Globalization, the modern lifestyle, immuno-suppressive agents, invasive surgical procedures, the loss of efficacies of existing drugs, and multidrug resistance are some of the factors used to explain the rise in fungal infections in recent years. Significant advances have been made in attempts to replace existing antifungal schedules, especially with synthetic targets. The identification of other platforms for drug discovery is now entrenched in research programs across the globe. Plants offer significant benefits owing to their numerical superiority, exceedingly broad chemical basis and appealing sustainability characteristics. Furthermore, plants have a long and rich historical association with traditional approaches towards fungal diseases. These have in numerous instances served as markers in the bioassay-guided identification of the active constituents. Although the plant family Amaryllidaceae is conventionally associated with cancer and motor-neuron disease chemotherapies, around 30 of its species have been examined for antifungal activities with microgram per millilitre inhibitory activities detected in several instances. This review focuses on the nearly 40 constituents from the family, mainly isoquinoline alkaloids, which have been screened against around 50 fungal pathogens. Encouragingly, microgram per millilitre growth inhibitory activities were applicable for several of the compounds with a minimum inhibitory concentration of 4 μg/ml seen to be the lowest. We thank Lambert and Steen ([12]) for the attention they have given to our paper (Bowne et al. [2]) and welcome productive discussions of how human activities affect turtle populations. We agree with Lambert and Steen that roads and urbanization can be and in many cases are threats to freshwater turtles, but we argue that male-biased sex ratio is not a universal result of anthropogenic activity. We also note that relative sex ratio is the standard most used in the literature on turtle sex ratio (e.g., Marchand & Litvaitis [14]; Steen & Gibbs [17]). We support any effort to improve our collective understanding of the relationship between turtle populations and urbanization by generating better population estimates, but that is different from treating relative population ratios as absolute population ratios. A new phytochemical study of the indigenous Brazilian species Hippeastrum papilio is reported herein. Three novel Amaryllidaceae alkaloids were isolated, including hippapiline ( 1), papiline ( 2), and 3- O-demethyl-3- O-(3-hydroxybutanoyl)haemanthamine ( 3). Their structures were determined by physical and spectroscopic methods. In addition, the known alkaloids, haemanthamine ( 4), galanthamine ( 5), narwedine ( 6), 11 β-hydroxygalanthamine ( 7), apogalanthamine ( 8), and 9- O-demethyllycosinine B ( 9) were identified. The unusual cis-B/ C-ring fusion for the new homolycorine representative hippapiline was ratified by NMR and CD spectroscopy. [ABSTRACT FROM AUTHOR]

Question To what extent are species, including native and non-native species, influencing one another's distribution and abundance in the herb layer of a Minnesota oak woodland? Location Oak woodland succeeding into a more mesic forest, on bluffland of the Mississippi River, east-central Minnesota. Methods We collected plant composition and species cover data in 182 1.0 × 0.5 m quadrats regularly spaced on a 6-ha study grid in the oak woodland. We also recorded slope, slope position, aspect, elevation and photosynthetically active radiation (PAR) at each quadrat. Results Presence and abundance of other plant species, topographic variables and light availability explained only a small portion of the variation (5-19%) in the distribution and abundance of individual species. The most common strongest predictor of cover for the ten most common species was species richness, with the association being positive. The non-native species, garlic mustard ( Alliaria petiolata) exhibited the strongest positive association with species richness. Only one of the 45 pair-wise comparisons of the ten species resulted in a negative relationship between the species. Abundance and distribution of two species were associated with topographic features, but this accounted for much less of the variation in abundance than did species richness. Conclusion We found little evidence that competition or any other interactions among common herb layer species, including the non-native Alliaria petiolata, play an important role in determining the abundance and the distribution of herb layer species in this oak woodland. Topographic factors may explain a small amount of the distribution and abundance patterns of a few species. But, for the most part, species are more likely to be present when other species are present, suggesting that they are simply establishing in microsites favourable to plants in general. A mechanistic understanding of the causes of naturalization in terrestrial plant communities, and the consequences for associated ecosystems, relies