On‐skin electronics can be conformably deployed on body for health monitoring, assisted living, and human/computer interfaces. However, developing corresponding energy devices is a critical challenge. Herein, a permeable and stretchable multifunctional liquid‐metal electronic skin that can generate electricity by recovering ambient electromagnetic pollution (from surrounding electrical appliances) and biomechanical energy (from body movements) is presented for epidermal energy and self‐powered sensing applications. To the best of the authors' knowledge, this is the first demonstration that a breathable on‐skin device can convert ambient electromagnetic pollution into useful electricity. The device is constructed using two stretchable microfibrous films sandwiching self‐organized mesh‐like and wrinkled liquid‐metal that affords electricity via induced electrification (±9.3 V, ±1.7 µA; 60 Hz) and triboelectricity (205.6 V, ±2.3 µA; 4 Hz). Its applicability in powering electronic devices is demonstrated. Moreover, it can serve as an epidermal self‐powered sensor for continuously monitoring whole‐body physiological signals and motions of the eyelids, face, throat, chest, and limbs, thus demonstrating its potential to remotely collect clinical and biomechanical information. Finally, it is used as an interface in diverse system‐level applications. Background: There is an increasing interest in using machine learning and artificial intelligence to estimate chronological age using neuroimaging data. The gap between chronological age and estimated brain age (brain age gap, BAG) is used as a measure of accelerated/resilient brain aging. Previously, BAG has been associated with cognitive status. However, whether the BAG varies across sex and cognitive status have been less explored. The present study examines these associations and validates a voxel‐based machine learning approach based on the elastic net regression (ENR) for BAG calculation. Method: Using data from the Atherosclerosis Risk in Communities Study (ARIC) study, the Wake Forest School of Medicine Alzheimer's Disease Research Center (WFSM‐ADRC) clinical cohort and Alzheimer's Disease Neuroimaging Initiative (ADNI), we examined associations of BAG across cognitive status and sex. We used structural MRI scans from 1853 ARIC participants (ages 67‐90, 60% females), 508 from the WFSM‐ADRC (55‐95 yo., 66% females) and 584 ADNI cognitively normal (CN) participants (55‐90 yo., 57% females). All images were aligned into a common template and the derived gray matter (GM) probability maps from ADNI MRIs were used as input to train the machine learning algorithm. Once the model was fitted the ARIC and WFSM‐ADRC GM probability maps were provided as input to the algorithm to estimate the BAG values. Finally, an age bias correction was applied. Linear regression methods were used to investigate differences between groups. Age, race, education, sex, and cognitive status were included in the model. Result: We found in both ARIC and WFSM‐ADRC participants that differences in BAG values between CN‐MCI and MCI‐Dementia participants were highly significant. In addition, when we examined differences in BAG values across sex per cognitive status, we found again in both cohorts that differences were only significant for CN individuals. Conclusion: Our analyses show that our approach to estimate chronological age using high‐dimensional ENR, produces BAG values which are strongly associated with cognitive status. Higher data transmission rates, broader bandwidths, better privacy, and security are indispensable for the current information‐explosive age. However, to achieve the desired functionalities still remains as a great challenge. In this study, a two‐terminal, vertical dual‐gate phototransistor, which can be controlled by light, electric and magnetic fields, is proposed, fabricated, and demonstrated. This phototransistor uses a tandem structure composed of an organic solar cell (OSC) of ITO/ZnO/P3HT:PC61BM/MoO3/Ag, a resistive random access memory (RRAM) of Ag/PMMA/Au and a magnetoelectric film with micropyramid structure of PDMS:FeNi/silver nanowires (AgNWs). This device integrates and couples the properties of each individual component enabling to respond to light, electric and magnetic fields and works as a dual‐gate transistor. Compared with conventional transistors, the proposed device carries several intriguing features, such as ultrafast photoresponse time of 3 µs, controllable photocurrent with a variety of external stimuli, high ON/OFF ratio greater than 103, and touchless human–machine interaction. All of the above features are beneficial for high‐speed optical communication and circuit miniaturization. Background: Brain tumor therapeutic responses can be quantified from magnetic resonance images (MRI) using 1‐ (1D) and 2‐dimensional (2D) linear and volumetric methods, but few studies in dogs compare these techniques. Hypotheses: Linear methods will be obtained faster, but have less agreement than volumetric measurements. Therapeutic response agreement will be highest with the total T2W tumor volumetric (TTV) method. Therapeutic response at 6‐weeks will correlate with overall survival (OS). Animals: Forty‐six dogs with intracranial gliomas. Methods: Prospective study. Three raters measured tumors using 1D and 2D linear, TTV, and contrast‐enhancing