A model to rate color combinations that considers human aesthetic preferences is proposed. The proposed method does not assume that a color palette has a specific number of colors, i.e., input is not restricted to a two-, three-, or five-color palettes. We extract features from a color palette whose size does not depend on the number of colors in the palette. The proposed rating prediction model is trained using a human color preference dataset. The model allows a user to extend a color palette, e.g., from three colors to five or seven colors, while retaining color harmony. In addition, we present a color search scheme for a given palette and a customized version of the proposed model for a specific color tone. This paper presents a procedure for modelling aggregates such as piles that consist of arbitrary components. The method generates an aggregate of components that need to be accumulated, and an aggregate shape represents the surface of the target aggregate. The number of components and their positions and orientations are controlled by five parameters. The components, the aggregate shape and the parameters are the inputs for the method which involves placement and refinement steps. In the placement step, the orientation and initial position of a component are determined by a non-periodic placement such that each component overlaps its neighbours. Objectives: MicroRNAs (miRNAs) are small functional RNAs that regulate mRNAs for degradation or translational suppression. In the present study, we aimed to reveal functional importance of miRNA-494 (miR-494) in A549 human lung cancer cells. Materials and methods: We established A549 cells that constitutively expressed miR-494. Next, we sought to investigate insulin-like growth factor 2 mRNA-binding protein 1 (IGF2BP1) mRNA as an miR-494 target. For this, we constructed a reporter plasmid bearing potential miR-494 binding sequences derived from the 3′-untranslated region (3′-UTR) of IGF2BP1 mRNA in the 3′-UTR of the luciferase gene. Results: Through comparison between miR-494 expressing cells and control cells, we revealed that miR-494 suppressed cell proliferation and colony forming activity, and induced senescence. Reporter activity was inhibited by miR-494. In addition, IGF2BP1 mRNA levels were down-regulated in A549 cells that constitutively expressed miR-494. IGF2BP1 has been shown to bind and suppress IGF2 mRNA, and this could be a reason why IGF2BP1 can regulate cell function. Therefore, we analysed IGF2 mRNA levels and revealed that IGF2 was up-regulated in A549 cells that constitutively expressed miR-494. Finally, elevated IGF2 mRNA levels in A549 cells that constitutively expressed miR-494 were suppressed to basal level by an miR-494 inhibitor. Conclusions: Taken together, IGF2BP1 and its downstream target IGF2 could be a crucial axis for miR-494 in regulation of the destiny of A549 cells. [ABSTRACT FROM AUTHOR]

The aim of this study is to verify the effectiveness of ordinary phenomenological constitutive relation of NiTi shape memory alloy under mechanical loading at a constant temperature, sufficiently. First, finite element analysis is performed by using ordinary phenomenological constitutive relation for rectangular plate with double notch under tensile loading at a constant temperature. Next, uniaxial tensile loading is carried out for 50.5Ni49.5Ti rectangular plate with double notch. At the same time, macroscopic stress-strain curve and local strain distribution are measured by using in-house measurement system on the basis of digital image correlation. As a result, it is found that the stress-strain curve obtained from finite element analysis is much different from those obtained experimental measurement, especially during stress-induced martensite transformation. The result can be derived from the phenomena of local strain band behavior arising in NiTi under mechanical loading. The phenomenological constitutive model used in present finite element analysis is constructed under assumptions that the material has isotropic characteristics and shows homogeneous deformation. However, this experimental result suggests that the material itself has anisotropy microscopically. Furthermore, material shows unique inhomogeneous deformation. Also, there is possibility that these anisotropic characteristic and inhomogeneous deformation behaviour may derive from its microstructure. In future, to sufficiently describe the macroscopic stress-strain curve of NiTi we should take into consideration the material microstructure. [ABSTRACT FROM AUTHOR]