on understanding whether and how native and non‐native plant traits differ. Traits of non‐native plants may either help them compete or avoid competition with natives, and depending on their interaction with the native community, the non‐native may act as more or less of an active driver vs. a passive passenger of change in the native ecosystem. Trait comparisons between native and non‐native plants are often laboratory‐based, providing insights that can be difficult to apply to natural communities. Studies of the impacts of non‐native plants, by contrast, are generally performed in natural communities, but usually do not compare observed impacts of the non‐natives with those exerted by dominant natives on each other and the community. Thus, the likely relative disruption of the non‐native to the community is unclear. In a four‐year observational study conducted at the microhabitat scale in the herbaceous layer of a Midwestern oak forest, we compared the non‐native herb Alliaria petiolata (garlic mustard) with dominant native herbaceous species in terms of correlational patterns related to three broad categories of traits: (1) environmental correlates (light, topography, soil nutrients) with presence and cover, (2) inter‐annual regeneration and phenology, and (3) associations with other species (competition and herbivory). We found that Alliaria differed strongly from dominant native herbs in all three categories. Compared with native herbs, Alliaria was more strongly associated with south‐facing slopes and high nutrient levels, displayed earlier phenology and enhanced dispersal ability, and was strongly avoided by the generalist native herbivore, Odocoileus virginianus (white‐tailed deer). Despite well‐described allelopathic effects of Alliaria on mycorrhizae and mycorrhizal plants, we found little evidence to suggest that Alliaria had negative interactions with native plants that were stronger than those of native community members. Overall, our data suggest that naturalization of Alliaria in this site is primarily due to differences from native plants in phenology, dispersal capability, and avoidance of herbivory, and that Alliaria is therefore more likely to be acting as a passenger than as a driver of change in our site. Introduction: In patients undergoing patent ductus arteriosus (PDA) ligation there is a significant risk of left vocal fold paralysis (LVFP) particularly in premature neonates who are small for gestational age. The objective of this study is to determine the incidence of LVFP in infants following PDA ligation and report on long-term outcomes in patients with LVFP., Methods: We performed a prospective study of patients undergoing PDA ligation in the newborn intensive care unit (NICU) between April 2004 and May 2014. Following PDA ligation, flexible laryngoscopy was performed to assess vocal fold mobility. Patients were then followed longitudinally to determine long-term outcomes., Results: A total of 163 infants underwent PDA ligation. Thirty-six patients (22%) developed LVFP following the procedure. Twenty-five percent of neonates <1500 g experienced LVFP versus 5% of patients >1500 g (p = 0.033). Patients with LVFP were more likely to require a feeding tube (64% vs. 19.6%; p < 0.05) and spent more time in the NICU (135 days vs. 106 days; p < 0.05). Twenty-four patients received long-term follow-up. Six (25%) had complete resolution of LVFP, 10 (42%) were compensated, and 8 (33%) demonstrated persistent LVFP with no improvement., Conclusions: The incidence of LVFP after PDA ligation is high especially in extremely low birth weight children. The majority of patients recovered well with time, but further surgical intervention was required in uncompensated cases. Long-term follow-up of these patients is needed to ensure improvement. Nitrogen is the most crucial element in the production of nutritious feeds and foods. The production of reactive nitrogen by means of fossil fuel has thus far been able to guarantee the protein supply for the world population. Yet, the production and massive use of fertilizer nitrogen constitute a major threat in terms of environmental health and sustainability. It is crucial to promote consumer acceptance and awareness towards proteins produced by highly effective microorganisms, and their potential to replace proteins obtained with poor nitrogen efficiencies from plants and animals. The fact that reactive fertilizer nitrogen, produced by the Haber Bosch process, consumes a significant amount of fossil fuel worldwide is of concern. Moreover, recently, the prices of fossil fuels have increased the cost of reactive nitrogen by a factor of 3 to 5 times, while international policies are fostering the transition towards a more sustainable agro-ecology by reducing mineral fertilizers inputs and increasing organic farming. The combination of these pressures and challenges opens opportunities to use the reactive nitrogen nutrient more carefully. Time has come to effectively recover used nitrogen from