High cannabidiol‐containing plants of Cannabis sativa (high‐CBD) growing in farms in Israel displayed foliar symptoms of interveinal chlorosis and yellowing, brittleness and occasionally necrosis. These symptoms, which were more apparent in older leaves, resembled those caused by the crinivirus lettuce chlorosis virus (LCV). However, this virus was not detected by reverse transcription (RT)‐PCR using specific primer sets. High‐throughput sequencing of viral RNA extracted from symptomatic leaves revealed the presence of cucurbit chlorotic yellows virus (CCYV), a crinivirus in the Closteroviridae family. The complete viral genome sequence was obtained using RT‐PCR followed by Sanger sequencing. The two CCYV RNA genomic segments shared 99.5%–99.85% nucleotide sequence identity with CCYV isolates from the GenBank. The virus was transmitted from symptomatic cannabis leaves to healthy plants of cannabis and Cucumis sativus 'King Star' (cucumber) by the whitefly Bemisia tabaci Middle Eastern Asia Minor 1 (MEAM1) species, causing disease symptoms identical to those of the donor plants. Cannabis‐CCYV was also transmitted between infected cucumber plants and cannabis seedlings of unknown genotype. Severe disease symptoms of yellowing and leaf‐edge necrosis were observed on high‐CBD and high Δ9‐tetrahydrocannabinol‐containing (high‐THC) flowering cannabis plants and were associated with mixed infections of LCV and CCYV. Cannabaceae are a relatively small family of angiosperms, but they include several species of huge economic and cultural significance: marijuana or hemp (Cannabis sativa) and hops (Humulus lupulus). Previous phylogenetic studies have clarified the most deep relationships in Cannabaceae, but relationships remain ambiguous among several major lineages. Here, we sampled 82 species representing all genera of Cannabaceae and utilized a new dataset of 90 nuclear genes and 82 chloroplast loci from Hyb‐Seq to investigate the phylogenomics of Cannabaceae. Nuclear phylogenetic analyses revealed a robust and consistent backbone for Cannabaceae. We observed nuclear gene‐tree conflict at several deep nodes in inferred species trees, also cyto‐nuclear discordance concerning the relationship between Gironniera and Lozanella and the relationships among Trema s.l. (including Parasponia), Cannabis + Humulus, and Chaetachme + Pteroceltis. Coalescent simulations and network analyses suggest that observed deep cyto‐nuclear discordances were most likely to stem from incomplete lineage sorting (ILS); nuclear gene‐tree conflict might be caused by both ILS and gene flow between species. All genera of Cannabaceae were recovered as monophyletic, except for Celtis, which consisted of two distinct clades: Celtis I (including most Celtis species) and Celtis II (including Celtis gomphophylla and Celtis schippii). We suggest that Celtis II should be recognized as the independent genus Sparrea based on both molecular and morphological evidence. Our work provides the most comprehensive and reliable phylogeny to date for Cannabaceae, enabling further exploration of evolutionary patterns across this family and highlighting the necessity of comparing nuclear with chloroplast data to examine the evolutionary history of plant groups. Marijuana (Cannabis) belongs to family Cannabaceae. This plant is a great natural source of cannabinoids, which have several medical applications such as it is used for the treatment of anxiety and psychosis, disorders of motility and epilepsy. However, this plant is also notorious for its pollen allergic reactions. Mainly two species, that is, C. sativa L. and C.indica Lam. are involved in allergic reactions. In this study, morpho‐palynological studies of both the species were conducted. A number of morphological differences were observed in the both species. C. sativa is tall with narrowed leaves and fibrous stalk plant while C. indica is short, bushier, broad leaves, and having woody stalk plant. Both the species are varied in their pollen characteristics. The polar diameter of C. sativa observed was 7.5 μm and equatorial diameter was 8 μm, and its shape was oblate‐spheroidal. C. indica had 7 μm polar diameter, 7.5 μm equatorial diameter but its shape was similar to C. sativa, that is, oblate‐spheroidal. About 40%‑50% people were affected with both these marijuana species and had symptoms of allergies like dry cough, congestion, itchy eyes, sore throat, nausea, a runny nose, watery eyes, and sneezing. It was concluded that both species' pollen are allergy causing and pollen of both species are different with variations in morphological and allergic symptoms. Two species of Marijuana, that is, Cannabis sativa L. and Cannabis indica Lam mainly responsible for these pollen allergies. Premise: Celtis is the most species‐rich genus of Cannabaceae, an economically important family. Celtis species have been described as wind‐pollinated and