volumetric (CEV) techniques on 143 brain MRI to determine agreement between methods, define therapeutic responses, and assess relations with OS. Results: Raters performed 1D the fastest (2.9 ± 0.57 minutes) and CEV slowest (17.8 ± 6.2 minutes). Inter‐ and intraobserver agreements were excellent (intraclass correlations ≥.91) across methods. Correlations between linear (1D vs 2D; ρ >.91) and volumetric (TTV vs CEV; ρ >.73) methods were stronger than linear to volumetric comparisons (ρ range,.26‐.59). Incorporating clinical and imaging data resulted in fewer discordant therapeutic responses across methods. Dogs having partial tumor responses at 6 weeks had a lower death hazard than dogs with stable or progressive disease when assessed using 2D, CEV, and TTV (hazard ration 2.1; 95% confidence interval, 1.22‐3.63; P =.008). Conclusions and Clinical Importance: One‐dimensional, 2D, CEV, and TTV are comparable for determining therapeutic response. Introduction: A data‐driven index of dementia risk based on magnetic resonance imaging (MRI), the Alzheimer's Disease Pattern Similarity (AD‐PS) score, was estimated for participants in the Atherosclerosis Risk in Communities (ARIC) study. Methods: AD‐PS scores were generated for 839 cognitively non‐impaired individuals with a mean follow‐up of 4.86 years. The scores and a hypothesis‐driven volumetric measure based on several brain regions susceptible to AD were compared as predictors of incident cognitive impairment in different settings. Results: Logistic regression analyses suggest the data‐driven AD‐PS scores to be more predictive of incident cognitive impairment than its counterpart. Both biomarkers were more predictive of incident cognitive impairment in participants who were White, female, and apolipoprotein E gene (APOE) ε4 carriers. Random forest analyses including predictors from different domains ranked the AD‐PS scores as the most relevant MRI predictor of cognitive impairment. Background: Seizures in the early postoperative period after intracranial surgery may affect outcome in dogs. Objectives: To determine the incidence of early postoperative seizures (EPS) in dogs with brain tumors, identify specific risk factors for EPS, and determine if EPS affects outcome. Animals: Eighty‐eight dogs that underwent 125 intracranial surgeries for diagnosis and treatment of rostrotentorial brain tumors. Methods: Retrospective cohort study. All patients with a diagnosis of rostrotentorial brain tumor from 2006 to 2020 were included. Early postoperative seizures were diagnosed by observation of seizure activity within 14 days of neurosurgery. Previously diagnosed structural epilepsy, perioperative anticonvulsant drug (ACD) use, magnetic resonance imaging (MRI), and tumor characteristics were evaluated. Outcome measures included neurologic and nonneurologic complications, duration of hospitalization, and survival to discharge. Results: Dogs with rostrotentorial brain tumors had EPS after 16/125 (12.8%) neurosurgical procedures (95% confidence interval [CI], 7%‐19%). Presence of previous structural epilepsy was not associated with EPS risk (P = 1). Perioperative ACD use also was not associated with EPS (P =.06). Dogs with EPS had longer hospitalization (P <.001), were more likely to have neurologic complications postsurgery (P =.01), and were less likely to survive to discharge (P =.01). Conclusions and Clinical Importance: It is difficult to predict which dogs are at risk of EPS because the presence of previous structural epilepsy and the use of perioperative ACDs was not associated with EPS. However, seizures in the early postoperative period are clinically important because affected dogs had prolonged hospitalization, more neurologic complications, and decreased short‐term survival. Background: Our study aim was to evaluate neuromuscular ultrasound (NMUS) for the assessment of taxane chemotherapy-induced peripheral neuropathy (CIPN), the dose-limiting toxicity of this agent.Methods: This cross-sectional study of breast cancer patients with taxane CIPN measured nerve cross-sectional area (CSA) by NMUS and compared with healthy historical controls. Correlations were determined between CSA and symptom scale, nerve conduction studies, and intraepidermal nerve fiber density (IENFD).Results: A total of 20 participants reported moderate CIPN symptoms at a median of 3.8 months following the last taxane dose. Sural nerve CSA was 1.2 mm2 smaller than healthy controls (P ≤ .01). Older age and time since taxane were associated with smaller sural nerve CSA. For each 1 mm2 decrease in sural nerve CSA, distal IENFD decreased by 2.1 nerve/mm (R2 0.30; P = .04).Conclusions: These data support a sensory predominant taxane neuropathy or neuronopathy and warrant future research on longitudinal NMUS assessment of CIPN. Crystallinity and crystal orientation have a predominant impact on a materials' semiconducting properties, thus it is essential to manipulate the microstructure arrangements for desired semiconducting device performance. Here, ultra‐uniform hole‐transporting material (HTM) by self‐assembling COOH‐functionalized P3HT (P3HT‐COOH) is fabricated, on which near single crystal quality perovskite thin film can be grown. In particular, the self‐assembly approach facilitates the P3HT‐COOH molecules to form an ordered and homogeneous monolayer on top of the indium tin