Background and Purpose: Acetylcholine receptor antibody (AChR‐Ab) detection is crucial in myasthenia gravis (MG) diagnosis and, currently, the radioimmunoassay (RIA) is the gold standard. However, RIA may detect AChR‐Ab against nonpathogenic intracellular epitopes. In this study, we performed fixed cell‐based assay (F‐CBA) in RIA‐AChR‐Ab positive subjects without MG symptoms, to assess whether F‐CBA could show a higher specificity compared to RIA in detecting pathogenic Abs. Methods: We reviewed medical records of patients referred to our MG outpatient clinic because of RIA‐AChR‐Ab detection. MG diagnosis was based on clinical examination, electrophysiology and Ab detection. AChR‐Abs were tested by RIA in the whole cohort. Serum samples from RIA‐positive asymptomatic subjects were retested by F‐CBA. Results: Of 605 subjects who tested RIA‐AChR‐Ab positive, MG diagnosis was confirmed in 599. Six subjects were RIA‐AChR‐Ab positive although they had never had MG symptoms; in four of these subjects AChR‐Abs were not detected by F‐CBA, whereas the remaining two (both non‐MG thymoma cases) were positive also by F‐CBA. Conclusions: RIA false positivity for AChR‐Ab is very rare. Previous literature has demonstrated that F‐CBA has higher sensitivity than RIA for MG, especially in ocular cases. Cancer frequency in muscle‐specific kinase myasthenia gravis (MuSK‐MG) has not yet been explored and the mechanisms leading to the formation of MuSK IgG remain elusive. We aimed to explore cancer frequency in MuSK‐MG patients and to assess MuSK expression in cancer cells from 2 tumors occurred in this cohort. Immunohistochemistry on tumor specimens revealed the expression of MuSK in the cancer cells from primary mediastinal B cell lymphoma and endometrial carcinoma. Twenty‐one males and 73 females were enrolled. Fifteen cancers occurred in 13 of 94 patients (13.8%). Patients with cancer were significantly older at time of MuSK‐MG onset. Given that people spend on average more than 80% of their time indoors, monitoring indoor air quality (IAQ) is of the utmost importance for the safeguard of human health. Indeed, due to penetration from outdoors and the presence of specific indoor sources, poor IAQ is not uncommon. The use of portable air cleaners (PACs) is one of many options to improve IAQ. The market offers many products, but not all of them have reliable documentation on their effectiveness. Literature standard methods for testing PACs toward pollutants require extensive resources. In this work, we developed a new laboratory scale methodology based on a small, easy to use, economical batch system to test the efficiency of PACs. With this method, three commercial PACs were successfully tested for effectiveness toward volatile organic compounds (VOCs) and particulate matter (PM). Background and Purpose: The oxidant sensor transient receptor potential ankyrin 1 (TRPA1) channel expressed by Schwann cells (SCs) has recently been implicated in several models of neuropathic pain in rodents. Here we investigate whether the pro‐algesic function of Schwann cell TRPA1 is not limited to mammals by exploring the role of TRPA1 in a model of chemotherapy‐induced peripheral neuropathy (CIPN) in zebrafish larvae. Experimental Approach: We used zebrafish larvae and a mouse model to test oxaliplatin‐evoked nociceptive behaviours. We also performed a TRPA1 selective silencing in Schwann cells both in zebrafish larvae and mice to study their contribution in oxaliplatin‐induced CIPN model. Key Results: We found that zebrafish larvae and zebrafish TRPA1 (zTRPA1)‐transfected HEK293T cells respond to reactive oxygen species (ROS) with nociceptive behaviours and intracellular calcium increases, respectively. TRPA1 was found to be co‐expressed with the Schwann cell marker, SOX10, in zebrafish larvae. Oxaliplatin caused nociceptive behaviours in zebrafish larvae that were attenuated by a TRPA1 antagonist and a ROS scavenger. Oxaliplatin failed to produce mechanical allodynia in mice with Schwann cell TRPA1 selective silencing (Plp1+‐Trpa1 mice). Comparable results were observed in zebrafish larvae where TRPA1 selective silencing in Schwann cells, using the specific Schwann cell promoter myelin basic protein (MBP), attenuated oxaliplatin‐evoked nociceptive behaviours. Conclusion and Implications: These results indicate that the contribution of the oxidative stress/Schwann cell/TRPA1 pro‐allodynic pathway to neuropathic pain models seems to be conserved across the animal kingdom. BACKGROUND: The first step in the contamination of leafy vegetables by human pathogens is their attachment to the leaf surface. The success of this is influenced strongly by the physical and chemical characteristics of the surface itself (number and size of stomata, presence of trichomes and veins, epicuticular waxes, hydrophobicity, etc.). This study evaluated the attachment of Salmonella enterica to 30 baby‐leaf salads and tested whether the differences found among them were related to the following leaf traits: hydrophobicity, roughness, and epicuticular waxes. RESULTS: Differences in susceptibility to contamination by S. enterica were found between the 30 baby‐leaf salads investigated. The lowest attachment was found in wild lettuce (Lactuca serriola L.) and lamb's lettuce 'Trophy F1' (Valerianella locusta [L.] Laterr.), with values of 1.63 ± 0.39 Log(CFU/cm2) and 1.79 ± 0.54 Log(CFU/cm2), respectively. Attachment was correlated with hydrophobicity (measured as contact angle) (r = −0.39) and epicuticular waxes (r = −0.81) but not with roughness (r = 0.24). The most important wax components for attachment were alcohols and, in particular, the three‐dimensional (3D) wax crystals of C26 alcohol, but fatty acids probably also had a role. Both these compounds increased hydrophobicity. The presence of thymol, whose antimicrobial properties are well known, was found in lamb's lettuce. CONCLUSIONS: The findings of this study can help to predict and control the attachment and contamination of leafy salads by enterobacteria. They also provide useful information for breeding programs aiming to develop cultivars that are less susceptible to human pathogens, enhancing the food safety of vegetables. © 2024 The Author(s). Despite national and international efforts to prevent non‐indigenous species' introductions, the spread of transboundary plant pests has increased dramatically in recent years, and it seems inevitable that many more species will enter the EU in the future. Identifying plant pests' entry points may offer some early insights to prevent new plant pest invasions and support the surveillance activities carried out in the EU territory. This document was prepared in the context of the EFSA grant GP/EFSA/ENCO/2020/02 and represents the final report of the "HoPPI: Hotspots for plant pests introduction" project. The main objectives of the project were to: i) make an inventory of the pests introduced in the EU in the last two decades; ii) identify hotspots of pests introduction in the EU; iii) identify and analyse the factors that determine their occurrence; iv) understand the role of world trade in affecting risk of introduction using network analysis; v) develop a tool for standardising the pathway model used for the entry step of the quantitative pest risk assessments carried out by EFSA. To meet Objective i, a dataset containing a comprehensive list of pests' first introduction records in the EU between 1999 and 2019, was compiled. The final database includes 278 pest species introduced in the EU, as well as detailed information on the specific species, their origin, and the pathways through which they might have entered the EU. The identification of hotspots and factors in Objective ii and iii was performed using two different methodologies, Getis G* and a Bayesian hierarchical spatial model, that pinpointed specific regions within the EU that are particularly vulnerable to plant pest