Alternative water uptake pathways through leaves and bark complement water supply with interception, fog or dew. Bark water-uptake contributes to embolism-repair, as demonstrated in cut branches. We tested whether bark water-uptake could also contribute to supplement xylem-water for transpiration. We applied bandages injected with 2H-enriched water on intact upper-canopy branches of Pinus sylvestris and Fagus sylvatica in a boreal and in a temperate forest, in summer and winter, and monitored transpiration and online isotopic composition (δ2H and δ18O) of water vapour, before sampling for analyses of δ2H and δ18O in tissue waters. Xylem, bark and leaf waters from segments downstream from the bandages were 2H-enriched whereas δ18O was similar to controls. Transpiration was positively correlated with 2H-enrichment. Isotopic compositions of transpiration and xylem water allowed us to calculate isotopic exchange through the bark via vapour exchange, which was negligible in comparison to estimated bark water-uptake, suggesting that water-uptake occurred via liquid phase. Results were consistent across species, forests and seasons, indicating that bark water-uptake may be more ubiquitous than previously considered. We suggest that water taken up through the bark could be incorporated into the transpiration stream, which could imply that sap-flow measurements underestimate transpiration when bark is wet. © 2022 John Wiley & Sons Ltd. Water inside plants forms a continuous chain from water in soils to the water evaporating from leaf surfaces. Failures in this chain result in reduced transpiration and photosynthesis and are caused by soil drying and/or cavitation-induced xylem embolism. Xylem embolism and plant hydraulic failure share several analogies to 'catastrophe theory' in dynamical systems. These catastrophes are often represented in the physiological and ecological literature as tipping points when control variables exogenous (e.g., soil water potential) or endogenous (e.g., leaf water potential) to the plant are allowed to vary on time scales much longer than time scales associated with cavitation events. Here, plant hydraulics viewed from the perspective of catastrophes at multiple spatial scales is considered with attention to bubble expansion within a xylem conduit, organ-scale vulnerability to embolism, and whole-plant biomass as a proxy for transpiration and hydraulic function. The hydraulic safety-efficiency tradeoff, hydraulic segmentation and maximum plant transpiration are examined using this framework. Underlying mechanisms for hydraulic failure at fine scales such as pit membranes and cell-wall mechanics, intermediate scales such as xylem network properties and at larger scales such as soil-tree hydraulic pathways are discussed. Understudied areas in plant hydraulics are also flagged where progress is urgently needed.

Heat waves occurring during droughts can have a devastating impact on yield, especially if they happen during the flowering and seed set stages of the crop cycle. Global warming and climate change are driving an alarming increase in the frequency and intensity of combined drought and heat stress episodes, critically threatening global food security. Because high temperature is detrimental to reproductive processes, essential for plant yield, we measured the inner temperature, transpiration, sepal stomatal aperture, hormone concentrations and transcriptomic response of closed soybean flowers developing on plants subjected to a combination of drought and heat stress. Here, we report that, during a combination of drought and heat stress, soybean plants prioritize transpiration through flowers over transpiration through leaves by opening their flower stomata, while keeping their leaf stomata closed. This acclimation strategy, termed ‘differential transpiration’, lowers flower inner temperature by about 2–3°C, protecting reproductive processes at the expense of vegetative tissues. Manipulating stomatal regulation, stomatal size and/or stomatal density of flowers could serve as a viable strategy to enhance the yield of different crops and mitigate some of the current and future impacts of global warming and climate change on agriculture.

Crop wild relatives, the closely related species of crops, may harbour potentially important sources of new allelic diversity for (a)biotic tolerance or resistance. However, to date, wild diversity is only poorly characterized and evaluated. Banana has a large wild diversity but only a narrow proportion is currently used in breeding programmes. The main objective of this study was to evaluate genotype-dependent transpiration responses in relation to the environment. By applying continuous high-throughput phenotyping, we were able to construct genotype-specific transpiration response models in relation to light, VPD and soil water potential. We characterized and evaluated six (sub)species and discerned four phenotypic clusters. Significant differences were observed in leaf area, cumulative transpiration and transpiration efficiency. We confirmed a general stomatal-driven ‘isohydric’ drought avoidance behaviour, but discovered genotypic differences in the onset and intensity of stomatal closure. We pinpointed crucial genotype-specific soil water potentials when drought avoidance mechanisms were initiated and when stress kicked in. Differences between (sub)species were dependent on environmental conditions, illustrating the need for high-throughput dynamic phenotyping, modelling and validation. Stomata play a central role in surface-atmosphere exchange by controlling the flux of water and CO

Increasing stomata movement is beneficial to improve plant water use efficiency and drought resilience. Contradictory results indicate that aquaporins might regulate stomatal movement. Here, we tested whether the maize plasma membrane PIP2;5 aquaporin affects stomatal closure under water deficit, abscisic acid (ABA) or vapor pressure deficit (VPD) treatment in intact plants, detached leaves, or peeled epidermis. Transpiration, stomatal conductance (gs) and aperture, and reactive oxygen species (ROS) in stomatal complexes were studied in maize lines with increased or knocked down (KD) PIP2;5 gene expression. In well-watered conditions, the PIP2;5 overexpressing (OE) plants transpired more than wild types (WT), while no significant difference in transpiration was observed between pip2;5 KD and WT. Upon mild-water deficit or low ABA concentration treatments, transpiration and gs decreased more in PIP2;5 OE, and less in pip2;5 KD lines, in comparison with WTs. In detached epidermis, ABA treatment induced faster stomatal closing in PIP2;5 OE lines compared to WT, while pip2;5 KD stomata were ABA insensitive. These phenotypes were associated with guard cell ROS accumulation. Additionally, PIP2;5 is involved in the transpiration decrease observed under high VPD. Soil-leaf hydraulic conductance determines canopy-atmosphere coupling in vegetation models, but is typically derived from ex-situ measurements of stem segments and soil samples. Using a novel approach we derive robust in-situ estimates for whole tree conductance (ktree ), 'functional' soil conductance (ksoil ), and 'system' conductance (ksystem , water table to canopy), at two climatically different tropical rainforest sites. Hydraulic 'functional rooting depth', determined for each tree using profiles of soil water potential (Ψsoil ) and sap flux data, enabled a robust determination of ktree and ksoil . ktree was compared across species, size classes, seasons, height above nearest drainage (HAND), two field sites, and to alternative representations of ktree ; ksoil was analysed with respect to variations in site, season and HAND. Key results: i) ktree was lower and changed seasonally at the site with higher vapour pressure deficit (VPD) and rainfall, ii) ktree differed little across species but scaled with tree circumference, iii) rsoil (1/ksoil ) ranged from 0 in wet season to 10x less than rtree (1/ktree ) in the dry season. Conclusions VPD and not rainfall may influence plot-level k; leaf water potentials and sap flux can be used to determine ktree , ksoil and ksystem ; Ψsoil profiles can provide mechanistic insights into ecosystem-level water fluxes.

