Development of real-time non-invasive imaging probes to assess infiltration and activation of cytotoxic T cells (CTLs) is critical to predict the efficacy of cancer immunotherapy, which however remains challenging. Reported here is an activatable semiconducting polymer nanoprobe (SPNP) for near-infrared fluorescence (NIRF) and photoacoustic (PA) imaging of a biomarker (granzyme B) associated with activation of CTLs. SPNP comprises a semiconducting polymer (SP) conjugated with a granzyme B cleavable and dye-labeled peptide as the side chain, both of which emit NIRF and PA signals. After systemic administration, SPNP passively targets the tumor and in situ reacts with granzyme B to release the dye-labeled peptide, leading to decreased NIRF and PA signals from the dye but unchanged signals from the polymer. Such ratiometric NIRF and PA signals of SPNP correlate well with the expression level of granzyme B and intratumoral population of CTLs. Thus, this study not only presents the first PA probes for in vivo imaging of immune activation but also provides a molecular design strategy that can be generalized for molecular imaging of other immune-related biomarkers. In this paper, the extended Direct Steady Cyclic Analysis method (eDSCA) within the Linear Matching Method Framework (LMMF) is combined with the Stress Modified Ductility Exhaustion method and the modified Cavity Growth Factor (CGF) for the first time. This new procedure is used to systematically investigate the effect of several load parameters including load level, load type and creep dwell duration on the creep–fatigue crack initiation process in a notched specimen. The results obtained are verified through a direct comparison with experimental results available in the literature demonstrating great accuracy in predicting the crack initiation life and the driving mechanisms. Furthermore, this extensive numerical study highlighted the possible detrimental effect of the creep–ratchetting mechanism on the crack growth process. This work has a significant impact on structural integrity assessments of complex industrial components and for the better understanding of creep–fatigue lab scale tests.

We report the synthesis of chemically asymmetric silica nanobottles (NBs) with a hydrophobic exterior surface (capped with 3-chloropropyl groups) and a hydrophilic interior surface for spatially selective cargo loading, and for application as nanoreactors and nanomotors. The silica NBs, which have a "flask bottle" shape with an average diameter of 350 nm and an opening of ca. 100 nm, are prepared by anisotropic sol-gel growth in a water/n-pentanol emulsion. Due to their chemically asymmetric properties, nanoparticles (NPs) with hydrophilic or hydrophobic surface properties can be selectively loaded inside the NBs or on the outside of the NBs, respectively. A high-performance nanomotor is constructed by selectively loading catalytically active hydrophilic Pt NPs inside the NBs. It is also demonstrated that these NBs can be used as vessels for various reactions, such as the in situ synthesis of Au NPs, and using Au NP-loaded NBs as nanoreactors for catalytic reactions.

Summary A comprehensive study of the two sub-steps composite implicit time integration scheme for the structural dynamics is presented in this paper. A framework is proposed for the convergence accuracy analysis of the generalized composite scheme. The local truncation errors of the acceleration, velocity, and displacement are evaluated in a rigorous procedure. The presented and proved accuracy condition enables the displacement, velocity, and acceleration achieving second-order accuracy simultaneously, which avoids the drawback that the acceleration accuracy may not reach second order. The different influences of numerical frequencies and time step on the accuracy of displacement, velocity, and acceleration are clarified. The numerical dissipation and dispersion and the initial magnitude errors are investigated physically, which measure the errors from the algorithmic amplification matrix's eigenvalues and eigenvectors, respectively. The load and physically undamped/damped cases are naturally accounted. An optimal algorithm-Bathe composite method (Bathe and Baig, 2005; Bathe, 2007; Bathe and Noh, 2012) is revealed with unconditional stability, no overshooting in displacement, velocity, and acceleration, and excellent performance compared with many other algorithms. The proposed framework also can be used for accuracy analysis and design of other multi-sub-steps composite schemes and single-step methods under physical damping and/or loading. Summary A degenerated shell element with composite implicit time integration scheme is developed in the present paper to solve the geometric nonlinear large deformation and dynamics problems of shell structures. The degenerated shell element is established based on the eight-node solid element, where the nodal forces, mass matrices, and stiffness matrices are firstly obtained upon virtual velocity principle and then translated to the shell element. The strain field is modified based on the mixed interpolation of tensorial components method to eliminate the shear locking, and the constitutive relation is modified to satisfy the shell assumptions. A simple and practical computational method for nonlinear dynamic response is developed by embedding the composite implicit time integration scheme into the degenerated shell element, where the composite scheme combines the trapezoidal rule with the three-point backward Euler method. The developed approach can not only keep the momentum and energy conservation and decay the high frequency modes but also lead to a symmetrical stiffness matrix. Numerical results show that the developed degenerated shell element with the composite implicit time integration scheme is capable of solving the geometric nonlinear large deformation and dynamics problems of the shell structures with momentum and energy conservation and/or decay. Prediction of time from corrosion initiation to corrosion cracking is a crucial factor in evaluating concrete structures' durability and integrity deteriorated by reinforcement corrosion. This paper presents a mathematical model to predict the time from corrosion initiation to corrosion cracking. A dynamic corrosion rate model based on Bulter–Volmer kinetics and a rust expansion model based on Faraday's law are developed. An analytical model is developed to demonstrate the mechanical consequences of rust buildup around the rebar. The time to corrosion cracking is found to be a function of surrounding concrete material properties, chemical composition of rust, ratio of concrete cover thickness to rebar diameter, and corrosion current density. Comparisons with available experimental results are conducted to show the accuracy and reliability of the present model. Further parametric analysis is carried out to show the influences of various parameters included in the model on the time to corrosion cracking.

