Components of these two developmental and cancer-associated pathways, including (Patched)PTCH,SMOH,adenomatous polyposis coli(APC),β-cateninandAXIN1show somatic mutations in sporadic MBs. In this study we analysedSUFU(human suppressor of fused), which acts as a negative regulator of both the Shh and Wnt signalling pathways and therefore represents a putative tumour suppressor gene, to find out if it is also involved in the pathogenesis of sporadic MBs. We screened 145 primitive neuroectodermal tumours (PNETs) including 90 classic MBs, 42 of the desmoplastic variant and two medullomyoblastomas as well as 11MB cell lines for mutations using single-strand conformational polymorphism (SSCP) and sequencing analysis. 18% of the MBs exhibited allelic losses on chromosome 10q. In contrast to a previous report, in which truncating mutations ofSUFUhave been identified in 9% of MBs, we were not able to identify somatic mutations ofSUFUin our large tumour panel. We uncovered single nucleotide polymorphisms (SNPs) in exon 4, 8, 11 and in intron 2 in theSUFUgene. Expression analysis by competitive reverse transciption-polymerase chain reaction (RT-PCR) revealed no difference inSUFUmRNA levels of both MB subtypes and normal foetal or adult cerebellar tissues. Medulloblastomas (MBs) are malignant primitive neuroectodermal tumours (PNETs) of the cerebellum occurring predominantly in childhood. The association of monosomy of chromosome 22 with MB is controversial. Atypical teratoid/rhabdoid tumours (AT/RTs) of the brain share clinical and histological features with MBs and supratentorial PNETs (sPNETs). In particular, AT/RTs can be misdiagnosed as MBs and sPNETs because AT/RTs frequently contain areas of primitive neuroepithelial cells similar to PNETs. Recently, mutations of the tumour suppressor gene hSNF5/INI1 , located on 22q11.23, have been described in AT/RTs, MBs and sPNETs, with conflicting data on the prevalence of hSNF5/INI1 mutations in the latter entities. Therefore, we screened MBs for point mutations and homozygous deletions of the hSNF5/INI1 tumour suppressor gene. In 90 MBs, no mutations of the hSNF5/INI1 gene were identified. Thus, our study virtually rules out hSNF5/INI1 as a tumour suppressor gene involved in the pathogenesis of medulloblastoma. [ABSTRACT FROM AUTHOR]

We previously used fhuF as a sensitive reporter gene of the iron status of Escherichia coli. In this report, the fhuF gene was identified as open reading frame f262b at 99.2 min on the genome sequence map of E. coli K-12. The FhuF protein was labeled with a His-tag and then purified to electrophoretic homogeneity. Based on sulfur determinations and Mössbauer and EPR spectroscopy, FhuF was identified as a [2Fe-2S] protein. The g values (gx = 1.886, gy = 1.961, gz = 1.994) and some of the Mössbauer parameters of FhuF obtained [oxidized protein as isolated : ΔEQ,4.2K = 0.474 mm s-1; Fe3+ (reduced protein) : ΔEQ = 0.978 mm s-1] are not typical of common [2Fe-2S] proteins and indicate that FhuF has unusual structural properties. The primary sequence of FhuF does not show any sequence similiarities to known [2Fe-2S] proteins. By site-directed mutagenesis, each of the six cysteines of FhuF was replaced by serine. EPR of the six reduced mutant proteins revealed that the terminal cysteine residues 244, 245, 256, and 259 form the [2Fe-2S]Cys4 cluster. Mutants having the Cys-to-Ser replacement at positions 244, 245, 256, or 259 did not complement a fhuF mutant. The motif Cys-Cys-Xaa10-Cys-Xaa2-Cys in FhuF differs considerably from the motif Cys-Xaa2-Cys-Xaa9-15-Cys-Xaa2-Cys found in other [2Fe-2S] proteins. The unusual Cys-Cys terminal group of the cluster may explain the atypical EPR and Mössbauer spectroscopic properties of the FhuF protein; possibly the tetrahedral symmetry at the ferric ion site is distorted. The phenotype of fhuF mutants and the structural features of the FhuF protein suggest that FhuF is involved in the reduction of ferric iron in cytoplasmic ferrioxamine B. [ABSTRACT FROM AUTHOR]