Interactions between climate change and UV penetration in the biosphere are resulting in the exposure of plants to new combinations of UV radiation and drought. In theory, the impacts of combinations of UV and drought may be additive, synergistic or antagonistic. Lack of understanding of the impacts of combined treatments creates substantial uncertainties that hamper predictions of future ecological change. Here, we compiled information from 52 publications and analysed the relative impacts of UV and/or drought. Both UV and drought have substantial negative effects on biomass accumulation, plant height, photosynthesis, leaf area and stomatal conductance and transpiration, while increasing stress-associated symptoms such as MDA accumulation and reactive-oxygen-species content. Contents of proline, flavonoids, antioxidants and anthocyanins, associated with plant acclimation, are upregulated both under enhanced UV and drought. In plants exposed to both UV and drought, increases in plant defense responses are less-than-additive, and so are the damage and growth retardation. Less-than-additive effects were observed across field, glasshouse and growth-chamber studies, indicating similar physiological response mechanisms. Induction of a degree of cross-resistance seems the most likely interpretation of the observed less-than-additive responses. The data show that in future climates, the impacts of increases in drought exposure may be lessened by naturally high UV regimes.

Reliable partitioning of micrometeorologically measured evapotranspiration (ET) into evaporation (E) and transpiration (T) would greatly enhance our understanding of the water cycle and its response to climate change. While some methods on ET partitioning have been developed, their underlying assumptions make them difficult to apply more generally, especially in sites with large contributions of E. Here, we report a novel ET partitioning method using Artificial Neural Networks (ANN) in combination with a range of environmental input variables to predict daytime E from nighttime ET measurements. The study uses eddy covariance data from four restored wetlands in the Sacramento-San Joaquin Delta, California, USA, as well as leaf-level T data for validation. The four wetlands vary in structure from some with large areas of open water and little vegetation to very densely vegetated wetlands, representing a range of ET conditions. The ANNs were built with increasing complexity by adding the input variable that resulted in the next highest average value of model testing R2across all sites. The order of variable inclusion (and importance) was: vapor pressure deficit (VPD) > gap-filled sensible heat flux (H_gf) > air temperature (Tair) > friction velocity (u∗) > other variables. Overall, 36 ANNs were analyzed. The model using VPD, H_gf, Tair, and u∗(F11), showed an average testing R2value across all sites of 0.853. In comparison with the model that included all 10 variables (F36), F11 generally performed better during validation with independent data. In comparison to other methods described in the literature, the ANN method generated more consistent T/ET partitioning results especially for more complex sites with large E contributions. Our method improves the understanding of T/ET partitioning. While it may be particularly suited to flooded ecosystems, it can also improve T/ET partitioning in other systems, increasing our knowledge of the global water cycle.

Temperature (T) and vapour pressure deficit (VPD) are important drivers of plant hydraulic conductivity, growth, mortality, and ecosystem productivity, independently of soil water availability. Our goal was to disentangle the effects of T and VPD on plant hydraulic responses. Young trees of Fagus sylvatica L., Quercus pubescens Willd. and Quercus ilex L. were exposed to a cross-combination of a T and VPD manipulation under unlimited soil water availability. Stem hydraulic conductivity and leaf-level hydraulic traits (e.g., gas exchange and osmotic adjustment) were tracked over a full growing season. Significant loss of xylem conductive area (PLA) was found in F. sylvatica and Q. pubescens due to rising VPD and T, but not in Q. ilex. Increasing T aggravated the effects of high VPD in F. sylvatica only. PLA was driven by maximum hydraulic conductivity and minimum leaf conductance, suggesting that high transpiration and water loss after stomatal closure contributed to plant hydraulic stress. Many woody plants produce large floral displays early in the spring when xylem transport can be variable and often reduced. To determine whether stem hydraulics impact floral water use, we quantified floral transpiration and tested whether it was correlated with stem xylem conductivity in five temperate woody species that flower before producing leaves. We measured inflorescence gas exchange, examined the relationship between diffusive conductance and inflorescence morphology, and estimated the amount of water supplied to an inflorescence by the phloem. We also tested for correlation between transpiration and native stem xylem conductivity for branches with leaves and branches with flowers. The flowers of our study species obtain most of their water from the xylem. Diffusive conductance was higher in small inflorescences, but water content and daily transpiration rates were greater for larger inflorescences. We found no correlation between floral transpiration per branch and stem xylem conductivity within species. The data suggest that inflorescence water loss during anthesis is not limited by the xylem in our study species. We highlight the impact of floral morphology on hydraulic traits and encourage exploration into temporal shifts in floral hydration.

The plasma membrane subfamily of aquaporins [plasma membrane intrinsic proteins (PIPs)], which facilitates the CO2 diffusion across membranes, is proposed to play an important role in mesophyll conductance to CO2 (gm ), a major limiting factor of photosynthesis. However, recent studies implied no causal relationship between gm and PIPs because they failed to repeat the previous observed differences in gm between PIP knockout lines and their wild-type. The contrasting results on the role of PIPs in gm were interpreted as the different growth conditions among studies, which has never been tested. Here, we assessed whether the differences in gm between wild-type and PIP knockout lines, Ospip1;1, Ospip1;2 and Ospip2;1, varied with growth condition (field versus pot condition) and growth stages in rice. Under field conditions, no differences were observed in plant performance, photosynthetic rate (A) and gm between PIP knockout lines and the wild-type. However, in agreement with previous studies, two out of three knockout lines showed significant declines in tiller number, plant height, A and gm under pot conditions. Moreover, we found that the differences in A and gm between PIP knockout lines and the wild-type varied with the growth stage of the plants. Our results showed that the differences in gm between PIP knockout lines and wild-type were depending on the growth environments and stage of the plants, and further efforts are required to reveal the underlying mechanisms.

Water stress may greatly limit plant functionality and growth. Stomatal closure and consequently reduced transpiration are considered as early and sensitive plant responses to drought and salinity stress. An important consequence of stomatal closure under water stress is the rise of leaf temperature (Tleaf ), yet Tleaf is not only fluctuating with stomatal closure. It is regulated by several plant parameters and environmental factors. Thermal imaging and different stress indices, incorporating actual leaf/crop temperature and reference temperatures, were developed in previous studies toward normalizing for effects unassociated to water stress on Tleaf , aiming at a more efficient water stress assessment. The concept of stress indices has not been extensively studied on the model plant Arabidopsis thaliana. Therefore, the aim of this study was to examine the different indices employed in previous studies in assessing rosette transpiration rate (E) in Arabidopsis plants grown under two different light environments and subjected to salinity. After salinity imposition, E was gravimetrically quantified, and thermal imaging was employed to quantify rosette (Trosette ) and artificial reference temperature (Twet, Tdry ). Trosette and several water stress indices were tested for their relation to E. Among the microclimatic growth conditions tested, RWSI1 ([Trosette - Twet ]/[Tdry - Twet ]) and RWSI2 ([Tdry - Trosette ]/[Tdry - Twet ]) were well linearly-related to E, irrespective of the light environment, while the sole use of either Twet or Tdry in different combinations with Trosette returned less accurate results. This study provides evidence that selected combinations of Trosette , Tdry , and Twet can be utilized to assess E under water stress irrespective of the light environment.