andromonoecious. However, the andromonoecy of Celtis has been debated because there are reports of monoclinous flowers with non‐opening anthers on short filaments. Our objective was to study the floral morphogenesis of Celtis to establish the breeding system and to better understand the developmental patterns that lead to the formation of reduced flowers in the genus. Methods: Flowers and floral buds of Celtis species were studied using scanning electron microscopy, high‐resolution x‐ray computed tomography, and light microscopy. Results: All flowers initiate stamens and carpels during early floral development, but either stamens or carpels abort during later stages. Thus, at anthesis, flowers are either functionally pistillate or functionally staminate. In pistillate flowers, stamens abort late and become staminodes with normal‐looking anthers. These anthers have no functional endothecium and, in most of the species studied, produce no viable pollen grains. The gynoecium is pseudomonomerous, and its vascularization is similar in the sampled species. In staminate flowers, the gynoecium aborts early resulting in small pistillodes. No vestiges of petals were found. Conclusions: The species studied are monoecious and not andromonoecious as described earlier. The absence of petals, the carpel and stamen abortion, and the pseudomonomerous gynoecium result in the reduced flowers of Celtis species. The use of high‐resolution x‐ray computed tomography was essential for a more accurate interpretation of ovary vascularization, confirming the pseudomonomerous structure of the gynoecium. Summary: We recently described, in Cannabis sativa, the oldest sex chromosome system documented so far in plants (12–28 Myr old). Based on the estimated age, we predicted that it should be shared by its sister genus Humulus, which is known also to possess XY chromosomes.Here, we used transcriptome sequencing of an F1 family of H. lupulus to identify and study the sex chromosomes in this species using the probabilistic method SEX‐DETector.We identified 265 sex‐linked genes in H. lupulus, which preferentially mapped to the C. sativa X chromosome. Using phylogenies of sex‐linked genes, we showed that a region of the sex chromosomes had already stopped recombining in an ancestor of both species. Furthermore, as in C. sativa, Y‐linked gene expression reduction is correlated to the position on the X chromosome, and highly Y degenerated genes showed dosage compensation.We report, for the first time in Angiosperms, a sex chromosome system that is shared by two different genera. Thus, recombination suppression started at least 21–25 Myr ago, and then (either gradually or step‐wise) spread to a large part of the sex chromosomes (c. 70%), leading to a degenerated Y chromosome. Summary: Tree stems undergo a massive secondary growth in which secondary xylem and phloem tissues arise from the vascular cambium. Vascular cambium activity is driven by endogenous developmental signalling cues and environmental stimuli. Current knowledge regarding the genetic regulation of cambium activity and secondary growth is still far from complete. The tropical Cannabaceae tree Parasponia andersonii is a non‐legume research model of nitrogen‐fixing root nodulation. Parasponia andersonii can be transformed efficiently, making it amenable for CRISPR‐Cas9‐mediated reverse genetics. We considered whether P. andersonii also could be used as a complementary research system to investigate tree‐related traits, including secondary growth. We established a developmental map of stem secondary growth in P. andersonii plantlets. Subsequently, we showed that the expression of the co‐transcriptional regulator PanNODULE ROOT1 (PanNOOT1) is essential for controlling this process. PanNOOT1 is orthologous to Arabidopsis thaliana BLADE‐ON‐PETIOLE1 (AtBOP1) and AtBOP2, which are involved in the meristem‐to‐organ‐boundary maintenance. Moreover, in species forming nitrogen‐fixing root nodules, NOOT1 is known to function as a key nodule identity gene. Parasponia andersonii CRISPR‐Cas9 loss‐of‐function Pannoot1 mutants are altered in the development of the xylem and phloem tissues without apparent disturbance of the cambium organization and size. Transcriptomic analysis showed that the expression of key secondary growth‐related genes is significantly down‐regulated in Pannoot1 mutants. This allows us to conclude that PanNOOT1 positively contributes to the regulation of stem secondary growth. Our work also demonstrates that P. andersonii can serve as a tree research system. Significance Statement: Parasponia andersonii Pannoot1 CRISPR‐Cas9 knockout mutants are altered for secondary growth development. Premise: How genetic variation within a species affects phytochemical composition is a fundamental question in botany. The ratio of two specialized metabolites in Cannabis sativa, tetrahydrocannabinol (THC) and