Today, in Central Otago, New Zealand, with competing claims for water beyond irrigation, farmers and water conservation interests are at loggerheads, and freshwater management has become a site of intense debate, competition and political action. We comment on the polarised politics of water governance in Central Otago, writing from the perspective of fish and gamebird management. Our commentary is allied to an interpretation of freshwater allocation for irrigation in New Zealand, which emphasises the emergence and dominance of a singular 'more water' discourse in rural development in the 2000s. Background: Matted nodes in human papillomavirus (HPV)‐mediated oropharyngeal squamous cell carcinoma (OPC) is an independent predictor of distant metastases and decreased overall survival. We aimed to classify imaging patterns of metastatic lymphadenopathy, analyze our classification system for reproducibility, and assess its prognostic value. Methods: The metastatic lymphadenopathy was classified based on radiological characteristics for 216 patients with HPV‐mediated OPC. Patient outcomes were compared and inter‐rater reliability was calculated. Results: The presence of ≥3 abutting lymph nodes with imaging features of surrounding extranodal extension (ENE), one subtype of matted nodes, was associated with worse 5‐year overall survival, overall recurrence‐free survival, regional recurrence‐free survival, and distant recurrence‐free survival (p ≤ 0.03). Other patterns were not significantly associated with outcome measures. Overall inter‐rater agreement was substantial (κ = 0.73). Conclusion: One subtype of matted nodes defined by ≥3 abutting lymph nodes with imaging features of surrounding ENE is the radiological marker of worst prognosis. [ABSTRACT FROM AUTHOR]

We discuss case studies of three South Island small towns: Ashburton, Timaru and Oamaru and their wider local authority jurisdictions, focusing on how local stakeholders are defining the issues facing these places and identifying, prioritising and investing in regeneration initiatives, sometimes with the support of central government and other external agencies. Our key finding is that small‐town regeneration is complex, demands a long‐term local collaborative approach, and significant investment in skills, information sharing and programme development, some of which needs to be provided by external, central government, agencies. Elevated arsenic in Bengal Basin aquifers threatens human health. Most deep (>150 m) groundwater in Pleistocene aquifers is low in arsenic; however higher concentrations have been reported in the southwest border region. Here, we establish that this extensive arsenic contamination at depth is not associated with well failure. A combination of geochemistry and flow modeling constrains the factors that contribute to arsenic contamination at depth in this region. Deep groundwater in the affected area is younger (2.0 ± 0.6 kyr) than deep, low‐arsenic groundwater elsewhere (12.0 ± 4.0 kyr) based on radiocarbon. Stratigraphic data indicate pre‐Holocene deposition of the contaminated aquifers, but few low‐permeability strata. Numerical modeling indicates that this stratigraphic anomaly permits a natural flow system that transports shallow groundwater to depth. Thus, in areas lacking low‐permeability layers, arsenic contamination can occur in pre‐Holocene aquifers and is probably not an early sign of future deep contamination in regions with interbedded low‐permeability strata. Plain Language Summary: Exposure to arsenic in untreated groundwater pumped from millions of shallow wells across rural South Asia causes life‐threatening cardiovascular disease and cancers in adults and reduces intellectual function in children. Deep (>150 m) groundwater low in arsenic is currently the most effective mitigation option in Bangladesh. This study shows that high concentrations of arsenic observed in deep aquifers in the border area between Bangladesh and India are likely confined to that region due to the absence of clay layers vertically dividing the sandy aquifer. Flow modeling constrained by groundwater dating shows that this contamination is therefore of natural origin and unlikely to expand to wider areas in the near future. On a sunny afternoon in December 2019, family, friends, colleagues and students met at Lincoln University to farewell Pyarali Memon, and celebrate his life and achievements. The connection with Africa remained strong however, and Ali and Farida took their three children, Sameer, Sohrabh and Neelu, back to Kenya to visit family, friends and familiar places several times. This article draws on the application of interpretive walks in a socio-geographical study of tourism-oriented entrepreneurial activity on multi-generational family farms in New Zealand. We highlight the great potential this method holds for tourism researchers interested in the ways tourist spaces are produced in processes of place-making. Mobile methods have been a feature of qualitative field research in several disciplines for some time, particularly in cultural