Predicting the magnitude of this impact urgently requires an understanding of the process connecting atmospheric drying to plant tissue damage. Recent episodes of forest mortality worldwide have been widely attributed to dry conditions causing acute damage to plant vascular systems. Under this scenario vascular embolisms produced by water stress are thought to cause plant death, yet this hypothetical trajectory has never been empirically demonstrated. Here we provide foundational evidence connecting failure in the vascular network of leaves with tissue damage caused during water stress. We observe a catastrophic sequence initiated by water column breakage under tension in leaf veins which severs local leaf tissue water supply, immediately causing acute cellular dehydration and irreversible damage. Tropical forest canopies cycle vast amounts of carbon, yet we still have a limited understanding of how these critical ecosystems will respond to climate warming. We implemented in situ leaf-level + 3°C experimental warming from the understory to the upper canopy of two Puerto Rican tropical tree species, Guarea guidonia and Ocotea sintenisii. After approximately 1 month of continuous warming, we assessed adjustments in photosynthesis, chlorophyll fluorescence, stomatal conductance, leaf traits and foliar respiration. Warming did not alter net photosynthetic temperature response for either species; however, the optimum temperature of Ocotea understory leaf photosynthetic electron transport shifted upward. There was no Ocotea respiratory treatment effect, while Guarea respiratory temperature sensitivity (Q10 ) was down-regulated in heated leaves. The optimum temperatures for photosynthesis (Topt ) decreased 3-5°C from understory to the highest canopy position, perhaps due to upper canopy stomatal conductance limitations. Guarea upper canopy Topt was similar to the mean daytime temperatures, while Ocotea canopy leaves often operated above Topt . With minimal acclimation to warmer temperatures in the upper canopy, further warming could put these forests at risk of reduced CO2 uptake, which could weaken the overall carbon sink strength of this tropical forest.

BACKGROUND Limited and erratic precipitation with inefficient irrigation scheduling often leads to an unstable crop yield and low water-use efficiency (WUE) in semi-arid and semi-humid regions. A 2-year field experiment was conducted to evaluate the effect of three irrigation strategies (conventional irrigation (CK), full-drip irrigation (FI), based on crop evapotranspiration and precipitation forecast, and deficit drip irrigation (DI) (75% FI)) on photosynthetic characteristics, leaf-to-air temperature difference (∆T), grain yield, and the WUE of summer maize. RESULTS The results showed that the daily average net photosynthetic rate (Pn) of DI and FI increased by 25.4% and 25.8% at jointing stage in 2018, and 26.3% and 26.5% at grain-filling stage in 2019 compared with CK, respectively. At jointing stage in 2018 and grain-filling stage in 2019, the transpiration rate (Tr) of DI was significantly lower than that of FI (P 0.05). Stomatal optimization models can improve estimates of water and carbon fluxes with relatively low complexity, yet there is no consensus on which formulations are most appropriate for ecosystem-scale applications. We implemented three existing analytical equations for stomatal conductance, based on different water penalty functions, in a big-leaf comparison framework, and determined which optimization principles were most consistent with flux tower observations from different biomes. We used information theory to dissect controls of soil water supply and atmospheric demand on evapotranspiration in wet to dry conditions and to quantify missing or inadequate information in model variants. We ranked stomatal optimization principles based on parameter uncertainty, parsimony, predictive accuracy, and functional accuracy of the interactions between soil moisture, vapor pressure deficit, and evapotranspiration. Performance was high for all model variants. Water penalty functions with explicit representation of plant hydraulics did not substantially improve predictive or functional accuracy of ecosystem-scale evapotranspiration estimates, and parameterizations were more uncertain, despite having physiological underpinnings at the plant level. Stomatal optimization based on water use efficiency thus provided more information about ecosystem-scale evapotranspiration compared to those based on xylem vulnerability and proved more useful in improving ecosystem-scale models with less complexity.

Bamboos are arborescent monocotyledons that have no secondary growth, but can continually produce conduits with diameters appropriate to the current size of the plant. Here, we studied bamboo hydraulic architecture to address the mechanisms involved in compensating for the increase in hydraulic resistance during ontogeny. We measured the hydraulic weighted vessel diameters (Dh ) at different distances from the apex along the stem of Bambusa textilis. The hydraulic resistance of different components and individuals of different heights were quantified using the high-pressure flowmeter method. The Dh showed tip-to-base widening with a scaling exponent in the range of those reported for trees. Although theoretical hydraulic conductivity decreased from base-to-tip, leaf-specific conductivity did not change. Leaves contributed the most to the whole-shoot hydraulic resistance, followed by the leaf-bearing branches. Roots contributed c. 13% to whole-plant resistance. Interestingly, taller individuals showed lower whole-shoot resistance owing to an increased number of resistances in parallel (side-branches), while leaf-specific resistance was independent of plant size. Tip-to-base vessel widening and height-independent constant leaf-specific conductance could be mechanisms for hydraulic optimization in B. textilis. Similar patterns have also been found in woody plants with secondary growth, but this bamboo exhibits them without secondary growth.

Elevated tropospheric ozone concentration (O3) significantly reduces photosynthesis and productivity in several C4 crops including maize, switchgrass and sugarcane. However, it is unknown how O3 affects plant growth, development and productivity in sorghum (Sorghum bicolor L.), an emerging C4 bioenergy crop. Here, we investigated the effects of elevated O3 on photosynthesis, biomass and nutrient composition of a number of sorghum genotypes over two seasons in the field using free‐air concentration enrichment (FACE), and in growth chambers. We also tested if elevated O3 altered the relationship between stomatal conductance and environmental conditions using two common stomatal conductance models. Sorghum genotypes showed significant variability in plant functional traits, including photosynthetic capacity, leaf N content and specific leaf area, but responded similarly to O3. At the FACE experiment, elevated O3 did not alter net CO2 assimilation (A), stomatal conductance (g s), stomatal sensitivity to the environment, chlorophyll fluorescence and plant biomass, but led to reductions in the maximum carboxylation capacity of phosphoenolpyruvate and increased stomatal limitation to A in both years. These findings suggest that bioenergy sorghum is tolerant to O3 and could be used to enhance biomass productivity in O3 polluted regions., Sorghum (Sorghum bicolor L.), a widely adapted and highly productive C4 grass, has potential as a bioenergy feedstock. In this study, we found that sorghum genotypes exhibit significant variability in plant functional traits, but a similar response to O3. Elevated O3 did not alter net CO2 assimilation (A), stomatal conductance (g s) and chlorophyll fluorescence in multiple genotypes of sorghum in both field and chamber experiments. We suggest that sorghum could be used to enhance biomass production for bioenergy in O3 polluted regions.