introductions, uncovering environmental, climatic, and anthropogenic factors contributing to the introduction of pests in specific regions. The application of network analysis in Objective iv sheds light on the intricate connections between international trade routes and the introduction of plant pests into the EU. The results highlight key pathways and trade networks that pose a higher risk of facilitating pest entry. In pursuit of Objective v, an R package named "qPRAentry" was developed. The growing demand for carbon fiber‐reinforced thermoplastic and thermoset (CFRT) composites, mainly in the aerospace, automotive, and energy industries, is due to obtaining lighter components. However, the amount of waste generated during the CFRT production process and at the end of its useful life has been increasing sharply, urgently requiring viable solutions that create less environmental impact in its final disposal. The mechanical recycling process has been presented as an economically and environmentally viable alternative to minimize the environmental impacts generated by the accumulation of this waste in the environment. Therefore, in this study, possible contaminations arising from the grinding process of waste composites reinforced with carbon fiber (CF) based on thermoplastics, polyamide 6 (CF/PA6), low melting poly (aryl ether ketone) (CF/PAEK), poly (phenylene sulfide) (CF/PPS), and fast‐curing epoxy thermoset, are investigated. The composites are ground in a knife mill, resulting in materials measuring (2.6 ± 1) mm. Through differential scanning calorimetry (DSC) analysis, it is observed that the thermal profile of the ground composites is not changed; only the CF/PA6 composite shows an increase of 10% in the value of the degree of crystallinity. X‐ray diffraction (XRD) and scanning electron microscopy (SEM) equipped with an energy‐dispersive X‐ray spectrometer show no metallic contamination from the grinding process. Only low levels of aluminum contamination are observed (below 2.7 wt%), attributed to the comminution step of the waste using an angle grinder adapted to cutting discs with abrasive aluminum oxide grains before the grounding process in the knife mill. The membrane separation process represents an advance in this area, and the use of nanocomposites enables their improvement. Polyamide 6 (PA6) membranes were produced with attapulgite (ATP) and montmorillonite (MMT) clays by melting through the hot pressing of thin films. Subsequently, the membranes were characterized by thermogravimetry (TGA), showing a significant mass loss at 330°C. Differential scanning calorimetry (DSC) revealed that the clay increased thermal stability. X‐ray diffraction (XRD) was performed, confirming the presence of characteristic peaks of polyamide 6, and with the introduction of clay, there were changes in the shapes. Contact angle measurements showed that the clays increased the hydrophilicity of the membranes. Atomic force microscopy (AFM) revealed that the presence of clays modified the surface roughness. The tensile test showed that the clays decreased the strength of the nanocomposites. Oxygen permeation confirmed a decrease in the permeability of compositions containing clay compared with pure PA6 membranes. Studies aimed at understanding the interaction of gases with the membrane material represent a small part of research on the selective barrier. This work aimed to obtain flat membranes from polyamide 6/clay nanocomposites to separate gaseous mixtures using the thin film hot‐press fusion method. Poly(N‐vinylcaprolactam) (PNVCL) is a thermoresponsive polymer, which presents a transition from hydrophilic to hydrophobic behavior close to the physiological temperature, being promising for biomedical applications. However, the production of finished products with PNVCL is difficult due to its high mechanical fragility in the solid state. In the present work, in order to prepare a thermoresponsive film with good mechanical properties, a blend of PNVCL with thermoplastic polyurethane (TPU) was prepared. Three compositions of PNVCL/TPU were studied with mass proportions of 90/10, 70/30, and 50/50, respectively. The films were flexible, miscible and presented different thermoresponsive behaviors, with changing from transparent to opaque varying from 31 to 40°C. Contact angle analysis showed significant changes in the hydrophilicity of the films' surfaces with temperature variation. Moreover, it was possible to induce a change in solid films from transparent to opaque by varying the temperature and verify the reversibility of this change. In addition, tensile tests were carried out, which proved, for PNVCL/TPU blends, the reduction of film brittleness with TPU increase. A helicase‐primase inhibitor, amenamevir (ASP2151), is the active pharmaceutical ingredient of a drug for the herpes zoster that is caused by reactivation of varicella‐zoster virus (VZV). Here we report a new amenamevir‐resistant VZV isolated under the selection pressure of amenamevir. The resistant virus has a nonsynonymous mutation K350N in the helicase gene ORF55. A recombinant virus artificially constructed harboring the ORF55 K350N also acquired amenamevir resistance, and thus the single amino‐acid substitution in helicase is revealed to be responsible for the resistance. We observed that the drug‐resistant virus and the ORF55 K350N recombinant virus have high resistance to amenamevir, as the EC50 values in a plaque reduction assay were > 100 μM, while the two viruses remained susceptible to the nucleoside analog drug acyclovir. No defect in viral growth was observed for these resistant viruses in a plaque size assay in human malignant melanoma cells. However, defect in plaque formation was observed from resistant virus in human fetal lung fibroblast cells, showing that the growth of the resistant virus is dependent on the cell type. We observed that the single amino‐acid substitution in the helicase induces amenamevir resistance, confirming the importance of the helicase in amenamevir's inhibition of virus growth. Our findings highlight the importance of regulating the clinical use of amenamevir to minimize the risk of the emergence of helicase K350N mutation, especially in the long‐term use of amenamevir by immunosuppressed patients. Transanal total mesorectal excision (TaTME) has emerged as a surgical method for treating rectal malignancies in the middle and lower third of the rectum over the past 15 years. This approach was adopted because of the challenges encountered in secure total mesorectal excision (TME). Patient selection criteria and the rationale for TaTME have evolved, leading to improved oncological outcomes and patient quality of life. The procedure includes inserting a unique platform through the anus, forming a purse‐string closure, and endoscopically sealing the lower rectum. The mesorectum is then removed laparoscopically following a 'down‐to‐up' approach, finalised with a transabdominal laparoscopic phase and anastomosis. Pelvic anatomy complexity poses challenges, including potential injuries to the urinary tract, surgical site leakage, sinus damage, sagittal vein harm, nerve injury, carbon dioxide embolism, bowel function disturbance, sphincter mechanism issues, and rectal abscess formation. Proficient anatomy knowledge, precise surgical techniques, and advanced technologies contribute to their prevention. In conclusion, TaTME offers a promising approach to rectal surgery with satisfactory oncological outcomes. However, vigilance is required to eliminate potential complications. [ABSTRACT FROM AUTHOR]

