Natural ester dielectric fluids are being used as alternatives for mineral insulating oils in oil-filled power apparatuses. Although partial discharges (PDs) are recognised as one of the main causes of degradation of dielectrics used in power equipment, their characteristic voltages appear to be temperature-dependent. In this contribution, the pre-breakdown and breakdown behaviour of two natural ester fluids, extracted from canola oil (NE1) and soybean oil (NE2) is reported. To assess the effectiveness of the temperature dependence, −5, 0, and + 20°C have been identified as test temperatures. It was found that the different pre-breakdown and breakdown characteristic voltages increase with increasing temperature for both types of esters. The delay time is always longer for canola oil when compared to soybean oil regardless of the test temperature. The analysis of the average propagation velocity of NE1 and NE2 indicates that PDs propagate faster in NE2 than in NE1. The field strength at the tip of the needle electrode during the inception of the streamers leading to breakdown indicates that streamers in both oils are propagating in the slow mode. In this Letter, a demodulation based pre-filtering technique is proposed for fast detection of the single-phase grid variables. The estimation of grid voltage parameters is achieved through a Type-I synchronous reference frame phase-locked loop (PLL). The modified bandpass filter in the pre-filtering stage ensure a better rejection of the DC-offset and harmonics. Thus, the grid variables are estimated rapidly with the help of a simple proportional controller. Consequently, in-loop filtering burden and the effort required to tune the PLL is minimal. However, steady errors are witnessed in the fundamental amplitude and the phase information which are mitigated by a curve fitting technique. Importantly, the proposed PLL scheme has improved the dynamic performance under off-nominal frequency conditions with immunity against grid disturbances. Study of alternate fluids for transformer and their combination with traditional ones is an interesting area of research. The intent of this work is to investigate the thermal ageing performance of ester oils (synthetic and natural), and mixed oil vis‐à‐vis mineral oil. All the samples were thermally stressed at 110, 140, 160, and 185°C for 2 weeks each. Dissolved decay contents in oil and linear thermal expansion coefficients of solid insulation have been studied using ultraviolet visible infrared spectroscopy and dilatometry analysis, respectively, as per ASTM standards. Change in functional groups of the insulation oils with ageing have been understood by Fourier transform infrared spectroscopy analysis. Additionally, diffusion of oil–moisture mixture into paper with ageing and the corresponding effect on dielectric strength of paper has been also examined using ASTM standards. It is observed that the use of synthetic ester (SE) and addition of SE to mineral insulating oil lead to improved performance of oil–paper insulation system with retarding the rate of degradation of the insulation system. This study reports experimental study of cellulose performance in mixture of mineral and synthetic ester (SE) oils. Solid insulation impregnated with mineral oil (MO) and mixture oil has been subjected to accelerated thermal stresses to understand the deterioration rate of cellulose. Diffusion of oil–moisture mixture into paper and corresponding variation in dielectric strength of paper has been examined. Compatibility of solid insulation with MO and mixture oil is analysed by thermal expansion coefficients, X‐ray diffraction analysis and dissolved decay products. Furthermore, chemical functional groups present in aged oils are identified by Fourier transform infrared spectroscopy analysis. It is observed that the performance of cellulose insulation is improved with addition of SE to MO. This is achieved by higher water solubility of esters, which enables higher moisture consumption during hydrolysis and keeps cellulose dry. [ABSTRACT FROM AUTHOR]

Background and Aim The usefulness of carcinoembryonic antigen ( CEA) in the diagnosis and prognosis of colorectal cancer ( CRC) is unclear. The aim was to analyze changes in the expression of CEA during CRC progression and metastasis, so as to determine the influence of tumor metastatic organ on the CEA expression by CRC cells. Methods The human biopsies of adenocarcinomas in colon and CRC liver and lung metastases were analyzed by immunohistochemistry for the expression of CEA. Expression of E-cadherin and β-catenin was also analyzed to localize the CRC neoplastic glands in metastatic tissues. Results The CRC neoplastic glands in colon and liver expressed significantly higher amount of CEA compared with crypts in normal colon. In contrast, CRC neoplastic glands formed in lung expressed