Acoustic telemetry and microchemical analysis of otolith strontium-calcium ratios were used to evaluate how exotic brown trout Salmo trutta have responded to Japanese riverine environments of south-western Hokkaido by observing their migratory patterns. The existence of anadromous S. trutta was also verified. Most S. trutta caught in rivers for otolith analysis were freshwater residents (95·6%), whereas those caught in the sea were mainly smolts (91·3%), which had just migrated from rivers during spring. Anadromous S. trutta ( n = 6) were captured in rivers and in the sea, confirming the existence of mature pre- and post-spawning fish. According to telemetry results, both mature and immature S. trutta used the river in winter, and their estimated sea-run timings showed individual differences. Through the combination of these two methods, migratory patterns on various spatio-temporal scales were observed. This first documentation of the presence of both male and female anadromous S. trutta in the same region within Japan indicated the risk of further colonization of exotic S. trutta via oceanic migration. We and others have reported that human NF-κB inhibitor-like-1 (NFKBIL1) was a putative susceptible gene for autoimmune diseases such as rheumatoid arthritis (RA). However, its precise role in the pathogenesis of RA is still largely unknown. In this study, we generated transgenic mice expressing human NFKBIL1 (NFKBIL1-Tg) and examined whether NFKBIL1 plays some role(s) in the development of autoimmune arthritis. In both a collagen-induced arthritis model and a collagen antibody-induced arthritis model, NFKBIL1-Tg mice showed resistance to arthritis compared to control mice, indicating that the gene product of NFKBIL1 was involved in the control of thusly induced arthritis. Total spleen cells of NFKBIL1-Tg mouse showed decreased proliferation to mitogenic stimuli, consistent with its resistance to arthritis. Unexpectedly, purified T cells of NFKBIL1-Tg mouse showed increased proliferation and cytokine production. This apparent discrepancy was accounted for by the impaired functions of antigen-presenting cells of NFKBIL1-Tg mouse; both T/B cell-depleted spleen cells and bone marrow-derived dendritic cells of the Tg mouse induced less prominent proliferation and IL-2 production of T cells. Furthermore, dendritic cells (DCs) derived from NFKBIL1-Tg mouse showed lower expression of co-stimulatory molecules and decreased production of inflammatory cytokines when they were activated by lipopolysaccharide. The study identified seasonal habitat use by endangered adult Sakhalin taimen Hucho perryi and the environmental characteristics of their habitat (water depth, amount of riparian forest and sinuosity). Fifteen adult H. perryi with acoustic tags were tracked by towing an acoustic receiver with a canoe in the Bekanbeushi River system in eastern Hokkaido Island, Japan, during each month from late April to late November 2008. Individuals mainly used midstream (shallower than downstream) habitats in all seasons. These locations were generally characterized by relatively dense riparian forests and high sinuosity, indicating the presence of pools. In spring, individuals used habitats with less riparian forest cover compared to mean value of the river channel. From spring to autumn, adult H. perryi selected limnologically complex habitats with meandering channels. From summer to autumn, individuals selected habitats with more riparian forest cover. The inverse relationship between H. perryi detection and riparian forest area in spring was a result of seasonal defoliation in deciduous riparian forests. [ABSTRACT FROM AUTHOR]

Trans and conjugated fatty acids may exhibit either beneficial or detrimental bioactive effects depending on their metabolic properties. This study was conducted to elucidate if isomerization and conjugation of eicosapentaenoic acid (EPA) and docosahexaenoic acid (DHA) demonstrate more favorable bioactivity on lipid metabolism compared to unmodified EPA and DHA. The effects of dietary intake of trans and conjugated forms of EPA and DHA on lipid metabolism were evaluated in animal trials and compared to a control group fed soybean oil. None of the experimental diets showed significant differences from the control in terms of body weight; however, the white adipose tissue weight of rodents fed trans DHA, conjugated EPA (CEPA), and conjugated DHA (CDHA) was significantly lower than the control. Triacylglycerol levels in plasma were significantly decreased in groups fed trans DHA (17.2 mg/dL) and CDHA (31.9 mg/dL) relative to the control (51.3 mg/dL). The total cholesterol concentrations were significantly lower than the control (68.0 mg/dL) in all experimental groups (47.3 to 53.7 mg/dL) except CEPA (58.3 mg/dL). Fatty acid compositions of lipids extracted from rodent livers were influenced by the dietary fatty acid profiles, with all groups showing higher concentrations of stearic acid and lower levels of linoleic acid compared to the control. Rodents fed trans DHA did not have detectable levels of these fatty acid isomers in their livers, suggesting either quick metabolism or a difficulty with bio-absorption. [ABSTRACT FROM AUTHOR]

