Distinct microbial habitats on glacial surfaces are dominated by snow and ice algae, which are the critical players and the dominant primary colonisers and net producers during the melt season. Here for the first time we have evaluated the role of these algae in association with the full microbial community composition (i.e., algae, bacteria, archaea) in distinct surface habitats and on 12 glaciers and permanent snow fields in Svalbard and Arctic Sweden. We cross-correlated these data with the analyses of specific metabolites such as fatty acids and pigments, and a full suite of potential critical physico-chemical parameters including major and minor nutrients, and trace metals. It has been shown that correlations between single algal species, metabolites, and specific geochemical parameters can be used to unravel mixed metabolic signals in complex communities, further assign them to single species and infer their functionality. The data also clearly show that the production of metabolites in snow and ice algae is driven mainly by nitrogen and less so by phosphorus limitation. This is especially important for the synthesis of secondary carotenoids, which cause a darkening of glacial surfaces leading to a decrease in surface albedo and eventually higher melting rates. Virus concentrations, virus-to-bacterium ratios ( VBRs) and levels of lysogenic phage were measured throughout the 110-m water column of epishelf Beaver Lake during the austral summer of 2003/2004. The aim was to determine which factor(s) controlled viral dynamics using detailed concomitant published data on inorganic nutrients, chlorophyll a, dissolved organic carbon ( DOC) and bacterial production for Beaver Lake ( Freshwater Biology, 51, 2006, 1119). Denaturing Gradient Gel Electrophoresis (DGGE) analysis of the bacterial community was also undertaken to investigate the potential relationship between bacterial community composition and viral dynamics., Virus concentration ranged between 1.43 × 104 and 302.4 × 104 viruses mL−1 and showed a clear increase over the summer, while bacterial concentrations exhibited no seasonal pattern (range: 9.6 × 104 to 44.6 × 104 cells mL−1). Consequently, VBR varied with highest values in January (3.32-7.33)., The percentage of lysogenic phage was low, ranging from 0 to 11.69%, suggesting that other life cycles (i.e. lytic or possibly pseudolysogeny) were taking place. Attempts to measure viral production using both the Td R incorporation technique and the dilution technique failed to produce consistent results, in common with other ultra-oligotrophic Antarctic lakes., There were significant correlations between viral concentration and each of chlorophyll a, DOC, ammonium and nitrate concentrations and bacterial production, but not between bacterial abundance and either soluble reactive phosphorus or temperature., Lack of change in the dominant bacterial community composition over summer suggests that changes in viral concentrations were a function of changes in other biological and physicochemical factors, rather than changes in host/phage infection., The results suggest a significant connection between viruses and bacteria, with the DOC pool acting as a conduit for the movement of carbon between the two components. Cryoconite holes, which can cover 0.1–10% of the surface area of glaciers, are small, water-filled depressions (typically <1 m in diameter and usually <0.5 m deep) that form on the surface of glaciers when solar-heated inorganic and organic debris melts into the ice. Recent studies show that cryoconites are colonized by a diverse range of microorganisms, including viruses, bacteria and algae. Whether microbial communities on the surface of glaciers are actively influencing biogeochemical cycles or are just present in a dormant state has been a matter of debate for long time. Here, we report primary production and community respiration of cryoconite holes upon glaciers in Svalbard, Greenland and the European Alps. Microbial activity in cryoconite holes is high despite maximum temperatures seldom exceeding 0.1 °C. In situ primary production and respiration in cryoconites during the summer is often comparable with that found in soils in warmer and nutrient richer regions. Considering only glacier areas outside Antarctica and a conservative average cryoconite distribution on glacial surfaces, we found that on a global basis cryoconite holes have the potential to fix as much as 64 Gg of carbon per year (i.e. 98 Gg of photosynthesis minus 34 Gg of community respiration). Most lakes and rivers are generally considered as heterotrophic systems, but our results suggest that glaciers, which contain 75% of the freshwater of the planet, are largely autotrophic systems. There is now compelling evidence that microbially mediated reactions impart a significant effect upon the dynamics, composition, and abundance of nutrients in glacial melt water. Consequently, we must now consider ice masses as ecosystem habitats in their own right and address their diversity, functional potential, and activity as part of alpine and polar environments. Although such research is already underway, its fragmentary nature provides little basis for developing modern concepts of glacier ecology. This paper therefore provides a much-needed framework for development by reviewing the physical, biogeochemical, and microbiological characteristics of microbial habitats that have been identified within glaciers and ice sheets. Two key glacial ecosystems emerge, one inhabiting the glacier surface (the supraglacial ecosystem) and one at the ice-bed interface (the subglacial ecosystem). The supraglacial ecosystem is characterized by a diverse consortium of microbes (usually bacteria, algae, phytoflagellates, fungi, viruses and occasional