Abscisic acid (ABA) signals regulating stomatal aperture and water loss are usually studied in detached leaves or isolated epidermal peels, and at infrequent timepoints. Measuring stomatal ABA-responses in attached leaves across a time-course enables the study of stomatal behaviour in the physiological context of the plant. Infra-red thermal imaging is often used to characterise steady-state stomatal conductance via comparisons of leaf surface temperature but is rarely used to capture stomatal responses over time, or across different leaf surfaces. We used dynamic thermal imaging as a robust, but sensitive, tool to observe stomatal ABA-responses in a whole plant context. We detected stomatal responses to low levels of ABA in both monocots and dicots and identified differences between the responses of different leaves. Using whole plant thermal imaging, stomata did not always behave as described previously for detached samples: in Arabidopsis we found no evidence for fast systemic ABA-induced stomatal closure, and in barley we observed no requirement for exogenous nitrate during ABA-induced stomatal closure. Thus, we recommend dynamic thermal imaging as a useful approach to complement detached sample assays for the study of local and systemic stomatal responses, and molecular mechanisms underlying stomatal responses to ABA in the whole plant context. The aim of the study was to understand the hydraulic response to salt stress of the root system of the comparatively salt-tolerant crop barley (Hordeum vulgare L.). We focused on the transcellular path of water movement across the root cylinder that involves the crossing of membranes. This path allows for selective water uptake, while excluding salt ions. Hydroponically grown plants were exposed to 100 mM NaCl for 5-7 days and analysed when 15-17 days old. A range of complementary and novel approaches was used to determine hydraulic conductivity (Lp). This included analyses at cell, root and plant level and modelling of water flow. Apoplastic barrier formation and gene expression level of aquaporins (AQPs) was analysed. Salt stress reduced the Lp of root system through reducing water flow along the transcellular path. This involved changes in the activity and gene expression level of AQPs. Modelling of root-Lp showed that the reduction in root-Lp did not require added hydraulic resistances through apoplastic barriers at the endodermis. The bulk of data points to a near-perfect semi-permeability of roots of control plants (solute reflection coefficient σ ~1.0). Roots of salt-stressed plants are almost as semi-permeable (σ > 0.8).

Drought is a cyclical phenomenon in natural environments. During dehydration, stomatal closure is mainly regulated by abscisic acid (ABA) dynamics that limit transpiration in seed plants, but following rehydration, the mechanism of gas exchange recovery is still not clear. In this study, leaf water potential (ψleaf ), stomatal conductance (gs ), leaf hydraulic conductance (Kleaf ), foliar ABA level, ethylene emission rate in response to dehydration and rehydration were investigated in four Caragana species with isohydric (Caragana spinosa and C. pruinosa) and anisohydric (C. intermedia and C. microphylla) traits. Two isohydric species with ABA-induced stomatal closure exhibited more sensitive gs and Kleaf to decreasing ψleaf than two anisohydric species which exhibited a switch from ABA to water potential-driven stomatal closure during dehydration. Following rehydration, the recovery of gas exchange was not associated with a decrease in ABA level but was strongly limited by the degradation of the ethylene emission rate in all species. Furthermore, two anisohydric species with low drought-induced ethylene production exhibited more rapid recovery in gas exchange upon rehydration. Our results indicated that ethylene is a key factor regulating the drought-recovery ability in terms of gas exchange, which may shape species adaptation to drought and potential species distribution.

Intrinsic transpiration efficiency (i TE), the ratio of photosynthesis (A) to stomatal conductance (g s), is considered a useful trait for improving productivity; however, higher i TE with high A is more desirable than that with low g s. This study dissects i TE of 20 sugarcane genotypes to understand its relationship with total dry matter (TDM) and cane yield (TCH) under irrigated and rainfed conditions. Water stress reduced mean A and g s by 56 and 61%, and mean TDM and TCH by 55 and 59%, respectively; however, genotype × irrigation treatment interaction was smaller than genotype variance. Mean i TE increased from 117.4 μmol mol−1 in the irrigated treatment to 130.6 μmol mol−1 in the rainfed treatment. In irrigated conditions, i TE had high heritability (H 2 b = 0.67) and significant genetic correlation with TDM (r g = 0.58) and TCH (r g = 0.72). Under water stress, at g s below 0.1 mol m−2 s−1, non‐stomatal limitation to A was evident and i TE had low heritability (H 2 b = 0.2). Whereas in the g s range of 0.1–0.4 mol m−2 s−1, heritability of i TE (H 2 b = 0.63) and its genetic correlation with TDM (r g = 0.78) and TCH (r g = 0.75) were maximised. There was significant genotypic variation in photosynthetic capacity (A c), and the differences were related to TDM and i TE. Selecting genotypes with higher i TE and A c could offer potential for improving productivity without the unfavourable effect of low g s.

While plant height is the main driver of variation in mean vessel diameter at the stem base (VD) across angiosperms, climate, specifically temperature, does play an explanatory role, with vessels being wider with warmer temperature for plants of the same height. Using a comparative approach sampling 537 species of angiosperms across 19 communities, we rejected selection favouring freezing-induced embolism resistance as being able to account for wider vessels for a given height in warmer climates. Instead, we give reason to suspect that higher vapour pressure deficit (VPD) accounts for the positive scaling of height-standardized VD (and potential xylem conductance) with temperature. Selection likely favours conductive systems that are able to meet the higher transpirational demand of warmer climates, which have higher VPD, resulting in wider vessels for a given height. At the same time, wider vessels are likely more vulnerable to dysfunction. With future climates likely to experience ever greater extremes of VPD, future forests could be increasingly vulnerable.

International audience; Plants continue to lose water from their leaves even after complete stomatal closure. Although this minimum conductance (g(leaf-res)) has substantial impacts on strategies of water use and conservation, little is known about the potential drivers underlying the variability of this trait across species. We thus untangled the relative contribution of water leaks from the cuticle and stomata in order to investigate how the variability in leaf morphological and anatomical traits is related to the variation ing(leaf-res)and carbon assimilation capacity across 30 diverse species from the Brazilian Cerrado. In addition to cuticle permeance, water leaks from stomata had a significant impact ong(leaf-res). The differential pattern of stomata distribution in the epidermis was a key factor driving this variation, suggesting the existence of a trade-off between carbon assimilation and water loss throughg(leaf-res). For instance, higherg(leaf-res), observed in fast-growing species, was associated with the investment in small and numerous stomata, which allowed higher carbon assimilation rates but also increased water leaks, with negative impacts on leaf survival under drought. Variation in cuticle structural properties was not linked tog(leaf-res). Our results therefore suggest the existence of a trade-off between carbon assimilation efficiency and dehydration tolerance at foliar level.

