Visceral hypersensitivity, a hallmark of disorders of the gut‐brain axis, is associated with exposure to early‐life stress (ELS). Activation of neuronal β3‐adrenoceptors (AR) has been shown to alter central and peripheral levels of tryptophan and reduce visceral hypersensitivity. In this study, we aimed to determine the potential of a β3‐AR agonist in reducing ELS‐induced visceral hypersensitivity and possible underlying mechanisms. Here, ELS was induced using the maternal separation (MS) model, where Sprague Dawley rat pups were separated from their mother in early life (postnatal day 2–12). Visceral hypersensitivity was confirmed in adult offspring using colorectal distension (CRD). CL‐316243, a β3‐AR agonist, was administered to determine anti‐nociceptive effects against CRD. Distension‐induced enteric neuronal activation as well as colonic secretomotor function were assessed. Tryptophan metabolism was determined both centrally and peripherally. For the first time, we showed that CL‐316243 significantly ameliorated MS‐induced visceral hypersensitivity. Furthermore, MS altered plasma tryptophan metabolism and colonic adrenergic tone, while CL‐316243 reduced both central and peripheral levels of tryptophan and affected secretomotor activity in the presence of tetrodotoxin. This study supports the beneficial role of CL‐316243 in reducing ELS‐induced visceral hypersensitivity, and suggests that targeting the β3‐AR can significantly influence gut‐brain axis activity through modulation of enteric neuronal activation, tryptophan metabolism, and colonic secretomotor activity which may synergistically contribute to offsetting the effects of ELS. Distributions of the natural radionuclide 210Po and its grandparent 210Pb along the GP15 Pacific Meridional Transect provide information on scavenging rates of reactive chemical species throughout the water column and fluxes of particulate organic carbon (POC) from the primary production zone (PPZ). 210Pb is in excess of its grandparent 226Ra in the upper 400–700 m due to the atmospheric flux of 210Pb. Mid‐water 210Pb/226Ra activity ratios are close to radioactive equilibrium (1.0) north of ∼20°N, indicating slow scavenging, but deficiencies at stations near and south of the equator suggest more rapid scavenging associated with a "particle veil" located at the equator and hydrothermal processes at the East Pacific Rise. Scavenging of 210Pb and 210Po is evident in the bottom 500–1,000 m at most stations due to enhanced removal in the nepheloid layer. Deficits in the PPZ of 210Po (relative to 210Pb) and 210Pb (relative to 226Ra decay and the 210Pb atmospheric flux), together with POC concentrations and particulate 210Po and 210Pb activities, are used to calculate export fluxes of POC from the PPZ. 210Po‐derived POC fluxes on large (>51 μm) particles range from 15.5 ± 1.3 mmol C/m2/d to 1.5 ± 0.2 mmol C/m2/d and are highest in the Subarctic North Pacific; 210Pb‐derived fluxes range from 6.7 ± 1.8 mmol C/m2/d to 0.2 ± 0.1 mmol C/m2/d. Both 210Po‐ and 210Pb‐derived POC fluxes are greater than those calculated using the 234Th proxy, possibly due to different integration times of the radionuclides, considering their different radioactive mean‐lives and scavenging mean residence times. Empathetic relationships and the social transference of behaviours have been shown to occur in humans, and more recently through the development of rodent models, where both fear and pain phenotypes develop in observer animals. Clinically, observing traumatic events can induce 'trauma and stressor‐related disorders' as defined in the DSM 5. These disorders are often comorbid with pain and gastrointestinal disturbances; however, our understanding of how gastrointestinal – or visceral – pain can be vicariously transmitted is lacking. Visceral pain originates from the internal organs, and despite its widespread prevalence, remains poorly understood. We established an observation paradigm to assess the impact of witnessing visceral pain. We utilised colorectal distension (CRD) to induce visceral pain behaviours in a stimulus rodent while the observer rodent observed. Twenty four hours post‐observation, the observer rodent's visceral sensitivity was assessed using CRD. The observer rodents were found to have significant hyperalgesia as determined by lower visceral pain threshold and higher number of total pain behaviours compared with controls. The behaviours of the observer animals during the observation were found to be correlated with the behaviours of the stimulus animal employed. We found that observer animals had hypoactivity of the hypothalamic–pituitary–adrenal (HPA) axis, highlighted by reduced corticosterone at 90 minutes post‐CRD. Using c‐Fos immunohistochemistry we showed that observer animals also had increased activation of the anterior cingulate cortex, and decreased activation of the paraventricular nucleus, compared with controls. These results suggest that witnessing another animal in pain produces a behavioural phenotype and impacts the brain–gut axis. [ABSTRACT FROM AUTHOR]