geography with its emphasis on human interactions in and with landscapes. The interpretive walk, known also as the walking interview, has been applied mainly in urban neighbourhood, health, transport, and housing research, where it has proven very useful for revealing human connections to place that have been difficult to elicit using stationary face-to-face interviews. This article is one of the few that reports on the use of the method in a farm tourism setting. It is also one of few applied studies seeking to understand the local geographies of farm tourism and their connections to the farm site as both family home and place of primary production. The method is characterised as an effective tool for navigating and interpreting the socio-spatial settings inwhich newrural tourism ventures emerge, evolve, and are embedded. The approach allows for unexpected encounters with spatial practices and strategies, projects, and objects, behind which lie stories of changing human relationships with the land, economy, and community, and of the exigencies of everyday life that are less readily unearthed using conventional interviews. Abstract: Fidelity is of key importance if virtual environments are to be used as authentic representations of real environments. However, simulating the multitude of senses that comprise the human sensory system is computationally challenging. With limited computational resources, it is essential to distribute these carefully in order to simulate the most ideal perceptual experience. This paper investigates this balance of resources across multiple scenarios where combined audiovisual stimulation is delivered to the user. A subjective experiment was undertaken where participants (N=35) allocated five fixed resource budgets across graphics and acoustic stimuli. In the experiment, increasing the quality of one of the stimuli decreased the quality of the other. Findings demonstrate that participants allocate more resources to graphics; however, as the computational budget is increased, an approximately balanced distribution of resources is preferred between graphics and acoustics. Based on the results, an audiovisual quality prediction model is proposed and successfully validated against previously untested budgets and an untested scenario. [ABSTRACT FROM AUTHOR]

The paper examines the value-generating strategies used in some of New Zealand's primary activities in a rapidly changing trading context in which the countries of East Asia increasingly figure. These strategies range from scaling up volume, with a consequent conspicuous impact on rural landscapes, to innovative value-enhancing relationships linking producers, processors and consumers in processes which value landscapes for the contribution they make to provenance stories. Our argument is that higher-value strategies are necessarily more intensely collaborative and involve a pursuit of engaged relationships, especially between onshore producers and manufacturers and consumers at a distance. We term this collaborative form of strategising as 'the relationships economy'. We illustrate our argument using two case studies, dairying and merino. While there are great differences in scale between them, and dairying focuses significantly on volume-generating capacity, there are players in dairying who are adopting higher-value relationships-based approaches similar to those developed in the merino industry. The common thread connecting the case studies is that of intentional enactment, which echoes earlier periods in New Zealand's trading history when considerable work was undertaken by industry participants to create new markets for primary products. Three novel HLA-Class II alleles, DRB1*03:112, DQB1*03:02:16 and DQB1*03:139, are described with predicted bearing haplotypes of A*02:01, B*40:01, C*03:04, DRB1*03:112, DQB1*02:01; A*23:01, B*15:01, C*03:03, DRB1*04:01, DQB1*03:02:16 and A*01:01, B*44:02, C*05:01/03, DRB1*04:01, DQB1*03:139. Serological tests showed that the DRB1*03:112 and DQB1*03:139 specificities failed to react as expected with some well-documented monoclonal antibodies. Subsequent examination of published HLA-Class II epitopes and inspection of amino acid motifs suggested that epitopes exist that include the positions of their single substitutions (F31C between DRB1*03:01:01:01 and DRB1*03:112, and R48P between DQB1*03:01:01:01 and DQB1*03:139 specificities). This suggests that the reactivity of the monoclonal antibodies used was dependent on these epitopes and that their loss from these rare allele products resulted in their aberrant serology. The new alleles were found after the sequence-based typing of 32 530 random UK European routine blood donors suggesting that each has a maximum carriage frequency of 0.0031% in the blood donor population resident in Wales. Declining abundance of Chinook Salmon Oncorhynchus tshawytscha across the Pacific Northwest is an issue of great concern ecologically, culturally, and economically. Growth during the first summer is vitally important for juvenile Chinook Salmon, as it influences not only life