International audience; Defining plant hydraulic traits is central to the quantification of ecohydrological processes ranging from land-atmosphere interactions, to tree mortality and water-carbon budgets. A key plant trait is the xylem specific hydraulic conductivity (K-x), that describes the plant's vascular system capacity to transport water. While xylem's vessels and tracheids are dead upon maturity, the xylem is neither inert nor deadwood, various components of the sapwood and surrounding tissue remaining alive and functional. Moreover, the established definition of K-x assumes linear relations between water flux and pressure gradient by tacitly considering the xylem as a "passive conduit". Here, we re-examine this notion of an inert xylem by systematically characterizing xylem flow in several woody plants using K-x measurements under constant and cyclic pressure gradients. Results show a temporal and pressure gradient dependence of K-x. Additionally, microscopic features in "living branches" are irreversibly modified upon drying of the xylem, thus differentiating the macroscopic definition of K-x for living and dead xylem. The findings highlight the picture of the xylem as a complex and delicate conductive system whose hydraulic behaviour transcends a passive gradient-based flow. The study sheds new light on xylem conceptualization, conductivity measurement protocols, in situ long-distance water transport and ecosystem modelling.

Water released from wood during transpiration (capacitance) can meaningfully affect daily water use and drought response. To provide context for better understanding of capacitance mechanisms, we investigated links between capacitance and wood anatomy. On twigs of 30 temperate angiosperm tree species, we measured day capacitance (between predawn and midday), water content, wood density, and anatomical traits, that is, vessel dimensions, tissue fractions, and vessel-tissue contact fractions (fraction of vessel circumference in contact with other tissues). Across all species, wood density (WD) and predawn lumen volumetric water content (VWCL-pd ) together were the strongest predictors of day capacitance (r2adj = .44). Vessel-tissue contact fractions explained an additional ~10% of the variation in day capacitance. Regression models were not improved by including tissue lumen fractions. Among diffuse-porous species, VWCL-pd and vessel-ray contact fraction together were the best predictors of day capacitance, whereas among semi/ring-porous species, VWCL-pd , WD and vessel-fibre contact fraction were the best predictors. At predawn, wood was less than fully saturated for all species (lumen relative water content = 0.52 ± 0.17). Our findings imply that day capacitance depends on the amount of stored water, tissue connectivity and the bulk wood properties arising from WD (e.g., elasticity), rather than the fraction of any particular tissue.

Improving plant water-use efficiency (WUE) is important to plant survival and crop yield in the context of water limitation. In this study, SlTLFP8 (Tubby-like F-box protein 8) was identified as an osmotic-induced gene in tomato. Transgenic tomato with up-regulated expression of SlTLFP8 showed enhanced water-deficient resistance, whereas knockout mutants generated by CRISPR/Cas9 were more sensitive to water deficit. SlTLFP8 overexpression significantly enhanced WUE by suppressing transpiration under both water-sufficient and water-deficient conditions. Further study showed that overexpressing SlTLFP8 significantly increased leaf epidermal cell size and thereby decreased stomatal density 10-20%, conversely SlTLFP8 knockout resulted in decreased cell size and thereby increased stomatal density 20-50%. SlTLFP8 overexpression and knockout modulated ploidy levels in leaf cells. Changes in expression of cell cycle related genes also indicated that SlTLFP8 affected cell size and stomatal density through endocycle transition. Despite changes in stomata density and transpiration, altering the expression of SlTLFP8 did not change photosynthesis. Additionally, biomass was not altered and there was little difference in fruit yield for transgenic and wild type lines under water-sufficient and water-deficient conditions. Our results demonstrate the effect of SlTLFP8 on endoreduplication and the potential of SlTLFP8 for improvement of WUE. BRIEF SUMMERY: This work found a new mechanism of TLP (Tubby like protein) response to water-deficient stress. SlTLFP8, a member of TLP family, regulates water-deficient resistance by modulating water loss via affecting stomatal density. Expression of SlTLFP8 was induced by osmotic stress. Transgenic tomato lines with SlTLFP8 overexpression or SlTLFP8 knockout showed significantly differences in water-use efficiency (WUE) and water-deficient resistance. The difference of leaf water loss caused by transpiration is the main explanation of the difference in WUE and water-deficient resistance. Additionally, overexpressing SlTLFP8 significantly decreased stomatal density, while SlTLFP8 knockout resulted in increased stomatal density, and SlTLFP8 affected stomatal density through endoreduplication and altered epidermal cell size. Despite changes in stomata density, altering the expression of SlTLFP8 did not result in distinct changes in photosynthesis, biomass and yield of tomato.

Water uptake from the soil via a vapor pathway was tested. Viburnum suspensum L. seedlings were divided into: (1) irrigated, (2) drought with vapor, and (3) drought without vapor treatments. Each plant was placed into a larger bucket containing deuterium labelled water as a vapor source (vapor treatment) or no water (drought and irrigation treatments). We also tested whether uptake via a vapor pathway could mitigate drought effects. Net CO2 assimilation (A), transpiration (E) and stomatal conductance (gs) were measured daily until the first visible signs of stress. Soil water content, stem water potential (Ψ), and the stable hydrogen isotope ratio (δ2 H) of soil and plant xylem water were then measured in all treatments. We show that water is taken up by plants through the vapor phase in dry soils. The δ2 H values of the soil water in the vapor treatment were highly enriched compared to the background isotope ratios of the non-vapor exposed irrigated and drought treatments. Stem water δ2 H values for the vapor treatment were significantly greater than those for irrigation and drought treatments not exposed to isotopically-enriched vapor. In this experiment, movement of water to the plant via the vapor phase did not mitigate drought effects. Net CO2 assimilation, E, plant Ψ and gs significantly decreased in the drought and vapor treatments relative to the controls, with no significant differences between vapor and drought treatments. Terminal drought substantially reduces chickpea yield. Reducing water use at vegetative stage by reducing transpiration under high vapor pressure deficit (VPD), i.e. under dry/ hot conditions, contributes to drought adaptation. We hypothesized that this trait could relate to differences in a genotype’s dependence on root water transport pathways and hydraulics. • Transpiration rate responses in conservative and profligate chickpea genotypes were evaluated under increasing VPD in the presence/absence of apoplastic and cell-to-cell transport inhibitors. • Conservative genotypes ICC 4958 and ICC 8058 restricted transpiration under high VPD compared to the profligate genotypes ICC 14799 and ICC 867. Profligate genotypes were more affected by aquaporin inhibition of the cell-to-cell pathway than conservative genotypes, as measured by the root hydraulic conductance and transpiration under high VPD. Aquaporin inhibitor treatment also led to a larger reduction in root hydraulic conductivity in profligate than in conservative genotypes. In contrast, blockage of the apoplastic pathway in roots decreased transpiration more in conservative than in profligate genotypes. Interestingly, conservative genotypes had high early vigour, whereas profligate genotypes had low early vigour. • In conclusion, profligate genotypes depend more on the cell-to-cell pathway, which might explain their higher root hydraulic conductivity, whereas water-saving by restricting transpiration led to higher dependence on the apoplastic pathway. This opens the possibility to screen for conservative or profligate chickpea phenotypes using inhibitors, itself opening to the search of the genetic basis of these differences.

