One of the major challenges in evolutionary biology is to understand the mechanisms underlying morphological dimorphism and plasticity, including the genomic basis of traits and links to ecology. At the yellow eel stage of the European eel ( Anguilla anguilla), two morphotypes are found: broad- and narrow-heads. This dimorphism has been linked to dietary differences, with broad-heads feeding on harder, larger prey than narrow-heads. However, recent research showed that both morphotypes could be distinguished at the glass eel stage, the nonfeeding predecessor of the yellow eel stage, implying that nondietary factors play a role in the development of this head shape dimorphism. Here, we used transcriptome profiling ( RNAseq) to identify differentially expressed genes between broad- and narrow-headed glass eels. We found 260 significantly differentially expressed genes between the morphotypes, of which most were related to defence and immune responses. Interestingly, two genes involved in growth ( soma and igf2) were significantly upregulated in narrow-heads, while nine genes involved in chemotaxis showed significant differential expression. Thus, we found support for the observation that head shape is associated with somatic growth, with fast-growing eels developing a narrower head. Additionally, observations in the wild have shown that slow-growers prefer freshwater, while fast-growers prefer brackish water. The differential expression of genes involved in chemotaxis seems to indicate that glass eel growth rate and habitat choice are linked. The anadromous–freshwater three-spined stickleback (Gasterosteus aculeatus) system allows for inferring the role of adaptation in speciation with a high level of accuracy because the freshwater ecotype has evolved multiple times from a uniform anadromous ancestor. A cause for concern is that independent evolution among drainages is not guaranteed in areas with a poorly resolved glacial history. This is the case for the west European great rivers, whose downstream valleys flanked the southern limit of the late Pleistocene ice sheet. We tested for independent and postglacial colonization of these valleys hypothesizing that the relationships among anadromous and freshwater sticklebacks correspond to a raceme structure. We compared the reduction in plate number accompanying this colonization to the genetic differentiation using 13 allozyme and five microsatellite loci in 350 individuals. Overall microsatellite differentiation (FST = 0.147) was twice as large as allozyme differentiation (FST = 0.066). Although habitat-specific gene flow may mask the ancestral relationships among both ecotypes, levels of microsatellite differentiation supported the hypothesis of raceme-like divergence, reflecting independent colonizations rather than the presence of two distinct evolutionary clades. Under an infinite alleles model and in the absence of gene flow, the observed freshwater divergence might be reached after 440 (microsatellites) to 4500 (allozymes) generations. Hence, the anadromous–freshwater stickleback system most likely diverged postglacially. We conclude that the reduction in plate number in two freshwater basins probably occurred independently, and that its considerable variation among populations is not in agreement with the time since divergence. European eel( Anguilla anguilla; Teleostei) is a valuable commercial species. However, over the past 25 years, the population of European eel has been declining to such a degree that major concerns have been raised for its long-term conservation. Since little information is available on the life-cycle and genetic structure of European eel, it has been difficult to evaluate the existence of any population substructuring. Molecular genetic methods contribute to a better knowledge of the demography and population structure in marine fish. In addition, management strategies and conservation goals must consider information on genetic substructuring as well as on life history patterns.The aim of the study is to provide more detailed knowledge on the genetic variability, demography and population substructuring of European eel by analysing and comparing natural and farmed individuals. Natural eel samples have been obtained in two geographical sites(Netherlands, France) including temporal samples in a short-scale(within years) and a long-scale(between years). Simultaneously, farmed glass eels have been grown in two separate batches during one year. Batches have been monitored and genetic samples have been obtained during the year.A combination of selection-sensitive(allozymes) and selection-neutral markers(microsatellites) has been used in the study since selection seems to play an important role in the determination of the quality of future eel spawners. Results suggest a positive correlation between growth and genetic variability since individuals attaining a large length and mass present significant higher heterozygosities. [ABSTRACT FROM AUTHOR]