oxide (ITO) electrode facilitate the perovskite crystalline film growth with high quality and preferred orientations. After detailed spectroscopy and device characterizations, it is found that the carboxylic acid anchoring groups can down‐shift the work function and passivate the ITO surface, retarding the interface carrier recombination. As a result, the device made with the self‐assembled HTM show high open‐circuit voltage over 1.10 V and extend the lifetime over 4,300 h when storing at 30% relative humidity. Moreover, the cell works efficiently under much reduced light power, making it useful as power source under dim‐light conditions. The demonstration suggests a new facile way of fabricating monolayer HTM for high efficiency perovskite devices, as well as the interconnecting layer needed for tandem cell. [ABSTRACT FROM AUTHOR]

Background: Stereotactic brain biopsy (SBB) allows for histopathologic diagnosis of brain tumors. Adverse events (AE) occur in 5 to 29% of dogs after SBB, but risk factors associated with developing AE are poorly described. Objective: Identify clinicopathologic, diagnostic imaging, or procedural variables that are associated with AE in dogs after SBB. Animals Twenty‐nine dogs with brain tumors. Methods: Retrospective, case‐control study. Dogs had laboratory investigations performed before SBB, as well as clinical examinations and diagnostic imaging of the brain before and after SBB. Cases experienced AE after SBB including transient exacerbation of preexisting neurologic deficits, transient new deficits, or permanent neurologic deficits. Controls had SBB performed without AE. Fisher's exact and Student's t tests were used to examine associations between the postulated risk factors and AE. Results: Adverse events occurred in 8/29 (27%) dogs, and 7/8 AE (88%) were transient. Cases were significantly more likely to have T2W‐heterogenous tumors (88 versus 38%; P =.04) and lower platelet counts (194.75 ± 108.32 versus 284.29 ± 68.54 ×103/mm3, P =.006). Dogs with gradient echo signal voids present on baseline imaging were significantly more likely to have hemorrhage present after biopsy, and 7/8 (88%) of cases had hemorrhage on imaging after SBB. Conclusion and Clinical Importance: Twenty‐seven percent of dogs undergoing SBB experience AE, with the majority of AE resolving with 1 week. Platelet counts should be ≥185 000/mm3 to minimize risk of SBB‐associated AE. Background: Intracranial hypertension (ICH) is often presumptively diagnosed based on clinical or imaging findings. Clinical or imaging surrogates of ICH are not usually validated with reference standard direct intracranial pressure (dICP) recordings. Hypotheses: Dogs with brain magnetic resonance imaging (MRI) or clinical features of presumed ICH would have higher dICP than dogs lacking those features. Animals Twenty dogs with gliomas and 3 normal controls. Methods: Prospective, convenience study. Dogs were presumptively categorized with normal ICP or ICH from scores generated from described clinical and brain MRI indicators of ICH. dICP was recorded in anesthetized dogs using an intraparenchymal microsensor and compared between groups. Results: dICP was not different between control (10.4 ± 2.1 mm Hg) and dogs with glioma (15.6 ± 8.3 mm Hg), or between dogs in clinically predicted ICP groups. Compared with dogs with MRI‐predicted normal ICP, MRI‐predicted ICH dogs had higher dICP (10.3 ± 4.1 versus 19.2 ± 7.9 mm Hg, P =.004), larger tumors (1.45 ± 1.2 versus 5.71 ± 3.03 cm3, P =.0004), larger optic nerve sheath diameters, and 14/14 (100%) displayed structural anatomical shifts on MRI. At a dICP threshold of 15 mm Hg, the sensitivity of MRI for predicting ICH was 90% and the specificity 69%. Conclusions and Clinical Relevance: dICP measurements are feasible in dogs with brain tumors. MRI features including brain herniations, mass effect, and optic nerve size aid in the identification of dogs with ICH. Background: MRI is a widely used modality to evaluate dementia risk. Here, we extend previous work by estimating the Alzheimer's disease pattern similarity (AD‐PS) scores for participants of a community‐dwelling older adult cohort to evaluate neuroanatomic risk of AD using MRI. Method: Baseline MRI data were available from 522 participants from the Wake Forest Alzheimer's Disease Research Center's (ADRC) Clinical Core with mean age 70.2, 81.2% White and 17.6% African American. Of those, 253 were cognitively impaired (dementia and mild cognitive impairment (MCI)) and 138 underwent [11C]PiB PET (MCI = 48 and dementia = 19). Global PiB SUVR was averaged from a cortical region of interest sensitive to early AD. Participants were classified as amyloid positivity using a previously defined threshold (≥1.21 SUVR). AD‐PS scores were estimated based on GM probability maps according to previously reported machine learning methodology[1]. High‐dimensional classifiers were trained using ADNI MRI data. To generate the AD‐PS scores, MRI data from the ADRC cohort was provided as input to the classifiers. We investigated associations that AD‐PS scores [range 0‐1] had with amyloid positivity and cognitive impairment. All analyses were adjusted for age, sex, race and education. AD‐PS scores discrimination of participants with dementia versus normal cognition (CN) was compared to hippocampal volume and cortical thickness in temporal