Despite representing a sizeable fraction of the canopy, very little is known about leaf sheath gas exchange in grasses. Specifically, estimates of sheath stomatal conductance, transpiration and photosynthesis along with their responses to light, CO2 and vapour pressure deficit (VPD) are unknown. Furthermore, the anatomical basis of these responses is poorly documented. Here, using barley as a model system, and combining leaf-level gas exchange, whole-plant gravimetric measurements, transpiration inhibitors, anatomical observations, and biophysical modelling, we found that sheath and blade stomatal conductance and transpiration were similar, especially at low light, in addition to being genotypically variable. Thanks to high abaxial stomata densities and surface areas nearly half those of the blades, sheaths accounted for up to 17% of the daily whole-plant water use, which -surprisingly- increased to 45% during the nighttime. Sheath photosynthesis was on average 17-25% that of the blade and was associated with lower water use efficiency. Finally, sheaths responded differently to the environment, exhibiting a lack of response to CO2 but a strong sensitivity to VPD. Overall, these results suggest a key involvement of sheaths in feedback loops between canopy architecture and gas exchange with potentially significant implications on adaptation to current and future climates in grasses.

Many plant water use models predict leaves maximize carbon assimilation while minimizing water loss via transpiration. Alternate scenarios may occur at high temperature, including heat avoidance, where leaves increase water loss to evaporatively cool regardless of carbon uptake; or heat failure, where leaves non-adaptively lose water also regardless of carbon uptake. We hypothesized that these alternative scenarios are common in species exposed to hot environments, with heat avoidance more common in species with high construction cost leaves. Diurnal measurements of leaf temperature and gas exchange for 11 Sonoran Desert species revealed that 37% of these species increased transpiration in the absence of increased carbon uptake. High leaf mass per area partially predicted this behaviour (r2 = 0.39). These data are consistent with heat avoidance and heat failure, but failure is less likely given the ecological dominance of the focal species. These behaviours are not yet captured in any extant plant water use model.

A mechanistic understanding of plant photosynthetic response is needed to reliably predict changes in terrestrial carbon (C) gain under conditions of chronically elevated atmospheric nitrogen (N) deposition. Here, using 2,683 observations from 240 journal articles, we conducted a global meta-analysis to reveal effects of N addition on 14 photosynthesis-related traits and affecting moderators. We found that across 320 terrestrial plant species, leaf N was enhanced comparably on mass basis (Nmass , +18.4%) and area basis (Narea , +14.3%), with no changes in specific leaf area or leaf mass per area. Total leaf area (TLA) was increased significantly, as indicated by the increases in total leaf biomass (+46.5%), leaf area per plant (+29.7%), and leaf area index (LAI, +24.4%). To a lesser extent than for TLA, N addition significantly enhanced leaf photosynthetic rate per area (Aarea , +12.6%), stomatal conductance (gs , +7.5%), and transpiration rate (E, +10.5%). The responses of Aarea were positively related with that of gs , with no changes in instantaneous water-use efficiency and only slight increases in long-term water-use efficiency (+2.5%) inferred from 13 C composition. The responses of traits depended on biological, experimental, and environmental moderators. As experimental duration and N load increased, the responses of LAI and Aarea diminished while that of E increased significantly. The observed patterns of increases in both TLA and E indicate that N deposition will increase the amount of water used by plants. Taken together, N deposition will enhance gross photosynthetic C gain of the terrestrial plants while increasing their water loss to the atmosphere, but the effects on C gain might diminish over time and that on plant water use would be amplified if N deposition persists.

Flowers underpin plant evolution, genetic legacy and global food supply. They are exposed to similar evaporative conditions as leaves, yet floral physiology is a product of different selective forces. We used Tanacetum cinerariifolium, a perennial daisy, to examine the response of flowers to whole-plant water stress, determining if flowers constitute a liability during drought, and how this species has adapted to minimize risk associated with reproduction. We determined the relative transpiration cost of flowers and leaves and confirmed that flowers in this species are xylem-hydrated. The relative water stress tolerance of leaves and flowers then was compared using xylem vulnerability measurements linked with observed tissue damage during an acute drought treatment. Flowers were a major source of water loss during drought but the xylem supplying them was much more vulnerable to cavitation than leaves. This xylem vulnerability segmentation was confirmed by observations that most flowers died whereas leaves were minimally affected during drought. Early cavitation and hydraulic isolation of flowers during drought benefits the plant by slowing the dehydration of perennial vegetative organs and delaying systemic xylem damage. Our results highlight the need to understand flower xylem vulnerability as a means of predicting plant reproductive failure under future drought.

Barley is described to mostly use sucrose for night carbon requirements. To understand how the transient carbon is accumulated and utilized in response to cold, barley plants were grown in a combination of cold days and/or nights. Both daytime and night cold reduced growth. Sucrose was the main carbohydrate supplying growth at night, representing 50-60% of the carbon consumed. Under warm days and nights, starch was the second contributor with 26% and malate the third with 15%. Under cold nights, the contribution of starch was severely reduced, due to an inhibition of its synthesis, including under warm days, and malate was the second contributor to C requirements with 24-28% of the total amount of carbon consumed. We propose that malate plays a critical role as an alternative carbon source to sucrose and starch in barley. Hexoses, malate, and sucrose mobilization and starch accumulation were affected in barley elf3 clock mutants, suggesting a clock regulation of their metabolism, without affecting growth and photosynthesis however. Altogether, our data suggest that the mobilization of sucrose and malate and/or barley growth machinery are sensitive to cold.