cannabidiol (CBD), can be grouped into three main classes (THC‐type, CBD‐type, and intermediate type). We tested a genetic model associating these three groups with functional and nonfunctional alleles of the cannabidiolic acid synthase gene (CBDAS). Methods: We characterized cannabinoid content and assayed CBDAS genotypes of >300 feral C. sativa plants in Minnesota, United States. We performed a test cross to assess CBDAS inheritance. Twenty clinical cultivars obtained blindly from the National Institute on Drug Abuse and 12 Canadian‐certified grain cultivars were also examined. Results: Frequencies of CBD‐type, intermediate‐type, and THC‐type feral plants were 0.88, 0.11, and 0.01, respectively. Although total cannabinoid content varied substantially, the three groupings were perfectly correlated with CBDAS genotypes. Genotype frequencies observed in the test cross were consistent with codominant Mendelian inheritance of the THC:CBD ratio. Despite significant mean differences in total cannabinoid content, CBDAS genotypes blindly predicted the THC:CBD ratio among clinical cultivars, and the same was true for industrial grain cultivars when plants exhibited >0.5% total cannabinoid content. Conclusions: Our results extend the generality of the inheritance model for THC:CBD to diverse C. sativa accessions and demonstrate that CBDAS genotyping can predict the ratio in a variety of practical applications. Cannabinoid profiles and associated CBDAS segregation patterns suggest that feral C. sativa populations are potentially valuable experimental systems and sources of germplasm. [ABSTRACT FROM AUTHOR]

Dispersal scenarios have been favored over tectonic vicariance as an explanation for disjunct distributions in many plant taxa during the last two decades. However, this argument has been insufficiently addressed in cosmopolitan groups showing disjunct patterns in both the temperate and tropical regions. In this study, we used the Cannabaceae, an angiosperm family distributed in tropical and temperate regions of both the New World and the Old World, to explore the role of dispersal in shaping disjunct patterns and species diversification of cosmopolitan plants. We reconstructed the phylogenetic relationships of all 10 genera and 75 species of Cannabaceae (ca. 64.1% of recognized species) based on eight DNA regions. Based on fossil calibrations, we estimated the divergence times and net diversification rates. We further inferred the ancestral geographical ranges with several models and compared the fitness of different models. The Cannabaceae and most genera were strongly supported as monophyletic except for the Parasponia being embedded within the Trema. The Celtis were resolved into two strongly supported clades primarily corresponding to temperate and tropical regions. We inferred that the Cannabaceae originated at ca. 93 Ma, and that subsequent rampant and widespread dispersals shaped the intercontinentally disjunct distribution of the Cannabaceae. Dispersal coincides with adaptation to drier and colder climate in the Northern Hemisphere, or humid and warm climate in the tropical regions, followed by rapid species diversification. This study advances our understanding as to the formation of distribution patterns and species diversification of a plant family with tropical to temperate disjunct distributions. Premise of the Study: This study details the unusual synorganization of the staminate flower in wind‐pollinated urticalean rosids to add the missing pieces that complete the puzzle of the explosive mechanism of pollen release in this group. Methods: Flower buds and flowers were analyzed using light and scanning electron microscopy. Key Results: The pistillode, stamens, and sepals form a floral apparatus that explosively releases pollen to be carried by the wind. The anthers dehisce when the stamens are still inflexed on the floral bud and are enveloped by the sepals and supported by an inflated pistillode. The distension of the filaments presses the pistillode, which decreases the pressure exerted on the anthers by releasing the air accumulated internally through its apical orifice. The extended filaments and the dehiscent free anthers move rapidly outward from the center of the flower. This movement of the filaments is then blocked by the robust basally united sepals, which causes a rapid inversion of the anther position, thus hurling the pollen grains far from the flower. The pollen grains are released grouped by the mucilage produced in high quantity in the cells found in all floral organs. Conclusions: The anatomical structure of the pistillode and the finding of mucilaginous cells are the main features that help in the understanding the explosive mechanism of pollen release in urticalean rosids. Summary: Background: Trema micrantha is a tree widely distributed throughout the Americas. The tree produces highly palatable leaves that