Meloidogyne incognita is the most economically important plant‐parasitic nematode. Meloidogyne incognita manipulates plant cell development and metabolism by injecting effectors from the oesophageal glands into the plant host. Chorismate mutase (CM) is one such effector that may contribute to successful parasitism by M. incognita. This investigation identified and functionally characterized a novel CM effector, Mi‐CM‐3, which is more similar to CMs from bacteria than from other phytoparasitic nematodes. Spatial and temporal expression assays showed Mi‐cm‐3 mRNA accumulates specifically in the subventral oesophageal glands and transcription is up‐regulated during the early parasitic stages of the nematode. In planta gene silencing of Mi‐cm‐3 attenuated nematode parasitism. In addition, Mi‐cm‐3 could fully restore the full virulence phenotype of the pathogenic bacterium Xanthomonas oryzae pv. oryzae by complementation when it was introduced into a mutant strain carrying a deletion in the CM gene. Transient expression of Mi‐CM‐3 caused a reduction in levels of salicylic acid (SA) and mRNA of gene PR1 in Nicotiana benthamiana in response to oomycete pathogen Phytophthora capsici infection, while confocal observations showed that Mi‐CM‐3 was localized within the cytoplasm and the nucleus, but not the plastids, of transfected N. benthamiana leaf cells. Constitutive expression of Mi‐CM‐3 in N. benthamiana plants inhibited root growth and increased susceptibility to M. incognita infection. These results provide direct experimental evidence to show that Mi‐CM‐3 may play an important role in suppressing plant immunity by regulating the SA pathway during the early parasitic stage of M. incognita so as to promote nematode parasitism. We describe 24 novel primers that amplify intron regions in housekeeping and structural genes of Heterorhabditis bacteriophora. The cross-amplification potential of these primers in seven other Heterorhabditis species was determined. The results obtained showed interspecific nucleotide, length and splice site variability in the sequenced introns and for one gene, an intron gain was observed. These primers will be useful tools for studying population genetics, genetic diversity and intron DNA evolution within the genus Heterorhabditis and other genera of rhabditid nematodes. Synopsis Cuticle scales are the most obvious feature of a hair fibre's outer surface and much research has focused on assessing the influence of surface topography on the associated hair fibre's properties. However, much of the research has either been qualitative or, if quantitative, employed relatively laborious analytical techniques to establish the necessary statistical robustness. In this study, we report on the application of a 3D image analysis package capable of producing 3D data from multiple 2D scanning electron microscope (SEM) images of hair fibres. Analysis of the surface profile can be carried out quickly and accurately, enabling quantification of the scale structure. To validate the novel technique and ensure that the scale heights measured were indeed accurate and reproducible, extensive calibration of the SEM and the3d software has been performed. In addition, scale heights on a single hair have been determined by using atomic force microscopy (AFM) and the results compared with analogous data produced from the same scale edges using the 3D image analysis technique. The data obtained indicate that a relatively quick, accurate and viable method to determine scale height in keratin fibres has been established. A further 3D-SEM analysis method has been developed which allows individual scales to be monitored and for the cuticular scale height to be quantified after repeated ‘smoothing’ treatments. Xiphinema americanum is listed as a quarantine nematode by The European Plant Protection Organisation because of its ability to transmit quarantine viruses. Detection of this nematode in soil samples, or in mixed nematode samples extracted from soil, is therefore of paramount importance. We recently described the use of magnetic beads (Dynabeads) in combination with probes such as lectins or antibodies for recovery of small endoparasitic nematodes including Meloidogyne spp. and Globodera rostochiensis from mixed nematode samples. Here we show that magnetic capture can be used with much larger nematodes such as X. americanum. We describe lectins and antisera that bind to the surface of this nematode and show that the antisera can be used in the Dynabeads system to recover and enrich X. americanum from mixed nematode samples.