International audience; Identifying the drivers of stomatal closure and leaf damage during stress in grasses is a critical prerequisite for understanding crop resilience. Here, we investigated whether changes in stomatal conductance (g(s)) during dehydration were associated with changes in leaf hydraulic conductance (K-leaf), xylem cavitation, xylem collapse, and leaf cell turgor in wheat (Triticum aestivum). During soil dehydration, the decline of g(s) was concomitant with declining K-leaf under mild water stress. This early decline of leaf hydraulic conductance was not driven by cavitation, as the first cavitation events in leaf and stem were detected well after K-leaf had declined. Soil drying is a limiting factor for crop production worldwide. Yet, it is not clear how soil drying impacts water uptake across different soils, species, and root phenotypes. Here we ask (1) what root phenotypes improve the water use from drying soils? and (2) what root hydraulic properties impact water flow across the soil-plant continuum? The main objective is to propose a hydraulic framework to investigate the interplay between soil and root hydraulic properties on water uptake. We collected highly resolved data on transpiration, leaf and soil water potential across 11 crops and 10 contrasting soil textures. In drying soils, the drop in water potential at the soil-root interface resulted in a rapid decrease in soil hydraulic conductance, especially at higher transpiration rates. The analysis reveals that water uptake was limited by soil within a wide range of soil water potential (-6 to -1000 kPa), depending on both soil textures and root hydraulic phenotypes. We propose that a root phenotype with low root hydraulic conductance, long roots and/or long and dense root hairs postpones soil limitation in drying soils. Near-term iterative forecasting is a powerful tool for ecological decision support and has the potential to transform our understanding of ecological predictability. However, to this point, there has been no cross-ecosystem analysis of near-term ecological forecasts, making it difficult to synthesize diverse research efforts and prioritize future developments for this emerging field. In this study, we analyzed 178 near-term (≤10-yr forecast horizon) ecological forecasting papers to understand the development and current state of near-term ecological forecasting literature and to compare forecast accuracy across scales and variables. Our results indicated that near-term ecological forecasting is widespread and growing: forecasts have been produced for sites on all seven continents and the rate of forecast publication is increasing over time. As forecast production has accelerated, some best practices have been proposed and application of these best practices is increasing. In particular, data publication, forecast archiving, and workflow automation have all increased significantly over time. However, adoption of proposed best practices remains low overall: for example, despite the fact that uncertainty is often cited as an essential component of an ecological forecast, only 45% of papers included uncertainty in their forecast outputs. As the use of these proposed best practices increases, near-term ecological forecasting has the potential to make significant contributions to our understanding of forecastability across scales and variables. In this study, we found that forecastability (defined here as realized forecast accuracy) decreased in predictable patterns over 1–7 d forecast horizons. Variables that were closely related (i.e., chlorophyll and phytoplankton) displayed very similar trends in forecastability, while more distantly related variables (i.e., pollen and evapotranspiration) exhibited significantly different patterns. Increasing use of proposed best practices in ecological forecasting will allow us to examine the forecastability of additional variables and timescales in the future, providing a robust analysis of the fundamental predictability of ecological variables. The expression of HaHB4® transcription factor reduces soybean sensitivity to abiotic stresses, such as water deficit. Studies that quantify the tolerance of HaHB4® soybean to the soil water content in comparison with cultivars currently sown in Brazil are lacking. The objective of this study was to determine the level of drought tolerance of soybean genotypes expressing the HaHB4® transcription factor (TS18-6-610108 and TS18-6-610084) and commercial cultivars (TMG 7063 IPRO and BS IRGA 1642 IPRO) subjected to water deficit during the vegetative phase. We used the fraction of transpirable soil water (FTSW) approach. Parameters related to leaf transpiration, dry matter accumulation, water use efficiency (WUE), and transpiration coefficient (TC) were evaluated in the four soybean genotypes and two treatments (T1-100% replacement of transpired water and T2-without replacement of transpired water). The FTSW threshold for the decline in transpiration was evaluated to identify the onset of water stress in soybean. TS18-6-610108 and BS IRGA 1642 IPRO maintains potential transpiration at low FTSW values. The TS18-6-610108 genotype has 14% higher WUE than the sensitive cultivar under water deficit. Under well-irrigated conditions, the HaHB4® genotypes showed the highest TC values, which indicate well-functioning physiological processes.

In summary, the results of Rodriguez-Dominguez & Brodribb (2020) show that stomatal closure prevents the formation of large gradients in water potential around the roots. They demonstrate the importance of hydraulic processes at the root–soil interface and call for accurate measurements of the hydraulic properties of the rhizosphere and approaches linking belowground and aboveground hydraulic properties. According to their measurements, the loss of conductance of the root–soil interface and of the rhizosphere, more than that of the soil, impacts the total hydraulic conductance of the soil–plant continuum; and this result is partly explained by the prompt closure of stomata. Testing this result across plant species, soil types and for variable atmospheric conditions should be a research priority for understanding the coordination between stomata and soil drying.

Root water uptake is a key ecohydrological process for which a physically-based understanding has been developed in the past decades. However, due to methodological constraints, knowledge gaps remain about the plastic response of whole plant root systems to a rapidly changing environment. - We designed a laboratory system for non-destructive monitoring of stable isotopic composition in plant transpiration of an herbaceous species (Centaurea jacea) and of soil water across depths, taking advantage of newly developed in-situ methods. Daily root water uptake profiles were obtained using a statistical Bayesian multi-source mixing model. - Fast shifts in the isotopic composition of both soil and transpiration water could be observed with the setup and translated into dynamic and pronounced shifts of the root water uptake profile, even in well-watered conditions. - The incorporation of plant physiological and soil physical information into statistical modelling improved the model output. A simple exercise of water balance closure underlined the non-unique relationship between root water uptake profile on the one hand, and water content and root distribution profiles on the other, illustrating the continuous adaption of the plant water uptake as a function of its root hydraulic architecture and soil water availability during the experiment

The CO2 transfer conductance within plant leaves (mesophyll conductance, gm ) is currently not considered explicitly in most land surface models (LSMs), but instead treated implicitly as an intrinsic property of the photosynthetic machinery. Here, we review approaches to overcome this model deficiency by explicitly accounting for gm , which comprises the re-adjustment of photosynthetic parameters and a model describing the variation of gm in dependence of environmental conditions. An explicit representation of gm causes changes in the response of photosynthesis to environmental factors, foremost leaf temperature, and ambient CO2 concentration, which are most pronounced when gm is small. These changes in leaf-level photosynthesis translate into a stronger climate and CO2 response of gross primary productivity (GPP) and transpiration at the global scale. The results from two independent studies show consistent latitudinal patterns of these effects with biggest differences in GPP in the boreal zone (up to ~15%). Transpiration and evapotranspiration show spatially similar, but attenuated, changes compared with GPP. These changes are indirect effects of gm caused by the assumed strong coupling between stomatal conductance and photosynthesis in current LSMs. Key uncertainties in these simulations are the variation of gm with light and the robustness of its temperature response across plant types and growth conditions. Future research activities focusing on the response of gm to environmental factors and its relation to other plant traits have the potential to improve the representation of photosynthesis in LSMs and to better understand its present and future role in the Earth system.