In contrast to the consumption of long-chain triacylglycerols (LCT), consumption of medium- and long-chain triacylglycerols (MLCT) reduces the body fat and blood triacylglycerols (TAG) level in hypertriacylglycerolemic Chinese individuals. These responses may be affected by BMI because of obesity-induced insulin resistance. We aimed to compare the effects of consuming MLCT or LCT on reducing body fat and blood TAG level in hypertriacylglycerolemic Chinese subjects with different ranges of BMI. Employing a double-blind, randomized and controlled protocol, 101 hypertriacylglycerolemic subjects (including 67 men and 34 women) were randomly allocated to ingest 25-30 g/day MLCT or LCT oil as the only cooking oil for 8 consecutive weeks. Anthropometric measurements of body weight, BMI, body fat, WC, HC, blood biochemical variables, and subcutaneous fat area and visceral fat area in the abdomen were measured at week 0 and 8. As compared to subjects with BMI 24-28 kg/m(2) in the LCT group, corresponding subjects in the MLCT group showed significantly greater decrease in body weight, BMI, body fat, WC, ratio of WC to HC, total fat area and subcutaneous fat area in the abdomen, as well as blood TAG and LDL-C levels at week 8. Based upon our results, consumption of MLCT oil may reduce body weight, body fat, and blood TAG and LDL-C levels in overweight hypertriacylglycerolemic Chinese subjects but may not induce these changes in normal or obese hypertriacylglycerolemic subjects.

Intended to avoid the complicated computations of elasto-plastic incremental analysis, limit analysis is an appealing direct method for determining the load-carrying capacity of structures. On the basis of the static limit analysis theorem, a solution procedure for lower-bound limit analysis is presented firstly, making use of the element-free Galerkin (EFG) method rather than traditional numerical methods such as the finite element method and boundary element method. The numerical implementation is very simple and convenient because it is only necessary to construct an array of nodes in the domain under consideration. The reduced-basis technique is adopted to solve the mathematical programming iteratively in a sequence of reduced self-equilibrium stress subspaces with very low dimensions. The self-equilibrium stress field is expressed by a linear combination of several self-equilibrium stress basis vectors with parameters to be determined. These self-equilibrium stress basis vectors are generated by performing an equilibrium iteration procedure during elasto-plastic incremental analysis. The Complex method is used to solve these non-linear programming sub-problems and determine the maximal load amplifier. Numerical examples show that it is feasible and effective to solve the problems of limit analysis by using the EFG method and non-linear programming. Copyright © 2007 John Wiley & Sons, Ltd.

A finite element (FE) buckling analysis of rotationally periodic laminated composite shells is performed in this paper. Because the buckling mode of such structures is characterized as rotationally periodic, a corresponding FE buckling analysis scheme is proposed to reduce the computational expenses. Moreover, a new kind of relative degrees-of-freedom element is developed, which can be connected to other solid elements with ease and can yield satisfactory results with a relatively coarse FE mesh. Numerical results of two laminated cylindrical shells subjected to lateral pressure are compared with theoretical ones. The good agreement of them shows the validity of this new computational strategy. Finally, a practical structure is analysed to demonstrate the advantage of this method.