Subnanometer metal clusters show advantages over conventional metal nanoparticles in numerous catalytic reactions owing to their high percentage of exposed surface sites, abundance of under‐coordinated metal sites and unique electronic structures. However, the applications of subnanometer metal clusters in high‐temperature catalytic reactions (>600 °C) are still hindered, because of their low stability under harsh reaction conditions. In this work, we have developed a zeolite‐confined bimetallic PtIn catalyst with exceptionally high stability against sintering. A combination of experimental and theoretical studies shows that the isolated framework In(III) species serve as the anchoring sites for Pt species, precluding the migration and sintering of Pt species in the oxidative atmosphere at ≥650 °C. The catalyst comprising subnanometer PtIn clusters exhibits long‐term stability of >1000 h during a cyclic reaction‐regeneration test for ethane dehydrogenation reaction. [ABSTRACT FROM AUTHOR]

Introduction: This observational study conducted across seven emergency care units compares the efficacy of four D‐dimer detection methods, namely HemosIL D‐dimer HS (HS), HemosIL D‐dimer HS‐500 (HS‐500), VIDAS D‐dimer (VIDAS), and HemosIL AcuStar D‐dimer (ACUSTAR). The primary focus is on patients with a clinical suspicion of deep venous thrombosis (DVT) or pulmonary embolism (PE). Methods: A total of 149 samples were collected from patients with suspected DVT or PE. The confirmation of DVT/PE was based on calf ultrasound or computed tomography‐Angiography. Direct comparisons were made between the different detection methods, considering both their analytical performance and clinical utility. Additionally, the impact of an age‐adjusted cut‐off on the diagnostic accuracy of each method was assessed. Results: The results revealed comparable negative predictive value, sensitivity, and specificity across the methods, with a notable exception of increased specificity for HS compared with HS‐500 (50.8% vs. 41.5%, p = 0.03). Further analysis incorporating an age‐adjusted cut‐off demonstrated a significant improvement in specificity for HS. When using the age‐adjusted cut‐off, HS exhibited a substantial increase in specificity compared with HS‐500 (63.1% vs. 49.2%, p = 0.004) and demonstrated significantly higher specificity compared with VIDAS (63.1% vs. 53.8%, p = 0.04). Conclusion: The study emphasizes the nonuniversal effect of an age‐adjusted cut‐off and discusses the potential necessity for different cut‐off values, particularly in the case of HS‐500. These findings contribute to the understanding of D‐dimer detection methods in the context of DVT and PE, providing insights into their relative performances and the potential optimization through age‐adjusted cut‐offs. Objective: Rare and complex epilepsies encompass a diverse range of disorders characterized by seizures. We aimed to establish a consensus on key issues related to these conditions through collaboration among experienced neurologists, neuropediatricians, and patient advocacy representatives. Methods: Employing a modified Delphi method, a scientific board comprising 20 physicians and 4 patient advocacy representatives synthesized existing literature with their expertise to formulate statements on contentious topics. A final 32‐member expert panel, representing diverse regions of Italy, validated these statements through a two‐round voting process, with consensus defined as an average score ≥7. Results: Sixteen statements reached a consensus, emphasizing the necessity for epidemiological studies to ascertain the true prevalence of rare epilepsies. Etiology emerged as a crucial factor influencing therapeutic strategies and outcome prediction, with particular concern regarding prolonged and tonic–clonic seizures. The importance of early implementation of specific drugs and non‐pharmacological interventions in the treatment algorithm for developmental and epileptic encephalopathies (DEEs) was underscored. Multidisciplinary care involving experts with diverse skills was deemed essential, emphasizing non‐seizure outcomes in adolescence and adulthood. Significance: This national consensus underscores the imperative for personalized, comprehensive, and multidisciplinary management of rare epilepsies/DEEs. It advocates for increased research, particularly in epidemiology and therapeutic approaches, to inform clinical decision‐making and healthcare policies, ultimately enhancing patients' outcomes. Background Objectives Methods Results Conclusions Gestational age significantly influences children's growth and development. Yet, the effect of postterm birth (gestation beyond 42 weeks) on children's growth outcomes remains underexplored.This study aimed to assess the impact of postterm birth on adverse growth outcomes in children using a nationally representative sample from China.A retrospective cohort study was conducted in China from 1 April 2018, to 31 December 2019. The final analysis included 141,002 children aged 3–6 years from 551 cities. Postterm birth was defined as children with postterm birth at a gestational age of 42 weeks or more. Obesity, overweight and thinness were assessed using body mass index‐for‐age (BMI‐for‐age) z‐scores, based on the World Health Organization (WHO) Child Growth Standards. Generalised additive models were employed to investigate the non‐linear relationship between maternal gestational age and BMI‐for‐age z scores. Poisson regression models and subgroup analyses with forest plots were performed to examine the associations between postterm birth and the risks of obesity, overweight and thinness in children.We included 141,002 mother–child pairs, of whom 7314 (5.2%) children were classified as postterm births. There exists a non‐linear relationship between gestational age and BMI‐for‐age z scores. Children born postterm exhibited a 46% increased risk of obesity, a 27% increased risk of combined overweight/obesity and a 13% increased risk of thinness. Similar associations were observed in most cases when further sensitivity and subgroup analysis were conducted.Postterm birth was associated with elevated risks of obesity, overweight and thinness in children aged 3–6 years, independent of sex. Background and Purpose: Fibromyalgia is a complex clinical disorder with an unknown aetiology, characterized by generalized pain and co‐morbid symptoms such as anxiety and depression. An imbalance of oxidants and antioxidants is proposed to play a pivotal role in the pathogenesis of fibromyalgia symptoms. However, the precise mechanisms by which oxidative stress contributes to fibromyalgia‐induced pain remain unclear. The transient receptor potential ankyrin 1 (TRPA1) channel, known as both a pain sensor and an oxidative stress sensor, has been implicated in various painful conditions. Experimental Approach: The feed‐forward mechanism that implicates reactive oxygen species (ROS) driven by TRPA1 was investigated in a reserpine‐induced fibromyalgia model in C57BL/6J mice employing pharmacological interventions and genetic approaches. Key Results: Reserpine‐treated mice developed pain‐like behaviours (mechanical/cold hypersensitivity) and early anxiety‐depressive‐like disorders, accompanied by increased levels of oxidative stress markers in the sciatic nerve tissues. These effects were not observed upon pharmacological blockade or global genetic deletion of the TRPA1 channel and macrophage depletion. Furthermore, we demonstrated that selective silencing of TRPA1 in Schwann cells reduced reserpine‐induced neuroinflammation (NADPH oxidase 1‐dependent ROS generation and macrophage increase in the sciatic nerve) and attenuated fibromyalgia‐like behaviours. Conclusion and Implications: Activated Schwann cells expressing TRPA1 promote an intracellular pathway culminating in the release of ROS and recruitment of macrophages in the mouse sciatic nerve. These cellular and molecular events sustain mechanical and cold hypersensitivity in the reserpine‐evoked fibromyalgia model. Background and Purpose: Magnetic resonance imaging (MRI) is heavily relied upon for the diagnosis and monitoring of multiple sclerosis (MS), a chronic, demyelinating disease of the central nervous system. Serum biomarkers may serve as an