Translocations are a widespread approach to conserve threatened populations. Given the rapid decline and genetic deterioration of many natural populations, translocations are expected to become even more common in the future. The success of translocations is, however, dependent on multiple context‐dependent factors, such as demographic and genetic status, habitat quality and animal behaviour. The Scandinavian arctic fox (Vulpes lagopus) exists in a small, fragmented population that is demographically vulnerable and exposed to inbreeding depression. In the early 2000 s, releases of arctic foxes from the Norwegian Captive Breeding Programme were initiated with the purpose of reintroducing populations to formerly inhabited areas and promoting connectivity. Since 2008/2009, 61 foxes have been released in Junkeren, Norway to re‐establish an unoccupied area. We used a combination of field observations and microsatellite genotyping from the release site and two neighbouring subpopulations to investigate (i) the probability of establishment and reproduction for released foxes at the release site and in neighbouring subpopulations, and (ii) the impact on litter size and genetic composition in the recipient populations. Results showed that 18% of the released foxes were established at the release site, or in neighbouring subpopulations and 11.5% reproduced successfully. The extent of post‐release dispersal into neighbouring subpopulations was also relatively high (11.5%). During the study period, the number of litters more than doubled in the subpopulations with released foxes contributing 29.5% to this increase, but no clear effect of immigration on litter size was found. There was a slight increase in genetic variation in one of the subpopulations, and a significant decline in genetic divergence between subpopulations. We conclude that despite extensive releases, demographic and genetic effects were highly context‐dependent. This study highlights the challenges of reinforcement programmes in small populations and reintroductions to unoccupied sites, especially for highly mobile species in a fragmented landscape. Nature‐based recreational and tourism activities can exert significant direct and indirect impacts on wildlife, through behavioral, physiological and distributional changes. Despite many studies demonstrating such changes, few attempts have been made to quantify the fitness consequences and evaluate their biological significance. Helagsfjällen in Sweden is a core area of the endangered Fennoscandian arctic fox Vulpes lagopus, and a popular area for recreational tourism. Some dens in the area experience daily disturbance from tourism during the summer season, while others are virtually undisturbed. We used a long‐term dataset (2008–2017) of 553 juveniles in 74 litters to investigate summer juvenile survival, which is an important fitness component for the arctic fox. We found that the mean juvenile survival rate increased from 0.56 at undisturbed dens to 0.83 at disturbed den during years of decreasing small‐rodent abundance, where predation on the arctic fox is presumed to be highest. We suggest that the increased survival could be mediated by a human activity‐induced predator refuge for the arctic foxes in close proximity of trails and mountain huts. Our study demonstrates a possible positive indirect effect of nature‐based tourism on wildlife and is one of a few studies attempting to quantify this impact. It highlights the importance of context for how animals are affected by disturbance. We also demonstrate that studying how the effects of tourism activity vary depending on the context could provide opportunities for identifying the mechanisms behind these effects, which can be an important link between scientific research and the management of wildlife and tourism activities. Objective: The formation of foam cells from monocyte-derived macrophages involves the uptake of modified lipoproteins by scavenger receptors. Antioxidants inhibit lipoprotein oxidation and may also modulate gene expression. We investigated the effect of the antioxidant N-acetylcysteine on the expression of the class A scavenger receptor (SR-A) types I and II in human macrophages.Design: Monocytes and macrophages from healthy blood donors and plaque-derived macrophages from patients undergoing carotid endarterectomy were used for experiments. SR-A mRNA was analysed with quantitative and semiquantitative reverse transcription-polymerase chain reaction, and ligand binding and uptake were assessed with 125I-labelled acetylated low-density lipoprotein (LDL).Results: Incubation of monocytes and monocyte-derived macrophages with N-acetylcysteine decreased both SR-A I and II mRNA expression. N-Acetylcysteine also reduced SR-A mRNA in lesion-derived cells. Binding and uptake of 125I-acetylated LDL was decreased after brief incubation with N-acetylcysteine. After longer periods of incubation with N-acetylcysteine we observed an increased degradation of lipoproteins.Conclusions: Our results imply that N-acetylcysteine leads to a decrease in SR-A mRNA and initially also to an attenuated uptake of modified lipoproteins. Hybridization between wild and captive-bred individuals is a serious conservation issue that requires measures to prevent negative effects. Such measures are, however, often considered controversial by the public, especially when concerning charismatic species. One of the threats to the critically endangered Fennoscandian arctic fox Alopex lagopus is hybridization with escaped farm foxes, conveying a risk of outbreeding depression through loss of local adaptations to the lemming cycle. In this study, we investigate the existence of escaped farm foxes among wild arctic foxes and whether hybridization has occurred in the wild. We analysed mitochondrial control region sequences and 10 microsatellite loci in samples from free-ranging foxes and compared them with reference samples of known farm foxes and true Fennoscandian arctic foxes. We identified the farm fox specific mitochondrial haplotype H9 in 25 out of 182 samples, 21 of which had been collected within or nearby the wild subpopulation on Hardangervidda in south-western Norway. Genetic analyses of museum specimens collected on Hardangervidda (1897–1975) suggested that farm fox genotypes have recently been introduced to the area. Principal component analysis as well as both model- and frequency-based analyses of microsatellite data imply that the free-ranging H9s were farm foxes rather than wild arctic foxes and that the entire Hardangervidda population consisted of farm foxes or putative hybrids. We strongly recommend removal of farm foxes and hybrids in the wild to prevent genetic pollution of the remaining wild subpopulations of threatened arctic foxes. [ABSTRACT FROM AUTHOR]