Stem xylem-specific hydraulic conductivity (KS ) represents the potential for plant water transport normalized by xylem cross section, length, and driving force. Variation in KS has implications for plant transpiration and photosynthesis, growth and survival, and also the geographic distribution of species. Clarifying the global-scale patterns of KS and its major drivers is needed to achieve a better understanding of how plants adapt to different environmental conditions, particularly under climate change scenarios. Here, we compiled a xylem hydraulics dataset with 1,186 species-at-site combinations (975 woody species representing 146 families, from 199 sites worldwide), and investigated how KS varied with climatic variables, plant functional types, and biomes. Growing-season temperature and growing-season precipitation drove global variation in KS independently. Both the mean and the variation in KS were highest in the warm and wet tropical regions, and lower in cold and dry regions, such as tundra and desert biomes. Our results suggest that future warming and redistribution of seasonal precipitation may have a significant impact on species functional diversity, and is likely to be particularly important in regions becoming warmer or drier, such as high latitudes. This highlights an important role for KS in predicting shifts in community composition in the face of climate change.

Vapour pressure deficit is a major driver of seasonal changes in transpiration, but photoperiod also modulates leaf responses. Climate warming might enhance transpiration by increasing atmospheric water demand and the length of the growing season, but photoperiod-sensitive species could show dampened responses. Here, we document that day length is a significant driver of the seasonal variation in stomatal conductance. We performed weekly gas exchange measurements across a common garden experiment with 12 oak species from contrasting geographical origins, and we observed that the influence of day length was of similar strength to that of vapour pressure deficit in driving the seasonal pattern. We then examined the generality of our findings by incorporating day-length regulation into well-known stomatal models. For both angiosperm and gymnosperm species, the models improved significantly when adding day-length dependences. Photoperiod control over stomatal conductance could play a large yet underexplored role on the plant and ecosystem water balances.

Salinity is known to affect plant productivity by limiting leaf-level carbon exchange, root water uptake, and carbohydrates transport in the phloem. However, the mechanisms through which plants respond to salt exposure by adjusting leaf gas exchange and xylem-phloem flow are still mostly unexplored. A physically based model coupling xylem, leaf, and phloem flows is here developed to explain different osmoregulation patterns across species. Hydraulic coupling is controlled by leaf water potential, ψl , and determined under four different maximization hypotheses: water uptake (1), carbon assimilation (2), sucrose transport (3), or (4) profit function - i.e. carbon gain minus hydraulic risk. All four hypotheses assume that finite transpiration occurs, providing a further constraint on ψl . With increasing salinity, the model captures different transpiration patterns observed in halophytes (nonmonotonic) and glycophytes (monotonically decreasing) by reproducing the species-specific strength of xylem-leaf-phloem coupling. Salt tolerance thus emerges as plant's capability of differentiating between salt- and drought-induced hydraulic risk, and to regulate internal flows and osmolytes accordingly. Results are shown to be consistent across optimization schemes (1-3) for both halophytes and glycophytes. In halophytes, however, profit-maximization (4) predicts systematically higher ψl than (1-3), pointing to the need of an updated definition of hydraulic cost for halophytes under saline conditions.

The aboveground portion of this pathway is well-described, yet the roots andtheir connection with the soil are still poorly understood due to technical limitations. Here we used a novel rehydration technique to investigate changes in the hydraulic pathwaybetween roots and soil and within the plant body as individual olive plants were subjected to arange of water stresses. Whole root hydraulic resistance (including the radial pathway from xylem to the soil–rootinterface) constituted 81% of the whole-plant resistance in unstressed plants, increasing to>95% under a moderate level of water stress. The decline in this whole root hydraulicconductance occurred in parallel with stomatal closure and contributed significantly to thereduction in canopy conductance according to a hydraulic model. Our results demonstrate that losses in root hydraulic conductance, mainly due to a disconnectionfrom the soil during moderate water stress in olive plants, are profound and sufficient to inducestomatal closure before cavitation occurs. Xylem vulnerability to embolism represents an important trait to determine species distribution patterns and drought resistance. However, estimating embolism resistance frequently requires time-consuming and ambiguous hydraulic lab measurements. Based on a recently developed pneumatic method, we present and test the "Pneumatron", a device that generates high time-resolution and fully automated vulnerability curves. Embolism resistance is estimated by applying a partial vacuum to extract air from an excised xylem sample, while monitoring the pressure change over time. Although the amount of gas extracted is strongly correlated with the percentage loss of xylem conductivity, validation of the Pneumatron was performed by comparison with the optical method for Eucalyptus camaldulensis leaves. The Pneumatron improved the precision of the pneumatic method considerably, facilitating the detection of small differences in the (percentage of air discharged [PAD] < 0.47%). Hence, the Pneumatron can directly measure the 50% PAD without any fitting of vulnerability curves. PAD and embolism frequency based on the optical method were strongly correlated (r2 = 0.93) for E. camaldulensis. By providing an open source platform, the Pneumatron represents an easy, low-cost, and powerful tool for field measurements, which can significantly improve our understanding of plant-water relations and the mechanisms behind embolism.

International audience; The vast majority of measurements in the field of plant hydraulics have been on small-diameter branches from woody species. These measurements have provided considerable insight into plant functioning, but our understanding of plant physiology and ecology would benefit from a broader view, because branch hydraulic properties are influenced by many factors. Here, we discuss the influence that other components of the hydraulic network have on branch vulnerability to embolism propagation. We also modelled the impact of changes in the ratio of root-to-leaf areas and soil texture on vulnerability to hydraulic failure along the soil-to-leaf continuum and showed that hydraulic function is better maintained through changes in root vulnerability and root-to-leaf area ratio than in branch vulnerability. Differences among species in the stringency with which they regulate leaf water potential and in reliance on stored water to buffer changes in water potential also affect the need to construct embolism resistant branches. Many approaches, such as measurements on fine roots, small individuals, combining sap flow and psychrometry techniques, and modelling efforts, could vastly improve our understanding of whole-plant hydraulic functioning. A better understanding of how traits are coordinated across the whole plant will improve predictions for plant function under future climate conditions.

Understanding the pronounced seasonal and spatial variation in leaf carboxylation capacity (Vc,max ) is critical for determining terrestrial carbon cycling in tropical forests. However, an efficient and scalable approach for predicting Vc,max is still lacking. Here the ability of leaf spectroscopy for rapid estimation of Vc,max was tested. Vc,max was estimated using traditional gas exchange methods, and measured reflectance spectra and leaf age in leaves sampled from tropical forests in Panama and Brazil. These data were used to build a model to predict Vc,max from leaf spectra. The results demonstrated that leaf spectroscopy accurately predicts Vc,max of mature leaves in Panamanian tropical forests (R2 = 0.90). However, this single-age model required recalibration when applied to broader leaf demographic classes (i.e. immature leaves). Combined use of spectroscopy models for Vc,max and leaf age enabled construction of the Vc,max -age relationship solely from leaf spectra, which agreed with field observations. This suggests that the spectroscopy technique can capture the seasonal variability in Vc,max , assuming sufficient sampling across diverse species, leaf ages and canopy environments. This finding will aid development of remote sensing approaches that can be used to characterize Vc,max in moist tropical forests and enable an efficient means to parameterize and evaluate terrestrial biosphere models.