rotifers, tardigrades, and diatoms) within the snowpack, supraglacial streams, and melt pools (cryoconite holes). The subglacial system is dominated by aerobic/anaerobic bacteria and most probably viruses in basal ice/till mixtures and subglacial lakes. A third, so-called englacial ecosystem is also described, but it is demonstrated that conditions within glacier ice are sufficient to make metabolic activity and its impact upon nutrient dynamics negligible at the glacier scale. Consideration of the surface and internal heat balances of the glacier show that all glacial ecosystems are sensitive to climate change, although at different timescales. Thus, while rapid, melt-driven habitat changes lead to melt-out, resuscitation, and redistribution of microorganisms in many supraglacial ecosystems, much slower climatic and glacial mass-balance processes effect such changes in the subglacial ecosystem. Paradoxically, it is shown that these forces have brought about net refreezing and the onset of cryostasis in the subglacial ecosystems of many Arctic glaciers subject to thinning in recent decades. Microbial blooms colonize the Greenland Ice Sheet bare ice surface during the ablation season and significantly reduce its albedo. On the ice surface, microbes are exposed to high levels of irradiance, freeze–thaw cycles, and low nutrient concentrations. It is well known that microorganisms secrete metabolites to maintain homeostasis, communicate with other microorganisms, and defend themselves. Yet, the exometabolome of supraglacial microbial blooms, dominated by the pigmented glacier ice algae Ancylonema alaskanum and Ancylonema nordenskiöldii, remains thus far unstudied. Here, we use a high‐resolution mass spectrometry‐based untargeted metabolomics workflow to identify metabolites in the exometabolome of microbial blooms on the surface of the southern tip of the Greenland Ice Sheet. Samples were collected every 6 h across two diurnal cycles at 5 replicate sampling sites with high similarity in community composition, in terms of orders and phyla present. Time of sampling explained 46% (permutational multivariate analysis of variance [PERMANOVA], pseudo‐F = 3.7771, p = 0.001) and 27% (PERMANOVA, pseudo‐F = 1.8705, p = 0.001) of variance in the exometabolome across the two diurnal cycles. Annotated metabolites included riboflavin, lumichrome, tryptophan, and azelaic acid, all of which have demonstrated roles in microbe–microbe interactions in other ecosystems and should be tested for potential roles in the development of microbial blooms on bare ice surfaces. Glacier and ice sheet surfaces host diverse communities of microorganisms whose activity (or inactivity) influences biogeochemical cycles and ice melting. Supraglacial microbes endure various environmental extremes including resource scarcity, frequent temperature fluctuations above and below the freezing point of water, and high UV irradiance during summer followed by months of total darkness during winter. One strategy that enables microbial life to persist through environmental extremes is dormancy, which despite being prevalent among microbial communities in natural settings, has not been directly measured and quantified in glacier surface ecosystems. Here, we use a combination of metabarcoding and metatranscriptomic analyses, as well as cell‐specific activity (BONCAT) incubations to assess the diversity and activity of microbial communities from glacial surfaces in Iceland and Greenland. We also present a new ecological model for glacier microorganisms and simulate physiological state‐changes in the glacial microbial community under idealized (i) freezing, (ii) thawing, and (iii) freeze–thaw conditions. We show that a high proportion (>50%) of bacterial cells are translationally active in‐situ on snow and ice surfaces, with Actinomycetota, Pseudomonadota, and Planctomycetota dominating the total and active community compositions, and that glacier microorganisms, even when frozen, could resume translational activity within 24 h after thawing. Our data suggest that glacial microorganisms respond rapidly to dynamic and changing conditions typical of their natural environment. We deduce that the biology and biogeochemistry of glacier surfaces are shaped by processes occurring over short (i.e., daily) timescales, and thus are susceptible to change following the expected alterations to the melt‐regime of glaciers driven by climate change. A better understanding of the activity of microorganisms on glacier surfaces is critical in addressing the growing concern of climate change in Polar regions, as well as for their use as analogues to life in potentially habitable icy worlds. Summary: Snow algae are predicted to expand in polar regions due to climate warming, which can accelerate snowmelt by reducing albedo. Green snow frequently occurs near penguin colonies, and red snow distributes widely along ocean shores. However, the mechanisms underpinning the assemblage of algae and heterotrophs in colored snow remain poorly characterized.We investigated algal, bacterial, and fungal communities and their interactions in red and green snows in the Antarctic Peninsula using a high‐throughput sequencing method.We found distinct algal community structure in red and green snows, and the relative abundance of dominant taxa varied, potentially due to nutrient status differences. Contrastingly, red and green snows exhibited similar heterotrophic communities (bacteria and fungi), whereas the relative abundance of fungal pathogens was substantially higher in red snow by 3.8‐fold. Red snow exhibited a higher network complexity, indicated by a higher number of nodes and edges. Red snow exhibited a higher proportion of negative correlations among heterotrophs (62.2% vs 3.4%) and