secondary resources and to upgrade it to a legal status of fertilizer. Organic nitrogen is a slow-release fertilizer, it has a factor of 2.5 or higher economic value per unit nitrogen as fertilizer and thus adequate technologies to produce it, for instance by implementing photobiological processes, are promising. Finally, it appears wise to start the integration in our overall feed and food supply chains of the exceptional potential of biological nitrogen fixation. Nitrogen produced by the nitrogenase enzyme, either in the soil or in novel biotechnology reactor systems, deserves to have a 'renaissance' in the context of planetary governance in general and the increasing number of people who desire to be fed in a sustainable way in particular., (© 2022 The Authors. Background Gait is a complex task requiring coordinated efforts of multiple brain networks. To date, there is little evidence on whether gait is altered in cerebral amyloid angiopathy (CAA). We aimed to identify impairments in gait performance and associations between gait impairment and neuroimaging markers of CAA, cognition, and falls. Methods and Results Gait was assessed using the Zeno Walkway during preferred pace and dual task walks, and grouped into gait domains (Rhythm, Pace, Postural Control, and Variability). Participants underwent neuropsychological testing and neuroimaging. Falls and fear of falling were assessed through self-report questionnaires. Gait domain scores were standardized and analyzed using linear regression adjusting for age, sex, height, and other covariates. Participants were patients with CAA (n=29), Alzheimer disease with mild dementia (n=16), mild cognitive impairment (n=24), and normal elderly controls (n=47). CAA and Alzheimer disease had similarly impaired Rhythm, Pace, and Variability, and higher dual task cost than normal controls or mild cognitive impairment. Higher Pace score was associated with better global cognition, processing speed, and memory. Gait measures were not correlated with microbleed count or white matter hyperintensity volume. Number of falls was not associated with gait domain scores, but participants with low fear of falling had higher Pace (odds ratio [OR], 2.61 [95% CI, 1.59-4.29]) and lower Variability (OR, 1.64 [95% CI, 1.10-2.44]). Conclusions CAA is associated with slower walking, abnormal rhythm, and greater gait variability than in healthy controls. Future research is needed to identify the mechanisms underlying gait impairments in CAA, and whether they predict future falls.

Apoptosis-inducing factor (AIF) is a mitochondrial flavoprotein involved in redox signalling and programmed cell death. The role of AIF has been well recognized in diabetes and obesity. However, the aspect of AIF deficiency in the development of hepatic steatosis and liver injury is unknown. Therefore, in the current study, Harlequin (Hq mutant) mouse with markedly reduced content of AIF was investigated to explore the role of AIF on the initiation of liver injury. The wild type (WT) developed physiological and pathological features of non-alcoholic fatty liver disease (NAFLD) that were not seen in the Hq mice with AIF deficiency, when fed on high fat high fructose (HFHF) diet. Following bile duct ligation (BDL), the liver associated pathological changes were less conspicuous in Hq mice as compared to WT mice. The expression of AIF protein and apoptosis was markedly lesser as compared to their respective control in Hq mice on HFHF diet. Furthermore, the genes involved in fatty acid metabolism were also altered in the group of treated Hq mice. In conclusion, Hq mice failed to develop diet induced hepatic steatosis, suggestive of a role of AIF mediated pathway in the initiation and progression of liver inflammation. Thus, partial loss of AIF appears to be hepatoprotective. SIGNIFICANCE OF THE STUDY: AIF deficiency has multiple roles in altered pathology processes and cellular metabolism, thereby compromising the cellular homeostasis. Considering the molecular functions of AIF in other organ pathology little is known about its role in diet induced liver injury. Hence, the aim of the current study was to investigate the role of AIF deficiency in liver injury and diseases with focus on NAFLD. Most cancer cases occur in areas of low resources. The diagnosis, treatment and monitoring of cancers is especially challenging in these locations. Unique partnerships exist between non-profit organisations and pharmaceutical companies to provide free drugs to CML patients throughout the world if the diagnosis can be rigorously and unambiguously established. But there lies the rub: How do you perform molecular diagnostics in areas where even electricity is unreliable? This review reflects the presentations and discussion at the 14th post-American Society of Hematology (ASH) International Workshop on Chronic Myeloproliferative Malignancies, which took place on the December 10 and 11, 2019, immediately after the 61st ASH Annual Meeting in Orlando, Florida. Rather than