history decisions (to smolt or not to smolt) but also subsequent river and ocean survival. Using Ecopath with Ecosim, we developed a food web model for a representative stream in the Salmon River basin, Idaho, to evaluate potential species‐specific and food web effects of three management strategies: (1) adding salmon carcasses or carcass analogs to promote primary production and detrital availability that were lost due to declining salmon returns; (2) removal of nonnative Brook Trout Salvelinus fontinalis, which are competitors with and predators on juvenile Chinook Salmon; and (3) stocking hatchery Chinook Salmon into streams to supplement wild production. Overall, juvenile Chinook Salmon responded strongly to increases in basal resources. Removal of Brook Trout had little effect on potential production for juvenile Chinook Salmon, but the responses of sculpins Cottus spp. were strong, primarily due to the sculpins’ high degree of dietary overlap with and predation by Brook Trout. Supplementation with hatchery‐origin juveniles depressed the production of wild juvenile Chinook Salmon, especially at the densities commonly applied to streams in this region. Our results suggest that efforts to enhance basal resources are likely to be the most effective in promoting the production of juvenile Chinook Salmon and nearly all food web groups considered in our model system. Removal of nonnative Brook Trout is unlikely to substantially affect salmon but could have a disproportionately large effect on nongame species, which are generally overlooked in single‐species management approaches. The objective of this research was to validate the use of Fleishman's human abilities taxonomy approach in predicting human performance in loading the U.S. Navy's Close-In Weapon System (CIWS). An experiment was conducted that compared actual and predicted performance of CIWS loading operations. Twenty-one male students and staff from the Close-In Weapon System School, Fleet Training Center, were participants in the experiment. They ranged in age from 20 to 35 years (mean = 27.2, standard deviation = 4.88) with an average of 81.1 months (standard deviation = 51.83) of military experience and 23.95 months of CIWS loading experience. Each participant was tested in the human abilities identified for CIWS loading operations, and the test results were correlated with recorded times of the loading tasks. Correlations were performed using bivariate regression analysis. The results of the experiment showed high linear relations between human abilities scores and CIWS loading times. This article reports research investigating the ways individuals and families in New Zealand have adapted to and created economic and social change through holding multiple jobs since the beginning of a period of restructuring in the 1980s. Our research has been stimulated by the very significant increase in multiple job holding in New Zealand over the whole workforce, and more particularly in rural areas over the last 25 years. Triage processes are commonly used to manage the interface between demand for, and supply of, health services. This dimension of service provision is particularly pertinent for mental health services in Australia, where demand outweighs services available. This paper draws on the experiences of using participant observation to explore mental health triage processes. Findings highlight the complexities of the researcher role and benefits of using an ethnographic approach to explore mental health triage patterns. Insider participant observation brings many challenges but also, in this study, enabled the researcher to uncover some roles and processes underpinning triage decisions in mental health services. Overall, about one-third of women operated upon because of suspected ovarian cancer turn out to have benign disease. This proportion will be even higher when the tumours are small. A more accurate method of determining which are malignant would reduce the number of unnecessary referrals to the cancer centre and increase the scope for conservative management. This pilot study of ultrasound and intravenous microbubble contrast in 20 women with small ovarian tumours enabled the identification of all four malignant lesions with good inter-observer agreement. These preliminary findings suggest that intravenous contrast improves the ultrasound identification of small ovarian malignancies and may provide an accurate way of selecting women who require to be referred urgently for surgery under the auspices of a gynaecological oncologist. Summary Expression of lactase in the intestine persists into adult life in some people and not others, and this is due to a cis -acting regulatory polymorphism. Previous data indicated that a mutation leading to lactase persistence had occurred on the background of a 60 kb 11-site LCT haplotype known as A (Hollox et al. 2001). Recent studies reported a 100% correlation of lactase persistence with the presence of the T allele at a CT SNP at -14 kb from LCT , in individuals of Finnish origin, suggesting that this SNP may be