In this study, we examined the effects of serotonin (5-HT)3 receptor antagonists (5-HT3RAs) including ramosetron, alosetron, and cilansetron on colonic nociceptive threshold in rats. Furthermore, we established a restraint stress-induced colonic hyperalgesia model in rats, and compared the inhibitory effects of 5-HT3RAs on restraint stress-induced colonic hyperalgesia and diarrhoea with those of loperamide, trimebutine, tiquizium and polycarbophil. The colonic nociceptive threshold was measured as the balloon pressure at the time the rat showed a nociceptive response during colonic distension by an intrarectally inserted balloon. Oral administration of ramosetron (3–30 μg kg−1), alosetron (30–300 μg kg−1), or cilansetron (30–300 μg kg−1) increased the colonic nociceptive threshold in a dose-dependent manner in non-stressed rats. Restraint stress for 1 h significantly decreased the colonic nociceptive threshold, but ramosetron (0.3–3 μg kg−1), alosetron (3–30 μg kg−1), cilansetron (3–30 μg kg−1) and trimebutine (100–1000 mg kg−1) significantly inhibited the decrease in the threshold. Loperamide (3–30 mg kg−1), tiquizium (100–1000 mg kg−1) and polycarbophil (1000 mg kg−1) did not affect the restraint stress-induced decrease in the colonic nociceptive threshold. All drugs tested in this study showed dose-dependent inhibition of restraint stress-induced diarrhoea in rats. These results indicate that, unlike existing antidiarrhoeal and spasmolytic agents, 5-HT3RAs have inhibitory effects on colonic nociception, and prevented restraint stress-induced both diarrhoea and hyperalgesia at almost the same doses in rats. This suggests that the 5-HT3RAs may be useful in ameliorating both colonic hyperalgesia and diarrhoea in patients with irritable bowel syndrome. [ABSTRACT FROM AUTHOR]

Kawasaki disease (KD) is vasculitis of unknown etiology in infants and young children. The diagnostic criteria for KD include major and minor symptoms, but various nail lesions are not described in detail. The aim of this study was to identify symptoms that are relatively found in nail of KD as diagnostic markers. After literature review, various nail lesions are classified as Beau's lines, leukonychia, onychomadesis, orange-brown chromonychia, and pincer nail deformity. The orange-brown chromonychia is the most common nail lesion in KD. In this study, the authors found three cases of KD with orange-brown chromonychia; two of these cases included rare dotted or splinter hemorrhages in the nail bed that were found on dermoscopic examination. The COL2A1 gene encodes the alpha-1 chain of procollagen type 2. Pathogenic variants in the COL2A1 gene are associated with several different types of skeletal dysplasia collectively known as type 2 collagenopathies. Type 2 collagenopathies have an autosomal dominant inheritance. Some germline or somatogonadal mosaicism cases have been reported. We investigated whether somatogonadal mosaicism occurred in a family with two children suspected of type 2 collagenopathies or related diseases. First, we detected a pathogenic variant in the COL2A1 gene in the two affected children by whole exome sequencing (WES). Next, we performed targeted deep sequencing to their parents without the variant by WES. A low level of COL2A1 mosaicism was revealed in the mother's tissues. We concluded that the mother had somatogonadal mosaicism with the COL2A1 mutation arose in the epiblast, and that the intrafamilial recurrence rate of the disease by the somatogonadal mosaicism was higher than by the germline mosaicism. Cole-Carpenter syndrome is a rare skeletal dysplasia associated with low-bone mass or an osteogenesis imperfecta (OI)-like syndrome. Only 3 and 6 variants in SEC24D have been reported in patients with Cole-Carpenter syndrome type 2 and autosomal recessive OI, respectively. We describe a 15-year-old Japanese boy with short stature of the short-trunk type and craniofacial abnormalities including ocular proptosis, marked frontal bossing, midface hypoplasia, and micrognathia. These features were consistent with a diagnosis of Cole-Carpenter syndrome. He had low-bone mineral density and basilar impression. Whole exome sequencing analysis identified biallelic variants in SEC24D (p.Arg484* and p.Arg313His) in the patient. Intervertebral disk (IVD) degeneration is a prevalent age-associated musculoskeletal disorder and a major cause of chronic low back pain. Aging is the main risk factor for the disease, but the molecular mechanisms regulating IVD