Herbivore exclusion is implemented globally to recover ecosystems from grazing by introduced and native herbivores, but evidence for large‐scale biodiversity benefits is inconsistent in arid ecosystems. We examined the effects of livestock exclusion on dryland plant richness and reproductive capacity. We collected data on plant species richness and seeding (reproductive capacity), rainfall, vegetation productivity and cover, soil strength and herbivore grazing intensity from 68 sites across 6500 km2 of arid Georgina gidgee (Acacia georginae) woodlands in central Australia between 2018 and 2020. Sites were on an actively grazed cattle station and two destocked conservation reserves. We used structural equation modeling to examine indirect (via soil or vegetation modification) versus direct (herbivory) effects of grazing intensity by two introduced herbivores (cattle, camels) and a native herbivore (red kangaroo), on seasonal plant species richness and seeding of all plants, and the richness and seeding of four plant groups (native grasses, forbs, annual chenopod shrubs, and palatable perennial shrubs). Non‐native herbivores had a strong indirect effect on plant richness and seeding by reducing vegetative ground cover, resulting in decreased richness and seeding of native grasses and forbs. Herbivores also had small but negative direct impacts on plant richness and seeding. This direct effect was explained by reductions in annual chenopod and palatable perennial shrub richness under grazing activity. Responses to grazing were herbivore‐dependent; introduced herbivore grazing reduced native plant richness and seeding, while native herbivore grazing had no significant effect on richness or seeding of different plant functional groups. Soil strength decreased under grazing by cattle but not camels or kangaroos. Cattle had direct effects on palatable perennial shrub richness and seeding, whereas camels had indirect effects, reducing richness and seeding by reducing the abundance of shrubs. We show that considering indirect pathways improves evaluations of the effects of disturbances on biodiversity, as focusing only on direct effects can mask critical mechanisms of change. Our results indicate substantial biodiversity benefits from excluding livestock and controlling camels in drylands. Understanding the relative strengths of intrinsic and extrinsic factors regulating populations is a long‐standing focus of ecology and critical to advancing conservation programs for imperiled species. Conservation could benefit from an increased understanding of factors influencing vital rates (somatic growth, recruitment, survival) in small, translocated populations, which is lacking owing to difficulties in long‐term monitoring of rare species. Translocations, here defined as the transfer of wild‐captured individuals from source populations to new habitats, are widely used for species conservation, but outcomes are often minimally monitored, and translocations that are monitored often fail. To improve our understanding of how translocated populations respond to environmental variation, we developed and tested hypotheses related to intrinsic (density dependent) and extrinsic (introduced rainbow trout Oncorhynchus mykiss, stream flow and temperature regime) causes of vital rate variation in endangered humpback chub (Gila cypha) populations translocated to Colorado River tributaries in the Grand Canyon (GC), USA. Using biannual recapture data from translocated populations over 10 years, we tested hypotheses related to seasonal somatic growth, and recruitment and population growth rates with linear mixed‐effects models and temporal symmetry mark–recapture models. We combined data from recaptures and resights of dispersed fish (both physical captures and continuously recorded antenna detections) from throughout GC to test survival hypotheses, while accounting for site fidelity, using joint live‐recapture/live‐resight models. While recruitment only occurred in one site, which also drove population growth (relative to survival), evidence supported hypotheses related to density dependence in growth, survival, and recruitment, and somatic growth and recruitment were further limited by introduced trout. Mixed‐effects models explained between 67% and 86% of the variation in somatic growth, which showed increased growth rates with greater flood‐pulse frequency during monsoon season. Monthly survival was 0.56–0.99 and 0.80–0.99 in the two populations, with lower survival during periods of higher intraspecific abundance and low flood