have been associated with natural poisoning in goats, sheep and horses, in which hepatic necrosis and hepatic encephalopathy have been observed. Objectives: This study describes malacia and haemorrhage in the central nervous system (CNS) due to T. micrantha consumption, with minimal to absent hepatic lesions. Study design: Retrospective case series. Methods: A total of 14 horses with a history of neurological signs and spontaneous consumption of T. micrantha leaves were submitted to necropsy and multiple samples were collected for histopathology. Details of clinical history and signs of the horses were obtained through inquiries to the owners and attending veterinarians. Results: All the 14 horses had neurological signs of ataxia, severe sialorrhoea, involuntary running movements, sternal and lateral recumbency, and death after a clinical course that lasted from 24 h to 9 days. For a few days prior to onset of clinical signs, all horses had spontaneously consumed, potentially toxic doses of T. micrantha leaves. All 14 brains had diffuse yellowish discoloration affecting the rhombencephalon, mesencephalon, diencephalon, telencephalon and corpus striatum. In all cases, the most severe lesions were observed in the pons. Spinal cord lesions were observed affecting the lumbar intumescence, which was swollen with darken and depressed areas at the dorsal and ventral horns, and at the sacral level, which on cut surface displayed a friable and yellowish grey matter. The lesions observed grossly in brain and spinal cord consisted microscopically of severe vasculitis and liquefactive necrosis of white and grey matter of the brainstem, cerebellum and spinal cord. Main limitations: This is a small retrospective series relying on clinical observations reported by owners and attending veterinarians. The mechanism of action of the plant toxin in the CNS is still unidentified. Background and Purpose: The aim of this study was to compare the abilities of cannabidiolic acid methyl ester (HU-580) and cannabidiolic acid (CBDA) to enhance 5-HT1A receptor activation in vitro and produce 5-HT1A -mediated reductions in nausea and anxiety in vivo.Experimental Approach: We investigated the effects of HU-580 and CBDA on (i) activation by 8-hydroxy-2-(di-n-propylamino)tetralin of human 5-HT1A receptors in CHO cell membranes, using [35 S]-GTPγS binding assays, (ii) gaping by rats in acute and anticipatory nausea models, and (iii) stress-induced anxiety-like behaviour, as indicated by exit time from the light compartment of a light-dark box of rats subjected 24 h earlier to six tone-paired foot shocks.Key Results: HU-580 and CBDA increased the Emax of 8-hydroxy-2-(di-n-propylamino) tetralin in vitro at 0.01-10 and 0.1-10 nM, respectively, and reduced signs of (i) acute nausea at 0.1 and 1 μg·kg-1 i.p. and at 1 μg·kg-1 i.p., respectively, and (ii) anticipatory nausea at 0.01 and 0.1 μg·kg-1 , and at 0.1 μg·kg-1 i.p. respectively. At 0.01 μg·kg-1 , HU-580, but not CBDA, increased the time foot-shocked rats spent in the light compartment of a light-dark box. The anti-nausea and anti-anxiety effects of 0.01 or 0.1 μg·kg-1 HU-580 were opposed by the 5-HT1A antagonist, WAY100635 (0.1 mg·kg-1 i.p.).Conclusions and Implications: HU-580 is more potent than CBDA at enhancing 5-HT1A receptor activation, and inhibiting signs of acute and anticipatory nausea, and anxiety. Consequently, HU-580 is a potential medicine for treating some nausea and anxiety disorders and possibly other disorders ameliorated by enhancement of 5-HT1A receptor activation. Our aims are to describe and explain the structure of the Cannabis Abuse Screening Test (CAST) across countries. Standard statistical analyses fail to describe and explain several variables simultaneously while taking account of the group structure of individuals. The 2011 European School Survey Project on Alcohol and other Drugs (ESPAD): 5204 last-year cannabis users aged 15-16 from 13 European countries. Multigroup principal component analysis (mgPCA) and multigroup partial least squares (mgPLS). MgPCA shows that the CAST has a two-dimensional structure (frequency of use/problems and non-recreational use/dependency symptoms). All the countries present a good concordance with the common structure, except Kosovo, Lichtenstein and Romania. MgPLS shows that three explanative variables (in a total of eight) are mainly related with the CAST (the frequencies of cannabis use in the last 12 months and in the last 30 days and the age at first cannabis use) while Kosovo, Lichtenstein and Romania also present specificities. The CAST structure appears stable in the 13 countries except for Kosovo, Lichtenstein and Romania that also show specific relationships between the CAST variables and their determinants. The role of olfactory cues for host search is much less investigated in day-active butterflies than in their relatives, the nocturnal