present a resume of the proceedings, we address some of the topical translational science research and clinically relevant topics in detail. We consider how recent studies using single-cell genomics and other molecular methods reveal novel aspects of hematopoiesis which in turn raise the possibility of new therapeutic approaches for patients with myeloproliferative neoplasms (MPNs). We discuss how alternative therapies could benefit patients with chronic myeloid leukemia who develop BCR-ABL1 mutant subclones following ABL1-tyrosine kinase inhibitor therapy. In MPNs, we focus on efforts beyond JAK-STAT and the merits of integrating activin receptor ligand traps, interferon-α, and allografting in the current treatment algorithm for patients with myelofibrosis., (© 2020 The Authors. Measurable residual disease (MRD) that persists after initial therapy is a powerful predictor of relapse and survival in acute lymphoblastic leukemia (ALL). However, the optimal use of this information to influence therapeutic decisions is controversial. Herein, we comprehensively review the role of MRD assessment in adults with ALL, including methods to quantify residual leukemia cells during remission, prognostic impact of MRD across ALL subtypes, and available therapeutic approaches to eradicate MRD. Background: Preceding release of the 2013 American College of Cardiology (ACC)/American Heart Association (AHA) cholesterol guidelines, prescribers aimed for specific low-density lipoprotein cholesterol (LDL-C) goals in patients with atherosclerotic cardiovascular disease (ASCVD). The 2013 guidelines changed this focus to treating patients with appropriate statin intensity given their ASCVD risk. We examined statin use and LDL-C levels before and after the 2013 ACC/AHA guidelines in patients with clinical ASCVD as defined in the guidelines., Methods and Results: We conducted a retrospective cohort study of adult commercial and Medicare Advantage health plan enrollees in the Optum Research Database. Patients had ≥1 claim with a diagnosis of clinical ASCVD between November 1, 2012 and December 31, 2014 and were continuously enrolled 6 months before (baseline) and 7 months after (follow-up) the first ASCVD visit. Patients were assigned to monthly cohorts based on ASCVD event month. Statin use and intensity were measured at baseline and first month of follow-up. LDL-C changes were assessed using ordinary least squares regression. For 90 287 patients, mean (SD) age was 68 (12) years; 50% were female; and 30% had commercial insurance. Statin use remained consistent before and after guidelines (32% and 31%, respectively). Of patients receiving statins, high-intensity use increased by 4 percentage points 1 year after guidelines ( P <0.001). Mean LDL-C levels were 2.4 mmol/L (94 mg/dL) both pre- and postguidelines., Conclusions: Statin use and mean monthly LDL-C before and after the guidelines remained largely unchanged; statin intensity increased modestly. Tyrosine kinase inhibitor (TKI) therapy yields sustained cytogenetic remissions in most patients with chronic-phase chronic myeloid leukemia (CML). Peripheral blood quantitative reverse transcription polymerase chain reaction (qRT-PCR) monitoring of the chimeric BCR-ABL1 mRNA transcript levels is a very sensitive method to measure disease burden in patients with cytogenetic remission. qRT-PCR allows identification of patients (1) at high risk of progression early (3-6 months) after treatment initiation, (2) with no response to TKI therapy, (3) with undetectable disease who could be eligible for TKI discontinuation trials. Precursor B-acute lymphoblastic leukaemias (pre-B ALLs) comprise the majority of ALLs and virtually all blasts express CD22 in the cytoplasm and on the cell surface. In the present study (Southwestern Oncology Group S0910), we evaluated the addition of epratuzumab, a humanized monoclonal antibody against CD22, to the combination of clofarabine and cytarabine in adults with relapsed/refractory pre-B ALL. The response rate [complete remission and complete remission with incomplete count recovery] was 52%, significantly higher than our previous trial with clofarabine/cytarabine alone, where the response rate was 17%. This result is encouraging and suggests a potential benefit to adding epratuzumab to chemotherapy for ALL; however, a randomized trial will be needed to answer this question., (© 2014 John Wiley & Sons Ltd.)

The AF1q gene is expressed in normal haematopoietic progenitors, but less so in differentiated blood cells. In 47 patients with myelodysplastic syndrome (MDS), AF1q copy numbers were 0-6.8 x 10(6)/microg RNA compared with 1.5-2.4 x 10(5)/microg RNA in normal marrow. AF1q levels correlated with international prognostic scoring system (IPSS) scores (P = 0.004) and the risk of post-transplant relapse (P = 0.05). Among IPSS high-risk patients, survival correlated inversely with AF1q levels (P = 0.04). Thus, AF1q levels correlate with high-risk MDS and may provide a marker for risk stratification.