accessible tool for increasing sensitivity, improving accessibility, corroborating symptoms, and providing additional data to guide clinical management. This scoping review investigates the current understanding of how the serum biomarker glial fibrillary acidic protein (sGFAP) relates to brain MRI metrics. Methods: We adhered to the Joanna Briggs Institute methodology for scoping reviews and the Preferred Reporting Items for Systematic reviews and Meta‐Analyses guidelines. The databases Medline (Ovid), Embase (Ovid), CINAHL (Ebsco), and Web of Science (University of British Columbia institutional access) were searched on August 24, 2023 using a combination of medical subject headings and keyword terms for the topic of serum biomarkers in MS. Results: A total of 9880 articles were retrieved in total of which 6271 unique titles and abstracts were screened. Twelve of the 259 resultant papers contained sGFAP data and proceeded to extraction. It was found that lesion MRI metrics generally had a positive relationship with sGFAP, while gray matter and white matter metrics, including normal‐appearing white matter, were related negatively or not at all. Conclusions: These results highlight that while sGFAP may not be specific for MS, it may have utility for increasing sensitivity in postdiagnosis monitoring of MS progression. [ABSTRACT FROM AUTHOR]

Cyber‐bullying represents a form of verbal or psychological aggression, carried out intentionally and repeatedly, and perpetuated using digital devices that has negative consequences on the health and well‐being of adolescents. This phenomenon is prevented by contextual and dispositional factors. For this reason, our study investigated both these factors, specifically, the role of the perceived class cohesion in presence of vulnerability factors, that is, low levels of positivity. The results collected on a sample of 689 high school students (Mage = 17.3) suggest that the association between perceived class cohesion and victimisation was moderated by positivity. When young people with low positivity perceived low levels of social cohesion in the group, they experienced more cyberbullying victimisation. However, when high levels of perceived group cohesion were present, these experiences were reduced, regardless of their level of positivity. These results underline the importance of building a positive climate in the class group to prevent cyber‐bullying in adolescence, especially for the most vulnerable individuals. While the conclusions remain contested, these assessments have largely neglected the ecological and conservation impacts of other key regional processes such as declining populations, ageing demographics and rural‐to‐urban migration.These demographic shifts are increasingly prevalent across many regions of the world, and will have significant direct effects on natural resource management and biodiversity conservation by altering individual consumption patterns, land use, land stewardship and natural disturbances. Given that the scientific foundation around this topic is still developing, we first present an initial examination of some of the key environmental impacts, aiming to elevate awareness and encourage further research in these areas.Beyond the ecological implications, declining populations, ageing demographics and rural‐to‐urban migration carry intricate social and cultural consequences that can affect people and nature interactions. Ecological studies that focus on single dimensions of biodiversity or ecosystem responses often overlook these complexities. Demographic changes are likely to be accompanied by shifts in environmental attitudes and connections with nature, all of which will influence our capacity to adapt to or mitigate environmental changes. Finally, environmental policy and practice frameworks are potentially unprepared and their success could be sensitive to these socio‐cultural and demographic shifts.Synthesis and applications: This brief overview demonstrates that population decline, ageing and rural‐to‐urban migration can have extensive implications for biodiversity and the socio‐cultural relationships between people and nature. However, the significance, dynamics and consequences of these processes are still largely overlooked. We believe that these changes warrant specific attention from the research, policy and practice communities, as understanding the outcomes and feedbacks associated with depopulation, ageing populations, loss of culture and tradition and ecological change could aid in designing landscapes and informing management that enhances both human well‐being and biodiversity conservation. The mediator complex subunit 13 (MED13) gene is implicated in neurodevelopmental disorders including autism spectrum disorder (ASD), intellectual disability, and speech delay with varying severity and course. Additional, extra central nervous system, features include eye or vision problems, hypotonia, congenital heart abnormalities, and dysmorphisms. We describe a 7‐year‐ and 4‐month‐old girl evaluated for ASD whose brain magnetic resonance imaging was suggestive of multiple cortical tubers. The exome sequencing (ES ‐ trio analysis) uncovered a unique, de novo, frameshift variant in the MED13 gene (c.4880del, D1627Vfs*17), with a truncating effect on the protein. Concern for the environment has increased the interest of the automotive and aerospace industries in evaluating the feasibility of mechanical recycling of waste carbon‐fiber‐reinforced polymer composites (CFRP) and waste from protective films used during the transport and storage of thermosetting composites, which are removed and discarded during manufacturing. Therefore, this work aimed to develop a recyclable composite from protective film waste by incorporating carbon fiber/poly(aryl ether ketone) (CF/PAEK) composite particles obtained through grinding in knife mills, followed by an analysis of the particle size distribution. Protective film residues (PEres) were cut to facilitate processing. A mixture of low‐density polyethylene and PEres (50/50) was used as the composite matrix, and 5, 10, 15, and 20 wt% CF/PAEK composite particles were added as the reinforcing agents. Then, these recycled composites were prepared by melt mixing using a twin‐screw extruder and injection molding to obtain standardized samples. Mechanical, thermal, electrical, and electromagnetic properties were evaluated. It was possible to observe that the increase in the addition of CF/PAEK composite particles contributed to a 290% increase in elastic modulus values and a 9.1% increase in Shore D hardness, electrical conductivity, and attenuation of the electromagnetic wave. Furthermore, satisfactory adhesion between filler and matrix contributed to a good interface and reinforcement of the final material. Aims: To utilize the estimated glucose disposal rate (eGDR) index of insulin sensitivity, which is based on readily available clinical variables, namely, waist circumference, hypertension and glycated haemoglobin, to discriminate between metabolically healthy and unhealthy phenotypes, and to determine the prevalence of prediabetic conditions. Methods: Non‐diabetic individuals (n = 2201) were stratified into quartiles of insulin sensitivity based on eGDR index. Individuals in the upper quartiles of eGDR were defined as having metabolically healthy normal weight (MHNW), metabolically healthy overweight (MHOW) or metabolically healthy obesity (MHO) according to their body mass index, while those in the lower quartiles were classified as having metabolically unhealthy normal weight (MUNW), metabolically unhealthy overweight (MUOW) and metabolically unhealthy obesity (MUO), respectively. Results: The frequency of impaired fasting glucose (IFG), impaired glucose tolerance (IGT), and IFG + IGT status was comparable among the MHNW, MHOW and MHO groups, while it increased from those with MUNW status towards those with MUOW and MUO status. As compared with participants with MHNW, the odds ratio of having IFG, IGT, or IFG + IGT was significantly higher in participants with MUOW and MUO but not in those with MUNW, MHOW and MHO, respectively. Conclusions: A metabolically healthy phenotype is associated with lower frequency of IFG, IGT, and IFG + IGT status across all body weight categories. [ABSTRACT FROM AUTHOR]