BSTRACT In order to utilize upstream chum salmon as a component of nutraceutical food, their defatted muscle proteins were hydrolyzed with 5% thermolysin. The resulting hydrolysate showed high inhibitory activity against angiotensin I-converting enzyme (inhibitory concentration50= 27.9 protein μg/mL) in vitro. A significant reduction of systolic blood pressure was observed when 500 and 2000 mg/kg of body weight were orally administered into spontaneously hypertensive rats. Angiotensin I-converting enzyme inhibitory peptides contained in the hydrolysate were isolated with various chromatographs. These 6 active peptides were Trp residue-containing dipeptides: Trp-Ala, Val-Trp, Trp-Met, Met-Trp, Ile-Trp, and Leu-Trp. SUMMARY The purpose of this study was (1) to assemble and verify a system to measure the three‐dimensional (3‐D) movement of the upper and lower complete dentures and the movement of the mandible simultaneously, and (2) to analyse the relation between denture movements and the path of closure of the mandible during function. A 3‐D motion capture system with four infrared TV cameras was used for this purpose. The relation between the dentures and the mandibular movements was analysed through the change of the inner product of normal vectors of the denture occlusal planes and mandibular planes. The mandibular movements were classified into two types the normal stroke (the path of closure was on the ipsilateral side of mastication) and the reverse stroke (on the contralateral side). The results showed that the system could measure the denture mobility within a 0·3 mm error. The mobility of the upper dentures had a correlation to the path of closure of the mandible regardless of the working side or nonworking side, and the lower dentures had a tendency to move toward the working side. Trilinoleoylglycerol (LLL), trilinolenoylglycerol (LnLnLn) and four synthetic triacylglycerols were autoxidized and the volatile products were investigated to determine the effect of fatty acid glyceride position on the mechanism of hydroperoxide decomposition. Capillary gas chromatography provided a sensitive method to follow the volatile oxidation products of mixtures of LLL and LnLnLn and of synthetic triacylglycerols containing linoleate and linolenate in different known positions. The relative amount of linoleate oxidation was determined by analyzing for hexanal, 2-heptenal and 2,4-decadienal, and the relative amount of linolenate oxidation by analyzing for 2,4-heptadienal and 2,4,7-decatrienal. The volatiles from pure monohydroperoxides of LLL and LnLnLn were compared with those of the corresponding triacylglycerols by capillary gas chromatography. Significant differences in the distribution of volatile products were observed depending on the triacylglycerols precursor. A 1∶1 mixture of LLL and LnLnLn autoxidized at 40°C showed an equal contribution of linolenate and linoleate volatiles at a peroxide value of 34. The synthetic triacylglycerols LLnL and LLLn (L, linoleic; Ln, linolenic acid) formed initially about the same total volatiles, whereas LnLnL formed more volatiles than LnLLn. The ratio Ln to L volatile products was the same for the diL triacylglycerols, and higher for LnLnL than for LnLLn. The hydroperoxides and secondary products formed from trilinoleoylglycerol autoxidized at 40°C were isolated and characterized to clarify their contribution to oxidative deterioration of vegetable oils. The products were purified by high performance liquid chromatography (HPLC) and identified, as intact triacylglycerols, by ultraviolet, infrared,H NMR andC NMR analyses, and after derivatization by lipolysis, gas chromatography, and gas chromatography-mass spectrometry. The main, primary products included mono-, bis- and tris-9-hydroperoxy- trans-10, cit-12-; 9-hydroperoxy- trans-10, trans-12; 13-hydroperoxy- cis-9, trans-11; and 13-hydroperoxy- trans-9, trans-11-linolenoyl glycerols. The structures of the minor secondary products analyzed after derivatization were consistent with known oxidative degradation products of linoleate hydroperoxides. HPLC analyses showed that the bis- and tris-hydroperoxides