causal of the lactase persistence polymorphism, and also reported a very tight association with a second SNP (GA –22 kb) (Enattah et al. 2002). Here we report the existence of a one megabase stretch of linkage disequilibrium in the region of LCT and show that the –14 kb T allele and the –22 kb A allele both occur on the background of a very extended A haplotype. In a series of Finnish individuals we found a strong correlation (40/41 people) with lactose digestion and the presence of the T allele. The T allele was present in all 36 lactase persistent individuals from the UK (phenotyped by enzyme assay) studied, 31/36 of whom were of Northern European ancestry, but not in 11 non-persistent individuals who were mainly of non-UK ancestry. However, the CT heterozygotes did not show intermediate lactase enzyme activity, unlike those previously phenotyped by determining allelic transcript expression. Furthermore the one lactase persistent homozygote identified by having equally high expression of A and B haplotype transcripts, was heterozygous for CT at the -14 kb site. SNP analysis across the 1 megabase region in this person showed no evidence of recombination on either chromosome between the –14 kb SNP and LCT . The combined data shows that although the –14 kb CT SNP is an excellent candidate for the cause of the lactase persistence polymorphism, linkage disequilibrium extends far beyond the region searched so far.... The appearance of IgG and IgE binding components in the medium of shake cultures of Aspergillus fumigatus has been studied. Cultures were grown in synthetic asparagine medium at 35°C and flasks harvested in duplicate 1, 2, 3, 4, 7 and 14 days after inoculation. The pH of the medium dropped from its initial value of 5.5 to pH 3, and then after 4 days gradually increased up to pH 7.5 in the 14-day medium. The weight of mycelium, after an initial peak followed by a slight decline, increased as the pH of the medium increased. Components able to bind IgC and IgE from pooled ABPA sera were detected by crossed immunoelectrophoresis/self-crossed radioimmunoelectrophoresis within 24 h of growth, but maximal release of both antigens and allergens coincided with the increase in pH of the medium and was seen in the 14-day culture filtrate. Two recognised "major" antigens, Ag 7 and Ag 13, detected using the relevant monospecific antisera, were present in the culture medium after 14 days of growth and similarly for Ag 3, the major allergen, although another allergen, Ag 1, was identified in the 1-day extract. Rapid, sequential serum creatine kinase measurements may be used to exclude acute myocardial infarction in patients presenting with acute chest pain. Two cases of cardiopathia fantastica (the cardiac variant of Munchausen's syndrome) are described. Both patients presented with symptoms of acute myocardial infarction and had electrocardiographic changes consistent with this diagnosis. Both patients also had a raised serum creatine kinase level at the time of presentation but were unable to mimic the rise in serum creatine kinase associated with acute myocardial infarction. Munchausen's syndrome was suspected when both patients responded poorly to news that their enzyme results excluded acute myocardial infarction. [ABSTRACT FROM AUTHOR]

We describe a simple method of performing sequential excision biopsies during liver reperfusion in the isolated perfused rat liver. After hepatectomy, four ligatures (5.0 silk) tied with a slip knot are placed around the pedicles of: (1) the inferior and (2) the superior parts of the caudate lobe, as well as (3) the inferior and (4) the superior parts of the right lateral lobe. At the time of biopsy, the prepared 5.0 silk ties are tightened with sufficient force to occlude the vascular pedicle, preventing leakage of circulating perfusate. The procedure provides four biopsies of more than 350 mg each without alteration of perfusate transaminases and tissue ATP contents. It has previously been shown that human neutrophils generate substantial quantities of LTB4 when stimulated with the calcium ionophore, A23187, or with unopsonised zymosan. We now report that normal human neutrophils produced substantial quantities of LTB4 (measured by radioimmunoassay and validated by RP-HPLC) when incubated with large non-phagocytosable IgG-coated beads (Sepharose 4B). LTB4 was identified in both the extra and intracellular compartments. The production of LTD. was dependent upon the number of IgG-coated particles and the concentration of IgG bound to the beads. Release was maximal after a 15-30 min incubation time and was enhanced by prior activation of the neutrophils with the synthetic bacterial product f-met-leu-phe. Comparable LTB4 production was also observed when neutrophils were incubated with antigen (Aspergillus fumigatus)-coated beads sensitized with purified IgG obtained from the sera of patients with allergic bronchopulmonary aspergillosis. These results suggest a further mechanism by which neutrophils may be activated to produce inflammatory mediators in the tissues. [ABSTRACT FROM AUTHOR]