Over the last two decades, ecological and conservation studies on pollinator insects have increased significantly. However, scientific evidence alone is not enough to translate knowledge into policy and into changes in behaviour. To reduce the gap between scientific knowledge on conservation actions and their actual uptake, one should understand the socio‐psychological drivers of people's willingness to undertake these actions.Here, we investigated the socio‐psychological factors influencing individual behaviour in favour of conservation interventions for pollinators in rural versus urban environments across three European countries: Germany, Italy and the Netherlands. We administered an online questionnaire to 4541 respondents stratified by nationality, environment, age and gender.Despite regional differences in socio‐cultural and economic conditions, individuals from Germany, Italy and the Netherlands living in both rural or urban environments shared similar socio‐psychological drivers to protect pollinators. People intended to take action to protect pollinators when they felt morally obliged to, when their social environment supported pollinator protection, when they believed their individual behaviour had an impact, and when they engaged frequently in outdoor activities.Interestingly, specific values held towards pollinators, such as their right to exist, seemed much more important predictors of activating norms that promote conservation actions compared to increasing general environmental concern. In all countries, among the conservation actions, the most likely to be implemented was planting flowers, while one of the most unlikely was participating in monitoring activities.Synthesis and applications. People from three contrasting European countries living in both rural and urban landscapes were mostly driven by the same socio‐psychological factors to help pollinator insects. Therefore, our results offer several practical recommendations to promote pollinator conservation across Europe. First, conservation practitioners interested in pollinator conservation should pay greater attention to values specific to pollinators, as they seemed more important behaviour predictors than general care for the environment. Second, engaging people in conservation efforts can be accomplished by increasing awareness about the vital roles that pollinators play within ecosystems. A flexible and stretchable vitrimer was synthesized using exclusively biobased building blocks and following green chemistry principles. Epoxidized soybean oil (ESO), vanillin, the biobased diamine, Priamine®, and oleic acid were used to create a material that can replace fossil-derived, non-biodegradable packaging. Vanillin and Priamine® were used to develop a crosslinker that could react with the epoxide groups of ESO without the need for solvents, with a catalystfree synthesis that does not generate any waste or byproducts. Oleic acid was incorporated, to tune the properties of the final material, since it can react with ESO and control the crosslink density. The vitrimers produced in this work showed excellent reprocessability and recyclability. They possess the ability to be molded, and they can also strongly adhere on surfaces. Moreover, they exhibited oxygen and water vapor barrier comparable to low density polyethylene, while showing very low migration into food simulant, which position them as a promising material for flexible food and general packaging market. Aim: To examine whether antenatal diagnosis modifies relationships between neonatal brain volumes and 18‐month neurodevelopmental outcomes in children with transposition of the great arteries (TGA). Method: In a retrospective cohort of 139 children with TGA (77 antenatally diagnosed), we obtained total brain volumes (TBVs) on pre‐ (n = 102) and postoperative (n = 112) magnetic resonance imaging. Eighteen‐month neurodevelopmental outcomes were assessed using the Bayley Scales of Infant and Toddler Development, Third Edition. Generalized estimating equations with interaction terms were used to determine whether antenatal diagnosis modified associations between TBVs and neurodevelopmental outcomes accounting for postmenstrual age at scan, brain injury, and ventricular septal defect. Results: Infants with postnatal diagnosis had more preoperative hypotension (35% vs 14%, p = 0.004). The interactions between antenatal diagnosis and TBVs were significantly related to cognitive (p = 0.003) outcomes. Specifically, smaller TBVs were associated with lower cognitive scores in infants diagnosed postnatally; this association was attenuated in those diagnosed antenatally. Interpretation: Antenatal diagnosis modifies associations between neonatal brain volume and 18‐month cognitive outcome in infants with TGA. These findings suggest that antenatal diagnosis may be neuroprotective, possibly through improved preoperative clinical status. These data highlight the need to improve antenatal diagnosis rates. This study introduces a systematic experimental approach aimed at enhancing the production process of poly(urea‐formaldehyde) (PUF) microcapsules filled with 5‐ethylidene‐2‐norbonene (ENB) using in situ emulsion polymerization. Essential parameters were identified through a prior Plackett Burman (PB) design, leading to the implementation of a 24‐1 fractional factorial design. The key parameters selected encompassed stirring speed, poly(ethylene‐alt‐maleic anhydride) (EMA) emulsifier content, pH of the reaction, and the quantity of 1‐octanol. Optimal conditions for polymeric microcapsule synthesis were determined using a statistical analysis of the complete factorial design, facilitating the development of representative mathematical models for the process. A comprehensive evaluation of response variables including microcapsule diameter, yield, encapsulated content, stability, and thermal degradation was conducted. Utilizing response surface analysis (RSM), it was determined that the PUF/ENB(2C) condition (stirring at 700 rpm, a reaction pH of 3.0, and EMA content of 0.425 g) resides within the optimized synthesis region, producing microcapsules exhibiting favorable morphological characteristics, yield of 52% and encapsulated content of 80%. A mini review focused on the chemistry of selenosulfones reported since 2019 is here presented. Initially, methods aimed to the preparation of such moiety are discussed. Successively 1,2‐, 1,3‐, 1,4‐ and 1,7‐addition reactions to C−C single and multiple bonds are illustrated. The forth chapter focuses on electrophilic aromatic substitutions. The fifth chapter deals with C−H functionalizations while the sixth one shows the reactions with aryl diazonium salts or its precursors. The radical coupling with redox active reagents, is the subject of section seventh. Efficient cyclopropanation reactions of aryl or styrylacetonitriles with vinyl selenones under mild conditions were described. By this approach, synthetically challenging 1‐vinylcyclopropane‐1‐carbonitriles were accessible in good yields. The predominant use of synthetic materials, such as fiberglass and polymeric foams, for thermal and acoustic insulation in the construction sector contributes to the recalcitrant waste accumulation in the environment and is not economically sustainable in the long term. This is because they are developed with linear economy standards, they are neither reusable nor recyclable, and, at their end of lifecycle, they are not compostable, with a great amount of them finishing in landfills. This work is focused