were formed from the mono-hydroperoxides during autoxidation at peroxide values above 18 and 28 meq/kg. Studies on the further oxidation of the mono-hydroperoxides support a mechanism for the consucutive formation of bis- and tris-hydroperoxides from the monohydroperoxides. HPLC analyses showed that no preferential oxidation occurred between the 1(3)- and 2-triglyceride positions. Four triacylglycerols containing linoleate (L) and linolenate (Ln) in specific positions were synthesized to determine the effect of fatty acid position on their relative rates and products of autoxidation. Analyses by reversedphase high performance liquid chromatography (HPLC) showed that autoxidation of L- and Ln-containing triacylglycerols form monohydroperoxides and hydroperoxy epidioxides as the main products. The peroxyl radicals of internal 12- and 13-mono-hydroperoxides of Ln triacylglycerol components cyclized rapidly and their relative triacylglycerol position had no influence on their rates of cyclization. A good linear relation was obtained between total HPLC peak areas (detected at 235 nm) of the main oxidation products and peroxide values. Reversed phase HPLC analyses thus provide a useful method to estimate oxidation of polyunsaturated triacylglycerols. The ratios of Ln to L mono-hydroperoxides were twice the ratios of Ln to L in the triacylglycerol substrates. Ln triacylglycerol components, therefore, oxidized twice as much as the L components. At 40°C, LnLnL oxidized slightly faster than LnLLn with respective induction periods of 45 and 47 hr. LLnL oxidized faster than LLLn with respective induction periods of 56 and 60 hr. Dilinolenoyl-linoleoylglycerols are, therefore, slightly less stable to oxidation when Ln is in the 1,2- than the 1,3-triacylglycerol position. The hydroperoxides and secondary products formed from trilinolenoylglycerol autoxidized at 40°C were isolated and characterized to clarify the mechanism of oxidative deterioration of polyunsaturated vegetable oils. The products were purified by high performance liquid chromatography (HPLC) and identified either as intact triacylglycerols spectrophotometrically, or after lipolysis (pancreatic lipase) and capillary gas chromatography and gas chromatography-mass spectrometry. The products included 9-, 12-, 13-, and 16-mono-, bis-, tris-hydroperoxy, 9- and 16-hydroperoxy epidioxy, 9- and 16-hydroperoxy bicycloendoperoxy and 9,12-, 13,16-, and 9,16-dihydroperoxy linolenoylglycerols. The mono-hydroperoxides and sydroperoxy epidioxides were the only main products initially formed at peroxide values (PV) below 30. Bis- and tris-hydroperoxides were formed consecutively as minor products from the mono-hydroperoxides at PV's between 31 and 47. Hydroperoxy bicycloendoperoxides and monodihydroperoxides were also formed as minor secondary products at PV's above 75. HPLC analyses show a small preference for the formation of 16-hydroperoxides on the 1(3)-position over the 2-position of trilinolenoylglycerol. However, there was no selectivity for the formation of the 9-, 12- and 13-hydroperoxides and for the hydroperoxy epidioxides between the 1(3)- and the 2-positions in trilinolenoylglycerol. Mono-hydroperoxides and hydroperoxy epidioxides of linolenate triacylglycerols may be important precursors of volatile compounds contributing to oxidative deterioration of vegetable oils. Scallop ( Patinopecten yessoensis) adductor muscles were heated using superheated steam (150 and 200 °C), boiling (98 °C), and normal steaming (95 °C). The amounts of amino acids, water-soluble peptides, and nucleotides, expressed as extractive nitrogen in scallop products, are very important elements of quality and taste. After 15-min heating of scallop muscles with normal steaming and boiling, respective losses of 50% and 64% of the extractive nitrogen were observed. However, most extractive nitrogen (> 86%) remained in the scallop muscles treated with superheated steam at 150 and 200 °C. Protective effects of superheated steam against elution loss of nitrogen compounds were also observed in amino acid analyses of the heated products. The scallop-muscle surface temperature during treatment with superheated steam increased more quickly than that with normal steaming and boiling. The rapid water loss and the surface protein denaturation engendered formation of a 30-μm-thick film covering the surface, which prevented extractive nitrogen loss from internal tissues. Superheated steam treatment at 200 °C caused browning, surface shrinkage, and 47% weight loss. In marked contrast, the appearance and the weight loss of sample treated at 150 °C were almost the same as those of normal steaming and boiling-treated