regions based meta‐ROI which are sensitive to AD. Result: AD‐PS scores were strongly associated with cognitive impairment: OR = 4.1, CI: 95% [2.9‐5.9], p<0.0001. In addition, they were associated with amyloid positivity with OR = 2.8,CI: 95% [1.7‐4.7], p<0.001, N = 138. These amyloid positivity results did not hold in analyses limited to CN participants only(N = 71). Performance when discriminating patients with dementia from CN individuals was highest for AD‐PS score (AUC = 92.2, CI: 95% [86.9‐97.5]), followed by hippocampal volume (AUC = 86.5, CI: 95% [81.1‐91.2]) and cortical thickness in temporal regions based meta‐ROI (AUC = 81.5 CI: 95% [74.4‐88.6], See‐Figure). Conclusion: This work highlights further the generalization performance of our machine learning algorithm. Background: Stereotactic brain biopsy (SBB) is a technique that allows for definitive diagnosis of brain lesions. Little information is available regarding the diagnostic utility of SBB in dogs with intracranial diseases. Objective: To investigate the diagnostic accuracy (DA) of SBB in dogs with brain tumors. Animals: Thirty‐one client‐owned dogs that underwent SBB followed by surgical resection or necropsy examinations. Methods: Retrospective observational study. Two pathologists blinded to SBB and reference standard diagnoses reviewed histologic specimens and typed and graded tumors according to World Health Organization and revised canine glioma classification criteria. Agreement between tumor type and grade from SBB were compared to reference standards and assessed using kappa statistics. Patient and technical factors associated with agreement also were examined. Results: Stereotactic brain biopsy specimens were obtained from 24 dogs with gliomas and 7 with meningiomas. Tumor type agreement between SBB and the reference standard was observed in 30/31 cases (κ = 0.95). Diagnostic concordance was perfect for meningiomas. Grade agreement among gliomas was observed in 18/23 cases (κ = 0.47). Stereotactic brain biopsy underrepresented the reference standard glioma grade in cases with disagreement. The DA of SBB was 81%, with agreement noted in 56/69 biopsy samples. Smaller tumors and fewer SBB specimens obtained were significantly associated with diagnostic discordance. Conclusions and Clinical Importance: The DA of SBB readily allows for the diagnosis of common brain tumors in dogs. Although glioma grade discordance was frequent, diagnoses obtained from SBB are sufficient to currently inform therapeutic decisions. Multiple SBB specimens should be collected to maximize DA. OBJECTIVES To evaluate the effect of hospitalizations on patterns of sedentary and physical activity time in mobility‐limited older adults randomized to structured physical activity or health education. DESIGN Secondary analysis of investigator‐blinded, parallel‐group, randomized trial conducted at 8 U.S. centers between February 2010 and December 2013. PARTICIPANTS Sedentary men and women aged 70 to 89 at baseline who wore a hip‐fitted accelerometer 7 consecutive days at baseline and 6, 12, and 24 months after randomization (N=1,341). MEASUREMENTS Participants were randomized to a physical activity (PA; n = 669) intervention that included aerobic, resistance, and flexibility training or to a health education (HE; n = 672) intervention that consisted of workshops on older adult health and light upper‐extremity stretching. Accelerometer patterns were characterized as bouts of sedentary (<100 counts/min; ≥1, ≥10, ≥30, ≥60 minute lengths) and activity (≥100 counts/min; ≥1, ≥2, ≥5, ≥10 minute lengths) time. Each participant was categorized as having 0, 1 to 3, or 4 or more cumulative hospital days before each accelerometer assessment. RESULTS: Hospitalization increased sedentary time similarly in both intervention groups (8 min/d for 1–3 cumulative hospital days and 16 min/d for ≥4 cumulative hospital days). Hospitalization was also associated with less physical activity time across all bouts of less than 10 minutes (≥1: −7 min/d for 1–3 cumulative hospital days, –16 min/d for ≥4 cumulative hospital days; ≥2: −5 min/d for 1–3 cumulative hospital days, −11 min/d for ≥4 cumulative hospital days; ≥5: −3 min/d for 1–3 cumulative hospital days, −4 min/d for ≥4 cumulative hospital days). There was no evidence of recovery to prehospitalization levels (time effect p >.41). PA participants had less sedentary time in bouts of less than 30 minutes than HE participants (−8 to −10 min/d) and more total activity (+3 to +6 min/d), although hospital‐related changes were similar between the intervention groups (interaction effect p >.26). CONCLUSION: Participating in a PA intervention before hospitalization had expected benefits, but participants remained susceptible to hospitalization's detrimental effects on their daily activity levels. Objectives To examine associations between objectively measured physical activity ( PA) and incidence of major mobility disability ( MMD) and persistent MMD ( PMMD) in older adults in the Lifestyle Interventions and Independence for Elders ( LIFE) Study. Design Prospective cohort of individuals aged 65 and older undergoing structured PA intervention or health education. Setting The LIFE Study was a multicenter (eight sites) randomized controlled trial designed to compare the efficacy of a long-term structured PA intervention with that of a