moths. The goal of this study was to investigate whether host-plant volatiles from foliar extracts of hop, Humulus lupulus L. (Cannabaceae), evoke electroantennographic ( EAG) responses, increase landing rates, and stimulate egg-laying behavior of gravid Polygonia c-album L. (Lepidoptera: Nymphalidae) females. Eighty-nine volatile compounds were detected in a non-concentrated methanol extract of hop by gas chromatography-mass spectrometry, 11 of which elicited an EAG response. Concentration of the crude extract significantly reduced landing rates on artificial leaves treated with the sample due to loss of volatile compounds, but after landing the oviposition response of gravid females was not affected. A mixture of eight commercially available EAG-active volatiles increased the landing rate of gravid females to their source but did not act as oviposition stimulants. Dividing the volatile compounds into two groups - consisting of (1) hexanal, ( E)-2-hexenal, octanal, nonanal, and decanal, and (2) sulcatone, humulene, and benzyl alcohol - obliterated effectiveness, revealing synergism between compounds. Although volatiles did not stimulate oviposition, they significantly contributed to the distribution of eggs by increasing the landing rates on treated artificial leaves. [ABSTRACT FROM AUTHOR]

Noil hemp fiber-reinforced polypropylene composites were fabricated using intermixer and injection molding machines. X-ray microtomography and Weibull statistical methods were employed to characterize the aspect ratio distributions of noil hemp fibers in the polypropylene matrices. The influence of fiber content (0-40 wt%) and compatibilizer addition (5 wt%) on IFSS (interfacial shear strengths) was evaluated by means of the modified Bowyer and Bader model. The evaluated IFSSs decreased from 9.7 to 7.2 MPa as the fiber content increased from 10 to 40 wt%. Also, the outcomes indicated increases to IFSSs for the maleic anhydride grafted polypropylene (MAPP)-coupled composites than uncoupled ones. They were used to predict theoretical tensile strength of the composites. A good agreement has been found between the theoretical and the experimental tensile strengths of composites indicating that the developed model has excellent capability to predict the tensile strength of noil hemp fiber reinforced polypropylene composites. Ultimately, the influences of interfacial shear strength; fiber strength and fiber aspect ratio were investigated using the developed model to predict composite tensile strengths. Urtica cannabina ( U. cannabina), a member of the Urticaceae family, is widely distributed throughout the temperate regions of the world and can be used as a nutritious feed for animals through the winter period. To provide high-quality forage all year-round, we treated freshly harvested U. cannabina without additives (control), but with corn flour ( CF) (5:1 w/w), molasses (2, 4, and 8% fresh weight), or Lalsi L Dry ( LD) inoculant (5, 10 and 20 mg kg−1 of fresh weight). We then assessed the chemical composition, in vitro digestibility and fermentative parameters of the products after 0, 3, 5, 15, 20 and 60 d of ensiling. The results showed that: (i) U. cannabina had large quantities of protein and some essential minerals, including calcium, potassium, sodium, zinc, copper and manganese, and was particularly rich in magnesium and iron. (ii) U. cannabina can be preserved as a highly nutritious silage. No additive treatment or the LD inoculant treatments produced badly preserved silages. The 2% molasses treatment produced badly preserved silage, but 4-8% molasses produced well-preserved silages. The CF treatment also produced well-preserved silage. We recommend the application rates of molasses at 4-8% of fresh weight or 5:1 CF to improve U. cannabina silage. [ABSTRACT FROM AUTHOR]

Cannabaceae includes ten genera that are widely distributed in tropical to temperate regions of the world. Because of limited taxon and character sampling in previous studies, intergeneric phylogenetic relationships within this family have been poorly resolved. We conducted a molecular phylogenetic study based on four plastid loci (atpB-rbcL, rbcL, rpsl6, trnL-trnF) from 36 ingroup taxa, representing all ten recognized Cannabaceae genera, and six related taxa as outgroups. The molecular results strongly supported this expanded family to be a monophyletic group. All genera were monophyletic except for Trema, which was paraphyletic with respect to Parasponia. The Aphananthe clade was sister to all other Cannabaceae, and the other genera formed a strongly supported clade further resolved into a Lozanella clade, a Gironniera clade, and a trichotomy formed by the remaining genera. Morphological ancestral state reconstructions indicated the complex evolution pattern of most analyzed morphological characters, and it is difficult to identify morphological synapomorphies for most clades