Neuroimaging studies in rapid eye movement sleep behavior disorder (RBD) can inform fundamental questions about the pathogenesis of Parkinson's disease (PD). Across modalities, functional magnetic resonance imaging (fMRI) may be better suited to identify changes between neural networks in the earliest stages of Lewy body diseases when structural changes may be subtle or absent. This review synthesizes the findings from all fMRI studies of RBD to gain further insight into the pathophysiology and progression of Lewy body diseases. A total of 32 studies were identified using a systematic review conducted according to PRISMA guidelines between January 2000 to February 2024 for original fMRI studies in patients with either isolated RBD (iRBD) or RBD secondary to PD. Common functional alterations were detectable in iRBD patients compared with healthy controls across brainstem nuclei, basal ganglia, frontal and occipital lobes, and whole brain network measures. Patients with established PD and RBD demonstrated decreased functional connectivity across the whole brain and brainstem nuclei, but increased functional connectivity in the cerebellum and frontal lobe compared with those PD patients without RBD. Finally, longitudinal changes in resting state functional connectivity were found to track with disease progression. Currently, fMRI studies in RBD have demonstrated early signatures of neurodegeneration across both motor and non‐motor pathways. Although more work is needed, such findings have the potential to inform our understanding of disease, help to distinguish between prodromal PD and prodromal dementia with Lewy bodies, and support the development of fMRI‐based outcome measures of phenoconversion and progression in future disease modifying trials. © 2024 The Author(s). In a smart city, IoT devices are required to support monitoring of normal operations such as traffic, infrastructure, and the crowd of people. IoT‐enabled systems offered by many IoT devices are expected to achieve sustainable developments from the information collected by the smart city. Indeed, artificial intelligence (AI) and machine learning (ML) are well‐known methods for achieving this goal as long as the system framework and problem statement are well prepared. However, to better use AI/ML, the training data should be as global as possible, which can prevent the model from working only on local data. Such data can be obtained from different sources, but this induces the privacy issue where at least one party collects all data in the plain. The main focus of this article is on support vector machines (SVM). We aim to present a solution to the privacy issue and provide confidentiality to protect the data. We build a privacy‐preserving scheme for SVM (SecretSVM) based on the framework of federated learning and distributed consensus. In this scheme, data providers self‐organize and obtain training parameters of SVM without revealing their own models. Voxel‐based morphometry (VBM) and surface‐based morphometry (SBM) are two widely used neuroimaging techniques for investigating brain anatomy. These techniques rely on statistical inferences at individual points (voxels or vertices), clusters of points, or a priori regions‐of‐interest. They are powerful tools for describing brain anatomy, but offer little insights into the generative processes that shape a particular set of findings. Moreover, they are restricted to a single spatial resolution scale, precluding the opportunity to distinguish anatomical variations that are expressed across multiple scales. Drawing on concepts from classical physics, here we develop an approach, called mode‐based morphometry (MBM), that can describe any empirical map of anatomical variations in terms of the fundamental, resonant modes—eigenmodes—of brain anatomy, each tied to a specific spatial scale. Hence, MBM naturally yields a multiscale characterization of the empirical map, affording new opportunities for investigating the spatial frequency content of neuroanatomical variability. Using simulated and empirical data, we show that the validity and reliability of MBM are either comparable or superior to classical vertex‐based SBM for capturing differences in cortical thickness maps between two experimental groups. Our approach thus offers a robust, accurate, and informative method for characterizing empirical maps of neuroanatomical variability that can be directly linked to a generative physical process. The African Sacred Ibis Threskiornis aethiopicus is an invasive bird species in Taiwan and has expanded its distribution range rapidly over the past three decades. There is limited information available on the invasion process of African Sacred Ibises in Taiwan and its genetic consequences. We investigated whether genetic factors reflected the expansion of African Sacred Ibises and determined the extent to which two behaviour‐associated genes may have facilitated invasion. The dopamine receptor gene (DRD4) and the serotonin transporter gene (SERT) have both been found to be associated with novel‐seeking and bold behaviour in birds. We hypothesized that: (1) selection on temperament traits may determine the polymorphisms in these two genes and (2) the likelihood of dispersal of African Sacred Ibis populations can be explained by the intraspecific variation at these two genes. To detect the signals of population