on the development of natural, self‐growing mycelium‐biocomposites as sustainable alternatives to these conventional synthetic materials. Specifically, fungal mycelium derived from the nonpathogenic fungal strain Pleurotus ostreatus is fed by coffee silverskin flakes, a lignocellulosic agrowaste from roasted coffee seeds, forming 3D biocomposites. The physicochemical properties of the obtained composite are thoroughly investigated, with a final focus on their thermal and acoustic insulation properties. As proved, the natural agrowaste‐mycelium composites possess high porosity and thus low density, good thermal properties, and satisfactory sound absorption capability. Such properties combined with the minimal energetic requirements for their growth and their fully compostable end‐of‐life nature make them valuable alternatives for thermal and acoustic insulation in building construction, among other applications, promoting environmental and economic sustainability. Adrenal hypoplasia congenita, attributed to NR0B1 pathogenic variants, accounts for more than 50% of the incidence of primary adrenal insufficiency in children. Although more than 250 different deleterious variations have been described, no genotype–phenotype correlation has been defined to date. We report a case of an adopted boy who reported the onset of an adrenal crisis at 2 weeks of age, requiring replacement therapy with mineralocorticoids and glucocorticoids for 4 months. For 3 years, he did well without treatment. At almost 4 years of age, the disorder was restarted. A long follow‐up showed the evolution of hypogonadotropic hypogonadism. Molecular studies on NR0B1 revealed a novel and deleterious deletion–insertion–inversion–deletion complex rearrangement sorted in the 5′‐3′ direction, which is described as follows: (1) deletion of the intergenic region (between TASL and NR0B1 genes) and 5′ region, (2) insertion of a sequence containing 37 bp at the junction of the intergenic region of the TASL gene and a part of exon 1 of the NR0B1 gene, (3) inversion of a part of exon 1, (4) deletion of the final portion of exon 1 and exon 2 and beginning of the 3′UTR region, (5) maintenance of part of the intergenic sequence (between genes MAGEB1 and NR0B1, telomeric sense), (6) large posterior deletion, in the same sense. The path to molecular diagnosis was challenging and involved several molecular biology techniques. Evaluating the breakpoints in our patient, we assumed that it was a nonrecurrent rearrangement that had not yet been described. It may involve a repair mechanism known as nonhomologous end‐joining (NHEJ), which joins two ends of DNA in an imprecise manner, generating an "information scar," represented herein by the 37 bp insertion. Objective: To investigate the effects of unilateral upper limbs' (ULM) neuromuscular electrical stimulation (NMES) superimposed on a voluntary contraction added to a protocol of intradialytic leg cycle ergometer exercise on muscle strength, functional capacity and quality of life of adult patients with chronic kidney disease (CKD). Methods: This randomized controlled clinical trial will be carried out at a Brazilian University Hospital. The patients will be evaluated and randomly allocated to an intervention group (i.e., unilateral NMES on the upper limb without hemodialysis fistula for 20 min and leg cycle ergometer for 30 min) or a control group (i.e., unilateral NMES‐Sham on the upper limb without hemodialysis fistula for 20 min and leg cycle ergometer for 30 min). The patients will be treated for 8 weeks, with three weekly treatment sessions totaling 24 sessions. Measurements: ULM muscle strength, functional capacity, quality of life and also the feasibility, safety and patient adherence to the exercise protocol. All physical measurements will be collected by trained researchers before treatment (week 0) and at the end of treatment (week 9), always in the second hemodialysis session of the week. It will be used in an intention‐to‐treat analysis. Results/Conclusions: The outcomes of this clinical trial protocol may help to know the possible benefits of unilateral ULM' NMES superimposed on a voluntary contraction added to a protocol of leg cycle ergometer for patients with CKD and to aid clinical decisions about future implementation or not of this technique (NMES) in intradialytic physical training programs. Objective Methods Results Conclusion Lung cancer is the leading cause of cancer‐related death globally and provides a major disease burden likely to substantially impact quality of life (QoL). Patient‐reported outcome measures (PROMs) have been identified as effective methods of evaluating patient QoL. Existing lung cancer–specific PROMs however have uncertain utility and minimal patient involvement in their design and development. This qualitative study aimed to evaluate the patient perspective of existing PROMs and to explore their appropriateness for population‐based descriptions of lung cancer–related QoL.A descriptive qualitative study was conducted consisting of semi‐structured interviews with 14 patients recruited from the Victorian Lung Cancer Registry and Alfred Hospital using purposive sampling. Interviews first explored the factors most important to lung cancer patients QoL, and second, patient's perspectives on the appropriateness of existing PROMs. Thematic analysis was used to develop themes, and content analysis was conducted to determine PROM acceptability.Five novel themes were identified by patients as being important impacts on QoL: Personal attitude toward the disease is important for coping; independence is valued; relationships with family and friends are important; relationships with treating team are meaningful; personal and public awareness of lung cancer is limited. These patient‐identified impacts are poorly covered in existing lung cancer–specific PROMs. Patients welcomed and appreciated the opportunity to complete PROMs; however, they identified problems with existing PROMs relevance, tone, and formatting.Existing lung cancer PROMs poorly reflect the five themes identified in this study as most important to lung cancer patients QoL. This study reaffirms the need to review existing PROMs to ensure utility and construct validity. Objectives: The purpose of this study was to assess the diagnostic accuracy and the inter‐rater agreement among general dentists when staging and grading periodontitis cases with the aid of a software application (SA) developed by the Italian Society of Periodontology and Implantology. Materials and methods: Ten general dentists were asked to independently assess 25 periodontitis cases using the SA. Accuracy was estimated using quadratic weighted kappa and examiners' percentage of agreement with a reference diagnosis provided by a gold standard examiner. Inter‐rater agreement was evaluated using Fleiss kappa statistics. Results: The overall case definition agreed with the reference diagnosis in 53.6% of cases. The agreements for each general dentist's pairwise comparisons against the reference definition were at least substantial in 100% of cases for stage, in 70% of cases for grade and in none of the cases for extent. Fleiss kappa was 0.818, 0.608, and 0.632 for stage, extent, and grade, respectively. The study recognized possible reasons that could lead to decreased accuracy using the SA. Conclusions: Supported by the SA, general dentists have reached substantial inter‐rater agreement and highly accurate assignments of stage and grade. Phenothiazines (PTZs) are an emerging group of molecules showing effectiveness toward redox signaling and reduction of oxidative injury to cells, via the activation on Kelch‐like ECH‐associated protein 1/nuclear factor erythroid 2‐related factor 2 (Nrf2). Although several electrophilic and indirect Nrf2 activators have been reported, the risk of "off‐target" effect due to the complexity of their molecular mechanisms of action, has