The interaction of the antitumor agent daunomycin (DN) with ferric iron has been analysed by Mōssbauer spectroscopy, EPR, extended X-ray absorption fine structure (EXAFS), and magnetic susceptibility measurements. In contrast to literature data, at millimolar iron and anthracycline concentrations no solitary Fc(DN)3 complexes are formed in appreciable amounts. The Mössbauer spectroscopic analysis revealed severe dependencies on temperature, on the preparation procedure, the time allowed for equilibration, and on the metal/ligand ratio. The Mössbauer spectra exhibit two components: a broad magnetic sextet and a quadrupole doublet at an Fe/DN molar ratio of 1:3 and exclusively a doublet at a molar ratio of 1:20, indicating an equilibrium of these two spectral components. The EPR spectra are dominated by a signal at geff = 2. Double integration of the EPR signals enabled the determination of their spin density and a correlation between EPR and Mössbauer spectra. The Mössbauer sextet species is EPR invisible and corresponds to magnetically ordered polynuclear aggregates with high magnetic anisotropy. EXAFS and susceptibility measurements provide additional evidence for the formation of polynuclear aggregates of ferric daunomycin. The quadrupole doublet species in the Mössbauer spectra correlates with the g = 2 signal in EPR. This species is also related to a magnetically ordered system, exhibiting, however, superparamagnetic behavior due to less magnetic anisotropy. Since daunomycin forms dimers in aqueous solution at millimolar concentrations, we conclude that the cooperative phenomena observed in EPR and Mössbauer spectra are a consequence of its stacking effects. To date it has barely been recognized that the nature of about 75% of the Escherichia coli iron pool is unknown. Here we report the isolation of two iron species representing major components of iron metabolism in various growth states of E. coli. In vivo Mössbauer spectroscopy was applied to obtain information on the intracellular distribution pattern of iron in E. coli K12 W3110. Only two types of iron could be detected in the cell spectra: hexacoordinated Fe2+ and Fe3+ high-spin complexes. Other iron-requiting compounds are at least one order of magnitude less abundant in E. coli. The Mössbauer parameters of these complexes fit neither cytochromes nor iron-sulfur proteins nor ferric holo-bacterioferritin. They are sensitive to metabolic changes and inhibitors. The ratio of Fe/subunit, Fe2+/Fe3+ interconversion, chromatographic and electrophoretic data exclude bacterioferritin as the main iron metabolite in E. coli. Bacterioferritin can be observed only at very high ferric ion concentrations in the medium. The 55Fe fluorograms of both cytoplasmic and membrane fractions exhibit two exclusive bands with apparent molecular masses of 17 and 15 kDa, respectively. The two bands comprised 70% of the applied radioactivity. In gel filtration the main iron peak elutes at 155 kDa yielding two bands with apparent molecular masses of 17 and 15 kDa on SDS/PAGE. We therefore conclude that the iron species form a protein with an apparent molecular mass of 155 kDa containing 17-kDa and 15-kDa subunits. The iron content of the protein is 44 μg Fe/mg protein which corresponds to approximately 13 iron ions/subunit. No iron protein exhibiting the observed features has been described so far. Additional Mössbauer experiments suggest that these novel iron proteins are not restricted to E. coli but that similar components are detectable in several bacterial and fungal systems, thus pointing to a general occurrence. 1. Mössbauer spectra of whole cells of Neurospora crassa arg-5 ota aga (a siderophore-free mutant) show that the siderophore coprogen is accumulated inside the cell as an entity. 57Fe from 57Fe-labeled coprogen is slowly removed from the complex (45% in 27 h). The rate of removal depends on the degree of iron starvation of the cells. 2. The distribution of 55Fe from [55Fe]coprogen in vacuoles, membranes, and cytoplasm has been also determined. From this it is clear that coprogen is accumulated in the cytoplasm. 3. In addition to its role as a siderophore, coprogen serves as an iron-storage compound. No holoferritins could be detected. We therefore conclude that this type of iron-storage protein is lacking in N. crassa. 4. Metabolized iron was found predominantly to exist as an envelope of Fe(II) high-spin (δ = 1.2-1.3 mm s-1; ΔEQ = 3.0- 3.1 mm s-1 at 4.2 K) and fast-relaxing Fe(III) high-spin species (δ ≈ 0.25 mm s-1 and 0.45 mm s-1; ΔEQ ≈ 0.6 mm s-1 and 0.55 mm s-1, respectively, at 4.2 K). An assignment of these major iron metabolites is difficult. The Mössbauer data of the Fe(II) species do not fit those reported for heine, cytochromes and ferredoxins. We therefore assume that this iron metabolite represents a novel internal iron compound. One of the Fe(III) species becomes the dominant component of the cell spectra after 65 h of metabolization and might correspond to an iron-storage compound with iron oxide cores similar to bacterioferritin. 5. After 27 h of growth in mycelia supplied with 57Fe-labeled coprogen, the siderophore ferricrocin was observed in the cell spectra. This is unexpected, since N. crassa arg-5 ora aga is unable to synthesize omithine. We assume that ferricrocin is synthesized by the use of coprogen degradation products. [ABSTRACT FROM AUTHOR]