frequency. Our results suggest translocations can contribute toward the recovery of large‐river fishes, but continued suppression of invasive fishes to enhance recruitment may be required to ensure population resilience. Background: Rare loss‐of‐function variants in TREM2 (Triggering Receptor Expressed in Myeloid cells 2) are associated with an increased risk of AD. There are ongoing efforts to therapeutically boost microglial neuroprotective functions by targeting TREM2. However, the intracellular mechanisms underlying these functions in AD are not fully understood. TREM2 signaling maintains metabolic fitness through activation of the mechanistic target of rapamycin (mTOR) allowing microglia to sustain their metabolism during amyloidosis (Ulland et al., 2017). Detrimental decreases in microglial mTOR signalling during amyloidosis independent of TREM2 loss‐of‐function has also been reported (Baik et al., 2019). The mTOR pathway acts upstream of the eukaryotic mRNA translation by suppressing the translation repressor, eukaryotic translation initiation factor 4E (eIF4E)‐binding proteins (4E‐BPs). mTOR inhibits the 4E‐BPs via phosphorylation, thereby releasing eIF4E and allowing mRNA translation to initiate (Hay and Sonenberg, 2004). Work from our group demonstrated that mTOR boosts mitochondrial metabolism through 4E‐BP‐controlled mRNA translation (Morita et al., 2013). Therefore, we aimed to understand the dysregulated mechanisms at the intersection of mTOR‐controlled mRNA translation and metabolism in microglia response to Amyloid‐β (Aß). Method: We used immunoblotting to investigate the effect on 4E‐BP phosphorylation following chronic exposure to Aß aggregates and TREM2 signaling inhibition in a microglia cell line. We manipulated the mTOR pathway by knocking out the downstream effectors, 4E‐BPs, to promote the translation of 4E‐BP‐controlled mRNAs in microglia in vitro and in vivo. We crossed the microglia‐specific 4E‐BPs knock out mouse with a RiboTag mouse to pull‐down ribosome‐bound mRNAs, providing a genome‐wide pool of actively translating mRNAs (the translatome) in the absence of 4E‐BPs in microglia. Result: We showed that 4E‐BP‐controlled mRNA translation is inhibited upon chronic exposure to Aß or TREM2 signaling inhibition. The deletion of 4E‐BPs in microglia in‐vitro leads to a decrease in expression of pro‐inflammatory mediators and microglia cell death, and an increase in reliance on oxidative phosphorylation upon exposure to Aß. We also observed that 4E‐BP deletion changes the microglia translatome and decreases expression of inflammatory mediators in vivo after an intracranial Aß infusion. Continental‐ and regional‐scale assessments of gaps in protected area networks typically use relatively coarse range maps for well documented species groups, creating uncertainty about the fate of unexamined biodiversity and providing insufficient guidance for land managers. By building habitat suitability models for a taxonomically diverse group of 2216 imperiled plants and animals, we revealed comprehensive and detailed protection opportunities in the conterminous United States. Summing protection‐weighted range‐size rarity (PWRSR, the product of the percent of modeled habitat outside of protected areas and the inverse of modeled habitat extent) uncovered novel patterns of biodiversity importance. Concentrations of unprotected imperiled species in places such as the northern Sierra Nevada, central and northern Arizona, the Rocky Mountains of Utah and Colorado, southeastern Texas, southwestern Arkansas, and Florida's Lake Wales Ridge have rarely if ever been featured in continental‐ and regional‐scale analyses. Inclusion of diverse taxa (vertebrates, freshwater mussels, crayfishes, bumble bees, butterflies, skippers, and vascular plants) partially drove these new patterns. When analyses were restricted to groups typically included in previous studies (birds, mammals, and amphibians), up to 53% of imperiled species in other groups were left out. The finer resolution of modeled inputs (990 m) also resulted in a more geographically dispersed pattern. For example, 90% of the human population of the conterminous United States lives within 50 km of modeled habitat for one or more species with high PWRSR scores. Over one‐half of the habitat for 818 species occurs within federally lands managed for biodiversity protection; an additional 360 species have over one‐half of their modeled habitat on federal multiple use land. Freshwater animals occur in places with poorer landscape condition but with less