aroused research interest toward non‐electrophilic and direct modulators of Nrf2 pathway, such as PTZs. This review represents the first overview on the roles of PTZs as non‐electrophilic Nrf2 activator and free radical scavengers, as well as on their potential therapeutic effects in oxidative stress‐mediated diseases. Here, we provide a collective and comprehensive information on the PTZs ability to scavenge free radicals and activate the Nrf2 signaling pathway, with the aim to broaden the knowledge of their therapeutic potentials and to stimulate innovative research ideas. Atopic dermatitis (AD) is a composite disease presenting disruption of the skin permeability barrier (SPB) in the stratum corneum (SC). Recent evidence supports derangement of the sebaceous gland (SG) activity in the AD pathomechanisms. The objective of this study was to delineate profiles of both sebaceous and epidermal lipids and of aminoacids from SG‐rich (SGR) and SG‐poor (SGP) areas in AD. Both sebum and SC were sampled from SGR areas, while SC was sampled also from SGP areas in 54 adult patients with AD, consisting of 34 and 20 subjects, respectively with and without clinical involvement of face, and in 44 age and sex‐matched controls. Skin biophysics were assessed in all sampling sites. Disruption of the SBP was found to be associated with dysregulated lipidome. Abundance of sapienate and lignocerate, representing, respectively, sebum and the SC type lipids, were decreased in sebum and SC from both SGR and SGP areas. Analogously, squalene was significantly diminished in AD, regardless the site. Extent of lipid derangement in SGR areas was correlated with the AD severity. The abundance of aminoacids in the SC from SGR areas was altered more than that determined in SGP areas. Several gender‐related differences were found in both controls and AD subgroups. In conclusion, the SG activity was differently compromised in adult females and males with AD, in both SGR and SGP areas. In AD, alterations in the aminoacidome profiles were apparent in the SGR areas. Lipid signatures in association with aminoacidome and skin physical properties may serve the definition of phenotype clusters that associate with AD severity and gender. [ABSTRACT FROM AUTHOR]

Aplastic anemia (AA) is a rare bone marrow failure disorder that is treated with either allogeneic stem cell transplant or immunosuppressive therapy (IST) consisting of antithymocyte globulin (ATG), cyclosporine (CSA), and eltrombopag. While outcomes are favorable in younger patients, older patients (>60) have significantly worse long‐term survival. The dose of ATG is often reduced in older patients and those with multiple comorbidities given concerns for tolerability. The efficacy and safety of dose‐attenuated IST in this population is largely undescribed. We performed a retrospective review of patients with AA treated with IST. Our analysis was confounded by changes in practice patterns and the introduction of eltrombopag. We identified 53 patients >60 years old, of which, 20 received dose‐attenuated IST, with no statistically significant difference in overall survival between full and attenuated dose cohorts. Overall response rates in both cohorts were similar at 6 months at 71% and 68%. There were more documented infectious complications in the full dose cohort (13 vs. 3). This supports the consideration of dose‐attenuated IST in older patients with concerns about tolerance of IST. Aim: To determine whether the crustacean Rh1 protein functions as a dual CO2/ammonia transporter and investigate its role in branchial ammonia excretion and acid–base regulation. Methods: Sequence analysis of decapod Rh1 proteins was used to determine the conservation of amino acid residues putatively involved in ammonia transport and CO2 binding in human and bacterial Rh proteins. Using the Carcinus maenas Rh1 protein (CmRh1) as a representative of decapod Rh1 proteins, we test the ammonia and CO2 transport capabilities of CmRh1 through heterologous expression in yeast and Xenopus oocytes coupled with site‐directed mutagenesis. Quantitative PCR was used to assess the distribution of CmRh1 mRNA in various tissues. Western blotting was used to assess CmRh1 protein expression changes in response to high environmental ammonia and CO2. Further, immunohistochemistry was used to assess sub‐cellular localization of CmRh1 and a membrane‐bound carbonic anhydrase (CmCAg). Results: Sequence analysis of decapod Rh proteins revealed high conservation of several amino acid residues putatively involved in conducting ammonia transport and CO2 binding. Expression of CmRh1 in Xenopus oocytes enhanced both ammonia and CO2 transport which was nullified in CmRh1 D180N mutant oocytes. Transport of the ammonia analog methylamine by CmRh1 is dependent on both ionized and un‐ionized ammonia/methylamine species. CmRh1 was co‐localized with CmCAg to the apical membrane of the crustacean gill and only experienced decreased protein expression in the anterior gills when exposed to high environmental ammonia. Conclusion: CmRh1 is the first identified apical transporter‐mediated route for ammonia and CO2 excretion in the crustacean gill. Nickel‐based layered hydroxides (LHs) are a family of efficient electrocatalysts for the alkaline oxygen evolution reaction (OER). Nevertheless, fundamental aspects such as the influence of the crystalline structure and the role of lattice distortion of the catalytic sites remain poorly understood and typically muddled. Herein, we carried out a comprehensive investigation on ɑ‐LH, β‐LH and layered double hydroxide (LDH) phases by means of structural, spectroscopical, in‐silico and electrochemical studies, which suggest the key aspect exerted by Ni‐vacancies in the ɑ‐LH structure. Density functional theory (DFT) calculations and X‐ray absorption spectroscopy (XAS) confirm that the presence of Ni‐vacancies produces acute distortions of the electroactive Ni sites (reflected as the shortening of the Ni−O distances and changes in the O−Ni−O angles), triggering the appearance of Ni localised electronic states on the Fermi level, reducing the Egap, and consequently, increasing the reactivity of the electroactive sites in the ɑ‐LH structure. Furthermore, post‐mortem Raman and XAS measurements unveil its transformation into a highly reactive oxyhydroxide‐like phase that remains stable under ambient conditions. Hence, this work pinpoints the critical role of the crystalline structure as well as the electronic properties of LH structures on their inherent electrochemical reactivity towards OER catalysis. We envision Ni‐based ɑ‐LH as a perfect platform for hosting trivalent cations, closing the gap toward the next generation of benchmark efficient earth‐abundant electrocatalysts. Introduction: Adverse reactions are relatively common during peanut oral immunotherapy. To reduce the risk to the patient, some researchers have proposed modifying the allergen to reduce IgE reactivity, creating a putative hypoallergen. Analysis of recently cloned human IgG from patients treated with peanut immunotherapy suggested that there are three common conformational epitopes for the major peanut allergen Ara h 2. We sought to test if structural information on these epitopes could indicate mutagenesis targets for designing a hypoallergen and evaluated the reduction in IgE binding via immunochemistry and a mouse model of passive cutaneous anaphylaxis (PCA). Methods: X‐ray crystallography characterized the conformational epitopes in detail, followed by mutational analysis of key residues to modify monoclonal antibody (mAb) and serum IgE binding, assessed by ELISA and biolayer interferometry. A designed Ara h 2 hypoallergen was tested for reduced vascularization in mouse PCA experiments using pooled peanut allergic patient serum. Results: A ternary crystal structure of Ara h 2 in complex with patient antibodies 13T1 and 13T5 was determined. Site‐specific