low CEA level. However, CEA expression was high in areas of tumor necrosis in lung. E-cadherin and β-catenin were cell membrane-bound in normal crypts and CRC neoplastic glands in colon and liver. Although these two proteins were also cell membrane-bound in a majority of CRC neoplastic glands in lungs, a significant proportion of these expressed β-catenin in the nucleus, which lacked either E-cadherin or β-catenin at the cell membrane. Conclusion Our findings indicate that lung microenvironment is unique in that it suppresses the expression of CEA by CRC cells forming neoplastic glands. In addition, lung microenvironment promotes nuclear localization of β-catenin, suggesting that the Wnt signaling pathway is relatively active highly in CRC metastasized to lung, when compared with liver or colon. Bioflavonoids are of considerable interest to human health as these serve as antioxidant and anticancer agents. Although epidemiological and experimental studies suggest that luteolin, a natural bioflavonoid, exhibits chemopreventive properties, its effectiveness as an antiproliferative agent against multidrug resistant (MDR) cancers is unclear. Thus, we assessed the antiproliferative effects of luteolin and associated molecular mechanisms using two MDR cancer cell lines that express high levels of P-glycoprotein and ABCG2. In this article, we demonstrate that luteolin induces apoptosis in P-glycoprotein- and ABCG2-expressing MDR cancer cells without affecting the transport functions of these drug transporters. Analysis of various proliferative signaling pathways indicated that luteolin-induced apoptosis involves reactive oxygen species generation, DNA damage, activation of ATR → Chk2 → p53 signaling pathway, inhibition of NF-kB signaling pathway, activation of p38 pathway and depletion of antiapoptotic proteins. Importantly, use of luteolin in these analyses also identified specific molecular characteristics of NCI-ADR/RES and MCF-7/MitoR cells that highlight their different tissue origins. These results suggest that luteolin possesses therapeutic potential to control the proliferation of MDR cancers without affecting the physiological function of drug transporters in the body tissues. Development of colorectal cancer (CRC) involves a series of genetic alterations with altered expression of proteins and cell signaling pathways. Here, we identified that galectin-4 (gal-4), a marker of differentiation, was down-regulated in CRC. The goal of this work was to determine the function of gal-4 in CRC. Toward this goal, the human colon biopsies and tissue microarrays containing a gradient of pathology were analyzed for gal-4 expression by immunohistochemistry. Cell proliferation, migration, motility, forced expression, knockdown, cell cycle and apoptosis assays were used to characterize gal-4 function. Immunohistochemistry identified that gal-4 expression was significantly down-regulated in adenomas and was essentially absent in invasive carcinomas. Forced expression of gal-4 in gal-4 −ve cells induced cell cycle arrest and retarded cell migration and motility. Further, gal-4 sensitized the cells to camptothecin-induced apoptosis. Gal-4 knockdown resulted in increased cell proliferation, migration and motility. Gal-4 was found to be associated with Wnt signaling proteins. Finally, gal-4 expression led to down-regulation of Wnt signaling target genes. This study demonstrates that loss of gal-4 is a common and specific event in CRC. This study also shows that gal-4 exhibits tumor suppressive effects in CRC cells in vitro. Through its ability to interact with and down-regulate the functions of Wnt signaling pathway, gal-4 reveals a new dimension in the control of the Wnt signaling pathway. Aims: Leiomyosarcomas (LMS) are malignant neoplasms composed of cells that exhibit distinct smooth muscle differentiation. The molecular and cytogenetic features of LMS are complex and no consistent aberrations have been reported to date. Mitogen inducible gene-2 (Mig-2), kindlin and migfilin are recently identified cell–matrix adhesion proteins. The aim was to determine the expression and distribution of these proteins in human smooth muscle tumours of somatic soft tissue. Methods and results: Immunohistochemistry was performed on a human LMS tissue microarray and on sections of human leiomyomas (LM) and normal smooth muscle. Migfilin was barely detectable in normal smooth muscle cells, whereas increased levels of migfilin were observed in the majority of LM and LMS. Furthermore, the cytoplasmic level of migfilin was strongly associated with higher tumour grades. Additionally, the cytoplasmic levels of migfilin and Mig-2 were correlated with each other, suggesting an association between the two in the cytoplasm. Kindlin was expressed in normal smooth muscle, LM and LMS, and its level did not correlate with tumour grade. Conclusions: Our results suggest a role for cytoplasmic migfilin in the progression of LMS and identify cytoplasmic migfilin as a potentially important biological marker for human LMS progression. We propose a molecular model for phospholipid membrane lysis by the ubiquitous plant toxins called thionins. Membrane lysis constitutes the first major effect exerted by these toxins that initiates a cascade of cytoplasmic events leading to cell death. X-ray crystallography, solution nuclear magnetic resonance (NMR) studies, small angle X-ray scattering and fluorescence spectroscopy provide evidence for the mechanism of membrane lysis. In the crystal structures of two thionins in the family,α1- andβ-purothionins (MW: approximately 4.8 kDa), a phosphate ion and a glycerol molecule are modeled bound to the protein.31P NMR experiments on the desalted toxins confirm phosphate-ion binding in solution. Evidence also comes from phospholipid partition experiments with radiolabeled toxins and with fluorescent phospholipids. This data permit a model of the phospholipid–protein complex to be built. Further, NMR experiments, one-dimensional (1D)- and two-dimensional (2D)-total correlation spectroscopy (TOCSY), carried out on the model compounds glycerol-3-phosphate (G3P) and short chain phospholipids, supported the predicted mode of phospholipid binding. The toxins’ high positive charge, which renders them extremely soluble (>300 mg/mL), and the phospholipid-binding specificity suggest the toxin–membrane interaction is mediated by binding to patches of negatively charged phospholipids [phosphatidic acid (PA) or phosphatidyl serine (PS)] and their subsequent withdrawal. The formation of proteolipid complexes causes solubilization of the membrane and its lysis. The immune mechanisms operating on the sheath and the cuticle of the Brugia pahangi microfilariae have been studied in vitro. The intact and exsheathed parasites were used for this purpose with serum from albino rats immunized with sonicated homogenates of the parasites as the source of anti-bodies. The IgG component of the serum was found to promote adherence of rat leucocytes and death of the sheathed and exsheathed parasites in presence of fresh normal serum. EDTA and EGTA abolished the adherence activity suggesting the involvement of complement components via the classical pathway. Both macrophages and neutrophils participate in this reaction. Eosinophils exhibit marginal activity only on exsheathed parasites. The intensity of adherence and killing by neutrophils in the presence of immune serum was greater with the exsheathed microfilariae than with the sheathed ones. [ABSTRACT FROM AUTHOR]

The multimammate rat Mastomys natalensis, when infected with the filarial parasite Acanthocheilonema viteae, develops amicrofilaraemia. Worm recovery and the duration and intensity of microfilaraemia were analysed and related to the humoral and cellular immune responses of the host by using an antibody-dependent cell-mediated cytotoxicity (ADCC) assay towards microfilariae (Mf). Mf were detected in the peripheral blood at 7 weeks post-infection (p.i,), reaching maximum levels by 20 weeks p.i., and then gradually decreasing to undetectable levels during the next 36 weeks. The cytotoxic antibodies appeared around 15-18 weeks p.i., and the serum at 36 weeks pi, induced 70% cytotoxicity to the Mf in vitroin the presence of host cells. The IgM fraction of the immune serum from a microfilaraemic Mastomys promoted ADCC to Mf both in vitro and in vivo. Macrophages were more potent in inducing cytotoxic effect than eosinophils and neutrophils. Platelets were ineffective in killing the Mf in the presence of immune serum. IgM antibody cleared the circulating Mf from the blood when given passively to infected Mastomys. The average recovery of adult worms was about 20% of the inoculated larvae. No live females could be recovered 56 weeks p.i. Thus protective immune responses built up over an extended period of time are elicited against the Mf and perhaps even to adults in Mastomys infected with A. viteae. [ABSTRACT FROM AUTHOR]

The complement of fresh normal rat serum was activated by filarial eggs and microfilariae (mf). C3 was deposited on the surface of Litomosoides carinii, Brugia pahangi, Brugia malayi and Dipetalonema viteae as seen by immunofluorescence. Inira-uterine and in vitro-derived mf did not bind C3. In contrast, C3 bound to the blood-derived mf of B. pahangi and B. malayi as well as exsheathed mf of L. carinii and B. malay. Significant consumption of complement was observed with eggs of all filarial species, as well as sheathed mf of B. pahangi, B. malayi and exsheathed mf of L. carinii and B. malayi. These experiments indicated that complement was activated by filarial parasites via the alternative pathway. The bound complement promoted neutrophil-mediated adherence and cytotoxicity.