within Cannabaceae. Pre-dispersal seed predation by granivorous birds has potential to limit fruit removal and subsequent seed dispersal by legitimate avian seed dispersers in bird-dispersed plants, especially when the birds form flocks. We monitored pre-dispersal seed predation by the Japanese grosbeak, Eophona personata, of two bird-dispersed hackberry species (Cannabaceae), Celtis biondii (four trees) and Celtis sinensis (10 trees), for 3 years (2005, 2007 and 2008) in a fragmented forest in temperate Japan. Throughout the 3 years, predation was more intense on C. biondii, which, as a consequence, lost a larger part of its fruit crop. Grosbeaks preferred C. biondii seeds that had a comparatively lower energy content and lower hardness than C. sinensis, suggesting an association between seed hardness and selective foraging by grosbeaks. In C. biondii, intensive predation markedly reduced fruit duration and strongly limited fruit removal by seed dispersers, especially in 2007 and 2008. In C. sinensis, seed dispersers consumed fruits throughout the fruiting seasons in all 3 years. In C. biondii, variation in the timing of grosbeak migration among years was associated with annual variation in this bird's effects on fruit removal. Our results demonstrate that seed predation by flocks of granivorous birds can dramatically disrupt seed dispersal in fleshy-fruited plants and suggest the importance of understanding their flocking behaviour. After an absence of 50 years, the currant pug moth, Eupithecia assimilata Doubleday (Lepidoptera: Geometridae), has reappeared as an important pest of hops, Humulus lupulus L. (Cannabaceae), in the UK. Pheromone gland extracts from virgin female E. assimilata moths were shown to contain (3 Z,6 Z)- cis-9,10-epoxyheneicosadiene (3 Z,6 Z- cis-9,10-epoxy-21:H) by gas chromatography linked to mass spectrometry. (3 Z,6 Z,9 Z)-heneicosatriene (3 Z,6 Z,9 Z-21:H) was also found as a minor component in one of two extracts. In field experiments, significant numbers of male E. assimilata moths were caught in traps baited with the (9 S,10 R)-enantiomer of 3 Z,6 Z- cis-9,10-epoxy-21:H but not in those baited with the (9 R,10 S)-enantiomer or racemic mixture. Addition of 3 Z,6 Z,9 Z-21:H at the ratio present in gland extracts greatly reduced the attractiveness of the epoxydiene. [ABSTRACT FROM AUTHOR]

Cannabinoids are important chemotaxonomic markers unique to Cannabis. Previous studies show that a plant's dry-weight ratio of Δ9-tetrahydrocannabinol (THC) to cannabidiol (CBD) can be assigned to one of three chemotypes and that alleles BD and BT encode alloenzymes that catalyze the conversion of cannabigerol to CBD and THC, respectively. In the present study, the frequencies of Bo and BT in sample populations of 157 Cannabis accessions were determined from CBD and THC banding patterns, visualized by starch gel electrophoresis. Gas chromatography was used to quantify cannabinoid levels in 96 of the same accessions. The data were interpreted with respect to previous analyses of genetic and morphological variation in the same germplasm collection. Two biotypes (infraspecific taxa of unassigned rank) of C. sativa and four biotypes of C. indica were recognized. Mean THC levels and the frequency of BT were significantly higher in C. indica than C. sativa. The proportion of high THC/CBD chemotype plants in most accessions assigned to C. sativa was <25% and in most accessions assigned to C. indica was >25%. Plants with relatively high levels of tetrahydrocannabivarin (THCV) and/or cannabidivarin (CBDV) were common only in C. indica. This study supports a two-species concept of Cannabis. Cannabinoids are increasingly being considered for the management of various painful conditions, and could be considered as an option for treating acute pain in sickle cell disease (SCD). The objective of this study was to determine the extent of use of cannabis in the community for pain and other symptom relief, and its side effects during self-administration in patients with SCD. Patients attending Central Middlesex Hospital in London were invited to complete a structured self-administered anonymous questionnaire. Eighty-six young adults with HbSS, HbSC and HbSbetathalassaemia disease (median age 30 years) participated in the study. Results showed that 31 (36%) had used cannabis in the previous 12 months to relieve symptoms associated with SCD. The main route in all but two patients was by smoking. The main reasons for use were to reduce pain in 52%, and to induce relaxation or relieve anxiety and depression in 39%. Symptoms related to sedation and mood effects were reported in 77% of patients. The majority of patients (58%) expressed their willingness to participate in studies of cannabis as a medicine. We conclude that research in the use of cannabinoids for pain relief in SCD would be both important and acceptable to adult patients.