mutants were designed that reduced 13T1, 13T5, and 22S1 mAbs binding by orders of magnitude. By combining designed mutations from the three major conformational bins, a hexamutant (Ara h 2 E46R, E89R, E97R, E114R, Q146A, R147E) was created that reduced IgE binding in serum from allergic patients. Further, in the PCA model where mice were primed with peanut allergic patient serum, reactivity upon allergen challenge was significantly decreased using the hexamutant. Conclusion: These studies demonstrate that prior knowledge of common conformational epitopes can be used to engineer reduced IgE reactivity, an important first step in hypoallergen design. Super‐resolution strategies expand the applicability of imaging modalities toward previously inaccessible spatial scales. Herein, photoactivated far‐infrared thermography is pushed from the millimeter to the micrometer scale by a 2D super‐resolution imaging approach capable of tackling the resolution barriers imposed by both diffraction‐limited signal collection and lateral spatiotemporal heat diffusion. The proposed imaging strategy relies on a full‐wave forward model of far‐infrared thermography and on an image‐inversion approach, which reconstructs the surface distribution of the sample photothermal absorbers via gradient‐descent optimization as the one yielding the best match between predicted and experimental images. With minute‐long acquisitions on a commercial low‐cost far‐infrared camera, less‐than‐5 μm spatial resolution is demonstrated on both synthetic samples and human liver biopsies ex vivo, thereby exemplifying the applicability of super‐resolution photothermal imaging to the fast nondestructive characterization of biological specimens well below the tissue spatial scale. Background: The purpose of this study was to evaluate the reliability and accuracy in the assignment of the case definitions of peri‐implant health and diseases according to the 2018 Classification of Periodontal and Peri‐implant Diseases and Conditions. Methods: Ten undergraduate students, 10 general dentists, and 10 experts in implant dentistry participated in this study. All examiners were provided with clinical and radiographic documentation of 25 dental implants. Eleven out the 25 cases were also accompanied by baseline readings. Examiners were asked to define all cases using the 2018 classification case definitions. Reliability among examiners was evaluated using the Fleiss kappa statistic. Accuracy was estimated using percentage of complete agreement and quadratic weighted kappa for pairwise comparisons between each rater and a gold standard diagnosis. Results: The Fleiss kappa was 0.50 (95% CI: 0.48 to 0.51) and the mean quadratic weighted kappa value was 0.544. Complete agreement with the gold standard diagnosis was achieved in 59.8% of the cases. Expertise in implantology affected accuracy positively (p < 0.001) while the absence of baseline readings affected it negatively (p < 0.001). Conclusion: Both reliability and accuracy in assigning case definitions to dental implants according to the 2018 classification were mostly moderate. Late gestational supine positioning reduces maternal cardiac output due to inferior vena caval (IVC) compression, despite increased collateral venous return. However, little is known about the impact of maternal position on oxygen (O2) delivery and consumption of the gravid uterus, fetus, placenta and lower limbs. We studied the effects of maternal positioning on these parameters in 20 healthy pregnant subjects at 36 ± 2 weeks using magnetic resonance imaging (MRI); a follow‐up MRI was performed 6‐months postpartum (n = 16/20). MRI techniques included phase‐contrast and T1/T2 relaxometry for blood flow and oximetry imaging, respectively. O2 transport was measured in the following vessels (bilateral where appropriate): maternal abdominal descending aorta (DAoabdo), IVC, ovarian, paraspinal veins (PSV), uterine artery (UtA) and external iliacs, and umbilical. Maternal cardiac output was measured by summing DAothoracic and superior vena cava flows. Supine mothers (n = 6) had lower cardiac output and O2 delivery in the DAoabdo, UtA and external iliac arteries, and higher PSV flow than those in either the left (n = 8) or right (n = 6) lateral positions during MRI. However, O2 consumption in the gravid uterus, fetus, placenta and lower limbs was unaffected by maternal positioning. The ratio of IVC/PSV flow decreased in supine mothers while ovarian venous flow and O2 saturation were unaltered, suggesting a major route of pelvic venous return unaffected by maternal position. Placental–fetal O2 transport and consumption were similar between left and right lateral maternal positions. In comparison to non‐pregnant findings, DAoabdo and UtA O2 delivery and pelvic O2 consumption increased, while lower‐limb consumption remained constant , despite reduced external iliac artery O2 delivery in late gestation. Key points: Though sleeping supine during the third trimester is associated with an increased risk of antepartum stillbirth, the underlying biological mechanisms are not fully understood.Maternal cardiac output and uteroplacental flow are reduced in supine mothers due to inferior vena caval compression from the weight of the gravid uterus.This MRI study provides a comprehensive circulatory assessment, demonstrating reduced maternal cardiac output and O2 delivery (uteroplacental, lower body) in supine compared to lateral positioning; however, O2 consumption (gravid uterus, fetus, placenta, lower limbs) was preserved.Unlike other mammalian species, the ovarian veins conduct substantial venous return from the human pregnant uterus that is unaffected by maternal positioning. Lumbar paraspinal venous flow increased in supine mothers. These observations may have important considerations during major pelvic surgery in pregnancy (i.e. placenta percreta).Future studies should address the importance of maternal positioning as a potential tool to deliver improved perinatal outcomes in pregnancies with compromised uteroplacental O2 delivery. Background and purpose: Different algorithms aiming to identify individuals at risk of Parkinson disease (PD) have been proposed. Comparative studies of these scores and their recent updates in the general elder population are needed. Methods: We have previously applied the "basic" PREDICT‐PD algorithm, designed for remote screening, and the original and updated Movement Disorder Society (MDS) criteria for prodromal PD to the longitudinal population‐based Bruneck study cohort. We have now additionally employed the "enhanced" PREDICT‐PD algorithm (which includes motor assessment, olfaction, probable rapid eye movement sleep behaviour disorder status, pesticide exposure, and diabetes as additional factors). Risk scores were calculated based on comprehensive baseline assessments (2005) in 574 subjects aged 55–94 years (290 females), and cases of incident PD were identified at 5‐year (n = 11) and 10‐year follow‐up (n = 9). We analysed the association of the different log‐transformed risk scores with incident PD at follow‐up (calculated per 1‐SD unit change). Results: The enhanced PREDICT‐PD algorithm was associated with incident PD over 10‐years of follow‐up, yielding higher odds for incident PD (odds ratio [OR] = 4.61, 95% confidence interval [CI] = 2.68–7.93, p < 0.001) compared with the basic PREDICT‐PD score (OR = 2.38, 95% CI = 1.49–3.79, p < 0.001). The updated MDS prodromal criteria yielded a numerically higher OR of 7.13 (95% CI = 3.49–14.54, p < 0.001) in comparison with the original criteria as well as the enhanced PREDICT‐PD algorithm, with overlapping 95% CIs. Conclusions: The enhanced PREDICT‐PD algorithm was significantly associated with incident PD. The consistent performance of both the enhanced PREDICT‐PD algorithm and the updated MDS prodromal criteria compared to their original versions supports their use in PD risk screening.