expansion and natural selection, we compared intra‐population variation and inter‐population differentiation of DRD4 and SERT with those of a mitochondrial gene (COX1) and an additional intron. We recovered contrasting patterns of nucleotide variation between the mitochondrial and nuclear genes with no single nucleotide polymorphisms detected in COX1 compared with allelic polymorphism in DRD4 and SERT. Populations showed decreasing genetic diversity at DRD4 as their distance to the initial invasive locality increased, consistent with a rapid expansion from one founder population. However, we found little evidence of selection on DRD4 or SERT, suggesting that these behaviour‐related genes are unlikely to have played a significant role in the successful invasion of the African Sacred Ibises in Taiwan. Lung cancer is the leading cause of cancer death worldwide, of which lung adenocarcinoma (LUAD) is the most common subtype. Metastasis is the major cause of poor prognosis and mortality for lung cancer patients, which urgently needs great efforts to be further explored. Herein, glutathione peroxidase 8 (GPX8) was identified as a novel potential pro‐metastatic gene in LUAD metastatic mice models from GEO database. GPX8 was highly expressed in tumor tissues, predicting poor prognosis in LUAD patients. Knockdown of GPX8 inhibited LUAD metastasis in vitro and in vivo, while it did not obviously affect tumor growth. Knockdown of GPX8 decreased the levels of p‐FAK and p‐Paxillin and disturbed the distribution of focal adhesion. Furthermore, GPX8 was overexpressed in cancer‐associated fibroblast (CAF) and associated with CAF infiltration in tumor microenvironment of lung cancer. GPX8 silence on fibroblasts suppressed lung cancer cell migration in the coculture system. BRD2 and RRD4 were the potential transcriptionally regulators for GPX8. Bromodomain extra‐terminal inhibitor JQ1 downregulated GPX8 expression and suppressed lung cancer cell migration. Our findings indicate that highly expressed GPX8 in lung cancer cells and fibroblasts functions as a pro‐metastatic factor in lung cancer. Identity‐based encryption (IBE), introduced by Shamir, eliminates the need for public‐key infrastructure. The sender can simply encrypt a message by using the recipient's identity (such as email or IP address) without needing to look up the public key. In particular, when ciphertexts of an IBE do not reveal recipient's identity, this scheme is known as an anonymous IBE scheme. Recently, Blazy et al. (ARES '19) analysed the trade‐off between public safety and unconditional privacy in anonymous IBE and introduced a new notion that incorporates traceability into anonymous IBE, called anonymous IBE with traceable identities (AIBET). However, their construction is based on the discrete logarithm assumption, which is insecure in the quantum era. In this paper, we first formalize the consistency of tracing key of the AIBET scheme to ensure that a ciphertext cannot be traced with the use of wrong tracing keys. Subsequently, we present a generic formulation concept that can be used to transform structure‐specific lattice‐based anonymous IBE schemes into an AIBET. Finally, we apply this concept to Katsumata and Yamada's compact anonymous IBE scheme (Asiacrypt '16) to obtain the first quantum‐resistant AIBET scheme that is adaptively secure under the ring learning with errors assumption without random oracle. Aim: This study aims to explore the change process among chronic kidney disease patients in shared decision‐making. Design: Qualitative research is the design applied. Method: Original transcripts from an earlier study involving a purposive sample of 31 patients with chronic kidney disease provided a source of information. The data collection was conducted from August 2017–January 2018. Data analysis applied the thematic analysis method. Results: Three themes were identified: from avoidance to being forced to accept, decision‐making process of action and facilitating factors. After making the psychological adjustments necessary for undergoing dialysis treatment, participants experience the process of decision‐making action. The improvement of knowledge and ability increases the patient's self‐determination consciousness and strengthens confidence in decision‐making. Ecological niche evolution can promote or hinder the differentiation of taxa and determine their distribution. Niche‐mediated evolution may differ among climatic regimes, and thus, species that occur across a wide latitudinal range offer a chance to test these heterogeneous evolutionary processes. In this study, we examine (a) how many lineages have evolved across the continent‐wide range of the Eurasian nuthatch (Sitta europaea), (b) whether the lineages' niches are significantly divergent or conserved and (c) how their niche evolution explains their geographic distribution. Phylogenetic reconstruction and ecological niche models (ENMs) showed that the Eurasian nuthatch contained six parapatric lineages that diverged within 2 Myr and did not share identical climatic niches. However, the niche discrepancy between these distinct lineages was relatively conserved compared with the environmental differences between their ranges and thus was unlikely to drive lineage divergence. The ENMs of southern lineages tended to cross‐predict with their neighbouring lineages whereas those of northern