health education (HE) program in reducing the incidence of MMD in mobility-limited older adults. Participants LIFE Study participants (n = 1,590) had a mean age±standard deviation of 78.9 ± 5.2, low levels of PA, and measured mobility-relevant functional impairment at baseline. Measurements Activity data were collected using hip-worn 7-day accelerometers at baseline and 6, 12, and 24 months after randomization to test for associations with incident MMD and PMMD (≥2 consecutive instances of MMD). Results At baseline, every 30 minutes spent being sedentary (<100 accelerometry counts per minute) was associated with higher rate of subsequent MMD (10%) and PMMD (11%) events. Every 500 steps taken was associated with lower rate of MMD (15%) and PMMD (18%). Similar associations were observed when fitting accelerometry-based PA as a time-dependent variable. Conclusion Accelerometry-based PA levels were strongly associated with MMD and PMMD events in older adults with limited mobility. These results support the importance of daily PA and lower amounts of sedentary time levels in this population and suggest that accelerometry may be a useful tool for assessing risk of mobility disability. Background Physical activity ( PA) reduces the rate of mobility disability, compared with health education ( HE), in at risk older adults. It is important to understand aspects of performance contributing to this benefit. Objective To evaluate intervention effects on tertiary physical performance outcomes. Design The Lifestyle Interventions and Independence for Elders ( LIFE) was a multi-centered, single-blind randomized trial of older adults. Setting Eight field centers throughout the United States. Participants 1635 adults aged 78.9 ± 5.2 years, 67.2% women at risk for mobility disability (Short Physical Performance Battery [SPPB] <10). Interventions Moderate PA including walking, resistance and balance training compared with HE consisting of topics relevant to older adults. Outcomes Grip strength, SPPB score and its components (balance, 4 m gait speed, and chair-stands), as well as 400 m walking speed. Results Total SPPB score was higher in PA versus HE across all follow-up times (overall P = .04) as was the chair-stand component (overall P < .001). No intervention effects were observed for balance (overall P = .12), 4 m gait speed (overall P = .78), or grip strength (overall P = .62). However, 400 m walking speed was faster in PA versus HE group (overall P =<.001). In separate models, 29% of the rate reduction of major mobility disability in the PA versus HE group was explained by change in SPPB score, while 39% was explained by change in the chair stand component. Conclusion Lower extremity performance ( SPPB) was significantly higher in the PA compared with HE group. Changes in chair-stand score explained a considerable portion of the effect of PA on the reduction of major mobility disability-consistent with the idea that preserving muscle strength/power may be important for the prevention of major mobility disability. Background: Prostate-specific antigen (PSA) screening may reduce death due to prostate cancer but leads to the overdiagnosis of many cases of indolent cancer. Targeted use of PSA screening may reduce overdiagnosis. Multimarker genomic testing shows promise for risk assessment and could be used to target PSA screening.Methods: To test whether counseling based on the family history (FH) and counseling based on a genetic risk score (GRS) plus FH would differentially affect subsequent PSA screening at 3 months (primary outcome), a randomized trial of FH versus GRS plus FH was conducted with 700 whites aged 40 to 49 years without prior PSA screening. Secondary outcomes included anxiety, recall, physician discussion at 3 months, and PSA screening at 3 years. Pictographs versus numeric presentations of genetic risk were also evaluated.Results: At 3 months, no significant differences were observed in the rates of PSA screening between the FH arm (2.1%) and the GRS-FH arm (4.5% with GRS-FH vs. 2.1% with FH: χ2 = 3.13, P = .077); however, PSA screening rates at 3 months significantly increased with given risk in the GRS-FH arm (P = .013). Similar results were observed for discussions with physicians at 3 months and PSA screening at 3 years. Average anxiety levels decreased after the individual cancer risk was provided (P = .0007), with no differences between groups. Visual presentation by pictographs did not significantly alter comprehension or anxiety.Conclusions: This is likely the first randomized trial of multimarker genomic testing to report genomic targeting of cancer screening. Objective: Assess cross-sectional relationships between body mass index (BMI), waist circumference (WC), pericardial (PAT), visceral (VAT), and subcutaneous adipose tissue (SAT) volumes with calcified plaque (CP) in African Americans (AAs) and European Americans (EAs) with type 2 diabetes.Methods: Computed tomography measured PAT, VAT, SAT, and CP in coronary arteries (CAC), carotid arteries, and aorta. Generalized estimating equations models were fitted to test for associations between adiposity and CP, stratified by ethnicity while accounting for familial correlations.Results: AAs (N = 753) vs. EAs (N = 562) had significantly lower PAT and VAT, despite equal or higher BMI. In multivariable models adjusting for