homeostasis during aging are unknown. The aim of this study was to investigate the function of FOXO, a family of transcription factors linked to aging and longevity, in IVD aging and age-related degeneration. Conditional deletion of all FOXO isoforms (FOXO1, 3, and 4) in IVD using the Col2a1Cre and AcanCreER mouse resulted in spontaneous development of IVD degeneration that was driven by severe cell loss in the nucleus pulposus (NP) and cartilaginous endplates (EP). Conditional deletion of individual FOXO in mature mice showed that FOXO1 and FOXO3 are the dominant isoforms and have redundant functions in promoting IVD homeostasis. Gene expression analyses indicated impaired autophagy and reduced antioxidant defenses in the NP of FOXO-deficient IVD. In primary human NP cells, FOXO directly regulated autophagy and adaptation to hypoxia and promoted resistance to oxidative and inflammatory stress. Our findings demonstrate that FOXO are critical regulators of IVD homeostasis during aging and suggest that maintaining or restoring FOXO expression can be a therapeutic strategy to promote healthy IVD aging and delay the onset of IVD degeneration., (© 2018 The Authors. Aim: We examined whether the paediatric assessment triangle (PAT) could predict the severity of Kawasaki disease., Methods: We enroled patients diagnosed with Kawasaki disease between July 2012 and June 2016 at the emergency department of Tokyo Metropolitan Children's Medical Center in Tokyo, Japan. Triage nurses assigned participants to unstable or stable PAT groups. We compared the incidence of coronary artery aneurysms (CAA), the Kobayashi score, which measures resistance to intravenous immunoglobulin treatment, and the incidence of initial treatment resistance., Results: Of the 420 participants, who were aged 0-145 months with a mean age of 31.2 ± 23.9 months, 66 (16%) were assigned to the unstable PAT group. The incidence of CAA was similar between the two groups. The percentage of unstable PAT group participants with a Kobayashi score of at least five points (39 versus 18%, p < 0.001) and initial treatment resistance (25 versus 15%, p = 0.047) were significantly higher than in the stable PAT group. Unstable PAT was an independent risk factor for initial treatment resistance (odds ratio 2.02, 95% confidence interval 1.05-3.90, p = 0.035)., Conclusion: An unstable PAT was able to predict the severity of Kawasaki disease when measured by a higher rate of initial treatment resistance., (©2018 Foundation Acta Paediatrica. An 18-year-old healthy female student noticed a brown macule measuring 21 mm in diameter on the left palm and visited our clinic concerned about a cancerous mole. Dermoscopic examination revealed a brown, fine-dotted and granule-like structure overlapping an amorphous light brown macule. However, unlike previous cases, analysis of the high dynamic range-converted image revealed the parallel ridge pattern frequently observed in malignant melanomas. Brown mycelia were detected on direct microscopic examination; black colonies were isolated on fungal culture and the fungus was identified as Hortaea werneckii. Purpose: To examine improvement with the use of Scheimpflug imaging of the anterior and posterior corneal surfaces in the accuracy of ray-tracing intraocular lens (IOL) power calculation for normal cataractous eyes., Methods: Prospective case series comprised 136 eyes of 136 consecutive patients who had undergone cataract surgeries. Scheimpflug imaging of the cornea was included with routine preoperative examinations. Postoperative refractions were predicted using three methodologies; ray-tracing calculation using Scheimpflug imaging and Placido topography, ray-tracing calculations using Placido topography, and the SRK/T formula using autokeratometry. Prediction errors from the manifest refraction at 1 month postoperatively were compared between the methods. Influence of the posterior corneal curvature was also evaluated., Results: Mean prediction errors were 0.008, -0.103 and -0.042 D, respectively without significant difference between the three methods (p = 0.23). The prediction errors were significantly correlated with the posterior corneal curvature when the Scheimpflug imaging was not used (p < 0.03)., Conclusion: Use of Scheimpflug imaging in ray-tracing IOL power calculation was as accurate as the other calculations in normal cases, showing no bias in the posterior corneal curvature, as is the case with the other calculations., (© 2013 Acta Ophthalmologica Scandinavica Foundation.