lineages generally matched with their abutting ranges. The coalescence‐based analyses revealed more stable populations for the southern lineages than the northern ones during the last glaciation cycle. In contrast to the overlapping ENMs, the smaller parapatric distribution suggests that the southern lineages might have experienced competitive exclusion to prevent them from becoming sympatric. On the other hand, the northern lineages have expanded their ranges and their current abutting distribution might have resulted from lineages adapting to different climatic conditions in allopatry. Abstract: Aims: To investigate the relationships among social support, health literacy, and self‐management, and the factors influencing self‐management of chronic kidney disease (CKD). Design: Cross‐sectional study. Methods: A random sample of 410 patients was recruited from nephrology clinics. Data were collected using structured questionnaires and chart reviews from January 2013 to February 2014. Hierarchical regression analysis was used to determine the predictive factors of self‐management behaviors and ∆R2 to determine each variable's explanatory power. Findings: Health literacy and social support were positively correlated with self‐management behaviors. Furthermore, social support, health literacy, and marital status were significant predictors of self‐management behaviors. Social support had a relatively greater explanatory power for self‐management behaviors than did health literacy. Particularly, healthcare provider support had the greatest influence on patients’ self‐management behaviors. Conclusions: Health literacy and social support play independent positive roles in self‐management behaviors of patients with CKD, with social support having a particularly dominant role. Further research using a systems approach to improving self‐management behaviors is necessary to clarify the role of social support. Clinical Relevance: Health literacy and social support are independently and positively related to self‐management. Social support, which is a system‐level factor, is a relatively stronger and crucial predictor than is health literacy. Despite recent breakthroughs in targeted- and immune-based therapies, rapid development of drug resistance remains a hurdle for the long-term treatment of patients with melanoma. Targeting metastatically spreading circulating tumor cells ( CTCs) may provide an additional approach to manage melanoma. This study investigates whether targeting cholesterol transport in melanoma CTCs can retard metastasis development. Nanolipolee-007, the liposomal form of leelamine, reduced melanoma metastasis in both a novel in vitro flow system mimicking the circulating system and in experimental as well as spontaneous animal metastasis models, irrespective of the BRAF mutational status of the CTCs. Leelamine led to cholesterol trapping in lysosomes, which subsequently shut down receptor-mediated endocytosis, endosome trafficking, and inhibited the major oncogenic signaling cascades important for survival such as the AKT pathway. As pAKT is important in CTC survival, inhibition by targeting cholesterol metabolism led to apoptosis, suggesting this approach might be particularly effective for those CTCs having high levels of pAKT to aid survival in the circulation system. Aim The effect of health depreciation in older people on medical care demand is not well understood. We tried to assess the medical care demand with length of hospitalization and their impact on profits as a result of health depreciation. Methods All participants who underwent comprehensive geriatric assessment were from a prospective cohort study at a tertiary hospital. A total of 1191 cases between September 2008 to October 2012 were investigated. Three sets of qualitative response models were constructed to estimate the impact of older adults' health depreciation on multidisciplinary geriatric care services. Furthermore, we analyzed the factors affecting the composite end-point of rehospitalization within 14 days, re-admission to the emergency department within 3 days and patient death. Results Greater health depreciation in elderly patients was positively correlated with greater medical care demand. Three major components were defined as health depreciation: elderly adaptation function, geriatric syndromes and multiple chronic diseases. On admission, the better the basic living functions, the shorter the length of hospitalization (coefficient = −0.35, P < 0.001 in Poisson regression; coefficient = −0.33, P < 0.001 in order choice profit model; coefficient = −0.29, P < 0.001 in binary choice profit model). The major determinants for poor outcome were male sex, middle old age and length of hospitalization. However, factors that correlated with relatively good outcome were functional improvement after medical care services and level of disease education. Conclusions An optimal allocation system for selection of cases into multidisciplinary geriatric care is required because of limited resources. ABSTRACT For the purpose of improving recombinant protein production from mammalian cells, an unbiased, high-throughput whole-genome RNA interference screen was conducted using human embryonic kidney 293 (HEK 293) cells expressing firefly luciferase. A 21,585 human genes were individually silenced with three different siRNAs for each gene. The