccination. The unvaccinated control cat developed clinical signs of FCV disease whereas clinical disease was less severe and of shorter duration in the vaccinated cats. FCV persisted in the unvaccinated control cat whereas vaccinated cats excreted virus for a shorter duration. Following challenge, a 51Cr release cell mediated cytotoxicity (CMC) assay showed that SMC from vaccinated cats mediated a transient weak direct i. e. antibody independent, response whereas SMC from the unvaccinated control cat mediated a strong indirect, i. e. antibody dependent, CMC. Eine Katzenkalizivirusantigen (Ag)-induzierte Lymphozytenblastogenese (LB) mit SI>2 ließ sich bis zum 3. Tag nach der ersten Impfung in den vier geimpften Katzen nachweisen. In 3 von 4 geimpften Katzen war nach der zweiten Impfung eine anamnestische Reaktion nachweisbar. Nach Belastung mit FCV (6 Monate nach der Impfung) sank der SI-Wert vor allem in gewaschenen Blutkulturen zunächst ab, während sich die SI-Werte in Ganzblutkulturen erhöhten. Am siebten Tag nach der Belastung kam es zu anamnestischen LB-Reaktionen in den 3 geimpften Katzen, während bei einer ungeimpften Kontrollkatze eine primäre LB-Reaktion am 10. Tag festgestellt werden konnte. Die Freisetzung von FCV-Antikörpern und die LB-Reaktionen korrelierten mit der Genesung von der FCV-Krankheit. Mit Hilfe von peripheren Blutleukozyten als Effektorzellen konnte in mit FCV infizierten autologen Katzenwelpennieren- und Katzenembryozellen eine Verminderung der Plaquezahlen und des Durchmessers der Plaques beobachtet werden. Leukozyten, die mit T-Lymphozyten angereichert waren, vermittelten eine auf den 'major histocompatibility-complex' (MHC) beschränkte Reaktion (die Reaktion konnte in AKK, aber nicht in FEmb Zellenmonoschichten nachgewiesen werden). Isolierte mononukleäre Zellen vermittelten eine MHC-unabhängige Reaktion, da sie sowohl in AKK und FEmb Targetzellen nachzuweisen war. Die Immunität gegen FCV wurde auch 6 Monate nach der zweiten Impfung getestet. Die ungeimpfte Kontrollkatze zeigte die klinischen Symptome einer FCV-Krankheit, während bei den geimpften Katzen die klinischen Erscheinungen weniger schwer und von kürzerer Dauer waren. FCV persistierte bei der ungeimpften Katze, während die geimpften Katzen das Virus nur kurz ausschieden. Eine 51Cr-freisetzende zellvermittelte Zytotoxizität (CMC) zeigte nach der Belastung, daß SMC vorübergehend bei geimpften Katzen eine schwache, unmittelbare, d. h. antikörperunabhängige Reaktion auslösen, im Unterschied zu den SMC bei der ungeimpften Kontrollkatze, die eine heftige, indirekte, d. h. antikörperabhängige, CMC auslösten. Serum haemagglutination inhibiting and plaque reduction neutralizing antibodies were detected in cats given 2 doses of an adjuvanted inactivated feline panleukopenia virus vaccine. Plaque reduction neutralizing antibody titres were approximately 3 to 1126-fold higher than haemagglutination inhibiting antibody titres. Suppression of in vitro concanavalin A-induced lymphocyte blastogenesis was observed following vaccination; the stimulation indices of both whole and washed blood cultures were approximately equally suppressed on day 3 to day 24 or 28. In vitro feline panleukopenia virus antigen-induced lymphocyte blastogenesis with stimulation indices > 2 were detected as early as day 3 following vaccination. A secondary feline panleukopenia virus antigen-induced lymphocyte blastogenesis was detected in the washed blood cultures but not in the whole blood cultures of the vaccinated cats following a second vaccination. Cell-mediated cytotoxicity for feline panleukopenia virus infected target cells was detected with peripheral blood leukocytes from the vaccinated cats. Two populations of cytotoxic effector cells were detected: (a) Effector