age, gender, education, HbA1c, statins, smoking, cardiovascular disease, hypertension, nephropathy, and C-reactive protein, PAT positively associated with presence of CAC in AAs (P < 0.001), not EAs (P = 0.68; ethnicity interaction P < 0.01). Inverse associations were detected between SAT and severity of aorta CP (P < 0.01) in AAs and between BMI, WC, and SAT with severity of aorta CP in all participants.Conclusions: Ethnic- and gender-specific differences in BMI, WC, PAT, SAT, and VAT were present in AAs and EAs with diabetes. Background: Recently, we compared the effect of a high‐fat and low‐carb Modified Mediterranean‐Ketogenic Diet (MMKD) and a low‐fat American Heart Association Diet (AHAD) on 11 cognitively normal (CN) and 9 with mild cognitive impairment (MCI) older adults as a suggestive preventative intervention for Alzheimer's disease (AD). We reported the promising effect of the MMKD on MCI patients as it was well‐tolerated and associated with increased cerebrospinal fluid (CSF) Aβ1‐42, decreased tau, and increased cerebral perfusion. Here, we characterized the plasma neuron‐derived exosomes (NDE) to further understand the molecular mechanisms underlying the efficacy of the MMKD against MCI. Method: The completed study consisted of a randomized crossover design in which participants (both CN and MCI) consumed either the MMKD or the control AHAD for six weeks. Total exosomes (TE) were isolated from plasma by precipitation method, and NDE were enriched using Biotin‐tagged CD171 antibody attached with streptavidin‐magnetic beads. The purity, concentration, and size of TE and NDE were assessed by flow cytometry, nanoparticle tracking, immunogold‐labeling, electron microscopy, and exosome antibody array. NDE was characterized for several AD and neurodegeneration‐related biomarkers, advanced glycation endproducts (AGEs), glutamate, glutamate receptors, and β‐hydroxybutyrate (BHB) by ELISA and colorimetric assays. NDE effect on NF‐kB activity in human monocytes was studied as a molecular surrogate for inflammation. Result: We confirmed the size, shape, and purity of both TE and NDE by multiple methods. Importantly, the MMKD treatment strongly reduced the level of Aβ1‐42 (by 50%, p=0.001), p(181)Tau (by 17%), and neurofilament L (by 55%, p=0.03) in the MCI group, but not with AHAD. MMKD reduced the AGEs in both CN and MCI groups and strongly increased the glutamate level in NDEs from the MCI group (6 fold, p<0.001) but did not affect glutamate in CSF. The MMKD also modulated the expression of various glutamate receptors (GRIN1, GRIA1, GRIN2A, and GRIN2b). Interestingly, we detected BHB in NDEs, though no significant change was observed. MMKD inhibited the NF‐κB activity in monocytes suggesting a strong anti‐inflammatory effect. Background: MRI based biomarkers are of increasing interest due to availability, low cost, and low patient burden compared to other biomarkers. Alzheimer's disease pattern similarity (AD‐PS) scores, an MRI‐based AD risk measure, have been previously investigated in predominantly white cohorts. The Atherosclerosis Risk in Communities (ARIC) study provides an opportunity to further evaluate the AD‐PS score in a diverse setting. Method: The ARIC study has collected cognitive data and MRI scans at visit 5 (2011‐2013) and visit 6 (2015‐2017). Participants were aged 76.0 (5.3) yrs at visit 5 and included both blacks and whites. MRI scans from the ARIC and Alzheimer's Disease Neuroimaging Initiative (ADNI) studies were aligned and segmented using the Advanced Normalization Toolbox software. A high‐dimensional classifier was trained using gray matter images (N = 598) from the ADNI study. Then ARIC MRI images were provided as input to the classifier to obtain the scores. Incident cognitive impairment (ICI) analyses were based on individuals diagnosed as cognitively normal (CN) (N = 868) at visit 5. Logistic regression considered ICI (either a diagnosis of mild cognitive impairment or dementia at visit 6) as the outcome. In PET analyses logistic regression was used to investigate associations of the AD‐PS scores with cross‐sectional β‐amyloid lesion burden (global cortical standardized uptake value ratio dichotomized at 1.2) derived from F18‐Florbetapir PET images available in a subset (N = 330, MCI=89). All analyses were adjusted for age, race, sex, education, site, and intracranial volume. The relevant variables were standardized. Analyses were further stratified by race and sex. Result: Among participants who were CN at visit 5, 125 were diagnosed with MCI and 28 with dementia at visit 6. The AD‐PS scores were significantly associated with ICI in: the full sample, whites, blacks, males and females (Table 1). The scores were also significantly associated with β‐amyloid burden in: the full sample, blacks and males. Conclusion: AD‐PS scores, an MRI based biomarker of AD, were associated with ICI and β‐amyloid burden derived from PET in the ARIC study. We report two methods for linkage disequilibrium mapping that involve incorporation of covariates through parametric modeling to utilize combined case-parent trios and unrelated case and/or control data. The proposed two combined methods were used to map the disease locus of hypertension in the angiotensin-converting enzyme ( ACE) gene with incorporation of ACE activity. The efficiencies in estimating the disease locus increased by 351- and 