stronger network stability, suggesting the red‐snow network is more resistant to external disturbance.Our study revealed that the red snow microbiome exhibits a more stable microbial network than the green snow microbiome. Cyanobacteria are important photoautotrophs in extreme environments such as the McMurdo Dry Valleys, Antarctica. Terrestrial Antarctic cyanobacteria experience constant darkness during the winter and constant light during the summer which influences the ability of these organisms to fix carbon over the course of an annual cycle. Here, we present a unique approach combining community structure, genomic and photophysiological analyses to understand adaptation to Antarctic light regimes in the cyanobacterium Leptolyngbya sp. BC1307. We show that Leptolyngbya sp. BC1307 belongs to a clade of cyanobacteria that inhabits near‐surface environments in the McMurdo Dry Valleys. Genomic analyses reveal that, unlike close relatives, Leptolyngbya sp. BC1307 lacks the genes necessary for production of the pigment phycoerythrin and is incapable of complimentary chromatic acclimation, while containing several genes responsible for known photoprotective pigments. Photophysiology experiments confirmed Leptolyngbya sp. BC1307 to be tolerant of short‐term exposure to high levels of photosynthetically active radiation, while sustained exposure reduced its capacity for photoprotection. As such, Leptolyngbya sp. BC1307 likely exploits low‐light microenvironments within cyanobacterial mats in the McMurdo Dry Valleys. [ABSTRACT FROM AUTHOR]

Subglacial environments are largely anoxic, contain organic carbon ( OC) overridden by glacier ice during periods of advance, and harbour active microbial communities. This creates favourable conditions for OC degradation via methanogenesis. It has been hypothesized that OC beneath ice sheets is converted to methane ( CH4) and may be released to the atmosphere during retreat. However, there are limited data available to support this contention. Here, we present new data on the abundance, diversity and activity of methanogenic archaea and the amount and character of OC in subglacial sediments from Arctic and Antarctic glacial systems based on different substrate types. We employed long-term laboratory incubations to quantify the CH4 production potential in different subglacial settings. Significant numbers of methanogens (up to 7 × 104 cells g−1) were detected in the samples and clones of Methanomicrobiales and Methanosarcinales were identified in clone libraries. Long lag periods (up to >200 days) were observed before significant CH4 concentrations were measured. We report order of magnitude differences in rates of CH4 production (101-105 fmol g−1 d−1) in different subglacial sediments, reflecting contrasts in the origin of the sediment and the OC character. Hence, we predict that contrasting rates of CH4 production are likely to occur beneath glaciers and ice sheets that overran different types of substrate. We subsequently estimated the potential for CH4 production beneath the Laurentide/Inuitian/Cordilleran and Fennoscandian Ice Sheets during a typical 85 ka Quaternary glacial/interglacial cycle. CH4 production from lacustrine-derived OC is likely to be an order of magnitude higher (~6.3-27 Pg C) than that from overridden soils (~0.55-0.68 Pg C), possibly due to a difference in lability between lacustrine and soil OC. While representing a fraction of the entire OC pool (~418-610 Pg C), this finding highlights the importance of considering the character of different OC pools when calculating subglacial CH4 production. The frequency of visibly phage-infected bacterial cells (FVIB) and the average number of phages per cell [i.e. burst size (BS)] were determined in Antarctic and Arctic ultra-oligotrophic freshwater environments. Water samples were collected from two Antarctic freshwater lakes and cryoconite holes from a glacier in the Arctic. Data from this bipolar study show the highest FVIB (average 26.1%, range 5.1% to 66.7%) and the lowest BS (average 4, range 2–15) ever reported in the literature. The bacterial density is low in these ultra-oligotrophic freshwater environments but a large proportion of the bacteria are visibly infected. Our results suggest that a constant virioplankton population can be maintained in these extreme environments even though host density is low and often slow growing. [ABSTRACT FROM AUTHOR]

We examined the response of bacteria and proto- and metazooplankton to photomodified dissolved organic matter (DOM). Sterile filtered water from a eutrophic and a humic lake, that was either exposed to artificial ultraviolet (UV) radiation or kept dark, was added to semicontinuous laboratory microcosms that lasted for 7 weeks. Bacterial production responded positively to photochemical modification of DOM regardless of lake type. Final heterotrophic biomass (bacteria + proto + metazooplankton) was 47 ± 5 and 37 ± 5 µg carbon (C) L-1 in microcosms with UV-exposed and unaltered eutrophic water DOM and 15 ± 4 and 11 ± 2 µg C L-1 in microcosms with UV-exposed and unaltered humic water DOM, respectively. For the eutrophic water, there were no significant differences in proto- or metazooplankton biomasses between microcosms receiving UV-exposed or nonexposed DOM. Differences between eutrophic water microcosms were not significant when flagellates, ciliates, cladocerans, and copepods were examined separately. In microcosms with UV-exposed humic water, biomasses of heterotrophic flagellates, rotifers, nauplii, and cladocerans were higher than in those with nonexposed DOM. Higher final metazooplankton biomass following addition of UV-exposed humic water indicates that photochemically modified DOM can be effectively transferred through the microbial loop.