Conservation psychology principles can be useful for aligning organizations and scaling up conservation programs to increase impact while strategically engaging partners and communities. We can use findings and recommendations from conservation psychology to inform organizational collaborations between zoos and aquariums to maximize efficiency and coordination. In this study, we developed and evaluated a collaborative conservation initiative for monarch butterflies built with conservation psychology principles. We present our process for collaborative program planning and the resultant collective conservation plan as well as our formative evaluation findings after 1‐year of collaboration. We share best practices for group facilitation and conservation planning along with our evaluation instruments to support future collaborative conservation initiatives. Deposition of aerosols to the surface ocean is an important factor affecting primary production in the surface ocean. However, the sources and fluxes of aerosols and associated trace elements remain poorly defined. Aerosol 210Pb, 210Po, and 7Be data were collected on US GEOTRACES cruise GP15 (Pacific Meridional Transect, 152°W; 2018). 210Pb fluxes are low close to the Alaskan margin, increase to a maximum at ∼43°N, then decrease to lower values. There is good agreement between 210Pb fluxes and long‐term land‐based fluxes during the SEAREX program (1970–1980s), as well as between GP15 and GP16 (East Pacific Zonal Transect, 12°S; 2013) at adjacent stations. A normalized fraction f(7Be, 210Pb) is used to discern aerosols with upper (high f) versus lower (low f) troposphere sources. Alaskan/North Pacific aerosols show significant continental influence while equatorial/South Pacific aerosols are supplied to the marine boundary layer from the upper troposphere. Lithogenic trace elements Al and Ti show inverse correlations with f(7Be, 210Pb), supporting a continental boundary layer provenance while anthropogenic Pb shows no clear relationship with f(7Be, 210Pb). All but four samples have 210Po/210Pb activity ratios <0.2 suggesting short aerosol residence time. Among the four samples (210Po/210Pb = 0.42–0.88), two suggest an upper troposphere source and longer aerosol residence time while the remaining two cannot be explained by long aerosol residence time nor a significant component of dust. We hypothesize that enrichments of 210Po in them are linked to Po enrichments in the sea surface microlayer, possibly through Po speciation as a dissolved organic or dimethyl polonide species. Sinking particles strongly regulate the distribution of reactive chemical substances in the ocean, including particulate organic carbon and other elements (e.g., P, Cd, Mn, Cu, Co, Fe, Al, and 232Th). Yet, the sinking fluxes of trace elements have not been well described in the global ocean. The U.S. GEOTRACES campaign in the North Atlantic (GA03) offers the first data set in which the sinking flux of carbon and trace elements can be derived using four different radionuclide pairs (238U:234Th ;210Pb:210Po; 228Ra:228Th; and 234U:230Th) at stations co‐located with sediment trap fluxes for comparison. Particulate organic carbon, particulate P, and particulate Cd fluxes all decrease sharply with depth below the euphotic zone. Particulate Mn, Cu, and Co flux profiles display mixed behavior, some cases reflecting biotic remineralization, and other cases showing increased flux with depth. The latter may be related to either lateral input of lithogenic material or increased scavenging onto particles. Lastly, particulate Fe fluxes resemble fluxes of Al and 232Th, which all have increasing flux with depth, indicating a dominance of lithogenic flux at depth by resuspended sediment transported laterally to the study site. In comparing flux estimates derived using different isotope pairs, differences result from different timescales of integration and particle size fractionation effects. The range in flux estimates produced by different methods provides a robust constraint on the true removal fluxes, taking into consideration the independent uncertainties associated with each method. These estimates will be valuable targets for biogeochemical modeling and may also offer insight into particle sinking processes. Access to carbon, iron, and other elements is important for marine organisms, which need them to survive. Furthermore, when the organic carbon produced by organisms is transported to depth by sinking, carbon dioxide has been effectively removed from the atmosphere and moved to the deep ocean. This carbon sink is one way that the ocean reduces the heat‐trapping potential of the atmosphere. To track how much of a given element descends on particles through the ocean, we use radioisotopes. These are elements that decay at a predictable rate. We can use them like a clock to determine how fast an element is moving from one location to another. Radioisotopes with varying decay rates can tell us about short‐term processes, like seasonal blooms, and longer term events, like the impact of ice ages. There were few ocean‐scale radioisotope data sets before GEOTRACES expeditions began about 10 years ago. For the first time ever, we present four types of radioisotope data from the U.S. GEOTRACES expedition across the North Atlantic and discuss how it improves our understanding of elemental budgets in the global ocean. Background: There are three main methods of obtaining population data on the incidence and/or prevalence of dementia: cross‐sectional surveys (which may be repeated over time); cohort studies that follow people initially without dementia and count newly diagnosed cases over time; and administrative health records (including linkage of records from multiple sources). The major challenges for all these methods are: how well the study sample represents the target population, the accuracy of diagnoses, and the costs of maintaining the data collection over time. Method: In a project to improve Australia's dementia statistics, we conducted a series of studies to compare population estimates of dementia obtained using different methods. Firstly, we used existing general health studies of community‐based cohorts, supplemented by linkage to administrative records of hospital and emergency department admissions, assessments for aged care support, medication prescriptions, and death certificates to estimate the cumulative incidence of dementia. Secondly, we created cohorts based on administrative records for entire populations. Thirdly, we assessed the validity of the identification of people with dementia in the record linkage cohorts in various ways, including linkage with studies that had obtained clinical diagnosis through the standardised assessment of participants. Result: We will present empirical results illustrating the strengths and limitations of these different approaches. In summary, community‐based cohort studies lack representativeness of national or regional populations due to recruitment biases and differential loss to follow‐up. Cohort studies are also costly to maintain over the long time needed for participants to develop dementia. In contrast, the use of administrative records is relatively inexpensive, but is subject to policy changes that impact on the continuity of data coverage and quality. Population coverage may also be problematic for administrative data if important sources of care for people with dementia are not included; for example, in Australia linkable primary care data are not available. The validation studies showed that accuracy was highly dependent on data sources, and identification of dementia type was unreliable. Conclusion: Prevalence and trends data of dementia obtained from multiple sources are needed to provide accurate population estimates, together with detailed contextual knowledge and careful analysis. The article discusses the results of a study which determined whether the point-of-care (POC) cardiac biomarker panel assessment will reduce the costs of health care. It describes the Randomized Assessment of Treatment using Panel Assay of Cardiac markers (RATPAC) as study method. Also discussed are the components of POC cardiac biomarker, including myoglobin and troponin. The study noted the linked between POC and higher emergency department (CD) costs.