samples. Dietary effects of herring roe lipids (HR-L) and proteins (HR-P) on plasma lipids and abdominal fat pad weight were determined. The main lipid class of HR-L was phospholipids (74%) and the main fatty acids were palmitic acid (16:0, 25.8%), DHA (22:6n-3, 21.6%), EPA (20:5n-3, 14.4%), and oleic acid (18:1n-9, 13.2%). A little increase in total cholesterol level was observed in plasma lipids of mouse fed with HR-L, although HR-L contained 9% cholesterol. This would be due to the lowering effect of EPA and DHA contained in HR-L on plasma cholesterol. Replacement of a part of dietary protein (5%) to HR-P reduced abdominal fat pad weight, but not significantly. On the other hand, combination of HR-P and HR-L significantly reduced the fat pad weight of the mouse as compared with the control. Objective: The treatment of nasal foreign bodies involves safe and reliable removal. Few reports have investigated the relationship between equipment and the incidence of complications., Methods: This retrospective study included 300 patients with nasal foreign bodies (average: 3.28 years, interquartile range: 2-4 years). Patients' background, characteristics of nasal foreign body, equipment to remove the nasal foreign body, and complications were obtained from medical records. Statistical analysis was performed using Pearson's chi-square test for associated factors and the incidence of epistaxis among the complications., Results: Nasal foreign bodies were found and removed in 256 patients. Forceps, hooks, suction, modified paper clips, and cotton swabs were mainly used to remove the nasal foreign bodies. Epistaxis due to the removal procedure was observed in 26 patients. The occurrence of epistaxis differed depending on the equipment (p = 0.077) and was less frequent in suction and paper clips than in forceps (p < 0.05 and p = 0.077). Epistaxis was not observed when a cotton swab was used. Aspiration and septal perforation were not observed. A statistical relationship was not detected between the hardness of foreign bodies and the occurrence of epistaxis (p = 0.251). Objectives/hypothesis: Recent developments in reconstructive techniques for mucosal defects using mucoperiosteal materials have enabled rapid recovery of physiological function after endoscopic sinus surgery. Clinical trials have described the advantages, disadvantages, and different outcomes of free graft and pedicled flap, which, respectively, sacrifice or preserve blood flow. However, histological changes, that affect the postoperative outcomes after reconstruction, remain unclear. We created an animal model for the reconstruction of mucosal defects using free grafts and pedicled flaps, and evaluated them histologically., Study Design: Animal study., Methods: We created mucosal defects in the left nasal septum of 20 rabbits and performed reconstruction with free grafts and pedicled flaps. The distribution of ciliary and goblet cells at the reconstruction site was evaluated after 7 and 28 days using hematoxylin and eosin-stained sections to calculate the Ciliary Cell Index and Goblet Cell Index. The severity of inflammation was assessed using the Cartilage Inflammatory Cell Score., Results: Crusting and changes in the mucosal morphology at the reconstruction site occurred only in the free graft group. In addition, the pedicled flap group had significantly greater preservation of ciliary and goblet cells and less inflammatory cell infiltration into the septal cartilage (P < .05) than the free graft group., Conclusions: After reconstruction procedures for mucosal defects, histopathological differences were observed between the free graft and pedicled flap. Reconstruction with pedicled flaps had advantages including preservation of healthy mucosal epithelium and suppression of inflammation on the reconstruction site. Objectives/hypothesis: Intranasal immunization with phosphorylcholine (PC) is known to reduce immunoglobulin (Ig)E production. However, its effects on the occurrence of allergic rhinitis (AR) are unknown. This study was performed to evaluate the effects of PC-keyhole limpet hemocyanin (PC-KLH) and to examine the effects on the occurrence of AR in a murine model of AR., Study Design: In vivo study using an animal model., Methods: Forty-five female BALB/c mice were divided into three groups; those pretreated with intranasal administration of PC-KLH followed by intraperitoneal sensitization and nasal challenge with ovalbumin (OVA) (group A), those untreated with PC-KLH followed by sensitization and nasal challenge with OVA (group B), and those untreated with PC-KLH or OVA as controls (group C). Nasal symptoms, allergic inflammation in the nasal