cells in a T lymphocyte-enriched preparation mediated a major histocompatibility complex-restricted cytotoxicity in autologous kitten kidney target cells, (b) A second effector cell in ficoll-diatrizoate separated mononuclear cells mediated a major histocompatibility complex-unrestricted cytotoxicity for a feline embryo cell line. Both effector cells were again detected in the peripheral blood of cats following a second vaccination. A natural cell-mediated cytotoxicity, distinctly different from the 2 immune specific cell-mediated cytotoxicities of the vaccinated cats was demonstrated throughout the experiments when peripheral blood leukocytes from an unvaccinated control cat were used as effector cells. Zusammenfassung Antikörper und zellvermittelte Immunreaktionen gegen eine inaktivierte Katzenpanleukopenievirus-Vakzine Hämagglutinationshemmende und Plaque-reduzierende, neutralisierende Serumantikörper wurden in Katzen nachgewiesen, die mit je 2 Dosen inaktivierter Katzenpanleukopenievirus-Vakzine (mit Adjuvans) geimpft wurden. Die Plaque-reduzierenden, neutralisierenden Antikörpertiter waren etwa 3-1126fach höher als die hämagglutinationshemmenden Antikörpertiter. Eine in vitro mit Concanavallin A-induzierte Lymphozytenblastogenese konnte mit der Impfung gehemmt werden; die Stimulationsindizes waren vom 3. bis zum 24. oder 28. Nach einer zweiten Impfung wurde nur in den gewaschenen Blutkulturen, aber nicht in den unbehandelten Ganzblutkulturen der geimpften Katzen eine sekundäre katzenpanleukopenievirus-antigen-induzierte Lymphozytenblastogenese festgestellt. Mit peripheren Blutleukozyten von geimpften Katzen konnte eine zellvermittelte Zytotoxizität für mit Katzenpanleukopenievirus infizierte Targetzellen nachgewiesen werden. Zwei Populationen mit zytotoxischen Effektorzellen wurden nachgewiesen: a) Effektorzellen in einer mit T-Lymphozyten angereicherten Suspension vermittelten einen MHC, der auf eine Zytotoxizität in autologen Katzenwelpennierentargetzellen begrenzt war. b) Eine zweite Effektorzelle aus mononukleären Zellen, die mit Ficoll-Diatrizoate getrennt wurden, vermittelte einen MHC mit uneingeschränkter Zytotoxizität für eine Katzenembryozellinie. Beide Effektorzellen wurden auch nach einer zweiten Impfung im peripheren Blut von Katzen festgestellt. Eine natürliche zellvermittelte Zytotoxizität, die sich aber deutlich von den 2 immunspezifischen zellvermittelten Zytotoxizitäten der geimpften Katzen unterschied, wurde während der gesamten Versuche nachgewiesen, wenn periphere Blutleukozyten von einer ungeimpften Kontrollkatze als Effektorzellen verwendet wurden. We describe an outbreak of infectious pustular vulvovaginitis caused by Caprine herpesvirus 1 (CpHV1) in a group of approximately 200, 8 month old virgin does that were imported to Victoria from New Zealand. CpHV1 was isolated in cell cultures from vaginal swabs from three of three affected does but not from two bucks that had been with the does. The identity of the virus as a herpesvirus was confirmed by negative stain electron microscopy. Restriction endonuclease DNA fingerprint analysis showed that the DNA fingerprints were similar, but not identical, to previously described CpHV1 isolates made in New Zealand, New South Wales, and in other parts of the world. Acute and convalescent phase sera from selected does supported the diagnosis of CpHV1 infection. It is most likely that the disease was initiated by reactivation of latent virus in at least one of four bucks that served the does, since each was positive for CpHV neutralising antibody when first tested. This is the first report of CpHV infectious pustular vulvovaginitis in goats in Victoria and to our knowledge appears to be one of the largest outbreaks recorded anywhere. [ABSTRACT FROM AUTHOR]