Objectives: We investigated the genetic history of southern African populations with a special focus on their paternal history. We reexamined previous claims that the Y-chromosome haplogroup E1b1b (E-M293) was brought to southern Africa by pastoralists from eastern Africa, and investigated patterns of sex-biased gene flow in southern Africa., Materials and Methods: We analyzed previously published complete mtDNA genome sequences and ∼900 kb of NRY sequences from 23 populations from Namibia, Botswana, and Zambia, as well as haplogroup frequencies from a large sample of southern African populations and 23 newly genotyped Y-linked STR loci for samples assigned to haplogroup E1b1b., Results: Our results support an eastern African origin for Y-chromosome haplogroup E1b1b (E-M293); however, its current distribution in southern Africa is not strongly associated with pastoralism, suggesting more complex demographic events and/or changes in subsistence practices in this region. The Bantu expansion in southern Africa had a notable genetic impact and was probably a rapid, male-dominated expansion. Our finding of a significant increase in the intensity of the sex-biased gene flow from north to south may reflect changes in the social dynamics between Khoisan and Bantu groups over time., Conclusions: Our study shows that the population history of southern Africa has been complex, with different immigrating groups mixing to different degrees with the autochthonous populations. The Bantu expansion led to heavily sex-biased admixture as a result of interactions between Khoisan females and Bantu males, with a geographic gradient which may reflect changes in the social dynamics between Khoisan and Bantu groups over time., (© 2018 Wiley Periodicals, Inc.)

The Khoisan populations of southern Africa are known to harbor some of the deepest-rooting lineages of human mtDNA; however, their relationships are as yet poorly understood. Here, we report the results of analyses of complete mtDNA genome sequences from nearly 700 individuals representing 26 populations of southern Africa who speak diverse Khoisan and Bantu languages. Our data reveal a multilayered history of the indigenous populations of southern Africa, who are likely to be the result of admixture of different genetic substrates, such as resident forager populations and pre-Bantu pastoralists from East Africa. We find high levels of genetic differentiation of the Khoisan populations, which can be explained by the effect of drift together with a partial uxorilocal/multilocal residence pattern. Furthermore, there is evidence of extensive contact, not only between geographically proximate groups, but also across wider areas. The results of this contact, which may have played a role in the diffusion of common cultural and linguistic features, are especially evident in the Khoisan populations of the central Kalahari., (Copyright © 2013 Wiley Periodicals, Inc.)

RECQL4 mutations cause genetic instability and increase the risk of malignant disease. We report on a patient with compound heterozygosity for two novel RECQL4 mutations: mutation c.1919_1924delTCACAG, p.L640_A642delinsP in exon 12 of the RECQL4 gene and mutation c.1704+1G>A in intron 10 of the RECQL4 gene. He subsequently developed large cell anaplastic T cell lymphoma at the age of 9 years, diffuse large cell B lymphoma and osteosarcoma when he was 14 years old, and finally acute lymphatic leukemia when he was 21 years old. The most remarkable clinical features are young age, spontaneous remission of diffuse large cell lymphoma, and severe CNS and skin toxicity of cytotoxic treatment., ((c) 2010 Wiley-Liss, Inc.)