Background and Aim: Locoregional therapies for hepatocellular carcinoma (HCC) are considered to confer a survival advantage, however, the patient group that should be targeted is not clearly defined. This study aimed to determine the impact on survival of locoregional therapies compared with supportive care, within prognostic categories as stratified by the Cancer of the Liver Italian Program (CLIP) scoring system. Methods: A prospective database was used to identify those patients who were treated with either locoregional therapy ( n = 128) or supportive care ( n = 92). Survival analysis was performed for groups matched by CLIP score at presentation. Comparison of important prognostic factors was undertaken and univariate and multivariate analysis was performed to assess determinants of survival. Results: Use of locoregional therapies was only associated with a survival benefit in patients with a CLIP score of 1 or 2. In this group, the median survival in patients who received locoregional therapies was 25.0 months (95% confidence interval 22.7–27.4) compared with 8.9 months (95% confidence interval 7.3–10.5) for supportive care ( P = 0.001). For patients with CLIP scores of 3 or greater, no survival benefit of locoregional therapies was observed. Multivariate analysis revealed locoregional intervention, CLIP score, tumor symptoms, α-fetoprotein level, bilirubin and alkaline phosphatase level as independent prognostic indicators. Conclusion: Locoregional therapies should be targeted specifically to patients with non-advanced hepatocellular carcinoma as assessed by validated scoring systems. Use of these therapies in patients with advanced disease does not appear to be associated with a survival benefit and may expose patients to unnecessary harm. [ABSTRACT FROM AUTHOR]