exposure to climate change than other groups, suggesting that habitat restoration is an important conservation strategy for these species. The results provide fine‐scale, taxonomically diverse inputs for local and regional priority‐setting and show that although protection efforts are still widely needed on private lands, notable gains can be achieved by increasing protection status on selected federal lands. Neuroinflammation contributes to Alzheimer's disease (AD) progression. Secondary inflammatory insults trigger delirium and can accelerate cognitive decline. Individual cellular contributors to this vulnerability require elucidation. Using APP/PS1 mice and AD brain, we studied secondary inflammatory insults to investigate hypersensitive responses in microglia, astrocytes, neurons, and human brain tissue. The NLRP3 inflammasome was assembled surrounding amyloid beta, and microglia were primed, facilitating exaggerated interleukin‐1β (IL‐1β) responses to subsequent LPS stimulation. Astrocytes were primed to produce exaggerated chemokine responses to intrahippocampal IL‐1β. Systemic LPS triggered microglial IL‐1β, astrocytic chemokines, IL‐6, and acute cognitive dysfunction, whereas IL‐1β disrupted hippocampal gamma rhythm, all selectively in APP/PS1 mice. Brains from AD patients with infection showed elevated IL‐1β and IL‐6 levels. Therefore, amyloid leaves the brain vulnerable to secondary inflammation at microglial, astrocytic, neuronal, and cognitive levels, and infection amplifies neuroinflammatory cytokine synthesis in humans. Exacerbation of neuroinflammation to produce deleterious outcomes like delirium and accelerated disease progression merits careful investigation in humans. [ABSTRACT FROM AUTHOR]

Junctional ectopic tachycardia (JET) is a tachyarrhythmia arising from the atrioventricular node and His bundle area. Enhanced normal automaticity has been postulated as the mechanism of JET in the majority of patients. It is more common in children and can be seen as congenital or in postoperative settings. It is often a narrow complex tachycardia but can present as a wide complex tachycardia as a result of aberrant conduction. Its differentiation from other arrhythmias especially atrioventricular nodal reentrant tachycardia (AVNRT) can be challenging. Medical treatment of JET is difficult, and catheter ablation remains the mainstay of treatment in refractory cases with a high risk of atrioventricular block and recurrence. [ABSTRACT FROM AUTHOR]

Objective: Although fetal growth restriction (FGR) is well known to be associated with adverse outcomes for the mother and offspring, effective interventions for the management of FGR are yet to be established. Trials reporting interventions for the prevention and treatment of FGR may be limited by heterogeneity in the underlying pathophysiology. The aim of this study was to conduct a systematic review of outcomes reported in randomized controlled trials (RCTs) assessing interventions for the prevention or treatment of FGR, in order to identify and categorize the variation in outcome reporting.Methods: MEDLINE, EMBASE and The Cochrane Library were searched from inception until August 2018 for RCTs investigating therapies for the prevention and treatment of FGR. Studies were assessed systematically and data on outcomes that were reported in the included studies were extracted and categorized. The methodological quality of the included studies was assessed using the Jadad score.Results: The search identified 2609 citations, of which 153 were selected for full-text review and 72 studies (68 trials) were included in the final analysis. There were 44 trials relating to the prevention of FGR and 24 trials investigating interventions for the treatment of FGR. The mean Jadad score of all studies was 3.07, and only nine of them received a score of 5. We identified 238 outcomes across the included studies. The most commonly reported were birth weight (88.2%), gestational age at birth (72.1%) and small-for-gestational age (67.6%). Few studies reported on any measure of neonatal morbidity (27.9%), while adverse effects of the interventions were reported in only 17.6% of trials.Conclusions: There is significant variation in outcome reporting across RCTs of therapies for the prevention and treatment of FGR. Here, we discuss the case of a man with a history of ischemic cardiomyopathy and cardiac resynchronization therapy defibrillator implantation, who presented to emergency department with decompensated heart failure due to the loss of resynchronization therapy.
