The S1222a marsquake detected by InSight on 4 May 2022 was the largest of the mission, at MwMa ${M}_{w}^{Ma}$ 4.7. Given its resemblance to two other large seismic events (S1000a and S1094b), which were associated with the formation of fresh craters, we undertook a search for a fresh crater associated with S1222a. Such a crater would be expected to be ∼300 m in diameter and have a blast zone on the order of 180 km across. Orbital images were targeted and searched as part of an international, multi‐mission effort. Comprehensive analysis of the area using low‐ and medium‐resolution images reveals no relevant transient atmospheric phenomena and no fresh blast zone. High‐resolution coverage of the epicentral area from most spacecraft are more limited, but no fresh crater or other evidence of a new impact have been identified in those images either. We thus conclude that the S1222a event was highly likely of tectonic origin. Plain Language Summary: During its time on Mars, NASA's InSight (Interior Exploration using Seismic Investigations, Geodesy and Heat Transport) mission recorded over 1,300 seismic events, known as "marsquakes." Of these, a number were identified as coming from meteoroid impact cratering events on the surface. The largest event identified by InSight, labeled S1222a, bore some similarities to two large impact events recorded earlier in the mission. In order to investigate whether the S1222a event might also have been caused by an impact event, we undertook a comprehensive search of the region in which the marsquake occurred. We did not identify any fresh craters in the area, implying that the marsquake was likely caused by geological processes. The domesticated rock pigeon (Columba livia) has been bred for hundreds of years to display an immense variety of ornamental attributes such as feather color and color patterns. Color is influenced by multiple loci that impact the type and amount of melanin deposited on the feathers. Pigeons homozygous for the "recessive red" mutation, which causes downregulation of Sox10, display brilliant red feathers instead of blue/black feathers. Sox10 encodes a transcription factor important for melanocyte differentiation and function, but the genes that mediate its promotion of black versus red pigment are unknown. Here, we present a transcriptomic comparison of regenerating feathers from wild‐type and recessive red pigeons to identify candidate SOX10 targets. Our results identify both known and novel targets, including many genes not previously implicated in pigmentation. These data highlight the value of using novel, emerging model organisms to gain insight into the genetic basis of pigment variation. [ABSTRACT FROM AUTHOR]

Abstract: Gale crater is the landing site of the Mars Science Laboratory (MSL) rover, Curiosity. Here we present Thermal Emission Imaging System Visible Imaging Subsystem (THEMIS‐VIS) mosaics in gray scale and in false color of Gale crater. We use these data products, in addition to THEMIS thermal infrared decorrelation stretch (DCS) mosaics, thermal inertia derivations, and near‐infrared spectra to investigate the MSL traverse area and sand across Gale crater. We identified several THEMIS‐VIS color units in the MSL traverse area that may correlate to the amount of sand cover that is present on each unit. This suggests that THEMIS‐VIS color is extremely useful in identifying dark basaltic sand, which appears as blue in THEMIS‐VIS false color images. We test this hypothesis by identifying small (~several pixels) blue patches on the southeast side of the central mound in Gale crater and confirming that they are patches of sand in High Resolution Imaging Science Experiment (HiRISE) images. Sand dunes on the crater floor exhibit variations in all data sets utilized in this study, with the Bagnold dunes and the western sand sea exhibiting the most notable differences. We propose that these differences are the result of a thin layer of dust that covers the western sand sea, although we do not rule out possible effects from grain size sorting. Understanding the molecular pathways that enhance β-cell proliferation, survival, and insulin secretion may be useful to improve treatments for diabetes. Nkx6.1 induces proliferation through the Nr4a nuclear receptors, and improves insulin secretion and survival through the peptide hormone VGF. Here we demonstrate that Nkx6.1-mediated upregulation of Nr4a1, Nr4a3, and VGF is dependent on c-Fos expression. c-Fos overexpression results in activation of Nkx6.1 responsive genes and increases β-cell proliferation, insulin secretion, and cellular survival. c-Fos knockdown impedes Nkx6.1-mediated β-cell proliferation and insulin secretion. Background: Genome editing techniques, including ZFN, TALEN, and CRISPR, have created a need to rapidly screen many F1 individuals to identify carriers of indels and determine the sequences of the mutations. Current techniques require multiple clones of the targeted region to be sequenced for each individual, which is inefficient when many individuals must be analyzed. Direct Sanger sequencing of a polymerase chain reaction (PCR) amplified region surrounding the target site is efficient, but Sanger sequencing genomes heterozygous for an indel results in a string of 'double peaks' due to the mismatched region. Results: To facilitate indel identification, we developed an online tool called Poly Peak Parser (available at ) that is able to separate chromatogram data containing ambiguous base calls into wild-type and mutant allele sequences. This tool allows the nature of the indel to be determined from a single sequencing run per individual performed directly on a PCR product spanning the targeted site, without cloning. Conclusions: The method and algorithm described here facilitate rapid identification and sequence characterization of heterozygous mutant carriers generated by genome editing.