100-fold in the hybrid study with respect to the two proposed methods when compared to the estimates from the trios study; and they changed by 1.4- and 0.4-fold, respectively, when compared to the case-control study. Efficiency of disease locus estimates was greatly improved in both simulations and hypertension studies based on the hybrid data, compared to case-parent trio studies only. These newly developed methods preserve the advantages of the previous methods, including flexible modeling and assessment of gene-gene and gene-covariate effects, while providing more power by using all the data combined. Objectives To assess the association between angiotensin converting enzyme inhibitors ( ACEis) and improvements in the physical function of older adults in response to chronic exercise training. Design Secondary analysis of the Lifestyle Interventions and Independence for Elders Pilot ( LIFE-P) study, a multisite randomized clinical trial to evaluate the effects of chronic exercise on the physical function of older adults at risk for mobility disability. Setting Four academic research centers within the United States. Participants Four hundred twenty-four individuals aged 70 to 89 with mild to moderate functional impairments categorized for this analysis as ACEi users, users of other antihypertensive drugs, or antihypertensive nonusers. Intervention A 12-month intervention of structured physical activity ( PA) or health education promoting successful aging ( SA). Measurements Change in walking speed during a 400-m test and performance on a battery of short-duration mobility tasks (Short Physical Performance Battery ( SPPB)). Results Physical activity significantly improved the adjusted walking speed of ACEi users ( P < .001) but did not of nonusers. PA improved the adjusted SPPB score of ACEi users ( P < .001) and of persons who used other antihypertensive drugs ( P = .005) but not of antihypertensive nonusers ( P = .91).The percentage of ACEi users deriving clinically significant benefit from exercise training for walking speed (30%) and SPPB score (48%) was dramatically higher than for nonusers (14% and 12%, respectively). Conclusion For older adults at risk for disability, exercise-derived improvements in physical function were greater for ACEi users than users of other antihypertensive drugs and antihypertensive nonusers. Insulin resistance is linked to general and abdominal obesity, but its relation to hepatic lipid content and pericardial adipose tissue is less clear. The purpose of this study was to examine cross-sectional associations of liver attenuation, pericardial adipose tissue, BMI, and waist circumference with insulin resistance. We measured liver attenuation and pericardial adipose tissue using the existing cardiac computed tomography scans in 5,291 individuals free of clinical cardiovascular disease and diabetes in the Multi-Ethnic Study of Atherosclerosis (MESA) during the study's baseline visit (2000-2002). Low liver attenuation was defined as the lowest quartile and high pericardial adipose tissue as the upper quartile of volume (cm3). We used standard clinical definitions for obesity and abdominal obesity. Insulin resistance was assessed by the homeostasis model assessment of insulin resistance (HOMAIR) index. In multivariate linear regression with all adiposity measures in the model simultaneously, all adiposity measures were significantly (P < 0.0001) associated with insulin resistance: regression coefficients (±s.e.) were 0.31 (±0.02) for low liver attenuation, 0.27 (±0.02) for high pericardial adipose tissue, 0.27 (±0.02) for obesity, and 0.32 (±0.02) for abdominal obesity. We found significant differences (P = 0.003) between standardized liver attenuation and insulin resistance by ethnicity: regression coefficients per 1 s.d. increment were 0.10 ± 0.01 for whites, 0.11 ± 0.02 for Chinese, 0.08 ± 0.2 for blacks, and 0.14 ± 0.01 for Hispanics. To determine whether the presence of high depressive symptoms (Center for Epidemiologic Studies Depression Scale (CES-D) score ≥14) diminished physical performance benefits after a comprehensive physical activity intervention in older adults. A post hoc analysis of data from the Lifestyle Interventions and Independence for Elders Pilot (LIFE-P) study, a single-blind randomized controlled trial comparing a moderate-intensity physical activity intervention (PA) with a successful aging control (SA). Multicenter U.S. institutions participating in the LIFE-P trial. Four hundred twenty-four sedentary, noninstitutionalized adults aged 70 to 89. Depressive symptoms were assessed using the CES-D. Physical performance tests included the Short Physical Performance Battery (SPPB) and 400-m walk time (400 mw) at baseline and 6 and 12 months. Of the participants, 15.8% had high depressive symptom scores (CES-D ≥14). For participants with low depressive symptoms, SPPB scores improved more in the PA than the SA group over 12 months (adjusted score difference +0.70; P<.001 at 6 months and +0.58; P=.004 at 12 months), and 400 mw times improved in the PA group at 6 months (adjusted score difference −0.41 minutes; P=.02). For participants with high depressive symptoms, the difference in improvement fell short of statistical significance on the SPPB between the PA and SA groups (adjusted score difference +0.76 ( P=.18) at 6 months and +0.94 ( P=.12) at 12 months). OBJECTIVES: To determine the effects of a long-term exercise intervention on two prominent biomarkers of inflammation (C-reactive protein (CRP) and interleukin-6 (IL-6)) in elderly men and women. DESIGN: Single-blind, randomized, controlled trial: The Lifestyle Interventions and Independence for Elders (LIFE) Trial. SETTING: The Cooper Institute, Dallas, Texas; Stanford University, Stanford, California; University of Pittsburgh, Pittsburgh, Pennsylvania; and Wake Forest University, Winston-Salem, North Carolina. PARTICIPANTS: Four hundred twenty-four elderly (aged 70–89), nondisabled, community-dwelling men and women at risk for physical disability. INTERVENTION: A 12-month moderate-intensity physical activity (PA) intervention and a successful aging (SA) health education intervention. MEASUREMENTS: CRP and IL-6. RESULTS: After adjustment for baseline IL-6, sex, clinic site, diabetes mellitus, treatment group, visit, and group-by-visit interaction, the PA intervention resulted in a lower ( P=.02) IL-6 concentration than the SA intervention. Adjusted mean IL-6 at month 12 was 8.5% (0.21 pg/mL) higher in the SA than the PA group. There were no significant differences in CRP between the groups at 12 months ( P=.09). Marginally significant interaction effects of the PA intervention according to baseline functional status ( P=.05) and IL-6 (above vs below the median; P=.06) were observed. There was a greater effect of the PA intervention on participants with lower functional status and those with a higher baseline IL-6. CONCLUSION: Greater PA results in lower systemic concentrations of IL-6 in elderly individuals, and this benefit is most pronounced in individuals at the greatest risk for disability and subsequent loss of independence. OBJECTIVES: To describe several recruitment parameters derived from the Lifestyle Interventions and Independence for Elders pilot (LIFE-P) study for use in a full-scale trial of mobility disability prevention. DESIGN: A description of the recruiting methods and baseline characteristics of a four-site randomized, controlled trial testing the effectiveness of a physical activity intervention at preventing mobility disability. SETTING: The Cooper Institute, Dallas, Texas; Stanford University, Stanford, California; University of Pittsburgh, Pittsburgh, Pennsylvania; and Wake Forest University, Winston-Salem, North Carolina. PARTICIPANTS: Community-living persons aged 70 to 89 who were able to walk 400 m within 15 minutes and were at high risk for disability (scoring<10 on the Short Physical Performance Battery (SPPB)) but without comorbidity severe enough to preclude full study participation. MEASUREMENTS: Measures of efficiency included number of randomized participants per recruitment technique and costs per randomized participant across randomization techniques. RESULTS: The 9-month recruiting period resulted in 3,141 telephone screens, of which 424 (13.5%) participants were randomized (68.9% women, 25.7% minorities, 41.5% with SPPB scores<8). Forty percent of telephone-screened participants were excluded primarily because of regular participation in physical activity, health exclusions, or self-reported mobility disability. Of the 1,252 persons attempting the physical performance assessments, 41% scored above the SPPB cutoff. Of the 566 remaining eligible, 9.9% could not complete the 400-m walk, and another 18.9% had various medical exclusions. Direct mailing was the most productive recruitment strategy (61.6% of all randomized participants). Recruitment cost approximately $439 per randomized participant. CONCLUSION: The LIFE study achieved all recruitment goals and demonstrated the feasibility of recruiting high-risk community-dwelling older persons for trials of disability prevention in diverse geographic areas. Triboelectric nanogenerators (TENGs) have revealed fascinating potential in multifaceted wearables. However, their reliance on physical contact and separation from human bodies presents drawbacks for efficient energy collection, especially over large areas. Herein, the first stretchable non‐contact TENG textile is proposed using a topological insulator nanocomposite coating (triboelectric layer) and a graphite‐like fabric (tribo‐charges reservoir). This design encompasses the capture, transporting, and storage of tribo‐charges, leading to enhanced device performance (452 V, 1.96 mA  m−2, and 179 mW m−2) and prolonged tribo‐charges retention time (5000 min). During non‐contact operation, the output remained at 382 V (318 µA m−2), 123 V (59 µA m−2), and 94 V (23 µA m−2) for separation distances of 0.1, 0.5, and 1 cm, respectively. Additionally, it exhibits excellent stretchability (>100% strain). Notably, its performance during non‐contact operation and mechanical freedom surpasses those of previous reports, enabling both wearable non‐contact biomechanical energy harvesting and deformable self‐powered proximity sensing. Its applicability is comprehensively examined for non‐contact harvesting of body‐motion energy in garments and driving electronics. Finally, its application as a self‐powered touchless interface for system‐level applications is demonstrated. These results provide new directions for developing non‐contact biomechanical energy harvesting and sensing, enabling advancements in autonomous wearables and Metaverse applications.