screen identified 56 genes that led to the greatest improvement in luciferase expression. These genes were found to be included in several pathways involved in spliceosome formation and mRNA processing, transcription, metabolic processes, transport, and protein folding. The 10 genes that most enhanced protein expression when downregulated, were further confirmed by measuring the effect of their silencing on the expression of three additional recombinant proteins. Among the confirmed genes, OAZ1-the gene encoding the ornithine decarboxylase antizyme1-was selected for detailed investigation, since its silencing improved the reporter protein production without affecting cell viability. Silencing OAZ1 caused an increase of the ornithine decarboxylase enzyme and the cellular levels of putrescine and spermidine; an indication that increased cellular polyamines enhances luciferase expression without affecting its transcription. The study shows that OAZ1 is a novel target for improving expression of recombinant proteins. The genome-scale screening performed in this work can establish the foundation for targeted design of an efficient mammalian cell platform for various biotechnological applications. Biotechnol. Hydroxyproline-rich glycopeptides (HypSys) are small signalling peptides containing 18-20 amino acids. The expression of IbpreproHypSys, encoding the precursor of IbHypSys, was induced in sweet potato ( Ipomoea batatas cv. Tainung 57) through wounding and IbHypSys treatments by using jasmonate and H2O2. Transgenic sweet potatoes overexpressing (OE) and silencing [RNA interference (RNAi)] IbpreproHypSys were created. The expression of the wound-inducible gene for ipomoelin ( IPO) in the local and systemic leaves of OE plants was stronger than the expression in wild-type (WT) and RNAi plants after wounding. Furthermore, grafting experiments indicated that IPO expression was considerably higher in WT stocks receiving wounding signals from OE than from RNAi scions. However, wounding WT scions highly induced IPO expression in OE stocks. These results indicated that IbpreproHypSys expression contributed towards sending and receiving the systemic signals that induced IPO expression. Analysing the genes involved in the phenylpropanoid pathway demonstrated that lignin biosynthesis was activated after synthetic IbHypSys treatment. IbpreproHypSys expression in sweet potato suppressed Spodoptera litura growth . In conclusion, wounding induced the expression of IbpreproHypSys, whose protein product was processed into IbHypSys. ABSTRACT Obtaining adequate quantities of functional mammalian membrane proteins has been a bottleneck in their structural and functional studies because the expression of these proteins from mammalian cells is relatively low. To explore the possibility of enhancing expression of these proteins using miRNA, a stable T-REx-293 cell line expressing the neurotensin receptor type 1 (NTSR1), a hard-to-express G protein-coupled receptor (GPCR), was constructed. The cell line was then subjected to human miRNA mimic library screening. In parallel, an HEK293 cell line expressing luciferase was also screened with the same human miRNA mimic library. Five microRNA mimics: hsa-miR-22-5p, hsa-miR-18a-5p, hsa-miR-22-3p, hsa-miR-429, and hsa-miR-2110were identified from both screens. They led to 48% increase in the expression of functional NTSR1 and to 239% increase of luciferase expression. These miRNAs were also effective in enhancing the expression of secretedglypican-3 hFc-fusion protein from HEK293 cells.The results indicate that these molecules may have a wide role in enhancing the production of proteins with biomedical interest. User identification plays an important role for access control in networks. Recently, Hsu and Chuang proposed a user identification scheme for distributed computer networks, which can also provide key agreement and user anonymity. However, Tsai pointed out the weakness of Hsu-Chuang's scheme and proposed an improved scheme. In this paper, we present two attacks to examine that some user identification schemes have a serious security loophole. The survival rate for breast cancer drops dramatically once the disease progresses to the metastatic stage. Selenium (Se) is an essential micronutrient credited with having high anticancer and chemopreventive properties. In our study, we investigated if dietary Se supplementation modified breast cancer development in vivo. Three diets supplemented with sodium selenite, methylseleninic acid (MSA) or selenomethionine (SeMet), as well as a Se-deficient and a Se-adequate diet were fed to mice before mammary gland inoculation of 4T1.2 cells. The primary tumor growth, the numbers of cancer cells present in lungs, hearts, livers, kidneys and femurs and several proinflammatory cytokines were measured. We found that inorganic selenite supplementation provided only short-term delay of tumor growth, whereas the two organic SeMet and MSA supplements provided more potent growth inhibition. These diets also affected cancer metastasis differently. Mice fed selenite developed the most extensive metastasis and had an increased incidence of kidney and bone metastasis. On the other hand, mice fed the SeMet diet showed the least amount of cancer growth at metastatic sites. The MSA diet also provided some protection against breast cancer metastasis although the effects