The article presents a study on blue nevus-like melanomas (BNLMs). The study reveals that BNLMs tended to demonstrate at a more advanced stage, with thicker main tumors, but had a metastatic pattern like other types of melanoma. It suggests that BNLMs should be dealt in the same way as other variants of melanoma based on pathologic prognostic indices and clinical staging.

The article discusses the recurrence rates of desmoplastic neurotropic melanoma (DNM) on patients after surgery and radiotherapy (RT) in Sydney, New South Wales. It notes that the head and neck are common recurrence location of DNM. Moreover, it highlights that surgical treatment and RT decrease the recurrence of DNM.

The split mouth study design in trials of Fissure sealants and restorative materials neatly controls for confounding by many of the variables associated with poor retention of sealants and occurrence of dental caries. Traditionally, the out- come measures used in split mouth trials have been material retention, (per cent) effectiveness and net gain. A survey of the literature revealed that a large proportion of split mouth studies report no statistical evaluation of outcome measures. In those studies in which statistical evaluation had been conducted, McNemar's χ[SUP2] was the test most frequently used. This statistic is appropriate for comparing differences between "success" and "failure" tooth pairs (or "positives" and "negatives" in split mouth terminology) but it cannot evaluate directly effectiveness and net gain. The distributions of effectiveness and net gain are different and it would be desirable to estimate confidence intervals for them. In this paper, we consider these statistics, suggest methods by which confidence intervals may be calculated, and provide examples of the calculations. We demonstrate the close relationship between effectiveness (as used in split mouth trials) and relative effect and relative risk (as used in general epidemiological analysis) and recommend that relative risk should be the preferred outcome measure for split mouth trials. Whatever outcome measure is chosen in split mouth trials should always he subjected to statistical evaluation. preferably by the calculation of confidence intervals. [ABSTRACT FROM AUTHOR]

This article emphasize on the need of having a valid format of statistical analysis of split mouth caries trials. Most of the case control studies are summarized in 2 by 2 tables, but the layout of 2 by 2 tables depends on weather the statistical analysis is based on paired data or otherwise. Authors of the article have clarified some of the technical problems in their original article. The thrust of the article, which recommends that relative risk be used as an outcome measure in split mouth caries trials, remain unchanged.

Summary This paper examines a number of methods of handling missing outcomes in regressive logistic regression modelling of familial binary data, and compares them with an EM algorithm approach via a simulation study. The results indicate that a strategy based on imputation of missing values leads to biased estimates, and that a strategy of excluding incomplete families has a substantial effect on the variability of the parameter estimates. In recent years philosophers and bioethicists have given considerable attention to the concept of care. Thus we have seen important work on questions such as: whether there is a uniquely female approach to ethics, whether ethics should be partial or impartial, and whether care must be supplemented by justice. Despite this valuable and extensive work, however, some important distinctions have gone largely undiscussed. This paper tries to fill a gap left in our understanding of the concept of care itself by distinguishing between compassion and two kinds of pity. While all three are kinds of caring, we should not give them similar moral evaluations. Consequently, the distinction between compassion and different kinds of pity gives us an important insight into the question of whether we can consider care a virtue for health care professionals.