Comprehensive population parameter data are useful for assessing effective conservation actions. The Fennoscandian arctic fox Alopex lagopus is critically endangered and the population size is estimated at 120 individuals that are fragmented into four isolated populations. Here, we use molecular tracking and visual observations to estimate population size and survival in one of the populations on the Swedish mountain tundra during a year of low food availability. We collected 98 arctic fox faecal samples during the winter of 2006 and recorded visual observations of ear-tagged individuals during the summer of 2005 and 2006. The faecal samples were analysed for variation in nine microsatellite loci and matched to the genetic profiles of previously ear-tagged individuals from 2001 to 2005. During winter 2006, the minimum number alive was 12 individuals using visual observations, 30 using molecular tracking and 36 by combining the datasets. Population size was estimated through mark–recapture for the molecular tracking and visual observation datasets and through rarefaction analyses for molecular tracking data. The mark–recapture estimate for visual observations was uninformative due to the large confidence interval (CI) (i.e. 6–212 individuals). Based on the molecular tracking dataset combined with the minimum number alive for visual observations and molecular tracking, we concluded a consensus population size of 36–55 individuals. We also estimated the age-specific finite survival rate during 1 year (July 2005 to July 2006) by combining molecular tracking with visual observations. Juvenile survival on a yearly basis was 0.08 (95% CI 0.02–0.18) while adults had a survival of 0.59 (95% CI 0.39–0.82). Juveniles displayed a lower survival than the adults during autumn ( P<0.01) whereas no age-specific survival difference during spring was found. Abstract Background Scavenger receptor-mediated uptake of cholesterol by macrophages in the arterial wall is believed to be proatherogenic. Thiazolidinediones are peroxisome proliferator-activated receptor γ (PPARγ)-agonists, which are used in the treatment of type II diabetes. They reduce atherogenesis in LDL receptor deficient and ApoE knockout mice, but up-regulate CD36, which may contribute to foam cell formation. The dyslipidaemia in type II diabetes is characterized by high levels of nonesterified fatty acids. Therefore we tested the effect of fatty acids and how fatty acids and the thiazolidinedione darglitazone interact in their effect on CD36 expression in human monocytes and macrophages. Materials and methods Flow cytometry and reverse transcription-polymerase chain reaction were used to study CD36 expression. Cellular lipids were analyzed with high performance liquid chromatography. Results Darglitazone increased CD36 mRNA and protein expression in human macrophage cells. In the presence of 5% human serum, darglitazone increased the accumulation of triglycerides, but did not affect cholesterol ester levels. In the presence of albumin-bound oleic or linoleic acid, darglitazone did not increase CD36 mRNA, cell-surface CD36 protein or triglyceride content. Fatty acids per se increased CD36 mRNA and protein. Discussion The increase in CD36 in macrophages suggests a role for fatty acids in the regulation of foam cell formation. The results also suggest that the potentially proatherogenic CD36 up-regulating effect of thiazolidinediones in macrophages might not be present when the cells have access to physiological levels of albumin-bound fatty acids. Abstract Background Oxysterols are biologically active molecules generated during the oxidation of low-density lipoprotein or formed enzymatically in vivo . In the atherosclerotic plaque newly recruited macrophages may be exposed to oxysterols present in the plaque. How these oxysterols affect the expression and secretion of inflammatory cytokines such as interleukin-1β (IL-1β) in macrophages is not known. Therefore the aim of the present study was to investigate how oxysterols regulate the expression and secretion of IL-1β in human monocyte-derived macrophages. Methods The IL-1β messenger RNA (mRNA) expression was analysed by reverse transcription-polymerase chain reaction, and the IL-1β protein secretion was measured by enzyme-linked immunosorbent assay. Results A significant, dose-dependent increase in the secretion of IL-1β was given by 25-hydroxycholesterol without the addition of lipopolysaccharide (LPS). At a concentration of 2·5 µg mL-1 this increase was similar to that obtained by endotoxin (LPS, 1 µg mL-1 ). A transient increase in IL-1β mRNA expression was found in macrophages incubated with 25-hydroxycholesterol compared with untreated controls. In addition, 25-hydroxycholesterol dramatically increased the IL-1β secretion induced by LPS. At a concentration of 5 µg mL-1 of 25-hydroxycholesterol the LPS-induced IL-1β secretion was increased by about 25-fold. A similar tendency, but not so consistent, was found for 27-hydroxycholesterol. Conclusions Our results show that oxysterols, and 25-hydroxycholesterol in particular, may modulate the inflammatory response in human macrophages. The crystal structures of three conformations, T(6), T(3)R(3) and R(6), of bovine insulin were solved at 1.40, 1.30 and 1.80 Å resolution, respectively. All conformations crystallized in space group R3. In contrast to the T(6) and T(3)R(3) structures, different conformations of the N-terminal B-chain residue PheB1 were observed in the R(6) insulin structure, resulting in an eightfold doubling of the unit-cell volume upon cooling. The zinc coordination in each conformation was studied by X-ray absorption spectroscopy (XAS), including both EXAFS and XANES. Zinc adopts a tetrahedral coordination in all R(3) sites and an octahedral coordination in T(3) sites. The coordination distances were refined from XAS with a standard deviation of <0.01 Å. In contrast to the distances determined from the medium-resolution crystal structures, the XAS results were in good agreement with similar coordination geometries found in small molecules, as well as in other high-resolution insulin structures. As the radiation dose for XRD experiments is two orders of magnitude higher compared with that of XAS experiments, the single crystals were exposed to a higher degree of radiation damage that affected the zinc coordination in the T(3) sites in particular. Furthermore, XANES spectra for the zinc sites in T(6) and R(6) insulin were successfully calculated using finite difference methods and the bond distances and angles were optimized from a quantitative XANES analysis.