were less significant than those of SeMet. The cytokine profiles indicated that serum levels of interlukin-2, interleukin-6, interferon γ and vascular endothelial growth factor were elevated in SeMet-supplemented mice. There was no significant difference in tumor growth and the patterns of metastasis between the Se-deficient and Se-adequate groups. In this study, the authors consider certificateless signature (CLS) schemes for strong non‐repudiation. They show that previous security models which ensure that any user can have a unique key pair cannot guarantee a CLS scheme to achieve strong non‐repudiation. The authors then fix the security model and propose a new CLS scheme which provides strong non‐repudiation under the new model, assuming the computational Diffie–Hellman problem is intractable. Micro RNAs (mi RNAs) are small noncoding RNAs which post-transcriptionally regulate gene expression by directing mRNA cleavage or translational inhibition. mi RNAs play multiple roles in the growth, development and stress responses in plants. However, little is known of the wounding-responsive mi RNAs and their regulation., Here, we investigated the expression patterns of micro R828 (mi R828) on wounding in sweet potato ( Ipomoea batatas cv Tainung 57). The expression of mi R828 was only detected in leaves, and was induced by wounding rather than by ethylene, hydrogen peroxide ( H2 O2), methyl jasmonate or nitric oxide ( NO). Moreover, cyclic guanosine monophosphate (c GMP) was necessary for mi R828 accumulation in leaves on wounding., Two mi R828 target candidates, named IbMYB and IbTLD, were obtained by cDNA cloning, and their mRNA cleavage caused by mi R828 was confirmed by cleavage site mapping, agro-infiltration and transgenics studies. The reduction in IbMYB and IbTLD expression coincided with the induction of mi R828, demonstrating that IbMYB and IbTLD might be mi R828 targets. Furthermore, transgenic sweet potato overexpressing miR828 precursor affected lignin and H2 O2 contents., These results showed that cGMP could regulate wounding-responsive mi R828, which repressed the expression of IbMYB and IbTLD. Recent developments in micromachining techniques at the University of Michigan have resulted in novel high-performance low-loss filters for microwave and millimeter-wave applications. The idea is based on suspending the filters on thin dielectric membranes to eliminate dielectric loss and dispersion problems, resulting in a pure TEM mode of propagation and conductor-loss-limited performance. The dielectric membrane and the surrounding cavities are built using chemical etching in Silicon and GaAs wafers. The filters are therefore compatible with low-cost IC fabrication techniques and can be combined with planar diodes and transistors to result in active filter networks. Several state-of-the-art filters have been realized including 15-GHz and 20-GHz bandpass interdigital suspended stripline filters, lowpass and bandpass microshield filters at 30 GHz and 90 GHz, microstrip 94-GHz bandpass filters, and a 250-GHz bandpass filter with 1.0–1.5-dB insertion loss. This article reviews the associated fabrication techniques, the different types of transmission lines achieved using this technology, and the measured performance from 15 to 350 GHz. Objective: Wegener's granulomatosis (WG) is a systemic inflammatory disease that is associated with substantial morbidity. The aim of this study was to understand the biology underlying WG and to discover markers of disease activity that would be useful for prognosis and treatment guidance., Methods: Gene expression profiling was performed using total RNA from peripheral blood mononuclear cells (PBMCs) and granulocyte fractions from 41 patients with WG and 23 healthy control subjects. Gene set enrichment analysis (GSEA) was performed to search for candidate WG-associated molecular pathways and disease activity biomarkers. Principal components analysis was used to visualize relationships between subgroups of WG patients and controls. Longitudinal changes in proteinase 3 (PR3) gene expression were evaluated using reverse transcription-polymerase chain reaction, and clinical outcomes, including remission status and disease activity, were determined using the Birmingham Vasculitis Activity Score for WG (BVAS-WG)., Results: Eighty-six genes in WG PBMCs and 40 in WG polymorphonuclear neutrophils (PMNs) were significantly up-regulated relative to controls. Genes up-regulated in WG PBMCs were involved in myeloid differentiation, and these included the WG autoantigen PR3. The coordinated regulation of myeloid differentiation genes was confirmed by GSEA. The median expression values of the 86 up-regulated genes in WG PBMCs were associated with disease activity (P = 1.3 x 10(-4)), and WG patients with low-level expression of the WG signature genes showed expression profiles that were only modestly different from that in healthy controls (P = 0.07). PR3 transcription was significantly up-regulated in WG PBMCs (P = 1.3 x 10(-5), false discovery rate [FDR] 0.002), but not in WG PMNs (P = 0.03, FDR 0.28), and a preliminary longitudinal analysis showed that the fold change in PR3 RNA levels in WG PBMCs corresponded to changes in the BVAS-WG score over time., Conclusion: Transcription of PR3 and related myeloid differentiation genes in PBMCs may represent novel markers of disease activity in WG.