mucosa, OVA specific IgE production, and cytokine profile were compared among those three groups. Dendritic cells (DCs) were isolated from splenic cells and PC-KLH-stimulated interleukin (IL)-12p40 production was measured., Results: The mice pretreated with PC-KLH showed lower allergic nasal symptoms and inflammation compared to untreated mice. The levels of total IgE and OVA-specific IgE in serum, and IL-4 production by nasal and splenic CD4 + T cells were significantly reduced by PC-KLH pretreatment. Furthermore, IL-12p40 production by DCs was induced by PC-KLH in a dose-dependent manner., Conclusions: Intranasal administration of PC-KLH suppressed allergic inflammation in nasal mucosa and antigen-specific IgE production by downregulating Th2-type immune response. Objective: Pneumococcal infection caused by Streptococcus pneumoniae is a major upper respiratory tract disease that causes severe illness and mortality. Therefore, it is important to develop safe and effective vaccines to prevent pneumococcal infections. The goal of the study was to investigate the effectiveness of transcutaneous immunization (TCI) for induction of pneumococcal surface protein A (PspA) responses in the upper respiratory tract., Methods: C57BL/6 mice were transcutaneously immunized with 1 μg of PspA and 2 μg of cholera toxin (CT) six times at weekly intervals and compared with transcutaneously treated controls (PBS alone/PspA alone/CT alone). Two weeks after the final immunization, nasal washes (NWs), saliva, and plasma samples were collected and subjected to a PspA-specific ELISA. Three weeks after the final immunization, mice were challenged with S. pneumoniae strain EF3030, and the numbers of CFUs in NWs and nasal passages (NPs) were determined., Results: Higher levels of PspA-specific IgM, IgG, and IgA Abs were noted in plasma of TCI with PspA plus CT compared with controls. Transcutaneous immunization mice also had significantly increased PspA-specific S-IgA Ab responses in NWs and saliva and, importantly, showed significantly lower numbers of bacteria CFUs in NWs and NPs compared with controls., Conclusion: These results show that TCI with PspA plus CT induces antigen-specific mucosal and systemic immune responses. This suggests that this method is an effective mucosal immunization strategy for induction of protective pneumococcal-specific Ab responses in blockade of S. pneumoniae colonization of the nasal cavity., Level of Evidence: NA. Long-term observation has identified a pattern of continuing relapse in limited stage diffuse large B-cell lymphoma (DLBCL) treated by three cycles of R-CHOP (rituximab, cyclophosphamide, doxorubicin, vincristine, prednisone) plus involved-field irradiation. We retrospectively analysed 190 untreated patients with limited stage DLBCL treated by R-CHOP alone. All the patients were scheduled to undergo primary therapy with six cycles of full-dose R-CHOP. Cases with a dose reduction of more than 20% were excluded from the study. Additional local irradiation was allowed in patients with partial response (PR). Five patients received additional local irradiation after PR at the end of the R-CHOP therapy. The median observation period was 52 months. Median age at diagnosis was 63 years. The responses to therapy were 180 complete responses, eight PR, and two progression of disease (PD). The 5-year progression-free survival and 5-year overall survival rates were 84% and 90%, respectively, both in plateau. During the observation period, 29 patients experienced PD. The progression sites were the primary sites in 15 patients, outside the primary sites in 10, and undetermined in four patients. These results suggest that the 'standard' strategy of three cycles of R-CHOP followed by involved-field radiotherapy for limited stage DLBCL could be effectively replaced by six cycles of R-CHOP alone., (© 2013 Blackwell Publishing Ltd.)

The intermittent administration of methamphetamine produces behavioral sensitization to methamphetamine. In the limbic forebrain, mainly including the nucleus accumbens, of mice that had been intermittently treated with methamphetamine, we found a significant increase in mRNA of a chemokine, CCR2. This increase was accompanied by a significant increase in histone H3 lysine 4 (H3K4) trimethylation at its promoter. Interestingly, the maintenance of sensitization to methamphetamine-induced hyperlocomotion was significantly decreased in CCR2 knockout mice. These findings suggest that increased CCR2 associated with epigenetic modification after the intermittent administration of methamphetamine may be associated with the maintenance of sensitization to methamphetamine-induced hyperlocomotion.
